Furostanol saponins from Tribulus terrestris   总被引:4,自引:0,他引:4  
An HPLC-ELSD-ESI-MS method has been developed for the analysis of the steroidal saponins in the aerial parts of Tribulus terrestris. Protodioscin, a new saponin (5,6-dihydroprotodioscin, neoprotodioscin) and their respective sulfates were detected. The structure of the new compound was elucidated on the basis of NMR and ESI-MS spectral analysis.  相似文献

采用砂堡培养基液体稀释培养法和薄层色谱分析法研究了洋葱挥发油抗真菌效果及机理,以期发现天然抗真菌药物,开发洋葱资源.结果表明,洋葱挥发油提取物对酿酒酵母、白假丝酵母、新生(?)球菌、黑曲霉、黄曲霉、红曲霉、青霉等真菌的生长有较强的抑制作用;其最低抑菌浓度酿酒酵母和白假丝酵母为0.25%,新生隐球菌为0.50%.薄层色谱分析表明:洋葱挥发油处理后的白假丝酵母的羊毛固醇、麦角固醇的含量降低,而角鲨烯的含量增高,说明洋葱挥发油可能抑制白假丝酵母麦角固醇的早期合成.  相似文献

Continued chemical investigation on the fruits of Aesculus pavia L. resulted in theisolation and identification of two new oleanolic acid saponins, namely vaccaroside A (1) andvaccaroside B (2). The isolated furostanol saponins were evaluated for cytotoxic activity againsthuman normal amniotic and human lung carcinoma cell lines using neutral red and MTT assays.In vitro experiments showed significant cytotoxicity in a dose dependent manner with IC50 valuesin the range of 27.80–79.02 μM.  相似文献

By using an agar dilution assay, the antifungal activity of aqueous extracts prepared from Allium cepa (onion; AOE) and Allium sativum (garlic; AGE) were evaluated against Malassezia furfur (25 strains), Candida albicans (18 strains), other Candida sp. (12 strains) as well as 35 strains of various dermatophyte species and compared with the activity of a known antifungal drug, ketoconazole (KTZ). All the AOE, AGE and KTZ were found to be able to inhibit growth of all fungi tested in a dose-dependent manner with maximum of 100% at defined concentrations. The results indicate that onion and garlic might be promising in treatment of fungal-associated diseases from important pathogenic genera Candida, Malassezia and the dermatophytes.  相似文献

The occurrence of the two new cis-fused A/B rings furostanol saponins (25S)-26-O-β-d-glucopyranosyl-5β-furostan-1β,3β,22α,26-tetraol-1-O-β-d-glucopyranoside and (25S)-26-O-β-d-glucopyranosyl-5β-furostan-1β,2β,3β,5β,22α,26-hexaol and the known compounds (25S)-26-O-β-d-glucopyranosyl-5β-furostan-3β,22α,26-triol-3-O-α-l-rhamnopyranosyl-(1→2)-O-β-d-glucopyranosyl-(1→2)-O-β-d-glucopyranoside and (25S)-26-O-β-d-glucopyranosyl-5β-furostan-3β,22α,26-triol-3-O-β-d-glucopyranosyl-(1→2)-O-β-d-glucopyranoside, trans-resveratrol, (+) catechin and (−) epicatechin in the rhizomes of Smilax aspera is reported. All saponins have been isolated as their 22-OMe derivatives, which were further subjected to extensive spectroscopic analysis. The isolated furostanol saponins were evaluated for cytotoxic activity against human normal amniotic and human lung carcinoma cell lines using neutral red and MTT assays. In vitro experiments showed significant cytotoxicity in a dose dependent manner with IC50 values in the range of 32.98-94.53 µM.  相似文献

The present study is designed to investigate the effect of methanolic extract of outer scales and edible portions of Allium cepa bulb on ischemia and reperfusion-induced cerebral injury. Global cerebral ischemia was induced by bilateral carotid artery occlusion for 10 min followed by reperfusion for 24 h. Pretreatment with methanolic extract of outer scales (100 mg/kg and 200 mg/kg) and edible portions (100 mg/kg and 200 mg/kg) of A. cepa bulb markedly reduced cerebral infarct size and attenuated impairment in short-term memory and motor coordination. The protective effect of methanolic extract of outer scales and edible portions of A. cepa bulb was accompanied by a marked decrease in mitochondrial TBARS.  相似文献

