In this work, the contents of oil, sugar and organic acids of olive fruit samples (Domat, Memecik and Uslu) were determined by using HPLC at different maturation stages. Olive fruit samples presented a common organic acid profile, composed of four constituents: oxalic, citric, malic and succunic acids. Total organic acid content of Domat variety was found to be 4787.1 ± 4.53 mg/100 g in January. Memecik variety had the highest value at the beginning of the ripening period (6385.08 ± 4.90 mg/100 g). The amount of organic acid in Uslu variety reached up to the value of 10942.5 ± 30.41 mg/100 g. The results were evaluated statistically to determine relationship between the oil, sugar and organic acid content in three olive varieties during maturation. A positive correlation was found between the total sugar and organic acid (r = 0.54, p < 0.01) whereas a low positive correlation was observed between the contents of oil and total organic acids (r = 0.46, p < 0.05) in olive varieties.  相似文献

An experiment was carried out in a young high-density olive grove (556 plants ha−1Olea europaea L., cv Coratina) located in Southern Italy to evaluate the effect of different soil water availability on the vegetative and productive performances of olive trees also looking into the quality of the resulting oils. Trials were carried out over a 3-year period on trees subjected to irrigation and grown under rainfed conditions. Vegetative tree response, as shoot elongation and trunk diameter, was evaluated. Yield per plant, fruit characteristics, oil quality indices (free fatty acid content, peroxide value, UV adsorption at 232 and 270 nm, total phenols, α-tocopherol content) were determined for both irrigated and non-irrigated treatments in the ‘on’ years 1997 and 1999 (6th and 8th year after planting, respectively).  相似文献

The composition of volatile components of the essential oils extracted from fruits of coriander (Coriandrum sativum L.) growing in two different Tunisian regions in both Menzel Temime and Borj El Ifaa was studied by GC and GC–MS. The highest oil yields were observed at final stages of maturity: 0.324 ± 0.09% and 0.327 ± 0.08%, in Menzel Temime and Borj El Ifaa, respectively. Essential oil yields were highly (P < 0.001) affected by the growing region, stages of maturity and their interaction. Essential oil composition changed significantly (P < 0.05) among the different stages of maturity and growing region. Linalool and geranyl acetate were the main compounds at full fruit maturity in the two studied regions. Growing region, stages of maturity and their interaction had a strong effect (P < 0.001) on 35 compounds. α-Pinene, sabinene, limonene, γ-terpinene, cis-dihydrocarvone and geranial were insensible to the regional factor.  相似文献

Volatile composition of virgin olive oil (VOO) from six Tunisian new cultivars was evaluated using headspace solid-phase microextraction method coupled to GC–MS and GC–FID. Various chemical classes were identified such as aldehydes, alcohols, esters, ketones, terpenes and carboxylic acids. The results showed that the most important contributors to olive oil aroma are C6 aldehydes. These compounds are biogenerated from polyunsaturated fatty acids through the lipoxygenase pathway. Furthermore, they pointed out the predominance of the linolenic acid oxidation over linoleic acid one. The main volatile compounds present in the oil samples were C6 derivatives such as (E)-hex-2-enal, (E)-hex-2-enol and hexanol. In addition to C6 compounds, the aroma of the studied VOOs contains reasonable amounts of various classes of C5 components. The tested oil samples showed different volatile profiles. Some components seem to occur exclusively in a specific oil samples and, consequently, their presence might be initially used to establish compositional differences. All results put in evidence that olive oil aroma compounds accumulate differently according to the cultivar.  相似文献

This work is an evaluation of the quality of three introduced European olive cultivars Arbequina, Coratina, and Koroneiki grown under a biological agricultural system in the arid zone of Sfax in Tunisia compared to Chemlali cultivars. Various olive parameters were analyzed, such as ripening index, pulp/stone (P/S) ratio and oil content. We have considered the regulated physicochemical analytical parameters, fatty acids and minor compounds for olive oil. The most of the quality indices and fatty acid composition showed significant variations among olive cultivars. The Koroneiki, Coratina and Chemlali Zarzis cultivars had highest values of oleic acid (62.7%) (76.8%, 75.8% and 73.9%, respectively), Arbequina, had lowest value of oleic acid and similar to Chemlali Sfax. Coratina was noteworthy for its higher content of phenolic compounds (287.8 mg kg−1), oil content (42.4%) and P/S ratio (4.7%). Although their low phenol contents, autochthonous cultivars presented high contents of α-tocopherol (577.8 and 434.6 mg kg−1 for Chemlali Boughrara and Chemlali Zarzis, respectively) except for Chemlali Sfax. In conclusion, European cultivars seem to have adapted to the area studied according to their oil biochemical composition while the increase observed in their α-tocopherol levels. The cultivar Arbequina showed a similar composition of Chemlali Sfax.  相似文献

