Forty-two kiwifruit orchards from Rize and Samsun provinces (Black Sea region, Turkey) exhibiting symptoms of root rot disease were examined between 2009 and 2010. Twenty-four fungal isolates recovered from affected roots were included in this study. Morphological characteristics of all isolates were recorded on potato dextrose agar, malt extract agar and Spezieller Nährstoffarmer agar. The effect of temperature on radial colony growth was also evaluated at 5, 10, 15, 20, 25, 30, and 35 °C. Histone H3 gene (HIS) was amplified with primers CYLH3F and CYLH3R and the amplified fragments were sequenced. The HIS phylogeny grouped all the isolates into six well-supported clades which were in agreement with phenotypical characteristics. Isolates were identified as “Cylindrocarponpauciseptatum, Cylindrocladiella parva, Ilyonectria liriodendri, I. torresensis, I. robusta and I. europaea, I. liriodendri being the most frequent species. All of them are reported for the first time on kiwifruit in Turkey, with the exception of I. liriodendri. Pathogenicity tests with selected isolates showed that 10 out of 11 isolates tested were able to induce typical root rot disease symptoms, affecting plant development and leading to the death of some plants. This study shows the high diversity of root rot pathogens found in kiwifruit trees in the Black Sea region of Turkey, being the first step towards a better understanding and management of the disease in local conditions.  相似文献

The presence, distribution or abundance of many invasive alien plants is positively correlated with roads, so roads need to be taken into consideration when planning a survey in a poorly studied area. During the two field surveys organized in the framework of the 2nd Workshop on Invasive Plants in the Mediterranean Type Regions of the World, 1 81 alien species were observed in the investigated area, i.e. 70 neophytes and 11 archeophytes (including 9 doubtful species), with 54 new records for the DAISIE inventory. Three of these species, Acalypha australis, Microstegium vimineum and Polygonum perfoliatum, were recorded near a tea factory, and the import of material for tea processing is expected to have been their pathway of introduction. The results of this survey in the region of Trabzon in North‐East Turkey show that roadside surveys are a useful tool for early detection efforts, in compiling and updating national or regional inventories (especially with time and budget constraints). This survey, being organized in the framework of an international workshop, enabled knowledge to be shared between experts in the field, and training of students and researchers. These survey methods could be adapted, improved, and used elsewhere by others seeking to use early detection as part of their overall weed strategy or to gather baseline data on invasive alien plants in a poorly studied area.  相似文献

Rhizoctonia solani and R. cerealis were isolated from diseased sugar-beet seedlings in Ireland. Isolates of R. solani were assigned to anastomosis groups AG-2, AG-4, AG-5 and an unidentified group that did not anastomose with recognized tester isolates. Cultures of AG-2 were similar to those of AG-5 on oatmeal agar (OA) and potato-dextrose-marmite agar (PDMA). Cultures of AG-4, the unidentified group and R. cerealis were morphologically distinct from one another, AG-2 and AG-5. The optimum temperature for growth of AG-2 was 225 C, with optimum growth of AG-4, AG-5 and the unidentified group at 275-C. R. cerealis grew slower than all groups of R. solani, with optimum growth at 225°C. Hyphae of R. cerealis were significantly narrower than those of the groups of R. solani studied. In glasshouse pathogenicity tests, some AG-2 and all AG-4, AG-5 and isolates from the unidentified group caused damping-off of beet seedlings. In controlled environments of 10-25°C, an AG-2 isolate was the most aggressive at 10 C whilst AG-4, AG-5 and the unidentified group caused most disease at or above 15°C. R. cerealis was also pathogenic to beet seedlings, causing damping-off at 10 and 15 C.  相似文献

