Direct link between mhc polymorphism,T cell avidity,and diversity in immune defense

Major histocompatibility complex (mhc)-encoded molecules govern immune responses by presenting antigenic peptides to T cells. The extensive polymorphism of genes encoding these molecules is believed to enhance immune defense by broadening the array of antigenic peptides available for T cell recognition, but direct evidence supporting the importance of this mechanism in combating pathogens is limited. Here we link mhc polymorphism-driven diversification of the cytotoxic T lymphocyte (CTL) repertoire to the generation of high-avidity, protective antiviral T cells and to superior antiviral defense. Thus, much of the beneficial effect of the mhc polymorphism in immune defense may be due to its critical influence on the properties of the selected CTL repertoire.     

GnIH在陆川仔公猪泌尿生殖系统及内分泌免疫系统中的分布

【目的】从形态学水平和分子水平探究促性腺激素抑制激素(GnIH)在陆川仔公猪泌尿生殖系统和内分泌免疫系统中的分布情况。【方法】采集30日龄健康陆川仔公猪的睾丸、附睾、附睾管、肾、肾上腺、甲状腺、腭扁桃体、脾、淋巴结、胸腺,采用免疫组织化学和半定量RT-PCR的方法检测GnIH在其泌尿生殖系统和内分泌免疫系统的分布情况。【结果】免疫组织化学分析结果显示,在睾丸、附睾、附睾管、肾、肾上腺、甲状腺、腭扁桃体、脾、淋巴结、胸腺均有不同数量的GnIH免疫阳性细胞分布。半定量RT-PCR的检测显示,GnIH mRNA除了在甲状腺、脾和淋巴结无表达外,在其余组织均有不同程度的表达。【结论】GnIH在泌尿生殖系统和内分泌免疫系统分布广泛,提示GnIH在生殖、免疫和内分泌等生理过程中可能有重要作用。     

Asef, a link between the tumor suppressor APC and G-protein signaling

The adenomatous polyposis coli gene (APC) is mutated in familial adenomatous polyposis and in sporadic colorectal tumors. Here the APC gene product is shown to bind through its armadillo repeat domain to a Rac-specific guanine nucleotide exchange factor (GEF), termed Asef. Endogenous APC colocalized with Asef in mouse colon epithelial cells and neuronal cells. Furthermore, APC enhanced the GEF activity of Asef and stimulated Asef-mediated cell flattening, membrane ruffling, and lamellipodia formation in MDCK cells. These results suggest that the APC-Asef complex may regulate the actin cytoskeletal network, cell morphology and migration, and neuronal function.     

Myosin: a link between streptococci and heart

Murine monoclonal antibodies to Streptococcus pyogenes reacted with skeletal muscle myosin. High molecular weight proteins in extracts of human heart tissue that reacted with an antibody to S. pyogenes also reacted with a monoclonal antibody to ventricular myosin. Adsorption of the antibody to streptococci with S. pyogenes simultaneously removed reactivity of the antibody for either S. pyogenes or myosin. These results indicate that myosin shares immunodeterminants with a component of S. pyogenes.     

Habituation and dishabituation: interactions between peripheral and central nervous systems in Aplysia

The withdrawal response of the isolated siphon of Aplysia habituates to a light stimulus and dishabituates to a tactile stimulus, and vice versa, with or without connections to the central nervous system. The peripheral nervous system can dishabituate or enhance the response mediated by the central nervous system and vice versa. Normally the adaptive siphon withdrawal response of the intact animal must be mediated by the integrated activity of the peripheral and central nervous systems.     

Diversity and function of adaptive immune receptors in a jawless vertebrate

Instead of the immunoglobulin-type antigen receptors of jawed vertebrates, jawless fish have variable lymphocyte receptors (VLRs), which consist of leucine-rich repeat (LRR) modules. Somatic diversification of the VLR gene is shown here to occur through a multistep assembly of LRR modules randomly selected from a large bank of flanking cassettes. The predicted concave surface of the VLR is lined with hypervariable positively selected residues, and computational analysis suggests a repertoire of about 10(14) unique receptors. Lamprey immunized with anthrax spores responded with the production of soluble antigen-specific VLRs. These findings reveal that two strikingly different modes of antigen recognition through rearranged lymphocyte receptors have evolved in the jawless and jawed vertebrates.     

Mast cells: a cellular link between autoantibodies and inflammatory arthritis

Previous studies have revealed that autoantibodies, complement components, and Fc receptors each participate in the pathogenesis of erosive arthritis in K/BxN mice. However, it is not known which cellular populations are responsive to these inflammatory signals. We find that two strains of mice deficient in mast cells, W/Wv and Sl/Sld, were resistant to development of joint inflammation and that susceptibility was restored in the W/Wv strain by mast cell engraftment. Thus, mast cells may function as a cellular link between autoantibodies, soluble mediators, and other effector populations in inflammatory arthritis.     

