Pigs experience biological stress such as physiological, environmental, and social challenges when weaned from the sow. The process of weaning is one of the most stressful events in the pig’s life that can contribute to intestinal and immune system dysfunctions that result in reduced pig health, growth, and feed intake, particularly during the first week after weaning. Technological improvements in housing, nutrition, health, and management have been used to minimize some of the adverse effects of weaning stress, but a greater understanding of the biological impact of stress is needed to improve strategies to overcome weaning stress. The focus of this review paper is to briefly describe how the biological stress associated with weaning impacts intestinal morphology, structure, physiology, and intestinal immune responses that can impact subsequent production efficiencies such as growth, intake, morbidity, and mortality.  相似文献

The use of the antimicrobial growth promoters (AGPs) carbadox and olaquindox has been banned in the European Union (EU) since September 1999. We studied the effects of the withdrawal on the health of weaned piglets on two types of piglet-producing farms (farrowing herds and farrow-to-finish herds) from the different regions of Finland. Farms with no major problems with post-weaning diarrhoea were selected for the study to better evaluate the effect of AGPs alone. Data on production, medication and incidence of diarrhoea were collected from 73 farms during 1 year after the withdrawal. On 29 of these farms, the data collection began 4 months before the withdrawal.

The health management of the pigs is considered good in Finland, and special attention has been paid to improve the husbandry practices and management of the farms. Eighty-two percent of the farms in the study were free of both Mycoplasma hyopneumoniae and Sarcoptes scabiei infection. Brachyspira hyodysenteriae infection was not detected in any of the farms. The median number of sows in the herds was 56.0 (IQR = 43.0; 72.5) in 2000.

The level of antimicrobial use in each herd was classified as low, moderate and high when the percentage of weaned pigs treated for diarrhoea during a 4-month period was 0–5%, 6–19% and ≥20%, respectively. Only on four herds (14%), there was an increase in the level of antibiotic use after the AGP withdrawal, when seasonally corresponding 4-month periods were compared. Fourty-one percent of these 29 farms were categorized as low users of antimicrobials, 38% as moderate users and 21% as high users.

The level of antimicrobial use for treatment of diarrhoea after weaning (and the incidence of diarrhoea in weaned piglets) did not increase significantly after the withdrawal of AGPs from weaner feeds according to farmers’ evaluations. In this study, the Escherichia coli infection was the most-common cause of diarrhoea in weaned pigs. The age at weaning did not change after the withdrawal of AGPs.  相似文献

The aim of this study was to evaluate the effects of dietary spray-dried porcine plasma (SDPP) on the productive performance, intestinal morphology and leukocyte cell subsets of piglets. Sixteen early-weaned piglets (20 ± 2 d) were distributed into two dietary treatments: 1) free access to control diet or 2) 6% SDPP in the control diet instead of soy-protein concentrate. Intestinal morphometry of the small and large intestine, haematology and immune cell flow cytometric analysis of blood, ileo-colic lymph node and ileal Peyer's patches were performed in all pigs. Although SDPP treatment did not increase growth performance, it improved feed efficiency. We observed that SDPP diminishes blood monocytes, and macrophages (SWC3+), B lymphocytes (CD21+) and γδ T cells (γδTCR+) in gut lymphoid tissues. SDPP-treated piglets also showed lower intraepithelial lymphocyte numbers and lamina propria cell density in the small and large intestine. All these results suggest lower activation of the immune system of the SDPP-piglets during the post-weaning period. Moreover, the use of SDPP can be considered a valid alternative to antibiotic growth promoters.  相似文献

In the weaning period, transition from sow's milk to the post-weaning diet causes the withdrawal of important nutrients as milk nucleotides, which are known to be determinant for the development of the gastrointestinal tract and immune function. The objective of these investigations was to study the effect of including these nucleotides in solid diets for piglets. Nucleotide composition of sow's milk was analyzed using 5 sows at 21 days of lactation. The average free nucleotide concentration was 102.8 ± 9.16 μmol/100 mL. Two experiments were performed to assess the effect of a product based on this composition (Nucleoforce Piglets®) on digestive adaptation and incidence of diarrhoea of nursery piglets. In Exp. 1, three groups of 6 piglets were weaned at 21 days of age and fed with a diet supplemented with 0 (control), 1000 or 2000 ppm of nucleotides, and a fourth group of 6 piglets was maintained in lactation. Seven days after weaning, piglets were euthanized and samples of jejunal mucosa were processed for histological measurements. Villus height decreased from 448 μm in un-weaned pigs to 275 μm in the control group 7 days after weaning. Although there were no differences in feed intake among groups, the reduction in villous height was less pronounced (P < 0.001) in nucleotide supplemented groups showing a villous height of 351 and 378 μm with the doses of 1000 or 2000 ppm respectively. In Exp. 2, 384 early weaned pigs were fed during 14 days with a diet supplemented with 0 (control), 750 and 1000 ppm of nucleotides. ADG and ADFI were not modified by the treatment, but nucleotide supplementation reduced the number of pigs treated with antibiotic as a result of diarrhoea (15.63% vs 3.13% and 1.56%; for control, 750 and 1000 ppm; P < 0.001). These results suggest that dietary supplementation with nucleotides from yeast might help to prevent post-weaning diarrhoea in piglets.  相似文献

