The effect of Bacillus S11 (BS11)‐ and/or Bacillus P11 (BP11)‐supplemented feeds on the growth performance, survival, immunoenhancement and disease resistance of cultured Pacific white shrimp, Litopenaeus vannamei, was evaluated. Four feeding treatments of (i) regular feed (control), (ii) BS11‐supplemented feed, (iii) BP11‐supplemented feed and (iv) BS11‐and BP11‐supplemented feed were prepared and used for shrimp cultivation in closed recirculating cement tanks (~400 L) in two trials, one for juvenile and PL‐30 shrimp at 60 and 90 days, respectively. The results showed that BS11 gave a higher probiotic potential than BP11 for both age groups of L. vannamei in cultivation, because the average weight and survival of shrimp fed BS11‐supplemented feed were significantly higher (P < 0.05) than those of the control and the other two groups. The survival of shrimp fed either BS11‐or both BS11‐and BP11‐supplemented feed was significantly higher (P < 0.05) than that of the control group. In addition, the highest total haemocyte and granular haemocyte counts and phenoloxidase activity were found in shrimp fed with the BS11‐supplemented feed. After challenge with Vibrio harveyi 639 (~107 CFU mL?1) by immersion, the lowest cumulative death (%) and disease resistance were clearly found in shrimp fed with the BS11‐supplemented feed. 相似文献

The objective of this study was to evaluate the effect of dietary supplementation of a probiotic bacterium, Bacillus subtilis, on the growth, immune response and antioxidant activities of shrimp (Litopenaeus vannamei). Shrimps with an average initial weight of 2.11±0.17 g were randomly assigned to four groups with three replicates. The control group was fed a basal diet, and three treated groups were fed diets supplemented with B. subtilis at doses of 1 × 104, 5 × 104 and 10 × 104 colony‐forming unit (CFU) g?1 feed respectively. After 40 days of culture, 10 shrimps from each replicate were taken randomly for the determination of immune response and oxidization resistance indices. The results showed that the shrimps fed with B. subtilis at a dose of 1 × 104, 5 × 104 CFU g?1 feed showed significantly better growth than that of the control diet. The phenoloxidase activities showed a tendency to increase with an increased dose of B. subtilis in diets but there was no significant difference among the three treated groups. In addition, phenoloxidase activities were found to be significantly higher (P<0.05) in the groups treated with B. subtilis than that of the control group. Shrimps treated with 5 × 104 CFU g?1 feed probiotic bacterium showed the highest lysozyme activity and it was significantly higher (P<0.05) than the other groups. However, there was no significant difference in acid phosphatase and alkaline phosphatase activity across all the groups. The total antioxidant capacity, superoxide dismutase and glutathione peroxidase activities in the probiotic‐treated groups were significantly increased (P<0.05) as compared with the control groups. Both maleic dialdehyde concentration and superoxide anion activities in the probiotic‐treated groups were significantly lower (P<0.05) than those of the control. The probiotic did not affect the nitric oxide synthase and the catalase activity in any of the control and treated groups. These results indicated that the probiotic B. subtilis could significantly promote the growth rate of shrimp by increasing the immune function and antioxidant capacity. The most effective dose of B. subtilis in the diet was 5 × 104 CFU g?1 feed. 相似文献

The explosive multiplication of phytoplankton caused by water eutrophication often occur in the intensive shrimp aquaculture. To comprehensively assess the diversity and community structure of phytoplankton in the waters of typical indoor industrial aquaculture system for Litopenaeus vannamei, a combination of high‐throughput sequencing and morphological identification methods were used in the present study. A total of 41 genera belong to nine phyla were detected by both methods. Chlorophyta, Cyanophyta and Bacillariophyta were found to be three dominant phyla. The high‐throughput sequencing revealed that green algae and cyanobacteria were the most dominant phytoplankton; however, diatoms were the first dominant phytoplankton by using the morphological identification. At the genus level, Picochlorum and Synechococcus were dominant, accounting for 20.94%–97.19% and 0.01%–52.81% of total phytoplankton, revealed by the high‐throughput sequencing. Therefore, more attention should be paid to their ecological impacts on the surrounding sea areas or potential toxicity to shrimp. Cyclotella was the most dominant genus revealed by the morphological identification. High‐throughput sequencing revealed a high diversity and small‐sized phytoplankton which were undetected by microscopy. Both methods provide similar information on the environmental drivers of phytoplankton community. NO3?, NH4+, DIP, DSi, DON and DOP concentrations were the main factors influencing the phytoplankton community structure and diversity. 相似文献

