Summary Black rot disease caused by Xanthomonas campestris pv. campestris is a limiting factor in the commercial production of the cauliflower crop. Crosses were attempted between SN 445, a mid season cultivar resistant to black rot and two highly susceptible commercial cultivars (Pusa Snowball-1 and K-1). Cocoa pod rot, caused by different Phytophthora species, is rife in all production zones. Phytophthora spp. control is therefore a major challenge for world cocoa cultivation, and selection of resistant material is a priority research theme for many producing countries. Various genetic trials have been set up over many years in zones affected by the disease. In Cameroon, a 6x6 diallel mating design was set up to study heritability of the resistance trait. Field observations led to the classification of parents according to their ability to transmit resistance to their progenies. These field measurements were long and laborious, so a resistance test based on artificial leaf inoculations was developed. This article studies the correspondence between results obtained with the test and observations of rot rates in the field. There was a significant genetic correlation between pod rot rate in the field and leaf test data, so using this resistance test could shorten cocoa tree selection cycles. Crosses were made between Waxy Globe, a small fruited hot type, resistant to fruit rot and four highly susceptible bell pepper commercial cultivars (Chinese Giant, California Wonder, Osh, Kosh and Yolo Wonder). Studies of the F1's, F2's and backcrosses indicated that Waxy Globe carries a dominant gene imparting resistance to fruit rot. After three backcrosses with California Wonder, lines were obtained combining the resistance gene of Waxy Globe with the large fruit size and low capsaicin of California Wonder.  相似文献

For the first time in the history of black pepper cultivation, a partly fertile interspecific hybrid having partial resistance to the dreaded disease Phytophthora foot rot was developed through hybridizing Piper nigrum with the wild species Piper colubrinnum. Hybridity of interspecific progenies was established through morphology, anatomy, cytology, and molecular studies. The hybrid, whose chromosome number is 2n = 39, is a triploid hybrid between a tetraploid and diploid species. The hybrid designated as Culture P5PC-1 exhibited distinct anatomical and morphological feature with a large number of long spikes with reduced setting percentage. The RAPD primers OPE 07 and OPG 08 were identified as hybrid specific molecular markers. Functional evaluation revealed partial introgression of genes—responsible for Phytophthora foot rot resistance—into the hybrid. This hybrid is considered as a successful breakthrough for introgression of resistance to the cultivated species Piper nigrum from the wild species Piper colubrinnum.  相似文献

Summary The inheritance of resistance in cauliflower to stalk rot (Sclerotinia sclerotiorum (Lib.) de Bary) was investigated in population from six generations of six crosses. Disease incidence was recorded on 4 parents, 6 Fs 1, 6 Fs 2 and 12 back-crosses in a screenhouse under artificially created epiphytotic conditions. Resistance to stalk rot in this set of parents was found to be polygenic and under the control of recessive genes and due primarily to additive gene action. A breeding strategy emphasizing recurrent selection should lead to improvement in resistance.  相似文献

Zingiber zerumbet (L) Smith, a wild clonal species related to the cultivated ginger (Zingiber officinale Roscoe), is a potential resistance donor for soft rot disease in ginger caused by Pythium aphanidermatum (Edson) Fitzp. In this study we evaluated the genetic diversity and P. aphanidermatum resistance of 74 Z. zerumbet accessions belonging to 15 populations from eight districts in Kerala state, India. The disease index (DI) of the accessions varied from 0% to 72.24% and the accessions could be separated into six frequency classes according to their DI values. More than 65% of the accessions had a DI < 20%. Eight accessions were found to be immune to the infection. The relative frequency of resistant accessions was higher in the central and northern regions of Kerala. Amplified fragment length polymorphism (AFLP) analysis of Z. zerumbet accessions using five primer combinations yielded 215 bands in total, of which 175 (81.4%) were polymorphic. Nei’s genetic diversity (h) of 0.2738 and Shannon information index (I) of 0.4012 revealed a high genetic diversity in Z. zerumbet unexpected for a clonal species. In the UPGMA dendrogram, accessions were clustered mostly according to their geographical origin and no clear correspondence was observed between the clustering pattern of accessions and their responses to Pythium aphanidermatum. The study revealed high genetic diversity and variability for pathogen resistance among Z. zerumbet accessions and confirmed the value of Z. zerumbet as a potential donor for soft rot resistance for the genetic improvement of ginger.  相似文献