This study was initiated to investigate the differences in germination percentages and rates between Corylopsis coreana Uyeki and Corylopsis sinensis var. calvescens Rehder & E.H. Wilson following a warm stratification (WS) and cold stratification (CS), and to study the effect of different WS temperatures interacting with different durations of CS. Warm stratification at 10 °C, 15 °C, 20 °C, and 25 °C was given for 1 month (1 M 10 °C, 15 °C, 20 °C, and 25 °C WS) followed by 0 M, 1 M, 2 M, and 3 M of CS at 5 °C (0 M, 1 M, 2 M, 3 M CS) and seeds were germinated in an air conditioned greenhouse maintained at 18.5 °C/18 °C. On average, less than 1% of C. coreana seeds germinated when sown without any WS and CS or with 1 M 15 °C, 20 °C, and 25 °C WS without CS treatment. However, 26% C. coreana seeds germinated after 1 M 10 °C WS without any CS treatment. Germination was not affected by WS temperatures when followed by 2 M 5 °C CS. It is concluded that C. coreana exhibited low seed germination at 10 °C and that this temperature could be considered the upper limit of CS for C. coreana. Only 2 M CS was required for more than 90% seeds to germinate. However, C. sinensis var. calvescens required longer than 3 M CS for more than 29% seeds to germinate. This clearly shows that there is an interspecific variation in optimum dormancy-breaking requirements.  相似文献

Carob (Ceratonia siliqua L.) is natively grown in the coastal areas of the Aegean and Mediterranean region in Turkey. The main carob growing areas (Mediterranean and Aegean) were surveyed and a total 70 promising wild and grafted carob genotypes were selected based on physical and chemical pod properties. Preliminary results showed that Turkey has very rich carob genetic resources containing considerable variations for most of the pod traits. In general, wild carob had smaller pods than the grafted ones. Pod mass ranged from 29.16 g to 120.28 g in the wild genotypes and 71.71 g to 147.91 g in the grafted genotypes with an average 107.61 g for the grafted and 63.72 g for the wild genotypes. The average pod dimensions (width, length and thickness) were 18.16 mm, 16.18 cm and 6.42 mm for the wild genotypes and 21.79 mm, 16.91 cm and 8.79 mm for the grafted genotypes. The average soluble solid content ranged from 59.42% in the wild and 64.06% in the grafted genotypes. The average total acidity of the wild and grafted genotypes was similar (0.78%). The pod traits of the wild genotypes differ significantly from those of the grafted genotypes, and the wild genotypes showed higher variations in these traits compared to the grafted genotypes.  相似文献

Changes in membrane lipid composition are important in the acclimation of plants. The influence of four day/night growing temperature combinations (18/12, 25/12, 25/22, and 30/22 °C) on membrane lipids of ‘Earliglow’ and ‘Kent’ strawberry (Fragaria × ananassa Duch.) were studied. The monogalactosyl diglyceride (MGDG) and digalactosyl diglyceride (DGDG) were the major galactolipids in the strawberry leaves. ‘Earliglow’ contained a higher amount of galactolipids in the leaves than ‘Kent’. The major phospholipids in the strawberry leaves, roots, and fruit were phosphatidylcholine (PC), phosphatidylinositol (PI), phosphatidylglycerol (PG), and phosphatidylethanolamine (PE). PC and PE were the two predominant phospholipids in the strawberry. The leaves and fruit of ‘Earliglow’ contained higher amounts of phospholipids compared to those of ‘Kent’, whereas ‘Kent’ strawberry roots had higher phospholipids. Palmitic (C16:0), stearic (C18:0), oleic (C18:1), linoleic (C18:2), and α-linolenic (C18:3) acids were major fatty acids in galacto- and phospholipids of the ‘Earliglow’ and ‘Kent’ strawberry. PC is very rich in linolenic acid in leaves compared to the fruit and root tissues. PC had the highest ratio of unsaturated to saturated fatty acids among all phospholipids. There was a significant increase in the content of galactolipids (MGDG, DGDG) and phospholipids (PC, PI, PG and PE) and unsaturation of their fatty acids in the cool day/night growth temperature. Increasing day/night growth temperatures decreased MGDG/DGDG ratios. The shifts in saturation and composition of fatty acids observed with strawberry may be an adaptation response of plants to the temperature changes.  相似文献

