The objective of this study was to evaluate the effects of coadministration of ractopamine-HCl (RAC) and trenbolone acetate plus estradiol (TBA) on LM fiber cross-sectional area (CSA), diameter, fiber-associated myonuclei, and satellite cell number. Culled crossbred beef cows (n = 98; 11 +/- 1.8 yr old; BCS 4.3 +/- 0.03) from a single ranch in south Florida were fed a concentrate diet for 92 d in a 2 x 2, randomized block design. Cows were blocked by BW on arrival into light (initial BW = 369.75 +/- 2.68 kg and end BW = 501.96 +/- 6.90 kg) and heavy (initial BW = 418.31 +/- 2.75 kg and end BW = 522.15 +/- 7.09 kg) groups before assignment to treatment. Factors included dietary treatment (0 or 15 ppm) and implant status (0 or 80 mg of trenbolone acetate + 16 mg of estradiol). Ractopamine was provided in the diet to 2 pens or half the treatments during the final 35 d of feeding. Cows were slaughtered on d 92. Forty-eight hours postmortem, the 6th-rib portions of the LM were obtained from 10 randomly selected carcasses from each treatment group (n = 40). Cryosections (12 mum) were immunostained for dystrophin and myosin heavy chain I or II for the measurement of fiber CSA and type, respectively. Fiber-associated nuclei and satellite cell numbers were measured in serial cryosections. There was a RAC x TBA interaction (P < 0.05). Type I fiber CSA and diameter were increased (P < 0.05) by TBA and RAC. Type I CSA and diameter were larger (P < 0.05) in TBA + RAC than RAC only. Type II fiber CSA and diameter were not affected by TBA (P = 0.48), RAC (P = 0.15), or TBA + RAC (P = 0.60). Satellite cell numbers and fiber-associated nuclei were not affected (P > 0.05) by implant status or ractopamine supplementation. These results indicate that TBA and RAC preferentially increase the size of type I fibers in cull cows.  相似文献

Yearling steers (n = 2,552; 314 kg of initial BW) were used to evaluate the effects of ractopamine-HCl (RAC) and days on feed on performance, carcass characteristics, and skeletal muscle gene expression in finishing steers. Treatment groups included serial slaughter dates of 150, 171, or 192 d on feed. Within each slaughter date, steers either received RAC (200 mg/steer) daily for the final 28 d or were not fed RAC. All steers were initially implanted with Revalor-IS and were reimplanted with Revalor-S after 75 d on feed. At slaughter, muscle samples from the semimembranosus were collected for mRNA analysis of the beta-adrenergic receptors (beta-AR). Ractopamine administration increased (P < 0.05) ADG, G:F, and HCW and increased (P = 0.08) LM area. Ractopamine did not affect the dressing percentage, USDA yield grade, or quality grade (P > 0.3). There was no change in overall feed intake across the entire feeding period; however, feed intake was increased during the 28-d period during which the steers were fed RAC (P < or = 0.05). Greater days on feed decreased (P < 0.05) ADG, G:F, DMI, and the number of yield grade 1 and 2 carcasses. Also, greater days on feed increased (P < 0.05) HCW, dressing percentage, and the number of prime and choice carcasses, as well as the number of yield grade 4 and 5 carcasses. Increasing days on feed decreased (P < 0.05) the abundance of beta(1)-AR and beta(3)-AR mRNA and increased (P < 0.05) the abundance of beta(2)-AR mRNA in skeletal muscle samples obtained at slaughter. Ractopamine had no effect (P > 0.10) on the abundance of beta(1)-AR or beta(3)-AR mRNA, but tended (P = 0.09) to increase beta(2)-AR mRNA. Additional time-course studies with primary muscle cell cultures revealed that advancing time in culture increased (P < 0.001) beta(2)-AR mRNA but had no effect (P > 0.10) on beta(1)-AR or beta(3)-AR mRNA. We conclude that days on feed and RAC are affecting beta-AR mRNA levels, which could, in turn, impact the biological response to RAC feeding in yearling steers.  相似文献

