Summary Potato tuber dormancy is usually defined as lasting from tuber initiation until a sprout of 2 mm long has been formed under storage conditions optimal for sprouting. We tried to find out whether there is a period during which buds of seed tubers do not grow and whether different batches of seed take the same time to grow sprouts 2 mm long. We measured changes in number of leaf primordia and length of tuber buds of cvs Diamant and Désirée over two years. After early haulm pulling, buds did not grow for at least 60 days (‘Diamant’) or 95 days (‘Désirée’). Buds in both cultivars and two tuber weights of ‘Diamant’ took about 20 days from the estimated onset of sprouting to grow 2 mm long. We question whether this period is always similar and thus whether the moment sprouts 2 mm long have formed is a good criterion for the end of dormancy.  相似文献

Summary In two indoor experiments under short day conditions, the effect of temperature during tuber bulking on dormancy of tubers was investigated for cvs Diamant and Désirée. Temperature treatments started after tuber initiation and lasted for 4 weeks, after which the haulm was removed. In Experiment 1, the day/night temperature regimes 18/12, 22/22, 26/18 and 32/12 °C (T18/12 etc.) were compared. In Experiment 2, three day temperatures (18, 24 and 30 °C) were combined with three night temperatures (12, 18 and 24 °C), resulting in nine treatments. The dormancy of cv. Diamant was shortest after very high day temperatures (30–32 °C), but intermediate day temperatures (22–26 °C) had no shortening effect compared to T18/12. Dormancy of cv. Désirée was not shortened, but rather tended to be prolonged by high temperatures (22–32 °C) during growth. High temperatures during growth resulted in more sprouts per tuber after dormancy had ended.  相似文献

In three experiments with cvs Diamant (short dormancy) and Désirée (long dormancy), the effect of a haulm application of gibberellic acid (GA) on the dormancy of seed potatoes harvested immature was investigated. Several storage temperature regimes were imposed to examine the interaction between GA and storage temperature. The storage regimes included 18 and 28 C continuously, hot pre-treatments of different duration (different periods at 28 C and subsequently 18 C) and a cold pre-treatment (20 days at 2 C and subsequently 18 C). A foliar spray of 375–750 g GA/ha 3–6 days before haulm killing shortened dormancy, and minimally induced sprouting before harvest. The magnitude of the GA effect depended on the cultivar and storage temperature regime. Compared to untreated tubers stored at 18 C, dormancy was shortened by about 40 days by a GA application and storage at 18 C (Diamant) or by about 90 days by a GA application and storage at 28 C (Désirée).  相似文献

Seed tubers that are planted soon after their harvest give low yields because of dormancy and low growth vigor. In the research reported in this paper, we investigated the advancing effect of a haulm application of gibberellic acid (750 g GA/ha) 6 days before haulm pulling and its interaction with storage temperature regimes on the growth vigor of immaturely harvested seed tubers of three cultivars. The effects on tuber yield were also examined in one experiment. The storage regimes were: 18 C continuously, hot pre-treatments of different duration (different periods at 28 C and subsequently 18 C) and a cold pre-treatment (20 days at 2 C and subsequently 18 C). Both a foliar spray with GA and storage at 28 C enhanced physiological aging of the tubers and greatly advanced the growth vigor, without negative effects on the morphology of the plants. There was a clear interaction between the GA treatment and the storage temperature regime. At early planting, the effect of the treatments on tuber yield were small for Diamant (short dormancy), but strongly positive for Désirée and Draga (long dormancy).  相似文献

Summary Eye-bearing slices, cut from healthy potato tubers and placed between Parafilm membranes, were inoculated with potato leafroll virus (PLRV) byMyzus persicae. PLRV was detected by ELISA and by transmission tests in tuber slices and in plants grown from the slices of the susceptible cv. Désirée, but not in those of the resistant cv. Arkula. These results suggest that PLRV replication and transport within tuber phloem is controlled by specific mechanisms of resistance.M. persicae was also able to acquire and transmit PLRV toPl floridana from slices cut from tubers of infected plants. The aphids effectively transmitted PLRV from slices cut from the sprouting rose end but they failed to transmit it from slices cut from the heel end of tubers.  相似文献

