The objective of the study has been to verify the hypothesis that the coat colour is regarded in the selection of Polish Thoroughbred horse population. Formally, the colour is not a selection criterion in this breed selected mainly for speed. The material consisted of twelve groups of foals registered in successive volumes of the Stud Book (11,688 foals, in total) and their parents selected to the breeding stud. The frequency of alleles in ASIP, MC1R and GREY loci controlling the coat colours was estimated from the recessive phenotype frequency square in the groups of foals. The inflow of foreign genes was limited and the population great, hence the migration effect was very low. The drift and Wahlund effect hardly influenced the genetic structure in the groups which enabled to analyze the population not divided. The total offspring frequency of recessive a, e and g alleles amounted to 0.1552, 0.4877 and 0.9773, respectively. Accuracy of the assessment of the a and e frequency was confirmed on the basis of test matings. The a, e and g alleles were more frequent in dams than in sires and the a alleles occurred more often in fillies than in colts. The frequency of a and e alleles was higher in the offspring than in the parents. The genotype distribution in the offspring differed from the expected one, assessed from the gamete frequency in sires and in dams. Fewer bay foals were born than anticipated. All the results show that the coat colour is not entirely disregarded in the breeding of Thoroughbred horses. The dominant A and E alleles producing the colour are preferred in the selection, particularly in the sires. This leads to some alterations in the phenotypic structure of the population. On the other hand, the horses are mated randomly, irrespective of the coat colour.  相似文献

The effect of used pine-shaving litter from broiler floor pens on Salmonella colonization resistance was evaluated in broiler chicks. One-day-old chicks were placed in floor pens on fresh unused litter or on used litter. All chicks were challenged orally with 10(4) S. typhimurium at 3 days of age. The study was replicated in three trials with used litter that was collected and stored for 1 day (Trial 1), 4 days (Trial 2), or 50 days (Trial 3) before the start of each trial. Cecal concentrations of volatile fatty acids (VFAs) were significantly higher (P < 0.05) in chicks placed on used litter than in chicks on new litter. In all three trials, the number of Salmonella in the cecal contents and the number of Salmonella cecal-culture-positive chicks was significantly lower (P < 0.01) at 10 days and 20 days of age in the chicks on used litter than in the chicks on new litter. The results indicated that newly hatched chicks reared on used litter had higher cecal VFA concentrations and higher resistance to Salmonella colonization than chicks reared on new litter.  相似文献

The aim of this study was to estimate the current level of inbreeding in the German cow population and for bull dams born in Germany, to find out sires most related to different subsets of their breed and to demonstrate the negative effect of homozygosity in the case of complex vertebral malformation (CVM). Further on, the application of optimum genetic contribution (OGC) theory for the selection of bull dams and bull sires in different breeding scenarios was investigated. Levels of inbreeding for the cow population were in a low range from 0.97% to 1.70% evaluating birth years from 1996 to 1999 in a total dataset of 244,427 registered Holstein cows. The inbreeding coefficient of 8030 bull dams was much higher, i.e. 3.71%, for the birth year 1999. Increases in inbreeding of 0.19% per year indicated an effective population size of only 52 animals. Individual sires like R.O.R.A. Elevation and Hannoverhill Starbuck were highly related to potential bull dams with coefficients of relationship of 13.4% and 12.9%, respectively, whereas P.F. Arlinda Chief (16.3%) and Carlin-M Ivanhoe Bell (16.1%) were highest related to the best available AI sires. Coefficients of relationship were calculated by classes of estimated breeding values (EBV) for production traits showing highest values above 7% in the two highest EBV-classes. The optimum genetic contribution theory using official EBVs and approximative, for zero inbreeding corrected EBVs, was applied for elite matings in a breeding program embracing 30 young bulls per year to find the optimal allocations of bull sires and bull dams. Compared with the actual breeding program applied in practice, OGC-theory has the potential to increase genetic gain under the same constraint for the increase of average relationship by 13.1%. A more relaxed constraint on increase in inbreeding allowed even higher expected genetic gain whereas a more severe constraint resulted in more equal contributions of selected bull sires. Contributions from 21 selected bull sires and 30 selected bull dams for a scenario at 5% constrained relationship were used to develop a specific mating plan to minimise inbreeding in the short term in the following generation applying a simulated annealing algorithm. The expected coefficient of inbreeding of progeny was 66.3% less then the one resulting from random mating. Mating programs can address inbreeding concerns on the farm, at least in the short term, but long-term control of inbreeding in a dairy population requires consideration of relationships between young bulls entering AI progeny test programs. Significantly better EBVs of CVM-free bulls compared with CVM-carriers for the paternal fertility justify the application of OGC for elite matings.  相似文献

