This study investigates a long-held belief that mares tend to ovulate during the night. If this were so, a strategic examination regimen might replace multiple daily examinations when the time of ovulation needs to be known to within a few hours. Over 500 mares were examined at least three times daily to determine the time, to within ±4.25 hours, of 896 ovulations. The time of ovulation was determined as midway between the pre-ovulatory and post-ovulatory examinations. Three broad peaks of incidence of ovulation occurred at 5:00 am, 2:00 pm, and 10:00 pm. The interval between peaks was 9, 8, and 7 hours. during which 34.2% (5:00 am to 2:00 pm), 44.3% (2:00 pm to 10:00 pm), and 21.3% (10:00 pm to 5:00 am) of ovulations occurred. The percentage of ovulations occurring in the period 2:00 pm to 10:00 pm was significantly higher (P < .001) than in the period 10:00 pm to 5:00 am and significantly higher (P < .01) than in the period 5:00 am to 2:00 pm. The data showed that mares ovulated at all times of the day and night, with some increased incidence between 2:00 pm and 10:00 pm. Therefore, no strategic examination regimen was possible.  相似文献

Selection of sheep for resistance to gastrointestinal nematodes has been considered as one option to control nematode infection. It is essential to understand the influence of selection on the diversification of protein metabolism in individual tissues. In this experiment we examined fractional protein synthesis rate (FSR) and polyamine (putrescine, spermidine and spermine) and histamine concentrations in tissues of Merino sheep selected for low faecal worm egg count (parasite resistance) and control sheep. Sixty 18-month-old Merino rams, 40 from the resistant flock and 20 from the control line, were kept in individual pens of an animal house and fed ad libitum. Half of each genotype group were dosed orally with infective larvae of Trichostrongylus colubriformis and Ostertagia circumcincta thrice weekly to deliver 10,000 larvae from each species per week for 18 weeks. At the end of week 18, FSR was measured using a flooding dose of l-[ring-d5]phenylalanine, and polyamine concentrations were determined in all the sheep. Regardless of sheep genotype, the nematode infection significantly increased FSR in the mesenteric lymph nodes (37.0% vs. 32.6%/day, P < 0.01) and abomasal lymph nodes (28.8% vs. 16.0%/day, P < 0.001) and the bone marrow (5.12% vs. 3.96%/day, P < 0.01), but reduced FSR in skin (17.4% vs. 20.6%/day, P < 0.001), muscle (2.46% vs. 2.73%/day, P < 0.05) and liver (24.1% vs. 25.8%/day, P < 0.05). Nematode infection did not affect FSR in the jejunum. Polyamine concentrations in the tissues were generally increased by the infection (P < 0.05) except for a decline in spermine concentration in the muscle (P < 0.01). Overall there was no significant effect of selection for nematode resistance on tissue FSR or the concentration of amines. It was concluded that nematode infection resulted in a divergence of protein synthesis between tissues, and selection for nematode resistance in Merino sheep did not result in significant change in tissue protein synthesis compared with control sheep.  相似文献

Sixty-one animals with different Halothane genes (homozygous halothane positive, n = 34; and homozygous halothane negative, n = 27) were fed with three diets (control group, with no supplement; magnesium (Mg) group with 1.28 g MgCO3/kg and tryptophan (Trp) group with 5 g l-Trp/kg) during the last 5 days before slaughter. Animals were submitted to minimal stress antemortem conditions. Pig behaviour was recorded at the experimental farm, raceway to the CO2 stunning system and during the stunning period. Corneal reflexes were recorded after stunning as well. There were no differences in feed intake among diets (P > 0.05) during the 5 days of treatment. The halothane positive (nn) group had lower intake than the halothane negative (NN) group (P < 0.01). The behaviour of the pigs in the raceway did not differ (P > 0.05) among treatments or halothane genotype. A significant (P < 0.001) interaction diet  halothane was found in the time to appear the first retreat attempt during the exposure to the CO2 system. In the nn group, the time of performing the first retreat attempt was later in the Mg (P < 0.05) than the control group. Moreover, in the Mg group, the nn had a later (P < 0.05) first retreat attempt than the NN. Thus, Mg supplementation could have a positive effect on welfare of nn pigs. The nn had a lower proportion of animals that showed corneal reflexes after stunning than NN, indicating a higher effectiveness of the stunning method in nn pigs. Neither Mg nor Trp affected carcass quality and meat quality parameters, although significant differences were found between genotypes.  相似文献