A field study was conducted to estimate seasonal differences in follicular development in weaned sows and to evaluate the implication of these differences on seasonal infertility. A total of 110 sows were selected at weaning during winter–spring (WS, n = 58) and summer–autumn (SA, n = 52). Ovaries were scanned once daily from weaning to the onset of oestrus and twice daily from then until ovulation. Six sows during WS were removed from study for not showing growing follicles at weaning. Oestrus was evaluated twice daily from day 1 after weaning to day 14 post-weaning. One of 52 (1.9%) sows in WS and 9/52 (17.3%) in SA showed no signs of oestrus within 14 days of weaning (P < 0.05). The diameters of the follicles at weaning, at the onset of oestrus and just before ovulation were smaller (P < 0.01) in SA sows than in WS sows. There were fewer follicles in SA sows than in WS sows just before ovulation (P < 0.05). Fifty of 51 (98.0%) sows in WS and 31/43 (72.1%) sows in SA experienced a weaning-to-oestrus interval (WOI) of 3–6 days (P < 0.05). Fifty-one of 52 (98.1%) sows in WS and 43/52 (82.7%) sows in SA were inseminated; the percentage of pregnant sows that failed to farrow was lower in WS (1/51, 2.0%) than in SA (5/43, 11.6%; P < 0.05). The percentage of farrowed sows was greater in WS (46/51, 90.2%) than in SA (32/43, 74.4%; P < 0.05). Sows in WS had on average 1.5 more piglets than sows in SA (P < 0.05). Sows with a WOI of 3–6 days had lower rates of pregnancy losses (P < 0.05) and higher farrowing percentages (P < 0.01) than those with a WOI > 6 days, irrespective of season.  相似文献

Chloride secretion in the intestines of pigs of different age (32 days and 4 months) was examined using the Ussing chamber technique. After stimulating chloride secretion by carbachol and forskolin, alternative chloride channels and finally Cystic Fibrosis Transmembrane Conductance Regulator (CFTR) channels were blocked. While basal short-circuit currents (Isc) were higher in the jejunum than in the colon of the piglets of 32 days, it was inverse for the 4-month-old fatteners. In the jejunum responses to stimulation and final levels of Isc were higher in the piglets than in the fatteners. The decrease in Isc after blocking CFTR channels was smaller in the fatteners than in the piglets in both intestinal segments. In conclusion, 32-day-old piglets show a higher basal chloride secretion as well as a higher response to stimulation in the jejunum than 4-month-old pigs.  相似文献

The objective of this experiment was to investigate the effect of non-starch polysaccharide (NSP) solubility and inclusion rate on gut health and development, performance and risk of post weaning enteric disorders (PWED) using NSP sources known not to affect digesta viscosity. The experiment consisted of a 2 × 3 factorial combination of NSP type (sNSP vs insoluble NSP (iNSP); inulin vs purified cellulose) and inclusion level (50 vs 100 vs 150 g/kg). Piglets were weaned at 28 days (day 0) and fed ad libitum until slaughter on day 14. There were no incidences of PWED. NSP solubility had little effect on performance, but sNSP diets resulted in lower caecal and colonic pH and higher colonic Lactobacilli to coliform ratio compared to iNSP diets. Increasing NSP levels significantly reduced caecal and colonic pH but decreased empty body weight percentage (P = 0.067). These results suggest that gut health may benefit most from diets containing appropriate sources of predominantly sNSP rather than iNSP. The optimal level of such sNSP will likely depend upon the level of risk of PWED in order to balance the trade off between maximising performance and improving gut health. Effects of NSP feeding under a higher infectious pressure remains to be assessed.  相似文献

During development, gut morphology and functions change. Some of these are mediated by milk-borne factors, e.g. insulin-like growth factor-1 (IGF-1) and epidermal growth factor (EGF), of which the effects are in part dictated by the distribution of their receptors (R).

