Brucellosis is a zoonotic disease worldwide distributed and having the economic as well as public health importance. The prevalence of brucellosis among sheep flock having history of abortions was studied. A total of 229 samples comprising of 157 blood and 72 clinical samples (vaginal swabs) were collected from 157 animals. Clinical samples were processed for the isolation of Brucella melitensis. Serum samples (n = 157) were tested by Rose Bengal plate test (RBPT) and i-ELISA. A total of 68 (43.31%) and 104 (66.24%) samples were positive by RBPT and ELISA, respectively. Brucella isolates (n = 2) were recovered from clinical samples. Both isolates demonstrated amplification for bcsp 31 and IS711 genes. On AMOS PCR, both the isolates amplified at 731 bp, i.e., belongs to B. melitensis species. The incidence of B. melitensis in a migratory flock warns the thorough testing and culling of Brucella-infected sheep from the flock on a continuous basis; otherwise, such incidence will be routine and poor farmers will be at a loss.  相似文献

A cross-sectional study was carried out to determine the status of Brucella infection in one-humped (Dromedary) camels in the North and Central senatorial districts of Katsina State, Nigeria. Nine hundred and eighty serum samples from live and slaughtered camels were tested. Modified Rose Bengal plate test (RBPT) and serum agglutination test (SAT) with ethylenediaminetetraacetic acid, (EDTA) were used as screening and standard tests, respectively. The prevalence of Brucella antibodies were 110 (11.2%) and 103 (10.5%) for RBPT and SAT, respectively. Of the 472 and 508 serum samples tested from the herds and abattoirs, respectively, 63 (13.3%) and 47 (9.3%) were positive by RBPT while 62 (13.1%) and 41 (8.1%) were positive by SAT, respectively. Based on the results, it was concluded that Brucella antibodies were present in camels in the study area. Poor management practices and mixing of camels with other species of livestock as well as unrestricted movement of camels were proposed to be the reasons for the prevalence of the disease in the study area. In view of the public health importance of the disease, it is recommended that there is the need to develop a strategic plan to decrease spread of brucellosis in the study area.  相似文献

In this study, we conducted an investigation to determine the true prevalence of bovine and ovine brucellosis in central-eastern Tunisia. A total of 1134 veterinary samples taken from 130 ruminant herds were screened for brucellosis using IS711-based real-time PCR assay. Sera collected from the ruminants were tested using the Rose Bengal test and indirect enzyme-linked immunosorbent assay. Based on serological and molecular results, the true adjusted animal population level prevalence was 23.5 % in cattle, against 13.5 % in sheep. In addition, the true adjusted herd level prevalence of brucellosis was 55.6 % in cattle and 21.8 % in sheep. A statistically significant association was found between vaginal and milk shedding for ruminants. In addition, our results showed that Brucella abortus could be responsible for bovine and ovine brucellosis. Multivariable logistic regression analysis at the animal population level indicated that age and origin variables were important risk factors for cattle. However, age and abortion variables were found to be associated with ovine brucellosis. At the herd level, risk factors for Brucella positivity were as follows: abortion and herd composition for cattle against herd composition, mortality rates, and hygiene for sheep. Animal hygiene, food quality, and sanitary practices on the farm should be applied as strategies to control brucellosis in herds.  相似文献

A cross-sectional epidemiological study was conducted to determine seroprevalence and risk factors influencing the presence of Brucella antibodies in donkeys of Borno State, north-eastern Nigeria. The study aimed at providing baseline information that may be used in planning a control policy against equine brucellosis. Blood samples were collected from 601 donkeys, comprised of 374 males and 227 females from the six agricultural zones of the state between March 2013 and September 2014. The sera obtained were tested for Brucella antibodies using Rose Bengal plate test (RBPT) and competitive enzyme-linked immunosorbent assay (cELISA). Of the 601 donkeys tested, 43 (7.2%) and 40 (6.7%) were seropositive by RBPT and cELISA, respectively. A seroprevalence of 8.6% was obtained in male and 3.5% in female donkeys. According to age, the highest seroprevalence of 9.6% was obtained from donkeys of age group 4–6 years. With respect to pregnancy status, a higher seroprevalence (6.8%) was obtained from pregnant donkeys compared to 3.8% obtained from the non-pregnant ones. There were statistically significant associations between the presence of antibodies and sex (p < 0.05) and the presence of antibodies and age (p < 0.05) of the studied donkeys. However, no statistically significant association (p > 0.05) was observed between the pregnancy status and presence of antibodies. The study concludes that Brucella infection is present in donkeys in all the agricultural zones of the state. The relatively high seroprevalence (7.2%) obtained is of public health concern because of the close interaction between donkeys, ruminants, and humans in the study area.  相似文献