The experiment investigated the ameliorative effects of l-glutamine administration on rectal temperature (RT), erythrocyte osmotic fragility (EOF), serum antioxidant enzyme activities and malondialdehyde (MDA) concentration in Red Sokoto goats during the hot-dry season. Twenty eight healthy Red Sokoto goats, comprising 14 experimental (administered 0.2 g/kg of l-glutamine dissolved in 10 mL of distilled water, once daily for 21 days) and 14 control (administered equivalent of distilled water) goats served as subjects. Rectal temperature (measured at 6:00, 13:00 and 18:00 h) and blood samples (taken at 8:00 h) were obtained from all subjects weekly, before, during and after l-glutamine administration. Data obtained were compared using one-way repeated-measures ANOVA, followed by Tukey’s post-hoc test. The dry-bulb temperature, relative humidity and temperature-humidity index for the study period ranged between 24.0 and 37.5 °C, 26.0 and 84.0% and 73.0 and 86.3, respectively. l-glutamine administration decreased (P < 0.05) RT, EOF and MDA and increased superoxide dismutase (SOD) activity in experimental group, compared to controls during weeks 1, 2 and 3. Glutathione peroxidase (GPx) and catalase activities were higher (P < 0.05) in the experimental group than in the controls only during week 1 of l-glutamine administration. In conclusion, l-glutamine administration mitigated increases in RT, EOF and serum MDA concentration and enhanced serum SOD, GPx and catalase activities and may be beneficial in heat-stressed goats during the hot-dry season.

In experiment 1, 6 pregnant mares received a concentrate that contained a trace mineral premix that provided 14.3 mg Cu, 40 mg Zn, 28 mg Fe, 28 mg Mn, 0.08 mg Co, 0.16 mg I, and 0.16 mg Se/kg concentrate (group A). Seven mares received the same concentrate plus 502 mg Zn and 127 mg Cu once daily (group B). No differences (P > .05) in foal growth data, or Cu, Zn, and Fe concentrations of mare milk, mare serum, or foal serum were observed. In experiment 2, 6 pregnant mares received the same concentrate as group A (group C), and 8 mares received the same concentrate fortified with 4× the trace mineral premix (group D). Group C mares had higher serum Zn concentration at 1 day (P < 0.01) and 56 days (P < 0.04). Group C mares had higher milk Fe concentration at 28 days (P < .01), and group D mares had higher milk Cu concentration at 56 days (P < .01). Group C foals had higher serum Cu concentration at 14 days (P < .03). The results from this study provide no evidence to indicate that supplementing late gestating and lactating mares with higher dietary trace mineral levels than those recommended currently by NRC has any influence on foal growth and development, or on the Cu, Zn, and Fe concentrations of the mare milk, mare serum, or foal serum.  相似文献

Eighty male pigs from 20 litters were used to evaluate dietary addition of 250 mg/kg of Zn from zinc amino acid complex (ZnAA) to sows during the last trimester of gestation and gastric intubation of 40 mg Zn from soluble zinc methionine (ZnMet) to suckling pigs at birth and on day 7 and 14 (weaning) on small intestinal morphology, Zn status and bacterial translocation in early-weaned pigs. At weaning, pigs were challenged with an intramuscular injection of saline without or with 120 μg/kg BW of lipopolysaccharide (LPS; from Escherichia coli O26:B6) and were euthanized 24 h later prior to collection of intestinal lymph nodes and small intestinal sections. Zinc concentration in serum 7 days after birth and at weaning were higher in pigs from ZnAA-supplemented sows and those receiving gastric intubation with ZnMet (P = 0.05 and P < 0.0001, respectively). Post-weaning liver tissue concentrations for Zn (P < 0.0001) and Fe (P = 0.04) were higher and for Cu lower (P < 0.0001) in pigs intubated with ZnMet. Pigs from ZnAA-supplemented compared with control-fed sows tended (P < 0.1) to have increased villi height and villus:crypt ratio in the jejunum and higher (P = 0.1) goblet cell counts in the ileum. Goblet cell counts of ZnMet-intubated (P = 0.03) and LPS-challenged pigs (P = 0.05) were also higher in the jejunum. Supplementation of ZnAA to gestating sows increased (P = 0.04) E. coli colony forming unit counts in the small intestinal mesenteric lymph nodes of early-weaned pigs. In conclusion, dietary addition of ZnAA to gestating sows and gastric intubation of ZnMet improved Zn status of suckling pigs.  相似文献