EGFR- and IGF-1R-immunoreactivity (IR) was evaluated in samples of the fetal, neonatal and weaned porcine small intestine.

EGFR-IR first appeared in the neonatal duodenum and was located basolaterally in villar and to a lesser extent in cryptal enterocytes. In weaned pigs, EGFR appeared apically in enterocytes with a region-dependent distribution along the villus–crypt axis. IGF-1R-IR was observed in smooth muscle cells in each age group. From the last trimester of gestation onward IGF-1R-IR was seen at the apical and basolateral side of villar enterocytes and in submucosal arterioles.

Thus, the age- and region-dependent presence of EGFR and IGF-1R has to be taken into account when evaluating effects of EGF and IGF-1 on growth or repair.  相似文献

The current study was conducted to investigate the effects of high dietary concentrations of Zn as zinc oxide and Cu as copper sulfate on the activity of digestive enzymes in the pancreas and the intestinal mucosa, intestinal morphology, and mucin histochemistry in pigs after weaning. Thirty-two pigs were weaned at 4 wk of age. The pigs were fed standard weaning diets supplemented with Zn (100 or 2,500 ppm) and Cu (0 or 175 ppm) in a 2 x 2 factorial arrangement of treatments for a 14-d period. In pancreatic tissue, the activity of amylase, carboxypeptidase A, chymotrypsin, trypsin, and lipase increased (P < 0.01) in pigs fed 2,500 ppm of Zn, whereas the activity of carboxypeptidase B and carboxylester hydrolase was unaffected. Copper had no effect on the activity of pancreatic enzymes. In small intestinal contents, the total activity of amylase and carboxypeptidase A was greater in pigs fed 100 ppm of Zn (P < 0.05), whereas feeding 2,500 ppm of Zn increased the chymotrypsin activity (P < 0.001). The remaining enzymes were unaffected by dietary Zn concentration. The villi were longer in the cranial small intestine (P < 0.001) in pigs fed 100 ppm of Zn than in pigs fed 2,500 ppm of Zn, but otherwise there were no clear effects of Zn and Cu supplementation on intestinal morphology. In the cranial small intestine, the activity of maltase (P < 0.001), sucrase (P < 0.001), and lactase was greater in pigs fed 100 ppm of Zn, even though there was a Zn x Cu interaction (P < 0.05) in lactase activity. In the middle and caudal small intestine, no clear differences between dietary treatments were observed. The activity of gamma-glutamyl transpeptidase in the intestinal mucosa was not affected by dietary Zn or Cu. In pigs fed 100 ppm of Zn, the activity of aminopeptidase N was greater in the caudal small intestine, but dietary Zn or Cu had no effect on aminopeptidase N in the cranial and middle small intestine. No effect of dietary Zn or Cu supplementation was found on carbohydrate histochemistry in the caudal small intestine, whereas high dietary Zn increased the area of neutral, acidic, and sulfomucins in the cecum (P < 0.01) and in the colon (P < 0.001). In summary, high dietary Zn increased the activity of several enzymes in the pancreatic tissue and increased the mucin staining area in the large intestine, whereas Cu had no clear effect on these variables. However, no definite answers were found as to how the growth promoting and diarrhea reducing effects of excess dietary Zn are exerted.  相似文献

In the small intestinal segment perfusion model the effect of osmolality on net fluid absorption in enterotoxigenic Escherichia coli (ETEC)-infected and non-infected small intestinal segments of piglets was investigated. In ETEC-infected segments net fluid absorption was reduced. Lowering the osmolality from 375 to 150 mOsmol/l by reducing sodium chloride concentrations increased net fluid absorption. There was a linear relationship between osmolality and net fluid absorption for both non-infected and ETEC-infected segments. Below 150 mOsmol/l the inverse relationship between osmolality and net fluid absorption disappeared. Substitution of sodium chloride with mannitol decreased net fluid absorption since mannitol cannot be actively absorbed. In ETEC-infected segments perfused with oral rehydration solution net fluid absorption significantly increased compared to a sodium chloride solution of similar osmolality, probably because of the high glucose concentration. Supplying hypotonic sodium chloride fluids to piglets can be beneficial in replenishing water and electrolyte losses in case of infectious diarrhoea.  相似文献