Sixty-two isolates of Rhizoctonia spp. were collected from Belgian cauliflower fields during 2005 and 2006. The majority of the isolates (60 out of 62) had multinucleate cells and were identified as Rhizoctonia solani . Characterization of anastomosis groups (AGs) was performed using pectic zymograms, PCR-RFLP and sequencing of the rDNA-ITS region. The most prevalent AG was AG 2-1 (55% of isolates), followed by AG 2-1 subset Nt (11%), AG 1-1C (8%), AG 5 (8%), AG 4 HGII (6%), AG 3 (5%) and AG 1-1B (3%). Pathogenic potential towards different vegetable crops and towards maize was determined. Damage to cauliflower and endive was caused by different AGs, with the isolates aggressive towards cauliflower belonging to AG 2-1, AG 2-1 subset Nt, AG 4 HGII, AG 1-1C, AG 1-1B and AG 2-2, and those aggressive towards endive belonging to AG 1-1B, AG 1-1C, AG 2-1 subset Nt, AG 2-2, AG 4 HGII and AG 5. The most aggressive isolates towards bean belonged to AG 2-1 subset Nt and AG 2-2, for lettuce to AG 1-1B and AG 2-1, on carrot to AG 4 HGII and towards maize to AG 2-2. Within the isolates of AG 2-1, variability was observed in PCR-RFLP pattern and in aggressiveness towards several crops, indicating this subgroup to be heterogeneous. This is the first study concerning the occurrence of R. solani AGs causing wirestem in Belgian cauliflower fields and the first report of aggressive isolates of AG 1-1C, AG 2-1 subset Nt and AG 4 HGII associated with cauliflower.  相似文献

Ninety-eight isolates of Rhizoctonia spp. were obtained from barley and wheat grown in Erzurum, Turkey. Of these, 78% were Rhizoctonia solani (AG-2 type 1, AG-3, AG-4, AG-5 and AG-11), 10% were binucleate Rhizoctonia (AG-I and AG-K) and the remainder were Waitea circinata var circinata ( Rhizoctonia sp.). Among the binucleate Rhizoctonia , AG-I was not recovered from barley. In pathogenicity tests on barley and wheat, the highest disease severity was caused by isolates of AG-4 and AG-11, whereas isolates of AG-2 type 1, AG-3, AG-5 and W. c . var  circinata were moderately virulent. Isolates of binucleate Rhizoctonia were all nonpathogenic. This is the first report of R. solani AG-11 and W. c . var  circinata from Turkey.  相似文献

A total of 147 samples of garlic ( Allium sativum ) bulbs affected by blue mould were obtained from a variety of agroclimatic districts between December 1999 and February 2000. Penicillium species were identified using both morphological and chemotaxonomic characteristics. Penicillium allii was the predominant species isolated (81·8%) in this survey and the only species proven to be pathogenic on garlic. Other species were isolated much less frequently: P. chrysogenum (13·7%), P. brevicompactum (2·8%), P. phoeniceum (0·9%), P. aurantiogriseum (0·6%) and P. flavigenum (0·2%). Colonies of P. allii could be classified into four morphotypes and their distribution seemed to be influenced by seed trade and agricultural practices. Penicillium allii isolates were grouped into three aggressiveness phenotypes (low, medium and high) based on their ability to cause disease during field trials on susceptible (Fuego INTA) and less susceptible (Castaño INTA) garlic cultivars. The number of surviving plants at 191 days after planting and postharvest bulb weight contributed the most towards aggressiveness modelling.  相似文献

Two-hundred and forty-eight isolates of Rhizoctonia spp, were obtained from 13 locations in Gifu Prefecture in Japan using the plant debris particles isolation, colonization of bait tissue, and soil-clump plating methods. Of the isolates, 143 were binucleate Rhizoctonia spp., 60 were R. solani and 45 were R. zeae. Three isolates of R. solani and 54 of binucleate Rhizoctonia spp, were hypovirulent on radish, whilst all isolates of R. zeae were highly virulent, Hypovirulent strains were isolated most frequently by the plant debris particles isolation method, Hypovirulent isolates of R. solani belonged to anastomosis group 4, whilst the hypovirulent binucleate Rhizoctonia isolates belonged to AG A, AG Ba, AG G, and AG O.
Thirty-two isolates of Rhizoctotria spp, selected for hypovirulence on radish were tested on cucumber in vitro. Only five binucleate Rhizoctonia isolates and one R. solani isolate were hypovirulent on both species, and these isolates were also hypovirulent on seven other crop species. Cucumber showed wide variation in disease susceptibility to different isolates but hypovirulent isolates exhibited a consistent reaction on five different host cultivars, Pathogenicity tests using cucumber grown in soil also showed consistent reactions with isolates selected either for hypovirulence or virulence. The results support the use of cucumber in bioassays for identifying hypovirulent isolates of binucleate Rhizoctonia spp.  相似文献

