Prolipid a known mixture of herbs is used as a plasma lipid lowering medicine. No side effects were registered. However, the bioactive substances of Prolipid were not investigated. Therefore in this investigation Prolipids bioactive compounds and antioxidant activity were studied. The contents of polyphenols and flavonoids were 19.87 ± 2.09 and 3.09 ± 0.31 mg gallic acid equivalent GAE/g DW and 2.09 ± 0.24 and 0.57 ± 0.05 mg catechin equivalent CE/g DW in water and methanol fractions, respectively. Anthocyanins (0.02 ± 0.001 mg/g DW) and flavanols (7.58 ± 0.81 μg CE/g DW) were found only in water fraction. The antioxidant activity of Prolipid, as determined by four different antioxidant assays [ferric-reducing/antioxidant power (FRAP); cupric reducing antioxidant capacity (CUPRAC); trolox equivalent antioxidant capacity (TEAC); 1, 1-diphenyl-2-picrylhydrazyl radical (DPPH)], was higher in water than in methanol fraction. The correlation coefficients between polyphenols, flavonoids and antioxidant activities of Prolipid water extracts with TEAC were 0.97 and 0.90, respectively. It can be concluded that the content of polyphenol compounds in Prolipid is very high and they are the main contributors to Prolipids overall antioxidant activity. Prolipid is widely used in human treatment without known side effects on patients and is comparable to other medicinal plants, and as a strong antioxidant mixture could be used as a supplement to known atherosclerosis preventing diets.  相似文献

The bioactivity of two kiwifruit’s cultivars growing under organic and conventional conditions were studied and compared. The bioactive compounds were extracted with water and ethanol using similar conditions which are applied in pharmaceutical applications and for daily fruit consumption such as tea drink. Antioxidant radical scavenging assays [ferric-reducing/antioxidant power (FRAP); cupric reducing antioxidant capacity (CUPRAC); 2, 2-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS)], fourier transform infrared (FT-IR) and ultraviolet spectroscopy, two (2D-FL) and three-dimensional (3D-FL) fluorometry were used for the detection of biologically active metabolites derived from kiwifruits (total phenols, flavonoids, chlorophylls, carotenoids and ascorbic acid). The correlation between the total phenol content (TPC) and other bioactive compounds, and their total antioxidant capacities (TAC) was calculated for studied kiwifruit’s extracts. The interaction between drugs and human serum albumin (HSA) plays an important role in the distribution and metabolism of drugs. The properties of kiwifruit’s phenol extracts showed their ability to quench HSA, forming the complexes similar to the ones between the proteins and pure flavonoids such as quercetin. The cultivar ‘Bidan’ exhibited significantly higher TAC than the classic ‘Hayward’. In conclusion, for the first time ‘Bidan’ organic kiwifruit was analyzed and compared with widely consumed ‘Hayward’, using its bioactive and fluorescence properties. The influence of physiologically active kiwifruit’s compounds on human health, through our investigations in vitro and scientifically proven information, was explained. Relatively high content of bioactive compounds, high antioxidant and fluorescence properties of kiwifruit justify its use as a source of valuable antioxidants.  相似文献

Substantial evidence exists to support the hypothesis that high fruit and vegetable consumption, rich in antioxidants, can reduce the incidence of several disease states. The aim of this study was to compare the results obtained by six spectrophotometric biochemical methods including the ferric reducing antioxidant power (FRAP), 2, 2-diphenyl-1-picryhydrazyl (DPPH?), 2,2-azinobis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS??), copper (II) reducing capacity (CUPRAC) and Cerium (IV) reducing antioxidant capacity (CERAC) assays as well as Folin-Ciocalteu method (FC) for the measurement of total antioxidant capacity (TAC) and total polyphenols (TP) in different commercially available vegetable juices. The aim of this investigation was to find a proper harvesting period and establishing fern number, which effects the spear yield, bioactive compounds and antioxidant activities of Asparagus officinalis L. Spears were harvested at 2, 4, and 6 weeks after sprouting. Control for comparison was used without harvest. Spears and total yield increased with prolonged spear harvest period. In harvest of 6 weeks long optimum spear yield was the highest and fern numbers were 5?~?8. Bioactive compounds (polyphenols, flavonoids, flavanols, tannins and ascorbic acid) and the levels of antioxidant activities by ferric-reducing/antioxidant power (FRAP) and cupric reducing antioxidant capacity (CUPRAC) assays in asparagus ethanol extracts significantly differed in the investigated samples and were the highest at 6 weeks harvest period (P?<?0.05). The first and the second segments from the tip significantly increased with the increase of catalase (CAT). It was interesting to investigate in vitro how human serum albumin (HSA) interacts with polyphenols extracted from investigated vegetables. Therefore the functional properties of asparagus were studied by the interaction of polyphenol ethanol extracts with HSA, using 3D- FL. In conclusion, antioxidant status (bioactive compounds, binding and antioxidant activities) improved with the harvesting period and the first segment from spear tip. Appropriate harvesting is effective for higher asparagus yield and its bioactivity.  相似文献