Skeletal muscle constitutes the largest organ system in the mammalian body and is essential for movement and force generation. Muscle tissue has the unique ability to adapt and remodel with regular exercise. Adaptation of equine contractile apparatus to exercise training with a different character occurs at the structural to the cellular and molecular levels and depends on age, breed, and sex. In Andalusian and Arabian horses from 3 months to 24 years of age, it was found that the mean cross-sectional area occupied by IIA and IIX fibers was greater in stallions than in mares. In muscle of Dutch Warmblood foals from birth to 1 year of age, a significant number of fibers coexpress either developmental and type IIa or cardiac–alpha and type I myosin heavy chain (MyHC) isoforms. Endurance training results in increased mitochondrial density, capillary supply, changes in key metabolic enzymes, and increased maximal oxygen uptake and promotes a transition from type II to type I muscle fiber. In horses, prolonged aerobic exercise training has been shown to induce a further decline in the percentage of type IIx MyHC isoform expression and an increase of type I and IIa MyHC isoform expression. Short-duration, high-intensity exercise training stimulates type IIA and hybrid (IIA/IIX) fibers. Therefore, intensive high-speed trotting facilitates muscle fiber hypertrophy and increases the oxidative capacity of type IIX fibers.  相似文献

Carcass characteristics, meat quality traits, and sensory attributes were evaluated in late-finishing barrows and gilts, weighing between 100 to 130 kg of BW, fed 0, 5, or 7.4 mg/kg of ractopamine hydrochloride (RAC) for the final 21 to 28 d before slaughter. Carcass data were collected from carcasses from barrows and gilts (n = 168), and all primal cuts from the right sides of these carcasses were fabricated to calculate primal yields as a percentage of the HCW. Subjective (National Pork Producers Council and Japanese) color, firmness, and marbling scores were determined on the LM of each loin and the semimembranosus muscle (SM) of the ham, whereas the moisture, extractable lipid, Warner-Bratzler shear force (WBSF), and trained sensory evaluations (juiciness, tenderness, and pork flavor) were measured on the LM samples only. Gilts produced heavier (P < 0.05) HCW than barrows, whereas feeding RAC increased (P < 0.05) HCW over pigs fed diets devoid of RAC. Carcasses from gilts also had greater (P < 0.02) primal cut and lean cut (P < 0.01) yields than barrows, and dietary inclusion of 5 mg/kg of RAC increased (P < 0.05) total boneless cut and lean cut yields when compared with carcass from pigs fed 0 or 7.4 mg/kg of RAC. Warner-Bratzler shear forces values were greater (P < 0.05) in the LM of gilts than barrows, but only juiciness scores were greater (P < 0.03) in LM chops from barrows than gilts. The LM from barrows had greater intramuscular lipid (P < 0.001) than the LM from gilts, and even though the LM from pigs fed 5 mg/kg of RAC had greater (P < 0.04) WBSF values than the LM from pigs fed 0 or 7.4 mg/kg of RAC, including RAC in the late-finishing diets for 21 or 28 d did not affect sensory panel rating or percentages of moisture and intramuscular lipid. In summary, addition of RAC in the late-finishing diet improved carcass and primal cut yields when it was fed at 5 and 7.4 mg/kg without altering pork quality traits regardless of whether RAC was fed for 21 or 28 d.  相似文献