新疆北疆棉田立枯丝核菌不同菌丝融合群致病力的研究   总被引:1,自引:0,他引:1  
从新疆北疆棉区采集了典型的棉花立枯病病苗及棉田土标样686份,按常规分离方法分离得到399个分离物,从中鉴定出272个纯化的立枯丝核菌(Rhizoctonia solani Kühn)菌株。用标准菌株,通过载玻片菌丝融合试验测定,将纯化的272个菌株划归为3个菌丝融合群:即AG-2、AG-4和AG-5,分别占总菌株的6.24%、84.2%和1.1%。另有23个菌株不与任何标准菌株融合,占8.46%,说明新疆北疆棉田立枯丝核菌的优势菌系是多核丝核菌的AG-4融合群。通过从10种不同配方培养基中筛选效果好的麦芽蛋白胨(MPDA)配方培养基(Ⅱ)进行对峙培养,将纯化获得的272个丝核菌菌株,划分为6个不同的营养亲和群。将多核的不同菌丝融合群及其各营养亲和群代表菌株在3种不同主栽棉花品种上(每品种都加不接菌的对照)进行温室盆栽致病力测定,结果以AG-4对棉花的致病力最强,其次是非融合类,AG-2和AG-5虽致病,但并不致死苗,其平均病指数分别为94.9、81.4、53.1、43.5。  相似文献

Four-hundred-sixty-eightFusarium andFusarium-like isolates were obtained from crowns and subcrown internodes of winter wheat grown in Erzurum, Turkey. Of these isolates, 34.8% wereFusarium acuminatum, 32.3% wereF. equiseti, 16.9% wereF. oxysporum, 15.0% wereMicrodochium nivale (formerlyFusarium nivale), 0.6% wereF. tabacinum and 0.4% wereF. solani. In pathogenicity tests on wheat, the highest disease severity was caused by isolates ofM. nivale, whereas isolates ofF. acuminatum, F. equiseti, F. oxysporum andF. solani were slightly virulent; isolates ofF. tabacinum were nonpathogenic. Chestnut blight caused by the introduced fungus Cryphonectria parasitica has been responsible for the decline of Castanea sativa in Turkey since the 1960s. In this study, 72 C. parasitica isolates were recovered from the Marmara and Black Sea regions of Turkey showing white or cream-coloured culture morphology and were subjected to various tests to determine if they were infected by Cryphonectria hypovirus 1 (CHV-1). The vast majority of the isolates (69 out of 72) were vc type EU-1. Both mating types were found among a subsample of the isolates. The hypovirus was detected in 55 isolates by dsRNA extraction and/or virus specific RT-PCR on total RNA extracts. All but one isolates showed no or only weak phenol oxidase activity on agar medium containing tannic acid, typical of CHV-1 infected isolates. Through sequencing of a specific region of the hypovirus genome, we found that 24 hypovirus isolates belonged to the CHV-1 subtype I and six to the CHV-1 subtype F2. The distribution of the two CHV-1 subtypes in Turkey showed a clear geographic pattern. CHV-1 subtype I was only detected in the Marmara and western Black Sea region, whereas subtype F2 was restricted to the eastern part of the Black Sea region. The effectiveness of 23 hypovirulent isolates was tested against a virulent isolate on 2–3 years old chestnut sprouts. Ten hypovirulent isolates, all infected by CHV-1 subtype I, prevented canker development by more than 80 % suggesting that they might be suitable for biological control of chestnut blight in Turkey.  相似文献