苦丁茶提取物多酚含量与抗氧化活性的测定   总被引:5,自引:1,他引:4  
首先用不同的有机溶剂分部萃取苦丁茶(Ilex kudincha C.J.Tseng)热水提取物(粗提物),得到氯仿萃取物、乙酸乙酯萃取物、正丁醇萃取物及萃取剩余物,然后采用Folin-Ciocalteu比色法测定粗提物和各萃取物的多酚含量,同时应用DPPH法、TEAC法和FRAP法分别测定粗提物和各萃取物的自由基清除能力和还原Fe3+能力。结果表明,苦丁茶提取物具有较高的多酚含量和较强的抗氧化能力;DPPH法和FRAP法测定各提取物抗氧化能力的结果为乙酸乙酯萃取物>正丁醇萃取物>粗提物>萃取剩余物>氯仿萃取物,TEAC法测定结果为乙酸乙酯萃取物>正丁醇萃取物>粗提物>氯仿萃取物>萃取剩余物;多酚含量与抗氧化能力之间、所用抗氧化测定方法之间均存在较好的相关性。  相似文献

为了进一步深入了解五指山水满茶的营养价值和保健功效,本研究对五指山水满茶及其他6种茶(普洱茶、白毛茶、白沙绿茶、西湖龙井、兰贵人和碧螺春)的提取物的抗氧化活性及其茶多酚、总黄酮含量进行了比较分析。结果表明:7种茶的75%乙醇提取液的抗氧化效果最好,且提取液的抗氧化活性与样品浓度成正比关系,整体热稳定性比较好。在7种茶中,水满茶清除ABTS自由基、超氧自由基和羟自由基的能力最强,且在测试浓度范围内清除能力均与样品浓度表现出量效关系,但水满茶清除DPPH自由基的能力最弱;还原能力是白毛茶最强,其次是白沙绿茶和水满茶。7种茶的总黄酮和茶多酚含量分别在3.06%~5.4%和14.6%~18.1%之间,其中,水满茶的茶多酚含量最低,但总黄酮含量却最高。因此,五指山水满茶具有较强的抗氧化活性,较高的总黄酮含量,具有较高的营养价值和保健功效。   相似文献

In this study, we analyzed eight phenolic compounds (tyrosol, hydroxytyrosol, oleuropein, pinoresinol, and caffeic, ferulic, vanillic, and p-coumaric acid) in 55 mono- and multivarietal extra virgin olive oil samples by high performance liquid chromatography (HPLC) coupled to a coulometric electrochemical array detector (ECD). The phenolic profile of olive oil samples differed depending on the geographical origin and olive variety. The total reducing capacity (total phenolics) of olive oils ranged from about 40 to 530 mg gallic acid equivalents/kg oil. Tyrosol, hydroxytyrosol and pinoresinol were the most abundant phenolic compounds in olive oils. The antioxidant capacity of the olive oil extracts was determined by ferric reducing antioxidant power (FRAP) and Trolox equivalent antioxidant capacity (TEAC) assays. Total reducing capacity was significatly correlated with FRAP (R 2 ?=?0.91, p?<?0.001) and TEAC (R 2 ?=?0.92, p?<?0.001) values. Total reducing capacity, TEAC and FRAP values were significantly correlated with tyrosol, hydroxytyrosol as well as oleuropein concentrations. Hydroxytyrosol, comprising over 40 % of total olive oil phenolics, mainly contributed to the antioxidant activity of olive oils. The present study provides a comprehensive database of polyphenols in olive oils from 9 different countries and four continents.  相似文献

