Background

The physiological levels of endocrine and metabolic parameters in Slovene autochthonous breeds of sheep are not yet well known, nor are the mechanisms of their adaptability and responses to climate and environmental factors. Therefore, the aim of this study was to evaluate fluctuations of cortisol, insulin and non-esterified fatty acids (NEFA) in growing ewes over an one-year period. Blood samples were collected monthly from 10 yearling Jezersko-Solchava, 10 Bovec and 10 Istrian ewes. Serum cortisol, insulin and NEFA were measured with commercial kits.

Results

Mean monthly cortisol values fluctuated with low levels in summer and high levels in autumn. Significant peaked cortisol values of 25.69 ± 6.89, 14.67 ± 2.43 and 21.11 ± 7.25 μg/L in Jezersko-Solchava, Bovec and Istrian breed, respectively, were found in September (Bovec breed) and October (Jezersko-Solchava and Istrian breed). Mean monthly insulin values increased during the observation period. The highest levels of 14.60 ± 3.15, 16.03 ± 5.35 and 12.56 ± 2.52 μIU/mL in Jezersko-Solchava, Bovec and Istrian breed, respectively, were observed in the last sample collection in May. NEFA concentrations were found to be low except in some autumn and spring months. The peak values were observed in March for Jezersko-Solchava and Istrian breed (0.60 ± 0.05 and 0.66 ± 0.10 mmol/L), and in April for Bovec breed (0.71 ± 0.11 mmol/L).

Conclusions

Monthly fluctuations of cortisol, insulin and NEFA were measured in all observed sheep breeds, but between-breed differences in monthly values of examined parameters were insignificant. Significantly increased serum cortisol levels were found in autumn for all breeds and were probably associated with the onset of puberty and low environmental temperature. A gradual increase of insulin level in the examined ewes was in parallel with their growth. Significantly higher NEFA values in spring suggest qualitatively insufficient feed supply during that period.  相似文献

The impact of pregnancy on ketone body disposal during a hyperketonaemic clamp was examined by tracer isotope dilution techniques in seven 12 h fasted sheep in three reproductive states, in the dry non-gestating period, late in gestation and during early lactation. After a sampling period of 60 min under basal conditions a DL-BHB racemate solution was continuously infused intravenously for 3 h at rates of 14.3-24.3 micromol/(kg min) to elevate the D-BHB concentration in blood plasma to values between 5 and 7 mmol/l. Two separate experiments were carried out with the same sheep in each of the three reproductive states. During pregnancy three ewes were pregnant with a single lamb and four ewes carried twins. Maximal D-BHB turnover rates fell significantly in late gestation by 26% relative to early lactation and by 22% when compared with the dry non-pregnant state. Reduction of maximal D-BHB disposal rate during late gestation was accompanied by a significant 297% (p<0.005) and a non-significant 49% increase in the basal BHB concentration in blood, a non-significant 10% and 4% decrease in the rate constant of D-BHB turnover and a non-significant 24% and 13% rise in the incremented increase of D-BHB concentration per unit D-BHB infusion, relative to the dry and the lactating period, respectively. Induction of hyperketonaemia significantly lowered NEFA and glycerol concentrations in blood by 67% and 57%, respectively, compared to the pre-infusional concentrations. The magnitude of this effect was the same in all reproductive states and was explained as a direct antilipolytic action of D-BHB on adipose tissue. It is concluded that the reduced ability of the late gestating ewe to utilize D-BHB promotes hyperketonaemia. Since hyperketonaemia exerts several adverse effects, e.g. on energy balance and glucose metabolism it appears that the impairment of ketone bodies disposal in late pregnancy facilitates development of pregnancy toxaemia, especially in ewes carrying twins.  相似文献

