1. Tissue-specific profiles of the expression of the antioxidant enzymes superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT) and the concentrations of reduced glutathione (GSH) during the development of the chick embryo were investigated. 2. The liver, brain, yolk sac membrane (YSM), kidney, lung, heart and skeletal muscles were collected at the following days of embryo development: 10, 11, 13, 15, 17, 19, 21 and 22 (day-old chicks). 3. The different tissues of the embryo displayed distinct development strategies with regard to the acquisition of antioxidant capacity. In the liver the specific activity of SOD increased between days 10 and 11 of development, then significantly decreased up to day 15 and remained at the same value during the rest of the developmental period. GSH-Px specific activity increased through the time of development. CAT had 2 peaks of specific activity at day 10 of the development and in day-old chicks. 4. The brain was characterised by comparatively high SOD-specific activity especially during the last days of incubation. The specific activities of GSH-Px and CAT were low throughout development. 5. In the YSM maximal GSH-Px and CAT-specific activities were found on day 15 of incubation. In the kidney and heart GSH-Px-specific activity increased at hatching time. CAT-specific activity in the kidney increased just after hatching. 6. It is concluded that each tissue studied expressed a profile of antioxidant defence mechanisms to deal with oxidative stress at hatching time.  相似文献

Tylosin tartrate, administered in the drinking water at a concentration of 0.55 g/litre for the first three days after hatching, was highly effective in controlling the adverse consequences of a Mycoplasma gallisepticum infection, established by air sac injection at one day of age, in turkey poults. Tylosin was ineffective in controlling M meleagridis infections established in embryo or at one day of age when administered in the drinking water of poults. Both mycoplasma isolates used were inhibited in vitro by a tylosin concentration of 0.1 mug/ml.  相似文献

A viral agent that was isolated from livers of commercial turkey poults that died at approximately two weeks of age was characterized as a reovirus. Experimental infection of day-old chickens with this reovirus isolate resulted in the development of tenosynovitis, hepatitis, and myocarditis. In vitro neutralization of the turkey reovirus isolate by antiserum against chicken reovirus correlated with in vivo protection of maternally immune chickens from day-old oral challenge with the turkey reovirus isolate.  相似文献

The aim of this study was to verify the hypothesis that the health and growth of turkey poults may be improved by supplementing diets fed to parent flocks with available selenium. Experimental poults originated from parent flocks fed with diets containing 0.3 mg/kg inorganic selenium (control group Se(M)) and organic selenium (experimental group Se(O)). Egg yolk selenium content was comparable in both flocks (0.72 and 0.70 mg/kg d.m., respectively). Eggs from the Se(O) flock had a significantly lower content of thiobarbituric acid reactive substances - TBARS (31.13 vs. 53.10 nmol/g, p > 0.001). Se(O) group poults were characterized by higher activity of glutathione peroxidase (7.54 vs. 5.92 U/mL, P = 0.001) and superoxide dismutase (89.30 vs. 79.23 U/mL, P = 0.026). The thigh muscles of Se(O) group birds had significantly higher selenium concentrations (0.74 vs. 0.57, p = 0.045) and a significantly lower TBARS content (38.42 vs. 65.01, p = 0.001). No differences were found between the groups with respect to the content of total protein, albumins and uric acid, and the activites of alanine aminotransferase (ALT), aspartate aminotransferase (AST) and lactate dehydrogenase (DLH) in day-old poults. On day 28, groups Se(O) and Se(M) differed in the activity of ALT (20.50 vs. 26.33, p = 0.05) and SOD (87.29 vs. 100.02 U/mL, p = 0.035). There were no differences between the groups regarding the percentages of T lymphocyte subpopulations CD4+, CD8+, CD4+CD8+ and B lymphocyte subpopulations (IgM+) at 1 and 28 days of age. Over the experimental period, mortality rates were similar in both groups (7.32 and 8.87%), and so were the final body weights of birds (1108 vs. 1135 g). The results of the study show that the dietary supplementation of organic selenium in turkey parent flocks reduces the rate of oxidation processes in the egg and in the tissues of newly-hatched poults, yet it has no effect on the analyzed parameters of cell-mediated immunity and the growth performance of birds during the first five weeks of their life.  相似文献

