Wolbachia (Rickettsiales: Alphaproteobacteria) infections induce abnormalities in the reproductive system and affect various biological traits of the host insects. The density of Wolbachia is one of the major parameters that influence induced phenotypes and interactions with the hosts. Wolbachia occurs naturally in populations of the leafhopper Yamatotettix flavovittatus Matsumura (Hemiptera: Cicadellidae), which transmits phytoplasma that cause white leaf disease in sugarcane. However, the quantity and dynamics of Wolbachia in this leafhopper are not well understood. In the current study, we estimated the number of Wolbachia by absolute quantification of the copy number of wsp, which encodes the outer surface protein, using real-time quantitative polymerase chain reaction (PCR). This investigation was performed using natural populations and laboratory colonies from three lineages of leafhoppers (designated as UD, KK, and SK). There was no significant difference in the number of wsp copies in most of field-collected adults. During the immature developmental stages, there were differences in the dynamics of Wolbachia infection between the UD lineage and the other two lineages. However, the number of wsp copies increased in the early instar and plateaued in the later nymphal instars. Sex had no influence on the number of Wolbachia within the same lineages. The number of Wolbachia was relatively constant during the adult stage in the UD lineage but fluctuated in the other two lineages. In conclusion, the present data provide a framework for exploring the relationship between Wolbachia and the leafhopper and could facilitate future research into management strategies using Wolbachia.  相似文献

The equilibrium moisture content of agricultural products is necessary to optimize drying process and helps to keep the quality of the product during the period of storage. The main aim of this research was to find the best model which could define well, the exchange of moisture between alfalfa (Medicago sativa) and the surrounding air. The desorption isotherms of alfalfa (stem and leaf) were determined separately by using the saturated salt solutions method at three temperatures (25, 50 and 70 °C) within a range of 5-90% relative humidity. Experimental curves of desorption isotherms were fitted to modified equations of Henderson, Halsey, Oswin and Chung-Pfost as well as the GAB model and then evaluated visually by using residual plots and also by some statistical error parameters. The modified Halsey model was found to be the most suitable for describing the relationship between equilibrium moisture content, relative humidity and temperature.  相似文献

The white-spotted globular bug Eysarcoris guttigerus (Thunberg) (Hemiptera: Pentatomidae) is widely distributed in East Asia and the Pacific region. In Japan, the species is found in grassy or composite weeds in the western area of the main islands and Ryukyu Islands of Japan. One notable characteristic of the Eysarcoris genus is the two white spots on the scutellum. This is not the case with the Ishigaki Island population, however, which sports red spots instead of white, suggesting that intraspecific variation exists in the species. Therefore, we investigated intraspecific variation in E. guttigerus using mitochondrial NADH dehydrogenase subunit 2 (ND2), cytochrome oxidase subunit 1 (CO1), cytochrome b (Cytb), tRNA-Serine (tRNAser), NADH dehydrogenase subunit 1 (ND1), and 16S ribosomal RNA (16SrRNA) genes from 13 populations of Japan. The obtained maximum likelihood phylogenetic tree was divided into three groups—Group 1: Mainland, Group 2: Central Ryukyu Islands (Okinawa-Amamioshima Islands), and Group 3: South Ryukyu Islands (Ishigaki Island). The Ishigaki population was significantly separated from the other populations with consistent differences in spot color. The estimated period of divergence between the Ishigaki population and the other populations was consistent with the period of formation of the Kerama Gap in the Ryukyu arc. Thus, the process of formation of the Kerama Gap may have influenced the intraspecific variation of E. guttigerus.  相似文献