This study examined mechanisms whereby the metabolic environment interacts with basic reproductive function. Ewes lambing during the breeding season were fed to maintain (MAINT, n = 10) or gain (GAIN, n = 11) body weight during the last 4 mo of gestation. From d 7 to 22 postpartum, ewes were infused iv with saline (n = 10) or glucose at a rate calculated to increase normal glucose entry rate by 75% (n = 11). Blood samples were collected daily to determine plasma concentrations of nutritive metabolites and insulin and at frequent intervals on d 14 and 21 to determine serum gonadotropin concentrations. Hypothalami and pituitaries were collected on d 22 to determine hormone content and receptor concentrations. Plasma concentrations of nutritive metabolites and insulin indicated that MAINT ewes mobilized more (P less than .01) body fat and protein reserves during gestation and early lactation than ewes in the GAIN group. Glucose infusion elevated plasma concentrations of glucose (P less than .05) and insulin (P less than .07) and reduced (P less than .05) fat and protein mobilization, even though it depressed feed intake (P less than .001), compared with saline infusion. Hypothalamic gonadotropin-releasing hormone (GnRH), pituitary luteinizing hormone (LH) and follicle-stimulating hormone (FSH) content and pituitary GnRH receptor concentration were similar between treatments.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Pregnancy toxaemia is a metabolic disorder with a high mortality rate and occurs in twin-bearing ewes in late gestation. Maternal hypoglycaemia is a characteristic symptom of the disease and has been attributed to an increase in glucose uptake by the twin-bearing uterus. The possibility that a reduced maternal glucose production rate might cause hypoglycaemia, has received little attention in the past. It was the aim of this study to investigate this explanation as a possible alternative. Six ewes were sequentially subjected to two types of hypoglycaemic stress, firstly by fasting for 14 h and secondly through induction of moderate hyperketonaemia. Glucose kinetics were assessed in each animal during the dry non-gestational period, during late gestation, and during early lactation. Application of these stress factors was associated with variation of plasma glucose concentration from 4.9 to 0.87 mmol L(-1). The plasma glucose concentration was always significantly related to the glucose production rate. The greatest stress-induced reductions in glucose concentration and glucose production rate were seen during late gestation in twin-bearing ewes. The decline in the glucose production rate after an overnight fast and during induced hyperketonaemia was greater in twin-bearing ewes than in single-bearing ewes (59% and 43%, respectively, p<0.05). The stress conditions resulted in the lowest levels of glucose concentration and glucose turnover rates in the stressed, hyperketonaemic, late gestation twin-bearing ewes. This illustrates that the glucose homoeostatic system of ewes bearing twins is significantly more susceptible to hypoglycaemic stress than that of ewes bearing single lambs. These findings also show that the primary cause of hypoglycaemia in late gestation twin-pregnant ewes is an increased susceptibility to a stress related reduction in glucose production rate. This metabolic condition leaves the twin-pregnant ewe predisposed for the development of pregnancy toxaemia.  相似文献

Increased plasma levels of non-esterified fatty acids (NEFA) may lead to several physiological changes e.g. increased insulin secretion with a concomitant reduction of blood glucose, decreased glucose utilization in heart and skeletal musculature and increased blood acetone levels. High NEFA levels also cause fat infiltration in various organs especially the liver. Recently Akgün & Rudman (1969) showed that ACTH induced NEFA mobilization in rabbits was followed by hypocalcemia. Serum calcium decreased about 30 %, while the calcium content in adipose tissue increased up to 1000 %. This finding could also be verified in vitro. When adipose tissue was incubated in serum containing lipolytic hormones, lipolysis was stimulated and there was a shift of calcium from serum to tissue. A negative correlation between serum calcium and NEFA in hypocalcemic cows was reported earlier (Luthman & Jonson 1969). The purpose of the present investigation was to study the effect of increased NEFA levels on serum calcium in sheep. The animals used were ewes in late pregnancy. Lipolysis was stimulated by norepinephrine (Norexadrin, Astra). The animals were given a continuous intravenous infusion during 8 hrs. at a rate of 1 μg/kg/min. The methods of analysis were the same as described before (Luthman & Jonson).  相似文献