This study aimed to evaluate the effect of autohydrolysis temperature of corn residues in the antioxidant activity of the phenolic compounds extracted from the liquid phase. The treatments were carried out at 160, 180, 190 and 200 °C for 30 min in a pressurized batch reactor. Two different methods for phenolic compounds extraction from the autohydrolysis liquors were investigated. For that purpose, solvent extraction with ethyl acetate and acidic precipitation were performed for phenolic compounds recovery. These methods have been compared in terms of extraction yield, physicochemical properties of obtained polyphenols (characterization by Fourier Transform Infrared Spectroscopy, Thermogravimetric Analysis and Gel Permeation Chromatography), total phenolic content, total antioxidant capacity and Trolox equivalent antioxidant capacity (TEAC) values, measured in DPPH (2,2-diphenyl-1-picrylhydrazyl) test system. The maximum phenolic contents ranged from 6.04 mg GAE/100 mg extract in acidic precipitated samples to 16.45 mg GAE/100 mg extract in ethyl acetate soluble fractions. The results indicated that the ethyl acetate fractions possessed the highest antioxidant activity, reaching after 30-60 min the same capacity reported for the reference synthetic antioxidants (Trolox).  相似文献

The present study evaluates variations among flaxseed varities in terms of fatty acid composition, phytochemical profile, and antioxidant activity determined by oxygen radical absorbance capacity (ORAC), 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and ferrous ion reducing antioxidant power (FRAP) assays. Significant variations in fatty acid composition, phenolic acids and lignan were observed in flaxseed varieties from different countries. Among them, unsaturated fatty acids accounted over 4/5 of total fatty acid content. The highest ratio of linolenic acid of total fatty acid was observed in USPEA, whereas the lowest one was found in Yexiao. USPEA showed the highest content of total phenolics, as well as flaxseed lignan. In general, total phenolics appeared to be the main contributors of antioxidant capacity of flaxseed, which presented significant positive correlation. Our study revealed that both cultivar and origin of seed significantly affected fatty acid composition, phenolic acids, lignans and subsequent antioxidant activity of flaxseed. These results provided new aspects of breeding resources of flaxseed cultivars by presenting their quality specification and possible commercial value.  相似文献

In this study polyphenol content, antioxidant activity, lipoxygenase (LOX) and Xanthine Oxidase (XO) inhibitory effects of n-hexane, dichloromethane, ethyl acetate and n-butanol fractions of aqueous acetone extracts from S. alba L., S. acuta Burn f and Cienfuegosia digitata Cav. were investigated. The total phenolics, flavonoids, flavonols and total tannins were determined by spectrophotometric methods using Folin-ciocalteu, AlCl3 reagents and tannic acid, respectively. The antioxidant potential was evaluated using three methods: inhibition of free radical 2,2-diphenyl-1-picrylhydramzyl (DPPH), ABTS radical cation decolorization assay and Iron (III) to iron (II) reduction activity (FRAP). For enzymatic activity, lipoxygenase and xanthine oxidase inhibitory activities were used. This study shows a relationship between polyphenol contents, antioxidant and enzymatic activities. Present results showed that ethyl acetate and dichloromethane fractions elicit the highest polyphenol content, antioxidant and enzymatic activities.  相似文献

选择由Benzie和Strain建立的铁离子还原法(ferric reducing /antioxidant power assay,FRAP)比较不同EGCG含量茶多酚的总抗氧化能力强弱以及茶多酚与其他天然抗氧化物质的协同抗氧化作用,测定结果表明EGCG在儿茶素中比例为40%~50%时抗氧化能力最强。抗氧化物质之间的协同抗氧化作用与抗氧化物质本身的抗氧化能力,抗氧化物质有效浓度的高低等因素有关。抗氧化物质之间的协同作用,在抗氧化物质本身有效浓度较低时协同作用也较低,随着有效浓度增加,协同抗氧化作用增加,达到一个峰值后,随着有效浓度的继续增加,协同作用会逐渐减弱。  相似文献

Wholegrain consumption is associated with several health benefits, in contrast to the consumption of refined grains. This can partly be related to the antioxidant compounds in the outer parts of the grain kernel. The bioaccessibility of these antioxidant compounds from the wholegrain matrix during gastrointestinal digestion is crucial for their absorption and bioavailability. In the current study, the bioaccessible compounds from aleurone, bran and flour were obtained from a dynamic in vitro model of the upper gastrointestinal tract. They were collected at 1 h time intervals to assess their antioxidant capacity (TEAC assay) and also their anti-inflammatory effect (TNF-α reduction in U937 macrophages stimulated with LPS). The bioaccessible compounds from aleurone had the highest antioxidant capacity and provided a prolonged anti-inflammatory effect, shown by the TNF-α reduction of a relatively late time-interval (3–4 h after start of digestion). The contribution of ferulic acid to those effects was minor due to its low bioaccessibility. Aleurone seems a promising wheat fraction for cereal products with a healthy added value.  相似文献

