硅营养对UV-B辐射条件下水稻酚类代谢的影响   总被引:1,自引:0,他引:1  
本研究在不同硅营养条件下,探讨了UV-B辐射增强对两个供试水稻品种(Lemont和Dular)硅营养特性和酚代谢物质的影响。结果表明:(1)在缺硅营养和UV-B辐射胁迫下,Lemont的硅营养吸收能力和生理利用率都比Dular强。(2)不同硅营养条件下,UV-B辐射胁迫促使Lemont叶片的PAL、PPO、 POD、CAT活性增强,总酚、类黄酮和绿原酸含量升高,而可溶性蛋白质含量降低;上述指标在Dular叶片中也表现出相同的变化趋势。但在缺硅营养和UV-B辐射共同胁迫下,Lemont表现出较强的酚类合成能力。(3)硅营养有利于水稻酚类代谢物质的积累,以减少UV-B辐射的伤害。综上所述,Lemont与Dular之间硅营养特性的不同,可能是引起两个供试水稻的酚类代谢差异,从而表现出对UV-B辐射抗性不同的原因之一。  相似文献

Inheritance patterns for table grape anthocyanins were investigated on three cross offspring populations during two successive years. Sixteen anthocyanins were detected, and all were monoglucoside derivatives. The proportion of anthocyanins in the maternal parent determined the proportion of anthocyanins in the offspring. But the absolute content of the maternal parent had no significant effect on progenies. Peonidin 3-O-glucoside and malvidin 3-O-glucoside were the most abundant anthocyanins, not only in the maternal parent but also in the progenies. The presence or absence of anthocyanins in grape skin was inheritance of a quality character controlled by oligogenes, and anthocyanins content was a quantitative character controlled by polygenes. Via principal component (PC) analysis, factors that affected the total content of cross progeny populations were peonidin 3-O-glucoside, malvidin 3-O-glucoside, delphinidin 3-O-glucoside, cyanidin 3-O-glucoside, petunidin 3-O-glucoside, peonidin 3-O-(6-O-coumaryl)-glucoside, and malvidin 3-O-(6-O-coumaryl)-glucoside. Anthocyanins content was a high broad sense heritability character (H2), and H2 was stable in different cross combinations (ranging from 0.65 to 0.98).  相似文献

The cabbage root fly Delia radicum L. (Diptera: Anthomyiidae) is one of the major pests of many Brassica crops in the temperate areas of Europe and North America. At present, turnip (B. rapa ssp. rapa L.) varieties resistant to the pest does not exist. With the aim to fill this gap, a no-choice tolerance test of 56 accessions among turnips, turnip tops and turnip greens was performed under controlled conditions by introducing D. radicum eggs. Plant survival, leaf and root conditions, pupae number and weight significantly varied among plant accessions. Ten putatively resistant and ten susceptible accessions (control group) were selected from this first screening, transplanted in the field and exposed to natural infestation to detect antibiosis and antixenosis mechanisms. Both in the laboratory and in the field, pupae number significantly varied within accessions and between resistant and susceptible group, although pupal weight did not, indicating the absence of antibiosis effect on this early stage. In the field, the number of galleries was significantly lower in the resistant group in comparison with the control. Resistant accessions had smaller size, and a smaller, white and mostly buried root. Within the resistant and susceptible accessions, larger plants harboured more pupae, however purple roots were those most preferred, and the hosted pupae weighed most. Three accessions from the resistant group (MBGBR0178, MBGBR0570 and MBGBR0371) stand out for resistance to D. radicum possibly through antixenosis mechanisms.  相似文献