Insulin resistance during late gestation may act as a predisposing factor of ovine pregnancy toxaemia (OPT). To evaluate the insulin action on energy metabolism in ewes with different susceptibilities to OPT, intravenous glucose tolerance tests (1 mmol glucose/kg body weight) were performed in 5.6 ± 0.7 year old, slightly underfed German Blackheaded Mutton ewes [high‐risk (HR) ewes] and 2.5 year old, overnourished Finnish Landrace ewes [low‐risk (LR) ewes] during mid and late pregnancy, during early lactation and during the dry period. Plasma samples were analysed for glucose, insulin, non‐esterified fatty acids (NEFA) and β‐hydroxybutyrate (β‐HB). The glucose elimination rate and the glucose‐stimulated first‐phase insulin secretion were significantly (p < 0.05) lower in the HR, in relation to the LR group combining the data of all gestational stages. The basal rate of lipolysis was significantly increased in the HR ewes during late pregnancy, but the NEFA clearance after the glucose load was similar in both groups during all reproductive stages. Plasma β‐HB concentrations decreased only in the LR ewes after the glucose load during late pregnancy. Results indicate an insulin resistance in the HR ewes regarding the glucose utilization and the ketone body formation during late pregnancy. The insulin resistance in the HR ewes may represent one predisposing factor responsible for the susceptibility to OPT. Further scientific work is necessary to elucidate whether this insulin resistance was due to breed, age or nutritional state.  相似文献

The objectives of this study were to examine the influence of body condition of cows on metabolic and antioxidative status, as well as to investigate the relationship between metabolic indicators of lipid mobilization and oxidative stress during transition period. The study was conducted on 24 Holstein‐Friesian dairy cows divided into 2 groups according to their body condition score (BCS) as optimal (n = 12; BCS from 3.25 to 3.75) or adipose (n = 12; BCS ≥4). Metabolic status (glucose, triglycerides, total cholesterol, HDL cholesterol, NEFA and BHB), paraoxonase‐1 (PON1) and apolipoprotein A‐I (ApoA‐I) were analysed in sera taken on days ?30, ?10, ?2, 0, 5, 12, 19, 26 and 60 relative to parturition. Adipose cows had significantly higher glucose concentration at parturition being significantly decreased after parturition on days 12 and 19. Total cholesterol and HDL‐C concentrations were the lowest at parturition and significantly higher on days 26 and 60 after parturition in both groups of cows. Both investigated groups had significantly higher NEFA concentration from parturition until day 19 after parturition, indicating energy deficit and an increased lipid mobilization after calving. There were no significant differences in BHB concentration during transition period in both groups. No significant differences were found in PON1 activity and ApoA‐I concentration during transition period in both groups of cows. However, in adipose cows, although not significantly different, PON1 was decreased from calving until day 19 after parturition indicating a disturbance in antioxidative status in adipose cows. PON1 significantly positively correlated with total cholesterol and HDL‐C concentrations and negatively with NEFA indicating a strong relationship of PON1 with lipid metabolism. Significant positive correlation between NEFA and BHB in both groups of cows points out on energy deficit during transition period that cows tend to overcome by lipid mobilization providing alternative source of energy needed for parturition and lactation.  相似文献

Previous studies have shown that shearing pregnant ewes at mid- or late-pregnancy is associated with an increase in lamb birth weight. The present study was designed to investigate metabolic responses that may underlie this response. Single- and twin-bearing ewes were either unshorn or shorn at mid-pregnancy (Day 69 of pregnancy; P69), and insulin, glucose and epinephrine challenges were conducted on P109–111 and P132–134. Shearing increased the birth weight of twin lambs by over 1 kg (P < 0.001) without having any effect on singleton birth weight. This response was associated with a 10–20% reduction in the insulin response to a glucose challenge (P < 0.05) without a change in glucose clearance following either glucose or insulin challenges. The lipolytic response to epinephrine challenge increased as pregnancy progressed, but was not associated with the increased birth weight of twin lambs born to shorn ewes. By late pregnancy, a 25% reduction in maternal IGF-I concentration and a two- to threefold increase in maternal IGFBP-1 concentration (P < 0.05) associated with shearing were observed. The increase in lamb birth weight associated with mid-pregnancy shearing may have been associated with an increase in the non-insulin dependent uptake of glucose by the placental-fetal unit.  相似文献

