Diverse procedures have been reported for the separation and analysis by HPLC of the two major glycoalkaloids present in potatoes, alpha-chaconine and alpha-solanine. To further improve the usefulness of the HPLC method, studies were carried out on the influence of several salient parameters on the analysis of the two potato glycoalkaloids. Effects on retention (elution, separation) times of the (a) composition and pH of the mobile phase (acetonitrile and phosphate buffer), (b) concentration of the phosphate buffer, (c) capacity values of column packing of four commercial HPLC amino columns, (d) column temperature were studied. Except for pH, all of the variables significantly influenced the retention times. The results make it possible to select analysis conditions that produce well-separated as well as symmetrical peaks of the two glycoalkaloids. This improved HPLC method (limit of detection of approximately 150 ng) was evaluated with extracts from the cortex of one whole potato variety (May Queen) grown in Japan and the freeze-dried peel and flesh from the following eight cultivars grown in the United States: Atlantic, Dark Red Norland, Ranger Russet, Red Lasoda, Russet Burbank, Russet Norkota, Shepody, and Snowden. In addition, the same samples were analyzed by GC-MS for the presence of two water-soluble nortropane alkaloids, calystegine A(3) and calystegine B(2), reported to be potent glycosidase inhibitors. The following ranges for the eight varieties of total glycoalkaloid and calystegine levels were observed: dry flesh, 5-592 and 6-316 mg/kg; dry peel, 84-2226 and 218-2581 mg/kg; dry whole potatoes, 40-883 and 34-326 mg/kg; wet flesh, 1-148 and 1-68 mg/kg; wet peel, 12-429 and 35-467 mg/kg; wet whole potatoes, 7-187 and 5-68 mg/kg. The possible significance of the results to plant and food sciences is discussed.  相似文献

Polyphenol oxidase (PPO) activity of Russet Burbank potato was inhibited by sense and antisense PPO RNAs expressed from a tomato PPO cDNA under the control of the 35S promoter from the cauliflower mosaic virus. Transgenic Russet Burbank potato plants from 37 different lines were grown in the field. PPO activity and the level of enzymatic browning were measured in the tubers harvested from the field. Of the tubers from 28 transgenic lines that were sampled, tubers from 5 lines exhibited reduced browning. The level of PPO activity correlated with the reduction in enzymatic browning in these lines. These results indicate that expression of tomato PPO RNA in sense or antisense orientation inhibits PPO activity and enzymatic browning in the major commercial potato cultivar. Expression of tomato PPO RNA in sense orientation led to the greatest decrease in PPO activity and enzymatic browning, possibly due to cosuppression. These results suggest that expression of closely related heterologous genes can be used to prevent enzymatic browning in a wide variety of food crops without the application of various food additives.  相似文献

Biofortification of staple crops like potato via breeding is an attractive strategy to reduce human micronutrient deficiencies. A prerequisite is metabolic phenotyping of genetically diverse material which can potentially be used as parents in breeding programs. Thus, the natural genetic diversity of thiamin and folate contents was investigated in indigenous cultivated potatoes (Solanum tuberosum group Andigenum) and wild potato species (Solanum section Petota). Significant differences were found among clones and species. For about 50% of the clones there were variations in thiamin and folate contents between years. Genotypes which contained over 2-fold the thiamin and 4-fold the folate content compared to the modern variety Russet Burbank were identified and should be useful material to integrate in breeding programs which aim to enhance the nutritional value of potato. Primitive cultivars and wild species with widely different amounts of thiamin and folate will also be valuable tools to explore their respective metabolic regulation.  相似文献

Manganese (Mn)- and zinc (Zn)-driven antagonistic interactions with high available phosphorus (P) can result in negative impacts on potato cropping systems. Two chelator-buffered hydroponic experiments were conducted with Russet Burbank potato to elucidate P and Mn relationships and associated interactions with Zn. In both experiments, a P concentration decline in new shoots, old shoots, and roots resulted as solution Mn changed from deficient to sufficient followed by a P concentration rise as solution Mn changed to excessive concentrations. New and old shoot Zn concentrations generally increased with augmented solution Mn in the variable Mn experiment, but no significant changes were found in root Zn contents. Available Mn was observed to control plant P concentrations and to influence Zn uptake and translocation; thus, Mn has considerable impact on uptake and distribution of P and Zn and on P-Zn interactions in potato.  相似文献

