Summary Cells of seven potato cultivars were selectedin vitro with culture filtrate (CF) ofPhytophthora infestans (Mont.) de Bary. Regenerated plants were tested for late blight resistance. The aim of the study was to check the efficacy of CF in the selection of potatoes for resistance to late blight and to evaluate the effects of additive factors on general resistance. One selection cycle, applied to a cell culture system, selected cells resistant to toxic metabolites of CF.In vivo screening of clones regenerated from selected cells was done in two steps: on whole plants, assessing foliage late blight, and on detached leaves, assessing single factors of horizontal resistance. In general, the frequency of resistant variants selected with CF did not differ from that of resistant somaclonal variants. Nevertheless, high concentrations of CF in the growth medium seemed to induce an improvement of some partial resistance factors as compared with the source plants.  相似文献

Coloured potatoes (Solanum tuberosum L.) rich in anthocyanins are gaining increasing interest on the markets, mainly due to beneficial health effects. Potato varieties containing colour pigments are therefore utilized more and more in actual breeding programmes. In this study, 10 purple-fleshed potato cultivars were compared in their resistance to soft rot caused by Pectobacterium carotovorum (Pc) with 10 white/yellow-fleshed potato cultivars. The results revealed that coloured potato cultivars exhibited on average better resistance to soft rot than white/yellow-fleshed potato cultivars. On tuber halves of purple-fleshed cultivars, the extent of rotting caused by Pc was on average 28.6% lower than on white/yellow-fleshed cultivars. The greater resistance of coloured potatoes coincided with the presence of anthocyanins, higher concentrations of soluble phenols and elevated polyphenol oxidase activity in tuber tissue. These three components were significantly correlated with the extent of rotting caused by Pc. However, coloured and white/yellow-fleshed cultivars were not very different in phenylalanine ammonia-lyase and peroxidase activity. There were also no significant differences in concentrations of dry matter, starch, crude protein and glycoalkaloids. Key findings reveal that total soluble phenols and anthocyanins contributed crucially to resistance expression in tuber tissue of coloured potato cultivars.  相似文献

Summary Potato cultivars of different maturity classes and levels of resistance toPhytophthora infestans were grown under several disease intensities in three field trials. Seasonal courses of ground cover by green foliage and final tuber yields were determined. Light use efficiencies (LUE) were calculated from regression analyses of yield on cumulative light interception. Late blight reduced tuber yields by decreasing cumulative light interception without affecting LUE. No differences in LUE between cultivars or cultivar classes were detected. Therefore, the maintenance of green leaf area is important when breeding potatoes for optimal performance in the presence of late blight. The results support the hypothesis that the correlation between lateness and reported resistance of potato cultivars is due to the vigorous foliage growth of late cultivars.  相似文献

Summary The susceptibility of 43 commercially available potato cultivars to dry rot caused byF. sulphureum, F. solani andF. oxysporum under similar conditions was examined over two years. The cultivars reacted with differing levels of resistance to the dry rot species.F. sulphureum was more aggressive than the other two species and the resistance to each species was independent. The overall cluster analysis classified the cultivars into six groups. Saturna, in an individual group, is the most resistant variety to all tested species, and also toF. surphureum. Panda, Fregate, Folva, Arian and a few others in a group are resistant toF. solani. AlthoughF. oxysporum was the least pathogenetic, almost half of the cultivars are included in a resistant group to this species. There was little correlation between the rank orders of susceptibility of the cultivars to the two main pathogens.  相似文献

Summary Foliar sprays of calonyctin, a natural plant growth regulator extracted from the leaves ofCalonyction aculeatum (L.) House, at a concentration 0.05 activity units, increased the tuber yield of six potato (Solanum tuberosum L.) cultivars by 7–19% (mean 15%) during 1993 and 1994. Physiological differences between calonyctin-treated and untreated potato plants were usually detected 30–40 days after spraying them with calonyctin. The chlorophyll content of treated potato leaves increased, and the chlorophyll a:b ratio decreased. By 40 days after treatment the root oxidizability and soluble protein contents of different parts of the plants also increased. Their subsequent decline as the plants aged was inhibited by calonyctin. It is concluded that calonyctin delayed the senescence of potato plants and increased tuber growth at later stages of development.  相似文献

Summary Variation in aggressiveness to tubers among isolates ofPhytophthora infestans sampled from three potato growing regions in the Netherlands was compared. Variation in the ability to infect tubers of cv. Bintje was found between isolates of each of the three regional populations. The most aggressive isolate of the old population matched the average level of the new population in its ability to infect tubers. As a consequence, the commonly used reference isolate VK 6C can no longer be considered to be representative of the present population ofP. infestans. Therefore it is recommended that testing tuber resistance for the official list of potato cultivars with this isolate should be discontinued. Tuber infection and subsequent spread of the fungus in the tuber tissues were not found to be correlated. The components of tuber pathogenicity studied were not correlated to pathogenicity factors in the foliage, as measured under growth chamber conditions.  相似文献

