脂肪严重沉积是当前养殖鱼类普遍发生的代谢障碍病症,其原因复杂多样。近年来,已有较多研究提示,广泛存在于自然水环境中的环境内分泌干扰物(EDCs)除了对多种鱼类的生长、发育和繁殖产生持久的慢性毒理效应之外,也对鱼类脂质代谢过程造成影响,导致脂肪沉积异常,并影响养殖鱼类的品质与食用安全性。本文首次系统回顾了近年来在EDCs影响鱼类脂质代谢研究领域中的重要研究成果,并针对持久性有机污染物(POPs)、重金属、环境雌激素和人工合成药物4大类污染物,系统总结了不同EDCs对鱼类脂质代谢的干扰作用。辨析了不同研究中EDCs对鱼类暴露方式、暴露时间和暴露浓度的差异,并由此分析同种EDC在不同的暴露环境下对鱼类脂质代谢产生的差异影响。本文在综述已有成果的基础上,提出了本领域今后的研究重点和方向,旨在为深入研究EDCs对鱼类脂质代谢的影响机制提供更多参考,进一步理解鱼类脂肪沉积的原因,为绿色可持续发展的水产养殖业提供重要的理论支持。  相似文献

本文旨在克隆鲤(Cyprinus carpio)激素敏感性脂肪酶a (hormone-sensitive lipase a, lipea)基因,探究其时空表达特征及其与脂肪沉积的关系,以期为鲤脂肪沉积分子调控机制研究奠定理论基础。本实验采用RACE技术克隆获得鲤lipea基因,其cDNA全长为3379 bp,包括230 bp的5′非翻译区(5′-UTR)、1067 bp的3′非翻译区(3′-UTR)和编码693个氨基酸的开放阅读框。多序列比对显示,鲤lipea与斑马鱼(Danio rerio)、牙鲆(Paralichthys olivaceus)和虹鳟(Oncorhynchusmykiss)的氨基酸序列相似性分别为88.17%、64.94%和63.35%,不同物种间序列差异主要集中在C-端的多肽序列上。蛋白质结构分析显示该蛋白含有3个功能域分别具有脂肪酶活性、乙酰基水解酶活性和水解酶活性。应用实时荧光定量PCR技术分析了lipea基因在鲤各组织、不同发育时期以及脂肪含量极端差异个体的肌肉和脂肪组织中mRNA表达量的变化。结果显示, lipea在各组织中均有表达,腹腔脂肪中相对表达量最高,其次是腹部肌肉,血液中最少。随着胚胎的发育, lipea的表达水平呈现下降趋势, 8细胞期表达量最高,其次是囊胚晚期,开口期最少。lipea表达量与背部肌肉脂肪含量呈正相关,但差异不显著(P0.05),与腹腔脂肪量呈负相关,且差异极显著性(P0.01),推测lipea基因对鲤腹腔脂肪沉积可能具有抑制作用。本实验结果可为进一步解析鲤lipea基因的功能和基因调控脂肪沉积分子机制研究提供参考。  相似文献

以鱼油为脂肪源,分别配制脂肪水平为3.7%、6.8%、9.6%、13.1%、15.6%的5种实验饲料,对平均初始体质量为(7.13±0.03)g的许氏平鲉进行为期60 d的饲养实验,探讨饲料脂肪水平对许氏平鲉形体指数、脂肪沉积、血液生化指标与脂肪代谢酶活性的影响。结果表明:肝体指数、脏体指数随着饲料脂肪水平的升高显著提高(P0.05),脂肪水平对肥满度没有显著影响(P0.05);许氏平鲉鱼体脂肪沉积随饲料脂肪水平的增加显著提高(P0.05),15.6%全鱼、肌肉、肝脏脂肪含量最高,显著高于3.7%、6.8%组(P0.05)。随着脂肪添加量的增加,许氏平鲉肝脏葡萄糖-6-磷酸脱氢酶(G6PDH)和苹果酸脱氢酶(MHD)活性显著降低(P0.05),15.6%与13.1%组G6PDH、MHD活性显著低于3.7%和6.8%组(P0.05);肝酯酶(HL)和脂蛋白酯酶(LPL)活性显著升高(P0.05),15.6%与13.1%组HL和LPL活性显著高于3.7%和6.8%组(P0.05)。血液甘油三酯(TG)和总胆固醇(CHO)含量随着饲料脂肪水平增加有下降的趋势,15.6%组的TG和CHO含量显著低于3.7%和6.8%组(P0.05);血液中高密度脂蛋白胆固醇(HDLC)、谷草转氨酶(AST)、谷丙转氨酶(ALT)与碱性磷酸酶(ALP)活性呈上升趋势(P0.05),15.6%组最高;低密度脂蛋白胆固醇(LDLC)呈先上升后下降的趋势(P0.05)。研究表明,饲料脂肪水平对许氏平鲉形体指数、脂肪沉积、血液生化指标及脂肪代谢酶活性影响显著,但过高的脂肪水平将使许氏平鲉肝脏脂肪沉积增多,当饲料脂肪水平高于9.6%时不利于鱼类健康生长。  相似文献

