The effects of nickel in combination with salicylic acid (SA) and sodium nitroprusside (SNP), a donor of nitric oxide (NO) on 21-day-old canola plants were evaluated. Exposure to 0.5 mM NiCl2·6H2O for 10 days resulted in toxicity symptoms such as chlorosis and necrosis at leaves. Addition of 0.2 mM SA or 0.2 mM SNP slightly reduced the toxic effects of nickel. After application of both SA and NO, these symptoms considerably decreased. Treatment with Ni resulted in a decrease in dry weight of roots and shoots and chlorophyll content of leaves. In Ni-treated plants, level of lipoxygenase activity and malondialdehyde (MDA), H2O2 and proline contents significantly increased, while the activities of the antioxidant enzymes such as catalase, guaiacol peroxidase and ascorbate peroxidase decreased in leaves. The results indicated that Ni caused an oxidative stress in canola plants. The Ni-stressed plants exposed to SA or NO, especially to SA + NO, exhibited an improved growth as compared to Ni-treated plants. SA or NO, especially both together considerably reduced root-to-shoot translocation of Ni and increased the activities of the antioxidant enzymes in leaves of Ni-stressed plants. Interaction of SA and NO improved the chlorophyll content and decreased the level of lipid peroxidation, H2O2 and proline accumulation in leaves. These results suggest that SA or NO in particular their combination counteract the negative effects of Ni on canola plants.  相似文献

AIM: To study the effects of nitric oxide (NO) on mitochondrial damage caused by exogenous calcium. METHODS: Normal myocardial mitochondria were divided into three groups; L-arginine control group (CG), Ca2+-damaged group (DG) and L-NAME-preserved group (PG). Mitochondria of all groups were incubated at 30 ℃ with reaction medium containing 20 μmol/L EDTA, 100 μmol/L CaCl2 and 1 μmol/L L-NAME with 100 μmol/L CaCl2 respectively. Then the NO2-/NO3- contents, mitochondrial viability and membrane potential were investigated. RESULTS: The NO2-/NO3- contents of DG was obviously higher than that of CG and PG, meanwhile, there was no obvious difference between CG and PG. Mitochondrial viability and membrane potential of DG were significantly lower than that of CG and PG, and negatively related to NO-2/NO-3 contents (r=-0.5297, P<0.01; r=-0.6041, P<0.01). But, the mitochondrial viability and membrane potential of PG were still lower than that of CG. CONCLUSION: Exogenous calcium could activate mitochondrial nitric oxide synthase resulting in NO production and the latter play an important role in decreasing mitochondrial viability and membrane potential.  相似文献

Nitric oxide (NO) is a key signaling molecule in different physiological processes of plants. To study the metabolism of endogenous NO in growth and development of apple fruit under normal physiological conditions, the changes of several relative components of NO metabolism including the contents of NO, l-arginine, S-nitrosothiols (RSNOs) and nitrite plus nitrate, the activities of l-arginine-dependent nitric oxide synthase (NOS) and nitrate reductase (NR) were analyzed using vegetal ‘Gold Delicious’ and ‘Fuji’ apples as tested materials. The endogenous NO and l-arginine contents, as well as the l-arginine-dependent NOS activity in young fruit reached the highest value. The NOS activity was dependent on l-arginine and β-NADPH, and inhibited by a well known NOS inhibitor. The NR activity in apple fruit had not been detected. Endogenous NO production depended on l-arginine-dependent NOS but not NR. The content of RSNOs tended to peak in the fruit enlarging period and then declined. Similarly, the nitrate content increased in early stage of fruit growth, but in late stage, it undulated greatly. Compared to RSNOs and nitrate, the change of nitrite content was in accordance with that of endogenous NO. In general, RSNOs and nitrite could reflect the removal mechanism of NO relatively well.  相似文献