 本研究从陕西分离得到61株苹果树腐烂病菌(Cytospora spp.),选取在PDA培养基上菌落特征差异明显的7个分离株,对其生物学特性及致病性进行研究。结果表明:各分离株在PDA、PSA、PMA和PEMA培养基上菌落生长较好,但在PDA上的菌落颜色和产孢情况差异很大。各分离株均能在5~32℃生长,但对35℃以上高温的适应性差异较大;所有分离株均能在pH4~9的条件下生长,最适pH为5~6;光照对各分离株的菌落生长有明显的促进作用。各分离株均可在Cza-pek培养基及供试的其它碳、氮源培养基上生长,其中以葡萄糖、麦芽糖和酵母膏为最佳碳、氮源。采用烫伤接种的方法,分别以菌饼和分生孢子悬浮液接种苹果离体枝条,两者均可侵染发病,但各分离株致病力差异显著,其中菌落颜色为黄褐色的菌株致病性强。因此,根据菌落颜色、致病性和生物学特性可将这7个分离株划分为3个类群:Ⅰ型为黄褐色强致病类群,但各分离株的产孢量有差异;Ⅱ型为乳白色不产孢的弱致病类群;Ⅲ型为灰褐色易产孢弱致病类群。其余54株分离物均属于Ⅰ型,这些结果说明陕西省苹果树腐烂病菌具有多样性和致病性分化现象。  相似文献

ABSTRACT Rhizoctonia spp. were commonly recovered from the roots of strawberry plants growing in nonfumigated soil in the central coastal region of California. With the exception of one multinucleate isolate of R. solani (frequency of recovery of 0.8%), all other isolates were binucleate and were in anastomosis groups (AG) A, G, or I. AGs-A and -I were recovered from all five collection sites, whereas AG-G was recovered from only two sites. AG-A was the most commonly isolated AG, followed by AGs-I and -G. Similar levels of virulence were observed among the different AGs, but differences in virulence were observed among isolates in the same AG. Evaluating anastomosis grouping by pairing isolates recovered from strawberry with known tester isolates did not always yield a positive anastomosis reaction, even though both isolates anastomosed with other members of the same AG. Subsequent investigations with multiple isolates in the same AG from the same collection location confirmed that there was a lack of anastomosis or weak anastomosis reactions for some combinations of pairings, highlighting the need for to use multiple tester isolates or molecular techniques for AG determination. Restriction fragment length polymorphism (RFLP) analysis of a polymerase chain reaction-amplified region of the rDNA was effective for differentiating AGs. Sixteen RFLP groups were observed after cluster analysis with data for the size of the amplified products and fragment sizes after digestion with four restriction enzymes. Although each AG had isolates in multiple RFLP groups, any one individual RFLP group contained isolates of only a single AG. There was no consistent correlation between RFLP group and location of isolate collection.  相似文献

In the last years, leafy vegetables cultivated as baby leaves have been established in the market and have attracted the interest of consumers throughout the world. During the growing seasons of 2019 and 2020, 97 isolates of Rhizoctonia solani and 112 isolates of Pythium spp. were obtained from baby leaf vegetables exhibited damping-off symptoms. Representative isolates of R. solani from each surveyed plant species were characterized using sequence analysis of the internal transcribed spacer (rDNA-ITS) region. Isolates were identified as belonging to four anastomosis groups (AGs): AG2-1, AG-IB, AG4-HGI and AG4-HGIII. AG4-HGI was the most prevalent group and phylogenetic analysis showed that the isolates were distinctly separated according to their AGs. Pathogenicity among the four AGs on 23 plant species varied considerably, from not susceptible to highly susceptible, while, in general, AGs did not exhibit host specificity. Furthermore, a total of 112 Pythium spp. isolates were obtained. The ITS region and the cytochrome oxidase II (coxII) gene were amplified, and three Pythium spp. were identified (P. ultimum, P. aphanidermatum and P. sylvaticum), which were used further for maximum-likelihood phylogenetic analysis. The pathogenicity of representative isolates was assessed in vitro and in vivo on 10 plant species. In general, all three tested Pythium spp. were virulent when used in vitro, while P. ultimum was the most virulent in vivo. This is the first comprehensive study aimed at determining the occurrence of specific R. solani AGs and Pythium spp. derived from baby leafy vegetables exhibiting damping-off symptoms in Greece.  相似文献