Summary The kinetics of accumulation of phytoalexins, glycoalkaloids and phenolics was studied in two potato cultivars differing in their degrees of field resistance when infected withPhytophthora infestans. Tuber slices and leaves of cvs Pampeana INTA (high degree of field resistance, free of R genes) and Bintje (susceptible) were infected with race C (complex race 1, 3, 5, 7, 11) ofPhytophthora infestans. Phytoalexins and phenolics accumulated in tuber and leaf tissues which had been inoculated. The levels of these compounds in the susceptible cv. Bintje were relatively low and similar to those found before inoculation. Leaves of cv. Pampeana INTA had a very high glycoalkaloid content, suggesting that glycoalkaloids may play a role in protection of leaves against the fungus. However, we could find no correlation between resistance and glycoalkaloid content of tubers. Our results suggest a major role of phytoalexins, phenolics and glycoalkaloids in the complex mechanisms of field resistance.  相似文献

Summary The resistance of ten potato cultivars (Agria, Ajax, Désirée, Liseta, Kennebec, Majestic. Monalisa, Prima, Spunta and Tonda di Berlino) toPhytophthora infestans was analyzed in vitro using 8 fungal strains. An assay based on electrolyte leakage was used for screening leaves and tuber tissues with fungal culture filtrates. With almost all cultivars the resistance of leaves did not correlate with the resistance of tubers. Cv. Ajax appeared the least susceptible in both leaf and tuber tests, while the cv. Summary Potato (cv. Bintje) was transformed with a gene encoding an oxalate oxidase from wheat under the control of the CaMV35S promoter. Transgenic potato plants produced high constitutive levels of H2O2 as visualized by 4-chloro-l-naphtol staining. The resistance of these plants was tested againstPhytophthora infestans. An increased level of resistance to the disease was marked by a reduced number of lesions as well as by a decreased number of sporangia formed per lesion. In addition, oxalate oxidase overexpressing plants also exhibited improved resistance toStreptomyces reticuliscabiei, the causal agent of netted scab. Increased expression of oxalate oxidase had no effect on the interaction withErwinia carotovora. These experiments show that overexpression of oxalate oxidase represents a potentially interesting approach for protection of potato to pathogens.  相似文献

In an experimental breeding scheme to improve late blight (Phytophthora infestans) and white potato cyst nematode (Globodera pallida) resistance of tetraploid potato over three generations of crossing and selection, 15 clones survived the final selection, and these were derived from 15 great-grandparents. There was no direct selection for resistance to Potato virus Y (PVY), but 14 out of the 15 great-grandparents were resistant to PVY and three had extreme resistance. Thirteen of the 15 descendants had PVY resistance and one extreme resistance. This was within the range expected for a random (unselected) sample from the genotypes of the great-grandparents. Hence, we found no evidence for any positive or negative association between PVY resistance and the attributes selected. The conclusion is that laborious selection is not required in every generation when many parents have PVY resistance, including some with more than one copy of a PVY resistance gene or resistance at more than one locus. However, in the future, determining the major virus resistance genes present in potential parents in each generation using diagnostic molecular markers would prevent susceptible × susceptible crosses being made and maximise the number of resistant × resistant ones.  相似文献

Summary A comparison of reactions of potato cultivars toS. endobioticum indicated that the degree of susceptibility observed in laboratory tests correlated well with the reaction of cultivars in field tests. Some cultivars found to be less resistant (Resistance Grade 2) in laboratory tests produced winter spores in the field test, and 10% of those derived from cv. Ausonia germinated in distilled water. Compost from the field plot was estimated to contain 14–18 apparently viable spores/g. and this resulted in 100% infection of the highly susceptible cv. Arran Chief. The implications of these findings for descheduling sites previously scheduled due to wart infestation are discussed.  相似文献

Summary Twenty seven cultivars of potato were screened for common scab grown in a commercial field in two successive years (1996–97 and 1997–98). Eight cultivars were least susceptible and the others ranged from medium susceptible to very highly susceptible. None of the cultivars was resistant. Summary Greenhouse experiments on the effects of white potato cyst nematode infestations (Globodera pallida Stone) on initial growth and development of a series of potato cultivars are compared with the results of field experiments on sandy and sandy-peat soils on the effect of nematode density on tuber yield. A simple greenhouse test, assessing root growth response to potato cyst nematode infection, provided a good insight into a cultivar's tolerance performance in the field early in the growing season. As a very limited number of plants is needed for the greenhouse test, screening for tolerance can be conducted in the early stages of a breeding programme.  相似文献

