Summary Although wild oats (Avena fatua L.) have been considered a potential source of genes for cultivated oat (Avena sativa L.) improvement, most progenies of A. sativa/A. fatua crosses have weak straw and are very susceptible to crown rust (Puccinia coronata CDA. var. avenae Fraser and Led.). Backcrossing to A. sativa has been suggested as a method of improving progeny lines while introducing new genes from wild oats to cultivated oats. In this study, A. sativa/A. fatua F1 hybrids were backcrossed twice to A. sativa, and lines from three backcross populations were selected on the basis of agronomic performance in segregating generations. The A. sativa recurrent parents were Dal (tall and late) and Stout (short and early).Backcross lines and recurrent parents were evaluated in five performance trails from 1983 through 1985. There was significant variation among backcross lines for most traits, but most backcross lines did not produce higher grain and straw yields than their A. sativa parent. Several backcross lines were higher than their recurrent parent in test weight and groat percentage. A line derived from Stout, 175BC2-6, was considered the most promising backcross line in the study. This line produced more grain, had heavier kernels, and headed 3.3 days earlier than Stout. Although 175BC2-6 does not have sufficient straw strength and crown rust resistance to be released as a cultivar, it is considered to be a new source of high grain yield, high test weight and earliness for oat breeding. 相似文献

Summary Two lines of descent were established from an F3 bulk lot of oats (Avena sativa L.) initiated by mixing seeds from approximately 250 crosses. For one line of descent, seeds were radiated with thermal neutrons or X-rays from F3 through F6, followed by five generations of bulk propagation. The second was propagated for 10 generations. No artificial selection was practiced in either line of descent. Grain yield data from 20 random strains from each of four generations from the radiated (F7, F8, F9, and F11) and five from the nonradiated (F3, F6, F7, F8, and F12) line of descent and 20 check cultivars tested in 14 environments were used for estimating regression stability indexes of oat strains.The 14 environments were assigned randomly to two sets of seven, and regression stability indexes were computed for the 180 experimental oat strains for both sets. Intrageneration correlations between regression stability indexes from the two sets of environments ranged from –0.35 to 0.64 (18 d.f.), and only one of nine was significant, indicating poor repeatability for estimates of this statistic computed from different sets of environments.Correlations between regression stability indexes from two sets of environments, one in which the environments varied by soil nitrogen levels and a second in which they varied by soil phosphorus levels, ranged from –0.01 to 0.28, none of which was significant.The relative magnitudes and ranking of the regression stability index values for the oat genotypes were nearly identical when environmental productivity indexes were assessed with any number of check cultivars from 2 to 20.Journal paper No. J-8080 of the Iowa Agriculture and Home Economics Experiment Station. Summary Gene action and heritability of groat protein percentage were determined in F1, F2, and F3 generations of nine crosses between three Avena sativa L. cultivars and three A. fatua L. selections. Relationships among groat protein percentage, grain yield, and 100-seed weight also were evaluated. The three A. sativa parents were Dal (high grain yield and intermediate groat protein percentage), Goodland (low grain yield and high groat protein percentage), and Stout (high grain yield and low groat protein percentage). The three A. fatua parents were chosen for the study on the basis of vigorous plant growth and high groat protein percentage. The study was conducted at Madison, Wisconsin in 1979 and 1980.There was partial dominance towards low groat protein percentage. Narrow sense heritability estimates for groat protein percentage were low in Dal and Goodland crosses and intermediate in Stout crosses. In the F2 generation, groat protein percentage was significantly higher in shattering than in nonshattering plants in 1979, but not in 1980. There were significant, positive correlations between groat protein percentage, 100-seed weight, and grain yield in F1 and F2 generations, but they were not large numerically. Relationships among these traits were either negative or nonsignificant in the F3 generation. Although our results indicated that selection for higher groat protein percentage is possible when a low protein A. sativa cultivar is used, most of our simple cross progenies from A. sativa x A. fatua crosses had weak straw and were susceptible to crown rust (Puccinia coronata

Summary The regular meiotic behaviour of the cultivated oat Avena sativa (2n=6x=42) is genetically controlled. The factors which control the diploid-like meiotic behaviour also restrict the amount of pairing that occurs between alien chromosomes and their homoeologues in A.sativa, and hence increases the difficulties of introducing desirable variation from wild species into the cultivated oat. A genotype of the diploid species A.longiglumis which interferes with the regular meiotic behaviour of A. sativa can be used to induce pairing between alien chromosomes and their corresponding chromosomes in A. sativa. Using this procedure the dominant gene conferring mildew resistance has been transferred from the tetraploid weed species A. barbata into the cultivated oat. 相似文献

