AIM: To observe the effects of Zhenwu decoction on the expression of podocin and nephrin in podocytes of adriamycin nephropathy (AN) rats, and to clarify the mechanism of Zhenwu decoction in decreasing adriamycin-induced proteinuria in rats. METHODS:Biochemical assay and pathological observation (HE staining, Masson trichrome staining and transmission electron microscopy) were used to evaluate the effects of Zhenwu decoction on renal function, pathological morphology and hydroxyproline (Hyp) content in AN rats with renal fibrosis. Western blotting was used to observe the effect of Zhenwu decoction on the expression of podocin and nephrin which are marker proteins in podocytes. RESULTS:In model group, the levels of urinary total protein (TP), blood urea nitrogen (BUN), serum creatinine (SCr) and renal Hyp were significantly increased, and the clearance of creatinine (CCr) level was decreased (P<0.05). The expression of podocin and nephrin was significantly decreased (P<0.05). Atrophic renal tubules, thickened basement membranes, fusion of foot processes, concentrating renal glomerules, expansion of some renal tubules, degeneration of renal tubular epithelial cells, protein casts, and proliferation of fibroblasts and infiltration of inflammatory cells in renal interstitium were also observed. After treatment with drugs (Zhenwu decoction and valsartan), the above-mentioned parameters were significantly changed. TP, BUN, SCr and Hyp were down-regulated to different levels, and CCr was significantly up-regulated (P<0.05). The expression of podocin and nephrin was up-regulated by treatment with Zhenwu decoction, and renal histological changes were alleviated compared with model group. CONCLUSION: Zhenwu decoction can reduce Hyp content in kidney tissue, alleviate kidney histological changes, and improve renal function in AN rats. It might protect kidney against adriamycin-induced proteinuria via increasing the expression of podocin and nephrin in podocytes.  相似文献

AIM: To investigate the effect of sodium valproate (VPA) on bleomycin-induced pulmonary fibrosis (PF) and its mechanism. METHODS: SD rats (n=42) were randomly divided into model group and treatment group. Bleomycin at dose of 5 mg/kg was intratracheally injected to establish a rat PF model. The rats in treatment group were given normal saline (NS, 0.5 mL/d), VPA (300 mg·kg-1·d-1) or dexamethasone (DEX, 0.6 mg·kg-1·d-1) via intraperitoneal injection from the 14th day to the 28th day after modeling. The rats in model group were sacrificed on 0 d, 14 day and 28 d after modeling . The rats in treatment group were killed at 14th day after treatment. The effects of VPA on PF were evaluated by HE and Masson staining. The hydroxyproline (HYP) content in the rat lung tissues was detected, and the expression of α-smooth muscle actin (α-SMA) and E-cadherin was determined by Western blotting. RESULTS: HE staining showed that the alveolar structure and interstitial morphology in VPA group were better than those in NS group and DEX group. The level of collagen in VPA group was significantly lower than that in DEX group and NS group by determining the HYP content and Masson staining. VPA reduced the expression of α-SMA, a mesenchymal marker protein of PF, while increased the expression of epithelial marker protein E-cadherin. CONCLUSION: VPA reduces collagen content and distribution, and up-regulates the expression of the epithelial marker protein E-cadherin, while down-regulates mesenchymal marker protein α-SMA, thereby preventing the rat lung tissues from bleomycin-induced PF.  相似文献

