OBJECTIVE: To relate trichostrongylid infections acquired by sheep during summer to prevailing weather conditions. PROCEDURE: Groups of worm-free 'tracer' sheep were put onto pastures, previously contaminated with trichostrongylid eggs, for successive periods of 2 weeks from December to March. After grazing the sheep were housed for 6 weeks. Weekly worm egg counts and worm counts were used to estimate the numbers of worms acquired and related to weather conditions during the grazing period. RESULTS: No worm eggs were detected in the faeces of sheep that grazed at the end of January when only 7 mm of rainfall was recorded. At other times rainfall between 12 and 24 mm occurred and strongyle egg counts were generally either < 50 or > 150 eggs per g (epg). Mean counts of 1,100 Ostertagia and Trichostrongylus adults gave rise to mean counts of about 350 epg whereas about 6,000 Nematodirus spp were associated with mean egg counts of about 200 Nematodirus spp epg. CONCLUSIONS: Rainfall events during summer determine the numbers of trichostrongylid larvae acquired by sheep in summer but further studies are necessary before the implications for strategic control programs in southern Australia can be fully assessed.  相似文献

The relative resistance of six breeds of sheep to Haemonchus contortus infection was investigated under field conditions on two stations situated in a semi-arid zone in Kenya. As judged by faecal egg counts, monitored throughout a two-year period, it was found that there were consistent differences between the breeds. The breeds studied in order of increasing susceptibility were Red Masai, Blackhead Persian, Merino, Dorper, Corriedale and Hampshire. Post mortem findings confirmed that the differences in faecal egg count were related to the establishment of worms rather than larval inhibition or the suppression of worm egg production. There was also evidence that the Red Masai showed self-cure more frequently than the other breeds.  相似文献

Twelve parasite-naive sheep were used to study the possible direct anthelmintic effect of a condensed tannin extract (quebracho) on the population and fecundity of the intestinal nematode Trichostrongylus colubriformis. The sheep were infected with a single dose of 20,000 L3 of T colubriformis. Twenty-eight days later, six of them were drenched daily for a week with quebracho extract at 8 per cent by weight of their food intake. All lambs were then slaughtered, and their small intestines removed to estimate the worm burdens and the numbers of eggs in utero. Two days after the first drench with tannin extract the faecal egg counts of the treated sheep were approximately 50 per cent of those of the control sheep (P<0.01), but there was no further reduction with continued drenching. In the treated sheep the worm burdens and number of eggs per gram faeces per worm were reduced by 30 per cent compared with the controls (P<0.05), but the sex ratios, the number of eggs in utero and length of the worms were not affected by drenching with tannin.  相似文献

Haemonchus cysteine proteases, because of their apparent critical function in worm physiology, are considered important candidates in the immunological control of haemonchosis in sheep. Only limited information is, however, available on the immunoprotective properties of these molecules in goats. In the present study cysteine proteases of Haemonchus contortus adult worms isolated from a goat strain (Gran Canaria, Spain) were enriched by affinity chromatography and evaluated as immunoprotective antigens against caprine haemonchosis. The eggs per gram of faeces averaged over the whole experiment for unvaccinated goats (550 +/- 13.5) was significantly greater (P < 0.001) than that of vaccinated goats (61 +/- 2.9). Accordingly, the worm burden was significantly different between the groups (P < 0.05), with mean values of 247.5 +/- 43.8 and 762.5 +/- 78.3 worms per animal in the immunized and nonimmunized goats, respectively. The percentage of egg (89%) and worm (68%) reduction approached those attained with other immunogens used in sheep.  相似文献

One-year old worm free, merino wethers, were each infected with 5000 H. contortus larvae of a strain resistant to mebendazole at a rate of 52 mg/kg body weight of sheep. After 21 days, they were assigned to two trials. The preliminary trial showed that mebendazole and levamisole acted synergistically on the H. contortus infection. In the second trial, sheep were treated with 0.35 mg/kg levamisole (one seventh the minimum effective dose against susceptible worms) or 40 mg/kg mebendazole (40 times the minimum effective dose against susceptible worms). In each case the anthelmintics did not reduce worm burdens, although mebendazole depressed egg production. However, when mebendazole and levamisole, at the above dose rates, were administered simultaneously, total worm counts in sheep were reduced by almost 60%. Similar results were obtained when the levamisole was administered 8 h or 14 h after mebendazole treatment. The implications of these observations for the treatment of benzimidazole-resistant haemonchiasis in sheep are discussed.  相似文献