采用30%葡萄糖或玉米淀粉作为糖源的配制两种纯化饲料,饲养初始体重为(35.94±1.86)g的两组草鱼,经过为期9周的生长试验,观察葡萄糖和玉米淀粉对草鱼生长和肠系膜脂肪沉积的影响.实验结果显示摄食葡萄糖饲料的草鱼其相对生长率、饲料效率和蛋白质效率均显著高于玉米淀粉组,而肠系膜脂肪占鱼体的百分比在葡萄糖饲料组和淀粉饲料组之间存在显著性差异,分别为1.85%±0.46%和3.56%±0.45%.由此可见,葡萄糖比玉米淀粉对草鱼的生长具有更好的作用,而玉米淀粉比葡萄糖更容易引起草鱼肠系膜脂肪沉积的增加.  相似文献

甘草次酸对团头鲂生长、脂肪沉积与抗氧化功能的影响   总被引:3,自引:2,他引:1  
为探讨甘草次酸对团头鲂生长、脂肪沉积和抗氧化功能的影响,选取均体质量为(15.63±0.04)g的团头鲂幼鱼420尾,随机分在15个网箱中,分别以甘草次酸水平为0、0.15、0.30、0.45和0.60 g/kg的5种饲料投喂8周。结果发现,饲料添加甘草次酸对团头鲂增重率、特定生长率、饵料系数没有显著影响(P0.05)。甘草次酸可以显著降低实验鱼脏体比、肝体比、腹脂率及肝脏脂肪含量(P0.05),但对全鱼体组成和肌肉脂肪含量无显著影响(P0.05)。比对血浆脂肪代谢酶可见,0.30~0.60 g/kg甘草次酸添加组血浆总胆固醇含量较对照组显著下降(P0.05);而甘油三酯、游离脂肪酸和高密度脂蛋白胆固醇含量无显著变化(P0.05)。肝脏中脂蛋白酯酶、肝酯酶和总酯酶活性在添加甘草次酸后显著降低(P0.05);0.30~0.60 g/kg甘草次酸添加组脂肪酶活性显著高于其他各组(P0.05)。饲料添加甘草次酸可以显著提高肝脏超氧化物歧化酶活性和还原型谷胱甘肽含量,降低丙二醛含量(P0.05)。研究表明,饲料中添加0.30~0.45 g/kg甘草次酸时,显著降低了团头鲂内脏团的脂肪沉积,改善了鱼体脂肪分布,这可能是由于甘草次酸加强脂解作用,提高脂肪代谢酶活性导致的;饲料中添加甘草次酸也可显著提高团头鲂的抗氧化能力。  相似文献

A feeding experiment was conducted for 10 weeks with Eurasian perchPerca fluviatilis juveniles (33.1–35.9ginitial body weight) using three dietary fat levels (11.7, 15 and 19.3%). Theresults showed that increasing dietary fat content increased growth, feedefficiency and protein utilization of perch (P < 0.05) but did not affectmortality. Lipid analysis showed that the composition of some perch tissueswerealso significantly affected by increasing dietary fat content, with an increaseof lipid deposition in the viscera (from 64.9% in fish fed 11.7 % fat to 78.8%in fish fed 19.3% fat) and liver (from 12.4 to 23.8%) (P < 0.05), while themuscle composition was not affected by the dietary treament. Abstract. During a 3-ycar study of the ruffe population in the highly polluted Elbe estuary, 551 fish were examined for grossly visible liver lesions. Three main types of abnormalities were distinguished, and the histological characteristics of representative livers are described. The rate of occurrence of each abnormality was recorded and correlated with the length of the affected fish. The livers in 11%; of the ruffe were uniformly discoloured due to excessive deposition of fat in the hepatocytes. In 39% of the livers, localized pale flecks, patches, or marbled patterns were observed. A subsample examined histochemically revealed that these were also due to fat deposition, except in a few cases resulting from excessive glycogen accumulation. Distinct from these two syndromes were pale nodules in the hepatic tissue seen in 8% of the ruffe. Microscopy showed the nodules to be neoplastic. A relationship between fish length and prevalence was found only in the case of these nodules. Possible causes of the various liver abnormalities found are discussed.  相似文献

