Stearoyl-CoA desaturase 1 (SCD1) catalyses the synthesis of conjugated linoleic acid (CLA) and mono-unsaturated fatty acids (MUFA) in the mammary gland of ruminant animals. Considerable variations in CLA and MUFA have been reported among animals of the same contemporary group. We hypothesized that single nucleotide polymorphisms (SNPs) in the 5' and 3' untranslated regions (UTRs) of the SCD1 gene would influence the production of SCD1 enzyme and consequently its activity in the mammary gland, which may account for some of the observed within breed variations in CLA and MUFA. The 5' and 3'UTRs of the SCD1 gene of 46 Holsteins and 35 Jerseys were analysed for SNPs by sequencing. No SNPs were identified in the 5'UTR, while 14 SNPs were identified in the 3'UTR region. Further analysis revealed three haplotype structures or regulatory variants in Holsteins: named H1, H2 and H3 and only H1 and H3 in Jerseys. An IRES motif was found in the H1 variant. A subsequent association study involving the milk fatty acid profiles of 862 Holstein cows found the H1 regulatory variant to be associated with higher C10 and C12 desaturase indices and consequently with higher contents of C10:1 and C12:1 relative to the H3 variant. The effects of the H2 variant were intermediate to those of H1 and H3. SNPs in the 3'UTR of the SCD1 gene could therefore explain some of the within-breed variations in MUFA content of milk fat.  相似文献

Conjugated linoleic acid (CLA) has been shown to have an effect on subcutaneous fatty acid composition and has been reported to decrease stearoyl coenzyme A desaturase (SCD) activity by decreasing mRNA expression and(or) catalytic activity in rodents and rodent cell lines. This investigation was designed to study the effects of CLA, corn oil, or beef tallow supplementation on s.c. adipose tissue fatty acid composition, adiposity, SCD enzyme activity, and the delta9 desaturase index in piglets. Eighteen crossbred barrows 16 to 18 d of age were adapted to diet for 1 wk and then assigned randomly to one of three treatments: 1.5% added CLA, 1.5% added corn oil, or 1.5% added beef tallow. Barrows were penned individually and fed the supplemental oils for 35 d (to 25.6 +/- 0.6 kg BW). Subcutaneous adipose tissue samples were obtained after slaughter. Fatty acid composition of the s.c. adipose tissue differed for each fatty acid measured due to diet with the exception of 18:3. The concentrations of CLA trans-10, cis-12 and cis-9, trans-11 were elevated from nondetectable to 1.62 and 2.52 g/100 g lipid, respectively (P < 0.001 for both isomers). Conjugated linoleic acid decreased the delta9 desaturase index (P < 0.01) and SCD enzyme activity, expressed as nanomoles of palmitate converted to palmitoleate/(7 min x g of tissue) (P = 0.075) and nanomoles of palmitate converted to palmitoleate/(7 min 105 cells) (P= 0.056). Tallow-fed pigs had a greater proportion of large adipocytes (> 700 pL) and the greatest SCD activity. These data provide the first direct evidence that dietary CLA depresses SCD enzyme activity in porcine adipose tissue, which may in part be responsible for the depression of adiposity by CLA observed by others in market weight pigs.  相似文献

The present study investigated (i) the effect of breed on the expression of stearoyl-CoA desaturase (SCD) protein and fatty acid composition in Semimembranosus muscle and subcutaneous adipose tissue of beef cattle and (ii) the relationship between SCD expression, cis-9, trans-11 conjugated linoleic acid (CLA) content, and monounsaturated fatty acid (MUFA) level. The study was conducted on the following breeds: Longhorn (L), Charolais cross with Holstein–Friesian (CX), Hereford (H), Belted Galloway (BG) and Beef Shorthorn (BS). Significant breed differences in the total fatty acid content, saturated fatty acid (SFA) level, MUFA and n−3 PUFA content were observed in subcutaneous adipose tissue but not in muscle. In the case of CLA, the breed differences were observed in both muscle and subcutaneous adipose tissue, with the highest level in L and the lowest level in H. In the case of subcutaneous adipose tissue, the breed with the highest CLA content (L) also had the highest SCD protein expression. The breed-specific pattern of SCD expression in subcutaneous adipose tissue was similar to the breed-specific pattern of one of the products of an SCD-catalysed reaction, C16:1 (BS < BG < H < CX < L). It has been concluded that (i) the mechanisms regulating SCD protein expression and CLA level in beef cattle are tissue-specific; (ii) breed-specific variations in SCD expression might contribute to breed variations in MUFA and CLA content in subcutaneous adipose tissue but not in Semimembranosus muscle.  相似文献