Three hydroponic experiments were conducted with Russet Burbank potato to elucidate zinc (Zn) and manganese (Mn) relationships and associated interactions with other nutrients at different levels of phosphorus (P). Except when P was optimal, root Mn concentration was reduced at optimal solution Zn relative to deficient or excessive Zn levels. Shoot Mn generally increased with augmented solution Zn. As solution Zn increased, root P declined while shoot P and copper (Cu) and root iron (Fe) and Cu increased. Increasing solution Mn generally decreased Zn in all plant tissues—especially in roots. The Mn influence on plant P, Cu, and Fe concentrations was nearly opposite the Zn impact, indicating that Zn and Mn have counter effects. Partitioning between root and shoot micronutrient concentrations occurred when available P was either deficient or excessive, which may also help explain poor plant health and adverse micronutrient relationships in potato when available P is not optimally maintained.  相似文献

Phosphorus (P) fertilizer is routinely applied to potato (Solanum tuberosum L.) in Manitoba, Canada; however, little information is available regarding P responses in this region. Field studies were conducted from 2003 through 2006 to determine the impact of P rate [0, 15, 29, 43 kg P ha?1 as broadcast incorporated monoammonium phosphate (MAP)] on Russet Burbank potato. Increasing P rate resulted in a linear increase in marketable yield in 2005, a quadratic increase in 2006, and no effect in 2003 and 2004. Phosphorus application had no effect on the yield of tubers <85 g, tuber number, or quality, but petiole P concentration and postharvest soil-test P levels increased with increasing P rate. Results of this and other studies show that P fertilization may improve potato yields, although increases are not completely predictable. Additional research is required to refine soil-test guidelines and petiole P sufficiency levels for this region.  相似文献

A chemical profiling technique for sub-lethal acetolactate synthase (ALS)-inhibitor herbicides (e.g., sulfonylureas, imidazolines, triazolopyrimidine sulfonanilides, and pyrimidyloxy salicylic) was developed using 2-aminobutyric acid, and was found to be directly proportional to application rates in field studies on two varieties of potato plants. An uncomplicated, benign-by-design analytical method for the determination of 2-aminobutyric acid in plant tissue was developed. The method is simple, fast, and automated, entailing a water-trichloroacetic acid extraction followed by precolumn on-line derivatization using o-phthalaldehyde (OPA) solution and liquid chromatographic analyses. Use of reagents and chlorinated organic solvents, and generation of waste, are minimized as compared to other ALS-inhibitor herbicide analytical techniques. Recoveries for a series of fortified plant tissues ranged from 82 to 103%. Two 20-day field trials on two potato varieties, Russet Burbank and Shepody, were conducted during the 2000 and 2001 growing seasons. The study demonstrated that the 2-aminobutyric acid method is an excellent, selective chemical marker technique for ALS-inhibitor herbicides for real world plant matrixes.  相似文献

Yield‐response correlations with old and improved soil extraction methods for boron (B) are needed. Russet Burbank potato (Solanum tuberosum L.) was grown with two, four, and six B treatments applied in 2004, 2005, and 2006, respectively. Zero and 1.1, 2.2, or 3.4 kg B ha?1 soil and 0.22 or 0.28 kg B ha?1 foliar treatments were applied. Boron fertilization did not significantly increase tuber yield or quality despite initially low hot‐water‐extractable B (0.34–0.50 mg kg?1), although postseason B for unfertilized treatments increased (0.51–0.57 mg kg?1). Soil‐applied B generally reflected B application relative to the untreated control and the low foliar rates in all three years for the four soil extractions utilized [hot water, pressurized hot water, diethylenetriaminepentaacetic acid (DTPA)–sorbitol, and Mehlich III]. Boron content of potato petiole did reflect application of B in 2 years, but tuber and peel tissues did not consistently reflect application of B.  相似文献

Abstract

Yield response of Idaho Russet Burbank potatoes to nitrogen fertilizer was related to soil test for inorganic N (NO3 and NH4 +) in a total of 27 field experiments over a 3‐year period using polynomial correlation and regression analysis. Nitrate plus ammonium nitrogen content in the surface foot of soil was found to be useful in predicting yield response of potatoes to applied nitrogen.