Molecular screening and genetic diversity of major rice blast resistance (R) genes were determined in 32 accessions of rice germplasm from North East and Eastern India with ten gene based single nucleotide polymorphisms and sequence tagged sites (STS) markers, namely z56592, zt56591, k39512, k3957, candidate gene marker, Pita3, YL155/YL87, YL183/YL87, Pb28, 195R-1 which showed close-set linkage to nine major rice blast resistance (R) genes, Piz, Piz-t, Pik, Pik-p, Pik-h, Pita/Pita-2, Pib and Pi9 and one susceptible pita gene. Among the 32 accessions, 13 were positive for Piz gene and six for Piz-t gene. Six accessions were positive for Pik gene, seven for Pik-p and 16 for Pik-h gene. One accession, Atte thima, was positive for three of Pik multiple genes. Out of 32, only two germplasm, Dudhraj and Nepali dhan, were detected with both Pita3 and YL155/YL87 marker for Pita/Pita-2 gene. The Pib gene appeared to be omnipresent and was detected in 31 of 32 germplasm with marker Pb28. The gene specific STS marker, 195R-1, for Pi9 gene produced positive bands in only two germplasm, Kalchatti and Bachi thima. The Uniform Blast Nursery (UBN) analysis showed that out of 32, six germplasm was resistant, ten moderately resistant and 16 germplasm were susceptible. Presence of Piz-t, Pita/Pita-2 and Pi9 gene ensured a resistant reaction in outdoor blast nursery whereas germplasm carrying Pib was susceptible when present alone. Presence of multiple genes, however, contributed to slow blasting resistance in the field. These results are useful in identification and incorporation of resistant genes from the germplasm into elite cultivars through marker assisted selection in rice breeding programs.  相似文献

This study assessed the altitudinal variations in the anthocyanin and isoflavone contents of six black seed coated soybean [Glycine max (L.) Merrill] cultivars. The black soybean cultivars Heugcheong, Seonheuk, Geomjeong 1, Geomjeong 2, Cheongja 2, and Cheongja 3 were planted at Milyang (12 m above mean sea level — low altitude) and Muju (600 m — high altitude), Korea on 10 June 2005 and 2006. The total anthocyanin and isoflavone contents and individual components were investigated by reverse-phase high performance liquid chromatography (HPLC). All black soybean cultivars cultivated in high altitude possessed significantly higher total anthocyanin (p < 0.01) and isoflavone (p < 0.01) contents than those grown in low altitude. For anthocyanin composition, cyanidin-3-O-glucoside, cyanidin-3-O-galactoside, and peonidin-3-O-glucoside contents were significantly higher while delphinidin-3-O-glucoside contents was significantly lower at high altitude. The composition of individual isoflavones, 6″-O-malonyldaidzin, and 6″-O-malonylgenistin contents significantly increased at high altitude.  相似文献

Visual quality is one of the major factors determining the market value of field pea (Pisum sativum L.). Breeding for improved visual quality of pea seeds is currently a challenging task, mainly because of the complexity and the lack of sound genetic knowledge of the traits. The objectives of this research were to characterize post-harvest cotyledon bleaching resistance in green pea at the biochemical and gene expression levels. Seed coats and cotyledons of two pea cultivars, CDC Striker (bleaching resistant) and Orb (bleaching susceptible) at three developmental stages [14, 21 and 28 days after flowering (DAF)] and following exposure to accelerated bleaching conditions after harvest (0, 3, 6, and 13 days after bleaching (DAB) were evaluated. CDC Striker had a slower rate of chlorophyll degradation in cotyledons, and a higher total carotenoids to chlorophyll ratio in seed coats, than Orb when seed samples were exposed to high intensity light. An oligo-nucleotide microarray (Ps6kOLI1) revealed that gene expression profiles of the CDC Striker and Orb seed coats were significantly different during seed developmental stages. A significant up regulation of genes involved in the production and accumulation of secondary metabolites responsible for antioxidant properties including epiafzelechin, epicatechin, epigallocatechin, kaempferide, kaempferol 3-O-β-d-sophorotrioside, O-quercetin and rutin, in the seed coats of CDC Striker were observed. Thus, bleaching resistance in field pea could be due to the accumulation of specific carotenoids and phenolic compounds which quench excess light or scavenge free radical singlet oxygen molecules. The candidate genes identified in this project will need to be validated using qPCR to further support their involvement with bleaching resistance before being used for the development of gene specific markers.  相似文献