Primiparous ewes and multiparous ewes show physiological differences during pregnancy, which can have an impact on the development of their offspring. The objective of this study was to compare the changes in the metabolic profile and in the size of some foetal bones throughout gestation between primiparous and multiparous ewes. Twelve primiparous (PM) ewes and 14 multiparous (MT) ewes were used. According to the dates of lambing, two groups of ewes were formed: Group 1 (G1, n = 6 PM and n = 7 MT) and Group 2 (G2, n = 6 PM and n = 7 MT). The body weight, body condition score, metabolic and foetal morphometric parameters were determined from before conception until the end of gestation. After lambing, the body weight and survival rate during the first 72 hr of life of lambs, as well as the maternal behaviour score were recorded. The PM ewes were lighter (p < .01) and had a greater mobilization of body reserves during gestation, mainly evidenced by a greater serum concentration of NEFAs and lower serum concentration of total proteins (p < .05) compared with the MT ewes. The parity did not affect the foetal morphometric variables. The lambs of MT ewes were heavier at parturition (p = .002) and tended to have a greater survival rate than those lambs of PM ewes (p = .09). In conclusion, PM ewes and MT ewes differ in their metabolic profile throughout the gestation. However, in the present study, we did not find parity differences in the dimensions of foetal bones during growth in gestation.  相似文献

The late pregnancy is a critical period of fetal growth and development. As approximately 80% of fetal body weight is obtained during late gestation, so that the amount of energy needed for the ewes at this period is much greater than the other periods. Now ewes with multiple fetuses are mutton sheep breeding cultivation objectives, however, under the grazing or confined feeding conditions where nutrient requirement can't satisfy the nutrient requirement of sheep, especially for pregnant ewes with twins or more lambs. The body glycogen and protein as well as body fat will be mobilized for energy needed for the fetal growth and reproduction, which would lead to bad influence on ewes (metabolic disorders, liver function damage, pregnancy toxemia, embryonic development and health) and also on lamb (weak lamb, dead lamb and health). Therefore, investigating the effects of dietary energy levels during late gestation on the ewe's health and growth performance of lambs are essential for the sustainability of livestock production systems. This review summarized the effect of different energy levels on the ewes' health, reproductive performance of the ewes, lactation performance, embryo development and the growth of lambs, then expounded the roles of energy in later gestation. When the dietary energy of ewes in late gestation are restricted, the weight, breast development, lactation capacity, pregnancy rate will all be decreased, and the growth rate and organ development of embryos and lamb will also be restricted.

German black headed mutton (GBM) ewes are recognized as being highly susceptible to ovine pregnancy toxemia (OPT). The present trial was performed to evaluate whether a breed-dependent gestational diabetes mellitus-like insulin resistance during late pregnancy might be responsible for the high incidence of OPT in the GBM breed. Modified frequently sampled intravenous glucose tolerance tests (300 mg glucose and 0.03 IU insulin per kg of BW) were performed during mid and late pregnancy, the periparturient, and the dry period in polytocous 3.5-yr-old GBM and Finnish Landrace (FL) ewes fed according to their requirements. The corresponding blood samples were analyzed for plasma concentrations of glucose, insulin, nonesterified fatty acids (NEFAs) and β-hydroxybutyrate (β-HB). In addition, the baseline plasma cortisol concentrations were determined during late pregnancy. The BW gain during pregnancy and the rearing success did not differ between the GBM and FL ewes. In both breeds, late pregnancy was associated with decreased basal plasma glucose concentrations and enhanced glucose disposal, as well as elevated baseline β-HB values. Only in the GBM ewes did the plasma NEFA concentrations increase significantly during advancing pregnancy. Moreover, significantly higher baseline plasma NEFA concentrations as well as lower (P < 0.05) basal plasma glucose values were recorded during late pregnancy in the GBM than in the FL ewes. The first-phase insulin secretion, the peripheral insulin sensitivity, and the baseline plasma cortisol values did not differ between both breeds during late pregnancy. It is concluded that increased lipolysis during late pregnancy is a characteristic of the GBM breed. Moreover, elevated plasma NEFA concentrations may contribute to impaired pancreatic insulin response and peripheral insulin resistance in GBM ewes and thus promote OPT.  相似文献

