Two‐line hybrid rice technology is an effective way to increase rice production and improve rice quality. In this study, three bacterial blight (BB) resistance genes, Xa7, Xa21 and Xa23, were introgressed separately into C815S, a popular thermo‐sensitive genic male sterile (TGMS) line to develop five BB‐resistant lines (Hua1005S, Hua1002S, Hua1009S, Hua1006S and Hua1001S) to be resistant against seven races of Xanthomonas oryzae pv. oryzae (Xoo). The two‐line hybrids with heterozygous Xa23 were resistant against seven Xoo strains. But, the hybrids with heterozygous loci for both Xa7 and Xa21 were only resistant against three Xoo strains and were moderately susceptible to the other four strains indicating the role of modifiers influencing the poor expression of dominant BB resistance genes under heterozygous state. Among them, Hua1006S was found to be a promising TGMS line with its higher degree of disease resistance level on account of broad‐spectrum resistance gene Xa23 besides possessing better plant type and rice grain quality features.  相似文献

Four bacterial blight (BB) resistance genes, Xa7, Xa21, Xa22 and Xa23, were introgressed into an elite hybrid rice restorer line Huahui 1035, by marker-assisted selection (MAS). Ten promising BB resistant lines identified by MAS approach in Huahui 1035 restorer background and their respective F1 hybrids with a cytoplasmic male sterile line i.e. Jinke 1A, three BB resistant gene donors and their recurrent parent were evaluated for BB resistance by inoculating them with eleven representative races of Xanthomonas oryzae pv. oryzae (Xoo) in China. Further, the ten marker assisted selection of BB (MAS-BB) resistant restorer lines and their F1 hybrids were also characterized for agro-morphological traits and grain yield. Results revealed that restorer lines with Xa23 introgression i.e. HBQ809 and HBQ810 were found to be resistant to all eleven Chinese representative Xoo races, showing broad spectrum resistance to BB. However, the lines possessing Xa7 or Xa7+Xa21 were resistant to ten out of eleven Xoo races. While restorer lines with Xa21 were resistant to nine out of eleven Xoo races. Interestingly, the F1 hybrids with Xa23, Xa7 or Xa7+Xa21 were resistant to two severe epidemic Xoo races of China. Restorer lines i.e. HBQ807 and HBQ808 possessing Xa22 were only resistant to six or five out of inoculated eleven Xoo races. Lesion length comparisons between restorer lines and their F1 hybrids for the above four BB resistance genes showed them to be dominant in heterozygote genotype. Three promising high yielding F1 hybrids with BB resistance were identified for immediate exploitation for hybrid rice production in China.  相似文献

水稻抗纹枯病QTL表达的遗传背景及环境效应   总被引:6,自引:4,他引:2  
利用水稻纹枯病菌强致病菌系RH-9人工接种Lemont导入到特青背景的213个近等基因导入系(TQ-ILs)群体和特青导入到Lemont背景的195个近等基因导入系(LT-ILs)群体,定位和分析了水稻抗纹枯病数量性状座位(quantitative trait loci, QTL)及其表达的环境与遗传背景效应。亲本Lemont对RH-9表现为高度感病,特青表现为中等抗病。人工接种后TQ-ILs群体的相对病斑高度(病斑高度与株高比)呈连续正态分布,LT-IL群体则明显偏向感病亲本Lemont。在不同年份和遗传背景下检测到影响纹枯病相对病斑高度的主效QTL 10个和互作QTL 13个,其中2006年在TQ-IL群体定位到的6个主效QTL在2007年均得到验证,表明这些QTL具有较好年度间的重复性。QSh4是唯一在双向导入系背景下表达的QTL,该位点特青等位基因降低相对病斑高度,提高抗性水平。在TQ-ILs群体中定位到位于第10染色体RM216~RM311区间的QSb10a与在LT-IL群体中定位到的位于相邻区间RM222~RM216的QSb10b的基因作用方向不同,推断这两个QTL存在紧密连锁关系。绝大多数在TQ-IL群体中表达的主效及互作QTL在LT-ILs群体中不表达,表明水稻抗纹枯病QTL具有明显的遗传背景效应。通过比较作图,本研究定位到的其中8个QTL在以往不同群体中同样被检测到,这些主效QTL对通过分子标记辅助选择(marker-assisted selection, MAS)培育水稻抗纹枯病育种可能具有应用价值。研究指出,标记辅助选择在不同遗传背景中能稳定表达的QTL或通过聚合不同抗病QTL是进一步提高水稻纹枯病抗性水平的一个有效途径。  相似文献