In this study, we characterized the genetic structure of Myzus persicae (Sulzer) (Hemiptera: Aphididae) populations in China using microsatellites. We expected that these data will reveal the genetic relationships among various populations of M. persicae and will be of value in the development of better methods for pest control. Four hundred sixty individuals from 23 areas over 13 provinces were collected in the early spring of 2010, all from their primary host, Prunus persicae. The markers analyzed were highly polymorphic, as demonstrated by the expected heterozygosity value (He = 0.861) and the Polymorphism Information Content (PIC = 0.847), which indicated that M. persicae maintains a high level of genetic diversity. Analysis of molecular variance revealed an intermediate level of population differentiation among M. persicae populations (FST = 0.1215). Geographic isolation existed among these populations, and, consequently, the genetic structure of the populations was split into a southern group and a northern group divided by the Yangtse River.  相似文献

The edibility of different Elaeis sp. breeding populations present in Benin was tested for the leaf miner Coelaenomenodera lameensis Berti (Coleoptera: Chrysomelidae), a major oil palm pest in Africa. Experiments carried out in sleeves revealed the oviposition capacities of females and the mortality rates for the different developmental stages by comparing the populations found on two breeding populations of Elaeis oleifera (HBK) Cortes, four of Elaeis guineensis Jacquin and four (E. guineensis × E. oleifera) × E. guineensis backcrosses. Females laid their eggs similarly on all breeding populations, with a preference for the E. guineensis La Mé origin. The average hatching rate reached 80% for the La Mé origin as opposed to 28% for the Deli origin. The mortality rates for the larval instars were greater on E. oleifera, on certain backcrosses and on the Deli origin of E. guineensis. Development at the second- and third- larval instars was the most affected, with a mortality rate of three to five times greater than that seen on La Mé. Epidermis and cuticle measurements indicated which breeding populations were suitable or unsuitable for the development of C. lameensis. E. guineensis, with its thin epidermis (12 µm) and cuticle (2 µm), proved to be highly susceptible to C. lameensis attacks. On the other hand, E. oleifera, which is very resistant, exhibited a thicker epidermis (17 µm) and cuticle (4 µm). The breeding populations were thus classified according to the positive or negative influence they exerted on the insect’s egg laying and feeding.RÉSUMÉ.

The most destructive enemy of the lychee, Litchi chinensis Sonn. (Sapindales: Sapindaceae), in India is a stink bug, Tessaratoma papillosa (Drury) (Hemiptera: Tessaratomidae). The population of T. papillosa on lychee trees varied from 1.43 ± 0.501 to 9.85 ± 3.924 insects per branch in this study. An increase in the temperature and a decrease in the relative humidity during summer months (April to July) favor the population buildup of T. papillosa. A forecasting model to predict T. papillosa incidences in lychee orchards was developed using the autoregressive integrated moving average (ARIMA) model of time-series analysis. The best-fit model for the T. papillosa incidence was ARIMA (1,1), where the P-value was significant at 0.01. The highest T. papillosa incidences were predicted for April in 2010, January in 2011, May in 2012, and February in 2013. A model based on time series offers longer-term forecasting. The forecasting model, ARIMA (1,1), developed in this study will predict T. papillosa incidences in advance, thus providing functional guidelines for effective planning of timely prevention and control measures.  相似文献

A field survey was conducted in Ataulfo mango (Mangifera indica L.) orchards in Chiapas, Mexico, with the objective of determining the natural enemies of the Frankliniella complex species (Thysanoptera: Thripidae). Seven species of this genus feed and reproduce in large numbers during the mango flowering. Two representative orchards were selected: the orchard “Tres A” characterized by an intensive use of agrochemicals directed against thrips, and the orchard “La Escondida” that did not spray insecticides. During mango flowering, five inflorescences were randomly collected every 5 d in both orchards, for a total of 18 sampling dates. Results revealed the presence of 18 species of arthropods that were found predating on Frankliniella. There were 11 species in the families Aeolothripidae, Phlaeothripidae, Formicidae, Anthocoridae and Chrysopidae; and seven species of spiders in the families Araneidae, Tetragnathidae, and Uloboridae. Over 88% of predators were anthocorids, including, Paratriphleps sp. (Champion), Orius insidiosus (Say), Orius tristicolor (White), and O. perpunctatus (Reuter). The orchard that did not spray insecticides had a significantly higher number of predators suggesting a negative effect of the insecticides on the abundance of these organisms.  相似文献