We have analyzed the influence of the stage of ripeness on L-phenylalanine ammonia-lyase (PAL) gene expression, accumulation of anthocyanins and total phenolics, and on antioxidant activity in the skin of table grapes treated with 20% CO2 + 20% O2 + 60 % N2 for 3 or 6 d at low temperature (0 °C). The residual effect of high CO2 treatment after transfer to air was also studied. In early harvested grapes, neither the anthocyanin content nor the accumulation of VcPAL mRNA was affected by any of the CO2 treatments applied. However, in late harvested grapes, the duration of high CO2 treatment determined its effect and a 6 d treatment with CO2 sustained higher levels of total phenolics and anthocyanin accumulation, and VcPAL expression than observed in untreated late harvested grapes. The increased antioxidant capacity was correlated with the total amount of phenolics and anthocyanins. Conversely, in grapes treated for 3 d with CO2 the phenylpropanoid pathway did not appear to be induced and a relationship between antioxidant activity and anthocyanins was not observed. Thus, further studies are needed to identify the most important antioxidants in these treated fruit.  相似文献

In wheat, the transition from the vegetative to reproductive stage is primarily controlled by the series of vernalisation (Vrn-1) genes located on the homoeologous group 5 chromosomes. Up to 2009, only two alleles at the Vrn-B1 locus were known: one dominant, spring, allele (now designated Vrn-B1a) and the other recessive, winter, (vrn-B1) allele. Recently, two additional dominant alleles, Vrn-B1b and Vrn-B1c, were described. In this study, we screened a range of hexaploid spring wheat germplasms for the presence of different Vrn-B1 alleles using new diagnostic molecular markers. Our results show that the Vrn-B1a allele was the most prevalent, being present in 55.3 % of the 2,495 accessions examined, followed by the recessive vrn-B1 allele, which occurred in 31.5 % of the accessions. The novel alleles Vrn-B1b and Vrn-B1c were found in 5.3 and 7.9 % of all accessions, respectively.  相似文献

Carrot sticks are increasingly in demand as ready-to-eat products, with a major quality problem in the development of white discoloration. Modified atmosphere packaging (MAP) and edible coating have been proposed as postharvest treatments to maintain quality and prolong shelf-life. The combined application of an edible coating containing 5 mL L?1 of chitosan under two different MAP conditions (10 kPa O2 + 10 kPa CO2 in Pack A and 2 kPa O2 + 15–25 kPa CO2 in Pack B) over 12 d at 4 °C was studied. Respiration rate, microbial and sensory qualities as well as the contents of vitamin C, carotenoids and phenolics of coated and uncoated carrot sticks were evaluated. The use of the edible coating containing chitosan preserved the overall visual quality and reduced surface whiteness during storage. Microbial populations were very low and not influenced by coating or MAP. Edible coating increased respiration rates of carrot sticks, although this was only noticeable in the package with the less permeable film (Pack B). Vitamin C and carotenoids decreased during storage particularly in coated carrot sticks. In contrast, the content of total phenolics markedly increased in coated carrot sticks stored under moderate O2 and CO2 levels, while it was controlled under low O2 and high CO2 levels. The combined application of edible coating containing chitosan and moderate O2 and CO2 levels maintained quality and enhanced phenolic content in carrot sticks.  相似文献

