蘑菇圈作为青藏高原高寒草甸一种常见生态学现象,也是草地群落植被更新重要场所。通过对青藏高原高寒草甸蘑菇圈区域群落植被特征和经济类群、生活型及演替顶极成分功能群生物量构成、Raunkiaer频度系数、演替度及Spearman秩相关特征进行分析,明晰蘑菇圈不同区域草地植被分异特征,解析蘑菇圈形成对草地植被变化和群落稳定性的作用。结果表明:1)蘑菇圈形成明显改变物种构成,圈上群落优势种为垂穗披碱草和赖草,而圈上和圈外群落皆无绝对优势种;2)蘑菇圈环的形成显著提高了圈上禾草、地下芽植物和降低种的生物量比例,降低其物种多样性;3)蘑菇圈圈上群落处于更高演替阶段,其植物种重要值与经济类群和演替顶极成分的生物量构成变化一致;4)圈上群落主要植物种对正负关联比为0.89,多数种对的种间联结松散,群落稳定性较低;圈内和圈外群落主要植物种对的正负关联比皆>1,多数种对间关联性较强,群落稳定性较强;且蘑菇圈群落中显著关联种对发生在亚优势种与伴生种或伴生种与伴生种之间。研究认为,蘑菇圈形成促进高寒草甸由莎草/杂类草群落向禾草/杂类草群落演替,但降低高寒草甸稳定性。  相似文献

Two groups of laying hens (each n=12) were administered 10 mg/kg enrofloxacin (ENRO) (group A) or 26.6 mg/kg flumequine (FLU) (group B) by gastric catheter daily for five consecutive days. A third group (n=6) was untreated controls. Eggs were collected from day one of treatment and up to 30 days after withdrawal of the drug. Egg white and yolk from each egg were separated, and ENRO, its metabolite ciprofloxacin (CIP) and FLU residues were analysed by a high-performance liquid chromatography method with fluorescence detection. The sum of ENRO and CIP was detectable in egg white on the first day of treatment in high-level concentrations (2007.7 μg/kg) and remained steady during administration. In egg yolk, residues were detectable at day one in lower concentrations (324.4 μg/kg), increasing to the end of treatment. After treatment, these residues decreased and were detectable up to day 8 in egg white, and day 10 in yolk. FLU residues during drug administration in white were detectable in high concentrations from day one to five (6788.4-6525.9 μg/kg), and in yolk, concentrations were lower during administration (629.6-853.9 μg/kg). After drug withdrawal, FLU residues remained longer in egg white (30 days) than in yolk (26 days). For both drugs, differences of concentrations between matrices were significant.  相似文献

OBJECTIVE: To determine the effects of xylazine on canine coronary artery smooth muscle tone. SAMPLE POPULATION: Hearts of 26 healthy dogs. PROCEDURE: Dogs were anesthetized with pentobarbital, and vascular rings of various diameters were prepared from the epicardial coronary arteries. Vascular rings were placed in tissue baths to which xylazine was added (cumulative concentrations ranging from 10(-10) to 10(-4) M), and changes in vascular ring tension were continuously recorded. Effects of the nitric oxide inhibitor NG-nitro-L-arginine methyl ester (L NAME; 5 mM), the alpha1-adrenoceptor antagonist prazosin (10 mM), and the alpha2-adrenoceptor antagonist atipamezole (10 mM) on xylazine-induced changes in vascular ring tension were determined. Results were expressed as percentage of maximal contraction for each vascular ring preparation. RESULTS: Xylazine induced vasoconstriction of small (< 500-microm-diameter) and medium (500- to 1,000-microm-diameter) vascular rings but not of large (> 1,000-microm-diameter) rings. For large vascular rings, L-NAME, atipamezole, and prazosin did not significantly affect the contractile response to xylazine. For small vascular rings, the contractile response following addition of xylazine to rings treated with L-NAME was not significantly different from the contractile response following addition of xylazine to control rings, except at a xylazine concentration of 10(-6) M. Xylazine-induced vasoconstriction of small vascular rings was blocked by atipamezole, but the addition of prazosin had no effect on xylazine-induced vasoconstriction. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that xylazine increases smooth muscle tone of small canine coronary arteriesand that this effect is predominantly mediated by stimulation of alpha2adrenoceptors.  相似文献