Background

Bovine brucellosis remains one of the most prevalent zoonotic infections affecting dairy cattle in developing countries where the applied control programs often fail. We analyzed the epidemiologic pattern of bovine brucellosis in a dairy cattle herd that showed several cases of abortions after regular vaccination with RB51 (B. abortus vaccine). In 2013 thirty dairy cows, from a Holstein-Friesian dairy herd with a population of 600 cattle, aborted five months post vaccination by a regular RB51 vaccine. Blood samples were drawn from milking cows and growing heifers, as well as heifers and cows pregnant up to 6 months. These samples were collected in June 2013 (n?=?257) and May 2014 (n?=?263) and were tested by real time (rt)-PCR as well as serological tests, in particular Rose Bengal Test (RBT), Enzyme-Linked Immunosorbent Assays (ELISA) and Fluorescence Polarization Assay. Tissue specimens were also collected from an aborted fetus and cultured. Isolates were subjected to bacteriological typing tests at the genus and species levels.

Results

Five months post vaccination with RB51 vaccine, Brucella (B.) DNA was detected in blood samples of cows by rt-PCR. The serological tests also revealed the spread of Brucella field strains within the herd in 2013. Four Brucella isolates were recovered from specimens collected from the aborted fetus. These isolates were typed as follows: one B. abortus RB51 vaccine strain and three isolates of B. abortus field strain. The seropositive cows with positive rt-PCR might indicate an infection by the Brucella field strain; while the positive rt-PCR results from seronegative animals may either be due to circulating RB51 vaccine DNA in vaccinated animals or to circulating field strain in infected animals before seroconversion.

Conclusion

The results herein suggest that PCR can be a good supplementary tool in an outbreak situation, if an assay is available that can differentiate vaccine and field strains with a high analytical sensitivity. We recommend using RBT and ELISA in parallel in outbreak situations, to identify as many infected animals as possible during the initial screenings. This test procedure should be repeated for at least three successive negative tests, with one month interval.

A cross-sectional study was conducted in three counties (Damxung, Maizhokunggar and Yadong) in Tibet in April and May 2015. A total of 1,523 yaks owned by 181 herders were randomly selected and blood sampled. Sera were tested using the rose bengal test (RBT) and a competitive immune-enzymatic assay (C-ELISA) and the test results interpreted in parallel. The individual yak prevalence was 2.8% (95% CI 2.0–3.7) with a herd prevalence of 18.2% (95% CI 12.9–24.6). At the individual level, two predictor variables, age and production system, were significantly associated with seropositivity by a binary logistic regression analysis. The odds of Brucella infection were significantly higher in older Yaks (3–5 years old, OR = 4.51; 95% CI 1.53–19.29; ≥6 years old, OR = 3.89; 95% CI 1.23–17.21) compared to those of younger yaks (≤2 years old). The odds of seropositivity for yaks managed under an agro-pastoral production system were 2.9 (95% CI 1.48–5.86) times higher compared to those managed under a pastoral production system. At the herd level, an association between the infection with Brucella and a history of abortions in the herd was observed (OR = 4.98, 95% CI 1.48–16.62). Surprisingly, vaccination was not associated with a lower level of infection (p = 0.49 and p = 0.99 for individual and herd level data, respectively). The results of the survey indicate that bovine brucellosis is endemic among the yak population in the plateau region of China, and the risk factors identified in the study should be considered in the epidemiology of the disease and when developing control programs for the disease.  相似文献