Roots of seedlings of wheat and barley affected by bare patch disease at a field site in Western Australia were assessed for root damage and plated to isolate fungi. The patches were variable in shape and size and had the most severely affected plants in the centre. Of the 165 isolates ofRhizoctonia spp. obtained, 90% were multinucleate and 10% binucleate, the former being predominant in the plants at the centre of the patch. The relative frequency of binucleate isolates increased with proximity to the periphery. The increase in activity of avirulent binucleate isolates towards the periphery of the patch may be related to the sharp and abrupt edging of the patch. A variety of other species of fungi such asFusarium spp.,Mortierella spp.,Bipolaris sorokiniana, Pythium sp. andTrichoderma sp. were encountered within the patches. The multinucleate isolates belonging to anastomosis groups (Ag) 2–1, 2–2 and 8 (Thanatephorus cucumeris) were most pathogenic to wheat. The binucleate isolates of Ag C, D, E, and K (Ceratobasidium sp.) were less pathogenic. It is suggested that the bare patch disease is caused by a complex of root rot fungi composed of one or more anastomosis groups ofRhizoctonia spp. and other associated fungi.Samenvatting Van kiemplanten van tarwe en gerst, afkomstig van een met kale-plekkenziekte besmet perceel in West Australië werd de mate van wortelbeschadiging bepaald en werden schimmels uit de wortels geïsoleerd. De meerkernige isolaten die tot de anastomosegroepen Ag 2–1, 2–2 en 8 (Thanatephorus cucumeris) behoren, waren voor tarwe het meest pathogeen. De tweekernige isolaten van de anastomosegroepen Ag C, D, E en K (Ceratobasidium sp.) waren minder pathogeen. ABSTRACT Isolates of Rhizoctonia spp. were obtained from rice in India during 2000-2003. Characterization by conventional techniques and polymerase chain reaction showed that from 110 isolates, 99 were R. solani and 11 were R. oryzae-sativae. Of 99 isolates identified as R. solani, 96 were AG1-IA, 1 was AG1-IB, and 2 were AG1-IC. Amplified fragment length polymorphism (AFLP) analyzes were used to determine genetic relationships in Rhizoctonia pathogen populations collected from different geographic regions. Cluster analysis based on the AFLP data separated isolates belonging to the three different intraspecific groups of R. solani AG1 and differentiated R. solani from R. oryzae-sativae. Analysis of molecular variance (AMOVA) revealed that geographic region was the dominant factor determining population structure of R. solani AG1-1A; host cultivar had no significant effect. Pathogenicity tests on Oryza sativa cv. Zenith revealed that isolates of R. solani AG1-1A and AG1-1B were more virulent than R. solani AG1-IC and R. oryzae-sativae isolates.  相似文献

ABSTRACT Rhizoctonia spp. were isolated from the roots of apple trees and associated soil collected in orchards located near Moxee, Quincy, East Wenatchee, and Wenatchee, WA. The anastomosis groups (AGs) of Rhizoctonia spp. isolated from apple were determined by hyphal anastomosis with tester strains on 2% water agar and, where warranted, sequence analysis of the rDNA internal transcribed spacer region and restriction analysis of an amplified fragment from the 28S ribosomal RNA gene were used to corroborate these identifications. The dominant AG of R. solani isolated from the Moxee and East Wenatchee orchards were AG 5 and AG 6, respectively. Binucleate Rhizoctonia spp. were recovered from apple roots at three of four orchards surveyed and included isolates of AG-A, -G, -I, -J, and -Q. In artificial inoculations, isolates of R. solani AG 5 and AG 6 caused extensive root rot and death of 2- to 20-week-old apple transplants, providing evidence that isolates of R. solani AG 6 can be highly virulent and do not merely exist as saprophytes. The effect of binucleate Rhizoctonia spp. on growth of apple seedlings was isolate-dependent and ranged from growth enhancement to severe root rot. R. solani AG 5 and AG 6 were isolated from stunted trees, but not healthy trees, in an orchard near Moxee, WA, that exhibited severe symptoms of apple replant disease, suggesting that R. solani may have a role in this disease complex.  相似文献