Two new saponins of 20, 26-epoxy derivatives of pseudojujubogenin, hoduloside XI (1) and hoduloside XII (2) were isolated from the seeds of Hovenia trichocarpa. The structures of the new compounds were established by extensive NMR experiments and chemical methods. Hoduloside XI was confirmed to be 3-O-{β-d-glucopyranosyl(1  3)-[β-d-xylopyranosyl(1  2)]-α-l-arabinopyranosyl}-20, 26-epoxypseudojujubogenin. Hoduloside XII was identified as 3-O-{β-d-xylopyranose(1  2)glucopyranosyl(1  3)[rhamnopyranose(1  2)]β-d-glucopyranosyl}-20, 26-epoxypseudojujubogenin. The in vitro cytotoxic activity of compounds 1 and 2 was assayed. They displayed inhibitive activities against human cancer cell lines HL60 and K562.  相似文献

The programmed cell death (PCD) characteristics of Ulmus pumila L. seeds were investigated. The seeds were treated at a high temperature of 37°C and 100% relative humidity for six days. DAPI (4′6-diamidino-2-phenylindole) staining revealed that the aging treatment induced condensation and margination of chromatin, as well as the formation of apoptotic bodies. DNA electrophoresis results of U. pumila seeds on an agarose gel showed a characteristic “ladder” pattern. Levels of electrolyte leakage of seed cells showed that membranes retained their integral form during almost the entire aging time. There was an immediate increase in the production rate of superoxide anion (O2 ) and in the amount of hydrogen peroxide (H2O2), which remained at a μmol level. At 15-20 degrees C, seeds of Eucalyptus sieberi L. Johnson need to maintain a water content equal to 30% or more of dry weight for approximately 60 h in order to germinate. Several shorter periods of imbibition at 30% water content separated by between 6 and 120 h desiccation were as effective as an unintenupted imbibition in bringing seeds to the point of germination. Even after 8 months' desiccation an earlier imbibition shortened the time required for the completion of germination, although it did not hasten the onset of germination. The water potential of seeds sufficiently hydrated to germinate was about -0.6 MPa, whereas that of the embryo was only about -4.5 MPa. The difference was caused by the inhibition of water uptake by the inner integument of the seed coat. During germination, reserve protein in a small group of cells above the collet was hydrolyzed. Subsequent expansion of these cells caused rupture of the inner integument thereby allowing further water uptake by the embryo. The balance of water absorption and evaporative loss by the outer integument was such that most seeds on a moist substrate but exposed to dry air (6.5-8 x 10(-4) MPa) did not maintain the 30% water content necessary for germination. Thus although able to germinate on seedbeds subject to intermittent drying, E. sieberi seeds have the capacity to avoid germination except under humid conditions.  相似文献

Five new triterpene saponins, Ilexpublesnins N–R (15), along with seven known analogs were isolated from the root of Ilex pubescens. Their structures were elucidated on the basis of extensive spectroscopic analysis, including 1D and 2D NMR experiments. Ilexpublesnin N (1) possessed a rare 20-hydroxyursolic acid scaffold from natural resource. These compounds were evaluated in vitro for their cytotoxic effects on human cancer cell lines HepG2, HLE, BEL7402, BEL7403, BEL7405, MCF-7, HeLa. Among them, only compounds 5 and 10 showed cytotoxic potentiality against BEl-7403 and HEL cell lines [inhibition (%): 35.38 and 45.12, respectively].  相似文献

A new triterpenoid saponin was isolated from the roots of Silene cucubalus and its structure was determined on the basis of one- and two-dimensional NMR spectroscopy as gypsogenin 3-O-beta-xylopyranosyl-(1-->3)-[beta-galactopyranosyl-(1-->2)]-beta-glucuronopyranoside. A known saponin, quillaic acid 3-O-beta-xylopyranosyl-(1-->3)-[beta-galactopyranosyl-(1-->2)]-beta-glucuronopyranoside, was also isolated.  相似文献