‘Minghui 63’ is a restorer line widely used in hybrid rice production in China for the last two decades. This line and its derived hybrids, including ‘Shanyou 63’, are susceptible to bacterial blight (BB), caused by Xanthomonas oryzae pv. oryzae (Xoo). To improve the bacterial blight resistance of hybrid rice, two resistance genes Xa21 and Xa7, have been introgressed into ‘Minghui 63’ by marker‐assisted selection and conventional backcrossing, respectively. The single resistance gene‐introgressed lines, Minghui 63 (Xa21) and Minghui 63 (Xa7) had higher levels of resistance to bacterial blight than their derived hybrids, Shanyou 63 (Xa21) or Shanyou 63 (Xa7). Both Xa21 and Xa7 showed incomplete dominance in the heterozygous background of rice hybrids by infection with GX325 and KS‐1‐21. The improved restorer lines, with the homozygous genotypes, Xa21Xa21 or Xa7Xa7, were more resistant than their hybrids with the heterozygous genotypes Xa21xa21 or Xa7xa7. To further enhance the bacterial blight resistance of ‘Minghui 63’ and its hybrids, Xa21 and Xa7 were pyramided into the same background using molecular marker‐aided selection. The restorer lines developed with the resistance genes Xa21 and Xa7, and their derived hybrids were evaluated for resistance after inoculation with 10 isolates of pathogens from China, Japan and the Philippines, and showed a higher level of resistance to BB than the restorer lines and derived hybrids having only one of the resistance genes. The pyramided double resistance lines and their derived hybrids have the same high level of resistance to BB. These results clearly indicate that pyramiding of dominant genes is a useful approach for improving BB resistance in hybrid rice.  相似文献

Bacterial leaf blight (BB) is among the “top 3” diseases in rice production. Breeding resistant cultivars has been the most effective strategy for BB management. The inbred cultivar Baixiangzhan (BXZ) showed a broad spectrum of resistance to five Xoo pathotypes in China, including the prevalent and highly virulent Xoo pathotypes Chinese Race V (CV), which can overcome the resistance of Xa4 and Xa21. The resistance heredity of BXZ has been explored in this study. A single recessive major resistance gene, which designated as xa42(t), confers resistance against the tested Xoo pathotypes CV. Linkage analysis lands xa42(t) on chromosome 6, and genetic mapping confines it to 3.9 cM region flanked by RM20558/RM20547 and RM20580. A further seven markers were developed from this interval for high‐resolution mapping, and the xa42(t) locus was narrowed to 34.8 Kb segment bounded by RM20572 and DT46. The only functionally predicted gene included in the target region is LOC_Os06g45960 if based on the ‘Nipponbare’ reference sequence. This candidate gene is predicted to encode a cytochrome P450 protein.  相似文献

以优质粳稻品种Lemont与高产籼稻品种特青为亲本培育的高代双向回交导入系为材料,在温室140 mmol L-1 NaCl胁迫条件下定位影响苗期叶片盐害级别(SST)、幼苗存活天数(SDS)、地上部K+浓度(SKC)和地上部Na+浓度(SNC)及人工气候室条件下影响地上部K+、Na+浓度的QTL。双向导入系的大部分遗传背景与各自的受体亲本相同,其中Lemont背景导入系中轮回亲本Lemont的基因组平均占83.8%,特青背景导入系中轮回亲本特青基因组平均占88.9%。各耐盐相关性状在两个背景群体中均出现超亲分离,多数性状的频率分布呈相互重叠状态,表明双亲作为供体相互导入各耐盐性状基因的效应大致相当。两个背景导入系群体中分别检测到影响上述耐盐相关性状的QTL各18个,同一性状在两个背景导入系中未能检测到任何相同表达的QTL,表明耐盐QTL表达具有很强的遗传背景效应,同时也说明这些耐盐QTL的效应可能较小。温室和人工气候室两种环境下仅在特青背景导入系中检测到1个影响SKC的相同QTL,表明耐盐QTL与环境的互作非常明显。虽然双亲均表现中等感盐,但QTL定位结果表明双亲中都存在一些提高耐盐相关性状的有利等位基因。研究认为,利用分子标记技术挖掘“隐蔽”于育成品种中的耐盐基因,进一步利用分子标记辅助选择技术对这些非等位耐盐基因进行聚合,完全有可能提高育成品种的耐盐水平。  相似文献