Linaria maroccana Hook. f. Ann., ‘Lace Violet’, Lupinus hartwegii ssp. cruikshankii Lindl. ‘Sunrise’ and Papaver nudicaule L. ‘Meadow Pastels’ seeds were directly sown into 105 cell plug trays and received either ambient light or supplemental high intensity discharge (HID) lighting. For each species, a 2 × 3 × 3 factorial was used with two light intensities during propagation, three transplant stages, and three night temperatures. Seedlings were transplanted at the appearance of 2–3, 5–6, or 8–9 true leaves. Transplanted Linaria and Papaver seedlings were placed at 5/11, 10/16, or 15/21 ± 1 °C night/day temperatures and Lupinus seedlings were placed at 15/24, 18/25, or 20/26 ± 2 °C night/day temperatures. For this study, the optimum production temperature for Linaria was 10/16 °C as the cut stems produced at 15/21 °C were unmarketable and production time was excessively long at 5/11 °C. At 10/16 °C, Linaria seedlings should be transplanted at the 2–3 leaf stage to maximize stem number, stem length and profitability. For Lupinus the optimum temperature was 15/24 °C due to long stems and high profitability per plant. Lupinus seedlings should be transplanted at the 2–3 leaf stage when grown at 15/24 °C to obtain the longest and thickest stems; however, $/m2 week was higher for plants transplanted at the 8–9 leaf stage due to less time in finishing production space. For Papaver, the 15/21 °C temperature was optimal as that temperature produced the longest stems in the shortest duration, resulting in the highest $/m2 week. At 15/21 °C Papaver plants should be transplanted at the 2–3 leaf stage. Supplemental HID lighting had no effect on any of the species.  相似文献

探讨了外源NO对水分胁迫下核桃叶片荧光参数的影响。结果表明,水分胁迫下,核桃叶片的荧光参数最大光化学量子效率(Fv/Fm)、PSⅡ反应中心的实际光化学效率(φPSⅡ)、光化学猝灭系数(qP)、非光化学猝灭系数(NPQ)均降低,经NO供体硝普钠(SNP)诱导处理后,荧光参数下降程度减小,且随着SNP处理浓度增大,缓解作用愈明显。表明外源NO处理一方面提高了光能转换效率和PSⅡ的潜在活性,另一方面增强了过剩光能的非化学耗散,有利于保护光合机构免受环境胁迫的伤害。  相似文献

AIM and METHODS:The animal model of acute lung injury (ALI) caused by intratracheal instillation of lipopolysaccharides(LPS) in vivo and human peripheral blood polymorphonuclear neutrophil (PMN) in vitro were used to study the effects of sodium nitroprusside (SNP), nitric oxide (NO) donor, on LPS-induced PMN accumulation, microvascular permeability and PMN apoptosis. RESULTS:①In vivo, PMN accumulation in lung, the protein content in bronchoalveolar lavage fluid (BALF) and the Evans blue dye and monastral blue dye extravasation in lung tissue of LPS group were markedly higher than those of both sham operation group and LPS+SNP group. ②In vitro, the apoptotic percentage of SNP group was much higher than that of control group, while compared with LPS group, SNP+LPS group has significantly higher apoptotic percentage. CONCLUSIONS:SNP intratracheal instillation attenuated LPS-induced microvascular permeability and alleviated ALI. PMN apoptosis induced by SNP may be one of the potential mechanisms underlying the decrease of PMN accumulation in lung tissue.  相似文献

A short postharvest life is the major constraint associated with cut Acacia flowers and foliage. Treatment with CuSO4 (Cu2+) has previously been shown to improve the longevity of cut Acacia holosericea stems. Towards refining the treatments, a range of Cu2+ and Cu+ salts were assessed for relative efficacy in improving vase life and water relations of A. holosericea. Five hour pulses with the Cu2+ salts CuSO4, CuCl2, (CH3COO)2Cu and Cu(NO3)2 at 2.2 mM gave equally longer vase lives by ∼2.5-fold over deionised water (DIW) and standard tap water (STW) controls. The same Cu2+ salts at 0.5 mM in the vase solution also gave significantly (P < 0.05) improved vase life, relative fresh weight and water uptake compared to the DIW control. For Cu2+ versus Cu+, optimum concentrations with Cu2+ could not be directly compared due to the low solubility of the Cu+ salt CuCl. However, Cu+ from CuCl at 0.415 mM also had positive effects on vase life compared to the DIW control. Thus, both Cu2+ and Cu+ treatments can enhance vase life parameters for cut A. holosericea foliage. The benefits were irrespective of the counter ion and, thus, Cu2+ and Cu+per se were responsible. The most effective Cu2+ pulse treatment decreased stomatal conductance of phyllodes initially, but did not cause sustained stomatal closure. Cu accumulated to greater levels in basal stem and phyllode tissues than in upper stem and phyllode tissues of cut A. holosericea stems. Possible mechanisms of Cu2+/Cu+ action are discussed.  相似文献