Genetic resistance to Salmonella infection in experimental animal models is well described. However, genetic resistance in domestic animals, which has potentially great value in terms of controlling Salmonella in the food chain, has been relatively poorly described. Recent advances in genetics and immunology have identified several factors that influence resistance in chickens and pigs in particular. Resistance to systemic salmonellosis in the chicken is encoded by a number of factors including Nramp1 (now termed Slc11a1) and a novel gene, SAL1 that leads to increased macrophage activity against Salmonella. Studies in outbred, and in particular, inbred chickens have revealed considerable differences in levels of colonization of the gastrointestinal tract and responses to vaccination. Factors influencing this appear to include innate immune function, MHC and Nramp. In pigs several immune factors, including polymorphonuclear cell activity, have been shown to influence resistance.  相似文献

Selection intensity and generation interval were evaluated in a Hereford cattle herd made up of 14 inbred lines and 14 linecross groups corresponding to the lines of inbred sires at the Suan Juan Basin Research Center, Hesperus, Colorado. Selection indexes practiced were calculated in retrospect. Analyses of the records collected from 1946 through 1973 involved weaning weight (WW) and postweaning traits in males and females. Analyses by line were performed for the inbreds, while pooled analyses were done on the inbred and linecross populations. From records of 1,239 calves weaned, age of sire averaged 3.75 yr compared with 4.52 yr for age of dam, showing faster generation turnover for sires than for dams. Generation interval determined as actual age of midparent was 4.13 yr. Selection applied for WW, evaluated as annual selection differentials within inbred lines and then pooled over all lines, averaged .55 standard deviations (sigma)/generation for sires. For females, selection was much less intense. Midparent selection differential amounted to .33 sigma/generation. For sires, pooled standardized selection differentials per generation over all lines during the postweaning gain period were .49 sigma, .46 sigma, .40 sigma, -.20 sigma, -.10 sigma and .69 sigma, respectively, for initial weight, final weight, feed consumed, feed efficiency (FE, unadjusted and adjusted) and average daily gain (ADG). Selection of females for postweaning traits was not intense. Selection index actually practiced in retrospect for sires was: IS = .4461 (WW) - .0092 (FE) + .6126 (ADG). The indexes for dams included WW, 12-mo weight (12W), 18-mo weight (18W), mature spring weight (SPW) and mature fall weight (FAW) and were: for inbred dams, ID = .1824 (WW) - .0284 (12W) + .0736 (18W) - .1097 (SPW) - .1097 (FAW); for linecross dams, ID = .2693 (WW) - .2960 (12W) + .0147 (18W) + .1185 (SPW) - .0354 (FAW). The corresponding index selection differentials were .818, .203 and .209. Sire index selection differentials represent about 79% of the total selection differentials.  相似文献

The effect of oral inoculation with anaerobic cultures of cecal microflora and providing lactose in the feed on colonization resistance to invasive Salmonella enteritidis was evaluated in newly hatched leghorn chicks. Salmonella colonization of the ceca, tissue invasion and organ colonization, horizontal transmission, and seroconversion were significantly decreased (P less than 0.01) in chicks inoculated with cecal flora. The addition of lactose to the feed, in the absence of cecal microflora, failed to provide protection. Dietary lactose enhanced colonization resistance in chicks that were inoculated with anaerobic cultures of cecal flora. The results indicated that establishment of normal cecal flora in layer chicks together with the addition of lactose to the diet markedly increases resistance to cecal colonization and organ invasion, and decreases horizontal transmission of S. enteritidis.  相似文献