Bacterial blight (BB), caused by Xanthomonas oryzae pv. oryzae (X00), is one of the most devastating diseases of rice world‐wide; it is also a serious problem of hybrid rice production in China. In this study, a molecular marker‐assisted introgression of Xa21, a gene highly resistant to a broad spectrum of Xoo strains, from ‘IRBB21’ was performed to improve the BB resistance of‘6078′, a new restorer line with high yielding potential. The entire process took one generation of crossing followed by three generations of backcrossing and one generation of selfing. The presence of Xa21 in each generation was determined by both polymerase chain reaction (PCR) and pathogen inoculation. Recombinations between Xa21 and flanking markers were identified by PCR analysis. Background selection was conducted in BC1F1 and BC2F1 using amplified fragment length polymorphism (AFLP) markers detecting a total of 129 polymorphic bands between‘6078’ and ‘IRBB21′. The individual selected in BC3F2, or‘6078′(Xa21), carried a fragment of less than 3.8 cM from the donor line in the Xa21 region on chromosome 11, and about 98.8% of the genetic background from the recurrent parent. The results showed that‘6078′(Xa21) had the same level and spectrum of BB resistance as the donor parent ‘IRBB21′, while maintaining the agronomic performance and combining ability of the original 6078. A significant increase in BB resistance was also achieved in the hybrid using 6078(Xa21) as the restorer line.  相似文献

Basmati rice is highly susceptible to bacterial blight (BB) caused by Xanthomonas oryzae pv. oryzae. Transfer of BB resistance genes from non‐Basmati sources to Basmati through cross‐hybridization requires strict monitoring for recovery of the desirable Basmati quality traits in the recombinants, which show complex inheritance pattern. We integrated background analysis using mapped microsatellite markers with foreground selection to identify superior lines that combine useful genes from a non‐Basmati BB resistance donor line IRBB55 with grain and cooking quality characteristics of the popular Basmati rice variety ‘Pusa Basmati 1’ (PB 1) employing backcross pedigree strategy. Foreground selection using linked markers ensured presence of two genes, xa13 and Xa21 for BB resistance from IRBB55, and the recurrent parent PB 1 allele for the waxy locus giving intermediate amylose content and maintainer allele at fertility restorer locus in the BC1F5 recombinants. Background analysis enabled selection of recombinants with recurrent parent genome to the extent of 86.3% along with the quality traits. The extent of introgression of non‐Basmati donor chromosome segments in the superior selections was estimated to be < 7.8 Mb and < 6.7 Mb in the xa13 and Xa21 linked genomic regions, respectively. Association mapping identified three quantitative trait loci, one each for 1000‐grain weight, fertile grains/panicle and cooked kernel length. The backcross‐pedigree breeding strategy facilitated recovery of additional desirable characteristics from the donor in some of the selections. The elite selection Pusa 1460‐01‐32‐6‐7‐67 with maximum genomic background and quality characteristics of the recurrent Basmati parent gave resistance reaction against BB, similar to that of the non‐Basmati resistant check variety and recorded an yield advantage of 11.9% over the best check in the multiplication agronomic trial in the Basmati growing region of India. This line, which has been released as a new variety in the name of ‘Improved Pusa Basmati 1’ for commercial cultivation in India, is an example of successful application of marker assisted selection to variety development.  相似文献