Characterization of Rhizoctonia solani from potato in Great Britain   总被引:1,自引:1,他引:1  
One hundred and thirty five isolates of Rhizoctonia solani were obtained from British potato crops between 2001 and 2003. Isolates were assigned to anastomosis group (AG) using conventional PCR assays for AG2-1 or AG3 or through the observation of hyphal interactions, where appropriate. A previously published primer set was modified in this study to enhance specificity for AG3PT. Most of the isolates (92·6%) belonged to AG3PT whilst some (6·7%) belonged to AG2-1. Only one isolate recovered (0·7%) belonged to AG5. Isolates of AG2-1 were diverse, with variation in both the length of the rDNA intergenic spacer 1 (IGS1) region and the categories of hyphal interaction observed between pairings of AG2-1 isolates. No variation in the length of the rDNA IGS1 region was observed amongst the AG3 isolates collected. Tests carried out on potato stems with a sub-set of the isolates revealed a wide range of aggressiveness amongst AG2-1 isolates. Sequencing of the rDNA internal transcribed spacer (ITS) region of the AG2-1 isolates and construction of a neighbour joining tree with other AG2-1 sequences available indicated that AG2-1 isolates with the short IGS1 region were closely related. This is the first investigation which provides evidence of the relative AG composition of R. solani populations causing disease in potato crops in Great Britain.  相似文献

Isolates of Rhizoctonia collected from the stems, roots, tuber sclerotia and soil of potato crops in Virginia and Lenswood, South Australia, were identified to anastomosis groups (AG). Of the 301 multinucleate isolates of Rhizoctonia solani tested, 90% were AG-3, 7% were AG-4 and 2% were AG-5; 12 isolates were binucleate Rhizoctonia spp. This is the first report of isolates of AG-4 and AG-5 causing disease in potato crops in South Australia. All AG-3, AG-4 and AG-5 isolates tested caused rhizoctonia disease symptoms on the potato cultivar Coliban in pathogenicity trials conducted under glasshotise conditions. Both AG-3 and AG-5 isolates caused black scurf and stem cankers, although symptoms of black scurf were less severe with AG-5. AG-4 isolates produced the most severe stem and stolon cankers of all isolates tested. The pathogenicity of tuber-borne inoculum was confirmed by growing plants from sclerotia-infested tubers. AG-8 isolates from diseased barley and wheat produced severe root cankers and caused loss of feeder roots on inoculated potato plants. Results suggest that rhizoctonia disease in potato fields in South Australia is caused by a combination of different anastomosis groups and this has important implications for crop rotations.  相似文献

ABSTRACT Recent reports have shown induction of resistance to Rhizoctonia root rot using nonpathogenic strains of binucleate Rhizoctonia spp. (np-BNR). This study evaluates the biocontrol ability of several np-BNR isolates against root and foliar diseases of cotton in greenhouse trials, provides evidence for induced systemic resistance (ISR) as a mechanism in this biocontrol, and compares the disease control provided by np-BNR with that provided by the chemical inducer benzothiadiazole (BTH). Pretreatment of cotton seedlings with np-BNR isolates provided good protection against pre- and post-emergence damping-off caused by a virulent strain of Rhizoctonia solani (AG-4). Seedling stand of protected cotton was significantly higher (P < 0.05) than that of nonprotected seedlings. Several np-BNR isolates significantly reduced disease severity. The combination of BTH and np-BNR provided significant protection against seedling rot and leaf spot in cotton; however, the degree of disease reduction was comparable to that obtained with np-BNR treatment alone. Significant reduction in leaf spot symptoms caused by Alternaria macrospora occurred on cotyledons pretreated with np-BNR or sprayed with BTH, and the np- BNR-treated seedlings had significantly less leaf spot than BTH-treated seedlings. The results demonstrate that np-BNR isolates can protect cotton from infections caused by both root and leaf pathogens and that disease control was superior to that observed with a chemical inducer.  相似文献
