AIDS is a slow virus disease caused by a lentivirus. The silent incubation period following infection usually lasts many years, during which the infected person is potentially infectious to others. Pathological changes accumulate imperceptibly. When major symptomatic disease develops it is eventually fatal. The epidemic spread of the virus is new in Africa, as well as in North America and Europe. Twenty-five per cent of people infected with the virus die within seven years of infection. The ultimate mortality 20 years after infection cannot yet be known, but it may turn out to be close to 100 per cent. It is already apparent that AIDS is the most lethal epidemic viral disease of humans known to medical science. Flaws in the generally accepted hypothesis that AIDS is a sexually transmitted infection are exposed. It has been, characteristically, a blood-borne infection in the early years of the epidemic. However, the AIDS virus is exceptionally unstable genetically and it is probable that means of transmission more efficient than through blood have already developed.  相似文献

The Becht strain of infectious bursal disease virus was compared with a virus isolated from Aedes vexans mosquitoes and designed 743 virus. The viruses were compared with respect to cell culture host range, cellular changes resulting from viral infections, growth curves, antigenic relationship, and physicochemical characteristics. The viruses are closely comparable in all these properties, and they are considered to be strains of the same virus. In cross comparisons by the enzyme-linked immunosorbent assay, 743 virus and infectious bursal disease virus were found to be antigenically identical, confirming the results of the neutralization test. The 743 virus differs from most strains of infectious bursal disease virus in that it is nonpathogenic for chickens.  相似文献

按照口蹄疫病毒(FMDV)聚合酶3D基因序列,设计合成了引物和探针,经各反应务件的优化,建立了实时荧光定量RT-PcR技术,对细胞培养物、水泡液、水泡皮及分泌物、血液中的FMDV进行了特异性检测和敏感性试验。结果,用300nmol/L的引物浓度和200nmol/L探针浓度,获得的CT值较小,而△Rn最大;可检测到相当于9.1TCID50的病毒RNA;与VSV和其他水泡性病毒不发生交叉反应;制作的标准曲线中各浓度范围内有极好的线性关系,且线性范围宽,相关系数为0.984;组内和组间试验重复性的变异系数(CV)分别为5.4%和6.7%;与常规RT-PCR相比较,该方法具有快速、特异、敏感、可定量,并可同时检测大量样品等优点。  相似文献

Akabane and bovine ephemeral fever viruses are exotic to the American continent. Both viruses are spread by insect vectors, and each causes disease of varying severity in food-producing animals. However, there are few other similarities between the agents and the diseases that they cause. They do not share the same insect vectors, the mammalian host range is different, and the clinical manifestations of virus infection vary markedly. Akabane virus is a cause of severe congenital defects, but adult animals show no signs of infection. In contrast, bovine ephemeral fever virus causes a febrile illness affecting mainly mature animals. If introduced to North America, it is probable that there would be significant economic losses, at least until endemic virus transmission patterns were established. Subsequently, it is likely that there would be patterns of alternate disease outbreaks followed by interepidemic periods in which there is a minor clinical effect.  相似文献

Transfer and low temperature storage of embryos are now proven techniques for a number of mammalian species. These techniques are useful in control of disease and in saving genotypes from infected animals. The place of embryos in the epidemiology of disease depends upon whether the causative organism can gain entry to the oocyte before or at fertilisation and on whether the young embryo can be invaded by organisms in the uterine environment. There is little evidence that important live-stock diseases are transmitted via gametes. The zona pellucida surrounding the embryo is an effective barrier against a number of important disease organisms; in some cases the embryo is susceptible once it has hatched from the zona pellucida. It is important therefore in considering the use of embryos in disease control, to ensure that virus is not attached to the surface of the zona pellucida from where it can infect the recipient and/or the embryo after hatching. Washing procedures have been devised together with the use of enzymes and antisera to remove virus from the surface of embryos. Some viruses enter pores and sperm tracks in the zona and removal of these may present a problem. African swine fever virus has been shown to resist removal by treatment with enzymes. There are no guidelines as to the likely interaction between a certain virus and embryos. Therefore each virus of interest must be tested to determine whether it can be transmitted via washed embryos. Nevertheless there are numerous instances of the use of embryo transfer to eradicate a specific disease or to save valuable genetic material from infected animals without transmitting disease.  相似文献