To improve salt tolerance of two elite rice varieties, Ce258 and Zhongguangxiang1 (ZGX1), two sets of introgression lines (ILs) each comprising 200 BC1F10 lines derived from a common donor, IR75862, and two recipient parents, Ce258 and ZGX1, were used for mapping of QTLs for four salt tolerance‐related traits at the seedling stage. Although the three parents were susceptible to salt, the two IL populations showed transgressive segregations for salt tolerance with 12 and 8 salt tolerance ILs in the Ce258‐ILs and ZGX1‐ILs. Eighteen main‐effect QTLs were identified for the four traits in the two IL populations, and the IR75862 alleles at most loci showed increased and decreased salt tolerance in the ZGX1 and Ce258 backgrounds, suggesting overwhelming genetic background effects on QTL detection for salt tolerance. The qDSS11 simultaneously detected in the two backgrounds was validated in a F2 population derived from a salt tolerance line and ZGX1. Our results indicated that salt tolerance‐enhancing allele could be identified in the elite susceptible breeding lines and that introgression of the favourable alleles could facilitate the development of superior lines with greater salt tolerance levels.  相似文献

灌溉与自然降雨条件下水稻高代回交导入系产量QTL的定位   总被引:5,自引:2,他引:3  
利用254个Lemont导入到特青背景的高代回交导入系定位了灌溉(对照)与自然降雨(干旱胁迫)环境下影响单株籽粒产量及其穗部相关性状的QTL。在两种环境下共检测到32个影响单株粒籽产量、千粒重和每穗实粒数的主效QTL,根据不同环境下表达的情况将其分成3类,第1类10个QTL,在两种环境下均被检测到;第2类14个QTL,只在对照条件下检测到;第3类8个QTL,受干旱胁迫诱导,只在胁迫条件下被检测到。此外还检测到9个影响胁迫与对照条件下性状差值的QTL。认为在两种条件下均检测到的相对稳定的3个QTL(QGn11b、QGn12和QGn11b)及影响两种条件下性状差值(即性状稳定性)的9个QTL可能对耐旱性有直接贡献。在所有12个耐旱QTL中,除在QGn5和QGy1的Lemont等位基因减小性状差值(即增强耐旱性)外,其余位点上增强耐旱性的等位基因均来自特青。另外通过与源自相同亲本的不同定位群体在不同环境下定位结果的比较,鉴别出一些受遗传背景和环境影响较小的QTL如QGn3b、QGw1、QGw5、QGy1、QGy5、QGy8和QGy10。对应用QTL定位结果进行标记辅助选择培育耐旱品种进行了探讨。  相似文献

Bacterial blight caused by Xanthomonas oryzae pv. oryzae (Xoo) is an important disease constraining rice (Oryza sativa L.) production worldwide. The XM6 line was induced by N-methyl-N-nitrosourea from IR24, an Indica cultivar that is susceptible to Philippine and Japanese Xoo races. XM6 was confirmed to carry a recessive gene named xa20, resistant to six Philippine and five Japanese Xoo races. The chromosomal gene location was found using 10 plants with the shortest lesion length in an F2 population consisting of 298 plants from a susceptible Japonica variety Koshihikari × XM6. Analysis using PCR-based DNA markers covering the whole rice genome indicated the gene as located on the distal region of the long arm of chromosome 3. The IKC3 line carries IR24 genetic background with Koshihikari fragment on chromosome 3 where a resistance gene was thought to be located. The F2 population from IKC3 × XM6 clearly showed a bimodal distribution separating resistant and susceptible plants. Further linkage analysis conducted using this F2 population revealed that xa20 is located within the 0.8 cM region flanked by DNA markers KIC3-33.88 (33.0 Mb) and KIC3-34.06 (33.2 Mb). This study yields important findings for resistance breeding and for the genetic mechanism of Xoo resistance.  相似文献

水稻第9染色体上存在1个抗纹枯病QTL,被命名为qSB-9,水稻品种特青在该QTL上携带抗性等位基因qSB-9Tq,而Lemont携带相对感病等位基因qSB-9Le。为精确地评价qSB-9Tq的抗病效应,分析其作用方式,利用分子标记进行前景选择和背景选择,从轮回亲本Lemont与特青回交后代群体中筛选到1个目标单株。连续3年对该单株的扩繁后代(BC6F2)及随后获得的近等基因系采用嵌入法进行接种鉴定试验。田间试验采取2种不同的设计。第一种是完全随机试验,即从BC6F2分离群体中筛选出目标区间为qSB-9TqTq纯合型、qSB-9LeLe纯合型和qSB-9TqLe杂合型个体,并对3种基因型个体间的病级平均数差异进行统计分析。第二种设计为随机区组设计,即在BC6F3和BC6F4代,分别对上述3种基因型的近等基因系群体,按3次重复的随机区组设计进行移栽和接种鉴定试验。结果表明,3年的试验结果表现出一致的趋势,即qSB-9Tq存在于分子标记RM242~Y92.5之间,可减轻病级1.0级(0~9级病情分级系统)左右,且其抗性表现为几乎完全的显性特征。本研究的结果为qSB-9Tq的精细定位和育种利用奠定了基础。  相似文献