In the chicken the tracheal cartilages are complete circular rings of unequal width around their circumference. They are made up of a thick central part and two winglike projections, one cranial, the other caudal, gradually thinning out towards the edges. In each ring one half, left or right, is alternatively broader than the other. At the level of the sagittal median plane, each cranial and caudal margin of the ring displays two notches; at this level the change-over point from the broad to the narrow half of the ring is seen to occur. The interconnections between contiguous rings are characteristic. The broad half of one ring with its winglike projections covers the narrow halves of the rings in front and behind, resulting in a characteristic tile-like pattern. The broader halves of the rings lie always superficially with respect to the narrow ones which are overlaid by them and are deeper situated. This arrangement allows for a high degree of sliding movement of the rings one over the other.  相似文献

Electrophysiological techniques were used to record afferent activity in the superior spermatic nerves of young lambs under general anaesthesia. Receptive fields were identified in the pampiniform plexus and the deep tissue of the testis in response to mechanical stimulation. Application of a standard rubber castration ring to the scrotal neck evoked vigorous afferent activity, including some from formerly silent units with receptive fields particularly in the pampiniform plexus. Some of this multi-unit discharge adapted rapidly within 10 s of the application of the ring and was followed by a discharge pattern which decayed exponentially over 90 min. The rate of decay of this discharge showed more than one exponent (time constant) with inflections at approximately 90 s and 16 min. After the application of the castration ring, quantitatively controlled scrotal compression continued to excite receptors, though a declining frequency was recorded over the period of observation. It was concluded that: (a) rubber castration rings initiated afferent activity which persisted for periods in excess of 90 min, a time course which is similar to the behavioural and humoral changes in the conscious animal; (b) both standard and small rubber castration rings were ineffective in rapidly producing neuronal pressure block of the slowly conducting afferent fibres in the superior spermatic nerve; (c) intra-testicular injection of local anaesthetic rapidly blocked afferent fibres running in the superior spermatic nerve.Abbreviations TTL transistor transistor logic  相似文献

1. Laying hens were given food containing per kg diet: 1 mg pyrimethamine, 10 mg sulphadimethoxine (SDM) or 1 mg pyrimethamine with 10 mg SDM, for 14 days. Residues in the eggs were determined by high performance liquid chromatography. 2. Pyrimethamine was mainly found in yolk. Its concentration was slightly higher when it was given with SDM. 3. The concentrations of SDM in eggs were not different when it was given alone or in combination with pyrimethamine. 4. After withdrawal, pyrimethamine residues decreased below the detection limit (0.02 mg/kg) in the yolk after 11 to 12 d and in the albumen after 2 to 3 d. SDM in yolk fell below the detection limit (0.01 mg/kg) on day 7, whereas in albumen it was not detectable after 2 to 3 d.  相似文献

OBJECTIVE: To evaluate mediolateral, axial, torsional, and craniocaudal bending behavior of 6 distal ring-block configurations commonly used to stabilize short juxta-articular bone segments in small animals. SAMPLE POPULATION: 8 circular external skeletal fixator constructs of each of 6 distal ring-block configurations. The distal ring-block configurations were composed of combinations of complete rings, incomplete rings, and drop wires. PROCEDURE: Constructs were nondestructively loaded in axial compression, craniocaudal bending, mediolateral bending, and torsional loading by use of a materials testing machine. Gap stiffness was determined by use of the resultant load displacement curve. RESULTS: Circular external skeletal fixator configurations and constructs significantly affected gap stiffness in all testing modes. Within each loading mode, gap stiffness was significantly different among most configurations. In general, complete ring configurations were significantly stiffer than similar incomplete ring configurations, and addition of a drop wire to a configuration significantly increased stiffness of that configuration. CONCLUSIONS AND CLINICAL RELEVANCE: When regional anatomic structures permit, the use of complete ring configurations is preferred over incomplete ring configurations. When incomplete ring configurations are used, the addition of a drop wire is recommended.  相似文献