Borna disease virus represents an unknown neurotropic agent. It causes encephalitis in horses and sheep. The same or a similar type of virus might be responsible for psychiatric disorders in man. So far, it has been impossible to purify this agent to such an extent that it could be analyzed biochemically or electronmicroscopically. Therefore, different conventional virus purification techniques are applied in order to develop a method for obtaining purified Borna disease virus from infectious rat brain or persistently infected cell culture material.  相似文献

Foot-and-mouth disease (FMD) is a highly contagious and economically devastating disease of cloven-hoofed animals in the world. The disease can be effectively controlled by vaccination of susceptible animals with the conventional inactivated vaccine. However, one major concern of the inactivated FMD virus (FMDV) vaccine is that it does not allow serological discrimination between infected and vaccinated animals, and therefore interferes with serologic surveillance and the epidemiology of disease. A marker vaccine has proven to be of great value in disease eradication and control programs. In this study, we constructed a marker FMDV containing a deletion of residues 93 to 143 in the nonstructural protein 3A using a recently developed FMDV infectious cDNA clone. The marker virus, r-HN/3A93–143, had similar growth kinetics as the wild type virus in culture cell and caused a symptomatic infection in pigs. Pigs immunized with chemically inactivated marker vaccine were fully protected from the wild type virus challenge, and the potency of this marker vaccine was 10 PD50 (50% pig protective dose) per dose, indicating it could be an efficacious vaccine against FMDV. In addition, we developed a blocking ELISA targeted to the deleted epitope that could clearly differentiate animals infected with the marker virus from those infected with the wild type virus. These results indicate that a marker FMDV vaccine can be potentially developed by deleting an immunodominant epitope in NSP 3A.  相似文献

A horse with neurological signs and severe meningoencephalitis caused by Aujeszky's disease is described. The diagnosis was established by immunohistochemistry, DNA-in situ hybridization and serological tests. Aujeszky's disease virus antigen and Aujeszky's disease viral DNA were detected in neurons of the cerebrum. In the serum of the horse antibodies against Aujeszky's disease virus were detected in a virus neutralization test, in a blocking ELISA which specifically detects antibodies against the glycoprotein I (Ig) of the virus, in an indirect double sandwich ELISA and with colloidal gold immunoelectron microscopy which detects antibodies directed against the envelope and nucleocapsid of the virus. Intranasal infection of two points with a high dose of Aujeszky's disease virus caused very wild and transient signs. Although the experimental infection induced virus neutralizing antibodies, it failed to induce gI specific antibodies.  相似文献

施马伦贝格病毒病(Schmallenberg virus,SBV)是一种新发现的动物传染病,因于2011年底在德国施马伦贝格镇首次发现而临时得名,随后蔓延于西欧(包括比利时、法国、德国、荷兰、意大利、卢森堡、西班牙、英国和丹麦),并分别在奥地利、波兰、瑞典和芬兰等国的牛、山羊、绵羊中检测到抗体。遗传分析显示该病毒与布尼亚病毒科(Bunyaviridae)正布尼亚病毒属(Orthobunyavirus)西姆布血清群病毒(Simbu serogroup viruses)的亲缘关系最密切,西姆布血清群病毒是已知的反刍动物病原,可通过节肢动物媒介(蚊、蠓)传播。施马伦贝格病毒病有2种不同的临床症状:成年牛出现短暂轻微/温和的病症(产奶量减少、发热、腹泻)和新生哺乳动物(牛、羊)死产和先天缺陷。因为同群类似的病毒不是人畜共患病病原,也无该病毒致人发病的证据,但现阶段尚不能完全排除。尽管目前没有特效的药物和疫苗,但因已有类似病毒(赤羽病)的疫苗,疫苗接种应是控制该病的可能选项。因施马伦贝格病毒是一种新发现的病毒,许多方面尚不清楚,还有待于进一步研究。  相似文献