Changes in the diet have been previously reported to alter the gut bacterial profile in several organisms, including shrimp. These shifts in microbial structure either promote beneficial effects to the host or cause diseases. Supplementation of 5‐aminolevulinic acid (5‐ALA), the precursor of tetrapyrroles, has been previously reported to enhance shrimp's immune response. To know whether 5‐ALA has effects on the gut bacterial structure in Pacific white shrimp (Litopenaeus vannamei), we investigated the bacterial communities in the intestine and stomach of L. vannamei using high‐throughput Illumina sequencing of 16S rRNA gene libraries. One week of 5‐ALA supplementation altered the bacterial community structure (beta diversity) in both tissues, as shown by the results of multi‐dimensional scaling (MDS) plots and analysis of similarities (ANOSIM). DESeq2 analysis revealed enrichment of differentially abundant taxa in the 5‐ALA group (e.g. Enhydrobacter and Oceaniovalibus) and control group (e.g. Tenacibaculum and Mycobacterium). Metagenomic predictions suggested that the control group had more KEGG pathways associated with ‘metabolism’ than the 5‐ALA group. This study suggests that 5‐ALA supplementation potentially promotes the formation of a beneficial bacterial community structure in shrimp. This is the first report on the effect of 5‐ALA supplementation on the bacterial community profile in any organism. 相似文献

采用体外模拟胃肠道环境与体内芽孢存活计数相结合方法,考察枯草芽孢杆菌WB600耐受凡纳滨对虾胃、肠道环境及其在凡纳滨对虾肠道内的定殖存活情况。试验结果表明,枯草芽孢杆菌WB600芽孢在不同的pH环境中均表现出很强的耐受性和良好的生长增殖趋势,而营养体的耐受性较差;在含不同浓度胰蛋白酶的模拟肠液中芽孢和营养体均能表现出良好的耐受性,其中芽孢的耐受能力更为显著;当温度高达80℃时,仍有93.9%的芽孢存活,而营养体基本失活;在虾的肠道内容物中能检测到大量的活芽孢,说明芽孢在进入虾肠道内是可以在其内存活并生长定殖的。表明枯草杆菌WB600芽孢比营养体对胃肠道环境和高温具有更强的耐受能力,且能顺利在虾肠道内定殖存活,有望在水产养殖领域发挥益生作用的同时成为新型的口服疫苗载体。 相似文献

To produce a thermostable and neutral phytase (phy) of Bacillus subtilis E20 in Escherichia coli HMS174 and evaluate its efficiency in improving growth performance. The phy C of B. subtilis E20 was expressed in E. coli HMS 174, and then the 42‐kDa recombinant phy C was purified by Ni‐NAT and analysed by SDS–PAGE. The recombinant phy C had optimal ranges of pH of 6 ~ 7 and temperature of 50 ~ 60 °C. A thermostability analysis showed that the enzyme is a thermostable phytase, and around 33% of residual activity was detected after being incubated at 90 ~ 100 °C for 10 min. The recombinant phy C‐pretreated soybean meal for feed preparation improved white shrimp, Litopenaeus vannamei, growth and feed efficiency. Overall, the neutral and thermostable phy C is suitable for aquafeed, and it is able to improve the nutritional utilization, resulting in enhanced shrimp growth and reduced feed costs. 相似文献

This study aimed to improve the nutritional value and utilization of soybean meal (SBM) used as feed ingredient for shrimp by an approach of solid‐state fermentation with Bacillus subtilis E20. The protein content and B. subtilis E20 proliferation significantly increased, as the initial moisture increased from 30% to 50% during fermentation. Compared with SBM, the protein content of FSBM increased by 19% after fermentation, accompanied by an increase of 18.75% in the total hydrolyzed amino acids. The free amino acid profile and content in FSBM also obviously increased by 374.9% compared with SBM. The FSBM is a good substitute for fish meal (FM) in a diet with 37% protein and 7% lipid content. The maximal replacement levels of FM in shrimp diet with SBM and FSBM were 37.42% and 61.67%, respectively, based on the feed efficiency estimated by a broken‐line analysis. The B. subtilis E20‐FSBM can be a potential protein source used as a replacement for FM in shrimp diet. 相似文献