Leptin gene has been found to be associated with various economic traits including milk production and fat quality in dairy animals. In the present study, we investigated genetic variations in intron 1 region of leptin gene in riverine buffaloes (Bubalus bubalis) using polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) and sequencing methods and associated them with milk traits. The study revealed three SSCP variants A, B and C among a total of 301 buffaloes from nine breeds. The frequency of variant C was found invariably high among all the breeds except in Marathwada buffalo. Variant A was found to be absent in Chilika, Nili-Ravi, Nagpuri and Pandharpuri breeds and also had the lowest frequencies in Mehsana, Jaffarabadi, Murrah and Toda breeds. Sequencing of SSCP variants revealed a total of five polymorphic sites, with three haplotypes. Statistical analysis revealed significantly high fat percentage at 150?days in SSCP variant B in Mehsana buffaloes. However, the associations of SSCP variants of leptin gene with total milk yield, 305?days milk yield and total fat yield were found to be non-significant. The present study is the first report on association analysis of leptin gene polymorphisms with milk production and milk quality traits in river buffalo.  相似文献

The amount of monounsaturated fatty acid (MUFA) is intimately related to adipose softness, melting point (MP) and flavor in beef. Stearoyl‐CoA desaturase (SCD) is a main gene involved in MUFA synthesis. Mature adipose tends to be highly saturated, whereas immature or maturing adipose is highly unsaturated when chronologically based, so the degree of non‐saturation can be an index of adipose maturity. In this study, three different adipose tissues (coelomic (CL), perirenal (PR), and subcutaneous (SC)) from three beef breeds with differing slaughter ages (Japanese Black (29.5 months), Holstein (20.1 month), and F1 crossbreed (25.6 months)) were examined to: (i) determine adipose maturity level as indexed by MUFA %; and (ii) determine SCD and other lipogenic gene messenger RNA (mRNA) expression levels in relation to unsaturated fatty acid content. Fatty acid composition was significantly different between adipose tissues (P < 0.05). MUFA amount was high in the following order: SC > CL > PR. This pattern corresponded to SCD mRNA expression profile showing higher expression in SC than CL and PR. However, Japanese black cattle are an exception with CL adipose containing similar UFA % as SC adipose, yet having the lowest SCD mRNA expression level among all adipose tissues tested. Therefore, SCD mRNA expression and MUFA % appear to be directly related; however, differences in SCD mRNA expression among three adipose tissues may reflect differences in the fat development characteristics affected by chronological age of the cattle breeds.  相似文献

乙酰辅酯A∶二酰甘油酰基转移酶(DGAT1)是甘油三脂合成过程中唯一的关键酶,对动物机体脂肪代谢、沉积起重要作用,已成为研究奶牛泌乳性状和肉牛主要经济性状的重要候选基因之一。近年来,随着相关研究的不断深入,研究者普遍认为DGAT1基因对奶牛的产奶量、乳脂率、乳蛋白率、乳糖率、乳脂肪酸及空怀天数、输精次数、初配年龄等繁殖性状均有显著影响,且与肉牛体脂也存在相关性。作者主要介绍了DGAT1基因的遗传多态性及其与经济性状关联分析在中国各地区不同品种奶牛、肉牛、水牛和牦牛上的研究进展,指出该基因对奶牛的产奶性能及肉牛的肉质性状、生长性状具有重要的调控作用。但由于环境条件、品种及选育背景等因素影响,DGAT1基因遗传多态性对不同地区不同牛群体的经济性状影响不尽相同。因此,要将DGAT1基因实际应用在品种选育工作中还需明确该基因对研究群体的具体遗传效应,并结合其他候选基因进行综合分析。  相似文献

The cis-9, trans-11 C18:2 (CLA) content in milk fat of four pure sheep breeds (Awassi, Lacaune, Friesland and Chios) was examined. All sheep were kept indoors all year round under the same feeding practices, without any grazing at all. Sheep nutrition was based on alfalfa hay and concentrates. A total of 237 individual milk samples were collected at three sampling times (December, January and March) from sheep of different parity and different days in milk, for CLA determination. The results showed that: (a) there was a large variation in milk fat CLA content among individuals consuming the same diet, (b) the CLA content of milk fat was lower in Friesland breed, compared to the other three breeds but without any significant difference among the four sheep breeds, (c) the CLA content of milk fat was not affected by ewes parity or days in milk, (d) there was a negative but not significant correlation between milk fat and CLA concentration, (e) the CLA content of milk fat was not correlated with milk yield. In conclusion, no others factors, such as breed, parity, days in milk, etc., can affect CLA content in milk fat, which means that the dietary ones remain the sovereign factors explaining the highest proportion of CLA content variability in sheep milk fat.  相似文献