AIM:To investigate the role of hypoxia-inducible factor-1α (HIF-1α) stable expression in myocardial inflammatory injury induced by ischemia and reperfusion (I/R) in rats. METHODS:Male Wistar rats were randomly divided into 4 groups:sham operation (sham) group, I/R group, HIF-1α stabilizer dimethyloxalyl glycine (DMOG)+I/R group and HIF-1α inhibitor YC-1+I/R group. The protein expression of myocardial Toll-like receptor 4 (TLR4) and nuclear factor-κB (NF-κB) was determined by Western blot. The mRNA levels of interleukin (IL)-1β, tumor necrosis factor-α (TNF-α), IL-6, TLR4 and NF-κB were detected by real-time PCR. The myeloperoxidase (MPO) activity in the myocardial tissues was measured. HE staining was used to observe the infiltration of inflammatory cells. RESULTS:HIF-1α decreased the infiltration of inflammatory cells, the MPO activity, and the mRNA levels of inflammatory factors IL-1β, IL-6 and TNF-α in the myocardial tissues. HIF-1α also reduced the expression of TLR4 and NF-κB at mRNA and protein levels (P<0.05). CONCLUSION:The stable expression of HIF-1α has an anti-inflammatory effect on the myocardial tissues after I/R injury in rats. The mechanism may be related to the inhibition of TLR4/NF-κB signaling pathway.  相似文献

AIM: To examine the effects of silencing of plasminogen activator inhibitor-1 (PAI-1) expression by small interfering RNA (siRNA) on bleomycin (BLM)-induced rat pulmonary fibrosis. METHODS: Total 72 Wistar rats were divided into 4 groups: control, BLM, BLM+non-specific siRNA (BLM+N), and BLM+ PAI-1 siRNA (BLM+P). Pulmonary fibrosis was induced by intratracheal injection of BLM (5 mg/kg), whereas equal volume of normal saline was used in control group. After the administration of BLM or normal saline, the rats were treated with tracheal injection of PAI-1-siRNA (7.5 nmol/0.2 mL per rat) in BLM+P group, non-specific siRNA (7.5 nmol/0.2 mL per rat) in BLM+N group, and 0.2 mL normal saline in BLM group and control group, twice a week, 8 times in 28 d. On day 7, 14, and 28, the rats (n=6 at each time point) were sacrificed. The bronchoalveolar lavage fluid (BALF) from the left lung was harvested to examine the activity of PAI-1. The mRNA expression of collagen type Ⅲ, α-smooth muscle actin (α-SMA) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in the middle lobe of the right lung was detected by RT-PCR. RESULTS: PAI-1 activity and the expression of collagen type Ⅲ, α-SMA and TIMP-1 were increased in BLM group on day 7, 14 and 28. Intratracheal injection of PAI-1 siRNA twice a week continuously reduced PAI-1 activity in the BALF (P<0.05),and decreased the expression of collagen type Ⅲ, α-SMA and TIMP-1 in the fibrotic lung tissues on day 7, 14 and 28. Statistical differences in the expression of collagen type Ⅲ, α-SMA and TIMP-1 between BLM+P group and BLM group at the same time point were observed. CONCLUSION: Intratracheal injection of PAI-1 siRNA twice a week continuously inhibits the expression of PAI-1. PAI-1 siRNA ameliorates BLM-induced pulmonary fibrosis by down-regulation of TIMP-1 expression.  相似文献

AIM: To observe the pathologic changes of cardiomyopathy in type 2 diabetic rats and the therapeutic effect of Xuefuzhuyu decoction.METHODS: The diabetic model was established by feeding the animals with high-fat diet and injecting a middle dose of streptozotocin (50 mg/kg) intraperitoneally in 42 Wistar male rats. After 8 weeks, the damage of the heart in the model animals was detected by electrocardiogram and echocardiography, and the serum level of glucose, total cholesterol and triglyceride were determined by the methods of clinical chemistry. The content of collagen was quantified by Masson staining. The apoptosis of cardiomyocytes was measured by TUNEL apoptosis kit. The structures of myocardial damage were observed under light and electronic microscopes.RESULTS: (1) Compared with normal group at the same time points, the contents of serum glucose, triglyceride and cholesterol in model group increased (P<0.05). At the 11th and 14th weeks, the thickness of LVDS was significantly increased (P<0.01), the structure of myocardial tissues was severely damaged and collagen fiber content increased obviously (P<0.01). The cell apoptosis was also increased. (2) Compared with control group at the same time points, the contents of serum glucose, cholesterol and triglyceride in Xuefuzhuyu decoction group significantly decreased (P<0.05). The thickness of LVDS at the 11th and 14th weeks was decreased (P<0.05) and LVM at the 14th week became significantly thinner (P<0.01). The damage of the myocardium and subcellular structure was slighter and the content of collagen was lower than that in control group (P<0.05). The cell apoptosis was also attenuated.CONCLUSION: The levels of blood glucose, total cholesterol and triglyceride and the content of collagen fibers increase when diabetic cardiomyopathy develops, with more cell apoptosis and severe damage in the cardiac structure. Xuefuzhuyu decoction decreases the level of blood lipid in diabetic cardiomyopathy, alleviates the pathological changes of myocardial fibrosis and delays the progression of diabetic cardiomyopathy.  相似文献