Salmonid aquaculture has focused for many years on enhancing the quantity of fish produced. However, optimization of the quality of salmonids may lead to improved consumer acceptance and higher prices for the farmed product. The present review evaluates how the quality of salmonids is affected by parameters such as feed type, level of dietary intake (ration) and growth. Feed composition has a major influence on the proximate composition of salmonids. In particular, whole body lipid as well as the lipid content in the edible fillet is directly related to dietary fat content, while the fatty acid composition of the fish flesh is also strongly influenced by the dietary fatty acid profile. While fish body composition appears to be largely influenced by feed composition, an increase in other parameters such as feed ration and fish size also results in enhanced adipose deposition and decreased water content in the fish body. The protein content, however, remains more or less stable. An increase in fish body lipid content is not necessarily a negative factor, depending upon the processing procedure that follows. However, an increase in body fat content is generally accompanied by a reduction in slaughter yield, owing to an increase in the weight of viscera in relation to body weight. Although salmonid flesh quality with respect to levels of proximate constituents and yield appears to be under the strong influence of feed composition and feed amount, the sensory characteristics, such as odour, flavour, texture, etc., are only governed by these factors to a small degree. Tactics for the rearing of salmonids for specific purposes should therefore take into consideration the fact that the level of proximate constituents in the whole body as well as the fillet are readily manipulated by feed composition and feeding strategies, whereas the sensory parameters are less affected by these variables.  相似文献

Macroscopically visible lipid deposition varying in size from pinpoint to 8‐mm diameter was found in spleens of a population of intensively farmed perch, Perca fluviatilis L. over a 24‐month rearing period. Large agglomerates of adipocytes distinguishable from surrounding normal tissue occurred in all individuals with spleen lipidosis. Several affected fish presented total dystrophy of large clusters of hepatocytes. Prevalence of lipidosis was 5.0% at 12 months and 16.6% at 24 months. There was no significant difference between fatty acid profiles of liver or perivisceral fat of perch with and without lipidosis except for linoleic, myristic, γ‐linoleic, cis‐eicosatrienic, palmitooleic acid. Body weight and hepatosomatic, perivisceral fat and splenosomatic indices were not associated with lipidosis. There was no significant effect of lipidosis on mortality or growth.  相似文献

A study was undertaken to determine the effect of various dietary carbohydrate‐to‐lipid ratios on growth performance, whole‐body composition and tissue lipid content in Senegalese sole (Solea senegalensis) juveniles. Data on the dietary regulation of key hepatic enzymes of the lipogenic and glycolytic pathways (glucose‐6‐phosphate dehydrogenase, G6PD; malic enzyme, ME; fatty acid synthetase, FAS; pyruvate kinase, PK and glucokinase, GK) were also generated. Four isonitrogenous (crude protein: 52% dry matter (DM)) diets were formulated to contain one of two lipid levels (11% and 21% DM). Within each dietary lipid level, the nature of the carbohydrate fraction (raw or extruded peas) was varied. Triplicate groups of 54 sole (initial body weight: 23.6±1.2 g) were grown in recirculated seawater over 67 days. Fish were fed using automated feeders. At the end of the study, whole‐body, liver, viscera and muscle samples were withdrawn for analyses. During the experimental period, the mean fish weight about doubled in all treatments. No significant differences were found in growth performance (ranging from 1.1% to 1.4% body weight day?1) among dietary treatments. High‐fat diets increased whole‐body fat content. Similarly, daily fat gain ranged from 0.54 to 0.78 g kg?1 day?1 and highest values were found in fish fed high‐lipid diets. Dietary treatments also affected tissue lipid content (liver, viscera and muscle), with highest values in fish fed high‐fat diets. The nature of dietary carbohydrates had little influence on performance criteria, but affected tissue lipid deposition. The activities of G6PD, ME and FAS were depressed by elevated levels of dietary lipid, confirming the inhibitory effect of dietary fats on lipid biosynthesis. At both dietary lipid levels, ME and FAS activities were little affected by dietary carbohydrate. Activities of PK and GK were not affected by the starch level of the diets. In Senegalese sole juveniles, the lipogenic pathway is more susceptible to modulation by dietary means (particularly through lipid intake) than the glycolytic pathway.  相似文献