During investigations on the virus diarrhea-mucosal disease complex, a virus (KO-114) was isolated from a six-month-old calf, and on the basis of physicochemical characteristics the virus was classified as an enterovirus. The hemagglutinating properties indicate that it belongs to La Placa’s group I. Examination of 103 bovine sera for neutralizing capacity against virus KO-114 gave very low titers only 13 % having an index between log. 1 and log. 2. Human sera did not neutralize this virus, which is not related to the virus diarrhea-mucosal disease complex.  相似文献

Classical swine fever (CSF) is a notifiable disease of domestic pigs and wild boar. It is caused by the highly contagious CSF virus and in its acute form the disease generally results in high morbidity and mortality. Due to the great economical impact an outbreak can cause to the pig industry it is one of the most important swine diseases worldwide. To limit the damage in the case of a new outbreak it is necessary to identify the virus as fast as possible. This information helps epidemiologists to trace the origin of the virus and to follow the virus spread. Genetic typing revealed that CSF virus genotypes, subgroups and types show a regional distribution making it an important tool for epidemiologists. Meanwhile, besides epidemiological data and nucleotide sequences from European isolates, information from isolates from South- and Central America, the Caribbean, Asia and recently from South Africa have become available. The data are stored in a database in the EU Reference Laboratory for CSF, accessible by the WWW (http://viro08.tiho-hanno ver.de). A new module was implemented that allows efficient automated genotyping.  相似文献

Peste des petits ruminants (PPR) is a viral disease of sheep and goats that is spreading through many countries in the developing world. Work on the virus is often restricted to studies of attenuated vaccine strains or to work in laboratories that have high containment facilities. We have created a helper cell dependent form of PPR virus by removing the entire RNA polymerase gene and complementing it with polymerase made constitutively in a cell line. The resultant L-deleted virus grows efficiently in the L-expressing cell line but not in other cells. Virus made with this system is indistinguishable from normal virus when used in diagnostic assays, and can be grown in normal facilities without the need for high level biocontainment. The L-deleted virus will thus make a positive contribution to the control and study of this important disease.  相似文献

The presence of Aujeszky's disease virus in cerebrospinal fluid of experimentally infected pigs was studied using the techniques of virus isolation and PCR. Pigs, some of which were previously vaccinated against Aujeszky's disease, were inoculated with different doses of the Aujeszky's disease NIA-3 strain. At the time of death or sacrifice, a sample of cerebrospinal fluid was taken and tested for the presence of virus using the mentioned techniques. Virus was isolated only from one sample, while it was detected by PCR in most of them. The higher sensitivity of the PCR technique and the possible presence of antiviral antibodies in the cerebrospinal fluid are reasons that can be argued to explain this fact. By PCR, the virus was detected more efficiently when digested cerebrospinal fluid cells were used as DNA source than when using whole cerebrospinal fluid, suggesting that the virus could be cell-associated. Aujeszky's disease virus could not be detected by PCR in pigs which survived the acute phase of the infection and were euthanased at 8 weeks post-inoculation, when they were latently infected. This indicated that the cerebrospinal fluid is not an adequate sample for the diagnosis of latency. Since Aujeszky's disease virus was detected from most of the tested samples, we believe that this could be an adequate procedure for the quick diagnosis of Aujeszky's disease.  相似文献

