四季豆是多种盲蝽的寄主植物,该研究旨在发展一个利用四季豆豆荚规模化饲养多种盲蝽的技术.研究结果表明:绿盲蝽与中黑盲蝽的若虫最佳饲养密度约为100头/盒(2L),成虫为60~80头/盒.绿盲蝽成虫喜好在切头豆荚的两端伤口处产卵,而中黑盲蝽喜欢在完整豆荚的两端产卵.养虫盒内添加一些弯曲的纸条有助于提高若虫存活率和成虫产卵量.产卵后的豆荚需晾干后放置在25~28℃、相对湿度60%~70%、光照16:8(L:D)的环境下,以减少豆荚腐烂和提高卵的孵化率.利用这一方法饲养盲蝽的若虫存活率、成虫产卵量和卵孵化率分别达60%、40粒和80%.  相似文献

Cucumber mosaic virus (CMV) has resulted in much damage to Danshen (Salvia miltiorrhiza Bunge) crops in China. A 5-year survey was conducted in Hebei, Shandong, Sichuan, Shanxi, Henan and Gansu Provinces, all major Danshen-growing areas. A total of 156 Danshen plant samples with CMV symptoms were collected and tested for the presence of CMV by a double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) using polyclonal antibodies against CMV and a positive control, according to the supplier’s instructions (Agdia). They were confirmed to be CMV by amplification of complete coat protein gene and analysis of the gene sequence. The effects of four planting patterns of bean (Phaseolus vulgaris) (bean only, maize–bean (MB), sorghum–bean (SB), and maize–bean–sorghum (MBS)) and four cropping systems (sole cropping, row, mixed, and broadcast intercropping) on the temporal epidemics of bean common bacterial blight (CBB) caused by Xanthomonas campestris pv. phaseoli were studied. The experiments were conducted during two consecutive spring and summer seasons in 1999 and 2000 in replicated field experiments. The Gompertz model described disease progress curves better than the logistic model. Intercropping delayed epidemic onset, lowered disease incidence and severity, and reduced the disease progress rate. The type of cropping system and planting pattern affected CBB incidence and severity at initial, final and overall assessments and also affected the rate of disease development. Statistical significance of treatment interactions based on disease assessments was found for incidence in all four experiments and for severity in three experiments. A slower disease progress rate and lower incidence and severity occurred on beans planted with maize or sorghum in row, mixed and broadcast intercropping than on bean planted alone. Incidence was reduced 36% and severity 20% in intercropping compared to sole cropping. The built-in disease delay and the slowing of the disease progress rate could provide protection for beans from severe CBB epidemics in intercropped systems. Investigations on the efficacy of various methods of managing root-knot nematodes in microplots and under field conditions revealed that soil solarization, Furadan 5G and Tagetes erecta applied separately or in combination with other control methods, were the most effective in reducing the numbers of three root-knot nematodes, and root gall and egg mass indices. These management methods also resulted in significant increases (P0.05) in number of pods per plant, number of seeds per pod and 100-seed weight, and increased seed yield by up to 96.7 percent. Farmyard manure and Crotalaria ochroleuca were the least effective treatments. The use of T. erecta was the most economical root-knot nematode control method.  相似文献

During a survey conducted from 1998 to 2002, Fusarium proliferatum was found associated with young and adult palms belonging to the genera Chamaerops, Phoenix, Trachycarpus and Washingtonia showing symptoms of wilt and dieback. The fertility and toxicological profile of 36 strains representing different locations and hosts were studied. All of them except two, which were infertile, belonged to mating population D. Both mating types (MATD-1 and MATD-2) were isolated from the same host species, showing a high potential of genetic recombination in the field. Additionally, eight strains were fertile once crossed as female. Toxin analysis showed differences in the ability of strains to produce fumonisin B1, moniliformin, beauvericin, fusaric acid and fusaproliferin. Only 17 of them produced all the toxins analyzed. Pathogenicity tests were conducted on Phoenix dactylifera and P. canariensis using nine F. proliferatum Spanish strains and two reference strains from Saudi Arabia. Eight months after inoculation all strains caused disease, with palms showing lesions on the bases of leaves and development of wilt symptoms similar to those originally observed in affected plants. This is the first report on the occurrence of F. proliferatum on P. dactylifera in Spain and also the first report of this pathogen on C. humilis, P. canariensis, P. reclinata, T. fortunei, W. filifera and W. robusta.  相似文献