The state of Morelos, Mexico has gradually become an important producer of gladiolus. Some preconditioning treatments of corms are empirically done causing uneven emergence and low quality of flowers. In this investigation, before planting, gladiolus corms var. ‘Blanca Borrego’ were dipped in chitosan (chitosan reagent and commercial chitosan Biorend®), in hot water at various temperatures and in treatments combined with Biorend® at 1.5% and hot water. Results indicated that the most influenced variables were corm germination, number of flowers per spike, number of cormlets and vase life. Overall, the commercial product Biorend® at 1.5% accelerated corm emergence in approximately 4 days, the number of flowers increased by 2–7 and the vase life extended for 3 days. The number of cormlets was also duplicated. Corms dipped in the commercial chitosan Biorend® at 1.5% at different intervals of time were not greatly affected except for the emergence and number of cormlets. However, for this experiment there were significant effects on the number of leaves and flowers because of the interactions between chitosan and the immersion time. The temperature of 55 °C affected plant development because emergence was delayed by 6 days; and there were less number of leaves, flowers and cormlets. On the other hand, the incidence of Fusarium oxysporum in naturally infected corms was 0% at temperatures of 55 °C and 50 °C. Immersion times (0, 10, 15 and 20 min) in hot water at 50 °C did not show significant effects on plant development and vase life. Corms dipped in Biorend® at 1.5% and hot water at 50 °C accelerated their emergence for about 1–7 days, the number of flowers increased by two, extended the storage life for 1–3 days and increased the number of cormlets. The integration of these two treatments -Biorend® and hot water- might be a good option for increasing the gladiolus plant quality and vase life.  相似文献

Post-harvest performance of cut viburnum inflorescences was examined in the present study. Harvesting viburnum at three developmental stages resulted in significant differences on flower opening rate (FOR) and flower abscission rate (FAR), but not on vase life. Harvesting at Stage III (>50% open flowers) resulted in highest flower opening percentage, whereas harvesting at Stage I (10–30% open flowers) in significantly lower flower abscission percentage. Pulsing with 20 or 40 mg/l AgNO3 for 24 h extended vase life by 1.6 and 1.9 days, respectively, compared to the controls. Furthermore, flower abscission was significantly reduced after 20 or 40 mg/l AgNO3 treatments by 51 and 62%, respectively, compared to the controls. In contrast, vase solutions containing 100 mg/l DICA or 100 and 200 mg/l methanol did not extend vase life of cut viburnum inflorescences, but significantly reduced flower abscission. Vase solutions containing 1 or 2% (w/v) sucrose negatively affected vase life, flower opening and flower abscission of viburnum inflorescence. Post-harvest treatments with 1-MCP at 10 μl/l in an ethylene-free environment resulted in extension of vase life and in significant reduction of FAR and respiration rates compared to the controls. Vase life of 1-MCP treated inflorescences was increased by 4.2 days compared to the controls. FAR of 1-MCP treated inflorescences remained significantly lower from the second to the eighth day of the vase life period.  相似文献

AIM: To study the up-regulation of inducible nitric oxide synthase (iNOS) in lung of pulmonary fibrosis and its relationship with fibrosis. METHODS: The changes of amount of iNOS positive stain cells and type Ⅰ?Ⅲ collagen were examined on the day 7, 14 and 30 after intratracheal administration of bleomycin A5. The contents of NO2-/NO3- (nitrite/nitrate) in out-flowing pulmonary blood (OPB), hydroxyproline in lung and the histological changes were detected after iNOS was blocked by aminoguanidine (AG). RESULTS: (1) The number of iNOS-positive stain cells increased significantly in BLMA5 7 d, 14 d and 30 d groups compared with that in control group (P<0.01). Furthermore, the increment of the number of iNOS-positive stain cells in BLMA5 7 d, 14 d groups was more than that in BLMA5 30 d group. There was an increment of collagen in BLMA5 14 d group and in BLMA5 30 d group , with an increase in type Ⅲ collagen in BLMA5 14 d group and an increase in type Ⅰcollagen in BLMA5 30 d group. (2) The high level of NO2-/NO3- in OPB and hydroxyproline level in lung could be reversed by AG, a selective inhibitor of iNOS. Large amount of fibroblasts and macrophages were also abated by AG. CONCLUSION: In the development of pulmonary fibrosis, the expression of iNOS is up-regulated, which induces nitric oxide (NO) production and promotes propagation of pulmonary fibrosis.  相似文献