Colorado potato beetle (CPB, Leptinotarsa decemlineata Say) (Coleoptera: Chrysomelidae) is one of the most serious insect pest feeding on wild and cultivated Solanaceae plants. This pest poses a significant threat to potato crops. CPB originated from North America but has become widespread and has adapted in new localizations. Currently, it is reported in many countries worldwide. Endosymbiotic bacteria might have an influence on insect adaptation to new conditions. They are known to play a role in invasiveness of insect hosts and to facilitate colonization of new niches; however, information on endosymbionts of the CPB is very limited. In this study, we screened CPB populations collected from 20 evenly distributed locations in Poland for the presence of Arsenophonus, Cardinium, Wolbachia, and Flavobacterium. We found the presence of Flavobacterium in the studied insects. Little is known about CPB–endosymbionts interactions, thus this study may provide a reference for future studies in this subject.  相似文献

The glassy-winged sharpshooter, Homalodisca vitripennis (Germar), is a serious pest of grapes and other crop and ornamental plants mainly through its role as a vector of the bacterium Xylella fastidiosa Wells. Citrus harbors large populations of this insect throughout much of the year in areas where the pest is problematic and improved understanding of the population dynamics and management of H. vitripennis on citrus may be key to its management in the broader agricultural landscape. In turn, the study of population dynamics and the development of management strategies require effective and efficient sampling methods. Within-tree sampling distribution studies revealed that adults and nymphs were more abundant and less variable in the upper strata of citrus trees (>1.5 m). They occurred in greater numbers on the southern quadrants of trees but relative variability did not differ due to cardinal direction. We developed and validated several fixed-precision sequential sampling plans for estimating the density of nymphs and adults of H. vitripennis using a pole bucket sampling method. Based on validation from resampling of independent data sets, Green’s sequential sampling model, based on the Taylor’s power law, provided the best overall performance in terms of providing mean density estimates with levels of precision equal to or better than the desired precision over a range of possible insect densities. Average sampling costs varied from about 21 to 189 min for a desired precision of 0.25 depending on insect density and whether the goal is to sample nymphs, adults or both stages combined. Further, the sampling plans developed on orange trees were robust, being equally effective on orange and lemon trees and on trees treated or not with insecticides.  相似文献

Intestinal bacterial community plays a crucial role in the nutrition, development, survival, and reproduction of insects. When compared with other insects with piercing-sucking mouthparts, the habitats of cicada nymphs and adults are totally different. However, little is known about the differences in the gut bacterial communities in the nymphs and adults within any cicada species. The diversity of bacteria in the gut of nymphs and adults of both genders of Meimuna mongolica (Distant) was studied using the denaturing gradient gel electrophoresis (DGGE) method. Few inter-individual variations among gut microbiota were observed, suggesting that M. mongolica typically harbors a limited and consistent suite of bacterial species. Bacteria in the genera Pseudomonas and Enterobacter were the predominant components of the gut microflora of M. mongolica at all life stages. Bacteria of Pantoea, Streptococcus, and Uruburuella were also widespread in the cicada samples but at relatively lower concentrations. The relative stability and similarity of the PCR-DGGE patterns indicate that all individuals of this cicada species harbor a characteristic bacterial community which is independent from developmental stages and genders. Related endosymbionts that could be harbored in bacteromes of cicadas were not detected in any gut samples, which could be related to the cicada species and the distribution of these endosymbionts in the cicada cavity, or due to some of the possible limitations of PCR-DGGE community profiling. It is worthwhile to further address if related cicada endosymbiont clades distribute in the alimentary canals and other internal organs through diagnostic PCR using group-specific primer sets.  相似文献