In this experiment the effect of digestible energy (DE) restriction on carcass traits and meat quality of rabbits selected divergently for body fat content was studied. Using a 2 × 3 factorial arrangement, Pannon White rabbits selected for high (HFAT) or low (LFAT) total body fat content were fed isocaloric diets between 4 and 12 weeks of age. Energy restriction was achieved by reducing the daily feed intake by 10% (M) and 20% (L) compared to the ad libitum fed group (H). Proportional to the reduction of feed intake the nutrient density of diets M and L was increased. Thus, the DE intake was reduced while the nutrient intake remained stable among the 3 dietary groups. Effect of genetic group on carcass traits and on meat quality parameters was less marked than DE restriction. Selection for high body fat content improved the dressing out percentage (58.3 and 57.3% in HFAT and LFAT respectively; P < 0.01), and reduced the percentage of the full gastrointestinal tract to slaughter weight (13.7 and 14.3% in HFAT and LFAT respectively; P < 0.05). Reduction of DE intake decreased the body weight and the weight of the chilled and reference carcass (P < 0.001), while it had no effect on dressing out percentage. In group H the percentage of fore part to reference carcass was higher (29.7 vs 29.0%; P < 0.01) while that of the hind part was lower than in group L (37.0 vs 38.2%; P < 0.001). The Longissimus lumborum meat from HFAT rabbits exhibited a lower content of moisture (76.1 and 76.5% in HFAT and LFAT respectively; P < 0.01) as well as lower L (53.5 and 54.4 in HFAT and LFAT respectively; P < 0.05) and b (0.42 and 0.81 in HFAT and LFAT respectively; P < 0.05) colour values. When compared with ad libitum fed rabbits, those restricted at 20% (L) exhibited a higher content of moisture (76.9 vs 75.8%; P < 0.01), as well as higher pHu (5.87 vs 5.73; P < 0.01) associated with lower cooking loss (17.8 vs 19.1%; P < 0.05). L rabbits also produced lower values of redness (a, 2.21 vs 3.35; P < 0.01) and yellowness (b, 0.23 vs 0.91; P < 0.01). No effect of the divergent selection for body fat content on the total lipid percentage and on most of the fatty acids in the meat of the hind leg was found. DE restriction resulted in lower SFA (L: 32.1 vs H: 35.8%; P < 0.01) and MUFA (L: 21.0 vs H: 28.6; P < 0.01) but higher PUFA contents (L: 45.5 vs H: 34.9%; P < 0.01) in the hind leg meat.  相似文献

Background

C‐reactive protein (CRP) is a major acute‐phase protein in dogs. Serum concentrations are low in healthy animals, but increase rapidly after inflammatory stimuli.

Objective

The aim of the study was to investigate CRP concentrations in various respiratory diseases of dogs and to determine if CRP can be used as a biomarker in the diagnosis of bacterial respiratory diseases.

Animals

A total of 106 privately owned dogs with respiratory diseases (17 with bacterial tracheobronchitis [BTB], 20 with chronic bronchitis [CB], 20 with eosinophilic bronchopneumopathy [EBP], 12 with canine idiopathic pulmonary fibrosis [CIPF], 15 with cardiogenic pulmonary edema [CPE], and 22 with bacterial pneumonia [BP]) and 72 healthy controls.

Methods

The study was conducted as a prospective cross‐sectional observational study. CRP was measured in serum samples. Diagnosis was confirmed by clinical and laboratory findings, diagnostic imaging, and selected diagnostic methods such as cytological and microbiological analysis of respiratory samples, echocardiography, and histopathology.

Results

Dogs with BP had significantly higher CRP concentrations (median, 121 mg/L; interquartile range, 68–178 mg/L) than dogs with BTB (23, 15–38, P = .0003), CB (13, 8–14, P < .0001), EBP (5, 5–15, P < .0001), CIPF (17, 10–20, P < .0001), or CPE (19, 13–32, P < .0001) and healthy controls (14, 8–20, P < .0001). Dogs with BTB had significantly higher CRP concentrations than dogs with CB (P = .001) or EBP (P < .0001) and healthy controls (P = .029).

Conclusion and Clinical Importance

These results indicate that CRP has potential for use as an additional biomarker, especially in the diagnostics of BP.  相似文献

Background

Long‐duration beta‐lactam antibiotics are used for empirical treatment in female dogs with uncomplicated bacterial cystitis. However, women with bacterial cystitis are treated with short‐duration potentiated sulfonamides because longer courses of beta‐lactams result in lower cure and higher recurrence rates.

Hypothesis/Objectives

Short‐duration potentiated sulfonamide treatment is more efficacious than long‐duration beta‐lactam treatment in achieving clinical and microbiological cures in female dogs with uncomplicated bacterial cystitis.

Animals

Thirty‐eight client‐owned female dogs.