AIM: To explore the protective effects of luteolin on the diabetic kidneys. METHODS: Male Sprague-Dawley rats were randomly divided into 5 groups: normal control group, diabetic model group and the groups of diabetic rats treated with luteolin at a low dose, a middle dose and a high dose. The diabetic model was induced by a single intraperitoneal injection of streptozotocin (STZ,65 mg/kg). Blood glucose, urine protein, the activity of superoxide dismutase and catalase in serum and kidney, and the content of malonaldehyde(MDA) in kidney were analyzed by biochemical methods. Western blotting was used to detect the protein expression of transforming growth factor-β1(TGF-β1) and plasminogen activator inhibitor-1(PAI-1) in the renal cortex. The morphological changes of the renal tissues were observed under microscope. RESULTS: Compared with diabetic model group, luteolin significantly reduced the level of blood glucose (P<0.01), the content of urine protein (P<0.01) and MDA (P<0.01) in the kidneys, and increased the activity of superoxide dismutase and catalase (P<0.01) in serum and kidneys in the diabetic rats. The protein levels of TGF-β1 and PAI-1 in the renal cortex were dramatically decreased as the rats were treated with luteolin. CONCLUSION: Luteolin may exert an important protective effect on diabetic kidneys by relieving oxidative stress and inhibiting the protein expression of TGF-β1 and PAI-1 in the renal tissues of STZ-induced diabetic rats.  相似文献

AIM: To assess the renal protective effect of the combination use of spirinolactone and cilazapril on streptozotocin(STZ)-induced diabetic rats with single nephrectomy. METHODS: Diabetic nephropathies were induced by intraperitoneal injection of STZ in the rats with single nephrectomy. The rats were randomly divided into 5 groups: normal control; diabetes; diabetes treated with spirinolactone; diabetes treated with cilazapril; diabetic rats treated with spirinolactone and cilazapril. The expression of NF-κB and PAI-1 in the glomeruli was detected by immunohistochemical staining. RT-PCR was performed to evaluate the mRNA expression of AT-1R. RESULTS: Increased 24 h urinary protein, decreased Ccr and the pathological injury of the renal tissues were improved by the treatment with either spirinolactone or cilazapril alone and further ameliorated by using the combination of the two drugs. The activity of NF-κB and PAI-1 was higher in the renal tissues of diabetic rats than that in control group, and further attenuated by the combination therapy in both cases (P<0.05). The over-expression of AT-1R mRNA observed in the diabetic rats was attenuated by treating with spirinolactone or cilazapril and further reduced by the combination use of the two drugs (P<0.05).CONCLUSION: The combination use of spirinolactone and cilazapril confers superiority over monotherapy on the effect of renal protection. The mechanism may be partly correlated with synergistic suppression of the increasing activity of NF-κB and PAI-1 as well as the over-expression of AT-1R mRNA in renal tissues.  相似文献