Sogatella furcifera (Horvath) is an important rice pest with the wing dimorphism, including macropterous and brachypterous morphs. The protein expression profiles in two wing-type adults and two wing-type disc fifth-instar nymphs were analyzed using two-dimensional gel protein electrophoresis and mass spectrometry. In adults and fifth-instar nymphs, 127 and 162 protein spots were detected, respectively. Fifty-five differentially expressed protein spots were identified between the long-winged adults and the short-winged adults, and 62 differentially expressed protein spots were found between the long-winged disc fifth-instar nymphs and short-winged disc fifth-instar nymphs. In long-winged and short-winged adults, six and seven specific protein spots were identified, respectively, with five and seven protein spots having more than threefold increased level, respectively. In long-winged and short-winged disc morph nymphs, 8 and 12 specific protein spots were identified, respectively, with 11 and 17 spots containing more than threefold increased level, respectively. Among the 16 identified proteins, five proteins are associated with muscle function, suggesting that muscle is a main tissue where the genes were differentially expressed between the two wing types. In addition, the content of a peptidase with an insulinase domain was higher (by 3.02 ± 0.59 fold) in the short-winged fifth-instar nymphs than in the long-winged fifth-instar nymphs, which suggests that this peptidase may be involved in wing differentiation by regulating insulin receptors. The results of this study provide some genetic clues for the wing differential development in S. furcifera and provide more references for future studies.  相似文献

Seasonal timing is assumed to involve the circadian clock, an endogenous mechanism to track time and measure day length. Some debate persists, however, and aphids were among the first organisms for which circadian clock involvement was questioned. Inferences about links to phenology are problematic, as the clock itself is little investigated in aphids. For instance, it is unknown whether aphids possess diurnal rhythms at all. Possibly, the close interaction with host plants prevents independent measurements of rhythmicity. We reared the pea aphid Acyrthosiphon pisum (Harris) on an artificial diet, and recorded survival, moulting, and honeydew excretion. Despite their plant-dependent life style, aphids were independently rhythmic under light–dark conditions. This first demonstration of diurnal aphid rhythms shows that aphids do not simply track the host plant’s rhythmicity.  相似文献

The white-backed planthopper, Sogatella furcifera (Horváth) (Hemiptera: Delphacidae), is a major pest of rice and has long-range migratory behavior in Asia. Microsatellite markers (simple sequence repeats) have been widely used to determine the origins and genetic diversity of insect pests. We identified novel microsatellite loci for S. furcifera samples collected from Laos, Vietnam, and three localities in Bangladesh from next-generation Roche 454 pyrosequencing data. Size polymorphism at 12 microsatellite loci was verified for 40 adult individuals collected from Shinan, South Korea. The average number of alleles per locus was 7.92. The mean values of observed (Ho) and expected heterozygosities (HE) were 0.615 and 0.757, respectively. These new microsatellite markers will be a resource for future ecological genetic studies of S. furcifera samples across more broad geographic regions in Asia and may assist in estimations of genetic differentiation and gene flow among populations for implementation of more effective management strategies to control this serious rice pest.  相似文献

The inclusion of the cry gene in corn may produce direct effects on non-target pests. Our research was focused on the relationship between Bt corn germplasm, expressing the cry1F protein to control the fall armyworm [Spodoptera frugiperda (Noctuidae)], and a non-target pest, the corn leafhopper [Dalbulus maidis (Cicadellidae)]. The aim of this contribution was to elucidate if Bt corn plants have influence on the oviposition preference of the leafhopper and to evaluate the effect of the transgenic plant on the hatching rate of egg. Female corn leafhoppers were released in cages each containing two potted plants in the V2 stage: a Bt germplasm and the corresponding isogenic hybrid. Laid eggs were counted and the number of hatched nymphs recorded. D. maidis females oviposited and laid more eggs in Bt plants. The egg hatching rate was negatively affected by the Bt germplasm. In addition, a field study was conducted in order to determine the abundance of D. maidis adults in Bt corn and the corresponding non-Bt isoline. Two corn plots sown with the same germplasms as used in the laboratory bioassays were sampled weekly. In the field, the population of the corn leafhopper was higher in the Bt corn plot than in the non-Bt isoline. Possible hypotheses for the differences in abundance of the vector in the field are: a) that pleiotropic effects of Bt corn could attract adults; b) the existence of a possible direct competition between the corn leafhopper and the target pest in order to utilize the whorls of corn plants as refuge and feeding sites, so the high populations of the vector could be due to the large supply of healthy whorls in the transgenic plot; and/or c) a differential attack of natural enemies occurring in non-Bt plots.  相似文献