A study was conducted to characterize genetic diversity in the Alentejana breed of cattle based on its demographic trends and to investigate the major factors affecting genetic erosion in this breed. Herdbook information collected between 1940 and 2004, including pedigree records on 100,562 animals in 155 herds, was used to estimate demographic parameters. The mean generation intervals were 6.0 +/- 2.4 yr and 6.8 +/- 3.2 yr for sires and dams of calves, respectively. Average inbreeding increased steadily over the period analyzed, with an annual rate of inbreeding of 0.33 +/- 0.004% (P < 0.01) and an effective population size of 23.3. In the reference population (28,531 calves born between 2000 and 2003) the average inbreeding was 8.35 +/- 9.02% and nearly 80% of the calves were inbred, whereas the average relationship among all animals was 0.026 +/- 0.040. Nevertheless, the mean relationship was 0.328 +/- 0.264 and 0.022 +/- 0.026 for animals born in the same and in different herds, respectively. The computed genetic contributions to the reference population resulted in estimates for the effective number of founders, ancestors, founding herds, and herds supplying sires of 121.6, 55.0, 17.1, and 26.9, respectively, the 2 most influential herds and ancestors contributing 24.2 and 15.1%, respectively, of the current genetic pool. Of the 671 founding sires, only 24 Y-chromosomes are currently represented, but 1 sire alone contributes nearly 60% of this representation, such that the effective number of Y-chromosomes is only 2.73. The observed inbreeding per herd was, on average, 0.053 +/- 0.071 lower than expected from the relationship among the generation of parents of calves in the reference population, indicating that producers have followed breeding strategies that have kept inbreeding at lower levels than anticipated with random selection and mating. When compared with other cattle breeds, Alentejana has some of the highest levels of mean inbreeding and annual rate of inbreeding, and an effective population size that is nearly half of the minimum recommended for maintenance of genetic variability. These critical indicators demonstrate the need to adopt strategies aimed at minimizing inbreeding to avoid further losses of genetic diversity.  相似文献

Records for pigs included in an experiment on reciprocal recurrent selection conducted from 1956 through 1971 at the USDA Beltsville Agriculture Research Center were analyzed to obtain estimates of heritabilities and genetic correlations and to derive prediction equations for estimating weight of lean cuts (WTLC) and percentage of lean cuts of shrunk slaughter weight (LCPC). Lean cuts growth rate (LCGR) was then estimated as WTLC/age of pig at slaughter. The base population consisted of two unrelated crossbred strains. A total of 1,294 records of F1 and F2 crossbred pigs were analyzed with one barrow and one gilt from each litter. Estimates of heritabilities and genetic correlations were computed with sire components of variance and covariance from a nested analysis of variance with an assumed model of years, strain-lines within years, sire within strain-lines, dams within sires and residual. Degrees of freedom were 307 for sires in strain-lines, 270 for dams in sires and 646 for residual. Heritability (h2) estimates were .42 +/- .13, .41 +/- .13 and .27 +/- .18 for WTLC, LCPC and LCGR, respectively, and .71 +/- .16, .38 +/- .13, .31 +/- .13 and .25 +/- .15 for carcass length, average backfat thickness, longissimus muscle area and ADG in BW, respectively. These estimates were apparently the first published genetic estimates involving LCGR based on carcass data. It was recommended that prediction equations to estimate WTLC, LCPC and LCGR for use in swine testing programs be derived from current meat-type pigs.  相似文献

Salmonella enterica serovar Enteritidis is a gram-negative bacterium that negatively affects human and animal health. Many eukaryotes use antimicrobial peptides (alpha-defensins, beta-defensins, gamma-defensins, and cathelicidins) in innate immune responses to fight bacterial infections. Poultry gallinacins are the functional equivalents of mammalian beta-defensins. Two related advanced intercross lines of chickens were analyzed for association of gallinacin genotypic variation with Salmonella Enteritidis burden levels in the cecum and spleen after infection. Thirteen genes of the chicken beta-defensin cluster (GAL1-13) were sequenced from individuals of each advanced intercross line, plus the founder broiler sire and representatives of the highly inbred Leghorn and Fayoumi founder dam lines. The mean was 17 single-nucleotide polymorphisms (SNPs) per kilobase. One single-nucleotide polymorphism per gene was genotyped with SNaPshot to test for statistical associations with Salmonella Enteritidis colonization after challenge. Among the 13 gallinacin genes evaluated, the single-nucleotide polymorphisms in all genes in a cluster of three adjacent genes (GAL11, GAL12, and GAL13) were associated with bacterial load in the cecal content in the broiler x Leghorn advanced intercross line (three-gene SNP genotype effect, P < 0.008). The results strongly suggest a role of the gallinacins in defense of poultry against enteric pathogens. The use of gallinacin single-nucleotide polymorphisms as molecular markers for genetic selection for Salmonella Enteritidis resistance might result in reduced bacterial burden via development of an enhanced innate immune response.  相似文献

