O bjectives : We described epidemiological, clinical, histopathological and ultrastructural features of ichthyosis in the golden retriever breed in a prospective study. We also investigated the mode of transmission of this disease.
M aterials and M ethods : We examined 150 golden retrievers, 73 of which were affected by ichthyosis (35 males and 38 females). We carried out detailed clinical and histopathological examinations for 40 affected dogs. Transmission electron microscopy was performed for two of them. We used pedigree analysis with the Cyrillic software to determine the mode of transmission.
R esults : Dermatological signs included a mild to moderate or severe generalised scaling with initially small to large whitish scales and progressively blackish scales. The ventral glabrous skin was hyperpigmented and rough, similar to sandpaper. Histopathological features were characterised by moderate to severe laminated or compact orthokeratotic epidermal hyperkeratosis without significant involvement of the stratum granulosum. Ultrastructural findings revealed laminated or compact keratin layers and numerous persistent corneodesmosomes within the stratum corneum. Analysis of the pedigree suggested an autosomal recessive inheritance.
C onclusion : The histopathological and ultrastructural characteristics strongly suggest that golden retriever ichthyosis is a retention ichthyosis, caused by absence of corneodesmosomal degradation, transmitted through an autosomal recessive mode.  相似文献

A scaling disorder specific to Golden Retriever dogs has been recognized by both dermatologists and pathologists, but to date has not been well characterized. At the University of Pennsylvania's Laboratory of Toxicology and Pathology, 46 cases of ichthyosis were diagnosed histologically in Golden Retriever dogs from January 2004 to January 2007. A total of 22 dogs had skin lesions documented at younger than 1 year of age; 3 dogs between 1 and 2 years of age; 13 dogs developed lesions at older than 2 years; and the time of onset was unknown for 8 dogs. A total of 25 dogs were female, and 21 were male. All dogs had strikingly similar histopathologic changes that consisted of mild to moderate laminar orthokeratotic hyperkeratosis with an absence of epidermal hyperplasia and dermal inflammation. Ultrastructural analysis using a ruthenium tetroxide fixation method was performed on punch biopsy samples from 5 dogs and compared with 2 control dogs (1 clinically and histologically normal sibling of an affected dog and 1 Cairn Terrier). All affected dogs had retained and convoluted membranes with crystalline structures in the stratum corneum. Scattered keratinocytes in the granular cell layer had prominent, clear, membrane-bound, cytoplasmic vacuoles. Pedigree analysis of 14 dogs was compatible with autosomal recessive inheritance, but incomplete dominance could not be ruled out. This unique hyperkeratotic/scaling disorder in Golden Retrievers has distinctive clinical, histologic, and ultrastructural features, which are consistent with a primary cornification defect.  相似文献

A 7-year-old, spayed female, golden retriever with a 1-year history of progressive stertor and reverse sneezing was referred for further diagnostics. Endoscopy and computed tomography of the head revealed a bilobed mass in the choanae. Cytologic and histopathologic examinations of the mass were consistent with lymphosarcoma. In the present study, blood samples were taken from clinically healthy puppies of the breeds Beagle, German Shepherd, and Golden Retriever between days 1 and 3 (n = 146), 8 and 10 (n = 137), 28 and 33 (n = 151), and 50 and 58 (n = 129) post natum. Measurements for red blood cell count, haemoglobin concentration, haematocrit, mean erythrocyte volume (MCV), mean corpuscular haemoglobin (MCH), and mean corpuscular haemoglobin concentration (MCHC) were performed by a semi-automatic blood cell counter; the normoblast number was counted visually. Between the 1st and 3rd day of life, the erythrocyte number of the puppies was 4.57 +/- 0.68 10(6)/microliter and, as such, was clearly below the reference range for adult animals. It further decreased by the 2nd measurement (8th to 10th day of life) to 3.59 +/- 0.41 10(6)/microliter, and then increased again to 4.75 +/- 0.68 10(6)/microliter (reference range: 3.73-6.25 10(6)/microliter, 2.5% to 97.5% percentile) by the final measurement (50th to 58th day of life). The measurement values of the haemoglobin concentration (13.5 +/- 2.0 g/dl) and haematocrit (41.0 +/- 6.5%) after birth were only insignificantly below or around the lower limit of the reference range for adult animals. Both parameters decreased to a more pronounced extent than did the erythrocyte count. They reached a minimum of 8.4 +/- 1.0 g/dl and 26.8 +/- 3.2%, respectively, between the 28th and 33rd day of life. Even at the end of the examination period (50th to 58th day of life), the values of these parameters (10.1 +/- 1.1 g/dl, reference range: 7.5-11.8 g/dl; 32.1 +/- 4.2%, reference range: 24.8 to 40.8%) were remarkably lower than the minimum of reference range for adult dogs. At the 1st sampling (between 1st and 3rd day of life), MCV (89.8 +/- 6.7 fl) and MCH (29.6 +/- 1.9 pg) were distinctly higher than the reference values for adult dogs. Both parameters decreased with increasing age. Thus, from the 50th-58th day of life, the results were comparable to those of adults. No considerable age dependence was found for MCHC. During the first days of life a relatively high number of normoblasts (8 +/- 7/100 Leukozyten) was found; it decreased rapidly. The study revealed significant differences between the breeds, e.g. German Shepherd dogs had lower initial values of erythrocyte count, haemoglobin concentration, and haematocrit when compared to the other breeds. Puppies of this breed also had higher normoblast numbers than the Beagle and Golden Retriever puppies at the 2nd and 3rd samplings. No clear sex differences in the studied parameters were observed. The results of this study reflect the replacement of fetal erythrocytes by postnatal erythrocytes. Moreover, they illustrate the need to use age as well as breed-specific reference ranges.  相似文献