There are a large number of microorganisms in the porcine intestinal tract. These microorganisms and their metabolites contribute to intestinal mucosal immunity, which is of great importance to the health of the host. The host immune system can regulate the distribution and composition of intestinal microorganisms and regulate the homeostasis of intestinal flora by secreting a variety of immune effector factors, such as mucin, secretory immunoglobulin A (sIgA), regenerating islet-derived III (RegIII)γ, and defensin. Conversely, intestinal microorganisms can also promote the differentiation of immune cells including regulatory T cells (Treg) and Th17 cells through their specific components or metabolites. Studies have shown that imbalances in the intestinal flora can lead to bacterial translocation and compromised intestinal barrier function, affecting the health of the body. This review focuses on the composition of the pig intestinal flora and the characteristics of intestinal mucosal immunity, discusses the interaction mechanism between the flora and intestinal mucosal immunity, as well as the regulation through fecal microbiota transplantation (FMT), dietary nutritional composition, probiotics and prebiotics of pig intestinal microecology. Finally, this review provides insights into the relationship between intestinal microorganisms and the mucosal immune system.  相似文献

试验旨在探索对仔猪床进行加温处理以及采用定时饲喂的方式的对早期断奶仔猪不同肠段黏膜形态和肠道发育的影响,从而为确定断奶仔猪适宜的饲养温度和科学的饲喂方式提供理论依据。试验采用2×2因子设计。选用21日龄杜长大断奶仔猪240头,随机分到4个处理组:1):28~30℃的仔猪床加温处理组;2)23~25℃的低温对照组(炉火暖气管道舍内加热);加温处理组和低温对照组中又根据不同的饲喂模式分为每天6次定时饲喂组和自由采食组。每个处理6个重复,每个重复10头猪。经过14 d的试验之后,在每圈仔猪中随机抽取1头,屠宰后取样检测小肠黏膜的绒毛高度、隐窝深度和肠壁厚度。结果表明:加温饲养组(28~30℃)的仔猪空肠中段的绒毛高度显著高于低温对照组(23~25℃)(P<0.05)。加温饲养组十二指肠的隐窝深度和空肠中段的肠壁厚度显著低于低温对照组(P<0.05)。定时饲喂饲养的仔猪空肠后段的绒毛高度显著高于自由采食饲养的仔猪(P<0.05)。定时饲喂组仔猪空肠中段的肠壁厚度和十二指肠隐窝深度显著低于自由采食组(P<0.05)。2因子的互作效应表明:加温定时饲喂组饲养的仔猪空肠后段的绒毛高度显著高于其他处理组。低温自由采食组饲养的仔猪空肠后段的绒毛高度极显著低于其他处理组(P<0.01)。加温定时饲喂组的仔猪空肠中段的肠壁厚度极显著低于其他处理组(P<0.01)。由结果可知,28~30℃的加温饲养组以及定时饲喂的饲喂模式有利于仔猪的肠道黏膜形态的改善,有益于肠道的发育和健康。  相似文献

The intestine is not only critical for nutrient digestion and absorption, but also is the largest immune organ in the body. However, in pig production, inflammation induced by numerous factors, such as pathogen infection and stresses (e.g., weaning), results in intestinal mucosal injury and dysfunction, and consequently results in poor growth of pigs. Dietary fatty acids not only play critical roles in energy homeostasis and cellular membrane composition, but also exert potent effects on intestinal development, immune function, and inflammatory response. Recent studies support potential therapeutic roles for specific fatty acids (short chain and medium chain fatty acids and long chain polyunsaturated fatty acids) in intestinal inflammation of pigs. Results of these new lines of work indicate trophic and cytoprotective effects of fatty acids on intestinal integrity in pigs. In this article, we review the effect of inflammation on intestinal structure and function, and the role of specific fatty acids on intestinal health of pigs, especially under inflammatory conditions.  相似文献