Five parents of common vetch (Vicia sativa L.) having orange/beige cotyledon colour, brown/white testa colour, purple/green seedling colour and purple/white flower colour were crossed as a full diallele set. The inheritance patterns of cotyledon, testa or seed coat colour, flower and seedling colour, were studied by analyzing their F1, F2, BC1 and BC2 generations. The segregation pattern in F2, BC1 and BC2, showed that cotyledon colour was governed by a single gene with incomplete dominance and it is proposed that cotyledon colour is controlled by two allelic genes, which have been designated Ct1 and Ct2. Testa colour was governed by a single gene with the brown allele dominant and the recessive allele white. This gene has been given the symbol H. Two complementary genes governed both flower and seedling colours. These flower and seedling colour genes are pleiotropic and the two genes have been given the symbols S and F. 相似文献

Long-term selection experiments provide germplasm to study the effects of selection in a closed population. Recurrent selection
to enhance grain yield in oat has been ongoing at the University of Minnesota since 1968. The objectives of this study were:
(i) estimate the GCA and SCA effects for three agronomic traits in the seventh cycle of selection, (ii) assess the effect
of the current methods of selection on parental contribution and unselected traits, and (iii) determine the direct and indirect
responses to seven cycles of recurrent selection for grain yield. Progeny of the Cycle 6 parents and parents for Cycles 0
through 7 were grown in two separate tests. Grain yield, heading date and plant height were evaluated in each test. Grain
yield was increased by 21.7% after seven cycles of selection. Evaluation of Cycle 6 progeny showed that GCA effects were significant
for all three traits studied, and SCA effects were significant only for grain yield. Four Cycle 6 parents did not have any
progeny selected as Cycle 7 parents. Results from this study indicate that long term recurrent selection has continued to
increase grain yield. Alternative selection strategies may be necessary to maintain the genetic variability in this population,
particularly when improvement of secondary traits is required.

Two molecular marker techniques: RAPD and simplified PstIAFLP have been compared in order to decide on, which technique is better suited to genetic characterization of oat (Avena sativa L.) cultivars. It was investigated, if the same pattern of variability is revealed by two approaches and whether the observed
molecular variability reflects pedigree-based relationships. Polymorphic RAPD and PstIAFLP markers were sufficient to distinguish all analysed cultivars, demonstrating the usefulness of both methods for cultivar
identification. Genetic similarity estimates derived from RAPD, simplified PstIAFLP and combined RAPD and PstIAFLP data were compared with coefficients of parentage (COP). Molecular markers-based mean genetic similarities were considerably
greater than mean COP value. Correlation coefficients between COP and genetic similarities calculated from RAPD, PstIAFLP and combined molecular data were very low and not significant. A better correlation (0.50) was found between similarity
estimates derived from RAPD and PstIAFLP markers. Four separate dendrograms were constructed based on pedigree and molecular analyses using a neighbor-joining
algorithm (NJ). The dendrograms were compared and found to be topologically different. The results of this study showed, that
both molecular techniques can be conveniently used for genetic characterization of oat cultivars, however PstIAFLP would be the method of choice due to the higher efficiency and reproducibility.

Summary Pollen grain size in many genera positively correlated with chromosome number. In this study, oat (Avena) pollen grain size was examined in diploids (2×=14, one species, for accessions), tetraploids (4×=28, five species, 20 accessions), hexaploids (6×=42, one species, eight cultivars), and in 10 octoploid (8×=56) accessions. Mature anthers ready to dehisce pollen were sampled from one to six plants per accession, and pollen grains were squeezed out of the anther with tweezers. Oat pollen grains are slightly elliptical, and the length of the major axis was found to be highly correlated with the minor axis width (r=0.94**). Pollen grain length, 39.3 m for diploids, 41.3 m for tetraploids, 47.0 m for hexaploids, and 48.8 m for octoploids, was positively correlated (r=0.86**) with ploidy level. No genomic or species effects appeared to influence this trait. Only tetraploidAvena vaviloviana accession PI 412767 produced two distinct class sizes of pollen grains, 99% normal (43.0 m) and 1% large (52.7 m). 相似文献