Enzyme-linked immunosorbent assays (ELISAs) have been developed to detect IgG antibodies to Salmonella enteritidis and S typhimurium in the yolk of hens' eggs. Better discrimination and more consistent results were obtained between eggs from experimentally infected and uninfected hens by using saline-dilution of yolk rather than chloroform extraction. Threshold absorbance values were determined in three salmonella-free flocks, and on the basis of these results ELISA optical density values greater than 0.25 were considered to be positive for antibodies to salmonella. Four flocks with a history of salmonella infection were examined; three contained birds which were seropositive for S enteritidis by ELISA and from which S enteritidis was isolated, and a large proportion of eggs from these birds contained antibody to S enteritidis. Eggs from the fourth flock had no detectable antibody, although serum antibody was detected in some birds. No salmonellae were isolated from the yolks of the eggs from any of the four flocks.  相似文献

1. Anticoccidials are widely used as food additives to prevent and treat coccidiosis. They are licensed for use in a prescribed concentration and during a specific time interval with broilers and pullets, but not for laying hens. 2. This study was set up to develop a new high pressure liquid chromatography (HPLC) method to detect clazuril (CZ: (+/-)-2-chloro-alpha-(4-chlorophenyl)-4-(4,5-dihydro-3,5-dioxo-1,2,4-triazin-2(3H)-yl)-benzeneacetonitrile) in egg yolk and albumen and in plasma; to investigate both the presence of residues of CZ in eggs and its pharmacokinetic behaviour in laying hens. 3. A single oral dose (3 mg/kg BW) and multiple oral doses (3 mg/kg BW for 5 d) were investigated. The analytical method gave very good recovery (64 to 74%) in the three different matrices (yolk, albumen and plasma); precision and accuracy were within 11%. 4. After a single dose no residue was detected in eggs collected for up to 10 d, while following multiple dose treatment, CZ residues were detected until 10 d after the end of treatment. The concentration of the drug was higher in yolk than in albumen with a maximum ratio of 10 : 1. 5. Pharmacokinetics of CZ in laying hens after a single dose showed a detectable concentration of the drug up to 24 h. It reached a steady state after the third administration in multiple dosing. 6. Although further studies are necessary, these results indicate that a single oral dose of CZ could be used as an anticoccidial for laying hens due to the lack of residues in eggs.  相似文献

Reasons for performing study: One of the causes of equine laminitis is hyperinsulinaemia, which may be associated with endothelial dysfunction and insulin resistance of vessels. Hypothesis and objectives: Insulin resistance can be induced in palmar digital vessels by continued exposure to insulin in vitro. The objective was to evaluate this in vitro model for future studies. Methods: Palmar digital vessel segments were collected immediately after euthanasia from horses with normal insulin/glucose blood values. Four arterial and 4 venous rings (3 mm wide) were prepared and each ring mounted in a tissue bath, containing Tyrode's solution at 37°C, 2 g tension was applied and the rings allowed to equilibrate for 45 min. Of the 4 rings of each vessel type, one was used as a control. One each of the remaining 3 rings was used for incubation with insulin (to induce resistance), wortmannin (to block PI3‐kinase) and PD‐098059 (to block MAP‐kinase), respectively, for 30 min. After the incubation period, the rings were contracted with phenylephrine. When the response reached a plateau, a single dose of insulin was added to the baths and the response of each ring monitored for 30 min. Results: Insulin relaxed the control rings and those treated with PD 098059 but contracted those pretreated with insulin and wortmannin. Normal relaxation responses of the rings were converted to contractions by insulin resistance. Insulin resistance was confirmed by the qualitative response of insulin‐incubated and wortmannin‐incubated rings. Conclusions: This study demonstrated successful induction of insulin resistance in both arterial and venous rings. It also suggested that the MAP‐kinase pathway plays a minor role in controlling vasomotor tone under normal physiological conditions. Potential relevance: The study suggests that the induction of insulin resistance in equine palmar digital vessel rings is reliable and provides a good in vitro model for studying the vascular insulin resistance which may occur in equine laminitis.  相似文献