The objective of this study was to benchmark carcasses and muscles from commercially identified fed (animals that were perceived to have been fed an increased plane of nutrition before slaughter) and nonfed cull beef and dairy cows and A-maturity, USDA Select steers, so that the muscles could be identified from cull cow carcasses that may be used to fill a void of intermediately priced beef steaks. Carcass characteristics were measured at 24 h postmortem for 75 carcasses from 5 populations consisting of cull beef cows commercially identified as fed (B-F, n = 15); cull beef cows commercially identified as nonfed (B-NF, n = 15); cull dairy cows commercially identified as fed (D-F, n = 15); cull dairy cows commercially identified as nonfed (D-NF, n = 15); and A-maturity, USDA Select grade steers (SEL, n = 15). Nine muscles were excised from each carcass [m. infraspinatus, m. triceps brachii (lateral and long heads), m. teres major, m. longissimus dorsi (also termed LM), m. psoas major, m. gluteus medius, m. rectus femoris, and m. tensor fasciae latae] and subjected to Warner-Bratzler shear force testing and objective sensory panel evaluation after 14 d of postmortem aging. Carcass characteristics differed (P < 0.05) among the 5 commercially identified slaughter groups for the traits of lean maturity, bone maturity, muscle score, HCW, fat color, subjective lean color, marbling, ribeye area, 12th-rib fat thickness, and preliminary yield grade. Carcasses from commercially identified, fed cull cows exhibited more (P < 0.01) weight in carcass lean than did commercially identified, nonfed cull cows. There was a group x muscle interaction (P = 0.02) for Warner-Bratzler shear force. Warner-Bratzler shear force and sensory overall tenderness values demonstrates that muscles from the SEL group were the most tender (P < 0.01), whereas muscles from the B-NF group were the least tender (P < 0.01). Sensory, beef flavor intensity was similar (P > 0.20) among cull cow carcass groups and more intense (P < 0.01) than the SEL carcass group. Muscles from the SEL group exhibited less (P < 0.01) detectable off-flavor than the cull cow carcass groups, whereas the B-NF group exhibited the most (P < 0.01) detectable off-flavor. Although carcass and muscle quality from commercially identified, fed, cull beef and dairy cows was not similar to A-maturity, USDA Select beef, they did show improvements when compared with nonfed, cull, beef and dairy cow carcasses and muscles.  相似文献

Two experiments were conducted to investigate the effects of ractopamine-HCl (RAC) and implant strategy or days on feed (DOF) on feedlot performance and expression of beta-adrenergic receptors (AR). In Exp. 1, 1,147 feedlot heifers weighing 282 +/- 3 kg were used with implant treatments of Revalor-200 (R200) at arrival, or Revalor-IH at arrival with reimplantation with Finaplix-H on d 58 (RF). Ractopamine (0 vs. 200 mg/d) was fed the last 28 d in both experiments. Treatments were randomly assigned to 16 pens. At slaughter, semimembranosus muscle tissue was excised for RNA isolation. Ractopamine administration increased (P < 0.05) ADG, G:F, HCW, and LM; decreased (P < 0.05) 12th rib fat depth; and improved (P < 0.05) yield grade. There was no effect (P > 0.10) on the expression of beta1-AR mRNA; however, there was a tendency (P = 0.10) for RAC feeding to increase beta2-AR mRNA levels. For beta3-AR mRNA, there was an implant by RAC interaction (P = 0.05), with RAC numerically increasing beta3-AR mRNA in heifers implanted with RF, but a decrease (P < 0.05) in expression in heifers implanted with R200. Ractopamine also decreased (P < 0.05) IGF-I mRNA in heifers implanted with RF. In Exp. 2, 2,077 heifers were used to investigate the effects of RAC and DOF. Days on feed were 129, 150, and 170, and RAC was administered the last 28 d. Ractopamine improved (P < 0.05) G:F, but had no other effects (P > 0.05) on performance. Average daily gain decreased (P < 0.05) as DOF increased. Hot carcass weight, LM area, 12th rib fat, G:F, calculated yield grade, and marbling score increased (P < 0.05) and the percentage of KPH fat decreased (P < 0.05) as DOF increased. These data aid in our understanding of the effects of steroidal implants, DOF, and RAC administration in feedlot heifers.  相似文献