Correlations between yield and extractable N were considerably better when the data from each cropping system were analyzed separately than when all locations were analyzed as one group. Additional improvement was obtained by including extractable ammonium nitrogen and nitrogen in the second foot of soil respectively. The best correlation with yield was found using (NO3 + NH4 +)‐N in the surface foot following grain and the top two feet following non‐grain crops with R2 values of 0.875 and 0.821 respectively.  相似文献

Soils under intensive potato (Solanum tuberosum L.) production are often low in organic matter content and microbial activity. Pulp fibre residue addition may restore the quality of these soils. An experiment was initiated in which raw or composted pulp fibre waste was added to a Fredericton sandy loam (Orthic Humo-Ferric Podzol) in New Brunswick (Canada). Each material was applied at 45 Mg (dry weight) ha-1 and 90 Mg (dry weight) ha-1 before planting. Soil was under continuous potato (cv. Russet Burbank) for 3 years. The raw and the composted pulp greatly increased soil organic matter (SOM) content, the C/N ratio, macroaggregation, C mineralization, microbial biomass C (MBC) and enzyme activities for the whole duration of the experiment but had no effect on its total N. Most of the SOM increase was found in the macroorganic matter fraction. The SOM, C/N ratio, macroaggregation, C mineralization and MBC decreased from the time of application. No major difference was found between materials except for CO2-C release in the incubation which was higher for the composted than for the raw pulp. This study indicated that pulp fibre, either applied raw or composted, increased SOM and macroaggregation and promoted microbial growth and activity in this potato soil which was low in C content. The beneficial effects on soil physical and biochemical properties were still present after 3 years of continuous cropping.  相似文献

Understanding how the environment and production and cultivation practices influence the composition and quality of food crops is fundamental to the production of high-quality nutritious foods. In this 3-year study, total phenolics, percent soluble solids, ascorbic acid, and the flavonoid aglycones quercetin, kaempferol, and luteolin were measured in two varieties of tomato (Lycopersicon esculentum L. cv. Ropreco and Burbank) and two varieties of bell peppers (Capsicum annuum L. cv. California Wonder and Excalibur) grown by certified organic and conventional practices in a model system. Significantly higher levels of percent soluble solids (17%), quercetin (30%), kaempferol (17%), and ascorbic acid (26%) were found in Burbank tomatoes (fresh weight basis; FWB), whereas only levels of percent soluble solids (10%) and kaempferol (20%) were significantly higher in organic Ropreco tomatoes (FWB). Year-to-year variability was significant, and high values from 2003 influenced the 3-year average value of quercetin reported for organic Burbank tomatoes. Burbank tomatoes generally had higher levels of quercetin, kaempferol, total phenolics, and ascorbic acid as compared to Ropreco tomatoes. Bell peppers were influenced less by environment and did not display cropping system differences.  相似文献

This paper explores the regulation of water quality protection downstream from the Ranger Uranium Project in the Alligator Rivers Region, an area of high conservation value which is both World Heritage- and Ramsar-listed. Available historical monitoring data for surface water quality in Magela Creek downstream of Ranger have been compiled and analysed with respect to hydrologic data and the Australian and New Zealand Environment and Conservation Council?CAgricultural and Resource Management Council of Australia and New Zealand (ANZECC?CARMCANZ) regulatory guidelines, introduced in late 2000. The paper focuses on the underlying scientific basis for the current approach and examines the complex inter-relationships of minesite water management, hydrology, climate, monitoring design, implementation and interpretation which are used to differentiate between natural variability and potential mine-derived solutes. The research found that the application of the ANZECC?CARMCANZ guidelines has clearly improved the regulation of water quality protection downstream from the Ranger Uranium Project. The scientific basis is more coherent than the previous regulatory regime; however, for U (a key parameter of indigenous Mirarr-Gundjeihmi and public concern), higher downstream concentrations are permitted than those observed through natural variability, leaving open the potential for an influence of mine-derived U loads while still being within regulatory limits. Another improvement that could be made to the current regulatory regime, to provide enhanced protection of the water quality in Magela Creek downstream of Ranger, would be to explicitly link the water quality monitoring regime with hydrologic flow conditions. The paper makes a valuable case study for the application of water quality guidelines, especially for controversial projects such as uranium mining surrounded by a World Heritage- and Ramsar-listed region on indigenous land??a context of clear relevance for many places around the world.  相似文献