为比较不同品种荔枝果皮酚类物质含量、组成及其抗氧化活性差异,用80%甲醇分别提取6个品种荔枝果皮中的酚类物质,测定其总酚和总黄酮含量。采用铁离子还原能力(FRAP)和自由基离子清除能力(ABTS)等方法评价其抗氧化活性。通过高效液相色谱(HPLC)鉴定不同的单体酚并分析其在不同品种荔枝果皮之间的差异。结果表明,不同品种荔枝果皮的酚类物质含量为28.69~68.48 mg/g,不同品种荔枝果皮的酚类物质含量和抗氧化能力存在显著的差异(P<0.05)。酚类物质的含量、FRAP和ABTS抗氧化活性能力均以‘荔枝王’为最高,分别为68.48 mg/g546.31 μmol/g和511.25 μmol/g。液相色谱结果鉴定出‘糯米糍’中的8种单体酚,而含量相对较高的是A型原花青素三聚体和原花青素A2。研究结果可为荔枝加工废弃物荔枝果皮的开发利用提供理论依据。  相似文献

采用高效液相色谱法(HPLC)对 4 种黔引迷迭香的多酚成分进行定性和定量分析,并利用 DPPH、ABTS自由基清除能力和铁离子还原能力(FRAP)3 种方法评价多酚成分的抗氧化能力。结果显示 4 种材料均富含类黄酮、酚酸和二萜酚等多酚类化合物,总酚含量介于 4 345.5~5 673.0 mg/100 g 。其中槲皮素、迷迭香酸、阿魏酸、绿原酸和鼠尾草酸是迷迭香最主要的酚类物质。黔引迷迭香 4 号的抗氧化活性最高,利用 DPPH、ABTS 和 FRAP等 3 种方法检测的结果依次为 184.4、290.0、155.6 μmol/g。相关性分析表明,总酚与总抗氧化活性能力显著相关,迷迭香酸对总抗氧化活性能力贡献最大,其次为鼠尾草酸、槲皮素和水杨酸。本研究对于了解黔引迷迭香抗氧化活性成分,以及资源的进一步开发利用具有重要指导意义。  相似文献

Sorghum is becoming more widely recognised around the world as a valuable crop for its polyphenol antioxidant health-promoting properties and adaptation to harsh environments. The antioxidant capacity of diverse polyphenols in sorghum grain can vary with climatic conditions and genotype. To explore this further, the potential role of UV radiation on the profile and concentration of polyphenols was investigated in six diverse sorghum genotypes in a controlled environment facility using natural UV and visible radiation under either UV-transmitting or UV-blocking treatments. The polyphenol content and antioxidant activity were significantly reduced under the UV-blocking treatment, with contents of individual polyphenols differing among the genotypes. This information is valuable for sorghum breeders enabling them to select genotypes and UV growth conditions to produce sorghum grain with target levels of polyphenols and antioxidant capacity for human foods to meet the nutritional and health needs of different consumers.  相似文献