Summary The effect of growth conditions (medium and photoperiod) on in vitro tuberisation and microtuber dormancy of the cv. Désirée (Solanum tuberosum subspeciestuberosum) and Imilla Negra (Solanum tuberosum subspeciesandigena) was evaluated. The short photoperiod and presence of CCC in the medium reduced the dry matter of plants as well as the size of microtubers. The two cultivars showed a difference, in favour of Désirée, in the percentage of plants having more than one tuber and in the average time of sprouting after storage. The average time of sprouting was more rapidly obtained under long days and in a medium without CCC. The CCC and short days caused the same dormancy conditions and the same K+ quantity in the microtubers. The microtubers mineral composition is involved in the dormancy status.  相似文献

Summary Seed tubers grown in the northern mountain region — the Golan Heights — were as productive as imported seed tubers. In cvs. Blanka and Désirée, both the locally grown and the imported seed tubers were similar when yield components and tuber fresh weight are considered. In Up-to-Date, the locally grown seeds produced higher yields of marketable tubers (over 60 g/tuber) than those imported. Désirée seed tubers grown for two generations in the Golan Heights did not differ significantly in their productivity as compared with common Golan seeds and with imported ones. Seed tubers harvested from plots which were protected by means of white coarse nets and weekly sprays with insecticides, were virtually virus-free and were used for growing commercial seeds in the Golan. Summary Seed tubers, cv. Désirée, derived from stocks treated annually with thiabendazole were infected with thiabendazole-resistant strains ofHelminthosporium solani andPolyscytalum pustulans. Samples of seed tubers were either untreated or immersed for 5 min in fungicide suspensions of thiabendazole, imazalil or thiabendazole plus imazalil and planted on four farms in 1988 and 1989. After harvest, tuber samples from each treatment were treated with thiabendazole and stored for 6 months. Applying imazalil or thiabendazole plus imazalil to seed tubers decreased the severity of silver scurf and skin spot on stored progeny tubers. Thiabendazole applied to seed tubers or to progeny tubers after harvest did not affect the severity of either disease, but post-harvest treatment decreased the incidence of black scurf after storage.  相似文献

Summary We have evaluated the effect that the, silencing of two genes specifically expressed in conditions of dormancy (A2-1) and sprouting (G1-1) had on tuber dormancy. For this purpose potato plants (Solanum tuberosum L. cv. Désirée) were transformed with the antisense of the genes G1-1 and A2-1 under the control of constitutive 35S CaMV promoter. A first generation of transgenic plants was propagated from axenic stem cuttings and a second generation by tuber planting. The plants obtined were analyzed for the length of dormancy, plant morphology and agronomic characteristics. Statistical analysis of dormancy in lines obtained from the original transformants for the antisense of G1-1 gene showed a significant increase in length as compared with different types of control plants, with few effects on plant vegetative habit and tuber production. In contrast, results obtained on A2-1 antisense transformed plants did not reveal any significant change on the length of dormancy. Here we report small-scale field trials performed with the aim to select and regenerate commercially exploitable potato plants with a stable transgene-dependent phenotype, affecting the length of dormancy.  相似文献

Summary The resistance of ten potato cultivars (Agria, Ajax, Désirée, Liseta, Kennebec, Majestic. Monalisa, Prima, Spunta and Tonda di Berlino) toPhytophthora infestans was analyzed in vitro using 8 fungal strains. An assay based on electrolyte leakage was used for screening leaves and tuber tissues with fungal culture filtrates. With almost all cultivars the resistance of leaves did not correlate with the resistance of tubers. Cv. Ajax appeared the least susceptible in both leaf and tuber tests, while the cv. Prima was the most susceptible in tuber tests.  相似文献