New clonal selections with increased vine vigor and stress resistance have been identified for the potato cultivar ‘Russet Norkotah’. However, the importance of clonal variation in nitrogen (N) uptake and root morphological properties is not well known. The objective of this study was to determine the effect of N fertilization on dry matter and N accumulation and root morphological parameters of two clonal selections of ‘Russet Norkotah’. A field experiment was conducted in 2002 using the standard ‘Russet Norkotah’ clone (SRC) and Texas selection 112 (TX112) of ‘Russet Norkotah’, grown at 0 and 150 kg N ha? 1. Whole plants were excavated at 54, 76, and 96 days after planting; partitioned into tubers, vines, roots, stolons, and fruits; and their dry matter and N accumulation were determined. Soil cores were obtained from 10 spatial locations relative to the plant, and used for determination of root length (RL), root length density (RLD), root average diameter (RAD), and root dry weight (RDW). Soil inorganic N content was also measured. Nitrogen fertilization increased tuber yield and dry matter and N accumulation. Fertilizer N application did not affect RL, RLD, or RDW, but resulted in a larger proportion of roots close to the top of the potato hill. Tuber yield and dry matter and N accumulation were similar for the two clonal selections. The TX112 clone, however, partitioned more dry matter and N to vines and less dry matter and N to tubers compared with the SRC clone. Soil nitrate concentration was significantly higher for SCR than for the TX112 clone in the fertilized treatment at 54 DAP, and was low and similar between clones thereafter. Root length and RLD were significantly higher for the TX112 clone compared with SRC, and both clones had a similar spatial distribution of roots. Under the conditions of this study where moisture and disease stress were limited and under a short growing season, the larger root system and increased vine vigor of the TX112 clone did not provide any advantage in terms of plant production as either dry-matter accumulation or tuber yield.  相似文献

Foliar concentrations of starch and major elements, nitrogen (N), phosphorus (P), potassium (K), calcium (Ca), and magnesium (Mg), along with specific leaf weight (SLW) were determined in the potato (Solanum tuberosum L.) cvs ‘Denali’, ‘Norland’, and ‘Russet Burbank’ grown for 35 days under CO2 concentrations of 500, 1,000, 1,500 and 2,000 μmol.mol‐1 at both 16°C and 20°C air temperature. The starch concentration, pooled from the three cultivars, increased with increasing CO2 concentration at both 16°C and 20°C and was consistently higher at 16°C than at 20°C. The SLW (g.m‐2) was positively related to the foliar starch concentration on the basis of leaf area or dry weight. The concentrations of N, P, Ca, and Mg in leaves were negatively related to starch concentration under =14% starch on a dry weight basis. Above 14% starch, there was no significant relationship between element and starch concentrations. Similar patterns were seen when the SLW and element concentrations were expressed on a starch‐free basis. In contrast, the leaf concentration of K was not closely related to the starch concentration because the K concentration was similar at varied CO2 levels. The results of this study indicate that the changes in SLW and concentrations of N, P, Ca, and Mg in potato leaves only partially resulted from the changed starch concentration.  相似文献

Chlorophyll and glycoalkaloid synthesis in potato (Solanum tuberosum L.) tubers occur in direct response to light. The two processes are concurrent, but independent. Color photographic indices to subjectively grade fresh market potatoes for the extent of greening were developed under lighting conditions consistent with those of retail markets. Total glycoalkaloid (TGA) and chlorophyll accumulation for four cultivars were determined over the respective greening scales, thus calibrating the scales for TGA content. On average, TGA concentrations in complete longitudinal sections of tubers (flesh samples) were highest in Dark Red Norland followed by Russet Norkotah, Yukon Gold, and White Rose. TGA concentrations of flesh samples of White Rose and Yukon Gold tubers were somewhat variable and did not increase in direct proportion to greening level and chlorophyll content, particularly at higher levels of greening. TGA concentrations in Dark Red Norland and Russet Norkotah tubers were highly correlated (P < or = 0.001) with greening level and chlorophyll concentrations. When averaged over greening levels, skin samples contained 3.4- to 6.8-fold higher concentrations of TGAs than flesh samples, depending on the cultivar. The TGA concentration in periderm samples ranged from 37 to 160 mg/100 g of dry wt. Regardless of greening level, concentrations of TGAs in the flesh samples (including attached periderm) remained within limits presumed safe for human consumption. Discrimination of greened tubers on the basis of perceived glycoalkaloid toxicity is likely unfounded for the cultivars and greening levels studied.  相似文献