Sources of partial resistance to crown rot caused by Fusarium pseudograminearum are detected in mature plants grown in artificially inoculated soil in the field. The resistance in most but not all of these sources can also be detected in seedlings. In order to determine whether partial resistance is related to depth of crown formation, this character was measured in 13 cultivars/lines with a range of reaction to crown rot. It was also measured in doubled haploid plants from the cross, Batavia/2–49. Crown depths varied from 17.1 mm to -2.3 mm (above ground) in pots in a waterbath at 25 °C and from 43.5 mm to 20.7 mm when plants were grown in the field. The correlation coefficient between relative susceptibility to crown rot (Field test) and crown depth of 13 cultivars/lines was 0.57 (p ≤ 0.05). With the exception of the cultivars, Sunco and Pelsart, partial resistance to crown rot was inversely related to depth of crown formation. This indicates that depth of crown formation may be partly responsible for the reaction of a cultivar/line to crown rot. Depth of crown formation was also measured in susceptible and partially resistant cultivars/lines grown from seed planted at different depths. As depth of seeding increased, depth of crown formation in partially resistant and susceptible cultivars/lines increased in similar proportions. Summary Disease progress and gradient curves of black rot on cabbage were evaluated in field plots of the cultivars Bartolo, Erdeno, Perfect Ball, and Roxy in The Netherlands during 1991 and 1992. Plots were inoculated by single sources in the centre of each plot. Individual plants were examined for disease incidence and severity. Disease progress was described by the Gompertz model. The overall measure of absolute rate (disease progress rate r multiplied with maximum disease intensity K) was used to compare cultivar effects on disease progress. Disease gradients were described by the negative exponential model. The percentile distance (distance from the source at which disease intensity reached 1% of the empirical maximum disease intensity) was used to compare cultivar effects on disease spread. Disease severity is more sensitive than disease incidence to calculate the disease progress and spread of black rot. Measures of progress and gradient were about equally effective to screen cultivars for field resistance to black rot. Perfect Ball was the most susceptible, Erdeno and Bartolo were intermediate and Roxy was the most resistant for incidence and severity measures. Increased levels of field resistance reduced the development of black rot in time and in space. Field resistance of black rot is thought to be composed of several mechanisms. Microplots provide a good instrument for the assessment of small differences in field resistance, expressed equally well in disease progress as in disease gradient curves.  相似文献

Three segregating F2 populations were developed by self-pollinating 3 black rot resistant F1 plants, derived from across between black rot resistant parent line 11B-1-12 and the susceptible cauliflower cultivar ‘Snow Ball’. Plants were wound inoculated using 4 isolates ofXanthomonas campestris pv. campestris (Xcc) race 4, and disease severity ratings of F2 plants from the three populations were scored. A total of 860 arbitrary oligonucleotide primers were used to amplify DNA from black rot resistant and susceptible F2 plants and bulks. Eight RAPD markers amplified fragments associated with completely disease free plants following black rot inoculation,which segregated in frequencies far lower than expected. Segregation of markers with black rot resistance indicates that a single, dominant major gene controls black rot resistance in these plants. Stability of this black rot resistance gene in populations derived from 11B-1-12 may complicate introgression into B. oleracea genotypes for hybrid production. Summary Fusarium bulb rot is a serious tulip disease. Breeding for resistance may contribute considerably to a solution of the problem.It has been demonstrated that juvenile and adult bulbs of the same cultivars in Fusarium contaminated soil showed good agreement in degree of resistance.From an incomplete diallel cross of these cultivars second-year bulblets of 62 progenies were planted in both contaminated and non-contaminated soil. The percentages of non-diseased bulbs harvested provided a criterion for resistance. The analysis of combining ability for the degree of resistance revealed that both the mean square of GCA and that of SCA were significant. The relative magnitudes of the GCA and SCA mean squares suggest that resistance is governed primarily by additive gene action. The GCA of individual parents could be estimated and tested. In general it corresponded with their phenotypic behaviour.  相似文献