The following case report documents the clinical presentation, ultrasonographic appearance, surgical management and outcome of a horse with a traumatic tear extending from the superficial and deep inguinal rings, across the aponeurosis of the external abdominal oblique muscle and inguinal ligament, disrupting the femoral ring, vascular lacuna and peritoneum. This subsequently resulted in fluid accumulation within the femoral canal. This unusual injury and presentation gives rise to discussion of the aetiology and nomenclature that should be used to properly describe this injury.  相似文献

The distribution of Chlorocholine chloride (CCC) in the eggs of laying hens was studied using 15N-CCC. Twelve layers (37 weeks old) were divided into four groups and used in this study consisting of three feeding phases. In phase one (7 days), all the hens received a CCC-free diet [165 g CP/kg dry matter (DM); 11.58 MJ ME/kg DM]. In phase two (11 days), four levels of 15N-CCC: 0, 5, 50 and 250 ppm were added to the respective diets, while in phase three (7 days), CCC-free feed was again offered. Egg samples were taken and the 15N content of egg yolk and albumin were determined. At the end of phase two, there was a significant (p < 0.05) increase in 15N content in egg yolk from hens fed the 50 and 250 ppm CCC diets and in albumin from hens fed the 250 ppm CCC diet. The estimated 15N-CCC residue was 1.71, 6.64, 28.80 ppm in egg yolk and 1.58, 1.08 and 4.50 ppm in albumin from hens fed 5, 50 and 250 ppm CCC, respectively. The CCC residue, from quantitative analysis ranged from 0.21 to 0.93 and 0.93 to 2.43 ppm in yolk of hens fed 50 and 250 ppm CCC, respectively, whereas a range of 0.40-1.46 ppm, was found in the albumin of hens fed 250 ppm. The difference in measured CCC in yolk and albumin and that estimated from 15N-CCC could have been due to breakdown products of 15N-CCC. Seven days after withdrawal of 15N-CCC, the estimated 15N-CCC residue in egg yolk decreased to 0.43, 2.45 and 15.59 ppm, on 5, 50 and 250 ppm CCC dietary treatments, respectively, and to 2.46 ppm in albumin from hens fed 250 ppm CCC. The higher increase in 15N content could have been due to a higher incorporation of 15N-CCC into yolk than albumin during the process of rapid yolk deposition. This experiment showed that consumed CCC is distributed both into yolk and albumin in a dose dependent manner and that CCC is metabolized in laying hens. However, the level of CCC in the diet which could lead to accumulation of detectable CCC levels in eggs as observed in this study, is much higher than the established maximum residual limits in grains.  相似文献

OBJECTIVE: To determine relative effects of ring diameter and wire tension on axial biomechanical properties of 4-ring circular external skeletal fixator constructs. SAMPLE POPULATION: 4-ring circular external skeletal fixator constructs and artificial bone models. PROCEDURE: 4-ring constructs were assembled, using 50-, 66-, 84-, or 118-mm-diameter rings. Two 1.6-mm-diameter fixation wires were attached to opposing surfaces of each ring at intersection angles of 90 degrees and placed through a gap-fracture bone model. Three examples of each construct were loaded in axial compression at 7 N/s to a maximum load of 400 N at each of 4 wire tensions (0, 30, 60, and 90 kg). Response variables were determined from resulting load-displacement curves (construct stiffness, load at 1 mm of displacement, displacement at 400 N). RESULTS: Ring diameter and wire tension had a significant effect on all response variables and had a significant interaction for construct stiffness and displacement at 400 N. Significant differences within all response variables were seen among all 4 ring diameters and all 4 wire tensions. As ring diameter increased, effect of increasing wire tension on gap stiffness and gap displacement at 400 N decreased. Ring diameter had a greater effect than wire tension on all response variables. CONCLUSIONS AND CLINICAL RELEVANCE: Although effects of wire tension decrease as ring diameter increases, placing tension on wires in larger ring constructs is important because these constructs are inherently less stiff. The differential contribution of ring diameter, wire tension, and their interactions must be considered when using circular external skeletal fixators.  相似文献