Five Merino and five Dorper sheep were artificially infested with the sheep scab mite Psoroptes ovis and the effect of infestation on their haematology, serum protein levels and live mass recorded for a period of 14 weeks. The reaction of the Merino sheep to infestation was more severe than that of the Dorper sheep. Haematological values fluctuated within the normal range during the assessment period. The mean haemoglobin concentration of the Merino sheep declined until antiparastic treatment was administered 10 weeks after infestation, after which it gradually increased. The lymphocyte counts of both breeds of sheep declined from 2 weeks to 10 weeks post-infestation, but increased after treatment, while the highest eosinophil counts were recorded in the Merino sheep at the height of the acute disease 8-10 weeks post-infestation. Serum albumin values for both breeds and serum globulin values for the Merino sheep were higher than normal during the entire 14-week observation period. A decrease in serum albumin and an increase in serum globulin concentration occurred at the height of infestation in both breeds. The mean live mass of a second group of five infested Merino sheep decreased by 6.4 kg over a 16-week period compared to a gain of 4.56 kg for five infested Dorper sheep.  相似文献

Infections with the zoonotic trematode, Schistosoma japonicum in pigs serves as a valuable model for studying natural definitive host/parasite relationships and a model for human schistosomosis japonica. In the present study the efficiency of a peroral infection route was compared with that of an intramuscular route of infection. Eleven specific pathogen-free Danish Landrace/Yorkshire/Duroc crossbred male and female pigs were divided into two groups of five and six pigs, respectively. Each pig was given 1000 cercariae, either placed in droplets on the mucosa in the buccal cavity, or as medium-suspended cercariae injected into musculus biceps femoris of one of the hindlegs. Ten weeks post infection, all pigs were killed with pentobarbital and the venous system perfused. Worm burdens and liver egg counts were determined and worm fecundity was calculated. S. japonicum infections were established in all individuals in both groups of pigs. When comparing the two groups, the peroral group had significantly higher number of immature worms, whereas the intramuscularly infected group had significantly more worm nodules. However, no difference was seen in total number of worms. No statistical significant differences were found in neither tissue egg counts nor worm fecundity when comparing the two groups. The results from the present study showed a delay in maturation of infection following a peroral infection as compared with an intramuscular infection, but comparability was seen between overall worm establishment and egg production.  相似文献

Thirteen groups of 4 South African mutton Merinos grazed for 4 weeks with the flock on Kikuyu pastures and were slaughtered for total and differential worm counts at necropsy. Subsequently 12 groups of 8 week tracers grazed on the pastures and were killed for worm counts post mortem. The following were present in most sheep: Teladorsagia (syn. Ostertagia) circumcincta, Trichostrongylus axei, Trichostrongylus colubriformis, Dictyocaulus filaria and Oesophagostomum venulosum. Haemonchus contortus, Nematodirus spathiger and Trichuris skrjabini were less frequently recovered. Optimal conditions for infestation of grazing sheep occurred from June (late autumn)--October (spring) when mean temperatures in any 4 week period were less than 20 degrees C and a total of greater than 40 mm of rain fell on 8 or more separate days. When the mean temperatures exceeded 20 degrees C pastures were safe, sheep acquiring less than 1,000 worms in 4 weeks.  相似文献