Introduction: Most of the morphological studies on the skin of birds refer to chickens. Little can be found about the specific fine structure of the foot‐pad epidermis of turkeys and almost nothing is known about the developing reticulate scales in turkey foetuses. This study deals with the question as to whether the development and morphology of the reticulate scales of turkeys are similar to those of chickens. Since the first signs of food‐pad dermatitis can be observed in turkey poults at the age of four days, it was additionally examined if these alterations can be found in turkey foetuses already. Methods: Foot pad samples of heavy strain BUT Big 6 turkeys were collected at different developmental stages (day 20, 23, and 28 of incubation). The specimens were compared by light‐ and electron‐microscopic methods. Results: On the 20th day of incubation, the reticulate scales cover the food pads as small knob‐like structures. Between the embryonic epidermal cells wide inter‐cellular spaces are visible, and in the intermediate cells multigranular bodies (MGBs) are detectable. Peridermal cells cover the reticulate scales. They are characterized by peridermal granules and protuberances of the outer cell membrane. On the 23rd day of incubation, many lipid droplets are scattered within the intermediate cells. Because a thin stratum corneum has formed, the periderm is almost completely sloughing off. On the 28th day of incubation, the turkey poults hatch. The epidermis has almost reached maturity and a broad stratum corneum is detectable. Conclusions: The development and morphology of the reticulate scale epidermis of turkey is comparable to that of chicken, although little differences were found. Besides former investigations, which described the peridermal protuberances as microvilli, scanning‐electron microscopy proved that they also show a fingerprint‐like surface structure that has not been observed for birds previously. Up to the hatching day, no signs of food pad alterations are seen in developing turkeys.  相似文献

Day-old specific-antibody-negative turkey poults were inoculated orally with cloned turkey reovirus isolate 81-68. Virus reisolations from 11 different tissues revealed widespread distribution at 3, 5, and 7 days postinoculation (PI). Virus was isolated from the intestines until 21 days PI. Virus was isolated from tendons until day 7 PI and again at day 28 PI. Reovirus serum-neutralization antibodies appeared as early as 7 days PI. All inoculated birds showed positive VN serum titers (greater than or equal to 1:20) by day 21 PI. No reovirus was isolated from control poults, and they remained antibody-negative during the entire experiment.  相似文献

Turkey viral hepatitis (TVH) was experimentally reproduced in two experiments in 1-day-old poults. In the first experiment, an infectious inoculum was prepared from filtered yolk materials harvested from dead embryonating chicken eggs (ECE) previously inoculated with suspensions of liver and pancreas tissues collected from TVH-affected birds in commercial turkey flocks. One-day-old poults given a yolk-sac inoculation or oral gavage with this preparation developed lesions in the liver and pancreas characteristic of TVH at 20 days postinoculation (PI) in 60% and 14% of the experimentally infected birds, respectively. With the identical inoculum, embryo mortality occurred at 8 and 10 days PI in embryonating turkey eggs (ETE) inoculated into the yolk sac. In the second experiment, an infectious inoculum was prepared from filtered yolk materials from dead ETE harvested in the first experiment. One-day-old poults given a yolk-sac inoculation with this filtered yolk material developed lesions in the liver and pancreas within 5 days PI. At 20 days PI, 67% of the experimentally infected birds had similar lesions. With the inoculum given to these poults, embryo mortality occurred at 6, 8, and 10 days PI in ETE inoculated into the yolk sac. Virus particles 26-28 nm in diameter with icosahedral morphology typical of picornaviruses were identified by EM in the yolk sacs of ETE that died in both experiments, and inoculated ETE that died following passage of filtered suspensions of pancreatic tissues collected from affected birds in the first experiment.  相似文献

A strain of Escherichia coli isolated from the yolk sac of stunted turkey poults was administered orally to day-old large white poults. Poults were inoculated with either 0.1 ml of sterile broth or 0.1 ml of a 10(-2) dilution of a 24-hr E. coli culture containing 3.4 x 10(8) viable bacteria per ml. Two levels of dietary protein (28 or 22%) were fed from 1 day to 3 weeks of age. Following E. coli inoculation of 3.4 x 10(5) viable bacteria at day one, body weight gain and feed consumption from 0 to 3 weeks of age were numerically increased 4.5 and 2.1%, respectively, and feed efficiency was significantly increased 2.4%. E. coli had a greater effect on performance of poults fed the 28% protein diet than on poults fed the 22% protein diet. Metabolism studies, conducted from 7 to 10 and from 17 to 20 days postinoculation, showed no significant changes in the measurements of nutrient utilization due to E. coli other than a 17% increase in nitrogen retention from 17 to 20 days by those poults fed the 28% protein diet.  相似文献