In vitro seeds germination and plantlet establishment of Labisia pumila were studied in this report. The seeds obtained from the mature fruits of L. pumila were sterilized and cultured on Murashige and Skoog (MS) solid media supplemented with 1–3 μM of 6-benzylaminopurine (BAP) and 3% (w/v) sucrose. The presence of BAP in the medium significantly affects seeds germination. High percentage of seeds germination (up to 90%) was successfully achieved after 2 weeks of culture on medium supplemented with 2 μM BAP. Up to 70% of explants produced shoots through direct regeneration from newly emerged epicotyls after 5 weeks of culture. The average of 8.1 ± 1.0 shoots per explant obtained on media treated with 2 μM BAP. Seedlings were further transferred to growth media fortified with different types of cytokinin. Result observed after 12 weeks showed that medium supplemented with 1 μM zeatin (ZEA) promote the highest growth with an average of 2.9 ± 1.0 cm shoot length and 7.7 ± 3.2 leaves per explant after 12 weeks. In addition, medium added with 2 μM BAP and supplemented with 3–4% (w/v) of sucrose promote the best growth i.e., 3.0 ± 0.6 shoots per explant, 2.27 ± 0.2 cm length and 4.3 ± 0.5 leaves per explant.  相似文献

The influence of bulb maturity at bulb harvest on growth and flowering response of Ornithogalum thyrsoides Jacq. ‘Chesapeake Starlight’ was investigated. Experiments were designed to determine if bulb maturity can be induced by bulb storage temperatures and whether bulb maturity can be evaluated by flowering responses. Bulbs with all senesced leaves at harvest were considered “mature” or with emerging young leaves and re-growing young roots were considered “immature”. Bulbs were potted after 0, 3, and 6 weeks of 30 °C or 2 weeks of 10 °C given either in the middle or at the end of 6 weeks of 30 °C. Mature bulbs, as compared to immature bulbs, took longer for leaves to emerge when control bulbs that did not receive any temperature treatment after harvest were planted upon harvest. Leaf emergence of the immature bulbs was significantly earlier than that of the mature bulbs. Mature bulbs which received 30 °C for 3 weeks (30 °C/3 week) flowered 31 days faster than immature bulbs and all bulbs flowered. Leaf emergence and flowering of mature and immature bulbs that received 30 °C/6 weeks or 2 weeks of 10 °C in the middle of 6 weeks of 30 °C (30 °C/2 weeks–10 °C/2 week–30 °C/3 weeks) did not differ from each other. Maturity can be induced by storing immature bulbs at 30 °C/6 weeks. Maturity, as evaluated by flowering percentage and days from leaf emergence to flowering, can be induced in O. thyrsoides. Immature bulbs can, therefore, be harvested for later forcing as long as bulbs are treated with 30 °C/6 weeks. It is proposed that maturity can be correlated with the speed of flowering and bulbs can be harvested at immature physiological state for forcing. Postharvest high-temperature treatment can be used to force immature bulbs that were harvested before the senescence of the leaves.  相似文献

Based on the antioxidant properties of essential oils, the effect of thyme, coriander, and rosemary was evaluated on the reduction of peroxidase in white cabbage “Brassica oleraceae var.capitata f. alba” and red cabbage “B. oleraceae var. capitata f. rubra”. To do this, pure, 50, 75, 100 and 200 μl/100 ml concentrations of essential oils were applied in vivo and in vitro of cabbages. Antioxidant activity of the essential oils was compared with that of ascorbic acid. Results revealed that the highest percentage of antioxidant activity was achieved in vitro using a pure concentration of coriander (52.65%) and rosemary (54.64%), and in vivo applying thyme (60.07%) of white cabbage. Applying in vitro pure concentration of coriander (46.39%) and 50 μl/100 ml of rosemary (41.64%), and in vivo 75 μl/100 ml of thyme (42.64%) in red cabbage showed high antioxidant activity. A high reduction of peroxidase was obtained in vitro using of essential oils in red and white cabbages.  相似文献