Co-mingling weaned pigs often results in aggressive behavior which can result in injury and reduced performance. This experiment examined the effect of Acclimate™, a product reported to reduce equine aggression, on weaned pig aggression and growth performance. Weanling pigs (n=214, avg. wt.=5.93 kg) were blocked by weight, sex and litter (n=25) and randomly assigned to either Acclimate™ treatment or a control group. Acclimate™ treated pigs were marked on the nose, face and neck. Pigs were individually identified and videotaped to determine frequency of aggressive sequence (FAS) and duration of aggression (DOA) on an individual pig basis. Pig weights and feed consumption were used to determine feed intake, total weight gain (TWG), average daily gain (ADG) and feed to gain ratio (F/G). Differences between litter, sex and treatment for FAS, DOA, F/G and ADG were tested using GLM, SAS. Neither Acclimate™ treatment or sex had a significant effect on FAS, TWG or F/G. However, there was a trend towards reduced duration and intensity of aggression in Acclimate™ treated pigs (p=0.1). There was a significant effect of litter (p<0.01) and TWG (p<0.001) on FAS at both 1 and 3 weeks. Acclimate™ treatment of weaned pigs does not decrease FAS, but may decrease the intensity and duration of aggression without negatively effecting growth.  相似文献

Intestinal fatty acid binding protein (I-FABP) is an intracellular epithelial protein in the intestinal mucosa of many animals. I-FABP appears in the circulation following epithelial damage, and in humans, is proven to be a parameter for damage to the mucosa. In this paper, an ELISA test designed for human I-FABP analysis was used to assay pig blood samples. The test recognized I-FABP cloned from pig small intestine and expressed in Escherichia coli. Furthermore, in our experimental model of (low flow) intestinal ischemia and reperfusion a significant rise in plasma I-FABP concentrations 15-30 min after clamping of the mesenteric artery was demonstrated. This is the first report that in pigs circulating I-FABP is a useful marker for (mild) intestinal injury, and could possibly be used to monitor (intestinal) health in clinical practice.  相似文献

Not nocuous bacteria are important for the maturation and the modulating activity of the gut immune system. However, the humoral immune response against commensal and probiotic bacteria is less documented, particularly in farmhouse animals. Blood serum and saliva were collected in two trials where probiotics, Lactobacillus rhamnosus GG (LbR), well-defined human isolate (Trial A), and a novel and abundant porcine commensal, Lactobacillus sobrius strain 001T (LbS) (Trial B), were supplemented to weaning pigs. Anti-LbR IgA were present in serum of pigs before treatment with LbR, but also after 1 or 2 weeks in control pigs, notwithstanding the absence of DNA from LbR in colon. Pigs fed or not LbS for 1 or 2 weeks had LbS-specific IgA, in saliva and in serum. Colon contents of control subjects were positive for DNA from LbS. We hypothesized that part of this IgA strain-specific activity is partially related to immune cross-reactivity between different Lactobacillus-species. With the procedure of Shu et al. [Shu, Q., Bird, S.H., Gill, H.S., Rowe, J.B., 1999. Immunological cross-reactivity between the vaccine and other isolates of Streptococcus bovis and Lactobacillus. FEMS Immunol. Med. Microbiol. 26, 153–158], after ELISA test on blood serum or saliva pre-incubated with LbR or LbS, each strain blocked a relevant part of IgA specific for the other. So bacteria with different affinity for the pig present reciprocal crossed immune activity. When probiotics are supplied to weaning pigs, the possible action of already present multi-effective IgA should be considered. The mechanism of IgA induction by certain probiotics needs to be addressed in further studies.  相似文献

To demonstrate whether that hypotrophied histological alterations of intestinal villi and epithelial cells are observed in chickens fed low crude protein (CP) diet, 36 male chickens were allotted into 10%, 16% and 22% CP diets groups for 35 days. The 10% CP group showed a lower value of weight gain and feed efficiency than the other two CP groups (p < 0.05). On the relative weight of visceral organs, the 10% CP group showed a heavier gizzard than the 22% CP group, a heavier jejunum than the other two CP groups and a heavier value of ileum and caeca than the 22% CP group (p < 0.05). On the relative length of intestines, the 10% CP group showed a longer value of duodenum and caeca than the other two CP groups, a longer jejunum than the 16% CP group and a longer colon than the 22% CP group (p < 0.05). The villus height and villus area of all intestinal segments did not change in all groups. The duodenal cell areas and cell mitosis numbers tended to be lower in 10% CP group than in the other two protein groups, and the jejunal cell area and duodenal cell mitosis numbers were decreased in the 10% crude protein group (p < 0.05). In relation to the protuberated cells in the higher CP groups, the 10% CP group showed only faintly protuberated cells on the duodenal and jejunal villus apical surface. The ileal villi did not show specific alterations among each group. The chronic feeding of low CP diet induced a hypotrophied histological alteration. This suggests that the hypotrophied histological alterations can assess that the fed‐diet is not so well‐balanced diets, nutritionally.  相似文献