Methods

Randomized, double‐blinded, placebo‐controlled clinical trial. Dogs were treated with TMPSMX (15 mg/kg PO q12h for 3 days followed by a placebo capsule PO q12h for 7 days; Group SDS; n = 20) or cephalexin (20 mg/kg PO q12h for 10 days; Group LDBL; n = 18). Dogs were monitored for clinical and microbiological cure during treatment and at short‐ and long‐term follow‐up.

Results

No statistically significant differences were found between treatment groups in clinical cure rates after 3 days of treatment (89% SDS, 94% LDBL; P = 1.00) and 4 days (85% SDS, 72% LDBL; P = .44) or >30 days (50% SDS, 65% LDBL; P = .50) after conclusion of treatment or in microbiological cure rates 4 days (59% SDS, 36% LDBL; P = .44) or >30 days (44% SDS, 20% LDBL; P = .40) after conclusion of treatment.

Conclusions and Clinical Importance

We did not identify a difference in cure rates between short‐duration sulfonamide and long‐duration beta‐lactam treatments in female dogs with uncomplicated cystitis. Long‐term cure rates in both treatment groups were low. In some female dogs, “uncomplicated” bacterial cystitis may be more complicated than previously recognized.  相似文献

To assess the relative bioavailability of bis-glycinate bound Cu, 90 Angus-cross steers (265 ± 21 kg) were blocked by body weight (BW) to pens with GrowSafe bunks and randomly assigned to dietary treatments (14 to 18 steers/treatment): 0 mg supplemental Cu/kg dry matter (DM; CON), 5 or 10 mg supplemental Cu/kg DM as Cu sulfate (CS5; CS10) or bis-glycinate bound Cu (GLY5; GLY10). Steers received a high antagonist growing diet (analyzed 4.9 mg Cu/kg DM, 0.48% S, and 5.3 mg Mo/kg DM). Steers were weighed at the beginning (days 1 and 0) and end (days 125 and 126) of the trial to determine average daily gain (ADG) and gain:feed (G:F). Blood was collected from all steers on days 0, 28, 56, 84, and 126. Liver samples were collected on days −3 or −2 and day 123 or 124. Data were analyzed using ProcMixed of SAS (experimental unit = steer; fixed effect = treatment and block). Plasma Cu was analyzed as repeated measures (repeated effect = day). Plasma and liver Cu concentrations were regressed against total Cu intake using ProcGLM to calculate relative bioavailability of GLY. Final BW and overall ADG were greatest for CS5 and CS10 and least for CON and GLY5 (P = 0.01). Overall, DMI was not affected by treatment (P = 0.14), but overall G:F tended to be greatest for CS5, CS10, and GLY5 and least for CON (P = 0.08). Total and supplemental Cu intake was greatest for steers supplemented either source at 10 mg Cu/kg DM and least for CON (P < 0.01). However, total and supplemental Cu intake was greater for CS5 than GLY5 (P < 0.01). Final liver Cu concentrations were greatest for CS10, least for CON, CS5, and CS10, and intermediate for GLY10 (P < 0.01). Final plasma Cu was greatest for steers supplemented either source at 10 mg Cu/kg DM (P < 0.01). Relative bioavailability of GLY was 82% compared to CS based on liver Cu (P < 0.01) but did not differ based on plasma Cu (P = 0.60). The lesser bioavailability of GLY relative to CS could be due to a high concentration of dietary antagonists and lower solubility of GLY (68.9% relative to CS) in pH conditions (5.2) similar to the ruminal pH of a beef animal consuming a high concentrate diet. Future studies should examine the effects of bis-glycinate bound Cu fed in blended combination with inorganic Cu sulfate to determine the most effective blend of sources for feedlot cattle experiencing varying amounts of dietary Cu antagonists.  相似文献