Rust control and ethylenebisdithiocarbamate (EBDC) residues in green beans ( Phaseolus vulgaris L. 'Morgan') were evaluated in two seasons of 1993 in Dschang, Cameroon. Four weekly applications of mancozeb (2.8 kg ha- 1), sulphur (2.24 kg ha- 1), or mancozeb+sulphur (1.4+1.12 kg ha- 1) were initiated from initial rust symptoms. In both seasons, treatments with mancozeb alone resulted in a low area under pustule count progress curve (AUPPC) and a significant increase in leaf area index, and pod and green fodder yields. Pod yields were negatively correlated with AUPPC. Pod yield loss was estimated at 61% and 32% in the early and late seasons, respectively, while losses in green fodder yields were 57% and 24% respectively. EBDC residue levels decreased significantly with a delay in pod harvestand when pods were washed in running tap water. Residue levels were high in the late season, possibly due to low rainfall. Pods exposed to the combined fungicidal treatments accumulated higher levels of residues than those sprayed with mancozeb alone, suggesting a potential risk of EBDC accumulation on pods when EBDCs are mixed with sulphur.  相似文献

Routine laboratory testing of 710 bean seed lots from various origins forPseudomonas syringae pv. phaseolicola (Psp) with immunofluorescence microscopy (IF) showed that 27.5% of the seed lots (five subsamples of 1000 seeds tested per sample) contained two or more IF-positive cells in a total of 500 microscope fields (magnification 500×). Simultaneously performed dilution-platings of IF-positive subsamples on King's medium B confirmed presence of Psp for one-third of these IF-positive seed lots. The grey area of disagreement between both laboratory tests was studied by comparison of test data and by field trials.The number of IF-positive cells per subsample was positively correlated with isolation and identification of Psp (R=0.85). The detection level of IF was ca. 102 Psp cells per ml of undiluted subsample extract. The detection level of Psp by isolation on King's medium B was variable, being inversely related with the saprophyte to Psp ratio. The high sensitivity of IF was in part due to high percentages of dormant or dead IF-positive cells in the sample extract. Field trials over two years with 10 000 seeds per seed lot, showed disease incidence for 9 of the 22 seed lots. Of ten IF-positive lots with five positive subsamples per sample, nine were positive in the field test plot (the negative lot gave primary infection spots of Psp when used for commercial growing). By isolation, seven of these ten IF-positive lots were positive. Of the five IF-positive lots with two or less positive subsamples, isolation and field trial were both negative. Based on data on seed transmission from literature, field incidence was unlikely for these five samples in a 10 000 seeds field trial. All seven IF-negative lots were negative in the field trial. The causal agent of cucurbit powdery mildew in southeastern Spain has been investigated since 1996. Of the 139 single-spore isolates obtained, all were identified asSphaerotheca fusca. Four physiological races (1, 2, 4 and 5) of the pathogen were detected. During the survey, a population shift ofS. fusca was observed: race 1 was progressively replaced by other races. In addition, race coexistence was observed in several cucurbit greenhouses. Four host range patterns or pathotypes were distinguished among the isolates ofS. fusca. All isolates were highly aggressive on melon and especially on zucchini cultivars. No clear relationships between races and pathotypes could be established, although isolates virulent on watermelon were preferentially associated with race 1. The interactions ofBotrytis cinerea and seven biological control agents (BCAs) were examined in controlled environments to determine the influence of selected relative humidities (RH) (90,95, and 100%) and air temperatures (20,24 and 28 C) on grey mold of bean. All main effects and interactions were significant (P0.05) among the 72 treatments. In the control, lesions of grey mold developed under all environmental conditions but were largest at 24 C×95 and 100% RH, and 28 C×95% RH. Interactions of environment, BCAs and grey mold were complex.Alternaria alternata, Drechslera sp.,Myrothecium verrucaria, Trichoderma viride, Gliocladium roseum and an unidentified pink yeast were all highly dependent on environment for biological control efficacy, and changes of 4 C or 5% RH were associated with variability in disease suppression that ranged from 15 to 100%. Efficacy ofEpicoccum purpurascens appeared independent of environment and this BCA suppressed disease by 100% in all of the environmental treatments. Suppression of grey mold by many of the BCAs was most effective under environmental conditions least conducive to disease. Therefore, evaluations of potential BCAs in environmental conditions that are marginal for disease can overestimate their efficacy in field environments. Assessments of biological control efficacy in various environments can be used to more accurately assess the potential of BCAs.  相似文献