Golden retriever and Labrador retriever muscular dystrophy are inherited progressive degenerative myopathies that are used as models of Duchenne muscular dystrophy in man. Thoracic lesions were reported to be the most consistent radiographic finding in golden retriever dogs in a study where radiographs were performed at a single-time point. Muscular dystrophy worsens clinically over time and longitudinal studies in dogs are lacking. Thus our goal was to describe the thoracic abnormalities of golden retriever and Labrador retriever dogs, to determine the timing of first expression and their evolution with time. To this purpose, we retrospectively reviewed 390 monthly radiographic studies of 38 golden retrievers and six Labrador retrievers with muscular dystrophy. The same thoracic lesions were found in both golden and Labrador retrievers. They included, in decreasing frequency, flattened and/or scalloped diaphragmatic shape (43/44), pulmonary hyperinflation (34/44), hiatal hernia (34/44), cranial pectus excavatum (23/44), bronchopneumonia (22/44), and megaesophagus (14/44). The last three lesions were not reported in a previous radiographic study in golden retriever dogs. In all but two dogs the thoracic changes were detected between 4 and 10 months and were persistent or worsened over time. Clinically, muscular dystrophy should be included in the differential diagnosis of dogs with a combination of these thoracic radiographic findings.  相似文献

Wellness and pre-anesthetic screening of blood and urine of geriatric companion animals are routinely recommended. In addition, there are occasional references to the use of imaging in clinically normal geriatric patients. However, the utility of wellness testing is not known, and there is limited information regarding the value of pre-anesthetic testing. Wellness testing, including complete blood cell count, biochemical profile, urinalysis, and abdominal ultrasound, was performed on 53 clinically normal, mature golden retriever dogs. Laboratory analysis revealed abnormalities in 54.7% (29/53) of the dogs. Abdominal ultrasound screening demonstrated abnormalities in 64.2% (34/53) of the dogs. As only a small number of dogs had follow-up diagnostic testing available, the significance of these abnormalities is unknown. Further study involving a larger cohort of animals and analysis of follow-up data is necessary to determine the utility of laboratory and imaging studies in clinically normal geriatric patients.  相似文献

A severe neurological disorder was observed during a very dry season, in sheep pasturing on land with mediterranean vegetation. During the crisis, lasting 5-20", ataxia and convulsions were the main clinical signs. Brain congestion, spongy appearance of the liver, interstitial nephritis and hyperpigmentation of visceral lymph nodes were the most important macroscopic lesions. Histopathological, histochemical and ultrastructural findings confirmed liver and kidney lesions and showed considerable neurovisceral storage of lipofuscin. Although further toxicological and experimental studies are needed to elucidate the origin of the disease, the Authors attribute the outbreak of the neurological syndrome to the ingestion of a neurotoxin containing plant.  相似文献