The clinical pathology of Schistosoma curassoni infection in sheep and goats was studied for 22 weeks following experimental infection with 7000 and 4000 cercariae, respectively. Excretion of eggs began at week 7 after infection: in goats the numbers increased to 30 to 50 eggs per gram faeces (epg) at weeks 8 to 18, followed by a reduction. In a pregnant goat, epg values increased markedly before and after parturition. The mean faecal egg counts in sheep were lower than in goats, increasing to a maximum level of 30 epg at weeks 16 and 17 after infection. Infected sheep maintained growth rates roughly comparable with controls, whereas infected goats failed to gain as much weight as the controls. Infected goats and sheep produced eosinophil counts of about 3 x 10(3) mm-3, five and eight weeks after infection, respectively. Sheep developed a progressive anaemia from week 11 after infection, in goats blood values remained within normal limits. Differences in serum protein concentration were observed between infected and uninfected goats about nine weeks after infection, but not in sheep. Increased total protein values, hyperglobulinaemia and lowered albumin to globulin ratios were features of infected goats. Serum glutamate oxaloacetate transaminase, glutamate pyruvate transaminase, gamma-glutamyl transferase, total lactate dehydrogenase and bilirubin were not significantly changed. The mean recovery in sheep was 608 worms, in goats 428 worms, but the total tissue egg counts were higher in the latter. Of the total eggs deposited in the goats 92 per cent were found in the liver with 51.5 per cent in the ovine liver. The histopathological changes were studied.  相似文献

The efficacy of milbemycin oxime against fourth-stage (L4) larvae and adults of Ancylostoma tubaeforme was investigated in a trial involving 24 young domestic shorthair cats. The animals were inoculated with approximately 300 infective stage three (L3) larvae and divided into three groups. After 12 days, eight cats (group 1) were treated with medicated tablets containing 4 mg milbemycin and 10 mg praziquantel to test the efficacy against L4 larvae; eight cats in group 2 were treated with the same tablets after 33 days to test the efficacy against adult worms; and eight cats in group 3 were treated with a placebo tablet. Faecal egg counts were determined periodically in each cat and after 40 or 41 days the number of worms in each animal was determined postmortem. The egg count reduction was determined by comparing the geometric mean numbers of eggs per gram of faeces in the placebo and medicated groups, and the worm reduction by comparing the geometric mean numbers of worms. The egg count reduction was more than 99 per cent in both treated groups, while the number of worms in groups 1 and 2 were reduced by 94.7 per cent and 99.2 per cent, respectively.  相似文献

The resistance of a tropical breed of sheep towards the nematode parasite Haemonchus contortus (Hc) was compared to that of a European breed after artificial infection with the parasite following a 2 x 2 x 2 experimental design: two isolates (Hc Gua from Guadeloupe versus Hc Fra from France), two breeds each reared in their native habitats (Black Belly (BB) in Guadeloupe, FWI, INRA 401 reared in France), and two groups of lambs with differing infection status (one naive, i.e. infected for the first time during the experiment, and one primed, i.e. which had previously received doses of 5000 L3 on Days -35 and -32 before the infection during this experiment). Both groups were composed of 10 male lambs, which were 20 weeks of age when they were given a dose of 10,000 L3 of the relevant isolate on Day 0. Resistance was assessed by regular fecal egg counts (FECs) and blood samples, and worm counts were processed on Day 35 p.i. The FEC was lower in the BB than in INRA 401 lambs, lower in the primed than naive groups, and lower in Hc Fra than in Hc Gua. The eosinophil count remained unchanged in the naive INRA 401 groups, but increased in all the other groups. The hematocrit fell after infection, but the live weight was not affected by the infection. Worm burdens were 1868 and 5237 in the naive BB and INRA 401 groups, respectively, and 596 and 4835 in the primed BB and INRA 401 groups. The daily production of eggs per Hc Gua female worm was 1.5-fold that of Hc Fra. The number of worms had no effect on worm length or Hc in utero egg count in the BB sheep, but a positive and significant correlation between these two traits was seen in the INRA 401 lambs. These findings are indicative of innate resistance, and confirmed the high acquired resistance of the BB sheep to both sympatric and allopatric isolates of Hc. The regulation of worm population and fecundity differed in the BB and INRA 401 sheep.  相似文献

The application of polymer matrix ear tags impregnated with 8.5% w/w cypermethrin to 6 wethers following shearing reduced lice to non-detectable levels on 4 of them at 29 weeks after tagging. At the conclusion of the study at 45 weeks the mean count of lice on tagged wethers was 3 per sheep compared to 158 on untreated wethers. In a second experiment, 6 non-infected sheep were treated at shearing with cypermethrin tags, 6 with 25 g/l cypermethrin backline formulation, 6 with tags plus backline and 6 were left untreated. Each group was exposed to 6 sheep with moderate to heavy infestations of lice. Compared to controls, all treatments delayed infestation, but cypermethrin tags gave no longer protection than backline treatment. All sheep were infested by 17 weeks after commencement of the study. At 45 weeks mean counts of lice were 38, 27, 20 and 74 respectively for sheep treated with tags, backline formulation, backline plus tags and untreated. Possible reasons for the better effect observed from applying tags to infested sheep than to sheep which were not infected at application but which were subsequently exposed to infested sheep are discussed.  相似文献