Fibroblast growth factor 21 (FGF21) is a hepatic hormone that regulates peripheral glucose tolerance, energy balance and lipid metabolism. Prior evidence suggests that FGF21 may have the potential to favorably reduce obesity. The objective of the present study was to identify single nucleotide polymorphisms (SNPs) of bovine FGF21 using 1255 animals representing the five main Chinese breeds and to investigate the effect of these SNPs on economic traits in Nanyang cattle. Four significant SNPs were identified, one was a synonymous mutation and the other three were in intronic regions. The polymorphism information content (PIC) analysis showed that four beef cattle populations (NY, JX, LX and QC) had a moderate genetic diversity at the four loci while the beef and dairy population (CRS) had a low level. Additionally, allele and genotype frequencies for the beef breeds were significantly different from CRS, implying that these mutations are possibly associated with some quantitative traits. Moreover, linkage disequilibrium analysis and haplotype frequencies were also reported. Seven different haplotypes were identified and haplotype TCCC was predominant in all five cattle breeds. Association analysis suggested that SNPs g.297C>G and g.940C>T of bovine FGF21 were associated with higher body weight at 18 months within NY cattle, which would contribute to cattle breeding and genetics through marker-assisted selection (MAS).  相似文献

为探究山羊CLAⅠα链基因的多态性,本试验根据GenBank中山羊CLAⅠα链基因序列设计引物,采用RT-PCR技术从10个品系山羊白细胞中克隆山羊CLAⅠα链基因,测序后进行生物信息学分析。结果表明,CLAⅠα链基因长度为1 074 bp,编码357个氨基酸。经核苷酸序列分析发现,10个品系山羊CLAⅠα链基因同源性集中在82.8%~99.5%之间。将10个品系山羊CLAⅠα链基因核苷酸推导的氨基酸序列进行比对,发现突变集中在α1区域(22-110位氨基酸)与α2区域(111-203位氨基酸)。遗传进化分析表明,GenBank中山羊CLAⅠα链基因与红寺湖绒山羊、河西绒山羊、陕北绒山羊、阿尔巴斯绒山羊的CLAⅠα基因遗传关系较近,与其他6个品系山羊,即简阳大耳山羊、金堂黑山羊、西农萨能奶山羊、美姑山羊、大足黑山羊、努比亚山羊的CLAⅠα基因遗传关系较远。本试验结果为进一步研究CLAⅠα链基因的多态性奠定了理论基础。  相似文献

本研究旨在探索中国荷斯坦牛SCD1基因多态性与产奶性状的相关性.以上海某奶牛场610头中国荷斯坦牛为试验材料,采用PCR-SSCP法对SCD1基因A293V位点遗传多态性进行分析,利用一般线性模型分析SCD1基因A293V位点突变对测定日产奶量、乳脂率、乳蛋白率、305天校正产奶量、乳脂量、乳蛋白量及体细胞评分7个泌乳性状的影响.共检测到AA、AV和VV 3种基因型,频率分别为0.634、0.323和0.043,等位基因A和V的频率分别为0.796和0.204.该位点突变对乳脂率、乳蛋白率的影响达到显著水平(P<0.05),对测定日产奶量、305天校正产奶量、305天乳蛋白量和305天乳脂量以及体细胞评分影响不显著(P>0.05).多重比较表明,VV基因型个体的乳蛋白率、乳脂率极显著高于AA、AV型个体(P<0.01).SCD1基因A293V位点突变对中国荷斯坦牛泌乳性状的遗传效应有一定程度的影响,但影响机理需要进一步的研究.  相似文献

The fatty acid composition of adipose tissue in beef has been recognized as an important trait because of its relationship with beef quality, including favorable beef flavor and tenderness. Over the last decade, we have tried to identify the genes responsible for the fatty acid composition in cattle, and have found the following. (i) Genetic polymorphism of stearoyl‐CoA desaturase (SCD) is one of the responsible genes associated with fatty acid composition. The average effects of gene substitution of the SCD type A gene on the monounsaturated fatty acid (MUFA) percentage and the melting point of intramuscular fat were approximately +1.0% and ?1.0°C, respectively. (ii) Intron polymorphism of sterol regulatory element binding protein–1 (SREBP‐1) also affected MUFA. (iii) No effect of SCD or SREBP‐1 genotypes on any representative carcass traits of Japanese Black in the field population was observed. (iv) Additional genetic markers adipocytes fatty acid binding protein 4 (FABP4) and liver X receptor α also affected the fatty acid composition. (v) SCD and FABP4 significantly affected fatty acid composition in Holstein steers. These findings will bring new insight into the fat‐related carcass traits of beef cattle and will thus contribute to the beef industry.  相似文献