AIM: To observe the effect on Yunpiheluo decoction (YPHL) on renal injury in type 2 diabetic rats and to explore the mechanism from the perspective of Sirt1-AMPK-autophagy. METHODS: Male Zucker diabetic fatty (ZDF) rats (n=24) were randomly divided into model group, Sirt1 over-expression group and YPHL group, and fed with high-fat and high-sugar diet for 10 weeks. ZL rats were used as normal control and fed with normal diet for 10 weeks. After 10 weeks, urine and blood were collected for renal function detection. The rats were sacrificed and specimen was submitted. In addition, the mRNA expression of Sirt1 was analyzed by real-time PCR. The protein levels of Sirt1, AMPK, p-AMPK, LC3 and P62 in the renal tissues wene determined by Western blot. The renal pathological changes were observed under light microscope with HE and Masson staining. RESULTS: Compared with control group, fasting blood glucose (FBG), urinary protein (UP), urinary albumin (U-ALB) and serum creatinine (SCr) in model group were obviously increased (P<0.01). The mRNA expression of Sirt1 was decreased (P<0.05). The protein levels of SIRT1, AMPK, p-AMPK and LC3-II/-I were decreased (P<0.01), and P62 was increased (P<0.01). Glomerular focal fibrosis, focal renal tubular epithelial cell vacuolation, necrosis, shedding and atrophy, tubular type, and renal interstitial fibrosis were observed. Compared with model group, FBG was obviously decreased in Sirt1 over-expression group (P<0.01), but it showed no significant change in YPHL group (P>0.05). SCr and U-ALB were decreased (P<0.05), Sirt1 mRNA was increased (P<0.05), the protein levels of SIRT1, AMPK, p-AMPK and LC3-II/-I were increased (P<0.01), and P62 was decreased (P<0.01) in Sirt1 over-expression group and YPHL group. HE and Masson staining showed that the renal damage in Sir1 over-expression group and YPHL group was attenuated. CONCLUSION: Yunpiheluo decoction may protect the kidney by increasing the expression level of Sirt1, activating AMPK, and regulating autophagy.  相似文献

AIM: To study the preventive and curative roles of Danshensu (DA) in bleomycin (BLM)-induced pulmonary fibrosis in rats. METHODS: Pulmonary fibrosis was induced in SD rats by intratracheal instillation of BLM. The rats were intraperitoneally injected with dexamethasone (1 mg·kg-1·d-1, DXM group), DA (15 mg·kg-1·d-1, DA group), or physiological saline (2 mL·d-1, BLM group). Normal controls (NC group) received physiological saline both intratracheally and intraperitoneally. At the 28th day after modeling, the histological changes of the lungs were evaluated by hematoxylin-eosin (HE) and Masson’s trichrome staining. The protein levels of α-smooth muscle actin (α-SMA) in the lung tissues were detected by the method of immunohistochemistry. The mRNA expression of transforming growth factor beta 1 (TGF-β1), Smad3 and Smad7 was assessed by real-time fluorescence quantitative PCR. RESULTS: Compared with BLM group, the degree of inflammation and fibrosis of the lung in DA group was obviously reduced, and so was the expression of α-SMA in the lung tissues. The mRNA expression of TGF-β1 and Smad3 in the lung tissues of the rats decreased and the mRNA expression of Smad7 increased. CONCLUSION: DA alleviates BLM-induced pulmonary fibrosis in rats in the early stage by inhibiting the expression of TGF-β1/Smad3 and stimulating the expression of Smad7 in the lung tissues.  相似文献