Common bacterial blight (CBB) in edible beans (Phaseolus vulgaris), incited Xanthomonas campestris pv. phaseoli, reduces bean yields and seed quality. The main objective of this study was to determine resistance to common bacterial blight in bean genotypes. Twenty-two bean genotypes grown in Turkey including common and snap bean cultivars/lines were collected from different parts of Turkey and tested for resistance against to Xanthomonas campestris pv. phaseoli strain MFD-11. All the common and snap bean lines/cultivars tested were moderately susceptible, susceptible or highly susceptible, except AG-7117 which was found resistant to Xanthomonas campestris pv. phaseoli. This is the first report of a resistance source in a common bean line (AG-7117) against Xanthomonas campestris pv. phaseoli.  相似文献

Southern bean mosaic virus (SBMV) has been identified as the cause of a new disease in greenhouse-cultivated common bean (Phaseolus vulgaris), in the south-east of Spain. The identification was based on host range comparisons, morphological and serological characteristics of the virus, the size of its dsRNA species and the nucleotide sequence of an 810-bp fragment from ORF2. The virus could be clearly discriminated from the related sobemovirus Southern cowpea mosaic virus. This is the first report of SBMV in Spain.  相似文献

The relationships between strains of Pseudomonas savastanoi pv. phaseolicola (P. sav. phaseolicola), P. syringae pv. tabaci (P. syr. tabaci) and P. syr. syringae which all cause disease on bean; the related species P. sav. glycinea and P. syr. actinidiae, and reference bacteria, were evaluated by studying the phenotypic and genetic diversity of a collection of 62 strains. All the P. sav. phaseolicola strains tested produced characteristic watersoaked lesions on bean pods. Other pathovars produced varying combinations of symptoms including necrotic lesions, with or without watersoaked centres and sunken tissue collapse of the lesion (P. syr. tabaci) and necrotic lesions with or without sunken collapse (P. syr. syringae). At the genomospecies level, all the strains of P. sav. phaseolicola, P. sav. glycinea and P. syr. tabaci, belonging to genomospecies 2, could be separated from P. syr. syringae strains (genomospecies 1) and P. syr. actinidiae strains (unknown genomospecies) by BOX-PCR and DNA/DNA hybridisation. To distinguish P. sav. phaseolicola, within genomospecies 2, from P. sav. glycinea and P. syr. tabaci, it was necessary to perform nutritional characterisations myo-inositol negative and p-hydroxy benzoate positive for P. sav. phaseolicola strains), PCR with specific primers designed from the tox region (positive for all of the P. sav. phaseolicola strains) and serotyping, as 71% of the P. sav. phaseolicola strains reacted as O-serogroup PHA1. Important intrapathovar variation was seen by genomic fingerprinting with REP and ERIC primers, as well as with RAPD primers (AE7 and AE10) and esterase profilings. While RAPD fingerprinting detected variability correlated with two race-associated evolutionary lines, REP, ERIC and esterase profiles revealed intrapathovar variation linked to some host origins, that separated the kudzu isolates, and the mungbean isolates, from the other P. sav. phaseolicola strains.  相似文献

Xanthomonas campestris pv. phaseoli and X. campestris pv. phaseoli var. fuscans, the causal agents of the common and fuscous bacterial blight of beans, appear to be phenotypically identical except that the latter can produce a melanin-like pigment in culture. Ten isolates of X. campestris pv. phaseoli and 12 isolates of X. campestris pv. phaseoli var. fuscans were examined using pulsed-field gel electrophoresis (PFGE) and restriction fragment length polymorphism (RFLP). The average genome sizes for X. campestris pv. phaseoli and X. campestris pv. phaseoli var. fuscans were 3850.6±48.9 and 3584.3±68.1kb respectively. The genetic relatedness of the isolates was determined from macrorestriction patterns generated using XbaI. Cluster analysis indicated that the non-fuscous and fuscous strains are distinct. RFLP results, based on the highly conserved hrp genes and a pectate lyase gene from Xanthomonas, also indicated that the two bacteria are genetically different. The results obtained in this study suggest that this pathovar can be segregated into two subgroups under a recently proposed reclassification of the Xanthomonas genus.  相似文献

Spatial disease pattern of Cercospora beticola was characterised during natural epidemics of Cercospora leaf spot (CLS) in sugar beet. We applied linear regression and geostatistical analyses to characterise CLS spatial patterns in three field trials, in long-established and recently-established CLS-areas, during two consecutive years. Linear regression showed a positive influence of average disease severity of within-row neighbouring plants (0.38 < < 0.88). Semi-variograms modelled the spatial dependence of disease severity for two directions per week in both years. Disease severity displayed strong spatial dependence over time. The within-row spatial dependence was the largest, but across-row dependence was irregular and weaker. Both long- and recently established areas showed strong spatial dependence of disease severity within row, decrease in variability between years and within the second trial year and a relation between and the relative nugget. Observed differences were more field than area specific. These spatial and temporal analyses indicated that disease severities of adjacent plants were dependent; hence, we concluded that C. beticola is dispersed mainly over short distances from plant to plant.  相似文献