The transition period of dairy cattle is characterized by dramatic changes in metabolism and host defense mechanisms that are associated with increased disease. Intense lipid mobilization from tissue stores is an important metabolic adaptation during the transition period that results in significant release of non-esterified fatty acids (NEFA) into the blood stream. Whereas these fatty acids are important sources of energy during times of increased metabolic demands, elevated concentrations of NEFA are known to disrupt several immune and inflammatory functions. This review will discuss the implications of lipid mobilization on inflammatory responses with special emphasis on leukocytes and endothelial cell functions during the transition period of dairy cows.  相似文献

Neonatal lamb mortality is a major factor affecting profitability in the sheep industry, and lamb thermogenesis is a key element in neonatal lamb survival. Increased lamb vigor has been reported when ewes were supplemented during late gestation with algae-derived docosahexaenoic acid (DHA); however, the effects of DHA on lamb thermogenesis and immunocompetence have not been investigated. Eighty twin-bearing Targhee ewes (ages 2 to 5 yr; 68.5 ± 3 kg) were assigned randomly to 1 of 2 supplement treatments to determine the effects of feeding DHA to ewes during late gestation and early lactation on lamb thermogenesis, serum metabolites and hormones, and lamb growth. Supplement treatments were 12 g·ewe(-1)·d(-1) of algae-derived DHA (DHA Gold Advanced Bionutrition Corp., Columbia, MD; algae-derived DHA); and no algae-derived DHA (control). Supplements were individually fed daily during the last 30 d (±7 d) of gestation and pen fed (6 pens/treatment with 6 or 7 ewes/pen) during the first 38 d (±7 d) of lactation. One hour after lambing and before nursing, twin-born lambs were weighed, blood sampled via jugular puncture, and placed in a dry cold chamber for 30 min (0°C), and rectal temperatures were recorded every minute for 30 min. Lambs were removed from the cold chamber, blood sampled, warmed for 15 min, and returned to their dam. Ewes were blood sampled, and colostrum samples were collected 1 h postpartum. Ewe and lamb sera were assayed for glucose, NEFA, cortisol, and leptin. Lamb rectal temperature, glucose, NEFA, cortisol, leptin, and birth weights did not differ between treatments. The BW at 38 d was greater (P = 0.03) for lambs born to control ewes than for lambs born to algae-derived DHA-supplemented ewes; however, the colostrum of algae-derived DHA-supplemented ewes had a greater specific gravity (P = 0.05) than for control ewes. Overall, despite a potentially positive effect on ewe colostral IgG concentrations, supplementation of algae-derived DHA during late gestation and early lactation had a negative effect on lamb BW and did not affect indices of lamb thermogenesis.  相似文献