Inclusion of lactose in the diets of chickens has been determined to reduce cecal colonization with Salmonella typhimurium. We hypothesized, therefore, that dietary lactose may be a practical means for reducing the prevalence of Salmonella contamination of chicken products. Because some strains of Salmonella are atypical and ferment lactose, we investigated the effects of dietary lactose on cecal colonization with lactose-fermenting S typhimurium. Broiler chicks were inoculated intracloacally with Lac+ S typhimurium selected for resistance to novobiocin and rifampicin. The chicks also were inoculated orally with certain anaerobes that do not effectively inhibit colonization by S typhimurium, but do appear essential for lactose mediated inhibition of cecal colonization. Control chicks were not given dietary lactose, and chicks in the experimental group were fed a diet containing 7% lactose. Enumeration of Lac+ S typhimurium in cecal contents revealed dietary lactose to be effective at controlling this organism. Control was correlated with changes in cecal pH and increases in undissociated volatile fatty acids, especially propionic acid.  相似文献

The pathogenicity of one isolate of Salmonella typhimurium, four isolates of Salmonella heidelberg, three isolates of Salmonella kentucky, two isolates of Salmonella montevideo, one isolate of Salmonella hadar, and two isolates of Salmonella enteritidis (SE), one belonging to phage type (PT)13a and the other to PT34, was investigated in specific-pathogen-free chicks. Three hundred eighty-four chicks were separated into 16 equal groups of 24 chicks. Thirteen groups were inoculated individually with 0.5 ml of broth culture containing 1 x 10(7) colony-forming units (CFU) of either S. typhimurium (one source), S. heidelberg (four sources), S. montevideo (two sources), S. hadar (one source), S. kentucky (three sources), SE PT 13a (one source) or SE PT 34 (one source) by crop gavage. Two groups of 24 chicks were inoculated in the same way with 1 x 10(7) CFU of SE PT4 (chicken-CA) and Salmonella pullorum. Another group of 24 chicks was kept as an uninoculated control group. The chicks were observed daily for clinical signs and mortality. Isolation of salmonella was done from different organs at 7 and 28 days postinoculation (DPI). All the chicks were weighed individually at 7, 14, 21, and 28 DPI. Two chicks chosen at random from each group were euthanatized and necropsied at 7 and 14 DPI and all the remaining live chickens, at 28 DPI. Selected tissues were taken for histopathology at 7 and 14 DPI. Dead chicks were examined for gross lesions and tissues were collected for histopathology. Chicks inoculated with S. pullorum had the highest mortality (66.66%), followed by S. typhimurium (33.33%). Chicks inoculated with S. heidelberg (00-1105-2) and SE PT4 (chicken-CA) had 12.5% mortality and 8.3% mortality, respectively, with SE PT 13a. Ceca were 100% positive for salmonellae at acute or chronic infection compared with other organs. Mean body weight reduction ranged from 0.67% (inoculated with S. kentucky 00-926-2) to 33.23% (inoculated with S. typhimurium 00-372) in the inoculated groups at different weeks compared with uninoculated controls. Gross and microscopic lesions included peritonitis, perihepatitis, yolk sac infection, typhilitis, pneumonia, and enteritis in some groups, especially those inoculated with S. typhimurium, S. heidelberg (00-1 105-2), SE PT4 (chicken-CA), and S. pullorum.  相似文献