Guilan is one of the main province olive oil producers in Iran. The culture of olive trees is of considerable importance. One problem that affects Iran olive orchards is a disease bacterial canker caused by Pseudomonas syringae pv. syringae. In this research, the bacterium was recovered from sunken brown stem lesions on 2-year-old olive trees cv. Marie, during survey from olive orchards in different areas of Guilan province. Samples were taken from infected tissues and were washed with sterile distilled water and crushed in peptone water. Then 50 μl of the extract was cultured on NA and King's B medium containing cyclohexamid antibiotic (50 μg/ml). After 48–72 h, bacterial colonies were selected. Based on morphological, physiological, biochemical, pathogenicity properties and PCR methods with specific primers the predominant pathogenic type was identified as P. s. pv. syringae. This is the first report of the existence of P. s. pv. syringae on olive trees in Iran.  相似文献

Studies were carried out on the oil content, fatty acids, tocopherols and sterols in the seeds of two carobs (Ceratonia siliqua), cultivated and wild, from Turkey. The oil yields of cultivated and wild carob seeds were established as 1.73 and 1.82% on a dry weight basis, respectively. The main fatty acids in cultivated and wild carob seed oils are linoleic (49.1% and 51.0%), oleic (30.4% and 26.5%), palmitic (10.3% and 12.0%) and stearic (3.5% and 4.6%), respectively. Tocopherols and phytosterols are important constituents of the unsaponifiable fraction of several seed oils. The major tocopherol in both seed oils was γ-tocopherol. The composition (mg/100 g) was as follows α-tocopherol (69.06 and 70.39 mg/100 g), P8 (22.29 and 24.78 mg/100 g), δ-tocopherol (8.70 and 10.66 mg/100 g), β-tocopherol (2.30 and 1.85 mg/100 g). The total tocopherol contents had 208.45 and 223.14 mg/100 g, respectively. The total content of sterols of both oils were determined as 16400.94 and 30191.55 mg/kg, with β-sitosterol as the predominant sterol that accounted for more than 70% of the total amount of sterols other sterols, campesterol (5.33–5.32%), stigmasterol (0.58–11.43%) 7-avenasterol (3.45–3.03%), 7-stigmasterol (2.16–2.4%), and chlerosterol (1.33–1.0%) were detected in both carob oils. As a result, the accurate quantification of these analyses has very important applications for the nutrition sciences.  相似文献

Two clonal selections of lemon trees (‘Verna-62’ and ‘Verna-50-2’), were studied in aiming to ascertain the influence of genetic (clone) and environmental (season) factors on the human-health bioactive compounds of the lemon juice (vitamin C and flavonoids) and the possible relationship between composition and in vitro antioxidant capacity (DPPH, ABTS and FRAP) of the juice. The average values determined in bioactives were 13–26 mg·100 mL−1 in total of analysed phenolics and 23–29 mg·100 mL−1 in vitamin C content. Variability in the weather conditions and internal physiological phenomena of the fruits such as biosynthesis and transport, could determine, at least in part, differences in the contents of lemon juice bioactives more importantly than the genetic background, providing the food industry with phytochemically rich and nutritive lemons for processing and functional ingredients.  相似文献

High-frequency somatic embryogenesis and shoot regeneration of broccoli (Brassica oleracea var. italica) were achieved. Cotyledon and hypocotyl explants from four varieties of broccoli were cultured on MS and modified MS media (mMS, supplemented with PG-96 organic components) with different combinations of growth regulator. The effects of genotypes, different explants, growth regulator combinations, organic components and AgNO3 on induction of calli and shoots were evaluated. The optimal media for inducting calli/shoots and roots were mMS medium containing 3% (w/v) sucrose and 0.8% (w/v) agar supplemented with NAA at 0.5 mg l−1, 6-BA at 3.0 mg l−1, AgNO3 at 4.0 mg l−1 and MS medium containing 3% sucrose and 0.8% (w/v) agar supplemented with NAA at 0.2 mg l−1, respectively. The callus induction percentages were over 90% in all four varieties; shoot induction percentage was 92.5% and the average number of shoot per explant was 4.1 from cotyledon explant in variety Bishan. In this study, we established high-efficient embryogenesis and shoot regeneration system of broccoli and analyzed genetic stability of regenerants at DNA level using RAPD molecular marker. Out of 62 arbitrary primers screened using PCR amplification, 79 polymorphic bands were amplified from 20 primers. The results demonstrated the genetic stability of regenerants from the same variety.  相似文献