The objective of this experiment was to determine if growth, carcass composition, and myofiber characteristics of lambs were affected by heterozygosity for a myostatin mutation (g+6723G>A) when offered differing allowances of feed administered with or without ractopamine. Heterozygote [MSTN A/G (n = 40)] and homozygote wildtype [MSTN G/G (n = 39)] castrate male lambs were individually fed ad libitum (HI; 1.8 × estimated ME(m)) or a restricted allowance (LO; 1.1 × estimated ME(m)) of a diet (191 g of CP/kg of DM and 12 MJ of ME/kg of DM), supplemented with (0.4 mg/kg of BW) or without the β-adrenergic agonist ractopamine (RAC or NO RAC) for 47 d. The lambs were scanned by computed tomography at the beginning and completion of the feeding experiment to calculate composition of lean, fat, and bone in the carcass component of the body. The MSTN A/G HI intake lambs had significantly greater total daily carcass growth (P = 0.045) and loin eye depth (P = 0.022) and tended to have a greater daily growth of lean (P = 0.09) in the carcass, compared with MSTN G/G HI intake lambs. Conversely, MSTN A/G LO intake lambs tended to have less daily lean deposition (P = 0.09), significantly less total daily carcass growth (P = 0.045), and had a greater percentage of type IIX myofibers (P < 0.01) and total myofiber area (relative area) of type IIX myofibers (P = 0.013). The inclusion of RAC increased final BW (P = 0.03) and ADG (P = 0.02), percentage of type IIC (P < 0.001) and IIA (P = 0.012) myofibers, cross-sectional area of types I (P = 0.04) and IIAX (P = 0.04) fibers, and the relative area of type IIC (P = 0.003) and IIA (P = 0.01) myofibers in the LM. The experiment demonstrated that including RAC in the diet of lambs increased final BW and ADG, but not HCW, and increased proportion of type IIC and IIA myofibers and cross-sectional area of type I and IIAX myofibers. Our data suggest that RAC and the heterozygous myostatin mutation act together to increase growth of muscle on a high plane of nutrition. The experiment also demonstrated that poor nutritional background of lambs heterozygous for the myostatin mutation may negatively influence their growth rates and myofiber characteristics.  相似文献

The objective of this experiment was to determine the effect of diverse production systems on pig performance, muscle characteristics, and their relation to pork quality measures. Birth and rearing conditions were evaluated using 48 barrows during the fall/winter months. Pigs were farrowed in either indoor crates or outdoor huts. At weaning, indoor- and outdoor-born pigs were allotted randomly to treatments arranged in a 2 x 2 factorial design with two birth (indoor vs. outdoor) and rearing (indoor vs. outdoor) environments. Pigs housed indoors were on concrete-slatted flooring (1.2 m2/pig), and pigs housed outdoors were on an alfalfa pasture (212 m2/pig). Body weight data were collected. Muscle samples were removed within 1 h postmortem from the longissimus (LM) and semimembranosus (SM) muscles. Muscle samples were stained histochemically to identify type I, IIA, and IIB/X muscle fibers. Boneless loins were collected from the left side of each carcass and aged for 14 d. Objective and subjective color measurements were taken on the longissimus muscle at the 10th rib on d 14 postmortem. Loin chops were evaluated for sensory attributes, shear force, and retail display features. Pigs born outdoors were heavier and had a greater ADG at most growth intervals postweaning (d 28, 56, and 112; P < 0.05) than pigs born indoors. Pigs reared outdoors were heavier (P = 0.02) at d 140 (120.1 vs. 112.9 +/- 4.9 kg), and had higher (P = 0.01) ADG (2.2 vs. 1.9 kg/d) and gain:feed ratios (0.41 vs. 0.37) than did pigs reared indoors. Birth x rearing environment interactions were not significant (P > 0.10) for most measures. Carcass and meat quality measures did not differ (P > 0.05) among treatment groups, but loin chops from outdoor born or reared pigs had higher (P < 0.05) a* values than chops from pigs born or reared indoors. The LM muscle of pigs born outdoors had a higher (P < 0.01) percentage of type I, and a lower (P < 0.05) percentage of type IIA fibers than did pigs born indoors. Pigs reared outdoors had a higher (P < 0.01) percentage of IIA fibers and a lower (P < 0.05) percentage of IIB/X fibers than did pigs reared indoors for the LM and SM muscles. Outdoor production systems may influence growth, pork color, and muscle fiber types.  相似文献