A 12monthoid neutered male golden retriever was presented with a history of lethargy and exercise intolerance. Clinical examination, electrocardiography, radiography and echocardiography supported a diagnosis of fixed subvalvular aortic stenosis with a Doppler pressure gradient of 77.5 mmHg. Surgical inspection also revealed gross structural abnormalities of the mitral valve consistent with mitral dyspiasia. intervention consisted of resection of the dysplastic mitral valve and the subvalvular aortic stenosis. The mitral valve was replaced with a bioprosthetic valve. Total cardiopulmonary bypass time was 65 minutes and aortic crossclamp time was 55 minutes. A full recovery was made and 11 months postoperatively the aortic transvalvular gradient was 30 mmHg. At the time of writing, 12 months after surgery, the dog was clinically normal and requires no medication.  相似文献

The objective of the study was to determine whether the plasma concentrations of atrial and brain natriuretic peptides (ANP and BNP, respectively) could be reliable markers of cardiac alterations during occult cardiomyopathy in Golden Retriever Muscular Dystrophy (GRMD). Fifty Golden Retrievers without any clinical or radiographic sign of heart disease were included in this study (21 GRMD dogs and 29 controls). Controls and GRMD dogs were divided into 2 subgroups according to age (< and > or =12 months old, respectively). All dogs underwent echocardiography and determination of BNP and ANP plasma concentrations by radioimmunoassay. No ventricular dilatation or dysfunction was observed in either control or GRMD dogs. ANP plasma concentration did not differ significantly between controls and GRMD dogs (mean +/- SD = 72 +/- 49 versus 58 +/- 23 pg/mL, respectively, P = .21). This finding was confirmed in both subgroups of dogs (ie, those < and > or =12 months old). In contrast, BNP plasma concentrations were significantly higher in GRMD dogs than in controls (mean +/- SD = 117 +/- 92 versus 46 +/- 22 pg/mL, respectively, P < .05). In dogs > or =12 months old, sensitivity and specificity of BNP for identifying GRMD with a cutoff of 65 pg/mL were 78 and 86%, respectively. For the same cutoff value, sensitivity dropped to 42%, whereas specificity reached 100% in dogs <12 months old. In conclusion, BNP may be a useful biochemical marker of asymptomatic cardiomyopathy. However, this peptide does not allow very early detection because its optimal discriminatory power was observed in adult dogs (ie, > or =12 months of age).  相似文献

A 20-week-old male golden retriever, which was not lame and showed no clinical signs of a fragmented medial coronoid process (FCP), was euthanased for another study and perfused intravenously with formaldehyde. Gross dissection revealed no abnormalities within the right elbow joint. The medial coronoid process was excised, embedded in methylmethacrylate, scanned in a microcomputed tomography (microCT) scanner and sectioned for histology. The microCT scans revealed a dense trabecular bone structure, much denser than in other dogs of similar age, which was considered to be responsible for the sclerosis visible at the base of the coronoid process in radiographs. Three-dimensional reconstructions indicated that there was a small step within the subchondral bone, extending from the apex towards the radial notch. Histology revealed a necrotic lesion between locally thickened articular cartilage and the subchondral bone, characteristic of osteochondrosis.  相似文献

Background: Ehlers-Danlos syndrome comprises a group of rare inherited connective tissue diseases characterized by skin hyperextensibility, joint laxity, skin and vessel fragility, and poor wound healing.
Objective: The purpose of this report was to describe the clinical, histologic, and ultrastructural findings in 2 dogs with collagenopathies consistent with Ehlers-Danlos syndrome.
Methods: Two dogs were examined clinically; skin extensibility index was calculated. Skin biopsies obtained from the dorsum were examined by light and electron microscopy.
Results: Both dogs had clinical signs of skin hyperextensibility and fragility, lower skin elasticity, vessel fragility, and poor wound healing. One dog had a hip dislocation, and the other had bilateral medial patellar luxation (grade II), subcutaneous hematomas produced by minimal trauma, and generalized periodontitis. Histologic and ultrastructural examination confirmed abnormalities in the structure and arrangement of collagen fibrils. Fibroblasts were characterized by variable dilatation of the rough endoplasmic reticulum, and anomalous elastic fibers (elaunin fibers) were present in the dermis.
Conclusion: Although the primary defects underlying collagenopathies in animals are still unknown, analysis of the ultrastructural changes in collagen fibrils and clinical findings could facilitate better characterization of these disorders in dogs.  相似文献

