Quorum sensing in Gram‐negative bacteria is regulated by diffusible signal molecules called N‐acyl‐l ‐homoserine lactones (AHLs). These molecules are degraded by lactonases. In this study, six Bacillus simplex isolates were characterized and identified as a new quorum‐quenching species of Bacillus. An aiiA gene encoding an AHL‐lactonase was identified based on evidence that: (i) it showed high homology with other aiiA genes of Bacillus sp.; (ii) the deduced amino acid sequence contained two conserved regions, 104SHLHFDH111 and 165TPGHTPGH173, characteristic of the metallo‐β‐lactamase superfamily; and (iii) the protein had zinc‐dependent AHL‐degrading activity. Additionally, the expression of the aiiA gene was significantly up‐regulated by 3‐oxo‐AHL. The AHL‐lactonase inhibited multiplication of the 3‐oxo‐C6‐AHL‐producing plant pathogen Erwinia amylovora sy69 both in vitro and in planta. The results provide support for the use of the quorum‐quenching functionality of B. simplex in the integrated control of the devastating fire blight pathogen.  相似文献

Cyclization of 3‐aryl‐1‐(2‐hydroxyphenyl)prop‐2‐en‐1‐ones with hydrazine hydrate in refluxing formic acid afforded the title ligands, 5‐aryl‐1‐formyl‐4,5‐dihydro‐3‐(2‐hydroxyphenyl)‐1H‐pyrazoles (HL1–HL4, Ar = Ph, 4‐CH3O‐C6H4‐, 2‐furyl, 2‐thienyl). Reaction of HL1–HL4 with the divalent metal ions, Mn2+, Co2+, Ni2+, Cu2+, and Zn2+, afforded novel complexes of the type [ML2] (M = metal ion; L = deprotonated ligand) which were characterized by elemental analyses, molecular weight determinations, molar conductances, magnetic moments and electronic and infrared spectral data. The ligands behaved as tridentate, coordinating through the phenolic oxygen after deprotonation, N‐2 of the pyrazole ring and oxygen of the 1‐formyl group. The ligands and their complexes were evaluated for growth‐inhibiting activity against four phytopathogenic fungi. Macrophomina phaseoli was generally most sensitive followed by Alternaria alternata and Colletotrichum falcatum while Fusarium oxysporum was least sensitive to the tested compounds. 1,5‐Diphenyl‐1‐pentanone (A) and 1,5‐diphenyl‐2‐penten‐1‐one (B) are natural products extracted for the first time from Stellera chamaejasme. Laboratory bioassay showed that the two products have strong contact activity and very good anti‐feedant activity against Aphis gossypii and Schizaphis graminum. Both products showed dose‐dependent relationships for both forms of activity against the two aphids, the contact activity of B being about twice that of A. Both products were inferior to methomyl in contact activity but superior in anti‐feedant activity against the two aphids. This is the first report of aphicidal activity in these two compounds, which may represent a new class of aphicide. © 2001 Society of Chemical Industry  相似文献

Summary Weeds cause yield losses and reductions in crop quality. Prior to the introduction of selective herbicides, the drudgery of manual weeding forced farmers to adhere to a suit of weed management tactics by carefully combining crop rotation, appropriate tillage and fallow systems. The introduction of selective herbicides in the late 1940s and the constant flow of new herbicides in the succeeding decades provided farmers with a new tool, ‘the chemical hoe’, putting them in a position to consider weed control more independently of the crop production system than hitherto. The reliance on herbicides for weed control, however, resulted in shifts in the weed flora and the selection of herbicide‐resistant biotypes. In the 1980s, the public concern about side‐effects of herbicides on the environment and human health resulted in increasingly strict registration requirements and, in some countries, political initiatives to reduce the use of pesticides were launched. Today, the number of new herbicides being introduced has decreased significantly and integrated weed management has become the guiding concept. Farmers also have the option of growing herbicide‐resistant crops where the biology of the crop has been adapted to tolerate herbicides considered safe to humans and environmentally benign. This paper discusses some of the recent developments in herbicide discovery, technology and fate, and sketches important future developments.  相似文献

Plant glutathione S‐transferase (GST) forms a major part of the herbicide detoxification enzyme network in plants. A GST cDNA was isolated from Echinochloa crus‐galli and characterised. The gene, designated EcGST1 (E. crus‐galli GeneBank no: JX518596 ), has a 684 bp open reading frame predicted to encode a 25 kD protein. Sequence alignment showed that EcGST1 is a GST homologue. Its expression in response to quinclorac treatment was monitored in seedlings (leaves and roots) and adult plants (leaves, roots, stems and seeds) of quinclorac‐resistant (R) and susceptible (S) biotypes of E. crus‐galli. EcGST1 expression was 1.5–3 times greater in the R plants than in the S plants. However, after exposure to quinclorac, the difference in the expression levels of EcGST1 in R plants, compared with S plants, increased to a ratio of 6–10. Enhanced EcGST1 levels should enable greater quinclorac detoxification following quinclorac stimulation in R plants. GST‐based metabolism may be partially responsible for resistance to quinclorac in E. crus‐galli. The results suggest a new resistance mechanism for this R biotype in Chinese rice fields.  相似文献

Reduction in the amount of active substance and reduced dependence on chemical plant production products are the main items in the Dutch Government Multiyear Crop Protection Plan. There are problems in reaching this goal, as weather conditions in The Netherlands are very beneficial to all kinds of fungal diseases. Such diseases have to be controlled by applications of preventive fungicides, and it is quite regular to use a spray interval of 6–7 days. Another problem is application at the wrong time. With the help of decision‐support systems (DSSs), it is possible to calculate the period of protection by a product, the danger of an infection period and the moment of highest efficacy. DLV‐Meteo offers advice based on five DSSs for individual pests (Prophy, onion leaf spot disease, Botrypré, Mycos and Contapré) and on a general DSS for application at the time of day that ensures highest efficacy (Gewis).  相似文献
