A field outbreak of facial eczema occurred during a vitamin B12 response trial in young growing sheep. Pasture cobalt levels were in the low range for sheep <0.08 mg/kg, 1.358 µmol/kg) and mean (of 3) liver vitamin B12 levels in the sheep were low (>100 nmol/kg) during the period in which facial eczema occurred.

Mean serum vitamin B12 levels of the untreated group were low (>185 pmol/1) for the two months (January and February) preceding the period of facial eczema. However, levels showed an approximate 3.5 fold increase in both cobalt supplemented and unsupplemented groups with the onset of facial eczema in March. From February to March the mean serum vitamin B12 and glutamyl transferase (GGT) activity showed parallel increases with a positive correlation (r = 0.73) between log serum vitamin B12 and log serum GGT activity during the period January to July for both groups. This finding suggested that the increase in serum vitamin B12 was due to sporidesmin induced liver damage.

The diagnostic implication is that, in areas where facial eczema is a problem, liver is the sample of choice for determining vitamin B12 status, because sporidesmin toxicity can elevate low serum vitamin B12 levels to diagnostically normal levels.  相似文献

AIM: To determine the effect of increasing doses of long-acting injectable vitamin B12 plus selenium (Se) given pre-mating on the vitamin B12 and Se status of ewes and their lambs from birth to weaning. METHODS: Four groups of 24 Poll Dorset ewes each were injected 4 weeks pre-mating with different doses of a long-acting vitamin B12 + Se product, containing 3 mg vitamin B12 and 12 mg Se per ml. The treatment groups received 5 ml (15 mg vitamin B12 + 60 mg Se), 4 ml (12 mg vitamin B12 + 48 mg Se), 3 ml (9 mg vitamin B12 + 36 mg Se), or no vitamin B12 or Se (control). Twelve of the twin-bearing ewes per group were selected for the study. Efficacy of the product was evaluated from changes in the concentrations of vitamin B12 in serum and liver, and of Se in blood, liver and milk in the ewes during gestation and lactation, and in their lambs from birth to weaning. Pasture samples in paddocks grazed by the ewes and lambs were collected at about 2-monthly intervals from 200-m transects. RESULTS: The flock was Se-deficient, as the mean initial concentration of Se in the blood of ewes was 182 (SE 20.3) nmol/L. Compared with untreated controls, all doses significantly (p < 0.01) increased concentrations of Se in the blood of ewes for at least 300 days. Selenium concentrations in milk were likewise increased throughout lactation, as were those in the blood and liver of lambs. The mean concentration of vitamin B12 in the serum of ewes was initially > 1,000 pmol/L, but this decreased within 28 days to < 460 pmol/L. Treatment with the 5-ml and 4-ml doses raised serum vitamin B12 concentrations of ewes for at least 176 days (p < 0.01), while their lambs had significantly greater concentrations of vitamin B12 in serum and liver for less than 37 days after birth. Tissue concentrations and duration of elevation of both vitamin B12 and Se were proportional to the dose administered. The mean concentrations of Se and cobalt (Co) in the pastures were 32 and 74 microg/kg dry matter (DM), respectively. CONCLUSIONS: Injecting ewes from a Se-deficient flock 4 weeks prior to mating with 48 or 60 mg Se and 12 or 15 mg vitamin B12 increased and maintained the Se status of ewes for at least 300 days, and of their lambs from birth to weaning. The vitamin B12 status of ewes was increased for at least 176 days and that of their lambs for less than 37 days. Due to the proportional nature of the response to increasing dosage, the dose rate of the formulation tested can be adjusted according to the severity of Se and Co deficiency in a flock. CLINICAL SIGNIFICANCE: A single subcutaneous injection of vitamin B12 + Se administered pre-mating to Se-deficient flocks is likely to prevent Se deficiency in ewes and their lambs until weaning, as well as increase the vitamin B12 status of ewes and their lambs until 5 weeks after lambing.  相似文献