Pansteatitis has been identified in wild populations of sharptooth catfish, Clarias gariepinus (Burchell), and Nile crocodiles, Crocodylus niloticus Laurenti, inhabiting the same waters in the Olifants River Gorge in the Kruger National Park, South Africa. Mesenteric and pectoral fat tissue was investigated microscopically and by fatty acid analysis in healthy and pansteatitis‐affected catfish from both captive and wild populations. Variation in fatty acid composition between pectoral and mesenteric fat was noted. Composition of mesenteric fat differed between fish from various localities as a result of differences in diet. Pansteatitis in the captive population, resulting from ingestion of high amounts of dietary oxidized fat, reflected higher levels of unsaturated fatty acids within the mesenteric fat. Mesenteric fat of pansteatitis‐affected wild catfish was characterized by an increase in moisture content, a decrease in fat content and a decrease in stearic and linoleic acids. The n‐3 to n‐6 fatty acid ratio of mesenteric fat was higher in pansteatitis‐affected wild catfish than in healthy catfish from the same locality, reflecting higher polyunsaturated fat intake by pansteatitis‐affected fish. The possible role of alien, invasive, phytoplankton‐feeding silver carp, Hypophthalmichthys molitrix (Valenciennes), in the aetiology of pansteatitis in both catfish and crocodiles in the Olifants Gorge is discussed.  相似文献

Carcass and flesh morphometric, reological and chemical traits of triploid (3n) and diploid (2n) shi drum (Umbrina cirrosa L.) were evaluated through 7 months. Three age groups, 17‐, 21‐ and 24‐month‐old fish, were investigated. The effects of ploidy were statistically evaluated and the weight of fish was included in the model as a covariate because triploids grew less than diploids. As expected, fish weight was found to be significantly correlated with all the investigated morphometric traits, but showed a negative correlation with some chemical (pH) and colour traits (lightness) of raw fillet. In comparison with controls, triploid shi drums were characterized by different morphological traits that involved a slender body shape. In triploids, a reduction in condition factor, backbone weight, dressing index and an increase in the agility index were also recorded. When the commercial size (i.e. over 300 g) was achieved, triploids exhibited larger coelomatic and fillet (dorsal white muscle) fat deposition than diploids. Among reological traits, colour and texture were affected by ploidy; raw fillet lightness and cooked flesh tenderness were higher among triploids in all the investigated age groups. As fish were fed with a restricted feeding regimen, fillet fat deposition was supposed to be limited. Thus, the effects of ploidy on reological traits may be only partially explained by lipid fillet amount and are supposed to be more related to different fibre muscle architecture.  相似文献

An experiment was conducted to evaluate the effects of dietary composition on apparent digestibility coefficients of protein, fat and carbohydrate of gilthead sea bream. In addition α‐amylase and proteolytic activities were measured in the digestive tract of fish, held at 20±1°C. Six experimental diets were formulated containing approximately 40%, 45% and 50% protein, 11% and 21% fat and a starch level which fluctuated from 14% to 36%. Fish having an average weight of 100–130 g were used, fed at 1.5% of their body weight daily, and digestibility was measured using an indirect method. Enzyme activities were measured in the digestive tract of fish, fed diets containing a combination of 40%, 50% protein with 11%, 21% fat at 0.5, 5, 10, 24 h after a single meal. Starch digestibility was reduced with its level in the diet. It also negatively affected fat digestibility as well as protein digestibility, the last only at the high fat level. Dietary fat level had a negative effect on starch and protein digestibility. Fat affected also strongly α‐amylase levels in the digestive tract, while its effect on protease activity was of smaller magnitude. These results indicate that digestive enzyme activities and nutrient digestibility values are affected by dietary composition, carbohydrates and fat indicating the strongest effect.  相似文献

Two extruded diets designed to have fat levels of 220 g kg−1 (F22) and 300 g kg−1 (F30) were fed to Atlantic salmon Salmo salar L. in two different experiments during the sea rearing period (from 0.2–0.3 kg to 3–4 kg). Each diet was fed restricted and isoenergetically at two feeding rates to fish in triplicate groups. In one of the experiments, a supplementary group of fish was fed to satiation with the F30 diet. All fish were slaughtered and evaluated for quality according to a commercial standard.
No difference in growth was observed between fish fed the two diets at similar feeding rates and the growth was proportional to the amount of dietary energy offered. Feed conversion ratios decreased according to higher energy content in the F30 diet, and the nitrogen and phosphorus retention increased significantly. Fish fed the F30 diet revealed a higher incidence of sexual maturity. Fat content in cutlets and dressed carcasses were significantly affected by feeding rate but not by dietary fat level. Fish fed the F30 diet had more visceral fat and, consequently, lower dress-out percentage. Mortality, liver size and liver colour were not significantly affected by dietary fat level.
These experiments showed that even large differences in dietary fat level employed for the entire sea rearing period of Atlantic salmon, did not, or only marginally affected the cutlet and dressed carcass fat content. The high fat diet improved the feed utilization, thus decreasing the discharge to the environments. Furthermore, it resulted in greater growth at ad libitum feeding.  相似文献