A cross-sectional survey was carried out to investigate the seroprevalence of ovine and bovine brucellosis in the livestock–wildlife interface area of Nechisar National Park, Ethiopia. Furthermore, producer’s knowledge about brucellosis and its zoonotic potential was assessed using a structured questionnaire. A total of 268 cattle and 246 goat sera were collected from 50 herds and 46 flocks and subjected to Rose Bengal test (RBT) and enzyme-linked immunosorbent assay (ELISA) in parallel to detect anti-Brucella species antibodies. Positive reactions were further confirmed with compliment fixation test (CFT). Flock and herd level seroprevalence rate was 12.8% (95% CI 4.8–25.7) and 32.0% (95% CI 19.5–46.7) in goats and cattle, respectively. An overall animal-level seroprevalence of 4.5% (95% CI 2.25–7.86) and 9.7% (95% CI 6.44–13.89) was recorded for goats and cattle, respectively. Seroprevalence showed an increasing trend with age, where adult cattle >?2 years. Goats (>?1 year) recorded relatively higher seroprevalence, but the differences were not statistically significant. Similarly, female cattle and goats recorded a relatively higher seroprevalence, 11 and 5.6%, respectively, compared to males but the difference was not significant. However, a significant (P <?0.01) variation of seroprevalence was noted for parity (bovine), higher in animals in second parity, and abortion history, in both species, higher in animals that experienced abortion. Interviews revealed lack of awareness about brucellosis and food safety related to the zoonotic potential from consuming raw animal products (milk and meat). Ninety-eight percent of respondents did not consider handling abortion material is risky, and only a very low proportion (8%, n?=?50) was able to mention limited zoonotic diseases (anthrax and Taenia cysticercosis) could be transmissible to people. The study indicated that brucellosis is endemic in domestic animals in the interface area and calls for further broad epidemiological investigation of the disease in livestock, human and wildlife following ‘one health’ unified research approaches beside enhancing public awareness.  相似文献

The objective of this study was to determine the effects of dietary supplementation of Nigella sativa L. seeds, Rosmarinus officinalis L. leaves and their combination on rumen metabolism, nutrient intake and digestibility, growth performance, immune response and blood metabolites in Dorper lambs. Twenty-four entire male Dorper lambs (18.68?±?0.6 kg, 4–5 months old) were randomly assigned to a concentrate mixture containing on a dry matter basis either, no supplement (control, T1), 1% R. officinalis leaves (T2), 1% N. sativa seeds (T3) or 1% R. officinalis leaves +1% N. sativa seeds (T4). The lambs had ad libitum access to urea-treated rice straw (UTRS) and were raised for 90 days. Supplemented lambs had greater (P?< 0.05) intake of DM and UTRS than the control lambs. The T4 lambs had lower (P?< 0.05) nutrient digestibility than those fed other treatments. Total and daily weight gain was greater (P?< 0.05) in T2 lambs than those fed other diets. The T3 and T4 lambs had greater (P?< 0.05) ruminal pH than the T1 and T2 lambs. Supplemented lambs had lower (P?<?0.05) ruminal total volatile fatty acids, acetate, propionate, NH3-N and C18:0 than the control lambs. The T4 lambs had lower (P?< 0.05) population of Fibrobacter succinogenes, Ruminococcus albus, methanogens and total protozoa compared with those fed other diets. Supplemented lambs had lower (P?< 0.05) neutrophils, basophils and serum urea and greater (P?<?0.05) serum IgA and IgG compared with the control lambs. The current results emphasised the variation in the efficacy of medicinal plants in ruminant nutrition.  相似文献

