Summary Advanced generation random lines from crosses of tolerant x susceptible parents were studied to determine the inheritance of tolerance to barley yellow dwarf virus (BYDV) in oats Avena sativa L. Clintland 64, a susceptible oat cultivar, was crossed with the BYDV tolerant oat lines Otee, FF64/74, M921 and CI4492. Randomly selected lines were advanced to F5 or F6 by single seed descent and were tested for their reaction to BYDV at Palmerston North, New Zealand and Winnipeg, Manitoba and Ste-Foy, Quebec, Canada.The reactions of the lines to BYDV infection from each cross at the three test sites fall into a relatively continuous distribution. Two to four quantitatively inherited genes appear to contribute to the tolerance of the four tolerant oats. At Palmerston North, Clintland 64 expressed some tolerance to the virus.The correlation between the three sites was significant (P=0.01) for all crosses, ranging from 0.36 to 0.69, according to the particular cross considered. A breeding programme was developed to obtain barley yellow dwarf virus (BYDV)-resistant winter genotypes using the Yd2 gene. The aim was to incorporate the Yd2 allele into the new high-yielding genotypes to release cultivars that allow barley cultivation in areas where BYDV is endemic. The resistant lines were developed using pedigree selection. An ICARDA resistant line (83RCBB130) carrying the Yd2 gene was crossed with three susceptible, high-yielding winter varieties and their F1 lines were either selfed or backcrossed to the matching susceptible parent. The best lines selected from subsequent selfing generations were evaluated in replicated trials in the presence or absence of BYDV, starting from F6 and BC1F5 to F8 and BC1F7 generations. Four genotypes with superior agronomic traits and BYDV resistance were selected.  相似文献

Common wheat × hexaploid triticale hybrids were produced and evaluated for tolerance to barley yellow dwarf virus disease (BYD). The BYD tolerance expression varied with wheat × triticale combination. The selection for BYD tolerance increased the recovery of tolerant genotypes in the next generations. Homozygous tolerant and susceptible lines were obtained in advanced generations. The rye chromosomes 1R, 2R, and 4R with 7R were transmitted as disomic or monosomic, disomic, and double disomic substitution to the late generations of ‘Musala’ (common wheat) בMuskox 658’ (triticale), ‘Encruzilhada’ (common wheat) בNord Kivu’ (triticale) and ‘Encruzilhada’× 12th. International Triticale Screening Nursery 267 (12ITSN267) (triticale), respectively. A clear association was established between the 1R chromosome of the ‘Muskox 658’ triticale line and the tolerance to BYDV. Results suggest that the 2R chromosome may be involved in BYD tolerance of ‘Nord Kivu’ triticale line.  相似文献

The Japanese barley cultivar, ‘Chikurin Ibaraki 1’, is partially resistant to the PAV serotype of barley yellow-dwarf virus (BYDV), but its induced mutant line, Ea52, is susceptible. The inheritance of resistance in cv. ‘Chikurin Ibaraki 1’ to BYDV-PAV was investigated. The F, and F2 plants of crosses of cvs ‘Chikurin Ibaraki 1’, Ea52, ‘Vixen’, carrying the Yd2 gene of resistance, and ‘Plaisant’, a susceptible French cultivar, were tested in growth chamber and field conditions. Isolate RG, against which ‘Chikurin Ibaraki 1’ is partially resistant in growth chamber and field conditions, and isolate 2t, which overcomes the partial resistance of ‘Chikurin Ibaraki 1’ in field conditions (Chalhoub et al. 1994) were used. The segregation of F2 plants of crosses between ‘Chikurin Ibaraki 1’ and the susceptible cultivars to isolate RG (one resistant to three susceptible) suggests that the resistance of ‘Chikurin Ibaraki 1’ is controlled by a single recessive gene. All 537 F2 plants of ‘Chikurin Ibaraki 1’בVixen’ tested with isolate RG in growth chamber and field conditions were resistant. The F2 plants of this cross were all resistant to isolate 2t in growth chamber conditions but segregated with a ratio of one resistant to three susceptible in field conditions owing to the susceptibility of ‘Chikurin Ibaraki 1’ to this isolate. Results suggest that the resistance gene in ‘Chikurin Ibaraki 1’ is tightly linked or allelic with the Yd2 gene in ‘Vixen’. However, it differs from this gene in ‘Vixen’ in that it can be overcome by isolate 2t in field conditions.  相似文献