日益严重的水资源危机使水稻的耐旱育种尤为迫切, 干旱环境下较高的植株水分含量有助于提高或维持作物产量的稳定性, 挖掘与耐旱性密切相关的分子标记有助于提高耐旱品种的选育效率。从供体Lemont (粳稻)导入到特青(籼稻)背景的254个高代回交导入系中筛选出覆盖供体全基因组的55个回交导入系, 采用PVC管栽培, 分析了干旱(胁迫)条件下水稻植株水分状况相关性状与籽粒产量、生物量的相关性并定位了相关QTL。研究表明, 植株水分相关性状(相对含水量、叶片水势、渗透势、卷叶度)均与籽粒产量显著相关。检测到7个相对含水量QTL, 7个叶片水势QTL, 5个渗透势QTL及5个卷叶QTL; 另检测到5个产量QTL, 7个生物量QTL。分析发现, 不仅QLwp5、QLr5、QRwc5和QY5同时分布在RM509~RM163区域, 且该区域还分布有对水分环境表现稳定的产量QTL(QGy5), 效应方向一致, 从遗传学角度解释了籽粒产量与水分相关性状之间的显著相关性。另外, QLr5、QRwc5、QY5、QLr2、QLr7、QLr8、QLr9、QRwc3、QRwc4a、QRwc12及QY7 等11个QTL曾在不同遗传背景群体中被检测到, 它们控制相同目标性状。研究认为RM509~RM163区域及QLr2、QLr7、QLr8、QLr9、QRwc3、QRwc4a、QRwc12和QY7所分布的染色体区域对水分环境或者遗传背景相对稳定, 在水稻分子标记辅助选择(MAS)耐旱育种实践中有较重要利用价值。  相似文献

DNAs of 693 isolates of bacterial blight pathogen of rice, Xanthomonas oryzae pv. oryzae (Xoo), were characterized using PCR-based primers pJEL1 and pJEL2. The pathogen populations were grouped into 97 haplotypes based on DNA-banding patterns. An un-weighted pair-group method using arithmetic averages (UPGMA) indicated a high level of diversity in the pathogen isolates (51 lineages of Xoo at a 70% similarity level). Among these, lineages 5, 7, 27, and 29 are widely distributed and others are localized in the northern region of India. The isolates represent lineage-27, were prevalent in the entire disease-prone area in the region except at Ferozepur. Pathotyping data of the representative isolates of each lineage also indicate 17 different reaction patterns on a set of isogenic lines. Resistance genes xa8 and Xa21 were the most effective followed by xa5, and Xa7 against Xoo isolates prevalent in northern India. Different genes in combinations (xa5+xa13, xa5+Xa21, xa13+Xa21, and xa5+xa13+Xa21) in IR24 genetic background provided better protection against all the pathogen isolates tested in this study than did the component genes. 铁和锌是水稻生长必需的微量元素,也是重金属污染元素。在低洼或酸性土壤中,水稻容易遭受亚铁和锌毒害,抑制水稻生长,造成生物量和产量下降。为探讨水稻苗期耐亚铁毒、锌毒的遗传机制,利用优质粳稻品种Lemont和高产籼稻品种特青为亲本构建的高代双向回交导入系和308个在染色体上均匀分布的SNP标记剖析耐亚铁毒、锌毒相关的QTL。从双向导入系共检测到42个影响耐亚铁毒、锌毒相关性状如苗高、苗干重、根干重以及胁迫与对照相对值的QTL,多数位点增强亚铁毒、锌毒抗性的有利等位基因来自Lemont。其中同时在2个背景下表达的QTL有4个,占定位QTL总数的9.52%,说明大多数QTL的表达具有明显的遗传背景效应。同一遗传背景下同时影响耐亚铁毒和锌毒的QTL有9个,其中QSdw5在2个背景中均被检测到,其效应大小和方向一致,说明水稻苗期耐亚铁毒、锌毒之间存在遗传重叠位点。因此,通过分子标记辅助选择从Lemont中导入或聚合有利的遗传重叠区域,可以提高特青对亚铁毒、锌毒的抗性水平。  相似文献