Abstract

Development of nitrogen (N)‐use‐efficient genotypes could reduce N fertilization and nitrate leaching; however, screening in field trials can be costly and time consuming. This study evaluated a rapid and economical methodology for screening of potato (Solanum tuberosum L.) cultivars for N‐use efficiency. A hydroponic experiment was conducted with two solution NO3 ? concentrations (0.05 and 1.0 mM) and five potato cultivars (Atlantic, Chieftain, Red Pontiac, Russet Norkotah, and Shepody). Plants were harvested 30 days after transplanting. Root dry weight in combination with plant N accumulation was considered as a good indicator of N uptake efficiency in low solution NO3 ? concentration. Plant total dry weight (TDW) and plant N accumulation (PN) were highly correlated; therefore, TDW can be considered as an alternative criterion for PN when screening for N‐uptake efficiency. Chieftain had high N‐uptake efficiency and high N‐utilization efficiency whereas Russet Norkotah had low N‐uptake efficiency.  相似文献

Polyphenol profiles of French cider apple varieties (Malus domestica sp.)   总被引:6,自引:0,他引:6  
The cortex of 14 French apple varieties (12 cider and 2 juice varieties), one English cider variety, and one dessert apple (i.e., Golden Delicious) were studied for their polyphenol composition. Total polyphenols were assayed by the Folin-Ciocalteu method, and the precise polyphenolic composition (monomeric catechins, proanthocyanidins, hydroxycinnamic acids, and dihydrochalcones) was obtained by HPLC following thiolysis. ESI-MS and ESI-MS/MS analyses showed that chlorogenic acid and p-coumaroylquinic acid were methylated under the conditions of thiolysis. Depending on the variety, the global polyphenol concentration varied from 1 to 7 g per kilogram of fresh cortex. Cider varieties globally showed a higher polyphenol concentration than the dessert apple Golden Delicious, bitter varieties being the more concentrated. The proportion of the polyphenol classes varied greatly from one cultivar to another. For all varieties, procyanidins were always the predominant class. They were mainly constituted of (-)-epicatechin units with a small proportion of (+)-catechin as a terminal unit. The average degree of polymerization ranged between 4.2 and 7.5 depending upon the variety with an exception for the sharp varieties Guillevic and Avrolles which showed significant concentrations of procyanidins with DPn of 40 and 50, respectively.  相似文献

Fifty per cent of European peatlands are in a damaged state. While intact peatlands are natural carbon sinks, degraded sites release important amounts of greenhouse gases into the atmosphere, contributing to global warming. Restoration of the hydrological functionality of peatlands has proved to be an efficient tool to avoid these emissions. In France, Tuffnell & Bignon's ministerial report (2019) emphasized the need for peatlands ‘integration into the National Low Carbon Strategy, targeting carbon neutrality by 2050. However, current knowledge regarding French peatlands’ distribution and carbon stocks is insufficient and does not allow decision makers and managers to prioritize areas for restoration. The most complete database to date is the 1949 Atlas, an inventory of exploitable peat deposits that was conducted during WWII for peat exploitation as fuel. Until its digitalization, the latter database was archived and never used in a scientific study. It provides detailed information about peatland surfaces, peat thicknesses and carbon contents at that time. We estimated peat carbon stocks from French peatlands to be 111 Mt C in 1949 for 63,290 ha identified as peaty sites, the equivalent of 3% of the organic carbon contained in the upper 30 centimetres of French soils. 34% of this stock was held in Lower Normandy (37.7 Mt C) and 12% in the Picardy's region (13.0 Mt C), in large lowland peatlands. However, not all peatlands were prospected in the 1949 inventory and the characteristics of the prospected peatlands may have changed with anthropic disturbances of the last decades, such as draining or climate change. These first results highlight the need for a recent inventory of French peatlands and carbon stocks based on local data aggregation. Data from the 1949 Atlas could help constituting this new inventory but should be validated before being used to describe the present.  相似文献