Phenolic contents of barley and malt extracts and their corresponding antioxidant activities were investigated using a chromatographic online antioxidant detection system. Ethyl acetate extracts of barley and malt were separated using reverse phase HPLC and compounds eluting from the column were submitted to two UV–visible detections: one for the phenolic compounds; and the other for the reduced form of the radical cation 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS•+) after the compounds were allowed to react online with it. Prodelphinidin B3 and procyanidin B3 were identified as two major contributors in the antioxidant activity of barley, in addition to catechin. Malting had a dramatic impact on these three compounds by resulting in a sharp decrease in their detected amounts and the associated antioxidant activities. Two other antioxidants, ferulic and sinapic acids, showed a better ability to withstand not only malting but also brewing steps. As for the overall phenolic content and antioxidant capacity, the study showed that malting allowed a better release and/or extraction of phenolic compounds, while the first brewing step caused the most significant damage by drastically decreasing the total polyphenols and their activity. Total phenolics, flavonoid contents and antioxidant capacity from a wide collection of rice germplasm were measured, and their relations to grain color, grain size and 100-grain weight were investigated. Highly significant genotypic differences were observed in total phenolics, flavonoid contents and 2,2-azino-bis-(3-ehylbenzothiazoline-6-sulphonic acid) diammonium salt (ABTS) radical cation antioxidant capacity. They displayed an increasing order in the white rice, red rice and black rice, yet several white rice had higher phenolics and flavonoids contents than the red rice. Significant positive pair-wise correlations were found among the phenolics, flavonoid contents and antioxidant capacity, and the coefficient between the phenolic contents and antioxidant capacity was extremely high (r = 0.96). Among all rice accessions, the grain color parameters had negative correlations with the phenolics, flavonoid contents and antioxidant capacity (p < 0.001). The negative correlation between a* and antioxidant capacity, and the positive correlation between H° and antioxidant capacity were consistent within the respective white rice and red rice groups. Flavonoid contents had positive correlation with grain length and length to width ratio, and had negative correlation with the 100-grain weight among all rice accessions. It was also found that 100-grain weight still had negative correlations with phenolics, flavonoid contents and antioxidant capacity within the white rice genotypes. These relationships may serve as indexes to indirectly select breeding lines high in the phenolics, flavonoids and antioxidant capacity. Principal component analysis including the information for phenolics, flavonoids, antioxidant capacity, grain color parameters, grain size and 100-grain weight extracted five principal components that explained 83.7% of the total variances. The results of this study may provide new opportunities for rice breeders and eventually commercial rice growers to promote the production of rice with enhanced nutritional quality.  相似文献

以铁皮石斛为原料,利用乳酸菌发酵生产风味饮料,对铁皮石斛汁发酵前后多糖、石斛碱、总酚、总黄酮含量及抗氧化能力变化情况进行研究。结果表明,植物乳杆菌对石斛多糖的消耗最高,比原汁减少了41.63%。植物乳杆菌与保加利亚乳杆菌按1∶1混合发酵对石斛碱的消耗最高,比原汁减少了4.92%。经乳酸菌发酵,总酚、总黄酮含量均比发酵前有提高,其中以植物乳杆菌与保加利亚乳杆菌按比例1∶1混合发酵提高幅度最大,总酚与总黄酮含量分别提高了24.17%和31.87%。铁皮石斛汁发酵后对羟基自由基、DPPH自由基和超氧阴离子自由基的清除能力均比发酵前有所增强。本研究说明,利用乳酸菌发酵可提高铁皮石斛活性物质的溶出率,从而提高铁皮石斛乳酸菌饮品抗氧化功能。  相似文献

大叶冬青苦丁茶经85%乙醇溶液脱脂、热水提取、S-8大孔树脂脱色、醇沉、干燥,得到大叶冬青苦丁茶粗多糖。粗ILPS经DEAE-52纤维素阴离子交换层析柱分离,得到4个多糖组分ILPS-1,ILPS-2,ILPS-3和ILPS-4。采用化学法分别测定了粗多糖及其纯化组分的体外清除自由基(DPPH.自由基,O2^-.自由基,.OH自由基)能力、还原能力以及螯合金属离子能力。结果表明,大叶冬青苦丁茶多糖具有较强的体外抗氧化活性,并且抗氧化能力与多糖浓度之间存在良好的相关性。  相似文献

Many analytical methods for antioxidant determination in foodstuffs and raw materials based on various principles have been published so far. However, not all of them are applicable to barley and malt. The results of total antioxidant capacity (TEAC) of barley and malt obtained with methods based on ability to eliminate radicals of 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) and 2,2-diphenyl-1-(2,4,6-trinitrophenyl) hydrazyl (DPPH) were compared and statistically evaluated. The average TEAC of barley determined using ABTS and DPPH ranged from 2.1 to 2.5 μmol g−1 and from 1.2 to 1.7 μmol g−1, respectively. The TEAC ranges in malt were 2.7–3.0 μmol g−1 (ABTS) and 1.8–2.6 μmol g−1 (DPPH). TEAC of barley and malt were affected by the weather conditions (year), variety and application of Zn2+ fertilizer. The ABTS and the DPPH methods represent an effective tool for the assessment of antioxidant capacity of spring barley and malt and the factors having an impact on it.  相似文献