OBJECTIVE: To determine the growth-related changes in metabolic and anatomic properties in equine muscle fiber type, including hybrid fibers identified with immunohistochemical analysis. ANIMALS: 24 2-, 6-, 12-, and 24-month-old female Thoroughbreds. PROCEDURE: Samples were obtained from the gluteus medius muscle of all horses. Expression of myosin heavy chain (MHC) isoforms MHC-I, -IIa, -IIb, and -IIx in each muscle fiber was detected by use of 4 primary monoclonal antibodies: BA-D5, SC-71, BF-F3, and BF-35, respectively. Five muscle fiber types (types I, I/IIA, IIA, IIA/IIX, and IIX) were immunohistochemically identified. The area and activity of succinic dehydrogenase (SDH) in each fiber type were determined by use of quantitative histochemical staining and image analysis. RESULTS: Although the proportion of type I and IIX fibers did not change with age, the proportion of type IIA and IIA/IIX fibers significantly increased and decreased, respectively, from 2 months to 24 months of age. The increase in proportion of type IIA fibers with growth may have been attributable to muscle fiber-type transition from type IIA/IIX fibers but not from type IIX fibers. Values for SDH activity and fiber area in hybrid fiber types were intermediate to those for their respective pure phenotypes. CONCLUSIONS AND CLINICAL RELEVANCE: Hybrid fibers have an important role for determining the proportion of muscle fiber type in horses < 24 months old, and the metabolic and anatomic properties of the hybrid fibers are well coordinated, as in mature horses.  相似文献

Previous studies have shown that supplementation of vitamin D3 to cow diets for 4 to 10 d before slaughter lowers Warner-Bratzler shear force (WBSF) values and increases sensory tenderness scores in beef cuts. The present study was conducted to evaluate the effects of vitamin D3 supplementation on muscle calcium concentration, WBSF values, and sensory tenderness ratings of LM and semitendinosus (ST) muscles from cull, predominately Angus, cows (eight cows per treatment). Treatments included 0 (control), 5 million IU, or 7.5 million IU of vitamin D3 supplemented daily for 7 d preslaughter. Twenty-four hours after slaughter, 2.54-cm-thick LM and ST muscle steaks were cut; aged for either 0, 7, 14, or 21 d (ST steaks aged for 7 d only); and frozen at -20 degrees C for WBFS and sensory analysis. Mean values for LM calcium concentration tended to increase (P = 0.14) with vitamin D3 supplementation (154, 176, and 183 microig/g, fresh basis, for 0, 5, and 7.5 million IU/d, respectively). After 7 d of aging, LM steaks from cows fed 7.5 million IU had lower (P < 0.05) WBSF values than 7-d steaks from controls and cows fed 5.0 million IU/d aged 7 d; however, vitamin D3 supplementation had no (P > 0.05) effect on WBSF values of ST steaks aged 7 d. Vitamin D3 supplementation did not (P > 0.05) affect sensory tenderness ratings for either LM or ST steaks at any aging period. Aging, however, had a linear (P < 0.001) effect on tenderness, with an increase in tenderness as aging time increased from 0 to 21 d. Thus, results from the present study indicate that vitamin D3 supplementation, at these levels and duration before slaughter, provided little benefit to muscle tenderness of beef from cull cows.  相似文献