In this study, the influence of a branched‐chain amino acid blend (BCAA composed of 3 l ‐leucine:1 l ‐valine:2 l ‐isoleucine) injected into the amniotic fluid was evaluated for embryonic growth, yolk‐sac (YS) utilization and development of gastrointestinal tract (GIT) and skeletal muscles of turkey embryos from day 24 of incubation (24E) to hatching, together with hatchability, poult quality and liver L* (lightness), a* (redness) and b* (yellowness) values at hatch. At day 22 of incubation, embryonated eggs (n = 240) were assigned to three treatments, that is, eggs were not injected (control, NC) or injected with 1.5 ml sterile solution with 0.9% salt (SA) or 0.2% BCAA blend (BCAAb). These solutions were injected manually into the amniotic fluid of the embryonated eggs. To determine weights and lengths (where appropriate) of the studied organs and tissues, four embryonated eggs and poults per treatment were selected at 24E and at hatch. While the BCAAb decreased the YS and embryo weight, hatchability and the liver L* value, it increased the weight and quality of poults and the weights of breast and thigh muscles at hatch. In conclusion, the in ovo feeding of the BCAA blend negatively affected hatchability but positively affected hatching weight and poult quality by improving development of skeletal muscles and by regulating energy metabolism.  相似文献

Turkey poults, free of antibodies to turkey rhinotracheitis (TRT) virus were treated with cyclophosphamide on days 1, 2 and 3 after hatching and vaccinated by eyedrop when 10 days old with a Vero cell-attenuated preparation of TRT virus. No ELISA antibodies to TRT virus developed in the sera of these poults but they were as resistant to virulent virus challenge 21 days later as vaccinated groups which were not cyclophosphamide-treated but produced humoral antibodies. Following challenge with virulent virus at 31 days old cyclophosphamide-treated unvaccinated poults developed a more severe clinical response than untreated birds and had higher virus titres in tracheal swabs. The findings show that the respiratory tract of turkeys may be resistant to TRT despite the absence of ELISA antibodies in the serum.  相似文献

Lasalocid and monensin are widely used to control coccidiosis in broilers, but not in turkey poults. Four feeding trials were conducted to determine the performance of turkey poults when these compounds were used singly or in combination with 100 ppm of furazolidone. Bodyweights and feed consumption were significantly depressed for five weeks after hatching by 150 ppm of lasalocid. Combining furazolidone with lasalocid ameliorated the toxic effect of lasalocid. Bodyweights were significantly depressed by 150 ppm of monensin in the fifth week after hatching, but there was no significant depression in feed consumption. Furazolidone exacerbated any toxic effects of monensin. Data indicate that monensin may be used safely at dosages greater than the recommended level of 60 to 99 ppm, but should not be used in combination with furazolidone.  相似文献

Listeria monocytogenes is a ubiquitous, environmental pathogen that has contaminated poultry ready-to-eat products resulting in large-scale recalls. Research is needed to determine the source of product and processing plant contamination with L. monocytogenes. The purpose of this study was to compare the oral and oculonasal routes of infection on the pathogenicity of L. monocytogenes in turkey poults under different housing conditions. One-day-old turkey poults were challenged by either route with the Scott A strain of L. monocytogenes and placed either in paper-lined battery-brooder cages for 1 wk or in floor pens on fresh pine-shaving litter. On day 7, birds challenged in battery cages were transferred to floor pens. Challenge by the oculonasal route resulted in higher mortality (P = 0.05) and lower body weights (P < 0.0001) compared with both nonchallenged controls and those challenged by the oral route. Birds contained in battery cages for 1 wk had higher mortality (P = 0.002) and higher body weights (P < 0.0001) compared with floor-pen-reared birds. Using direct plating, the challenge strain was isolated from the gall bladder, brain, and knee joint of only one dead poult challenged by the oculonasal route. These results suggest that day-old turkey poults may be more susceptible to an oculonasal challenge with L. monocytogenes than to an oral challenge and that containment in battery cages for the first week increased contact exposure to the challenge.  相似文献