For the purpose of developing barley varieties suitable for light acid soils a collection of about 670 varieties of spring barley was tested for tolerance to low pH. In the investigations the root development of plants in the seedling stage was taken as a criterion for assessment. In 61 varieties a good tolerance to low pH was found. On the basis of root development of seedlings in offspring of crosses between tolerant and non-tolerant varieties, plants with tolerance to acid conditions can be recognized.  相似文献

Five molecular markers associated to two frost tolerance QTLs ( Fr-H1 and Fr-H2 ) were tested both on nine Turkish accessions, classified by breeders as highly frost-tolerant, and on a previously described sample of 26 barleys, winter, facultative and spring. Accessions were characterized in terms of frost tolerance under both field conditions and artificial freezing test at −12°C. The Turkish lines resulted to be equal or superior to the most tolerant European genotypes tested, showing that they can be used to improve the frost tolerance of the EU barley germplasm. The marker Hv BM5A ( Vrn-H1 and Fr-H1 ) resulted to be the best predictor for assisted selection within this germplasm, because of its high correlation between allelic variation and phenotypic traits. Only Hv CBF4 of the three Hv CBF markers tested at Fr-H2 was associated to the trait, but at lower significance than HvBM5A . The PCR-based molecular marker of Vrn-H1 can thus be used in barley breeding not only for selection of facultative and winter types, but also for fast routine selection of frost tolerant genotypes.  相似文献

研究分子标记鉴定大麦抗黄矮病基因Yd2的有效性,可为Yd2基因在大麦抗病育种中的广泛应用提供快速有效的分子辅助选择工具。利用与Yd2基因紧密连锁的YLM、CAPS-Ylp和ASPCR-Ylp标记同时检测52份国内外大麦品种(系)与4份大麦F1杂种的Yd2基因型,同时结合生物学抗性检测的表型分析其有效性。通过对Yd2基因型已知的20份大麦品种(系)及4个F1杂种的Yd2基因型分析,表明YLM、CAPS-Ylp与ASPCR-Ylp标记可以有效判断大麦Yd2基因型。进一步用这3个标记检测32份Yd2基因型未知的大麦的基因型,鉴定出基因型为Yd2-/Yd2-的品种(系) 27份,基因型为Yd2+/Yd2+的品种(系) 5份。在回交育种的分子辅助选择实例中,从BC2F2世代中选出了16个基因型为Yd2+/Yd2+的单株。3个分子标记结合应用能够快速有效地鉴定大麦Yd2基因型,可用于Yd2基因回交育种中的大规模分子标记辅助选择。  相似文献

The effects of the Yd2 gene on tolerance to barley yellow dwarf virus (BYDV) and other agronomically important characters in spring barley were evaluated in a set of randomly selected doubled haploid (DH) lines of an‘Igri’/‘Atlas 68’ cross and three crosses between CIMMYT Yd2 materials and the Czech malting barley ‘Akcent’. The cleaved amplified polymorphic site (CAPS) diagnostic marker Yd2 was used for identification of the Yd2 gene and this analysis showed high agreement with the results of field infection tests. Yd2 lines exhibited significantly lower symptom scores and lower reductions of some grain yield characters, but their resistance level was not consistent over the years. The presence of secondary stresses (high temperature/drought) in 2000 led to relatively higher sensitivity to BYDV infection, strengthened by the long life cycle of genotypes. In cases where secondary stresses were mild (in 2002), the longer life cycle significantly increased sensitivity to BYDV infection only in the absence of the Yd2 gene (in susceptible genotypes). The examination of different vegetative, grain yield and malting quality characters separately for groups of Yd2 and non‐ Yd2 lines did not show any evidence of adverse effect of the Yd2 gene on any character.  相似文献