Sixteen nonsibling sheep, approximately 12 months old, that were raised in a helminth-free environment, were used for 2 protection studies 6 months apart. Sheep were vaccinated weekly for 5 weeks by IM injection of fibrinogen-degrading proteins derived from the intestinal tract of adult Haemonchus contortus. Ten days after the last vaccination, sheep were given 2,500 infective H contortus larvae by intraruminal injection. Vaccinated sheep produced specific antibodies, and were protected from the worm challenge. Significant differences in mean fecal worm egg counts for 56 days after worm challenge, in mean numbers of H contortus worms, and female fecundity ratios at necropsy were detected in vaccinated sheep, compared with those in control sheep. These data suggest that the fibrinogen-degrading proteins have a protective role in vaccination of sheep against H contortus.  相似文献

In a series of three experiments, 64 jirds, Meriones unguiculatus, were infected with 700 infective larvae of Trichostrongylus colubriformis for the study of several biological parameters of this laboratory host-parasite model. In jirds the third stage larvae of T. colubriformis were shown to reach the fourth larval stage by the Day 6 post-infection (PI). By Day 10 PI, all the worms harvested had reached the immature adult stage. All immature adult stages of T. colubriformis developed to sexually mature adult stage by Day 12 PI when the female worms showed developing eggs in their uteri. Developed eggs were observed in the uteri of females on Days 13 and 14 PI. The first eggs of T. colubriformis appeared in the faeces of jirds on Day 13 PI. The peaks of egg production were recorded between Day 21 and Day 31 PI. Immunosuppression of jirds infected with 700 L3, by administration of dexamethasone from Day 57 to Day 94 PI led to increased faecal egg count when compared to untreated controls. All the worms were located in the small intestine. The jirds of dexamethasone treated group harboured higher number of adult worms than those of the untreated group. The number of adult worms was significantly higher in the first part of the small intestine than in the three other similar parts of the small intestine. The coefficient of correlation between the faecal egg count and worm number on the day of necropsy of jirds ranged between r = 0.58 and r = 0.89. Patent infections in jirds were maintained till the end of experiment on Day 100 PI, indicating that in this host and unlike other laboratory hosts, T. colubriformis is responsible for long lasting infections similar to what happens in domestic ruminants. The results of the present study suggest that the jird is a suitable laboratory model to study various aspects of this host-parasite relationship.  相似文献

Nineteen Friesian steers infested with between 5 000 and 45 000 cercariae of Schistosoma mattheei were slaughtered at intervals between 7 and 107 weeks post-infestation and the parasites were recovered by perfusion of the blood vessels. Adult parasites were eliminated as infestation progressed, the greatest loss occurring between 18 and 40 weeks. Females in the mesentery became shorter as infestation progressed but males remained unchanged. Males from the gastric veins were sometimes longer than those from the mesentery while males and females from liver and lungs were often shorter. Females from the mesentery contained 0–107 eggs, with a mean of 36, and those from liver and lung contained fewer eggs. The prepatent period was 6–7 weeks. A sharp reduction in faecal egg count between 15 and 25 weeks post-infestation was caused by a reduction in oviposition. The marked changes in the number of parasites present, their length and their egg-laying capacity appeared to be mediated by the immune response of the host and were manifestations of the establishment of a biological equilibrium which limited the pathogenic effects of the parasite.  相似文献