A new monodesmosidic spirostanol saponin, along with three known saponins was isolated from Agave macroacantha Zucc leaves. The structure of the new saponin was established as hecogenin-3-O-α-l-rhamnopyranosyl (1→4) β-d-xylopyranosyl (1→3)[β-d-glucopyranosyl (1→2)] β-d-glucopyranosyl (1→4) β-d-galactopyranoside. The 1H and 13C resonances of the four compounds were assigned using a combination of 1D and 2D NMR techniques including 1H, 13C, COSY, TOCSY, ROESY, HSQC and HMBC NMR and confirmed by mass spectrometry.  相似文献

Stratified loblolly pine (Pinus taeda L.) seeds were placed at three depths in a reconstructed forest floor and subjected to simulated prescribed winter burns. Within the forest floor, pine seeds were placed at the L/upper-F interface, upper-F/lower-F interface, and lower-F/mineral-soil interface. Wind was generated by electric box-fans. Seeds that survived the burns were transferred to moist sand flats for 30-day germination tests. About 96% of seeds placed at the two upper layers in the forest floor were either destroyed by the fires or failed to germinate. Germinative capacity of seeds placed at the lower-F/mineral-soil interface averaged 79% as compared to 97% for unburned control seeds.  相似文献

Phytochemical investigation of the leaves of Aralia elata has led to the isolation of four new compounds, 3-O-β-D-glucopyranosyl (1→3)-β-D-glucopyranosyl (1→3)-β-D-glucopyranosyl oleanolic acid (1), 3-O-[β-D-glucopyranosyl (1→3)-β-D-glucopyranosyl (1→3)]-[β-D-glucopyranosyl (1→2)]-β-d-glucopyranosyl hederagenin 28-O-β-D-glucopyranoside (2), 3-O-{[β-D-glucopyranosyl (1→2)]-[β-d-glucopyranosyl (1→3)-β-d-glucopyranosyl (1→3)]-β-D-glucopyranosyl} oleanolic acid 28-O-β-D-glucopyranosyl ester (3) and 3-O-[β-D-glucopyranosyl (1→2)]-[β-D-glucopyranosyl (1→3)]-β-d-glucopyranosyl caulophyllogenin (4) and two known compounds, 3-O-[β-D-glucopyranosyl (1→3)-α-l-arabinopyranosyl]-echinocystic acid (5) and 3-O-α-L-arabinopyranosyl echinocystic acid (6). The structural determination was accomplished with spectroscopic analysis, in particular (13)C-NMR, 2D-NMR and HR-ESI-MS techniques. Compounds 1-6 were tested for their inhibition of the growth of HL60, A549 and DU145 cancer cells. Compound 1 showed significant cytotoxic activity against HL60 and A549 cancer cells with IC(50) values of 6.99μM and 7.93μM respectively. In addition, compounds 5 and 6 showed significant cytotoxic activity against HL60 cancer cells with IC(50) values of 5.75μM and 7.51μM, respectively.  相似文献

A protocol was discussed for high efficient plant regeneration from seven bluegrass (Poa pratensis L.) cultivars via an indirect callus induction and somatic embryogenesis method. Mature seeds were used as explants for callus initiation. Callus induction and proliferation efficiencies were investigated on NB, modified MS (MMS) and MS media, supplemented with 2.0 mg·L-1 2,4-dichlorophenoxyacetic acid (2,4-D). The MMS medium performed best. Based on the MMS medium, direct and indirect callus induction effects of bluegrass from mature seeds were compared at the range of 1-5 mg·L-1 2,4-D contained in the medium. Under the direct callus induction method, the most suitable 2,4-D concentrations varied among cultivars. Under the indirect callus induction method, a significantly high callus induction frequency (93.33%-98.33%) was obtained and there were barely any statistically significant differences among the tested genetically diverse cultivars. Somatic embryos were promoted on the MMS medium supplemented with 3 mg·L-1 2,4-D, 0.1 mg·L-1 kinetin and 0.8 mg·L-1 CuSO4. Embryogenetic calli developed into plantlets on the MMS medium containing different concentrations of thidiazuron (TDZ), and the differentiation frequencies varied in the range from 20.15% to 77.65%. The 0.25 mg·L-1 TDZ was generally the most suitable concentration for the tested cultivars.  相似文献