Hazardous practices regarding antibiotics misuse, unsanitary milking procedures, and the commercial sales of raw milk and unpasteurized dairy products are currently being practiced by livestock farmers in the Sumapaz region (Colombia). The purpose of this study was to screen for food-borne and zoonotic pathogens associated with local livestock practices. We evaluated 1098 cows from 46 livestock farms in the Sumapaz region that were selected by random. Of the total population of cattle, 962 animals (88%) were tested for bovine TB using a caudal-fold tuberculin test and 546 (50%) for brucellosis by a competitive ELISA. In the population tested, 23 cows were positive for Brucella sp. representing a 4.2% seroprevalence and no cases of bovine tuberculosis were found. In addition, food-borne contamination with Escherichia coli and Staphylococcus aureus was assessed together with antibiotic susceptibility for ten different antibiotics in milk samples from 16 livestock farms. We found that 12 of the farms (75%) were contaminated with these food-borne pathogens. Noteworthy, all of the isolated pathogenic strains were resistant to multiple antibiotics, primarily to oxytetracycline and erythromycin. Our findings suggest that livestock products could be a source of exposure to Brucella and multidrug-resistant E. coli and S. aureus strains as a result of unhygienic livestock practices in the Sumapaz region. Training in good farming practices is the key to improving safety in food production.  相似文献

This study aims to evaluate the conjunctiva colour-based FAMACHA score (FS) coupled with a body condition score (BCS), haemogram and stressor hormone level estimation, in identifying post-mortem (PM)/coproscopically proven individuals wanting therapy for economically important gastrointestinal (GI) helminths, Haemonchus contortus, in the small ruminants. The incidence of haemonchosis was significantly (p < 0.05) higher (60.81%) in the ruminants with FS = 3. The H. contortus count in the animals with FS 2, 3 and 4 was 23.2 ± 0.37, 62 ± 2.5 and 74 ± 3.2 (p < 0.05) [positive correlation (r = 0.841 in goats; r = 0.828 in sheep, p < 0.05)], respectively, with corresponding 2.8 ± 0.15, 2 ± 0.3 and 2 ± 0.16 BCS (negative correlation, p > 0.05). The infected animals of FS 2, 3 and 4 measured 8.2 ± 0.0, 7.5 ± 0.23 and 6.7 ± 0.34 g/dl Hb (r = ?0.452, p = 0.01) in goats/9.3 ± 0.8, 8.6 ± 0.5 and 7.6 ± 0.3 g/dl Hb (r = ?0.511, p = 0.05) in sheep with 21.2, 19.8 ± 1.8 and 17.8 ± 0.2% PCV (r = ?0.369, p = 0.05) in goats/26.7 ± 1.2, 22.2 ± 0.2 and 20.9 ± 0.6% PCV (r = ?0.251, p = 0.03) in sheep, respectively. The FS 2, 3 and 4 infected goats/sheep measured 6.1 ± 0, 7.9 ± 1.0 and 9.5 ± 0.9 (p < 0.05)/5.8 ± 2.3, 6.9 ± 1.2 and 7.8 ± 0.2% (p < 0.05) mid-granulocyte [(r = 0.928 (goats)/0.834 (sheep), p < 0.05], while the cortisol level was 15.6, 23 ± 4.5 and 42 ± 2.3 (p = 0.23)/12.1 ± 0, 15.9 ± 1.2 and 24 ± 3.4 (p = 0.29) μg/dl, respectively. The infected ruminants recorded low (p < 0.05) level of Hb/PCV while high level of mid-granulocytes/cortisol. Specificity of FAMACHA test was maximized (100%) when FS = 4 was considered anaemic, but sensitivity was low (35.29% in goats; 25% in sheep). The false negatives was 5.9 (goat)/12.5 (sheep)% when FS ≥ 3 was considered anaemic. The small ruminants with FS ≥ 3, BCS ≤ 2.5, Hb ≤ 7.5 g/dl (goats)/8.6 g/dl (sheep), PCV ≤ 19.8% (goats)/22.2% (sheep) and mid-granulocyte ≥7.9% (goats)/6.9 ± 1.2% (sheep) can be subjected to target-selective treatment for haemonchosis in the field simultaneously maximizing the economic benefit to the farmers.  相似文献

The aim of the current study is to diagnose Brucella spp. infection using methods such as serology, bacterial isolation, and molecular analysis in buffaloes bred in Maranhão State. In order to do so, 390 samples of buffalo serum were subjected to serological tests, to Rose Bengal Plate Test (RBPT) and to 2-mercaptoethanol (2-ME) combined with slow agglutination test (SAT). Vaginal swabs were collected from seropositive animals and subjected to bacterial isolation and to generic PCR. According to the serological test, 16 animals had a positive reaction to the confirmatory test (2-ME/SAT). As for bacterial isolation, three samples resulted in the isolation of Brucella spp.-characteristic colonies, which were confirmed through PCR. These results confirmed Brucella spp. infection in the buffalo herd from Maranhão State.  相似文献