AIM:To investigate the effect of platelet inhibitor from Agkistrodon halys venom (AHV-PI) on lipopolysaccharide (LPS) induced human umbilical vein endothelial cells (HUVECs) injury in vitro, and to explore its mechanism. METHODS:Cultured HUVECs were induced inflammatory injury by LPS (1 mg/L). The experiment was divided into blank control group, LPS group, AHV-PI group and AHV-PI+LPS group. The viability of HUVECs was measured by MTT assay. The morphological changes of HUVECs were observed under inverted microscope. The optimum concentration of AHV-PI at 5 mg/L was selected. Flow cytometry was used to detect apoptosis of HUVECs. Immunohistochemical method was used to observe the expression of tissue type plasminogen activator (t-PA) and plasminogen activator indhibitor-1 (PAI-1) of HUVECs. ELISA was used to detect the concentrations of intercellular adhesion molecule-1 (ICAM-1) and tissue factor (TF) in the supernatant. The activation and translocation of NF-κB subunit p65 were observed by immunofluorescence staining. RESULTS:The HUVECs were spindle shaped, the ratio of length to width was increased, the cells were fibroblast-like, and granular substance appeared in the cytoplasm in LPS group. The viability and morphological changes of HUVECs were not significantly affected as treated with AHV-PI at concentration of 0~5 mg/L, but the viability of HUVECs induced by LPS was inhibited and the morphological changes were alleviated. Compared with the blank control group, the levels of TF and ICAM-1 in the supernatant increased, and the expression of t-PA and PAI-1 in the HUVECs was decreased in LPS group. Compared with the LPS group, the contents of TF and ICAM-1 in the supernatant were significantly decreased, the expression of t-PA and PAI-1 in the HUVECs was increased and the expression of nucleus NF-κB p65 was decreased in AHV-PI+LPS group (P<0.05). CONCLUSION:AHV-PI reduces HUVECs damage. The protective mechanism is related to the inhibition of cytokine secretion and NF-κB activation.  相似文献

AIM: To investigate the protective effect of pyrrolidine dithiocarbamate (PDTC) on the kidneys in type 2 diabetic rats. METHODS: High-fat diet and a small dose (27 mg/kg) of streptozotocin-induced diabetic rats were treated with or without PDTC (50 mg穔g-1-1, ip) for 1 week, and age-matched nondiabetic animals were also used for comparison. The concentration of malondialdehyde (MDA)and the activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were determined by commercial kit. The ratio of urine microalbumin/creatinine was measured by an automatic biochemical analyzer. The morphological changes of renal glomerulus were observed by HE/Masson staining and transmission electron microscopy. The expression of inducible nitric oxide synthase (iNOS) and nitrotyrosine (NT) in the renal tissues was examined by the method of immunohistochemistry. RESULTS: PDTC-treated rats had lower blood glucose level and urine microalbumin/creatinine ratio than those in untreated diabetic rats. The levels of tissue MDA in diabetic rats were significantly higher, and the activity of SOD and GSH-Px was lower than those in normal control rats (P<0.05). The renal damage in diabetic rats was significantly improved after PDTC treatment. PDTC administration markedly attenuated the expression of iNOS and the production of NT in renal glomerulus and tubule in diabetic rats. CONCLUSION: PDTC not only reduces blood glucose level, but also protects the diabetic rats from diabetic nephropathy by diminishing the expression of iNOS and the production of NT.  相似文献

AIM: To investigate the combined effect of octreotide and Dachaihu decoction on the treatment of severe acute pancreatitis(SAP). METHODS: Wistar rats(n=50) were randomly divided into sham group, SAP group, octreotide group, Dachaihu decoction group and combination group. The quantity of ascites was measured. The levels of amylase, alanine aminotransferase and creatinine in the serum were examined. The morphological changes of the pancreatic tissues were observed by HE staining. The activation of NF-κB and IκBα expression were determined by Western blot. The mRNA expression with ICAM-1 and IL-1 was detected by qPCR.RESULTS: Combined treatment with octreotide and Dachaihu decoction effectively reduced the quantity of ascites and the levels of amylase, alanine aminotransferase and creatinine in the serum in SAP rats. Moreover, combined treatment significantly inhibited SAP-induced activation of NF-κB and decrease in IκBα protein expression, accompanied by a decrease in ICAM-1 and IL-1 mRNA expression. CONCLUSION: Combination of octreotide with Dachaihu decoction effectively attenuates SAP by inhibiting NF-κB signaling pathway and ICAM-1 and IL-1 expression.  相似文献