AIM: To determine the impact of ingestion of soil on the iodine (I), selenium (Se), copper (Cu) and cobalt (Co; vitamin B12) status of young sheep. METHODS: Twenty young sheep were divided into two groups; one group was fed lucerne pellets, while the other group was fed lucerne pellets plus 100 g soil, for 63 days. At the end of the study the animals were blood-sampled, slaughtered, and the liver removed, and concentrations of I, Cu, vitamin B12 and Se were determined. RESULTS: The ingestion of soil significantly increased concentrations of I and vitamin B12 in serum, but had no effect on concentrations of Cu and Se in serum/blood and liver, and vitamin B12 in liver. CONCLUSION: Ingested soil can be a significant source of I and Co (vitamin B12) for grazing sheep.  相似文献

AIMS: To assess the efficacy of a soluble Vitamin B12 injection in lambs by measuring changes in the serum and liver Vitamin B12 concentrations. METHODS: Thirty-six lambs were injected subcutaneously with 2 mg of soluble Vitamin B12 while another group of 36 served as untreated controls. Blood and liver biopsy samples for Vitamin B12 determinations were collected just before the injection and at days 1, 2, 5, 8, 16, 24, 30 and 45. RESULTS: The serum Vitamin B12 concentrations of the Vitamin B12 treated lambs increased rapidly compared to the untreated lambs. Concentrations peaked at day 2, decreased rapidly to day 8, and then decreased more slowly until day 24 when there were no longer differences between the groups. Liver Vitamin B12 concentrations of the Vitamin B12 treated lambs were significantly greater over days 8-24. CONCLUSION: A subcutaneous injection of 2 mg of soluble Vitamin B12 was effective in increasing and maintaining the Vitamin B12 status of lambs for about 24 days. CLINICAL SIGNIFICANCE: This Vitamin B12 product is only effective for preventing cobalt deficiency in lambs for about 4 weeks.  相似文献

A trial was devised to assess whether the administration of selenium and cobalt together with the anthelmintic mebendazole (Ovitelmin S&C) was safe and could improve the supplies of selenium and cobalt for adult sheep fed a whole grain diet, low in both elements, which produced a steady decrease in blood glutathione peroxidase (GSHPx) and plasma vitamin B12 concentrations. Ovitelmin S&C, when given orally in a single dose as a suspension containing 0.34 mg selenium/ml, and 0.44 mg cobalt/ml (to provide 0.11 mg selenium and 0.15 mg cobalt/kg liveweight) significantly increased the GSHPx activity in blood. After a second dose given 28 days later the rate of change increased from 2.5 to 3.5 u/g haemoglobin/day. The responses in GSHPx were similar for a preparation which contained twice the concentration of selenium. Ovitelmin S&C increased the concentration of vitamin B12 in the plasma by about 1000 pg/ml for four to seven days after each dose and the increases were similar to those observed in sheep treated with an Ovitelmin preparation containing 45 times more cobalt (providing 6.7 mg cobalt/kg liveweight). After 63 days, liver vitamin B12 concentrations were 43 per cent higher in the cobalt treated than in the untreated groups (P less than 0.01) with no differences among the groups given cobalt. Neither adverse reactions nor signs of toxicity followed the administration of Ovitelmin S&C or Ovitelmin containing the higher concentrations of selenium and cobalt.  相似文献

AIM: To develop a long-acting Vitamin B12 injection to prevent Co deficiency in sheep. METHODS: Formulations of microencapsulated Vitamin B12 in lactide-glycolide polymers were injected intramuscularly or subcutaneously into the anterior neck region of groups of 10 lambs and their efficacy determined from changes in serum and liver Vitamin B12 concentrations. RESULTS: The 95:5 lactide glycolide and the 100 lactide formulations containing more than 12.5% Vitamin B12 w/w significantly increased and maintained serum Vitamin B12 concentrations for at least 210 days as well as liver Vitamin B12 concentrations in treated lambs when compared with untreated controls. CONCLUSIONS: Injections of microencapsulated Vitamin B12 in lactide/glycolide copolymers are able to increase and maintain the Vitamin B12 status of lambs for at least 210 days. CLINICAL SIGNIFICANCE: Another option for the prevention of Co deficiency in sheep is now available using a long acting injectable Vitamin B12.  相似文献