Bananas are typical to tropical and subtropical climate regions, where they have better conditions due to high air humidity and soil water availability. The production of drought-tolerant varieties is an alternative to expand the cultivation sites to drier areas. In this work, we present a hydroponic system for the genotype selection of Musa sp. (AA) subjected to water stress. A total of 14 banana genotypes were tested; 12 diploids and two commercial varieties with a known response to drought tolerance. The results show that the improved diploids have different responses with respect to water deficit, with the PMGB043 and PMGB044 genotypes considered more susceptible and the PMGB075 and PMGB099 genotypes more tolerant. Under water deficit conditions, most banana diploids maintain a high relative water content, but there is a reduction in aerial part growth and an increase in root dry mass. The accumulation of alcohol dehydrogenase, especially in control treatment plants, indicate that the glycolytic pathway was used to supply energy demand, showing potential flood-tolerant genotypes. The results are essential for the preimprovement of banana cultivars.  相似文献

YLM, a codaominant polymerase chain reaction (PCR) marker linked to Yd2, could substantially improve the precision and efficiency of barley yellow dwarf virus (BYDV) resistance breeding. The aim of this study was to assess the effectiveness of YLM in a marker‐assisted introgression programme and to quantify associations between the presence of Yd2 and other agronomic and quality traits. The Yd2 gene was introgressed into a BYDV‐susceptible background through two cycles of marker‐assisted backcrossing. BC2 F2‐derived lines, either carrying or not carrying the YLM allele associated with resistance, were compared in the presence and absence of BYDV. The YLM marker was shown to be effective in the introgression of Yd2. Lines carrying the YLM allele associated with resistance produced significantly fewer leaf symptoms and showed a reduction in yield loss when infected with BYDV. There were no deleterious effects associated with the introgression of Yd2 on grain yield, grain size or malting quality. The implications of marker‐assisted selection for Yd2 on barley improvement are discussed.  相似文献

To identify scorable marker traits that can be used in cereal breeding programs for selecting drought tolerant individuals, we investigated the correlation among the drought-associated traits in two F2 populations derived from the crosses made between drought tolerant and sensitive barley and wheat parental genotypes. The parental genotypes of these crosses also differed by at least three other traits – paraquat tolerance, leaf size, and the relative water content. These three traits were scored in two F2populations of 80 individuals for each barley and wheat cross. Analysis of results indicated that the enhanced tolerance to paraquat was correlated with reduced leaf size and increased relative water content, two traits associated with water stress phenotypes of the drought tolerant barley and wheat parents. Our results suggested that the selection based on paraquat tolerance istechnically less demanding and thus useful for rapid screening of individuals for enhanced drought tolerance in segregating populations. Selection of tomato plants supposedly tolerant to tomato yellow leaf curl virus (TYLCV), based solely on the absence of symptoms in an infested field can be misleading. An inoculation routine was therefore established to avoid escapes and to overcome difficulties associated with the age of the plant at the time of infection. The inoculation routine was applied to a selection of resistant/tolerant individuals generated through a diallel F1 cross and to F2 segregating populations originating from three wild tomato species described as tolerant to TYLCV: Lycopersicon peruvianum EC 104395, Lycopersicon pimpinellifolium Hirsute and Lycopersicon chilense LA 1969. Clear differences were observed between susceptible symptomatic and tolerant symptomless tomato genotypes, indicating that the uncertainty resulting from escapes, from different levels of inoculum, and from the time of inoculation, can be eliminated. The genes involved in tolerance provided different levels of protection; combinations of various tolerant sources and levels in a single genotype gave a higher level of tolerance. Differences in level of protection were found between genes from the same source and between sources; none of the sources tested had complete dominance. The results obtained with the F2 segregating population showed that tolerance from L. pimpinellifolium is controlled by one major gene, that from L. chilense by two genes, and that from L. peruvianum by three genes with no dominant effect. The combination of sources for resistance can thus have positive or negative synergistic effects, or no effect. We suggest that a maximal level of tolerance can be obtained by the additive effect of the partly dominant genes from L. pimpinellifolium and L. chilense.  相似文献