A 14-year-old neutered male Persian cat was evaluated because of an acute exacerbation of a chronic cough of 2-3 years of duration. Physical examination was normal except for the auscultation of accentuated breath sounds and wheezes cranially on both sides of the chest. Complete blood count, biochemical parameters and urinalysis were normal. Thoracic radiographs showed a generalised nodular pattern with multiple mineral opacities. Oral prednisone and doxycycline were prescribed. Two weeks later, the frequency of the cough was significantly reduced. Terbutaline was recommended for relief of acute exacerbations. Three years later the cat was evaluated again due to a non-related disease that led to the euthanasia of the cat. Concerning its respiratory disease, the cat had experienced nearly asymptomatic periods of 3-6 weeks of duration punctuated by acute exacerbation periods of 7-10 days, during which terbutaline was useful to relieve the cough. Thoracic radiographs showed a mild increase in the size and extent of the pulmonary mineralisation. Histopathologically, mild bronchitis and bronchiectasis were evident, accompanied by calcified bronchial plugs and marked hyperplasia and hypertrophy of the seromucinous glands. Based on clinical and pathoanatomical findings, a final diagnosis of miliary broncholithiasis and bronchiectasis was made. Broncholithiasis should be considered in differential diagnosis of pulmonary mineralisation in cats. When no concomitant diseases are present, this rare disease appears to have a slowly progressive evolution that does not appear to carry a bad prognosis and may be satisfactorily managed with combinations of bronchodilators and corticosteroids.  相似文献

Objective  To describe the microscopic features and lineage of proliferating/infiltrating pigmented cells in ocular melanosis of Cairn Terriers.
Animals studied  Forty-nine globes removed from 45 Cairn Terriers with ocular melanosis and three globes from control dogs were available for microscopic examination.
Procedures  All globes were examined histologically, eight affected and three control globes were also examined by immunohistochemistry, and three affected and three control globes by transmission electron microscopy.
Results  Large round pigment-laden cells infiltrated the anterior uvea, obscured the drainage angle and were present within the sclera and episclera of affected globes. Similar pigmented cells were present in lower numbers in the posterior segment of the globe, the optic nerve meninges and periphery of the optic nerve. Changes due to chronic glaucoma were present in many globes and some had evidence of uveitis. Many of the pigmented cells were immunoreactive to HMB45 and some were MITF and vimentin positive. One globe, which was inflamed when removed, had many pigmented cells that were CD18 immunoreactive. The other eyes had lower numbers of CD18 positive cells. The pigmented cells were not immunoreactive to smooth muscle actin, S-100, MART/Melan A, chromogranin A/B, PGP 9.5, synaptophysin, MNF116, AE1/AE3, and CD45. Ultrastructurally many of the pigmented cells had features typical of melanocytes while a smaller number appeared to be melanophages.
Conclusions  Ocular melanosis in Cairn Terriers is characterized by an infiltration of pigment-laden cells predominantly, but not exclusively, within the anterior uvea and anterior sclera. Most of these cells appear to be melanocytes although a variable proportion are pigment-laden melanophages.  相似文献

The purpose of the present study was to establish normal electroretinogram (ERG) parameters using 56 normal eyes of four dog breeds common in Thailand: poodle, Labrador retriever, Thai ridgeback, and Thai Bangkaew. Standard ERG findings were bilaterally recorded using a handheld multi-species ERG unit with an ERG-jet lens electrode for 28 dogs under preanesthesia with diazepam, anesthesia with propofol, and anesthesia maintenance with isoflurane. There were significant differences in the mean values of ERG amplitudes and implicit times among the four dog breeds (p < 0.05) except for the b-wave implicit time of the photopic 30 Hz flicker response with 3 cd.s/m2 (p = 0.610). Out of the four breeds, Thai Bangkaew had the longest implicit time (p < 0.001) of scotopic low intensity responses, b-wave of scotopic standard intensity responses (3 cd.s/m2), a-wave of the higher intensity response (10 cd.s/m2), and a-wave of the photopic single flash response (3 cd.s/m2). For the b/a ratio, only the ratio of the Cone response was significantly different among the different breeds. In this summary, normal ERG parameters for four dog breeds were reported. Data from the investigation supported the hypothesis that determination of breed-specific limits of normality for ERG responses is necessary for individual clinics and laboratories.  相似文献