Summary Three populations of oats, each with 790 lines, were derived from CI 7555: (a) one (M population) consisted of M2-derived lines obtained from EMS treatment of naked seeds, (b) one (O population) consisted of F2-derived lines from crosses of M1 with check plants, and (c) one (C population) consisted of check lines.About 98% of the grain yield (GYD) variation in each population was due to variation in growth rate (GR) and harvest index (HI).There was greater variation for both GR and HI in M and O than in the C population, showing that mutations were induced for both traits. Summary Improved grain yields in lines of oats from matings of Avena sativa x A. sterilis were found to be due to increased plant growth rate. Growth rates of oats were quantitatively inherited, with the minimum number of effective factor pairs segregating in the interspecific matings ranging from 3 to 9. Heritability values for this trait averaged 0.4. Growth rate was highly and positively correlated with bundle weight, straw yield, grain yield, and unit straw weight, but it was uncorrelated with heading date and harvest index. Correlations with plant height were low. Complementary genes for purple grain colour Pp1, Pp2, Pp3 (now designated Pp1, Pp3b, Pp3a, respectively) were mapped using crosses between purple-grained hexaploid wheats ‘Purple Feed’ – Pp1Pp1/Pp2Pp2 (Pp1Pp1/Pp3bPp3b), ‘Purple’ – Pp1Pp1/Pp3Pp3 (Pp1Pp1/Pp3aPp3a) with non-purple-grained cultivars ‘Novosibirskaya 67’ (‘N67’) and ‘Saratovskaya 29’ (‘S29’). The genes Pp2 (Pp3b) and Pp3 (Pp3a) were inherited as monofactorial dominant when purple-grained wheats were crossed to ‘N67’. Both were mapped in the centromeric region of the chromosome 2A. Therefore, they were suggested being different alleles at the same locus and designated Pp3a and Pp3b. In the crosses between purple-grained wheats and ‘S29’ a segregation ratio of 9 (purple) to 7 (non purple) was obtained suggesting a complementary interaction of two dominant genes, Pp1 and Pp3. To map Pp1 as a single gene, the influence of the other Pp gene was taken into consideration by determining the Pp3 genotype of the F2 plants. The gene was mapped on chromosome 7BL, about 24 cM distal to the centromere. The Pp1gene was shown to be non allelic to the Rc-1 (red coleoptile) and Pc (purple culm) genes, contrary to what was previously suggested. The colouration caused by the Pp genes has no effect on pre-harvest sprouting. 相似文献