The aim of this work was to compare energy and lipid metabolism during the peripartum period between Modenese (MO) and Italian Friesian (IF) cows. The study was carried out on 33 pluriparous pregnant cows, 19 IF and 14 MO, reared together in the same herd and kept under equal conditions of management and nutrition. Blood was sampled from jugular vein starting 4 week before expected calving date until 4 week post‐calving, once weekly. Plasma was analysed for glucose, NEFA, BHBA, triglycerides and cholesterol concentrations. Body condition score (BCS) was assessed weekly after blood sampling. Data from antepartum (a.p.) and post‐partum (p.p.) periods were separately analysed as repeated measures by a linear mixed models with the effect of breed, time and their interaction as main factors and random cow within breed. The energy status differed between the two breeds during the peripartum period. We observed higher BCS a.p. and p.p. and lower BCS variations p.p. in MO compared to IF group. Modenese cows showed lower glucose and cholesterol concentrations (p < 0.001), but higher NEFA values, NEFA to cholesterol and NEFA to albumin ratios (p < 0.001) during a.p.; on the contrary, IF cows had higher (p < 0.05) cholesterol, NEFA, BHBA levels and NEFA to albumin ratio than MO ones during p.p. The differences observed between the two breeds suggest how MO cows are subjected to lipid mobilization during late gestation; on the contrary, IF cows are predisposed to mobilize their lipid reserves at the beginning of lactation to support high production. The results indicate a diverse ability to cope with metabolic stress and suggest the hypothesis that the differences in concentrations of plasma parameters and their variation amplitude around the calving period might depend on the different genetic merit for milk production between the two breeds.  相似文献

In 11 Awassi ewes of a flock of 30 animals continuous investigations were carried out on the changes of somatogenic (growth hormone-GH, prolactin-PRL, insulin-like growth factor I-IGF-I) and steroid hormones (progesterone--PG, estradiol-17 beta-E2) in blood plasma during a gestation period. Eight animals were pregnant (3 with twins), 3 non-pregnant ewes served as control animals. Pregnancy is associated with an increase in the concentrations of the steroid hormones in blood plasma, they decrease markedly already prepartal (PG) or directly at term (E2), reflecting the placental origin of both hormones. Ewes with twins have higher concentrations of both steroid hormones in their blood plasma than animals with a single fetus. During pregnancy the concentrations of IGF-I in blood plasma increase continuously and normalize rapidly after birth. The relationship between the duration of the pregnancy and the concentration of IGF-I can be described by a quadratic regression, further positive correlations exist between the concentrations of IGF-I and PG or E2 in blood plasma, but a negative correlation between IGF-I and PRL. The concentration of PRL diminishes during the gestation, which reflects increasing seasonal influences (decreasing day length and environmental temperature). The concentration of GH shows a steady level throughout the gestation period. Both hormones increase markedly at term, which represents an attendant circumstance of the birth. From all hormones investigated in blood plasma of the ewes only the concentration of IGF-I exhibits a strong positive correlation with the birth weight of their lambs. This can be taken as a first hint to the ability of IGF-I to promote directly the fetal growth.  相似文献

A high prevalence of metabolic disease was evident in a large flock of sheep early in the lambing season. Pregnancy toxemia and secondary hypocalcemia were diagnosed on the basis of history, physical examination findings, and results of serum biochemical analyses. To decrease costs to the owner, pooled serum samples were used to determine the metabolic health status of the flock. Ewes close to the time of lambing were found to be in severe negative energy balance, which resulted in excessive fat mobilization as well as clinical signs associated with pregnancy toxemia and hepatic lipidosis. By nutrient analysis, it was determined that the ewes had an inadequate amount of fermentable carbohydrates, which provide glucose to support the additional nutrition required during late gestation. The diet provided to sheep during late gestation should contain sufficient glucose precursors to maintain adequate glucose availability to maternal and fetal tissues. To minimize the physiologic decline in dry-matter intake during late gestation (which results in accentuation of negative energy balance), high-quality feed ingredients should be fed to ewes during this period.  相似文献

AIM: To determine whether short-term progesterone supplementation post mating or shearing of ewes in early pregnancy affected either the proportion of ewes that lambed or that had multiple lambs, or the birthweight of lambs.