Sixty crossbred cull cows were used to determine the combined effects of a trenbolone acetate-estradiol implant and feeding zilpaterol hydrochloride on performance, carcass characteristics, and subprimal yields of mature cows fed for 70 d. Cows were assigned to 1 of 5 treatments: 1) grazing native grass pasture (G); 2) concentrate-fed (C) a grain sorghum-sorghum silage diet; 3) concentrate-fed and implanted (CI) with Revalor-200 (trenbolone acetate-estradiol); 4) concentrate-fed and fed Zilmax (zilpaterol hydrochloride) beginning on d 38 of the feeding period (CZ); and 5) concentrate-fed, implanted, and fed Zilmax beginning on d 38 (CIZ). The concentrate diet consisted primarily of ground grain sorghum and sorghum silage. During the last 34 d of the feeding trial, concentrate-fed (C, CI, CZ, and CIZ) cows had greater (P < 0.05) gains than G cows. Hot carcass weights and dressing percentages were greater (P < 0.05) for the concentrate-fed cows than for G cows. Longissimus muscle area was largest (P < 0.05) for CIZ cows, whereas subprimal weights from the chuck were heavier (P < 0.05) from CIZ cows than C and G cows, and carcasses from CI and CZ cows had heavier (P < 0.05) chuck subprimal weights than G cows. Rib and round subprimal weights were heavier (P < 0.05) for concentrate-fed cows compared with G cows. In addition, carcasses from CIZ cows had heavier (P < 0.05) total subprimal weights, and total subprimals were a greater percentage of their initial BW than C cows. Rib cut-out and total soft tissue weights from the 9-10-11th rib were less (P < 0.05) for G cows than concentrate-fed cows. Feeding cull cows a concentrate diet increased carcass weight, dressing percentage, and subprimal yields compared with feeding cows a grass-based pasture diet, and the combination of a trenbolone acetate-estradiol implant and feeding zilpaterol hydrochloride can maximize trimmed beef yields from cull cows fed a high-concentrate diet.  相似文献

Effects of ractopamine hydrochloride (RAC) supplementation on growth performance and carcass characteristics of feedlot steers differing in biological type were investigated using British, Continental crossbred, and Brahman crossbred calf-fed steers (n = 420). Steers of each type were weighed at reimplantation [British, mean BW = 375 kg (SD = 38 kg); Continental crossbred, mean BW = 379 kg (SD = 42 kg); Brahman crossbred, mean BW = 340 (SD = 32 kg)] and sorted into 7 BW blocks, each block consisting of 2 pens (10 steers per pen) per type. Pens within a block x type subclass were randomly assigned to RAC treatments (0 or 200 mg x steer(-1) x d(-1) fed during the final 28 d of the finishing period). The type x RAC interaction did not affect (P > 0.05) any of the traits evaluated in this study. Feeding RAC improved (P = 0.001) ADG (1.50 vs. 1.73 +/- 0.09 kg) and G:F (0.145 vs. 0.170 +/- 0.005), but did not affect (P = 0.48) DMI of steers. Dressing percentage, adjusted fat thickness, KPH percentage, and yield grade were not affected by RAC supplementation. Carcasses of steers fed RAC had heavier (P = 0.01) HCW (359 vs. 365 +/- 4.9 kg), larger (P = 0.046) LM areas (81.7 vs. 84.0 +/- 1.1 cm(2)), and tended (P = 0.07) to have lower mean marbling scores (487 vs. 477 +/- 5.2; Slight = 400, Small = 500) than did carcasses of control steers. Among the 3 biological types, Brahman crossbred steers had the lowest DMI and produced the lightest-weight carcasses that had the lowest mean marbling score (P < 0.05). Compared with Continental crossbred and Brahman crossbred steers, British steers produced carcasses with the greatest (P = 0.001) mean marbling scores. Continental crossbred steers had the heaviest BW and greatest dressing percentages and produced the heaviest carcasses with the largest LM areas (P < 0.05) compared with British and Brahman crossbred steers. In the present study, 28 d of supplementation with RAC at a dosage rate of 200 mg x steer(-1) x d(-1) elicited consistent responses in growth performance and carcass traits among 3 diverse biological cattle types.  相似文献