Fusarium head blight (FBH) is a serious disease of cultivated oats (Avena sativa L.). The objective of this study was to screen a set of oat genotypes from the VIR Avena sativa germplasm collection for the FHB resistance. Of the 155 screened genotypes 42 % belonged to both hulled and naked cultivars, 55 % were landraces and the rest were breeding lines. Screening nurseries were planted at two locations: in the Northwestern Russia in 2007–2008 with artificial inoculation by F. sporotrichioides strains and in the Russian Far East in 2009 (natural infections). The resistance data were based on three different parameters: (a) percentage of grain showing evidence of Fusarium damage, (b) the DNA content of the trichothecene-producing Fusarium species and (c) the trichothecene mycotoxin accumulation; the sum of which permitted identification of genotypes with multicomponent resistance to FHB. The highest degrees of resistance were observed in the naked cvs. Iymay (k-11014, China) and Numbat (k-14851, Australia) and by the hulled cv. Kuromi (к-11632, Japan). The most resistant landraces were the hulled oats originating from the Asian region (k-2513, k-6963, k-7766, and k-8479). Considerable similarity was observed between the resistance reactions of oat genotypes to the Fusarium disease under different environmental conditions. The identified resistant accessions may serve as crossing partners in oat breeding efforts.  相似文献

Pericarp colour of litchi fruit is an important quality attribute that determines its market value and consumer acceptance. Plant growth regulators (PGR) such as abscisic acid (ABA) and ethephon are known to play important roles in peel colour development during maturation and ripening of non-climacteric fruits (e.g. grape and litchi). Our aim was to investigate the effects of preharvest application of ABA, ethephon and their combination on pericarp colour and fruit quality of litchi (cv. Calcuttia) and also to assess the potential effects on postharvest performance of fruit. Exogenous application of ABA (150 or 300 mg L−1) at the colour-break stage significantly increased the concentration of total anthocyanins and cyanidin-3-O-rutinoside, the major anthocyanin contributing ∼71–96% of the total anthocyanins, in litchi pericarp compared to ethephon (500 μL L−1). Among different anthocyanins quantified, the relative contribution of cyanidin-3,5-diglucoside to the total anthocyanins was significantly higher in all PGR-treated fruit compared to the control, but the concentration of cyanidin-3-O-glucoside was specifically enhanced by ABA. No significant effect on the concentrations of epicatechin, and quercetin-3-O-rutinoside was observed in response to PGR treatments. Ethephon (500 μL L−1) treatment did not significantly increase the anthocyanin levels in pericarp, but it caused more degradation of chlorophyll pigments than control. Aril quality with regard to firmness, soluble solids and acidity was not significantly affected by PGR treatments, except that ethephon-treated fruit showed significant softening and lower acidity. Postharvest changes in fruit quality attributes including pericarp browning during cold storage at 5 °C for 14 d were mainly related to the storage duration effect, rather than PGR treatment. In conclusion, ABA treatment (150 or 300 mg L−1) at the colour-break stage enhanced anthocyanins accumulation in litchi pericarp without adversely affecting postharvest quality and storage stability for 14 d.  相似文献

Screening of pea germplasm for resistance to powdery mildew   总被引:1,自引:0,他引:1  
Powdery mildew caused by Erysiphe pisi DC results heavy losses in the yield and quality of pods and seeds of pea crop. Germplasm comprising 701 accessions of garden and field pea originating from 60 countries were screened for powdery mildew resistance under natural epiphytotic conditions and 64 accessions found resistant in field screening for 2?years at one location were further screened both in field at two locations and artificially in laboratory to four isolates. The information was also obtained on the amount of genetic diversity and agronomic superiority in resistant accessions. Fifty-seven accessions showed resistant reaction for 3 consecutive years in field screening but only 14 accessions originating from 10 countries showed resistant reaction in laboratory screening against the four most prevalent isolates of E. pisi collected from different places in the area of experiment. Germplasm lines showed both complete and incomplete levels of resistance and variable reactions to different isolates. There was sufficient genetic diversity and agronomic superiority in the resistant accessions e.g. EC598655, EC598878, EC598704, IC278261, and IC218988, which may serve as useful genetic material to plant breeders for breeding pea varieties for powdery mildew resistance and high yield.  相似文献