In the present study, we investigated the effect of grape seed and skin supplement (GSSS), on lactating dairy ewes’ production. Ten dairy pregnant ewes from northern Tunisia were allocated to two groups: control diet (C) and supplemented with 20 % (w/w) GSSS. The experiment lasted for 8 weeks and took place after 2 months of lambing. During the experiment, daily milk yield and milk composition were determined. Supplementation of the diet with GSSS increased milk production (P?<?0.001), calcium (P?<?0.01), free iron (P?<?0.01) and urea content (P?<?0.001) but had no effect on milk fat nor protein. From these data, it is concluded that the inclusion of GSSS in sheep diets increased significantly ewes’ milk yield.  相似文献

Twenty nine Pelibuey × Katahdin hair ewes rearing single lamb were used during 42 days of lactation to evaluate the effect of including the Mucuna pruriens whole pods in the diets of lactating ewes on milk production and offspring performance. Animals were distributed at random into three experimental diets: a control diet without Mucuna (M0), and two more diets with the inclusion of 13 (M13) and 26% (M26), of milled pods of Mucuna pruriens, respectively. Dry matter intake, was not significantly different (P?>?0.05) among diets. Milk yield (P?>?0.05) was 734, 786, and 694 g/day for diets M0, M13, and M26, respectively. Milk fat (P?>?0.05), milk protein (P?>?0.05), and milk lactose (P?>?0.05) did not differ between treatments. Lambs had similar daily weight gain (P?>?0.05) among diets (180, 174, and 171 g/day for diets M0, M13, and M26, respectively). Diets with Mucuna whole pods were 5.6 and 12.9% more profitable (M13 and M26, respectively) than control diet. Under the conditions of this work, Mucuna pruriens whole pods can be included up to 26% in the diets of lactating crossbred hair ewes without negatively affecting their productive performance during the first 6 weeks of lactation.  相似文献

The genus Brucella causes significant economic losses due to infertility, abortion, stillbirth or weak calves, and neonatal mortality in livestock. Brucellosis is still a zoonosis of public health importance worldwide. The study was aimed to optimize and evaluate PCR assays used for the diagnosis of Brucella infections. For this aim, several primers and PCR protocols were performed and compared with Brucella cultures and biological material inoculated with Brucella. In PCR assays, genus- or species-specific oligonucleotide primers derived from 16S rRNA sequences (F4/R2, Ba148/928, IS711, BruP6-P7) and OMPs (JPF/JPR, 31ter/sd) of Brucella were used. All primers except for BruP6-P7 detected the DNA from reference Brucella strains and field isolates. In spiked blood, milk, and semen samples, F4-R2 primer-oriented PCR assays detected minimal numbers of Brucella. In spiked serum and fetal stomach content, Ba148/928 primer-oriented PCR assays detected minimal numbers of Brucella. Field samples collected from sheep and cattle were examined by bacteriological methods and optimized PCR assays. Overall, sensitivity of PCR assays was found superior to conventional bacteriological isolation. Brucella DNA was detected in 35.1, 1.1, 24.8, 5.0, and 8.0% of aborted fetus, blood, milk, semen, and serum samples by PCR assays, respectively. In conclusion, PCR assay in optimized conditions was found to be valuable in sensitive and specific detection of Brucella infections of animals.  相似文献