Bacterial soft rot caused by Erwiniacarotovora subsp. carotovora is amajor disease in Zantedeschia spp.,particularly in cultivars from the sectionAestivae. The disease can partiallybe controlled by cultivation measures, so acombination with resistant cultivars couldeffectively protect the crop. However,resistant commercial Aestivaecultivars are not available yet. By meansof a recently developed non-destructiveresistance test, variation inaggressiveness was observed among fiveisolates of Erwinia carotovora subsp.carotovora without interactionsbetween the isolates and three Zantedeschia accessions. Within elevenaccessions of Z. aethiopica,variation was observed from almost completeto moderate resistance, while theZ. odorata accession was susceptible.All 21 Aestivae cultivars weresusceptible. Within the Aestivaespecies, Z. elliotiana and Z.pentlandii were also susceptible, butwithin twelve accessions of Z.albomaculata, as well as in six accessionsof Z. rehmannii, variation was foundfrom susceptible to moderately resistant.Hence, new sources of resistance wereidentified that show good potentials forresistance breeding.  相似文献

Cassava root rot disease is an increasing problem in Africa where yield losses of about 80% have been recorded. We evaluated 290 African landraces and 306 improved genotypes from the germplasm collections of the International Institute of Tropical Agriculture (IITA), for sources of resistance using root slice laboratory assay. Disease severity was assessed quantitatively by direct percentage estimation (PS) and by use of a rating scale (RS). Both methods of assessment were compared for identification of variability in the germplasm, and genotypes were classified into response groups using an enlarged rank-sum method that combined the PS and RS assessments. The two scoring methods revealed continuous variation (P < 0.001) for resistance in the sets of germplasm. Disease assessments based on PS and RS were highly correlated in both the improved germplasm (r = 0.75) and the landraces (r = 0.72). Based on PS assessment, 50 improved genotypes (16.3%) and 53 landraces (18.3%) showed significantly lower disease scores than the resistant control. The rank-sum method separated each set of collections into highly resistant, resistant, moderately resistant, moderately susceptible, susceptible and highly susceptible groups. Fifty-nine improved genotypes (16.4%) and 61 African landraces (16.9%) were identified as either highly resistant or resistant. Generally, these genotypes exhibited resistance by limiting the growth of the pathogen (reduced amount of invaded surface area). This type of rate-reducing resistance is highly heritable and a quantitative trait which can be harnessed in breeding. Genotypes subsets were identified for further studies into the genetic basis of resistance to root rot disease.  相似文献

Fusarium basal plate rot (FBR), caused by Fusarium oxysporum f. sp. cepae, is an important soil-borne disease of onions worldwide. The causal organism infects the basal stem plate of the bulb and eventually kills the entire plant through degradation of the basal plate. F. o. f. sp.cepae infections in dormant bulbs during storage allow secondary infections to occur. The primary method of infection by F. o. f. sp. cepaeis through direct penetration of the basal stem plate. Infection can also occur through wounded tissue particularly roots and basal portions of bulb scales. The most cost-effective methods of control are crop rotation and host plant resistance. Current research suggests that a single gene, two genes, or multiple genes govern resistance to FBR. Breeding programs have successfully used screening procedures to develop intermediate- and long-day, FBR-resistant cultivars. Stalk rot, also called as charcoal rot in India, caused by Macrophomina phaseolina, is an economically important, soil borne disease in major sorghum growing areas across the world. A population of F9 generation recombinant inbred lines (RILs), derived from IS22380 (susceptible) × E36-1 (resistant), along with parents were phenotyped in sick plots at two locations (Dharwad and Bijapur, Karnataka, India). A total of 85 polymorphic marker loci (62 nuclear and 4 genic SSRs, 19 RAPDs) was available for the construction of genetic map, spanning 650.3 cM in all the ten linkage groups. Analysis with QTL Cartographer (2.5b), adopting composite interval mapping method (LOD > 2.0) at both locations, revealed 5 QTLs at Dharwad and 4 QTLs at Bijapur locations for the component traits of charcoal rot disease resistance. QTLs for number of internodes crossed, length of infection and per cent lodging accounted for 31.83, 10.76 and 18.90 per cent at Dharwad location and 14.87, 10.47 and 26.44 per cent phenotypic variability at Bijapur location, respectively. The QTLs for number of internodes crossed by the rot, length of infection and percent lodging were common across two locations. These QTLs, consistent over environments for the component traits, are likely to assist in marker-assisted selection (MAS) for charcoal rot resistance in sorghum.  相似文献