The objectives of this study were to determine the effects of supplementation with a single β-adrenergic agonist (β-AA) or a sequence of β-AA on cow performance, carcass characteristics, and mRNA relative abundance of cull cows implanted and fed a concentrate diet. Sixty cull cows were implanted with Revalor-200 (200 mg of trenbolone acetate and 20 mg of estradiol) and assigned to 1 of 4 treatments (n = 15/treatment): CON = fed a concentrate diet only; RH = supplemented with ractopamine-HCl for the last 25 d before slaughter; ZH = supplemented with zilpaterol-HCl for 20 d before a 3-d withdrawal before slaughter; RH + ZH = supplemented with RH for 25 d, followed by ZH for 20 d before a 3-d withdrawal before slaughter. Ractopamine-HCl was supplemented at a dose of 200 mg·animal(-1)·d(-1), and ZH was supplemented at 8.33 mg/kg (100% DM basis) of feed. All cows were fed a concentrate diet for 74 d. Each treatment had 5 cows per pen and 3 replicate pens. Body weights were collected on d 1, 24, 51, and 72. Muscle biopsies from the LM were collected on d 24, 51, and at slaughter from a subsample of 3 cows per pen. Carcass traits were evaluated postslaughter. The 2 ZH treatments averaged 15.3 kg more BW gain, 0.20 kg greater ADG, and 7.8 cm(2) larger LM area than CON and RH treatments, and 21 kg more HCW than CON, but these differences were not significant (P > 0.10), likely due to a sample size of n = 15/treatment. The sequence of RH followed by ZH tended to optimize the combination of HCW, LM area, percent intramuscular fat, and lean color and maturity compared with the ZH treatment. Abundance of β(2)-adrenergic receptor (AR) mRNA was not altered in the RH + ZH treatment during RH supplementation from d 24 to 51 of feeding. However, the abundance of β(2)-AR mRNA increased (P < 0.05) the last 23 d of feeding for the RH treatment and tended (P = 0.10) to increase in ZH cows during ZH supplementation. For all cows, abundance of type IIa myosin heavy chain (MHC-IIa) mRNA decreased (P < 0.05) after 24 d of feeding. Abundance of MHC-IIx mRNA increased (P < 0.05) for ZH and RH + ZH treatments the last 23 d of feeding during ZH supplementation. Although few significant differences were observed in performance or carcass traits, mRNA quantification indicated that β-AA supplementation elicited a cellular response in cull cows. Implanting and feeding cull cows for 74 d, regardless of β-AA supplementation, added economic value by transitioning cows from a cull cow to what is referred to in industry as a white cow market in which cows have white fat resulting from grain feeding.  相似文献

The aim of the present study was to investigate the effect of oral supplementation of creatine on the muscular responses to aerobic training. Twelve purebred Arabian horses were submitted to aerobic training for 90 d, with and without creatine supplementation, and evaluated with respect to BW and BCS and to the area and frequency of the different types of muscle fibers in the gluteus medius. Supplementation consisted of the daily administration of 75 g of creatine monohydrate mixed into the ration for the 90 d of training. Physical conditioning was conducted on a high-performance treadmill, and training intensity was stipulated by calculating the velocity at which blood lactate reaches 4 mmol/L, determined monthly for each animal. The individual intensity of physical force at 80% of aerobic threshold was established. Morphometry of gluteus medius muscle fibers was performed on frozen sections processed for histochemical analysis of myosin adenosine triphosphatase and immunohistochemistry of slow-contracting myosin. The results demonstrated that the animals maintained a moderate BCS without alteration of BW during the course of training, providing evidence of equilibrium between food intake and caloric expenditure during the study period. The present study demonstrated that aerobic training for 90 d caused hypertrophy of fiber types I (P = 0.04), IIA (P = 0.04), and IIX (P = 