METHODS: Romney ewes (n=457) were synchronised in oestrus using controlled internal drug-releasing (CIDR) devices containing progesterone, and mated to Romney rams over a 5-day period. The mid-point of mating (Day 0) occurred 2 days after the withdrawal of CIDR devices. Ewes mated (n=397) were randomly allocated to one of four treatment groups: shearing at Day 5, shearing at Day 30, no shearing, and no shearing plus progesterone supplementation using a CIDR device inserted on Day 3 for 6 days. During the period from Day 5 to Day 27, six harnessed Suffolk rams were placed with the ewes and matings recorded. At Day 48, all ewes that did not return to the Suffolk rams were scanned for pregnancy using ultrasound. At Day 140, single- and multiple-bearing ewes were set-stocked at 15.1 and 12.2 ewes/ha, respectively, and equivalent numbers of ewes from each treatment group were placed in each paddock. Blood samples from 10 unshorn and 10 progesterone-supplemented ewes were collected on Days 3, 6 and 9, and analysed for plasma progesterone concentrations. Lambs were identified to their dam and weighed within 12 h of birth, and again at 27 and 93 days after the mid-point of lambing. The ewes were weighed at regular intervals throughout the trial.

RESULTS: Plasma progesterone concentrations of supplemented ewes were higher than those of unsupplemented ewes (3.28 vs 1.75 ng/ml) on Day 6 (p=0.02) but not on Day 9 (4.58 vs 4.63 ng/ml). Treatment of ewes had no effect on either the proportion of ewes which lambed to the synchronised mating period or that had multiple lambs. Lambs born to ewes shorn at Day 30 tended (p=0.09) to be heavier at birth (by 0.28 kg) than those born to unshorn ewes but this effect was not evident when data were corrected for length of gestation. Neither shearing at Day 5 nor progesterone supplementation had any effect on the birthweight of lambs, and the treatment of ewes had no effect on the survival rate of lambs to weaning.

CONCLUSIONS: Progesterone supplementation for 6 days beginning 3 days post mating did not increase either the proportion of ewes that lambed or that had multiple lambs, or the birthweight of lambs. Shearing 5 days after mating had no significant effect on the reproductive performance of ewes and need not be avoided, but is unlikely to result in an increase in lamb birthweight. Shearing ewes at Day 30 may result in an increase in the birthweight of lambs but, ideally, ewes should be further advanced in pregnancy before shearing is undertaken.  相似文献

The transition of pregnancy to lactation, with the concomitant negative energy balance during early lactation, requires substantial adaptive performance of the cow. Apart from clinical disease problems, the identification of cows with suboptimal adaptation is relevant to be able to adequately treat these animals or modify the ration. Effective approaches are necessary to provide maximal information at the earliest time possible. We therefore aimed to identify a measurement that, when applied at a defined point in time relative to calving, was relevant as much as possible to other information on metabolic and health status during early lactation. Blood samples were collected weekly from 4 wk antepartum to 12 wk postpartum from 38 high-yielding Holstein-Friesian cows. Nonesterified fatty acids, beta-hydroxybutyrate, IGF-I, and leptin were measured in serum, and BCS was recorded. Health status was characterized using the concentrations of haptoglobin, the number of leukocytes and neutrophils, as well as the activity of glutamate dehydrogenase (GLDH) in blood to evaluate liver status. Using the factors related to fat mobilization, the animals were classified according to their values recorded at one defined point in time or time interval as being above or below certain thresholds. For each criterion, the groups classified were compared with regard to the time-course yielded from all recordings. From 7 criteria of classification, the most closely related to the variables of fat mobilization was obtained when using NEFA and IGF-I (thresholds of 0.5 mM and 39 ng/mL in wk 1 postpartum, respectively). Both items were then combined into to the criterion NEFA + IGF-I. Applying these criteria, the relations to indices of health and liver status were detectable on the basis of NEFA- and NEFA + IGF-I-classes, which yielded differences in both GLDH and leukocyte numbers. Animals with NEFA > 0.5 mM showed increased GLDH activity but decreased leukocyte numbers. The time and effort required for measuring the IGF-I-concentration in addition to NEFA is not justified for evaluating the metabolic status. Nonesterified fatty acid values > or = 0.5 mM during the first week of lactation were considered as the most suitable criterion for identifying limited adaptive performance.  相似文献