Summary The effect of repeated short periods of water stress during the hot season on six potato cultivars was investigated. The number of tubers per plant tended to decrease in response to water stress in cultivars which had more than 9.8 tubers per control plant (Désirée, Alpha, Elvira). In the other cultivars, no consistent effect of water stress on tuber number per plant was observed. Except for Spunta, the percentage of large tubers (>60 g/tuber) was reduced in the water-stressed plants. Consequently, the average weight of these tubers was greater than that of tubers harvested from the unstressed plants, indicating that the sink force of the individual tuber was not impaired by drought. However, tuber yield per plant was reduced due to drought in all cultivars tested. The smallest decrease in dry matter accumulation as well as the greatest lowering of the osmotic potential in the tuber tissue was found in Alpha. No consistent effect of water stress on the percentage of misshapen and sprouting tubers was observed, indicating the marked effect of the high temperature on these traits. Summary The effects of a single treatment of heat or drought stress imposed at different stages of tuber growth, were investigated in two potato cultivars, Désirée and Cara. Both stresses impaired dry matter accumulation in tubers, and tuber yields. Stress imposed at early stages of tuber growth, when tubers were less than 20 mm in diameter, had little effect on dry matter accumulation, whereas stress imposed later resulted ingnificant losses of dry matter in tubers. Stress imposed during later tuber growth increased the percentage of tubers sprouting in the soil. Tuber malformation was less affected. The adverse effects of heat on tuber yield and quality seemed to be greater than those of drought. Drought increased the content of total soluble solids (TSS) and lowered the osmotic potential in the tuber tissue of both cultivars. These effects were detectable even 55 days after stress relief. Summary Different amounts ofRhizoctonia solani inoculum were applied to sprouted and non-sprouted seed tubers cultivar Désirée during planting in field experiments at Rothamsted in 1984 and 1985. Severity of stem canker and stolon infection increased with increasing amounts of inoculum; with small or medium amounts these infections were most severe on plants from sprouted seed. In 1985 plants grown from non-sprouted, seed had most stem canker when large amounts of inoculum were applied. Severe infection following high inoculum levels delayed shoot emergence, increased the variability in stem height and decreased the mean height of stems and weight of foliage but did not affect the number of stems per plant or final plant populations. Tuber initiation was delayed and tuber numbers slightly decreased by severe infection and at harvest the size distribution of tubers was altered. Tuber yields from severely infected plants were decreased by 15% (sprouted seed) and 19% (non-sprouted seed) in 1984 and by 17 and 28% respectively in 1985.  相似文献

Summary The expression of antioxidant genes has been analyzed in a potato plant and during tuber dormancy. Manganese superoxide dismutase (MnSOD), cytosolic copper and zinc superoide dismutase (Cu/ZnSOD), catalase class II, cytosolic ascorbate peroxidase (APX) and glutathione peroxidase (GPX) are expressed at the RNA level in all the contexts analyzed. By contrast, the expression of the iron superoxide dismutase (FeSOD) and plastidic Cu/ZnSOD seems to be limited to green tissues, as shown by northern blots and native gels. A complex DAB-peroxidase isozyme pattern (using diaminobenzidine as substrate) has been observed in different developmental contexts analyzed, but hardly observed in tubers. During tuber dormancy, MnSOD and cytosolic Cu/ZnSOD activity was relatively constant in both Désirée and Bintje varieties while catalase activity decreases. Moreover, tuber dormancy breakage did not involve significant changes in the activity of these enzymes. On the basis of these results, the possible link between active oxygen species (AOS) metabolism and dormancy is discussed.  相似文献