Boluses of controlled release glass containing cobalt and weighing approximately either 60 g or 14.5 g were administered to 22 steers and 21 sheep respectively. The steers were housed and slaughtered at intervals between 17 and 145 days after dosing. The boluses released more than 0.85 mg cobalt daily. In both untreated and dosed animals serum and liver vitamin B12 concentrations were at the upper end of the normal range. Two types of glass were administered to sheep. In five wethers one glass released 0.07 mg cobalt per day, and in 16 grazing lambs a second glass released more than 0.15 mg cobalt per day. Fourteen of the boluses were recovered from the lambs up to 276 days after dosing. The concentration of B12 in serum of lambs increased significantly from a mean +/- sd of 1.64 +/- 0.47 to 2.02 +/- 0.04 ng/ml serum and the concentration in liver from 3.84 +/- 0.85 to 4.99 +/- 0.72 micrograms/g dry weight liver.  相似文献

The efficacy and retention of prototype and commercial (Cosecure) soluble-glass boluses containing selenium, cobalt and copper and having a similar size, density and composition were evaluated in separate 12-month trials with sheep grazing low selenium pastures but with adequate cobalt and copper levels. In both trials, sheep confirmed by X-ray as containing a bolus grew at a significantly greater rate than control sheep. This was attributed to the correction of a selenium deficiency. Although liver copper, and liver and serum vitamin B12 levels were significantly greater in treated sheep than controls on some occasions, the differences were not as great or consistent as with blood selenium levels. The rate of bolus loss was high and in both trials, approximately 40% of the sheep had lost the bolus after 6 months. No prototype boluses were present after 12 months. Intact Cosecure boluses were recovered from 3 sheep out of 30 after 12 months whilst one animal contained a part bolus. The recovered boluses were approximately 45% lighter than when originally administered. Bolus loss did not appear to be due to complete dissolution. In sheep which had lost the bolus, blood selenium levels fell with a half-life of 43 +/- 10 days.  相似文献

The efficacies of four methods, used for the prophylaxis of cobalt deficiency in sheep as measured by the elevation of liver and serum vitamin B12 levels, were compared in marginally deficient sheep over 14 weeks. The methods used were weekly drenches of either cobalt sulphate or cobalt chelate (EDTA) three-weekly injections of hydroxocobalamin, and ruminal cobalt pellets. On the basis of elevated liver and serum vitamin B12 levels, chelated cobalt was shown to be available to rumen microflora for the synthesis of vitamin B12. However, at no stage were liver and serum vitamin B12 levels of sheep receiving the chelate significantly different from those receiving the same amount of cobalt as the sulphate. After five, three-weekly injections of hydroxocabalamin liver vitamin B12 levels were significantly higher (p<0.01) than for the other treatments, with the exception of cobalt sulphate. Cobalt pellets led to an initial rapid and significant rise in serum vitamin B12 when compared with the other treatments. However, at four weeks there was no significant difference between treatment groups for serum vitamin B12. Fourteen weeks after the administration of cobalt pellets, serum and liver.vitamin B12 levels in this group were not significantly different from those of untreated sheep. Vitamin B12 deficiency was induced in 15 small East African goats by feeding cobalt deficient Chloris gayana hay (containing 0.02 mg of Co/kg dry matter) over a 25 week experimental period. Cobalt was supplemented as an oral drench to supply 0.3 g of Co/goat/week to 15 treated goats. At intervals of 3-4 weeks, serum concentrations of Vitamin B12 , total thyroxine (TT4), free tetra-iodothyronine (FT4) and free tri-iodothyronine (FT3) were determined by radioimmunoassay, while the rate of resting metabolism was determined by measuring the goats' rate of oxygen consumption. Serum Vitamin B12 concentration was significantly higher (p<0.01) in cobalt-treated (289.6 +/- 40.76 pg/ml) than in control (142.8 +/- 28.27 pg/ml) goats. The mean serum TT4 concentration was significantly (p<0.01) higher in control (59.0 +/- 1.70 nmol/l) than in cobalt-treated (51.6 +/- 2.45 nmol/l) goats. However, the levels of FT4, FT3 and the rate of resting metabolism were unaffected by the goats' cobalt status. Furthermore, the goats did not lose weight or become anaemic.  相似文献