以24个冬小麦品种(或品系)为试材,研究测定了冬小麦对莠去津反应的敏感性。结果表明,在苗前土壤喷3000mL/hm^2莠去津的条件下,出苗后30d有6个品种表现不敏感(T),7个品种表现中感(MS),4个品种表现中抗(MT);小麦根部受莠去津的为害重于地上部。观察了不同反应型小麦品种之间杂种F1、F2和BC1的敏感和不敏感植株分离比例,小麦对莠去津反应受细胞核基因控制,与细胞质基因无关。该基因属单基因遗传,敏感性为隐性,不敏感性为显性。研究了莠去津对小麦幼胚培养的影响,低浓度莠去津对小麦幼胚愈伤组织诱导率没有明显影响,高浓度莠去津降低了愈伤组织的诱导率。无论培养基中的浓度高低,莠去津对继代培养中愈伤组织的生长状态都有影响。  相似文献

Acid soil and associated aluminium toxicity are considered as the number one abiotic factor limiting crop production. Over 2 billion hectares of acid soils exist world-wide, both in tropical and moderate climatic zones. In Poland acid soils represent up to60% of arable land. At soil pH < 5.0 Al ions become soluble in water and toxic to plants. Genetic improvement of Al tolerance in crops is the only alternative to soil liming, a traditional but short term and expensive agricultural cure to raise soil pH. Of the various cereals, barley is the most sensitive to Al toxicity. The known sources of Al tolerance in barley are limited to old cultivars and landraces. While they represent multiple alleles of a single locus, there is no potential to improve Al tolerance through recombination of non-allelic additive genes. In the Department of Genetics, Silesian University we have employed induced mutations for rapid creation of variability for Al tolerance in barley. Thirteen mutants with increased levels of tolerance to Al toxicity have been selected in M3 generation after mutagenic treatment of four barley varieties with N-methyl-N-nitroso urea (MNH) and sodium azide. Six further Al tolerant mutants were identified in the collection of semi-dwarf mutants of the Department. All selected mutants confirmed Al tolerance with the use of three different methods of screening, i.e., root re-growth, root tolerance index and hematoxylin staining. Fourteen mutants exhibited significant root re-growth after 48 hour incubation with 3 ppm Al+3 and two of them, namely RL819/2 and RL820/6 were tolerant even to 6 ppm Al+3. Crosses of two selected mutants with their respective parent varieties indicated that Al tolerance in each mutant was controlled by a single recessive gene. Out of three methods tested, the root re-growth method facilitated by hematoxylin staining proved to be the most reliable technique for large scale testing. Double treatment with MNH or combined treatment with sodium azide and MNH and 6hinter-incubation germination between treatments were the most successful treatment combinations for induction of aluminium tolerance in barley. Summary In four segregating populations of barley, grown in a naturally highly acid soil good plants and less vigorous ones were marked in a 3–4 leaf stage of development.Some agronomic characters of mature plants were studied in relation with the level of tolerance to high soil acidity of their progenies when tested in a seedling stage in a laboratory method.As compared with the good groups the reductions proved highly significant viz. 60% for mean number of tillers per plant, 25% for average culm length, 35% for mean number of kernels per spike and about 12% for 1000-grain weight.On the average more than 95% of the good plants showed a high level of tolerance to high soil acidity, indicating a close correlation between low-pH tolerance and agronomic performance.Although a rather high percentage (40%) of the progenies among the plants marked as bad was found to be tolerant, those lines showed a poor general performance.This goes to show that selection for tolerance to high soil acidity before flowering is an advisable procedure, without the risk of including sensitive lines or losing otherwise valuable genotypes.This study was carried out during a fellowship of the Spanish Ministry of Agriculture.  相似文献