Sixty-nine entire male pigs with different halothane genotype (homozygous halothane positive — nn-, n = 36; and homozygous halothane negative — NN-, n = 33) were fed with a supplementation of magnesium sulphate (Mg) and/or l-tryptophan (Trp) in the diet for 5 days before slaughter. Animals were housed individually and were submitted to stressful ante mortem conditions (mixed in the lorry according to treatments and transported 1 h on rough roads). Individual feed intake was recorded during the 5-d treatment. At the abattoir, pig behaviour was assessed in the raceway to the stunning system and during the stunning period by exposure to CO2. Muscle pH, colour, water holding capacity, texture and cathepsin activities were determined to assess meat quality. The number of pigs with an individual feed intake lower than 2 kg/day was significantly different among diets (P < 0.05; Control: 8.7%; Mg&Trp: 43.5%; Trp: 17.4%) and they were considered to have inadequate supplement intake. During the ante mortem period, 15.2% of pigs included in the experiment died, and this percentage decreased to 8.7% in those pigs with a feed intake > 2 kg/day, all of them from the stress-sensitive pigs (nn). In general, no differences were observed in the behaviour of pigs along the corridor leading to the stunning system and inside the CO2 stunning system. During the stunning procedure, Trp diet showed shorter periods of muscular excitation than control and Mg&Trp diets. The combination of a stressful ante mortem treatment and Mg&Trp supplementation led to carcasses with high incidence of severe skin lesions. Different meat quality results were found when considering all pigs or considering only those with adequate supplement intake. In this later case, Trp increased pH45 (6.15) vs Control diet (5.96) in the Longissimus thoracis (LT) muscle (P < 0.05) and pH at 24 h (Trp: 5.59 vs C: 5.47) led to a higher incidence of dark, firm and exudative (DFD) traits in SM muscle (P < 0.05). Genotype affected negatively all the meat quality traits. Seventy-five percent of LT and 60.0% of the SM muscles from nn pigs were classified as pale, soft and exudative (PSE), while none of the NN pigs showed these traits (P < 0.0001). No significant differences were found between genotypes on the incidence of DFD meat.Due to the negative effects observed in the Mg&Trp group in feed intake and carcass quality, the utilization of a mixture of magnesium sulphate and tryptophan is not recommended.  相似文献

The exposure of pigs to heat stress (HS) appears to damage their intestinal epithelia, affecting the absorption of amino acids (AAs). Arg is involved in the restoration of intestinal epithelial cells but HS reduces Arg intake. The effect of dietary supplementation with Arg on the morphology of intestinal epithelia, AA transporter gene expression, and serum concentration (SC) of free AAs in HS pigs was analyzed. Twenty pigs (25.3 ± 2.4 kg body weight) were randomly assigned to two dietary treatments: Control (0.81% Arg), wheat–soybean meal diet supplemented with l-Lys, l-Thr, dl-Met, and l-Trp, and the experimental diet where 0.16% free l-Arg was supplemented to a similar Control diet (+Arg). All pigs were individually housed and exposed to HS, fed ad libitum with full access to water. The ambient temperature (AT), recorded at 15-min intervals during the 21-d trial, ranged on average from 29.6 to 39.4 °C within the same day. Blood samples were collected on day 18 at 1600 hours (AT peak); serum was separated by centrifugation. At the end of the trial, five pigs per treatment were sacrificed to collect samples of mucosa scratched from each small intestine segment. The expression of AA transporters in intestinal mucosa and the SC of AAs were analyzed. Villi height (VH) was higher (P < 0.01) in the duodenum, jejunum, and ileum but the crypt depth did not differ between the Control and the +Arg pigs. Supplementation of l-Arg increased the mRNA coding for the synthesis of the cationic AA transporter b0,+ (P < 0.01) and the neutral AA transporter B0 (P < 0.05) in the duodenum by approximately 5-fold and 3-fold, respectively, but no effect on mRNA abundance was observed in the jejunum and ileum. The supplementation of l-Arg increased serum Arg, His, Met, Thr, Trp, and urea (P < 0.05) and also tended to increase Val (P < 0.10) but did not affect Ile, Lys, Leu, and Phe. These results indicate that supplementing 0.16% l-Arg to the Control diet may help to improve the function of the small intestine epithelium, by increasing the VH, the abundance of AA transporters, and the SC of most indispensable AAs in pigs exposed to HS conditions. However, the lack of effect of supplemental Arg on both Lys SC and weight gain of pigs suggests that increasing the Lys content in the +Arg diet might be needed to improve the performance of HS pigs.  相似文献