Little is known about the levels of resistance to Orobanche crenata available in narbon vetch. A germplasm collection of 200 accessions of narbon vetch (Vicia narbonensis L.) originating from different countries was screened for resistance to crenate broomrape under field conditions. Thirteen accessions were selected for more detailed screening under controlled conditions and for additional field testing. Resistance to O. crenata was manifested by lower germination of broomrape seeds, reduced emergence of Orobanche shoots and fewer root attachments per narbon vetch plant. Differences in pod yield among narbon vetch accessions were also found in response to Orobanche infestation. Finally, eight accessions were selected and identified as new sources of narbon vetch resistant to broomrape.  相似文献

Host plant resistance is an effective protection strategy to control aphids in many crops. However, the evolution of insensitive aphid biotypes necessitates the search for new resistance sources. Wild relatives of crop plants can be important sources for resistance genes to be introgressed into new cultivars. Nasonovia ribisnigri (Mosely) (Homoptera: Aphididae) is an important pest of cultivated lettuce, Lactuca sativa. Since 1982, resistance introduced into lettuce cultivars has relied on the Nr-gene, originating from a wild relative, Lactuca virosa. In 2007 first reports appeared that Nr-based resistance had become ineffective against certain populations of N. ribisnigri. The objective of this study was to establish if the original donor of the Nr-gene L. virosa accession, IVT 280, is resistant against recently emerged virulent (Nr:1) N. ribisnigri biotypes. To this end we investigated feeding and penetration behaviour of virulent and avirulent (Nr:0) aphids on the resistant L. virosa, IVT 280, and two susceptible L. virosa accessions, using the electrical penetration graph method. Additionally, aphid performance was analysed in terms of survival, development time and reproduction on these accessions. L. virosa accession IVT 280 was resistant against all populations of N. ribisnigri tested. The ingestion of phloem was strongly reduced on the resistant accession compared to the susceptible L. virosa accessions. Additionally, none of the aphids survived on the resistant accession that, therefore, constitutes a good source of resistance in lettuce against both biotypes of N. ribisnigri.  相似文献

Mist-chamber, field, and detached leaf inoculation procedures identified plants resistant to bacterial soft rot [Erwinia carotovora subsp. carotovora, (Ecc)] in Brassica rapa and related species. The mist-chamber seedling inoculation provided the best correlation of mean disease severity ratings with the field plant inoculation (r = 0.67**) and was used to identify resistant materials. The optimum mist-chamber incubation conditions to distinguish the resistance of accessions were 23 °C and 100% RH and were used for primary screening. A total of 752 accessions of B. rapa and related Cruciferae were screened. In general, accessions of B. oleracea were more resistant than accessions of B. rapa. Within B. rapa, subspecies pekinensis and chinensis were more susceptible than other subspecies. No completely resistant material was found. In species of B. rapa, only 7% of accessions showed some degree of resistance with plant-to-plant variation within the accessions. G30444, G30449, and AVRDC2837 were identified as the most resistant materials in B. rapa by both mist-chamber and field inoculations. The resistance was correlated between an USA isolate (Geneva-1) and two Chinese isolates (RL4-1 and RL-19) of Ecc. Thirty-nine sea beet [Beta vulgaris L. ssp. maritima (L.) Arcang.] accessions of the Adriatic coast were screened genetically and for their adaptive morpho-functional root traits in order to identify new sources of abiotic resistances for sugar beet breeding programs. Genetic diversity was evaluated with 21 microsatellites markers that identified 44 polymorphic alleles. Sea beets grouped into two main clusters: the West and the East Adriatic coast groups, with the latter showing higher genetic diversity. Among sea beet accessions with desirable root traits, four accessions have proved to be interesting for sugar beet [B. vulgaris (L.) ssp. vulgaris] breeding aimed to improve tolerance to nutritional stresses. Lastovo (ID 29) and Zut (ID 34) accessions were characterized by the highest values of RER, TRL, FRL and RSA still maintaining a high value of RTD, while Grado (ID 21) an Portic (ID 23) accessions were characterized by the highest RTD, but with low values of RER, TRL, FRL and RSA parameters.  相似文献