This study determined the effects of feed supplementation during the postpartum period on the weight gain, milk yield, blood profiles and reproductive performance of Sanga and Friesian-Sanga cows grazing on natural pasture. 20 Sanga and 20 Friesian-Sanga cows were randomly allocated either to serve as a control on grazing only or to be supplemented with 2.5 kg of concentrate a day for 10 weeks during the dry season. Each week, all cows were weighed and scored for body condition. Partial milk yield of cows was determined daily. Plasma concentrations of blood metabolites were assessed fortnightly from weeks 1 to 10 postpartum. Resumption of postpartum ovarian activity was determined by measuring progesterone concentration in the plasma from weeks 1 to 10. Supplemented cows had a better body condition score (6.2 versus 5.8; P?<?0.05) and higher partial milk yield (1.94 versus 1.55 L/day; P?<?0.01) than non-supplemented cows. Sanga cows had a better body condition score (6.2 versus 5.8; P?<?0.05) but lower milk yield (1.58 versus 1.92 L/day; P?<?0.01) than the Friesian-Sanga crossbreds. Total protein (P?<?0.05) and albumin (P?<?0.01) concentrations were higher in the supplemented than in the non-supplemented cows. Sanga cows recorded higher globulin (P?<?0.05) and total cholesterol (P?<?0.01) but lower albumin (P?<?0.01) concentrations than Friesian-Sanga crossbred cows. Feed supplementation did not affect (P?<?0.05) the interval from calving to resumption of ovarian activity, and the days to resumption of ovarian activity in the Sanga and Friesian-Sanga cows were also similar (P?>?0.05). The results demonstrate the beneficial effects of feed supplementation in terms of improved body condition and metabolic status and increased milk yield.  相似文献

The objective of the study was to determine growth performance, viscera organ development, economic benefit and sensory characteristics of meat from broilers finished on Acacia anguistissima leaf meal-based diets. One hundred and eighty Cobb 500-day-old chicks were used in the study. Acacia anguistissima was used as a protein source and included in broiler diets at 0, 50 and 100 g/kg. Total feed intake (TFI), average daily gain (ADG), and gain to feed ratio (G:F) and cost benefit analysis were determined. There were no differences in TFI of broilers across treatments diets. Average daily gain and gain to feed (G:F) ratio were low (P?<?0.05) for broilers feeding on diet with100 g/kg acacia meal. Highest returns from weight gain were obtained from broiler chickens fed on diet with 50 g/kg inclusion level of A. anguistissima (P?<?0.05). Intestinal length increased with inclusion level of A. anguistissima whereas liver and spleen weight decreased with inclusion level of A. anguistissima (P?<?0.05). Female consumers gave higher scores for overall flavour intensity than male consumers (P?<?0.05). Treatment diets influenced consumer sensory characteristics of broiler meat (P?<?0.05). High sensory scores were observed for overall flavour intensity (OFI) for both boiled and deep fried meat from broilers fed 100 g/kg A. anguistissima. It can be concluded that A. anguistissima meal can be used effectively up to 50 g/kg in broiler diets without compromising performance of chickens and can influence a few sensory characteristics.  相似文献

The aim of this study was to evaluate the effects of dietary supplementation of Morinda lucida leaf meal (MLLM) on the haematology, biochemical and organ changes of broiler chickens. One hundred and ninety-eight day-old Marshall broiler chicks were completely randomised into 6 treatments in a 3?×?2 factorial arrangement of three levels of M. lucida leaf meal supplementation (0, 0.1 and 0.2 g/kg) with or without medication. The treatment consisted of both negative (without MLLM and routine medication) and positive (containing no MLLM but with routine medication) control groups while each treatment was replicated thrice. MLLM-supplemented diets and routine medication decreased (p?<?0.05) the white blood cell count compared to the negative control. Dietary supplementation with MLLM in combination with normal routine medication increased (p?<?0.05) total serum protein when compared with treatment group without MLLM and routine medication. Dietary supplementation with MLLM and routine medication reduced (p?<?0.05) serum creatinine concentration of the broiler chickens. Birds fed with 0.2 g/kg MLLM supplement coupled with medication and those on negative control had higher (p?<?0.05) creatinine values. Serum enzyme activities reduced (p?<?0.05) following supplementation. MLLM supplementation recorded no significant effect (p?>?0.05) on the liver, kidney, heart and gizzard. M. lucida leaf meal can be compared to routine medication for improved health status of broiler chickens. Dietary inclusion with 0.1 g/kg MLML combined with routine medication could be used in producing healthy and safe chickens.  相似文献