Tolerance to drought-stress (DS) of the citrus rootstock Forner–Alcaide no. 5 (FA-5) was tested and compared with that of its parents, Cleopatra mandarin (CM) and Poncirus trifoliata (PT). Nine-month-old seedlings of CM, PT and FA-5 and 15-month-old grafted trees of ‘Valencia’ orange scions on these three rootstocks were cultivated in sand under glasshouse conditions and irrigated with a nutrient solution. Plants were drought-stressed by withholding irrigation until leaves were fully wilted. Survival time of both seedlings and grafted trees under DS was linked to the water extraction rate from the soil, which depended mainly on leaf biomass and on transpiration rate. Seedling responses to DS affecting leaf water relationships and gas exchange parameters varied among genotypes. FA-5 seedlings survived longer than the other seedlings, maintaining the highest levels of water potential, stomatal conductance, transpiration rate and net CO2 assimilation towards the end of the experiment, when water stress was most severe. Thus, FA-5 was more resistant to DS than its parents (CM and PT). Moreover, rootstock affected the performance of grafted trees under water stress conditions. The higher drought tolerance induced by FA-5 rootstock could be related to the greater osmotic adjustment (OA), which was reflected by smaller reductions in leaf relative water content (RWC) and in higher turgor potentials and leaf gas exchange than the other rootstocks.  相似文献

AIM:To observe the dynamic changes of plasma levels of nitric oxide(NO) and endothelin (ET-1) in portal veins of the rats during prehepatic portal hypertension, and investigate the role of them in hyperdynamic circulation.METHODS:The models of prehepatic portal hypertension were established in Sprague-Dawley rats by means of partial portal vein ligation (PVL). The plasma levels of nitrite/nitrate (NO2-/NO3-) and ET-1 in the portal veins were detected by the method of nitric reductase and radioimmunoassay, respectively. In this study, rats were divided into normal, sham operation (SO) and PVL group. SO and PVL rats were divided into several subgroups according to different time after operations. Meanwhile, the changes of several hemodynamic indexes in these rats were also measured.RESULTS:The levels of NO2-/NO3- were significantly increased and ET-1 were significantly decreased in rats at different time after PVL compared with normal control, whereas the hemodynamic indexes changed accordingly.CONCLUSION:The portal hypertensive rats are in hyperdynamic circulatory state (HCS). NO and ET-1 may play an important role in the induction and maintenance of HCS.  相似文献

AIM: To observe the kinetic alteration of nitric oxide formation in the lungs in the development of pulmonary fibrosis in the rat. METHODS: The contents of hydroxyproline in the lungs, NO2-/NO3- (nitrite/nitrate) in out-flowing and in-flowing pulmonary blood (OPB, IPB) were assayed on the day 7, 14, 21, 30 and 70 after intratracheal administration of bleomycin A5 . The content of NO2-/NO3- in supernatants of culture of the alveolar macrophages (AMs) and the amount of iNOS positive stain cells in lung tissue section were also observed in the rat on 14th day after-bleomycin A5 administration. RESULTS: The content of lung hydroxyproline had no change on the 7th day, increased on the 14th day (P<0.05), increased significantly on the 21th day, 30th day and 70th day post-bleomycin A5 compared with control rats. On the 7th day and 14th day, the content of NO- 2 /NO3- increased in OPB and decreased in IPB (P<0.01). On the 21th day, the content of NO2-/NO3- abated in OPB (P>0.05) but still decreased in IPB (P<0.01). On the 30th day and the 70th day, the NO2-/NO3- level recovered both in OPB and IPB. AMs from rats on the 14th day post-bleomycin A5 showed significant elevation (P<0.01) in NO2-/NO3- level. The amount of iNOS positive stain cells increased in rats on the 14th day post-bleomycin A5. CONCLUSION: The amount of NO in the lungs was high in the initial phase of fibroproliferative reaction induced by bleomycin A5 ,and these might be associated with the enhanced ability of AMs to release NO and the increased amount of iNOS.  相似文献