This study was conducted to evaluate the influence of a vancomycin pretreatment on the ability of marker (nalidixic-acid resistant) Salmonella Enteritidis (SE(M)), field Salmonella Enteritidis (SE(E)), and marker Salmonella Typhimurium (ST(M)) strains to colonize within the intestinal and reproductive tracts and translocate to other organs of leghorn laying hens. In each of three trials, caged laying hens (76, 26, and 33 wk ofage) were divided into six groups designated to receive SE(M), SE(F), or ST(M), and half were pretreated with vancomycin (n = 11-12 hens). Vancomycin-treated hens received 10 mg vancomycin in saline/kilogram body weight orally for 5 days to inhibit Gram-positive bacteria within the intestines. On Day 6, all hens were concurrently challenged by oral, intravaginal, and intracolonal routes with Salmonella and placed into separate floor chambers by Salmonella strain. Two weeks postinoculation, all hens were euthanatized and the ceca, spleen, liver/gall bladder (LGB), upper (URT), and lower (LRT) reproductive tracts, and ovarian follicles were aseptically collected, and analyzed for Salmonella. Results did not differ for the three hen's ages and were therefore combined. The vancomycin pretreatment also had no significant effect on the colonization ability of SE(M), SE(F) or ST(M), and therefore results were combined within Salmonella strain. The marker strain of Salmonella Enteritidis was recovered from 21% of ceca, 4% of LGB, 9% of LRT, and 17% of the fecal samples. The field strain of Salmonella Enteritidis was recovered from 88% of ceca, 96% of spleen, 92% of LGB, 30% of LRT, 4% of URT, 13% of follicle, and 42% of the fecal samples. The marker strain of Salmonella Typhimurium was recovered from 100% of ceca, 74% of spleen, 91% of LGB, 30% of LRT, 9% of URT, 9% of follicle, and 100% of the fecal samples. Among ceca, spleen, LGB, and fecal samples, SE(F) and ST(M) colonization was significantly greater than SE(M) colonization. Overall prevalence of Salmonella in the reproductive tracts of challenged hens was relatively low, ranging from 4% to 30%.  相似文献

Feed artificially contaminated with various levels of nalidixic-acid-resistant Salmonella montevideo was fed to newly hatched chicks for 7 days. Cloacal and cecal swabs were obtained from the chicks at 7, 14, and 21 days of age to monitor Salmonella colonization relative to the feed contamination level. In one of three trials, less than one Salmonella montevideo per gram of feed was sufficient to establish colonization in 1-to-7-day-old chicks.  相似文献

One-day-old broiler chicks were divided into four groups: untreated controls and three groups treated with the immunosuppressive drugs cyclophosphamide, cyclosporin, or dexamethasone on days 1-4 of age. On day 5, all groups were challenged orally with 10(4) Salmonella typhimurium. Suppression of immunocompetency was demonstrated in the treatment groups by significant decrease (P less than 0.05) in weights of bursas of Fabricius, and diminished mitogen-induced T and B lymphocyte blastogenesis and cutaneous basophil hypersensitivity responses. There were no significant differences among the four groups in the number of Salmonella in the cecal contents or in the number of cecal-culture-positive chicks on day 10 post-challenge at 15 days of age. Results indicated that impairment of B or T lymphocyte responses had little influence on the resistance of young chicks to Salmonella cecal colonization.  相似文献

The effect of providing lactose in feed and inoculation with volatile fatty acid-producing anaerobic cultures (AC) of cecal flora on Salmonella typhimurium colonization was evaluated in broilers. One-day-old chicks were divided into four groups and provided 1) no lactose, no AC; 2) AC, no lactose; 3) AC and lactose on days 1-10; or 4) AC and lactose on days 1-40. All groups were challenged per os with 10(6) Salmonella on day 3 and with 10(8) Salmonella on day 33. Salmonella growth in the cecal contents was significantly decreased (P less than 0.01) on day 10 in the chicks provided lactose from day 1-10. However, after the removal of lactose from the diet, the chicks were susceptible to Salmonella colonization. The number of Salmonella in the ceca was significantly reduced (P less than 0.05) in the chicks provided lactose throughout the 40-day growing period. Dietary lactose decreased the pH of the cecal contents and was accompanied by marked increases in the concentrations of undissociated bacteriostatic volatile fatty acids in the cecal contents.  相似文献