Cucurbit powdery mildew caused by Podosphaera fusca limits crop production in Spain. Since its management is strongly dependent on chemicals, the rational design of control programmes requires a good understanding of the fungicide resistance phenomenon in field populations. Fifty single-spore isolates of P. fusca were tested for sensitivity to three quinone-outside inhibiting (QoI) fungicides: azoxystrobin, kresoxim-methyl and trifloxystrobin. Minimum inhibitory concentration (MIC) values for QoI-sensitive isolates were found to range from 0.25 to 10 μg ml−1 for azoxystrobin to 5–25 μg ml−1 for kresoxim-methyl, using a leaf disc-based bioassay. High levels of cross-resistance to QoI fungicides were found. Eleven isolates showed resistance to the three QoI fungicides tested with MIC and EC50 values >500 μg ml−1 resulting in RF values as high as >715 and >1000 for trifloxystrobin and azoxystrobin, respectively. A survey of P. fusca QoI resistance was carried out in different provinces located in the south central area of Spain during the cucurbit growing seasons in 2002, 2003 and 2004. Examination of a collection of 250 isolates for QoI resistance revealed that 32% were resistant to the three fungicides tested; the provinces of Ciudad Real, Córdoba and Murcia being the locations with the highest frequencies of resistance (44–74%). By contrast, no resistance was found in Badajoz, and relatively low frequencies were observed in Almería and Valencia (10–13%). Nearly 50% of resistant isolates were collected from melon plants. Based on these data, recommendations about the use of QoI fungicides for cucurbit powdery mildew management in the sampled areas are made.  相似文献

In surveys conducted in 2011 and 2012 to identify the viruses causing diseases on pepper and tomato in the department of Alibori in northern Benin, 451 samples of pepper and tomato were analyzed by ELISA using 11 specific antibodies. The highest virus incidence among the surveyed districts was recorded on pepper in Malanville (56.18%), followed by Karimama (39.32%). The most frequently found viruses were Pepper veinal mottle virus (PVMV), Cucumber mosaic virus (CMV), and Potato virus Y-necrotic (PVY-n), accounting respectively for 22.39%, 21.73% and 15.96% of the collected samples. Tomato yellow leaf curl virus (TYLCV) was detected in only 2.43% of the samples, whereas Alfalfa mosaic virus (AMV), Chilli veinal mottle virus (ChiVMV), Tomato mosaic virus (ToMV) and Tomato spotted wilt virus (TSWV) were not detected in any of the samples tested. Double and triple infections involving different virus combinations were found, respectively, in 14.86% and 4% of the samples. Five plant species (Euphorbia hirta Linnaeus, Moringa oleifera Lam, Leucas martinicencis (Jacquin) R. Brown, Combretum micranthum G. Don, Abelmoschus esculentus L. Moench.) out of 30 samples belonging to 13 botanical families, collected within or nearby tomato and pepper fields, were found infected with PVMV, PVY-n, and CMV. Control measures to reduce the impact of viruses on pepper and tomato production are discussed. This is the first report of viruses infecting pepper and tomato in Benin.  相似文献

The relation between the frequency of legume crops in a rotation and the root rot severity in pea was examined in a field survey. Additionally, greenhouse experiments were performed with soil samples from legume rotation trials or from farmers' fields. The frequency of pea crops in current rotations proved to be much less than the recommended value of one in six years. The correlation between pea root rot and the number of years that pea or other legumes were not grown on the field under consideration (called crop interval) was weak. Root rot severity correlated better with the frequency of peas or legumes in general over a period of 18 years, but the frequency still explained only a minor fraction of the variation in disease index. Some experimental data pointed to the occurrence of a highly specific pathogen microflora with continuous cropping of only one legume species, but this phenomenon probably does not occur in farmers' fields. In field samples, root disease index for pea correlated well with that for field bean. The survival of resting structures of pathogens such asAphanomyces euteiches probably explains why the frequency of legume cropping has a higher impact than crop interval on root disease incidence. Pea-free periods and legume frequencies have a poor predictive value for crop management purposes.  相似文献