Haemonchus contortus is a parasite of major economic importance of most sheep-rearing areas of the world. A cross-sectional study was conducted from November 2015 to March 2016 with objectives of determining the prevalence and associated risk factors of Haemonchus contortus infection in sheep slaughtered at Jimma town municipal abattoir, Ethiopia. Of the total 7000 sheep slaughtered during the study time period, 384 sheep were randomly selected and examined for parasites. The overall prevalence of Haemonchus contortus infection was 264 (68.75%). Infection rate of poor body condition animals was significantly (P?=?0.000) higher than good body conditioned animals. The mean packed cell volume (PCV) values (%) of parasitemic and aparasitemic sheep were 23.73?±?3.7 SD and 37.66?±?4.736 SD, respectively. The prevalence in adult sheep (>?1 year) was insignificantly (P?=?0.653) higher than that of young sheep (≤?1 year). The highest prevalence was recorded in sheep that originated from Seka district (73.3%) and the lowest in Kersa district (63.4%) with non-significant variation (P?=?0.691). The highest monthly mean worm burdens and prevalence of Haemonchus contortus infection were recorded in November and lowest in March. The current study revealed that Haemonchus contortus infection is an important and common parasitic disease and requires special attention to its control.  相似文献

A total of 1,734 cattle faecal samples from 296 dairy-keeping households were collected from urban settings in Nairobi, Kenya. Modified Ziehl–Neelsen staining method and an immunofluorescence assay were used to identify those samples with Cryptosporidium oocyst infection. Oocysts from positive faecal samples were isolated by Sheather's sucrose flotation method and picked from the concentrate using cover slips. Genomic DNA was extracted from 124 of the faecal samples that were positive for Cryptosporidium and was used as template for nested PCR of the 18S rRNA gene. Twenty-five samples (20 %) were PCR-positive for Cryptosporidium, and 24 of the PCR products were successfully cloned and sequenced. Sequence and phylogenetic analysis identified 17 samples (68 %) as Cryptosporidium parvum-like, four samples (16 %) as Cryptosporidium ryanae, three samples (12 %) as Cryptosporidium andersoni and one sample (4 %) as Cryptosporidium hominis. To the best of our knowledge, this is the first genotyping study to report C. parvum-like, C. andersoni and C. hominis in cattle from Kenya. The results of this study show Cryptosporidium infections in calves and cattle may be potential zoonotic reservoirs of the parasite that infects humans.  相似文献

Objective

To examine the maturational competence, embryo development and expression of genes involved in oocyte maturation and cumulus expansion (GDF9, BMP15, HAS2, TNFAIP6, FGF17 and FSHr) following two standard methods of bovine COCs vitrification.

Methods

Bovine cumulus-oocyte complexes (COCs) were aspirated from slaughtered ovaries and then distributed into three groups: non-vitrified COCs (control), vitrification 1 group (V1); vitrification was performed by 15% ethylene glycol (EG) and 15% DMSO in holding media (TCM-199 with 20% FCS); and vitrification 2 group (V2); vitrification was performed by 40% EG in holding media. After vitrification, COCs were warmed in two steps and cultured and then evaluated for nuclear maturation, embryo development and gene expressions.

Results

The mean (±SD) percentages of nuclear maturation and blastocyst/cleaved were higher in control group (79.5 ± 8.0 and 31.0 ± 5.1%) than the V1 (34.8 ± 9.1 and 4.4 ± 5.1%) and V2 (47.8 ± 11.7 and 7.1 ± 5.8%) groups (P < 0.05), respectively. Further, COCs in V2 group showed higher mean (±SD) percentages of cleavage compared to V1 group (31.8 ± 1.0 vs 21.7 ± 2.8%; P < 0.05). GDF9 and BMP15 expression levels were higher in COCs in the control than of the vitrification groups (P < 0.05). In addition, expression level of GDF9 and BMP15 was higher in V2 group than in V1group (P < 0.05). The expression of HAS2 and FGF17 in V1 group was lower (P < 0.05) than that of the V2 groups.

Conclusions

Expression of oocyte maturation genes was affected by vitrification procedure and conditions. Using EG alone for vitrification of bovine immature COCs, resulted in higher expression of GDF9, BMP15 and production of more in vitro matured and cleaved oocytes.