Sixty crossbred (Iberian dam × Duroc sire) females, 80 days of age (17.6 ± 0.13 kg body weight, BW), was used to investigate the effect of castration on productive performance, carcass and meat quality and fatty acid profile of backfat (BF). There were 2 treatments (intact females, IF; castrated females, CF) and 5 replicates of 6 pigs per treatment. Pigs were reared indoor under an intensive production system, ovariectomized at 92 days of age (26.1 ± 0.19 kg BW) and slaughtered at 267 days of age (143.6 ± 6.49 kg BW). Meat samples were taken at longissimus dorsi muscle at the level of the last rib and BF samples were taken at the tail insertion. For the entire experiment (18 to 144 kg BW), IF ate less feed and were more efficient than CF (P < 0.05). Also, IF had less carcass yield (P < 0.01) and fat thickness at the gluteus medius muscle (P < 0.05) and tended to have lower backfat depth (P < 0.10) than CF. However, IF had higher shoulder yield at 2 and at 24 h post mortem (P < 0.05) and after trimmed (P < 0.10) than CF. The pH24 of the semimembranosus muscle tended to be lower for IF than for CF. Also, IF had more moisture (710 vs. 691 g/kg) and less fat (66.4 vs. 91.2 g/kg) in the longissimus dorsi muscle than CF (P < 0.05). Meat from IF was more lightness (higher L value; P < 0.01), redder (higher a value; P < 0.001) and had more intensive color (higher c value; P < 0.001) than meat from CF. Backfat was more saturated in CF than in IF (P < 0.05), mostly because of the higher palmitic acid (P < 0.05) and the lower linolenic acid (P < 0.05) content. We conclude that intact females have better productive performance and shoulder yield but less carcass yield than castrated females and that castration does not improve meat quality. Therefore, when animal welfare, cost of castration, productive performance and carcass and meat quality traits are considered, the use of intact females rather than castrated females is recommended for the production of Iberian pigs reared under intensive management systems.  相似文献

Background

Immune‐mediated polyarthopathy (IMPA) is common in dogs, and is monitored by serial arthrocenteses.

Hypothesis/Objectives

Plasma C‐reactive protein (CRP), interleukin‐6 (IL‐6), and CXCL8 (interleukin‐8) would serve as noninvasive markers of joint inflammation in IMPA.

Animals

Nine client‐owned dogs with idiopathic IMPA; 6 healthy controls.

Methods

Prospective study. Plasma CRP, IL‐6, and CXCL8 were measured by ELISA at baseline, 2, and 4 weeks during treatment with prednisone at 50 mg/m2/day. Arthrocenteses, the canine brief pain inventory (CBPI), and accelerometry collars were used to assess joint inflammation, lameness, and mobility at all 3 time points.

Results

C‐reactive protein concentrations were higher in IMPA dogs (median 91.1 μg/mL, range 76.7–195.0) compared with controls (median <6.3 μg/mL, <6.3–13.7; P = .0035), and were significantly lower at week 2 (10.6 μg/mL, <6.3–48.8) and week 4 (<6.3 μg/mL, <6.3–24.4; P < .001).C‐reactive protein was correlated with median CBPI scores (r = 0.68; P = .0004), joint cellularity (r = 0.49, P = .011), and mobility by accelerometry (r = −0.42, P = .048). Plasma IL‐6 concentrations were also higher in IMPA dogs (median 45.9 pg/mL), compared with controls (median <15.7 pg/mL; P = .0008). IL‐6 was lower in IMPA dogs by week 4 (<15.7 pg/mL; P = .0099), and was modestly correlated with CBPI scores (r = 0.47, P = .023). CXCL8 did not differ significantly between IMPA and healthy dogs.

Conclusions

Plasma CRP and IL‐6 might be useful surrogate markers of synovial inflammation and disease activity in dogs with IMPA.  相似文献

Background

Cyclooxygenase‐2 (COX‐2) is a key enzyme in the synthesis of pro‐inflammatory prostaglandins and 5‐lipoxygenase (5‐LO) is the major source of leukotrienes. Their role in IBD has been demonstrated in humans and animal models, but not in dogs with chronic enteropathies (CCE).

Hypothesis

COX‐2 and 5‐LO are upregulated in dogs with CCE.

Animals

Fifteen healthy control dogs (HCD), 10 dogs with inflammatory bowel disease (IBD), and 15 dogs with food‐responsive diarrhea (FRD).

Methods

Prospective study. mRNA expression of COX‐2, 5‐LO, IL‐1b, IL‐4, IL‐6, TNF, IL‐10 and TFG‐β was evaluated by quantitative real‐time RT‐PCR in duodenal and colonic biopsies before and after treatment.

Results

COX‐2 expression in the colon was significantly higher in IBD and FRD before and after treatment (all P < .01). IL‐1b was higher in FRD in the duodenum after treatment (P = .021). TGF‐β expression was significantly higher in the duodenum of HCD compared to FRD/IBD before treatment (both P < .001) and IBD after treatment (P = .012). There were no significant differences among groups and within groups before and after treatment for IL‐4, IL‐6, TNF, and IL‐10. There was a significant correlation between COX‐2 and IL‐1b in duodenum and colon before treatment in FRD and IBD, whereas 5‐LO correlated better with IL‐6 and TNF. IL‐10 and TGF‐β usually were correlated.