Without effective pain relief, rubber ring castration of lambs is acutely painful and can also produce chronic pain. The potential of novel, smaller rubber rings to reduce this pain substantially has been investigated. Three groups of eight 2-3 day old lambs, were castrated either with conventional rubber rings (cRR), or novel smaller rubber rings without (nRR) and with local anaesthetic treatment (nRR+La). Behavioural responses and chronic lesions indicative of pain were compared. No major reductions in behavioural responses were produced by castration with nRRs compared with cRRs, but injection of local anaesthetic did reduce these responses significantly. Lambs in all groups developed chronic inflammatory lesions but behavioural evidence of chronic pain was rarely seen. After 28 days, 6/8 necrotic scrotums had been shed by nRR+La; 4/8 by nRR and 2/8 by cRR lambs. The dimensions of rubber rings on necrotic scrotums, suggest that cRRs contracted to their un-stretched size before being shed, whereas nRRs did not. It is concluded that neither of these rubber rings exerted sufficient pressure on trapped nerves to produce anaesthesia rapidly and this may be explained by the physical properties of the tissues at the neck of the scrotum which support the ring.  相似文献

AIMS: To compare two methods of applying rubber rings to the pedicles of yearling stags to induce analgesia in the antler prior to removal. To compare the application of a rubber ring with that of a lignocaine ring block of the antler pedicle on the efficacy and time course of the analgesia produced in yearling stags.

METHODS: Rubber rings were applied to the pedicles of 36 yearling stags that required velvet antler removal. The standard method, a doubled-over ring expanded and lowered down from the distal end of the antler and released midway down the pedicle, was compared with a cable-tie method on the other pedicle, where a ring was pulled around the pedicle by an electrical cable tie threaded through the ring. Brief electrical stimulation (train-of-four mode) was applied proximal and distal to the ring before, and at regular intervals for 1 hour after, application of the ring to a level that produced an auriculopalpebral reflex response.

In a second experiment, each pair of antlers per yearling stag (n=36) was allocated to one of three pairs of treatments, viz no treatment (control) and the cable-tie method as described above, control and local anaesthesia (a ring block of 2 ml 2% lignocaine per cm pedicle circumference), or the cable-tie method and local anaesthesia. Electrical stimulation (tetanic mode) was applied to each antler approximately 25 mm distal to the pedicle/antler junction before, and at intervals up to 1 hour after, application of treatments at a level required to produce a head/neck avoidance behavioural response.

In a third experiment, the two electrical stimulation protocols used above were directly compared by measuring the response of stags (n=8) to one protocol on each pedicle/antler prior to, and at intervals for 1 hour after, application of a rubber ring. At the end of each treatment in all three experiments, analgesia of the antler was established as a nil behavioural response of the stag to a saw cut to the antler (the ‘nick test’).

RESULTS: For both methods of application of a ring the minimum electrical stimulation required distal to the ring to elicit a reflex response increased from around 16 to 55 mA by 60 minutes. In contrast, the electrical stimulation required proximal to the ring remained low (~17.0 mA) throughout. No stag subjected to either of the methods of application responded to the nick test 60 minutes after application of the ring.