Summary Four potato lines of cv. Désirée that express the pectate lyase (PL) isoenzyme 3 ofErwinia carotovora subsp.atroseptica were examined in a 4-year field experiment with respect to plant development, tuber yield and resistance of tuber tissue toErwinia soft rot. The PL3 degrades plant cell wall pectin into unsaturated oligogalacturonates eliciting plant defence responses. In one line, enzyme expression was controlled by the CaMV 35S promoter (C) and in three lines it was driven by the potato patatin B33 promoter (D). Plant development of the D-lines in field plots was not distinguishable from that of the non-transgenic counterpart. Also tuber yield was not too different. By contrast, plants of the C-line were smaller than those of the nontransformed counterpart and also showed reduced tuber yield. There were no significant differences in dry mass, starch and protein content of tuber tissue between PL transgenic and non-transgenic potatoes. But compared with the latter, field-grown tubers expressing the PL displayed an enhanced resistance toErwinia soft rot. Thus, average rotting caused byEc-bacteria was diminished in tubers of PL-transgenic lines by 34.1%. The resistance of tubers toEc soft rot was significantly correlated with the PPO activity in tuber tissue.  相似文献

Summary Foliar sprays of 3,5-dichlorophenoxyacetic acid (3,5-D) in June or early July decreased the incidence of common scab by about 30% in four field trials on cv. Maris Piper and cv. Désirée; however, significant decreases in yield and in mean tuber weight were also common. The effects of 3,5-D on potatoes resembled those of the growth retardant daminozide in some ways, but 3,5-D was not persistent enough to have a useful effect on scab in the field.  相似文献

Summary ExpressingE. coli inorganic pyrophosphatase in transgenic plants demonstrated that long distance sucrose transport is dependent on cytosolic pyrophosphate. It was speculated that removal of cytosolic pyrophosphate would impair sucrose utilization during storage of potato tubers and thereby prevent tuber sprouting. To explore this hypothesisSolanum tuberosum var. Désirée plants were transformed with a chimeric PPa gene. FollowingAgrobacterium mediated transformation, metabolite and carbohydrate contents of growing and stored tubers were measured. There was a large accumulation of soluble sugars and a decrease of starch at all developmental stages investigated. The PPase activity in PPaII tubers was parallel with a decrease of PPa, an increase of fructose-2,6-bisphosphate and an increase of UDP-glucose. As expected the amount of hexose-6-phosphates and glycolytic intermediates decreased. As a consequence PPaII tubers did not sprout even after a prolonged storage period of two years. Since the energy status of PPaII tubers is unaltered inhibition of sprouting is most likely due to reduced sucrose export and its subsequent utilization.  相似文献

Summary The cultivars Bintje and Désirée were growth with their shoots, roots and stolons at different temperatures, to study the effects on tuber development. More tuber sites were initiated in ‘Bintje’ when the air, root or stolon temperature was increased but a combined increase of air and root temperature greatly reduced the number of sites. In contrast, each temperature increase reduced the number of tuber sites in ‘Désirée’ but an increase in temperature in one compartment usually increased the number of tubers set. The relative tuber set was enhanced by a combination of high stolon temperature and low root temperature. Especially an increase in air temperature reduced tuber yield and tuber dry-matter content, but increasing stolon temperature had the largest effects on the induction of second growth. Stolon and tuber development, dry-matter partitioning and initiation of second growth responded differently to variation in temperature around different plant parts suggesting that these processes are affected by different regulation mechanisms.  相似文献

Summary One tonne boxes of seed potatoes cv. Désirée contaminated withErwinia carotovora were held in four different types of farm stores. In general, contamination fell rapidly immediately after harvest and rose later in the storage season. The extent of these changes varied considerably with store type. The greatest reduction was from a log count of 5.5 to 2.1 after one month storage, followed by a rise to 5.1 prior to planting. No relationship was found between store temperature, relative humidity and change in contamination. There was some evidence that reduction in contamination was related to duration of ventilation, and rise in contamination to onset of sprouting. A sample held at a constant 4°C showed little reduction in contamination, and no rise occurred later in the storage season.  相似文献

Summary An isolate ofRhizoctonia solani AG8 from a barley plant affected with barley stunt was incorporated into soil and caused severe stunting of cv. Désirée potatoes grown in pots. Fresh weight of foliage, stem bases, roots and tubers were greatly decreased. Most main roots were pruned off and a large number of lateral roots developed, resulting in an unusually fibrous root system; many of these lateral roots were also pruned off. Infected plants did not have stem canker or stolon infection nor was black scurf found on progeny tubers.  相似文献