Gonadotropin releasing hormone and its receptor (GNRHR) play a critical role in sexual differentiation and reproduction. Available evidence shows a strong genetic component in the timing of puberty. In bovines, there are significant differences within and among beef breeds in the time when bulls reach puberty. Despite its economic importance, there are not many SNPs or genetic markers associated with this characteristic. The aims of the study were to identify DNA polymorphism in the bovine GNRHR by re-sequencing analysis, determine haplotype phases, and perform a population study in a selected tag SNP in six breeds. Eight SNPs were detected, including: one in the Upstream Regulatory Region (URR), five in the coding regions, and two in non-coding regions. This polymorphism level corresponds to one variant every 249.4 bp and a global nucleotide diversity of 0.385. Two haplogroups comprising nine haplotypes and two linkage blocks were detected. Despite 5 tag SNPs were required to capture all variability, just one SNP allowed to define both haplogroups, and only two SNPs were needed to differentiate the most common haplotypes. An additional taq SNP was necessary to identify both URR variants. Allele-frequency analysis of a selected taq SNP among breeds showed a geographical cline. European Bos taurus breeds had lower frequencies of the C allele than B. indicus type cattle, while Creole cattle and Wagyu breeds had intermediate frequency. There was a significant correlation between frequency profile and timing of puberty among the studied breeds, which seems to suggest that genetic variation within bovine GNRHR gene could explain at least part of the reported variability.  相似文献

The objectives of the study were to evaluate allelic frequencies and to test the association of polymorphisms in the calpastatin (CAST) and μ-calpain (CAPN1) genes with meat tenderness in 3 French beef breeds. A total of 1,114 Charolais, 1,254 Limousin, and 981 Blonde d'Aquitaine purebred young bulls were genotyped for 3 SNP in the CAST gene and 4 SNP in the CAPN1 gene. Two of these markers, 1 in each gene, can be found in Australian or American commercial genetic tests. Others have previously been reported in American studies or are newly evidenced SNP. The quantitative traits studied were Warner-Bratzler shear force and a tenderness score evaluated by trained sensory panels. All the SNP were informative in the 3 breeds. Associations of individual markers or haplotypes with traits were analyzed. The results differed in the 3 breeds. The G allele of a CAST marker (position 97574679 on Btau4.0) was found to exert a significant effect on the shear force (+0.18 phenotypic SD; RSD) and tenderness score (-0.22 RSD) in the Blonde d'Aquitaine breed. In the same breed, this marker was associated with another CAST SNP (position 97576054 on Btau4.0) such that the GA haplotype appeared to be associated with tougher meat. Two CAPN1 markers (positions 45221250 and 45241089 on Btau4.0) had a significant effect on both traits in the Charolais breed (from |0.11| to |0.25| RSD). In the same breed, these markers were associated with another CAPN1 SNP (position 45219395 on Btau4.0) such that the ACA and AGG haplotypes appeared to be associated with a tender meat and a tougher meat, respectively. Consequently, the present results indicate that the effects of the markers studied are breed-specific and cannot be extended to all Bos taurus breeds. Further studies are also required to identify other more appropriate markers for French beef breeds.  相似文献

本研究探讨了引进牛种(西门塔尔牛、利木赞牛)和山东地方黄牛(鲁西黄牛、利鲁牛)在20个微卫星位点上的多态性。试验从不同地区的种公牛站采集鲁西黄牛、利鲁牛、利木赞牛和西门塔尔牛的血液或精液样本,分别为56、27、31和30个,共计144个样本,血液样本采用Lab-Aid基因组DNA分离试剂盒提取基因组DNA,精液样本采用高盐法提取基因组DNA。利用20个微卫星标记,采用荧光标记毛细管电泳方法,对4个肉牛品种的DNA多态性进行分析,主要研究了每个群体的遗传变异指标(等位基因数、多态信息含量和杂合度)及群体间的遗传关系(F-统计量和基因流)。结果表明,肉牛基因组DNA条带整齐,无拖尾现象,质量较好,可用于本试验。20个微卫星座位中共检测到211个等位基因,平均等位基因数为10.6个。群体的平均观测杂合度在0.6147~0.7176之间,平均期望杂合度在0.6735~0.7862之间。在所有群体中各位点的多态信息含量在0.4853~0.8714之间,平均为0.7284,但在各个群体内的差异较大。近交系数及基因流分析结果表明,4个肉牛品种近交系数较低,群体间平均近交系数为0.085,每个微卫星位点的基因流均>1。总体来看,所选用的20个微卫星座位多态信息含量高,可作为有效遗传标记用于肉牛品种间遗传多样性分析。  相似文献