Red clover (Trifolium pratense) is an important perennial forage crop that is widely cultivated in Europe. Clover rot remains a major disease in red clover, but resistance breeding is hampered by the lack of available sources of resistance. Moreover, little is known about the factors that influence clover rot resistance. In this paper we evaluated the variation in clover rot susceptibility among a diverse collection of 113 red clover accessions, with the aim of identifying more resistant accessions. Clover rot susceptibility was assessed with a high throughput bio-test on young plants. We found significant variation in clover rot susceptibility, within and among accessions. ‘Tedi’, ‘Maro’ and ‘No 292’ were the most resistant accessions. Fifteen diploid accessions were more susceptible than the average accession with the cultivar ‘Nemaro’ being the most susceptible. Clover rot susceptibility was not correlated with isoflavone levels from Mullaney et al. (Agronomy abstract. ASA, Madison, p 195, 2000). Cultivars were more resistant than landraces and wild accessions and tetraploid cultivars were more resistant than diploid cultivars. Besides the in-depth analysis for clover rot susceptibility, possible correlations with plant architecture and other diseases were investigated. Growth habit, branching, plant yield, flowering date and susceptibility to mildew, virus and rust diseases were investigated in a 3-year field trial. Unlike previously suggested, clover rot susceptibility was not correlated with branching or with plant yield over three years. On the other hand, late flowering accessions and accessions with erect growth habit were less susceptible to clover rot. Clover rot susceptibility was not correlated with susceptibility to rust disease (Uromyces trifolii) or viral diseases, but negatively with susceptibility to mildew (Erysiphe polygoni). Because no completely resistant accessions were found, the best way to improve clover rot resistance would be to select recurrently for resistant genotypes among diverse cultivars and landraces with lower susceptibility. Tetraploidisation of diploid populations with a higher resistance level can provide an additional level of protection.  相似文献

An investigation was carried out with 25 populations of Rauwolfia serpentina to assess variability, association of yield components and their direct and indirect effects on yield. Analysis of variance revealed significant differences among 25 populations of R. serpentina for all the traits. Heritability estimates in broad sense were high for most of the traits and moderate for number of primary root branch. As in case of heritability, expected genetic advance was the highest for plant height, number of primary stem branch, reserpine content (%) in root, the most important trait for commercial utilization. Correlation analysis indicated positive and significant phenotypic correlation of reserpine content with root length and total alkaloid content (%) and significant negative association with root diameter. The partitioning of genotypic correlation coefficient into direct and indirect effects revealed total alkaloid content and root yield were the most important direct contributors to reserpine percent in root. It is concluded that accessions with more alkaloid content and root length are likely to yield more reserpine content in root and these characters can be considered to be reliable traits for selection of high reserpine content in root.  相似文献

A total of 324 Japanese rice accessions, including landrace, improved, and weedy types were used to 1) investigate genetic variations in blast resistance to standard differential isolates, and 2) across the genome using polymorphism data on 64 SSR markers. From the polymorphism data, the accessions were classified into two clusters. Accessions from irrigated lowland areas were included mainly in cluster I, and upland and Indica types were mainly in cluster II. The accessions were classified into three resistance subgroups, A2, B1 and B2, based on the reaction patterns to blast isolates. The accessions in A2 were postulated to have at least two resistance genes Pish and Pik-s, whereas those in B1 had various combinations of the resistance genes Pish, Pia, Pii, Pi3, Pi5(t), and Pik alleles. The B2 accessions were resistant to almost all isolates, and many accessions of cluster II were included, and had Pish, Pia, Pii, Pi3, Pi5(t), certain Pik, Piz and Pita alleles, and unknown genes. The frequencies of accessions of B1 originating in Hokkaido, and those of B2 originating in the Kanto and Tohoku regions were remarkably higher than in the other regions.  相似文献