The electrical stimulation required to produce a behavioural response increased very rapidly in stags treated with local anaesthetic and at a slower rate in those treated with the cabletie method but showed no significant increase in control stags. After 4 and 30 minutes, for local anaesthesia and the cable-tie method, respectively, 95% of stags were not responding to 80 mA. A significantly greater proportion of stags with antlers treated with local anaesthetic and the cable-tie method did not respond to the nick test than controls, and there was no significant difference in the frequency of the response between stags with treated antlers.

The minimum current required to produce a response proximal to a rubber ring was slightly higher on average for train-of-four electrical stimulus (mean 18.1 (SD 2.6) mA) than for the tetanic mode (mean 11.9 (SD 2.5) mA). The increase in minimum current required to produce the respective response to stimulation distal to the ring was similar for both methods, although the maximum predicted value (67.4 mA) was lower for train-of-four than for the tetanic mode (84.5 mA). No stag responded to the nick test =60 minutes after application of the ring.

CONCLUSIONS: The cable-tie method was no different from the standard method as a procedure for producing analgesia in the antlers of yearling stags and should be accepted as an appropriate procedure for applying analgesic rings to yearling stags. The analgesia produced in the antlers of yearling stags by rubber rings applied by the cable-tie method to the pedicle was similar to that of a lignocaine ring block, but the time course for the development of analgesia was markedly different. Given that a lignocaine ring block is accepted as an adequate method of pain relief for antler removal, the application of rubber rings followed by a period of =30 minutes after application can be advocated as a viable alternative for pain relief.  相似文献

Objective— To report a technique for repair or prevention of inguinal hernia in horses using intracorporeal suture closure of the internal inguinal and vaginal rings.
Study Design— Case series.
Animals— Foals (n=6) and geldings (n=2).
Methods— After selection of a peri-umbilical telescope portal, instrument portals were made unilaterally or bilaterally, as needed. Herniated viscera was reduced, castration (foals) or spermatic cord remnant transection at the internal inguinal ring (adults) performed, and then simple interrupted intracorporeal sutures of synthetic absorbable suture material were used to close the internal inguinal and vaginal rings.
Results— There was no recurrence of inguinal hernia. Postoperative morbidity was minimal and cosmetic outcome was comparable to that achieved with conventional, open techniques.
Conclusion— Laparoscopic, primary closure of the internal inguinal ring by use of intracorporeal sutures provides a secure barrier to herniation and costs considerably less than using endoscopic staples.
Clinical Relevance— Laparoscopic suture closure of the internal inguinal and vaginal rings provides safe and effective repair of inguinal hernia in horses.  相似文献

AIM: To compare the changes in plasma concentrations of noradrenaline, adrenaline and cortisol in lambs after ring castration plus tailing and in calves after dehorning with or without prior local anaesthesia. METHODS: Male lambs were castrated and tailed with rings and calves were dehorned by amputation using a scoop with or without prior local anaesthesia. Blood samples were taken before and after treatment and plasma concentrations of noradrenaline, adrenaline and cortisol were determined. RESULTS: Castration plus tailing of lambs resulted in a rapid increase in noradrenaline concentrations, a lack of an adrenaline response and a marked increase in cortisol concentration. There were similar changes in catecholamine concentrations in calves that were dehorned both with and without local anaesthetic, with adrenaline being elevated within 5 min of treatment and noradrenaline exhibiting a more protracted response. Dehorning caused a marked cortisol increase which was reduced to control concentrations by local anaesthesia for as long as the associated nerve blockade lasted. CONCLUSIONS: The very short-lived adrenaline responses in calves were attributed to dehorning-induced nociceptor input leading to sympathetic stimulation of the adrenal medulla. The longer lasting noradrenaline responses in lambs and calves were thought be due to 'wash-out' of noradrenaline from damaged tissue associated with rings and amputation wounds, respectively.  相似文献