The white shrimp Litopenaeus vannamei, fed immunostimulant‐free, 0.2%β‐glucan and 0.06% glycyrrhizin diets for 18 days, respectively, were challenged with Vibrio alginolyticus at 6.4 × 104 CFU shrimp?1. The total haemocyte count (THC), phenoloxidase (PO) activity, respiratory burst (RB) and superoxide dismutase (SOD) activity changes for a 120‐h period were investigated, and shrimp mortality was also recorded. The results showed that PO activity, RB and SOD activity were significantly higher in shrimp fed the two immunostimulant diets after 18 days than those in shrimp fed immunostimulant‐free diets. The THC and SOD activity decreased significantly from 0 to 24 h post challenge, and then reverted to normal levels at 96 and 72 h respectively. The values for PO activity and RB increased from 0 to 48 h post challenge. Compared with those fed the control diets, shrimp fed immunostimulants had significantly higher PO activity and RB values at 120 h post challenge. Mortalities after challenge with V. alginolyticus were significantly lower in shrimp fed with β‐glucan or glycyrrhizin than in those fed with a diet without immunostimulants. It was concluded that dietary β‐glucan and glycyrrhizin increased the shrimp immunity. Furthermore, β‐glucan caused an increase in some immune parameters 12 h earlier than glycyrrhizin after V. alginolyticus challenge. 相似文献

Acute hepatopancreatic necrosis disease (AHPND) is a devastating condition impacting marine shrimp production worldwide. The objective of this study was to evaluate the effects of four probiotic formulations on Pacific white shrimp (Litopenaeus vannamei) infected with pathogenic Vibrio parahaemolyticus causing AHPND. In addition, bacterial community composition analyses of shrimp gastrointestinal tract (GIT) and aquaculture water before and after infection were conducted by sequencing variable region 4 of the 16S rRNA gene on the Illumina MiSeq platform. Treatments included: (1) Lactobacillus casei (P1), (2) L. casei and Rhodopseudomonas palustris (P2), (3) L. casei, Saccharomyces cerevisiae and R. palustris (P3), and (4) a commercial probiotic EM® (EM), which showed shrimp survival of 11.7%, 26.7%, 36.7% and 73.3% respectively. Treatments causing lower survival showed greater relative abundance (>60%) of family Vibrionaceae in the GIT compared to treatments with higher survival. Diversity indices from GIT samples revealed that treatments showing higher survival had higher Shannon index values (4.69 ± 0.133), compared with those of treatments with lower survival (0.17 ± 0.004). Diversity indices from water samples did not show significant differences after infection (Shannon index 4.64 ± 0.53). The results showed that probiotics could effectively mitigate AHPND while maintaining diverse microbial composition in shrimp GIT, thus maintaining sustainability in the shrimp aquaculture industry. 相似文献

Pyropia yezoensis is one of the most important economical seaweeds across the world and major efforts are underway to increase the production. However, its genome is relatively unexplored. In this study, genome sequence of wild‐type strain LS was determined through paired‐end sequencing of small‐size fractionated genomic library. In total, 283,606 contigs were assembled and 20,620 SSR‐containing sequences were identified. Most of the SSRs contained tri‐ and di‐nucleotide motifs (95.42% and 2.34% respectively), of which GCC was the most abundant (15.7%). Furthermore, 1,253 pairs of primer sets were designed and 124 of them were selected randomly for validation. The results showed that 120 pairs were successful in PCR, and the remaining four failed to generate PCR product at various conditions. Among the 120 pairs of which the PCR products were subsequently sequenced by Sanger sequencing, 104 pairs amplified SSRs with the same motif and repeat times, three pairs with the same motif but different repeat times and 13 pairs with different motifs. Moreover, 21 primer pairs amplified polymorphic products between LS and an improved strain HT, and tri‐nucleotide SSRs showed higher polymorphism frequency when compared to di‐nucleotide SSRs. The purpose of the present study is to profile the microfloral structure in the intestines of the white shrimp, Litopenaeus vannamei, after being fed a control diet, in comparison with the diets containing Bacillus subtilis E20‐fermented soybean meal (FSBM) or an antimicrobial peptide (AMP) isolated from B. subtilis E20‐FSBM (diets designated FSBMD and AMPD, respectively) for 60 days. Metagenomic data showed detection of eight phyla, 20 classes, 40 orders, 68 families and 96 genera. Despite no statistically significant difference, an evaluation of microbial diversity recorded higher species richness, Shannon–Weaver diversity index and evenness in the AMPD and FSBMD groups, compared to the control diet group. Venn diagrams showed that 58 of the operational taxonomic units (OTUs) were shared among the control, FSBMD and AMPD groups, but additional beneficial microbes were only found in the FSBMD and AMPD groups. In addition to the augmentation of beneficial bacteria in the FSBMD and AMPD groups, the abundance of potential pathogens, Vibrio and Flavobacterium, was lower in the gut of shrimp fed the FSBMD and AMPD. The results clearly suggest that the FSBMD and AMPD have the capability to change the microfloral structure of shrimp intestines and could be used for disease prevention in shrimp aquaculture. 相似文献