0.01), as well as an increase in the relative area occupied by type I fibers (P = 0.02) at the expense of type IIX fibers (P = 0.03), resulting in modifications of the contractile and metabolic characteristics of the gluteus medius muscle. Residual feed intake (RFI) is defined as the difference between the observed feed intake and that expected based on requirements for maintenance and production. A divergent selection was conducted during 4 generations in Large White male pigs to produce low and high RFI lines. The present study aims at determining the influence of this selection on biochemical and histological traits of skeletal muscle, and relating these changes to correlated effects on growth, carcass composition, and meat quality traits. At 8 d preslaughter, biopsies from the LM were taken in the fed state on 14 females from each RFI line fed ad libitum. Animals were slaughtered at 107.8 ± 8.0 kg of BW without any previous fasting. Samples of LM, semimembranosus (SM), biceps femoris (BFM), and rhomboideus muscles were taken at both 30 min and 24 h postmortem. Myofiber typing was only assessed in LM. Low RFI pigs ("efficient") had leaner carcasses with greater muscle content (P < 0.001), less backfat thickness (P < 0.001), and less intramuscular fat content in all 4 muscles (P < 0.01 to P = 0.04). Their greater muscle content was associated with hypertrophy of all fast-twitch fibers. Glycogen content in all glycolytic muscles (i.e., LM, SM and BFM), was greater in low than high RFI pigs. The greater accumulation of glycogen in LM of low RFI pigs was specifically located in the fast-twitch glycolytic IIBW fibers, which correspond to fibers containing IIb, IIb + IIx, or IIx myosin heavy chains. The difference in muscle glycogen content between RFI line pigs was more significant in the living animals (P = 0.0003) than at 30 min postmortem (P = 0.08). This was associated with a decreased ultimate pH (P = 0.001), and greater lightness of color (P = 0.002) and drip loss (P = 0.04) in LM of low than high RFI line pigs, suggesting that selection for reduced RFI may impair some meat quality traits, such as water-holding capacity. Pigs from the low RFI line exhibited a greater (P = 0.02) percentage of IIBW fibers in LM and tended (P < 0.10) to have less lipid β-oxidative capacity in LM, SM, and BFM. In contrast, no difference (P > 0.10) between lines was found for citrate synthase and lactate dehydrogenase activities, mitochondrial activity, and expression of genes coding for uncoupling proteins 2 and 3. Differences between RFI pigs in plasma leptin, cortisol, and thyroid hormone concentrations are presented and discussed. In conclusion, selection for low RFI influenced muscle properties in a way favoring muscle mass, but likely impairing meat quality.  相似文献

Crossbred pigs (n = 216) were used to test the interactive effect, if any, of ractopamine (RAC) and dietary fat source on the performance of finishing pigs, pork carcass characteristics, and quality of LM chops during 5 d of simulated retail display (2.6 degrees C and 1,600 lx warm-white fluorescent lighting). Pigs were blocked by BW and allotted randomly to pens (6 pigs/pen), and, after receiving a common diet devoid of RAC for 2 wk, pens within blocks were assigned randomly to 1 of 4 diets in a 2 x 2 factorial arrangement, with 5% fat [beef tallow (BT) vs. soybean oil (SBO)] and RAC (0 vs. 10 mg/kg). Diets were formulated to contain 3.1 g of lysine/Mcal of ME and 3.48 Mcal/kg of ME. Across the entire 35-d trial, pigs fed RAC had greater (P < 0.01) ADG and G:F, but RAC did not affect (P = 0.09) ADFI; however, performance was not affected (P >or= 0.07) by dietary fat source. Carcass weight, LM depth, and lean muscle yield were increased (P < 0.01), whereas fat depth was decreased (P = 0.01), in carcasses from RAC-fed pigs; however, carcass composition measures were similar (P >or= 0.27) between fat sources.