Measurements of sap flux were carried out from May 2003 to March 2004 on 6 year-old irrigated olive trees of cultivar Chétoui cultivated at 6 m × 6 m spacing in Mornag (36.5°N, 10.2°E), Northern Tunisia. The aim of the research is to evaluate the sap flux technique for its applicability with young olive trees and to estimate their water consumption under field conditions. Three thermal sensors were implanted in the trunk of three olive trees following to North (N), South-East (SE) and South-West (SW) directions. Data were analyzed following to the procedure of Do and Rocheteau (2002b) that derives from Granier (1985). In this paper, data on probe calibration, wood conductive section estimation and sap flux spatial-variability are presented and discussed. Relationships between sap flux measurements, climate and soil water status have been investigated. Results show that sap flux values vary with sensor position, soil water content and climate demand. Good agreements between sap fluxes and global radiation and reference evapotranspiration measurements were observed. Some variations were recorded under water shortage conditions. Maximum and minimum daily fluxes of 4.5 l and 41.0 l per tree were found in February 2003 and in August 2003, respectively. Maximum transpiration represented only 53% of the crop evapotranspiration as determined by the F.A.O. method.  相似文献

Heat treatments have been applied in fruit postharvest technology for insect disinfestations, decay control, ripening delay and modification of fruit responses to other stresses. Heat treatment affects several aspects of fruit ripening, such as ethylene production and cell wall degradation probably through changes in gene expression and protein synthesis. In this paper, strawberries (Fragaria × ananassa Duch., cv Camarosa) at 50–75% red stage of ripening were heat-treated at 45 °C during 3 h in an air oven and then stored at 20 °C for 0, 4, 18, 24 and 48 h. Fruit firmness was determined and the expression of a set of expansin genes (FaEXP1, FaEXP2, FaEXP4, FaEXP5 and FaEXP6) was analyzed. The firmness of treated fruit was higher than that of control fruit 24 h after treatment, though the differences disappeared after 48 h at 20 °C. The analysis by northern-blot indicated that heat treatments affected differently the expression of expansin genes. The expression of FaEXP1, FaEXP2 and FaEXP6 was lower in treated fruit during the following 24 h post-treatment. The lower expression of these expansin genes could contribute to delay softening after heat treatment.  相似文献

Pogonatherum paniceum (Lam.) Hack. is a rock plant with good potential for vegetative recovery on naked lands. A high frequency in vitro regeneration system was developed for P. paniceum. Calli were induced from explants of mature seeds, seedlings, young leaves, and stem segments on Murashige and Skoog (MS) medium supplemented with 1.0 mg L−1 2,4-dichlorophenoxyacetic acid (2,4-D), 2.0 mg L−1 α-naphthalene acetic acid (NAA) and 0.2 mg L−1 6-benzylaminopurine (BAP). High induction rates (59.57%) and regeneration rates (100%) were obtained from mature seed explants; calli were sub-cultured for over 2 years and still retained a high regenerative capacity. One seed explant resulted in 69,997 plants in 1 year. Shoot buds derived from calli were used for encapsulation in liquid MS medium containing 3% sucrose and two different alginate matrices (3% sodium alginate (w/v) + MS medium containing 3% sucrose and 3% sodium alginate + 1% activated carbon (w/v) + MS medium containing 3% sucrose) with a 20-min exposure to 2% CaCl2 and 0.3% bavistin (w/v). The capsule with 3.0% sodium alginate (w/v) and 1% activated carbon (w/v) showed a higher conversion rate (61.58%) and stronger plantlets under non-aseptic conditions. These systems are useful for the rapid clonal propagation and dissemination of artificial seed material of P. paniceum for eco-recovery.  相似文献