The development of cobalt deficiency was studied in 30 Scottish Blackface lambs grazing pasture on a soil containing 0-17 ppm cobalt. By the end of an eight-week period 50 per cent of lambs were subjectively appraised as showing signs of cobalt deficiency. After a further period lasting four weeks, during which three groups of lambs were studied (one group, least affected by cobalt deficiency, acted as control, the second received a single cobalt bullet and the third received oral doses of 200 mg cobalt chloride at the beginning of the period and three weeks later). Mean urinary formiminoglutamic acid (FIGLU) concentrations were inversely related to serum vitamin B12 concentrations and increased from 0-08 to 0-20 mumole per ml in group 1, and decreased to virtually zero within one week of treatment in groups 2 and 3. The use of serum vitamin B12 and urinary FIGLU concentrations in the diagnosis of cobalt deficiency in sheep are discussed.  相似文献

Acid-base reaction cements, containing salts of copper, cobalt and selenium, were placed in the reticulum of sheep and cattle, and the rate of release of cobalt and selenium from them was measured. The rate of release of cobalt decreased with time but was sufficient to provide adequate supplementation for at least 6 months. After an initial rate of release of 4 mg Se/day for 1-2 weeks, the mean subsequent rate of release of selenium over a period of 4 months was 0.1 mg Se/day (identical to 5 micrograms Se/day/cm2 surface area). Field trials in lambs and growing steers showed that the cement increased the selenium concentration of their blood throughout a summer grazing period.  相似文献

Herbage selenium (Se) concentration is generally low in Norway. It is unknown whether feeding practices on Norwegian organic farms fulfil the dietary needs of Se and vitamin E for sheep and dairy cattle. Therefore we analysed Se in soil and herbage, and Se and vitamin E in animal blood in the indoor feeding season at 14 organic dairy and 14 organic sheep farms. The herbage Se concentration was low. Approximately 50 and 35% of all samples in the first and second cut, respectively, had Se concentrations below the detection limit of 0.01 mg/kg dry matter (DM). The median (10th, 90th percentile) Se concentrations were <0.01 (<0.01, 0.03) and 0.02 (<0.01, 0.06) mg/kg DM in the first and second cuts, respectively. Whole blood Se concentrations were 0.10 (0.04, 0.15) μg/g in dairy cattle and 0.14 (0.03, 0.26) μg/g in sheep. Vitamin E concentrations were 4.2 (2.7, 8.4) mg/l in dairy cattle and 1.3 (0.9, 2.4) mg/l in sheep. None of the soil or plant variables explained the variation in herbage Se concentration, although Se in soil and plant tended to be correlated. Herbage Se concentration was inadequate to meet the dietary Se requirements. Vitamin E requirement was only met in dairy herds. We recommend Se and vitamin E supplementation to ruminants on organic farms.  相似文献

SUMMARY: Immunity in 12 weaner Merino sheep fed a low selenium (Se) diet (low Se sheep) was compared with that in 10 matching sheep fed the same diet but each given an intraruminal Se pellet (high Se sheep), while the sheep were housed in individual, sheltered pens. All sheep were challenged with killed Brucella abortus cells (days 0 and 28), rabbit red blood cells (days 0, 7 and 28) and corynebaclerium pseudotuberculosis toxoid (days 0 and 28), and serum antibody titres were measured weekly for 8 weeks from day 0. The sheep were then experimentally infected with Haemonchus conforfus, and slaughtered 8 weeks later. The mean antibody titre to B. abortus, measured by 4 different tests, was significantly higher in the high Se sheep on occasions during the primary immune response phase (Rose Bengal test - day 21 (p < 0.05), day 28 (p < 0.025); complement fixation - day 7 (p < 0.05); enzyme-llnked immunosorbent assay - day 14 (p < 0.01); serum agglutination - no differences), but not during the secondary phase. The mean antibody titre to rabbit red blood cells, measured by haemagglutination test, was marginally higher in the high Se sheep on day 49 (p = 0.049). The mean antibody titre to C. pseudotuberculois, measured by enzyme-linked immunosorbent assay, was not significantly different between the groups at any time during the trial. In addition, the mean invitro responsiveness of peripheral blood lymphocytes to stimulation with phytohaemagglutinin in the high Se sheep was significantly greater than that in 10 sheep from the low Se group on day 22 (p < 0.01), but not day 50. However, there were no significant differences in the mean number of sheep in which the infection with H. contortus established, time to first shedding of eggs in faeces, abomasal worm burdens at necropsy, or inflammatory response in the abomasal mucosa in the sheep in each group. The results showed that the low Se sheep produced strong overall immune responses that were largely comparable to those in the high Se sheep.  相似文献