AIM:To investigate the expression of sterol regulatory element binding protein-1 (SREBP-1) and sterol regulatory element binding protein-2 (SREBP-2) in the kidney of type 1 diabetic rats.METHODS:The triglyceride (TG) and cholesterol content in the kidney of experimental rats were measured by the assay kits and oil red O staining.Furthermore, the expressions of SREBP-1 and SREBP-2 protein were detected by the methods of Western blotting and immunohistochemistry.The analysis of SREBP-1 mRNA was performed by in situ hybridization.RESULTS:Renal triglyceride content was markedly higher in diabetic rats than that in normal rats and the result of oil red O showed that lipid deposited in the renal tubular epithelium.Immunohistochemistry and Western blotting presented similar results that SREBP-1 protein was up-regulated in renal tubular epithelium in diabetic rats.On the other hand, SREBP-2 protein didnt show difference between diabetic rats and normal control rats.In situ hybridization confirmed the increasing of SREBP-1 mRNA in renal tubular epithelium in diabetic rats.CONCLUSION:Above results suggest that altered SREBP-1 may play an important role in the pathogenesis of renal lipid accumulation in type 1 diabetic rats.  相似文献

AIM: To investigate the effects of dexmedetomidine on hemorrhagic shock/resuscitation (HS/R)-induced acute kidney injury (AKI) in rats, and to explore the possible mechanisms. METHODS: Wistar rats (n=32) were randomly divided into 4 groups (n=8):normal saline control group (NS group), dexmedetomidine group (D group), HS/R group and HS/R+D group. The animals were sacrificed at 6 h after resuscitation. The levels of serum creatinine (Cr) and blood urine nitrogen (BUN) were examined. The kidneys of all rats were removed for evaluation of histological characteristics, and the levels of malondialdehyde (MDA), tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and superoxide dismutase (SOD) were measured. The expression of nuclear factor-κB (NF-κB) and hemeoxygenase-1 (HO-1) was determined by Western blot. RESULTS: Compared with NS group, the levels of Cr, BUN, MDA, TNF-α and IL-1β were obviously increased in HS/R group, which were obviously decreased in HS/R+D group (P<0.05). Compared with NS group, the SOD activity was obviously decreased in HS/R group, which was obviously increased in HS/R+D group (P<0.05). Compared with NS group, the protein expression of NF-κB was obviously increased in HS/R group, which was obviously decreased in HS/R+D group (P<0.05). Compared with NS group, the protein expression of HO-1 was increased in HS/R group. Compared with HS/R group, the protein expression of HO-1 was obviously increased in HS/R+D group. Compared with NS group, HS/R induced marked kidney histological injury, which was less pronounced in HS/R+D group.CONCLUSION: Dexmedetomidine effectively protects rats against AKI caused by HS/R, and its mechanism may be associated with the increase in HO-1 expression and the inhibition of NF-κB expression.  相似文献

AIM:To investigate the effect of salidroside on alcoholic hepatic injury in rats. METHODS:The SD rats were randomly divided into 5 groups:negative control group, model group, bifendate group, and low-and high-dose salidroside groups. The rats in model group were administered with 56% alcohol, while the rats in negative control group was administered with saline. The rats in bifendate group and salidroside groups were administered with corresponding drugs every day. The blood and the liver tissues were collected to measure triacylglycerol (TG), alanine aminotransferase (ALT), aspartate aminotransferase (AST), malondialdehyde (MDA) and superoxide dismutase (SOD). The pathological changes of the liver tissues were observed with HE staining. Tumor necrosis factor-α (TNF-α) and nuclear factor-κB (NF-κB) were measured by ELISA and the protein and mRNA expression levels of TNF-α and NF-κB were detected by Western blot and RT-PCR. RESULTS:Compared with model group, the levels of TG, ALT, AST, MDA, TNF-α and NF-κB were reduced, while the activity of SOD was enhanced in salidroside group (P<0.05). The liver tissue injury was significantly attenuated. CONCLUSION:Salidroside improves the pathological changes, reduces inflammation, increases the activity of antioxidant enzymes and reduces lipid peroxidation in the liver with alcohol-induced injury. This effect may be related to regulating the NF-κB-mediated inflammatory responses.  相似文献