We designed this study to compare the replication potential of turkey coronavirus (TCV) and its effect in chickens and turkeys and to study the effect of singleand combined infection of turkey poults with TCV and astrovirus. We studied the pathogenicity of TCV in experimentally inoculated turkey poults and chickens by observing the dinical signs and gross lesions. Two trials were conducted with 1-day-old and 4-wk-old specific-pathogen-free turkey poults and chickens. One-day-old turkey poults developed diarrhea at 48 hr postinoculation. Poults euthanatized at 3, 5, and 7 days postinoculation had flaccid, pale, and thin-walled intestines with watery contents. The 4-wk-old turkeys had no clinical signs or gross lesions. One-day-old and 4-wk-old chicks developed no clinical signs or gross lesions although the TCV was detected in gut contents of the birds throughout the experimental period (14 days). In another experiment, mean plasma D-xylose concentrations in 3-day-old turkey poults inoculated with TCV, turkey astrovirus, or a combination of both viruses were significantly lower than in the uninoculated controls.  相似文献

Poults free from hemorrhagic enteritis (HE) antibody were vaccinated by gavage at 1 day or 2 weeks of age with a live HE vaccine virus that had been propagated in a Marek's disease (MD)-induced B-lymphoblastoid cell line of turkey origin. Vaccinated and unvaccinated poults were challenged with a virulent HE virus at various times postvaccination. One hundred tissue-culture-infectious doses of the vaccine virus per poult were sufficient to induce a serological response as well as to protect poults against HE lesions and mortality. Vaccinated poults were protected against the disease as early as 1 week and as late as 8 weeks PV. The vaccine was efficacious by several routes of application. The vaccine virus spread horizontally from vaccinated to contact-exposed poults, as indicated by seroconversion and resistance of contact-exposed poults to challenge. The vaccine had no detectable harmful effects on the humoral immune response to particulate antigens or on weight gain of vaccinated poults. The vaccine proved to be free from MD virus, as indicated by the absence of MD lesions and antibody in 8-week-old chickens inoculated intra-abdominally with the vaccine at hatching. These findings indicate that the cell-culture-propagated HE vaccine is efficacious and safe.  相似文献

Parthenogenetic development was found in 16.7 per cent of 934 Beltsville Small White turkey eggs in 1952. This development, occurring during incubation, was chiefly of an unorganised type, blood or embryos being encountered only on rare occasions. In 1954, a selective breeding programme was initiated which was designed to intensify this trait. This has resulted in marked yearly increases in the average level of parthenogenetic development. Since 1952 there has been a three‐fold increase in the overall incidence of parthenogenesis and presently 45 per cent of all unfertilised eggs incubated can be expected to undergo development to some extent. The most marked increases have come in the number of eggs giving rise to well‐formed embryos. At the present time from 12–13 Per cent of all eggs can be expected to contain embryos, representing a 65‐fold increase over the level found in eggs of the original stock. Corresponding increases have likewise occurred in embryo viability. The few embryos encountered before 1954 rarely attained the size of a normal 5–6 day embryo. During the last three years approximately 34 per cent of all parthenogenetic embryos lived for 21 days or longer and 7 of every 100 of them emerged or were helped from the shell on the 29th or 30th day of incubation. One hundred and six parthenogenetic poults were hatched from 10,060 eggs in 1961, 94 from 8519 eggs in 1962, and 23 from 5906 eggs in 1963.