Conclusions and Clinical Importance

COX‐2 is upregulated in IBD and FRD, whereas IL‐1b and TGF‐β seem to be important pro‐ and anti‐inflammatory cytokines, respectively. The use of dual COX/5‐LO inhibitors could be an interesting alternative in the treatment of CCE.  相似文献

Two 6 × 6 Latin-square designed experiments examined the glycemic responses to 10 common equine feeds in six Quarter Horse mares. At 8:00 am of the sampling days, horses were offered a meal of 1 of 10 common feeds. The amount of feed offered was calculated on an isocaloric basis at 4 Mcal digestible energy per meal. Jugular blood samples were collected for 300 minutes after feeding, and plasma was analyzed for glucose concentration. The results showed that plasma glucose values peaked approximately 90 to 120 minutes after eating, whereas glycemic index (GI) values ranged from 7 to 129 based on a standard of oats set at a GI of 100. The four feeds with the highest GI values (sweet feed, corn, jockey oats, and oats) were greater (P < .05) than the four feeds with the lowest GI values (beet pulp, alfalfa, rice bran, and soy hulls). Barley and wheat bran had intermediate GI (55–69) values, but were not significantly different from either the high or low group. Whereas large variations in the data were found, conservative utilization of the ranking or categories of GI values of the 10 common horse feeds examined may be useful for formulating rations with different glycemic objectives.  相似文献

Background

In EMS‐associated laminitis, laminar failure may occur in response to energy failure related to insulin resistance (IR) or to the effect of hyperinsulinemia on laminar tissue. 5′‐Adenosine‐monophosphate‐activated protein kinase (AMPK) is a marker of tissue energy deprivation, which may occur in IR.

Hypothesis/Objectives

To characterize tissue AMPK regulation in ponies subjected to a dietary carbohydrate (CHO) challenge.

Animals

Twenty‐two mixed‐breed ponies.

Methods

Immunohistochemistry and immunoblotting for total AMPK and phospho(P)‐AMPK and RT‐qPCR for AMPK‐responsive genes were performed on laminar, liver, and skeletal muscle samples collected after a 7‐day feeding protocol in which ponies stratified on body condition score (BCS; obese or lean) were fed either a low‐CHO diet (ESC + starch, approximately 7% DM; n = 5 obese, 5 lean) or a high‐CHO diet (ESC + starch, approximately 42% DM; n = 6 obese, 6 lean).

Results

5′‐Adenosine‐monophosphate‐activated protein kinase was immunolocalized to laminar keratinocytes, dermal constituents, and hepatocytes. A high‐CHO diet resulted in significantly decreased laminar [P‐AMPK] in lean ponies (P = .03), but no changes in skeletal muscle (lean, P = .33; obese, P = .43) or liver (lean, P = .84; obese, P = .13) [P‐AMPK]. An inverse correlation existed between [blood glucose] and laminar [P‐AMPK] in obese ponies on a high‐CHO diet.

Conclusions and Clinical Importance

Laminar tissue exhibited a normal response to a high‐CHO diet (decreased [P‐AMPK]), whereas this response was not observed in liver and skeletal muscle in both lean (skeletal muscle, P = .33; liver, P = .84) and obese (skeletal muscle, P = .43; liver, P = .13) ponies.  相似文献

The objective of this experiment was to examine the effects of feeding a vitamin A deficient diet (VAF) to pigs at different times on day 100 of foetal and days 0, 1 and 2 of neonatal development. Three treatments included a control (n = 12), VAF for 100 days before mating and during the first month of pregnancy (n = 13; VAF–control), and VAF during the oestrous cycle before mating and throughout pregnancy (n = 13; control–VAF). On day 100 of pregnancy, maternal liver and plasma retinol concentrations were reduced in both groups of gilts fed a vitamin A free diet compared to controls (P < 0.001 and P < 0.01, respectively). Day 100 foetal liver retinol concentrations were not affected by dietary treatment, whereas foetal plasma concentrations were higher in foetuses carried by gilts fed the VAF–control diet (P < 0.05). Piglets born to mothers fed the control–VAF, but not the VAF–control diet had consistently lower hepatic and plasma liver retinol concentrations (P < 0.05). Moderate reductions in maternal vitamin A at either stage of pregnancy did not affect pregnancy rate, litter size, progesterone secretion or the allometric relationships between foetal or neonatal organ and total body size. Reduced vitamin A during conception and early pregnancy, but not during later pregnancy, was associated with increased within-litter uniformity in birth weight (P < 0.05) and a tendency for fewer low birth weight piglets, but this needs to be confirmed in a greater number of sows. The mechanism underlying this effect is not known, but appears to not involve an alteration in progesterone production.  相似文献