The aim of this study was to assess the importance of pedigree depth when performing optimal contribution selection as implemented in the software program EVA. This was done by applying optimal genetic contribution in the breeding program of the major Danish Dairy breed Danish Holstein. In the analyses earlier breeding decisions were considered by including all AI waiting- and young bulls and contract matings. Twenty potential sires, 2169 potential dams, 1421 AI-bulls and 754 contract matings plus pedigree animals were included. Results showed that the outcome was very dependent on quality of pedigree, also for information going more than 25 years (5–7 generations) back. The analyses showed that EVA works satisfactorily as a management tool for planning of breeding schemes with respect to contributions of sires of sons at population level in maximising the genetic gain, while controlling the increase in future inbreeding. The more weight put on the average additive genetic relationship in next generation relative to genetic merit, the lower the average merit of the matings, and the lower average additive genetic relationship among the chosen matings and the present breeding animals. Furthermore more weight on average additive genetic relationship gives a more diverse use of sires of sons. Given the potential sires and dams the average additive genetic relationship among the selected matings and the present breeding animals can be reduced from 0.1621 to 0.1495 at the cost of 0.7 genetic S.D. units at the total merit index. This reduction was obtained when selection was only on reduction of average relationship compared to selection only on genetic merit. Optimal genetic contribution selection is a promising tool for managing breeding schemes for populations facing inbreeding problems, such as Danish Holstein and other dairy breeds. However sufficient pedigree information is a necessity.  相似文献

Production traits of a Rhode Island Red layer line were analyzed to detect carrier animals with major genes that influenced a particular trait by applying the major gene index (MGI) approach. This method was used to examine the deviation of predicted breeding values between parents and offspring. Advantages of this method include its simplicity and time‐reducing, cost‐saving benefits compared with any other statistical approach for screening major genes. The layer line had been selected based on the desired gain index for 6 years at the National Livestock Breeding Center, Okazaki station. The line consisted of 125 sires and 2986 dams. Two economic traits as breeding objectives were studied – age at sexual maturity (ASM, days) and egg production efficiency (EP, %). The MGI detected nine sires and 23 dams as carriers with major genes for ASM, and five sires and 26 dams for EP. They were identified as important breeding stock, and pedigree information provided candidates possessing major genes with favorable effect at the founding of the selected layer line. It seems likely that the simple approach of the MGI could be a useful preliminary method for detecting carrier animals with major genes before applying molecular techniques on a sampled population to identify in more detail the existence of major genes and their carriers.  相似文献

From a collection of over 2800 Salmonella enterica subspecies Enterica serotype Typhimurium F98 Tn5-TC1 insertion mutants 14 were identified as expressing growth-non-suppressive phenotype under strict anaerobic conditions. Sequence analysis of regions flanking the Tn insertions revealed that most of the selected mutants were defective in genes contributing to the anaerobic fumarate uptake and generation (insertions in dcuA, dcuB and aspA), or to the anaerobic L-arginine utilisation pathway (insertions in STM4467 encoding a putative arginine deiminase, and in between speF encoding ornithine decarboxylase and kdpE coding a response regulator protein). Mutants defective in flagellum synthesis (flhA) were also identified. In contrast to the in vitro results, all the mutants colonised 1-day-old chicks efficiently and suppressed the super-infection of chicks by the parent strain. This clearly indicates that neither of the metabolic pathways mentioned above nor motility play essential roles in lower intestinal tract colonisation.  相似文献

The estimation of (co)variance components for multiple traits with maternal genetic effects was found to be influenced by population structure. Two traits in a closed breeding herd with random mating were simulated over nine generations. Population structures were simulated on the basis of different proportions of dams not having performance records (0, 0.1, 0.5, 0.8 and 0.9): three genetic correlations (-0.5, 0.0 and +0.5) between direct and maternal effects and three genetic correlations (0, 0.3 and 0.8) between two traits. Three ratios of direct to maternal genetic variances, (1:3, 1:1, 3:1), were also considered. Variance components were estimated by restricted maximum likelihood. The proportion of dams without records had an effect on the SE of direct-maternal covariance estimates when the proportion was 0.8 or 0.9 and the true correlation between direct and maternal effects was negative. The ratio of direct to maternal genetic variances influenced the SE of the (co)variance estimates more than the proportion of dams with missing records. The correlation between two traits did not have an effect on the SE of the estimates. The proportion of dams without records and the correlation between direct and maternal effects had the strongest effects on bias of estimates. The largest biases were obtained when the proportion of dams without records was high, the correlation between direct and maternal effects was positive, and the direct variance was greater than the maternal variance, as would be the situation for most growth traits in livestock. Total bias in all parameter estimates for two traits was large in the same situations. Poor population structure can affect both bias and SE of estimates of the direct-maternal genetic correlation, and can explain some of the large negative estimates often obtained.  相似文献