This experiment was conducted to ascertain whether the growth of Salmonella Enteritidis (SE) or Salmonella Typhimurium (ST) would be suppressed in the presence of antibodies contained in egg yolks. Specific pathogen-free chickens (102 days of age) were subcutaneously immunized with oil-adjuvanted bacterin of SE or ST, twice within a four-week interval. During 160 to 170 days of age, eggs were collected, the yolks were removed and mixed with an equal volume of physiological buffered saline, inoculated with ten colony forming units (CFU) of SE or ST, and incubated at 37 degrees C or 20 degrees C for 23 hr. The growth of organisms in each yolk solution was examined. The egg yolk derived from non-immunized hens was examined in the same manner as the controls. There was no difference in the growth titer between the antibody-positive yolk and the negative yolk. The result suggests that the antibodies in the yolk do not influence the growth of each organism, even if the hens are highly immunized.  相似文献

In the developing avian embryo, the main energy source is the yolk. Toward the end of the incubation period, the remaining yolk sac is internalized into the abdominal cavity. At hatch, the remaining yolk comprises 20% of the chick's BW and provides the nutrients needed for maintenance. Posthatch, chicks rapidly initiate the transition from yolk dependence to the utilization of exogenous feed. However, at present, it is not known what types of bacteria are found to be associated with unabsorbed yolk sacs from market-age broilers. For Experiment 1, one hundred 6-wk-old defeathered broiler carcasses were obtained from a commercial processing facility during each of 3 visits. In the second experiment, one hundred 8-wk-old defeathered broiler carcasses were obtained from a different commercial processing plant on 4 separate occasions. For both experiments, each carcass was aseptically opened and inspected for the presence of an unabsorbed yolk sac. Three to 5 carcasses containing a free-floating yolk sac (within the abdominal cavity) and the yolk stalk (without a yolk sac) and 3 to 5 carcasses containing an attached yolk and yolk stalk from each repetition were randomly selected and analyzed for levels and types of total aerobic bacteria (APC), Enterobacteriaceae (ENT), and for the presence of Campylobacter spp. and Salmonella serovars. The APC ranged from log 3.3 to >log 6.0, and the ENT ranged from log 2.8 to >log 6.0. Staphylococcus spp. and Streptococcus spp. were the predominant organisms in APC, whereas Escherichia coli and Hafnia alvei were found to comprise the ENT. Campylobacter spp. was found in 29% of the yolk stalks, 32% of the attached yolk sacs, and 13% of the free-floating yolk sacs. All Campylobacter isolates were determined to be Campylobacter jejuni, except for 1 attached yolk and yolk stalk, which was Campylobacter coli. Salmonella serovars were found in 26% of the yolk stalks, 48% of the attached yolk sacs, and 23% of the free-floating yolk sacs, and the majority of Salmonella isolates were Salmonella Typhimurium. The significance of these bacterial reservoirs and carcass contamination during processing is yet to be determined.  相似文献

Rapid diagnosis of paratuberculosis in infected cattle is important for the successful control of Johne disease within herds. Thus, improving culture methods for Mycobacterium avium subsp. paratuberculosis (M. paratuberculosis) will aid in the identification of asymptomatic animals. Egg yolk is a component of the media used for growing M. paratuberculosis, but its requirement as a supplement has not been reported. Using the ESP II liquid culture system, 2 different sources and 5 concentrations (3.3%, 1.6%, 0.8%, 0.4%, and 0%) of egg yolk were analyzed. Egg yolk source did not affect either recovery rate or time to detection, but both parameters were significantly improved when the 3.3% egg yolk concentrations (final volume) were used over media containing no egg yolk. This study also assessed the recovery of M. paratuberculosis from fecal samples that were cultured multiple times using Herrold egg yolk agar (HEY). Specimens containing greater than 70 cfu/g feces could routinely be identified as positive for M. paratuberculosis after only 1 culture attempt, whereas specimens with fewer bacteria were only intermittently positive, even after 5 replicate cultures. Therefore, this study indicates that the sensitivity of the Trek Diagnostic ESP II liquid culture system for M. paratuberculosis is affected by egg yolk concentration and that single culture attempts using HEY solid media may not identify specimens containing low numbers of bacteria.  相似文献