The experiment was conducted at Dilla Agricultural Technical Vocational Education and Training (ATVET) College in Ethiopia using twenty-four yearling intact male Sidama goats with a mean body weight (BW) of 16.8 ± 1.14 kg (mean ± SD). The objective of the experiment was to examine the effect of different levels of cottonseed meal (CSM) supplementation on the feed intake, digestibility, BW gain, and carcass parameters of Sidama goats. The experiment consisted of 90 days of feeding trial and 10 days of digestibility trial followed by evaluation of carcass parameters at the end of the experiment. The treatments included ad libitum feeding of hay (T1, control) and daily supplementation of CSM at 200 g (T2), 300 g (T3) and 400 g (T4) per head on dry matter (DM) basis. The experimental design was a completely randomized block design. The experimental goats were blocked on initial BW and six goats were randomly assigned to each treatment. Hay DM, crude protein (CP) and neutral detergent fiber (NDF) intake were higher (P < 0.001) in the non-supplemented than in the supplemented goats, however, the contrary was true for total CP intake. Total DM intake was higher (P < 0.001) for goats supplemented with the high level of CSM than those on the control treatment. Among CSM supplemented goats, the intake of CSM DM, CP, NDF (P < 0.001) and total CP (P < 0.01) were higher in the order of T4 > T3 > T2. The high level of supplementation with CSM resulted in better (P < 0.05) apparent digestibility coefficient (DC) of DM and organic matter (OM) compared to the non-supplemented goats. The apparent DC of CP was higher (P < 0.01) for supplemented than non-supplemented goats. The non-supplemented goats had lower daily BW gain, empty BW, dressing percentage on slaughter weight base, rib-eye muscle area (P < 0.01), BW change (P < 0.001), final BW (P < 0.05) and higher feed conversion ratio (P < 0.01) than the supplemented ones. The medium level of supplementation resulted in higher daily BW gain (P < 0.01), BW change (P < 0.001), final BW (P < 0.05) and lower feed conversion ratio (P < 0.01) than the lower level of supplementation, but with no differences (P > 0.05) in these parameters with the high level of supplementation. Therefore, it is concluded that supplementation of CSM at 300 g DM per head per day resulted in better animal performance parameters in Sidama goats.  相似文献

Calcium (Ca) requirement increase rapidly by pregnant ewes at late gestation as a result of high fetal growth. Furthermore, Ca deficiency results in poor growth and bone development in growing newborn lambs. The aim of this study was to evaluate the effect of calcium supplementation (Calcium carbonate–CC) and vitamin AD3E injection (75,000 IU/shot) to pregnant Awassi ewes at late gestation (− 45 days prepartum) on their mineral levels and performance of their newborn lambs. Forty pregnant Awassi ewes at late gestation (65 ± 6.1 kg) were selected before parturition (− 45 days; using hormonal sponges for estrus synchronization) and randomly divided to 4 dietary treatments. The treatments were: group 1 Control (1.4% CC); group 2 (2.4% CC); group 3 (1.4% CC + 2 shots vitamin AD3E at − 45 and − 21 days prepartum); group 4 (2.4% CC + 2 shots vitamin AD3E at − 45 and − 21 days prepartum). Blood samples were collected from ewes (− 45, 0 days prepartum: 30 and 60 days postpartum) and lambs (0, 30 and 60 days old). Average milk yield and samples were collected. Body weights of lambs were taken at birth, 30 and 60 days old. Blood serum and milk samples were prepared and analyzed by AAS for Ca, Mg, Co, Mn and Cu concentrations.Calcium concentrations in bloods serum of ewes, 0 and 60 days postpartum, and lambs at weaning (60 days old) from group 4 were significantly (P < 0.05) higher compared to other groups. No significant effect (P > 0.05) of treatment on Co, Mg and Cu in blood serum of ewes and lambs except for Cu concentration in ewes. Moreover, a significant increase (P < 0.01) in milk inorganic% (30 and 60 days samples) in ewes from group 2 when compared with other groups. The average milk yield (AMY) of ewes from group 2 and 4 at late stage of lactation (45–60 days) were significantly higher (P < 0.05) compared to groups 1 and 3. In addition, Ca% in colostrums samples from ewes in groups 2 and 4 were significantly higher (P < 0.01). The average daily gain (ADG) and total body gain for lambs up to weaning (60 days old) were significantly higher (P < 0.05) in groups 1 and 2 compared with the other groups.In conclusion, increasing level of Ca intake in feed of Awassi ewes at late gestation, with or without vitamin AD3E injections, improve the calcium concentration in colostrums and blood serum of ewes and their lambs without negative effect in their health and performance.  相似文献