Itokawa dust particles: a direct link between S-type asteroids and ordinary chondrites

The Hayabusa spacecraft successfully recovered dust particles from the surface of near-Earth asteroid 25143 Itokawa. Synchrotron-radiation x-ray diffraction and transmission and scanning electron microscope analyses indicate that the mineralogy and mineral chemistry of the Itokawa dust particles are identical to those of thermally metamorphosed LL chondrites, consistent with spectroscopic observations made from Earth and by the Hayabusa spacecraft. Our results directly demonstrate that ordinary chondrites, the most abundant meteorites found on Earth, come from S-type asteroids. Mineral chemistry indicates that the majority of regolith surface particles suffered long-term thermal annealing and subsequent impact shock, suggesting that Itokawa is an asteroid made of reassembled pieces of the interior portions of a once larger asteroid.     

The Fusarium graminearum genome reveals a link between localized polymorphism and pathogen specialization

We sequenced and annotated the genome of the filamentous fungus Fusarium graminearum, a major pathogen of cultivated cereals. Very few repetitive sequences were detected, and the process of repeat-induced point mutation, in which duplicated sequences are subject to extensive mutation, may partially account for the reduced repeat content and apparent low number of paralogous (ancestrally duplicated) genes. A second strain of F. graminearum contained more than 10,000 single-nucleotide polymorphisms, which were frequently located near telomeres and within other discrete chromosomal segments. Many highly polymorphic regions contained sets of genes implicated in plant-fungus interactions and were unusually divergent, with higher rates of recombination. These regions of genome innovation may result from selection due to interactions of F. graminearum with its plant hosts.     

The Neurospora checkpoint kinase 2: a regulatory link between the circadian and cell cycles

The clock gene period-4 (prd-4) in Neurospora was identified by a single allele displaying shortened circadian period and altered temperature compensation. Positional cloning followed by functional tests show that PRD-4 is an ortholog of mammalian checkpoint kinase 2 (Chk2). Expression of prd-4 is regulated by the circadian clock and, reciprocally, PRD-4 physically interacts with the clock component FRQ, promoting its phosphorylation. DNA-damaging agents can reset the clock in a manner that depends on time of day, and this resetting is dependent on PRD-4. Thus, prd-4, the Neurospora Chk2, identifies a molecular link that feeds back conditionally from circadian output to input and the cell cycle.     

浙东白鹅2周龄体重、体尺和免疫器官的发育

对2周龄的浙东白鹅体重、体尺和免疫器官生长发育情况进行了测定分析,结果表明,2周龄浙东白鹅平均体重为452.73±60.46g,平均日增重为24.45 g.体重和背长分别比1989年初浙东白鹅品种选育5世代种鹅高21.25%和22.47%,差异均达极显著,同时群体均匀度提高.体重与背长的相关性较大(r=0.609 3).法氏囊、胸腺、脾脏随着体重的增加而不断增大.胸腺重个体差异缩小(CV值由初生时的60.53%下降到2周龄的21.01%).     

Understanding Conflicts between People and Parks at Ranomafana, Madagascar

The national park model originating in the unique circumstances of mid-19th century North America has been widely applied in the developing countries of the late 20th century, provoking numerous land-use conflicts between parks and resident peoples. Key factors in understanding these conflicts are examined using the field experience of the Ranomafana National Park in Madagascar. A conflict management strategy is suggested for alleviating such antagonism and facilitating the investigation of mutually acceptable conservation and development pathways.     

The metamorphosis of supernova SN 2008D/XRF 080109: a link between supernovae and GRBs/hypernovae

The only supernovae (SNe) to show gamma-ray bursts (GRBs) or early x-ray emission thus far are overenergetic, broad-lined type Ic SNe (hypernovae, HNe). Recently, SN 2008D has shown several unusual features: (i) weak x-ray flash (XRF), (ii) an early, narrow optical peak, (iii) disappearance of the broad lines typical of SN Ic HNe, and (iv) development of helium lines as in SNe Ib. Detailed analysis shows that SN 2008D was not a normal supernova: Its explosion energy (E approximately 6x10(51) erg) and ejected mass [ approximately 7 times the mass of the Sun (M(middle dot in circle))] are intermediate between normal SNe Ibc and HNe. We conclude that SN 2008D was originally a approximately 30 M(middle dot in circle) star. When it collapsed, a black hole formed and a weak, mildly relativistic jet was produced, which caused the XRF. SN 2008D is probably among the weakest explosions that produce relativistic jets. Inner engine activity appears to be present whenever massive stars collapse to black holes.     

Structural relaxation of polymer glasses at surfaces, interfaces, and in between

We analyzed the glassy-state structural relaxation of polymers near surfaces and interfaces by monitoring fluorescence in multilayer films. Relative to that of bulk, the rate of structural relaxation of poly(methyl methacrylate) is reduced by a factor of 2 at a free surface and by a factor of 15 at a silica substrate interface; the latter exhibits a nearly complete arresting of relaxation. The distribution in relaxation rates extends more than 100 nanometers into the film interior, a distance greater than that over which surfaces and interfaces affect the glass transition temperature.