The goal of these studies was to evaluate the influence of tuber age on the expression of resistance to Phytophthora infestans in five selected potato genotypes (three cultivars and two clones) varying in tuber resistance and maturity type, and also to select the best time for testing potato tubers for resistance to blight. Resistance was evaluated by inoculating whole tubers at 13 different times as they progressively aged during growth and storage. Tuber age was expressed as weeks after planting. Tubers were rated from week 13 (late July) until week 43 (late February) for tubers planted on 27–28 April, over three consecutive seasons (2001–2003). Analysis of variance performed on tuber resistance data showed significant effects of genotype, year and tuber age. Significant contributions of genotype × year, genotype × tuber age, year × tuber age and year × genotype × tuber age interactions were also detected. A slight increase in tuber resistance with tuber age was observed for cvs Bzura, Sokół and Irga, while the opposite trend was observed for the clones 97-A-63 and DG 92-227. Changes in tuber resistance with age of tested genotypes were not related to their maturity type. The period of most stable expression of tuber resistance was observed when tubers were tested between the 16th and 28th week after planting.  相似文献

Summary Potato cultivars were grown in a glasshouse and plants periodically transferred to a growth chamber. When the ambient temperature in the growth chamber was raised from 15 to 40°C in steps of 5°C per hour, net photosynthetic rate decreased at temperatures above 20°C. At 40°C the rate was 37% of the rate at 20°C. A greater decrease in net photosynthetic rate occurred with plants of the cultivar Up-to-Date than with cultivars R100 and BP13. Low values of leaf diffusive resistance were recorded and changes in photosynthetic rate could not be explained by changes in this factor. When a constant ambient air temperature of 20°C was maintained while soil temperature was increased, net photosynthetic rate decreased.  相似文献

Summary The acquisition of potato leafroll virus (PLRV) byMyzus persicae nymphs from the top leaves of potato plants was studied throughout a growing season in relation to the antigen titre in those leaves and the feeding behaviour of the aphid. Secondarily-infected plants of eight potato genotypes with different levels of field resistance served as virus sources. Early in the growing season, plants were efficient sources for virus acquisition. The amount of viral antigen detected inM. persicae nymphs fed on the top leaves was strongly correlated with the titres of viral antigen in these leaves. Virus acquisition from the top leaves of older potato plants was markedly impaired and could not be correlated with their virus titre. With increasing age of the potato plants and the development of virus symptoms, the virus titre in the leaves declined and the initial weak correlation between the virus titre and field resistance ratings disappeared. Thus, screening secondarily-infected potato plants for field resistance to PLRV based on the concentration of viral antigen in leaves or in aphids fed on them should be avoided later in the growing season. The feeding rate ofM. persicae, measured by the number of honeydew droplets excreted, did not account for the reduced uptake of virus from older plants since it was not influenced by the age of the plant. Throughout the growing season, the feeding rate ofM. persicae nymphs on PLRV-infected plants was higher on genotypes with low levels of field resistance to PLRV than on genotypes with high ones.  相似文献

Summary Optimum storage conditions to identify resistance to silver scurf among commercially grown potato cultivars were determined in a series of experiments. Inoculation of field-grown tubers with a conidial suspension ofH. solani (concentration 104 conidia per ml of water) and incubation at 15°C with 95% RH for 1 month followed by 2 months at 85% RH produced the greatest differences in disease severity between potato cultivars. Results with glasshouse-grown tubers were comparable with those from field-grown tubers, indicating that the test could be used to identify resistance in wild species of potato and to understand the inheritance of this resistance.  相似文献

Summary Synergism between fungicides could be used to reduce the application dosage. Synergism between cymoxanil and mancozeb was studied using potted potato plants under field conditions. In three experiments the preventive efficacy of the fungicides and fungicide mixtures in the treatments were studied using a bio-assay. Potato plants were sprayed with the fungicides or fungicide mixtures. Leaves were picked from the potato plants with regular intervals after fungicide application. The efficacy of the fungicides was studied in a bioassay by inoculating the leaves withPhytophthora infestans zoosporangia. The Abbot method was used to show synergistic effects of the cymoxanil/mancozeb mixtures tested. In two of three experiments synergy of the fungicide mixtures was found.  相似文献

Summary Fourteen genotypes of potato were assessed for resistance toAlternaria solani Sor., both in the glasshouse at the Scottish Crop Research Institute in Scotland and in the field in the Negev, Israel. Glasshouse assessments agreed well with field results, identifying the most resistant and susceptible genotypes. The effects on the glasshouse test of plant age, inoculum concentration, and length of time plants were kept damp after inoculation, were examined. The best agreement with field results was shown by plants inoculated at flowering, and the best discrimination between genotypes was achieved by keeping the foliage damp for 24 h after inoculating with a spore suspension containing 2.5×103 spores/ml.  相似文献