About 25 per cent of the parthenogenetic poults which have hatched survived to maturity. Some of these have produced semen containing viable spermatozoa. More than 25 parthenogenetic males thus far have demonstrated their ability to sire offspring.  相似文献

Tissue responses of 4 different tocopherols found in a basal diet (BD) and the effect of 2 physiologic levels of dl-alpha-tocopheryl acetate (25 and 150 mg/kg) on tissue tocopherol content were studied in the mink. The BD contained a total of 7.1 mg vitamin E/kg, with alpha-, beta-, gamma-, and delta-tocopherol in a ratio of 1:0.07:0.55:0.10, respectively. The corresponding ratios in the tissues were: liver, 1:0.04:0.12:0; plasma, 1:0:0.13:0; and adipose tissue, 1:0:0.19:0. After mink were fed diets containing vitamin E, alpha- and gamma-tocopherol were distributed in similar proportions in plasma and liver, but gamma-tocopherol was in a slightly higher proportion in adipose tissue. Addition of 25 or 150 mg/kg of alpha-tocopheryl acetate to the BD decreased the gamma-tocopherol levels in all 3 tissues; this was considered to be a dilution effect of other tocopherols in BD with added alpha-tocopheryl acetate. The beta-tocopherol content in the liver remained unchanged, irrespective of the dietary amount of alpha-tocopheryl acetate. Plasma alpha-tocopherol had a linear relationship to log dietary dose, with an apparent half-saturation of the vitamin E binding capacity at 13 mg vitamin E/kg diet. At the given dietary levels, liver and adipose continued to accumulate alpha-tocopherol. The correlation between total plasma lipids and plasma alpha-tocopherol was significant (P less than 0.001) only in the group fed the BD. The effect of maternal antibody (MAB) to hemorrhagic enteritis (HE) on the response of turkeys to infection with virulent and avirulent strains of HE virus (HEV) was examined. The influence of age at exposure and treatment with HEV antibody on development of clinical HE also was studied. MAB protected poults from clinical HE for up to 6 weeks of age. MAB also interfered with vaccination against the disease for at least 5 weeks after hatching, as indicated by absence of HEV antigen in spleens and by poor seroconversion at 6 days and at 3 weeks post-vaccination, respectively. The incidence of clinical HE in MAB-negative poults was significantly higher in poults inoculated with virus at 15 days of age or older than in poults inoculated at 1-13 days of age. Further, MAB-negative poults embryonally inoculated with virulent or avirulent strains of HEV did not develop disease; these poults developed antibody and resisted challenge with virulent virus at 6 weeks of age. Poults treated with HE antibody within 1 hour of challenge or at 1, 3, or 5 weeks before challenge with virulent virus were protected against lesions and mortality induced by HEV. These results suggest that MAB may influence susceptibility of turkeys to infection with HEV for at least 5 to 6 weeks after hatching, unlike the case with most other viral infections of poultry. The results confirm that early age resistance to clinical HE is independent of MAB and suggest that such resistance persists for up to 13 days of age. The data also suggest that turkeys lacking MAB can be immunized against HE by embryo vaccination.  相似文献

1. The effect of supplementing the diet of the parent hen with vitamin E on the vitamin E content of the yolk and of embryonic and neonatal tissues was evaluated and the effects of elevated tissue concentrations of vitamin E on peroxidation susceptibility was examined. 2. Laying hens (Ross 1 broiler-breeder strain) were maintained on diets containing either 147 (control diet) or 365 (high vitamin E diet) microg vitamin E/g feed. 3. In the day-16 embryo, the concentrations of of vitamin E in the yolk sac membrane, liver, brain and lung were respectively 5.0, 4.3, 1.7 and 5.6 times greater for those derived from the hens on the high vitamin E diet compared with those from the control group. 4. In the day-old chick, the concentrations of vitamin E in the yolk sac membrane, liver, brain and lung were respectively 14.8, 2.8, 3.0 and 5.1 times greater for those derived from hens on the high vitamin E diet compared with those from the control group. 5. Homogenates of tissues from the day-old chick were incubated in the absence and presence of Fe2+ in order to determine the extent of spontaneous and iron-stimulated peroxidation as measured by the generation of thiobarbituric acid reacting substances. For the chicks derived from hens on the control diet, the brain was markedly more susceptible to both spontaneous and iron-stimulated peroxidation than were the other tissues. Tissues from the chicks derived from the hens on the high vitamin E diet exhibited significantly reduced susceptibilities to peroxidation. In particular, the susceptibility of the brain was reduced to the same level as that of the other tissues. 6. It is concluded that the high peroxidative susceptibility of the chick's brain can be normalised by supplementation of the parent hen with vitamin E.  相似文献