Sera from more than 100 alpacas (Lama pacos) from the Peruvian southern sierra were examined for antibodies to 8 viruses known to infect other domestic animals. On the basis of these serologic findings and previously published serologic or clinical data, it is now known that the alpaca can be infected with the following viruses: parainfluenza-3, bovine respiratory syncytial virus, bovine herpesvirus-1, bluetongue virus, border disease virus, influenza A virus, rotavirus, rabies virus, vesicular stomatitis virus, foot-and-mouth disease virus, and contagious ecthyma virus.  相似文献

An outbreak of mucosal disease that occurred among Friesian heifers aged 15 to 18 months is described as it occurred. Japanese encephalitis (JE) virus is a mosquito-borne flavivirus that can cause encephalitis and death in horses and humans. It is an emerging disease of international concern because it has been spreading into previously nonendemic areas. Major epidemics may occur where the virus moves into new areas, but many infections are subclinical. This article presents information on the virus, its epidemiology, and what little information has been published on the disease in horses. The methods available for the diagnosis and control of JE are described and issues raised, particularly for those countries faced with the threat of imminent incursions as the virus extends its range.  相似文献

Ephemeral fever remains a viral disease of considerable importance to many countries including Australia. The virus has been only partly characterised and still awaits final classification. Although BEF virus was first thought to contain 6 structural proteins there is increasing evidence to suggest that it contains the 5 proteins characteristic of the Rhabdoviridae. Although BEF is thought to be arthropod borne, the vector has yet to be identified but it is clear from the distribution of BEF that more than one vector is capable of transmitting the disease. Despite rigorous investigation of the clinical signs and the pathology of ephemeral fever, little progress has been made on the pathogenesis of the disease. This has been partly due to the difficulty of propagating BEF virus in vitro and the inability to define the site of replication. However, there is mounting evidence to suggest that BEF is immunopathologic in nature and that the clinical expression of the disease is influenced by the release of one or more mediators of inflammation. The disease is characterised by a number of haematological and biochemical changes and early and prolonged treatment with phenylbutazone is capable of reversing a number of these changes. The intravenous administration of calcium can now be considered a justifiable addition to the treatment regimen together with prolonged phenylbutazone therapy. The vaccines currently available are prepared from either live attenuated or killed virus and may be less than reliable. There appears to be a need for a reliable, inexpensive, cold-chain independent alternative vaccine.  相似文献

The current, generally accepted approach to formulating health requirements for the international movement of embryos is to base them on the health status of the male and female donor animals. The alternative approach of basing them on the health status of the embryos themselves has been blocked by the lack of scientific information about the potential of the early embryo to transmit agents of infectious disease. Consequently, most research into infectious disease transmission by embryos has had the objective of assessing the potential of the embryo to transmit infectious disease, at the stage of development at which it is transferred commercially, with the thought in mind that, for some diseases, it may be possible in the future to focus on the embryo rather than the donor when drawing up health requirements for import permits. Results from experiments involving the bovine leukemia virus, bluetongue virus, infectious bovine rhinotracheitis virus, foot and mouth disease virus and Brucella abortus are encouraging to the point where, with the exception of foot and mouth disease virus, they could and should be put to the test in field studies. Research on several other bovine pathogens is underway, but the studies are not sufficiently advanced for a judgement to be made on the potential of embryos to transmit them. There is evidence that the research done is starting to have a positive effect through the relaxation of some health requirements for the international movement of embryos.  相似文献

The spread of antibody to Aujeszky's disease virus through a susceptible pig herd was monitored after the probable introduction of infection by a recently purchased boar. The infection spread slowly through the herd but no clinical signs of Aujeszky's disease were seen. The strain of virus isolated was designated NIA-6. It has been characterised by a series of experimental infections and extends the known range of virulence of isolates of Aujeszky's disease virus made in Northern Ireland. The strain caused no disease in four-week-old piglets and is therefore less virulent than other isolates from Northern Ireland pigs. However, it killed rabbits and a proportion of experimentally infected two-week-old piglets, which differentiates it from the avirulent bovine isolate (NIA-4).  相似文献