In northern China, Adelphocoris suturalis, Adelphocoris lineolatus and Adelphocoris fasciaticollis (Hemiptera: Miridae) are common pests of cotton and several other crops. These species have vastly diverse geographic distribution, seasonal dynamics and abundance, the underlying causal factors of which are poorly understood. In this study, the importance of a broad range of plant species as overwintering hosts for each Adelphocoris sp. was compared. Nymphal emergence from a total of 126 plant species was monitored at two distinct locations. The eggs of A. suturalis successfully eclosed from un-plowed cotton field soil and 115 plant species, primarily pastures, weeds and agricultural crops. The eggs of A. lineolatus successfully eclosed from 40 plant species, mainly pastures and weeds. Finally, A. fasciaticollis overwintered on 35 plant species, primarily tree species, weeds and agricultural crops. In conclusion, the most common and widely distributed mirid species, A. suturalis, overwintered on a comparatively broader range of plants compared to the other two species. These observations help to understand the differences in geographical distribution and abundance of the three Adelphocoris species, and constitute the basis for forecasting and pest management protocols for Adelphocoris spp. in China.  相似文献

The aim of this research was to evaluate acute toxicity of the essential oil of leaves of Chinese chives, Allium tuberosum Rottler ex Spreng (Asparagales: Alliaceae) and its major constituents against Apolygus lucorum Meyer-Dür (Hemiptera: Miridae). The essential oil of A. tuberosum leaves was obtained by hydrodistillation and analyzed by gas chromatography and gas chromatography-mass spectrometry. The major constituents of the oil were sulfur-containing compounds, including allyl methyl trisulfide (36.24%), diallyl disulfide (27.26%), diallyl trisulfide (18.68%), and dimethyl trisulfide (9.23%). The essential oil of A. tuberosum leaves exhibited acute toxicity against Ap. lucorum with an LD50 value of 20.03 μg per adult. Among the main compounds, diallyl trisulfide (LD50 = 10.13 μg per adult) showed stronger acute toxicity than allyl methyl trisulfide (LD50 = 21.10 μg per adult) and dimethyl trisulfide (LD50 = 21.65 μg per adult). The LD50 value of diallyl disulfide against Ap. lucorum was 28.10 μg per adult. The results indicated that the essential oil of A. tuberosum and its major constituents may have a potential to be developed as botanical insecticides against Ap. lucorum.  相似文献

Stink bugs, primarily southern green stink bug, Nezara viridula (Hemiptera: Pentatomidae), are a major pest complex of soybeans (Glycine max) throughout the southern United States. Densities sometimes peak during late R6 and R7 soybean growth stages when soybeans are approaching physiology maturity and the rate of injury from stink bugs is reduced. Field cage trials were conducted from 2005 to 2008 to examine the type and extent of soybean damage caused by southern green stink bugs during the R7 growth stage. The yield response was variable, but overall was not significant. The impact of southern green stink bugs on quality was more consistent. Test weight decreased, and heat damage and total damage increased as stink bug density increased. Based on these data, three economic injury models were developed using different assumptions. The model that assumes no yield loss, does not predict economic injury within the range of stink bug densities tested. However, if the statistically non-significant yield losses are accepted as real, then the models suggest that the southern green stink bug economic injury level and action threshold for soybeans during R7 stage is generally between nine and 15 stink bugs per row m.  相似文献