The introgression lines (ILs) of the wild tomato species Solanum pennellii have been widely used to identify genes related to yield, texture, disease resistance and stress tolerance. In addition to flavour, fruit firmness is an important evaluation index and essential trait indicating tomato fruit quality. Quantitative trait loci (QTL) for fruit firmness have been identified through hand squeezing and pericarp puncturing. However, these techniques hardly reveal the force value of the whole fruit suffering from rupture or deformation. In this study, S. pennellii ILs were used to identify QTLs related to fruit firmness through flat‐plate compression. Nine QTLs for enhancing and sixteen QTLs for decreasing fruit firmness were successfully identified. Compared with that of ‘M82’, the amount of QTLs that enhance fruit firmness increased by 8.76% to 21.00%, and the amount of QTLs that reduce fruit firmness decreased by ?8.27% to ?30.80%. The QTL Crf12a and Crf‐R‐7b is the strongest and weakest QTL, respectively, while they are very stable in all independent biological trials. Six QTLs should be further confirmed through open‐field trials.  相似文献

Bacterial blight (BB) of rice caused by X. oryzae pv. oryzae is a major production constraint in commercial hybrid rice production in the Philippines because most of the parental lines used in hybrid production do not carry resistance genes against the pathogen. In this study, three bacterial blight resistance genes, Xa4, Xa7 and Xa21, were introgressed to a temperature-sensitive genetic male sterile (TGMS1) line. A three-way cross of AR32-19-3-3/TGMS1//IRBB4/7 (PR36944) was made to produce 1,364 F2 plants carrying various combinations of Xa4, Xa7 and Xa21. Individual plants were characterized for reaction to bacterial blight PXO61 (race 1), PXO86 (race 2), PXO99 (race 6) and pollen sterility. Of 144 F2 plants demonstrating resistance against PXO61, PXO86 and PXO99, 22 exhibited highly resistant phenotypes with mean lesion lengths ranging from 0.37–2.97 cm. Analysis of disease reaction identified 20 potential TGMS F2 plants containing Xa4, Xa7 and Xa21 while 78 plants with Xa4 + Xa7. Phenotypic and polymerase chain reaction (PCR) analyses confirmed PR36944-450, PR36944-473 and PR36944-700 as homozygous for Xa7 and Xa21 and highly resistant to all three Xoo races. Fertility of PR36944-450 and PR36944-700 was restored at permissive temperature in a growth chamber. BB-resistant TGMS lines should facilitate breeding two-line hybrids in the tropics.  相似文献

Forty resistant rice cultivars were studied for theinheritance of resistance to bacterial blight usingPhilippine races of Xanthomonas oryzae pv.oryzae (Xoo). Results showed that all thevarieties have at least two recessive genes forresistance. One of these genes governs resistance torace 1 (PXO61) while the other gene confers resistanceto race 6 (PXO99). In addition to the recessivegenes, nine of the varieties possess another dominantgene which also confers resistance to race 1.Allelism tests revealed that the recessive genesgoverning resistance to race 1 in 39 varieties areallelic to xa5 while the dominant genes in thenine varieties are allelic to Xa4. Therecessive gene conferring resistance to race 1 incultivar Sada Diga is inherited independently of xa5. One-hundred and sixty-seven accessions belonging to 13 wild Oryza species were screened against the bacterial leaf blight (BB) pathogen (Xanthomonas oryzae pv. oryza) in Punjab, India. Nine species were identified as carrying resistant genes against the four most prevalent pathotypes under conditions in Punjab. These included O. barthii (eight accessions), O. longistaminata (five accessions), O. meridionalis (three accessions), O. nivara (five accessions), O. rufipogen (two accessions), and one accession each from O. punctata, O. minuta, O. malampuzhaensis and O. latifolia. This is the first report showing the availability of BB resistance in two species namely, O. meridionalis and O. punctata. The resistant accessions, especially those with a similar genome (AA) could potentially be used for the transfer of BB resistance to well-adapted high-yielding rice cultivars.  相似文献