AIM: To evaluate the efficacy of a long-acting injectable microencapsulated formulation of Vitamin B12 in dairy calves. METHOD: Fifty calves, average liveweight 110kg, were randomly allocated to 5 groups of 10 animals and injected subcutaneously in the anterior neck with 0.12, 0.18, 0.24 and 0.3 mg Vitamin B12/kg liveweight using a formulation of microencapsulated Vitamin B12 in a lactide: glycolide copolymer. The untreated calves were injected with the same vehicle, without Vitamin B12. Subsequent changes in serum and liver Vitamin B12 concentrations were followed for 244 days. RESULTS: The microencapsulated Vitamin B12 significantly increased, then maintained serum and liver Vitamin B12 concentrations higher than those of untreated controls for at least 110 days. CONCLUSIONS: Injection of the microencapsulated Vitamin B12 at dose rates of 0.12 to 0.24 mg/kg liveweight will increase and maintain the Vitamin B12 status of calves for at least 110 days.  相似文献

The rapidity and duration of the response of sheep to graded oral doses of cobalt, given as the sulphate or as a cobalt-supplemented anthelmintic Panacur SC, was assessed by the determination of serum vitamin B12 and methylmalonic acid concentrations. The rapidity of the response to the treatments, which ranged from 1 to 250 mg cobalt, was unaffected by dose rate but the effective duration was dose related. There was some evidence of a cumulative effect from repeat treatments of the anthelmintic at three weekly intervals. It is suggested that the optimum inclusion rate for cobalt in anthelmintics lies within the range 21 to 100 mg/treatment.  相似文献

AIM: To evaluate the efficacy of a new long acting injectable selenium ( Se ) formulation to increase the Se status and prevent Se deficiency in red deer. METHODS: Groups of weaned red deer (four stags and six hinds/group) grazing pastures containing <30 mg Se/kg DM were injected subcutaneously with either 0.5, 1.0 or 2.0 mg Se/kg as a new formulation of BaSeO4 (Deposel Multidose), 1.0mgSe/kg of a current formulation (Deposel), or not treated. Blood Se concentrations and liveweight were measured nine times at intervals over 377 and 270 days, respectively. RESULTS: Both formulations of Se elevated blood Se concentrations from 105 nmol/l pre-injection for at least 377 days with peak levels of 1894, 1395 and 818 nmol/l for high, medium and low doses of Deposel Multidose, respectively, at 73141 days, and 1508 nmol/l at 73-141 days for the medium dose of Deposel, which persisted at similar levels for the duration of the study. Deposel Multidose produced fewer and less severe subcutaneous tissue reactions than Deposel. Pastures contained 10 to 30 mg Se/kg DM. There was no significant difference in growth rate between treated and control deer. There was a significant (p<0. 01) linear relationship (y = 1.25x + 71.6, R2=0.86) between blood (x) and liver (y) Se concentrations in the range of 120 - 2100 nmol/l for blood concentrations, and 200 - 3000 nmol/kg for liver concentrations. CONCLUSION: Injections of BaSeO4 in both formulations studied were effective in increasing the Se status of deer but the new formulation produced fewer and less-severe tissue reactions. Young growing red deer appear less sensitive to Se deficiency as measured by weight gain, than sheep and cattle, suggesting that reference ranges for those species are not appropriate for deer. There was a linear correlation between blood and liver selenium concentrations.  相似文献