Trans10, cis12 conjugated linoleic acid (t10c12‐CLA) is well‐established in decreasing milk fat content and causing milk fat depression (MFD) in dairy cattle and goats. However, the detailed mechanisms of its effect are not completely understood. Therefore, we used goat mammary epithelial cells (GMECs) to further study the molecular mechanisms whereby t10c12‐CLA regulates milk fat synthesis. The optimal concentration of t10c12‐CLA (100 μmol/L) for cell culture was determined through a cell vitality and morphology assay, and evaluation of abundance of apoptosis‐related proteins. Oil red O stain indicated that t10c12‐CLA increased concentration of cytoplasmic lipid droplets. Furthermore, t10c12‐CLA increased the intracellular triacylglycerol (TG) content (< 0.05). Among 16 genes related to lipid metabolism that were measured by quantitative real‐time PCR, t10c12‐CLA down‐regulated (< 0.05) genes involved in de novo fatty acid synthesis including FASN, ACACA and SCD1, and also down‐regulated the protein expression of FASN and SCD1 but up‐regulated (< 0.05) the expression of CD36 and ADRP. Overall, the data indicate that a side effect of de novo fatty acid synthesis inhibition by t10c12‐CLA is the up‐regulation of fatty acid uptake and accumulation of lipid droplets in GMECs. The biologic reason for such an effect merits further study.  相似文献

Conjugated linoleic acids (CLA) are a group of polyunsaturated fatty acids (PUFA) with a single pair of conjugated double bonds. The major natural CLA isomer is 18:2 cis-9, trans-11 (c9, t11) linoleic acid, or rumenic acid (RA). Chemically synthesized CLA is also available, mostly as a mixture of RA and 18:2 trans-10, cis-12 (t10, c12) isomers in equal amounts (50:50). Consumption of ruminant meat (beef and lamb) and dairy products (milk and cheese) is the main source of dietary exposure to CLA. Despite numerous studies on animal and human models (tumorigenesis, obesity, immune response) it has not been established whether additional supplementation of CLA is of benefit. Moreover, some studies, conducted both in animals and in humans, reveal that CLA isomers may induce insulin resistance. Presently, balanced diet rich in CLA from natural sources is recommended. The purpose of this review was to sum up the results available in the literature.  相似文献

Feeding sheep concentrate-based diets increases the oleic acid content of their tissues, whereas the cis-9, trans-11 conjugated linoleic acid (CLA) content is increased by feeding forage diets. Both these metabolic transformations could be attributable to increased activity of stearoyl-CoA desaturase (SCD). Therefore, the effect of forage or concentrate feeding regimens on the fatty acid composition of sheep tissues were investigated to determine whether any changes are related to an alteration of SCD mRNA levels. Twenty-four ewe lambs were randomly allotted to one of three dietary treatment groups: 1) dehydrated grass pellets, 2) concentrate diet fed to achieve a growth rate similar to that of the dehydrated grass pellets, and 3) the same concentrate diet approaching ad libitum intake. As expected, animals fed ad libitum concentrates grew at a greater (P = 0.001) rate (280 g/d) than those fed either of the other two diets (180 g/d), which were similar. In samples of liver and the three adipose tissue depots studied, the concentration of oleic acid from sheep fed either level of the concentrate diet was greater (P < 0.001) than from animals fed forage. This was associated with an increase (P < 0.05) in the ratio of SCD to acetyl-CoA carboxylase mRNA in adipose tissue and liver. Compared with concentrate-fed, the forage-fed lambs had increased (P < 0.05) levels of the cis-9, trans-11 isomer of CLA and C18:1, trans-11 in all their tissues, although the levels of SCD mRNA were lower. It therefore seems that the increased oleic acid content of sheep tissues in response to concentrate-rich diets is associated with an increase in SCD gene expression. By contrast, the increased concentration of CLA in animals fed forage-based diets is associated with an increase in substrate (C18:1 trans-11) availability.  相似文献