Challenge with an equal mix of drug-resistant and drug-susceptible larvae of Teladorsagia circumcincta resulted in infections in groups of lambs (n = 6) either untreated or given controlled-release capsules, containing either albendazole or ivermectin. Lambs treated with albendazole capsules contained similar numbers of adult worms at necropsy to the other groups but had no detectable faecal egg count. Animals treated with ivermectin capsules had similar worm burdens and faecal egg counts to the control group but the worms had significantly higher numbers of eggs in utero. These results provide evidence for suppression of egg production by both anthelmintic treatments. The observation that albendazole caused a significant reduction in the developmental success of parasite eggs also has implications for the use of faecal egg count as an indicator for pasture contamination with resistant parasites. In two further groups of lambs, either untreated or given albendazole capsules, treatment caused a significant reduction in egg count and adult worm burden of Trichostrongylus colubriformis. No significant effects were observed on in utero egg counts or egg viability and the apparent effect on the number of eggs produced in faeces per adult female was not significant (p = 0.077). There was, therefore, no evidence that albendazole controlled-release capsules caused suppression of egg output in this species.  相似文献

Twelve each of Red Masai and Dorper sheep, aged between 6 and 9 months, were acquired from a Fasciola-free area of eastern Kenya. Each breed was divided into two groups of 6. The sheep in one group of each breed were experimentally infected with 400 viable metacercariae of Fasciola gigantica. The other group of 6 sheep of each breed remained as uninfected controls. The animals were monitored regularly for any evidence of disease. Blood samples taken weekly revealed a general reduction in red cell counts and packed cell volume, which was much faster in the infected Dorper sheep than in the Red Masai. This reduction started from the tenth week after infection and persisted to the end of the experiment 18 weeks post infection (PI). The absolute eosinophil counts rose in all the infected animals, but the values were higher among the Dorper than among the Red Masai. Patency occurred at weeks 12 and 13 PI in the Red Masai and Dorpers, respectively, with the latter shedding significantly more fluke eggs. The worm recovery rates were higher among the Dorpers than among the Red Masai, though not significantly so. On the basis of egg counts and clinicopathology, the Dorper sheep were considered to be more susceptible to F. gigantica infections.  相似文献

In a study designed to test the effect of molybdenum (Mo) on resistance to trichostrongylosis, the Mo content of the diet during primary infection of 8-month-old Merino lambs with Trichostrongylus colubriformis was found to affect the number of worm eggs found in the faeces during challenge and the total number of worms in the jejunum after six weeks of challenge infection. The optimal intake of Mo in this experiment was in the range of 4-8 mg sheep(-1)day(-1), approximately equivalent to feeding a diet containing 6-10 mg Mo kg(-1)dry matter (DM) and to 0.15-0.30 mg Mo kg(-1)liveweight. Lambs fed Mo at this rate showed a 90 per cent reduction in faecal egg count and total worm count six weeks after challenge compared with lambs fed quantities of Mo outside this range. The mechanism by which Mo exerted these effects was not defined, although the interactions of molybdenum, worm establishment and faecal egg count suggested that this trace element may be acting via an effect on the host's acquired immune response. This hypothesis is supported by the observed enhancement of immune responses (intestinal antibody and granulocyte numbers and in vitro worm-specific proliferation of lymphocytes) associated with Mo intake of 0.15-0.3 mg kg(-1)Lwt.  相似文献

A single Merino sheep, artificially infested with the sheep scab mite, Psoroptes ovis, and a similarly infested Dorper sheep were placed with 9 uninfested Merino or 9 uninfested Dorper sheep respectively during winter and the rate of spread of infestation on the uninfested sheep observed. The same procedure was repeated in summer. It took 14 and 8 weeks respectively in winter before all sheep in the 2 groups displayed lesions of sheep scab, whereas in summer it took 10 and 12 weeks before all sheep had lesions.  相似文献

Worm egg counts of yearling Angora or Merino wethers grazing in mixed flocks at either 4/ha or 8/ha were observed over a 5 month period. Based on faecal egg counts infection levels in the sheep increased for the first 3 months and then decreased as the animals developed resistance. Egg counts in the goats did not decrease during the experiment. Post mortem worm counts indicated that there was a significant host species--stocking rate interaction on worm burdens of Haemonchus contortus adults and Ostertagia circumcincta adults and larvae. Goats had more H. contortus and O. circumcincta adults than sheep at the 8/ha stocking rate. At 4/ha goats had fewer O. circumcincta fourth stage larvae than sheep. There was no stocking rate effect on the numbers of Trichostrongylus axei or T. colubriformis. Sheep had more T. axei and fewer T. colubriformis than goats.  相似文献