Low seed germination is a major problem in commercial rose propagation and breeding and is species-dependent. The present work selected four rose species previously un-examined to explore effective methods for improving seed germination and the relevant dormancy mechanism and its levels in seven experiments. The results showed that both pulp and achenes from the four rose shrubs had chemical substances that significantly inhibited seed germination with the inhibitory effect was more pronounced in pulp extract than of achenes. Single treatments of H2SO4 scarification, short-term cold stratification (<16 weeks) or warm stratification were less effective in breaking dormancy as indicated by lower germination index than their combinations. Comprehensive comparisons showed that among the six treatments the most effective for breaking dormancy was H2SO4 scarification followed by warm plus cold stratification, then H2SO4 scarification followed by cold stratification and finally warm plus cold stratification. Scarification with H2SO4 for 2–4 h ordinal followed by warm stratification at 20 °C for 4 weeks and cold stratification at 5 °C for 8 weeks was the best pretreatment for increasing seed germination percentage for Rosa multibracteata (81.4 ± 2.9%), Rosa hugonis (13.1 ± 6.0%), and Rosa filipes (62.7 ± 5.7%); and H2SO4 scarification for 4 h followed by cold stratification at 5 °C for 12 weeks was the best pretreatment for Rosa sericea (46.7 ± 8.7%). Our results suggest that these four species have only physiological dormancy caused by integrative roles of pulp, pericarp and embryo. The level of physiological dormancy was ranked as R. hugonis > R. sericea > R. filipes > R. multibracteata.  相似文献

Twelve commercially produced Dutch green waste composts were evaluated for their suitability to replace 20% (v/v) peat substrate in the cultivation of ornamentals. Salt concentrations were determined in water extracts of the composts and disease suppressive effects were assessed against various soilborne diseases. The Cl-concentration of the compost extract appeared to be the limiting factor for use of the composts in potting mixtures. The Cl-concentrations in 7 and 1 composts, respectively, were too high to replace 20% of peat for growing salt sensitive and moderately salt sensitive plants, according to guidelines set for these groups of plants. The suppressive effects of the composts were tested in peat-based potting mixtures using three bioassays: Phytophthora cinnamomi—lupin, Cylindrocladium spathiphylliSpathiphyllum and Rhizoctonia solani AG2-1—cauliflower. Disease levels in compost-amended mixtures were compared with the non-amended controls. None of the composts induced suppressiveness against P. cinnamomi; 3 and 9 composts significantly induced suppressiveness against C. spathiphylli and R. solani, respectively. No significant disease enhancement was observed in any of the bioassays. The pH of the potting mixture showed a negative correlation with suppression of the Rhizoctonia disease (R2 = 0.56). The effect of pH (pH 4–6) on suppression of R. solani and P. cinnamomi was further studied in non-amended peat. Disease suppression of R. solani in cauliflower decreased with increasing pH in two different kinds of peat, while there was no effect on P. cinnamomi. The suppressive effect of 3 composts was assessed in two experiments against Fusarium wilt in Cyclamen persicum (caused by F. oxysporum f. sp. cyclamini) and Begonia eliator (caused by F. foetens) under near-commercial conditions. None of the composts had a significant effect on Fusarium wilt in Cyclamen. Two and 3 composts significantly induced suppressiveness against Fusarium wilt in Begonia in the first and second experiments, respectively. No significant differences were observed in growth characteristics between Begonia plants grown in compost-amended and non-amended potting mixture in both experiments. In the second experiment, Cyclamen plants grown in compost-amended potting mixture had significant lower number of flowers than plants grown in non-amended potting mixture, which may have been due to lower concentrations of N in the compost-amended potting mixtures. In conclusion, most composts of the origin and composition tested can replace 20% peat in potting mixtures for moderately salt sensitive and salt tolerant plants. Amendment of these composts can contribute to control of Fusarium wilt in Begonia plants.  相似文献