AIM: To explore the expression of Snail 1 in renal tissues of diabetic rats, and to investigate its contribution to the progression of diabetic nephropathy. METHODS: Streptozotocin-induced diabetic rats were randomly divided into 2, 4, 8, 12, 16, 20, 24 weeks groups and 16 week A, 20 week A and 24 week A groups. A groups were treated with insulin to control blood glucose to normal level from the 13th week. Control groups were set up in age-matched time points. Blood glucose, 24 h urine protein, serum creatinine (Scr) and kidney index of rats were measured. Periodic acid-silver (PAS) staining was used to observe the renal pathological changes. The mRNA and protein expressions of Snail 1 and FN in renal cortex were detected by RT-PCR and immunohistochemical staining, respectively. Western blotting was employed to detect the expression of Snail 1 protein in the renal cortex. RESULTS: The levels of blood glucose, Scr, kidney weight index were increased remarkably in diabetic rats as compared with those in control groups (P<0.05, P<0.01), and decreased remarkably in the insulin-treated rats as compared with those in the diabetic rats (P<0.05, P<0.01). The Snail 1 protein was not detected by immunohistochemical staining in normal renal tissues. However, strongly positive staining was observed in renal tubules of diabetic rats. A time-dependent loss of Snail 1 expression was detected in the kidney in insulin-treated rats. The Snail 1 protein and mRNA of Snail 1 and FN were significantly up-regulated in the diabetic rats as compared with those in controls (P<0.01), while down-regulated in the insulin-treated diabetic rats (P<0.01). A close positive relationship existed between the mRNA expression of Snail 1 and FN (r=0.800, P<0.01). The level of Snail 1 protein expression was positively correlated with blood glucose, urine protein, Scr, kidney index (r=0.877, 0.694, 0.522, 0.875, P<0.01).CONCLUSION: These findings suggest that Snail 1 gene and protein expression are up-regulated in the kidney of rats with diabetes and may be involved in the pathogenesis of diabetic nephropathy.  相似文献

AIM: To investigate the effects of tumor necrosis factor (TNF) receptor associated factor 3 (TRAF3) on the signaling pathway of NF-κB and the expression of Bax and Bid in renal collecting duct epithelial cells of polycystic kidney disease (PKD), and to observe the tubulogenesis of TRAF3 transgenics for exploring the protective effect of TRAF3 on the cystogenesis and development of PKD. METHODS: The signaling changes of NF-κB and expression of Bax and Bid in PKD renal collecting duct epithelial cells and TRAF3 transgenic PKD renal collecting duct epithelial cells were determined by the Western blot. The percentage of apoptotic cells was measured by flow cytometry with Annexin V staining. The caspase-3 activity was also measured. The 3D Matrigel culture was performed to examine abnormal tubulomorphogenesis in vitro. RESULTS: The over-expression of TRAF3 significantly inhibited the signaling pathway of NF-κB in PKD renal collecting duct epithelial cells and significantly downregulated the expression of Bax and Bid, and also decreased the cell apoptosis. More branch structures were observed in the TRAF3 transgenic PKD renal collecting duct epithelial cells. CONCLUSION: TRAF3 is a negative regulatory inhibitor for Bax and Bid by regulating the NF-κB signaling and may be a new target for inhibiting the cyst formation in PKD.  相似文献