Summary The potential for breeding for high groat-oil content in oats was investigated by (a) conducting generation means analyses on data from three matings among adapted Avena sativa L. cultivars, (b) practicing one cycle of phenotypic recurrent selection in a segregating population derived from eight species backcrosses (Avena sativa x (A. sativa x A. sterilis)) among 24 parents, and (c) identifying transgressive segregates from interspecific (A. sativa x A. sterilis) matings.Additive gene action was the most important component in explaining the variation among generation means for groat-oil content. Dominance and epistatic interactions involving dominance were not significant in any mating. Significant residual genetic variation occurred in one mating, even after additive, dominance, and three digenic interactions were fitted. The importance of additive genes action implies that desired allelic combinations for high groat-oil content can be obtained in pure-line cultivars.One cycle of phenotypic recurrent selection using single plants as the selection units resulted in a genetic gain of 1.7 to 2.1% in groat-oil content. Individual plants selected for initiating the second cycle had from 9.5 to 12.6% groat oil.Over all 12 interspecific matings, the F2 progeny means were similar to the midparent values. Only two were significantly deviant. Summary Eight oat cultivars and experimental lines from four germplasm sources were crossed in a diallel mating design without reciprocals. F1 heterosis for grain yield was evaluated in two experiments, and 48 F2-derived lines from each of the 28 matings were evaluated for bundle weight, grain yield, straw yield, harvest index, height, and heading date in two experiments. Number of transgressive segregates per trait and generalized genetic variance were calculated for each mating. Genealogical distance for each mating was obtained by using coefficient of kinship based on the pedigree of the parents. The relationship between genealogical distance and the three types of breeding behaviour was examined via correlation and regression. Significant correlations occurred only for genealogical distance with numbers of transgressive segregates for height and with generalized genetic variances. Both were positive. Significant heterosis was observed for matings of more distantly related parents. Regressions on genealogical distance, when significant, were linear. Summary Test weight, seed weight, and groat percentage are three common measures of grain quality for oat (Avena sativa L.). There is considerable disagreement, however, about the utility of each of these traits as measures of grain quality, so it is important to understand the genetic interrelations among them and between them and other agronomic traits. In this study, 50 random F2-derived F3 lines from each of 13 biparental oat crosses were evaluated. Genotypic variances, heritabilities, and genotypic correlations for test weight (TW), seed weight (SW), groat percentage (GP), grain yield (GY), harvest index (HI), plant height (PH), and date of heading (DH) were used to predict and compare direct and indirect gains from single-trait selection. Direct selection was always the most effective method for improving single traits; average predicted improvement of population means was 4% for TW, 10% for SW, 1% for GP, and 14% for GY when 10% selection intensity was applied. Genotypic correlation coefficients, averaged for all crosses, were 0.04 for TW with SW, 0.37 for TW with GP, -0.09 for SW with GP, and 0.29, 0.36, and 0.30 for GY with TW, SW, and GP, respectively. Thus, direct selection for TW, SW, or GP should not greatly affect the other two grain quality indicators. Summary Factors favouring callus proliferation and subsequent regeneration of plants from pollen grains of rice anthers (Oryza sativa L., cvx. Bahia, Girona, Balilla × Sollana and Sequial) were determined. Cultivar differences in response were found, such as a high rate of haploid plant regeneration.In addition, isolated pollen grain culture was used to induce tissue proliferation outside the anther walls. The frequency of callus formation from isolated pollen grains was very low. It was necessary to preculture the anthers before pollen grain separation, in order to accomplish a successful development later. Summary A study of derivatives from the hybrid Avena sativa × A. magna backcrossed to A. sativa clearly indicate that it is possible to introduce characters of the wild oat into the cultivated oat. The genotypes of Avena sativa were crossed to A. magna Murph. et Terrell, from the sterile pentapliod F1 amphiploids were successfully developed after colchicine treatment. The C1 plants were observed for their morphology, cytology and fertility. These plants were characterized by longer and broader glumes, swollen pedicel attachment, pubescent lemmas, bold seeds and persistent spikelets. Other characters, viz., final plant height, rachis length, peduncle length and spikelets per panicle were intermediate. All plants hail the chromosome number 2n = 70 or close to. 7C. Due to stickiness of chromosomes, meiosis of only two decaploid plants could be studied in detail. The average chromosome Association was 1.3 I + 28.56 II + 1.4 III + 1.77 IV + 0.07 VI and 0.13 I + 28.7 II + 0.14 III + 2.09 IV + 0.59 VI; but in some cells up to 35 bivalents were observed. Summary Thirty-five rice (Oryza sativa L.) varieties, including 18 japonica, 5 javanica and 12 indica subspecies and 12 lettuce (Lactuca sativa L.) varieties were identified taxonomically, using PCR with originally designed 21 RAPD (Random Amplified Polymorphic DNA) primers and 8 sequence-specific primers, used for amplifying four specific DNA fragments. Use of these primers revealed polymorphisms among varieties in rice and lettuce and facilitates DNA fingerprinting. Dendrograms of both species based on polymorphisms were constructed and genetical relationships were established. In rice, half the number of amplified bands were polymorphic and almost all varieties differentiated. However, differentiation of minor genetic alterations among somaclonal variants or mutants and their mother varieties was not feasible. In L. sativa, 47% of the amplified fragments were polymorphic and all 12 varieties were differentiated. Some of the PCR fragments were variety or type specific, which could be used for indicators for type-selection. The dendrogram obtained showed differentiated clusters of crisphead, leaf and butterhead type, findings in good accord with the classification based on the genetic background. 相似文献

β-葡聚糖是燕麦发挥保健作用的重要功能因子,分离其合成关键基因有助于解析燕麦β-葡聚糖积累的分子机制。以高β-葡聚糖燕麦地方品种夏莜麦为材料, 通过RACE和染色体步移的方法克隆了燕麦β-葡聚糖合酶基因AsCSLH及其基因组序列并分析其序列结构特征,采用半定量RT-PCR方法研究该基因组织表达特性。分离出的燕麦CSLH基因包含6个内含子, 内含子剪接方式均符合GT/AG剪接规则,编码区全长2 277 bp,在序列上与水稻CSLH基因的编码区序列相似性最高并且所预测的编码蛋白含有相关多糖合成酶保守结构域。AsCSLH基因在燕麦各组织中均有表达,在灌浆期籽粒中表达水平最高。推测AsCSLH基因在燕麦籽粒的β-葡聚糖合成中起重要作用。 相似文献