AIM:To determine whether vasorelaxation induced by cAMP is dependent on eNOS activation.METHODS:A constant perfusion model of rabbit femoral artery was used to observe the changes of perfusion pressure before and after intra-arterial infusing different concentrations of dbcAMP(a membrane-permeable cAMP analogue) or forskolin(a direct stimulator of adenyl cyclase) and the effects of NOS inhibitor L-NAME on their actions.RESULTS:Both dbcAMP and forskolin induced concentration-dependent relaxation of rabbit femoral artery bed and NOS inhibitor L-NAME partly inhibited this vasorelaxation.CONCLUSION:Vasorelaxation induced by cAMP is mediated partly through eNOS activation.  相似文献

AIM: To observe the role of nitric oxide (NO) in the reactivity of isolated lymphatics to substance P (SP),which presents a biphasic change, in the hemorrhagic shock (HS) rats with the technique of lymphatic perfusion in vitro. METHODS: Male Wistar rats were randomly divided into control group (surgical procedure only) and shock group (the rats were further divided into shock 0.5 h and shock 2 h groups after the HS model was established). A segment of lymphatics was pressed and perfused in vitro at transmural pressure of 3 cmH2O after thoracic ducts were separated from the rats at the corresponding time points in each group. The lymphatics of shock 0.5 h and shock 2 h were incubated with different drugs for changing the activity of No and nitric oxide synthase (NOS), respectively. The end-systolic diameter, end-diastolic diameter, contraction frequency (CF) and passive diameter of isolated lymphatics were measured, while the contraction amplitude (CA), tonic index (TI) and fractional pump flow (FPF) were calculated after stimulated with gradient SP. Different values between pre-and post-administration of SP in CF, CA, TI and FPF were calculated and expressed as ΔCF, ΔTI, ΔCA and ΔFPF for further assessing the reactivity of lymphatics. RESULTS: NO donor L-Arg reduced ΔCF, ΔTI and ΔFPF of 0.5 h-shocked lymphatics treated with different concentrations of SP. The effect of L-Arg was obviously suppressed by a soluble guanylate cyclase inhibitor ODQ. ΔCF, ΔTI and ΔFPF increased strikingly compared with shock 0.5 h+L-Arg group in the presence of SP at certain concentration, and ΔCF and ΔFPF increased remarkably compared with control group. NOS inhibitor L-NAME elevated ΔCF, ΔTI and ΔFPF of 2 h-shocked lymphatics treated with different concentrations of SP and the manifestation of lymphatics exceeded the values of control levels. In the experiment of 2 h-shocked lymphatics treated with L-NAME+phosphodiesterase inhibitor aminophylline (AP), the effect of L-NAME was suppressed significantly, which manifested by the decrease in ΔCF, ΔTI and ΔFPF as compared with the values of shock 2 h+L-NAME group in the presence of SP at the concentrations of 1×10-8 mol/L and 3×10-8 mol/L. CONCLUSION: These data indicate that NO involves in the biphasic modulation of shocked lymphatics and the effect might be involved in the action of cyclic guanosine monophosphate.  相似文献

Asiatic citrus canker caused by Xanthomonas citri subsp. citri (Xcc) is one of the most detrimental diseases affecting citrus production. ‘Meiwa’ kumquat (Fortunella crassifolia) and ‘Newhall’ navel orange (Citrus sinensis) are two genotypes with contrasting Xcc resistance. In the current study, efforts were made to perform pairwise comparison between them in terms of structural difference and biochemical response to Xcc attack. Pinprick inoculation demonstrated that ‘Meiwa’ had lower disease incidence rate and smaller lesion size, concurrent with significantly less bacterial number at the inoculation sites relative to ‘Newhall’. Microscopic observation demonstrated that stomatal density and size of ‘Meiwa’ were significantly smaller than ‘Newhall’. Stomatal apertures of ‘Meiwa’ were smaller than ‘Newhall’ measured at 2 h after their leaves were immersed in Xcc suspension, consistent with the bacteria number in the internal tissues counted at 4 d. Epicuticular wax content of ‘Meiwa’ was significantly higher than that of ‘Newhall’, while no difference in palisade and spongy tissue was observed between them. Enzymatic activity assay showed that after Xcc inoculation, ‘Meiwa’ exhibited significantly higher activities of catalase (CAT), peroxidase (POD) and phenylalanine ammonia-lyase (PAL) in comparison with ‘Newhall’ at most of the sampling time points. Our data showed that the two genotypes with contrasting Xcc resistance exhibited clear difference in the structure and physiological response, which may provide information to facilitate our understanding of Xcc resistance.  相似文献