番茄晚疫病是危害番茄生产的重要病害之一。在我国开展番茄晚疫病菌致病性分化研究对番茄抗病品种选育和番茄晚疫病的综合防治意义重大。以往的研究结果表明,番茄晚疫病菌具有明显的生理小种分化现象,且致病性分化变异频繁。2001年,从北京地区采集并获得了一份番茄晚疫病新菌株,以番茄晚疫病菌的5个鉴别寄主品种为材料,采用苗期喷雾接种法对其致病性进行了研究。根据致病性表现初步得出结论,该菌株属于晚疫病生理小种T1.3。同时,利用该菌株对7份番茄一代杂种和10份育种材料接种鉴定表明,7份一代杂种品种均高度感病;而10份番茄育种材料中,7份表现感病或高感病,3份耐病。另外,在2份耐病品系和3份感病材料中均存在一定比例的抗病单株;而两个感病亲本杂交后,其F1抗性明显增强。可见,番茄晚疫病抗性并非简单的由单显性基因控制的遗传模式。  相似文献

The enzyme-linked immunosorbent assay (ELISA) was used to determine the concentration of barley yellow dwarf virus (BYDV) in leaves of spring wheat Triticum aestivum L. cv. ‘Fukuhokomugi’, Leymus angustus (Trin.) Pilger (Altai wild rye) and their F1 hybrids inoculated at the 2- to 3-leaf stage. The BYDV isolate “Cloutier” (serotype PAV) was used. The results showed that L. angustus confers immunity or strong resistance to the virus. Significant multiplication of the virus occurred in ‘Fukuhokomugi’ which expressed an intermediate level of tolerance to the disease under field conditions. The F1 haploid hybrids of ‘Fukuhokomugi’×L. angustus expressed a level of resistance almost equal to that of their wild parent, with a low concentration of virus appearing 16 days after inoculation and no more afterwards. The discovery of BYDV resistance in L. angustus and the expression of this character in the F1 hybrids provide new opportunities for the enlargement of the gene pool for BYDV resistance in wheat.  相似文献

Soil flooding or waterlogging is a major abiotic stress in upland crops. In barley, there have been several reported studies of selection for flooding-tolerant genotypes, but it is difficult to obtain varietal rankings that are consistent among researchers. Our objectives were to establish experimental conditions that could be applied by other research groups and to verify the varietal ranking conducted in an earlier study. We conducted greenhouse experiments on 14 barley varieties. At the 2.5-leaf stage, they were flooded with 0% or 0.1% soluble starch solution (mimicking reducing conditions). At 13 to 15 days after the start of treatment, the degree of leaf injury and the shoot dry weight ratio (treatment:control) were recorded. Reliable and highly repeatable results were obtained for the criterion of leaf injury under reducing conditions, whereas shoot dry weight ratio was unstable. The varieties OUJ820 and OUA301 were highly tolerant, whereas OUA002 and OUJ247 were sensitive; these results matched those of the earlier study. The experimental conditions that we developed here may be useful for selection testing and genetic analysis of flooding tolerance in other laboratories.  相似文献

Rice is susceptible to cold during early developmental stages. Most tolerant cultivars have been developed for other conditions than those occurring in South America’s rice growing areas, or their grain type is not suitable for the local markets. If locally adapted varieties were available, growers could anticipate sowing date, making flowering time coincide with the moment of maximum solar radiation and increasing yields. In this work, 116 rice inbred lines and varieties of diverse origin within the South American gene pool were tested for seedling survival and germination percentage under low temperature in controlled conditions. As a result, lines used as controls responded as expected, whereas lines with similar behavior to controls, intermediate between susceptible and tolerant controls and more extreme than controls were detected at both seedling and germination stages. Allelic variation at candidate genes OsGSTZ1, OsGSTZ2 and OsCDPK13 was analyzed in a subset of ten contrasting lines. Ten out of thirty-four polymorphisms detected in all three genes were associated with cold tolerance in these lines. A functional marker, designed from an aminoacid-changing SNP found in gene OsGSTZ2, was tested in the 116 lines. As a result, a tight association was observed between cold tolerance and marker score. In conclusion, wide variability for cold tolerance at early developmental stages has been detected in rice advanced breeding materials that are adapted to local growing conditions. Also, a functional marker tightly associated with the trait is available for performing marker assisted selection.  相似文献