The pedigree method is often used for developing inbred lines in maize (Zea mays L.). This study was conducted to assess the effectiveness of pedigree selection for improving resistance to Gibberella ear rot in four maize populations. Selection was based on the severity of ear rot symptoms after inoculation with macroconidial suspensions of Fusarium graminearum (Schwabe) into the silk channel (for two populations) and into the developing kernels (for two other populations). Samples of the selfed families (S1 to S5), recovered from remnant seed from the selection programs, were evaluated for disease resistance during three years, using inoculation and evaluation protocols similar to those used during selection. Among-family selection was effective in both of the populations selected after silk inoculation and in one of the populations selected after kernel inoculation. Responses to selection were more evident in later than in earlier generations for both types of inoculation. Changes in the estimated genetic gain over generations were consistent with changes in the variances among families, which tended to increase in early generations and to decrease in later generations. Selection after kernel inoculation seemed to have been more effective than selection after silk inoculation in developing families with more stable resistance. Based on the results obtained here, it seems that responses to family selection could be accelerated without increasing operational costs by increasing selection intensity in later generations and inoculating fewer plants per family.  相似文献

Summary The content of coumarins, as probable phytoalexins, was analysed in four sunflower genotypes that ranged in responses to head rot from highly susceptible to highly resistant. Low levels of all coumarins (scopolin, scopoletin and ayapin) were detected in the three most susceptible genotypes irrespective of time after inoculation. However, in the resistant genotype there was a clear time-dependent disease-induced increase of all coumarins that reached a maximum after 10–14 days. Detailed comparison of the most susceptible and the resistant genotype showed that in the resistant but not the susceptible, scopoletin peroxidase activity increased during the course of the experiment. Results confirmed a clear negative correlation between coumarin content and disease symptoms and in particular for scopolin. Furthermore we show for the first time that scopolin is inhibitory to Sclerotinia at similar doses to scopoletin. As scopolin is known to be less phytotoxic than ayapin and scopoletin, its accumulation may well confer head rot resistance with minimal plant damage and might be one of the bases for resistance to Sclerotinia.  相似文献

为了阐明黑腐病对苗期花椰菜的根系形态和生理的影响,对建立花椰菜抗黑腐病根系育种新途径提供依据,利用黑腐病菌感染花椰菜抗病品种雪峰的叶片后,研究植株根系发生的一系列形态和生理变化。结果表明,接种7天后的植株根长、根干重、地上部干重、根系活力比接种后2天的显著增加。接种处理的植株根长、根干重和地上部干重明显比未接种对照(CK)低,但是接种幼苗的根系活力比未接种对照的根系活力高。接种3天后,接种处理的根系可溶性蛋白质含量一直显著高于未接种对照;接种后0~4天,接种处理的根系超氧化物歧化酶(SOD)活性明显低于未接种对照,但到接种后5~7天,接种处理的根系SOD活性又高于未接种对照;根系多酚氧化酶(PPO)活性和脱落酸(ABA)含量在植株接种后都显著高于未接种对照。但是,黑腐病病原菌没有提高根系过氧化物酶(POD)活性。以上结果说明,黑腐病菌引起了花椰菜幼苗根部一系列形态变化,并通过诱导植株根系中防御酶活性和调控内源激素等抵御病原菌的进一步侵染。  相似文献

Resistance to bacterial soft rot caused by Erwinia carotovora subsp. carotovora is a quantitative trait. The narrow-sense heritability was from 42% to 60% in the studied populations. Griffing's diallel analysis and generation mean analysis indicated that additive genetic effects were most significant, and the resistant sources involved in this study appeared to have different genes or alleles. Recurrent phenotypic selection was used for improving the resistance level by combining different resistance genes from the selected genotypes of Chinese cabbage. After 3 cycles of recurrent selection, the level of resistance increased markedly. When the cycle 3 population was compared with the cycle 0 population, the mean disease severity rating was reduced 38% from 6.3 to 3.9, and the percentage of plant survival increased from 65% to 97% based on the mean of two years experiments using mist-chamber seedling inoculation methods.
