Spermatozoa and developing spermatids showing neck region abnormalities have been studied in material from 2 genetically related boars. In both boars the defects were abaxial implantation of the tails and lack of substance in the neck region. In many spermatozoa, a too wide space between the capitulum and the basal plate was more pronounced in epididymal spermatozoa compaired to testicular material. This implies that the defect aggravated, and might be connected with the migration of the cytoplasmic droplet in the epididymis. Since the defects were observed in spermatids, it its concluded that the defects were heriditary. This conclusion was further supported by the observation of similar defects in 6 other related boars, examined by light microscopy only.  相似文献

This study investigates the association of semen traits with boar fertility. The fertility outcome (farrowing rate – FR and total piglets born – TB) of 14 boars was obtained from a field trial conducted during 10 week of breeding period on a commercial farm using multiparous sows (n = 948) through single‐sire mating with 2 × 109 motile sperm cells per artificial insemination (AI) dose. Sperm motion parameters, evaluated with computer‐assisted semen analysis system in raw and stored semen at 17°C for 240 h, in addition to morphological sperm defects, measured on the collection day, were included in the analysis to determine which semen traits were important to discriminate the fertility potential of ejaculates from these boars. The data underwent multivariate cluster, canonical and discriminant analyses. Four clusters of boars were formed based on fertility outcome. One boar, with the lowest FR and TB values (89.7% and 11.98), and two boars, with the highest FR and TB values (97.8% and 14.16), were placed in different clusters. The other boars were separated in two distinct clusters (four and seven boars), including boars with intermediate TB (12.64 and 13.22) but divergent values for FR (95.9% vs 91.8%). Semen traits with higher discriminatory power included total motility, progressive motility, amplitude of lateral head displacement and cytoplasmatic droplets. Through multivariate discriminant analysis, more than 80% of the 140 ejaculates were correctly classified into their own group, showing that this analysis may be an efficient statistical tool to improve the discrimination of potential fertility of boars. Nevertheless, the validation of the relationship between fertility and semen traits using this statistical approach needs to be performed on a larger number of farms and with a greater number of boars.  相似文献

Sperm morphology and the fertilizing capacity of ejaculated spermatozoa were examined in 6 Swedish Landrace boars before and after heat stress. The boars were exposed to 35° C during 100 h in a climatic room. Fertility was measured by insemination of gilts before and at various times after heat stress. Each gilt (n = 44) was inseminated with a total of 5×109 spermatozoa diluted to 10O ml with EDTA-glucose diluent and fertilization was assessed by examining recovered ova 2 days after insemination.Changes in semen quality varied among the boars from a very weak response in 2 boars to pronounced semen alterations occurring 2–6 weeks after heat stress in the other boars. A close relationship was found between seminal changes and fertilization rates, all ejaculates which had high fertilization rates being of the same quality as the pre-exposure ejaculates. The ejaculates that had poor fertility were characterized by lowered sperm motility and increased numbers of spermatozoa with abnormal heads, proximal cytoplasmic droplets and nuclear pouch formations.  相似文献

Sperm of 28 boars of the Landrace breed was evaluated with the help of the thermoresistance test (TRT). The principle of the TRT is that the sample of fresh sperm is left in a 38 degrees C bath for 120 min and after that time the activity of the sperm is evaluated. 308 ejaculates were evaluated in this way and 1342 sows were inseminated by these ejaculates after a short storage. The highest conception rate was stated after insemination with the sperm whose activity was above 50% at the end of the TRT. 366 of 400 sows became pregnant, i.e. 82.43%, whereas out of 370 sows inseminated with sperm whose activity was 0-30% at the end of the TRT only 269 conceived, i.e. 72.70%; difference was statistically highly significant. No statistically significant difference was recorded between the fertility (number of piglets per one litter) and the TRT results. A significant dependence was observed between the sperm survival after 72 hours and conception (P less than 0.05). The same significant relation was found between survival within 72 hours and TRT in fresh ejaculates. This justified the recommendation to introduce the TRT among the test used for evaluation of the quality of fresh boar ejaculate.  相似文献

The aim of this study was to examine the incidence of sperm agglutinations and their relationships with sperm quality and fertility. Semen samples of 40 boars of an AI-station were investigated. Nineteen of the 40 investigated boars showed a constantly low (< 10% agglutinated sperm), 3 an intermediate (10-20%) and 6 boars a high level (> 20%) of agglutination in raw semen. The degree of agglutination in sperm samples of 12 boars varied distinctly during the investigation period. During summer more (P < 0.05) agglutinated sperm were observed (11.0 +/- 11.6%) than during winter (6.2 +/- 7.3%). There was no association between bacterial contamination and incidence of agglutinations (P > 0.05). After dilution in extender the percentage of agglutinated sperm decreased from 6.2 +/- 7.3% to 1.1 +/- 1.4% (P < 0.0001). Twenty-four hours after dilution the percentage of progressively motile sperm was 7.4% lower (P < 0.05) in ejaculates with an initially high degree of agglutination (> 20% agglutinated sperm) compared to samples with an initially low degree of agglutinated sperm (< 10%). Plasma membrane integrity, mitochondrial membrane potential, acrosome reaction and chromatin structure were independent (P > 0.05) from the level of agglutination. Fertility data did not differ (P > 0.05) between boars with low and high numbers of agglutinated sperm in raw semen. The results show that there are individual, ejaculatory and seasonal variations in the incidence and degree of agglutination. Agglutinations have a negative effect on motility of sperm and disappear to a large extent after dilution in sperm extender. They have no negative consequences on fertility.  相似文献

The objective of the present research was to determine the effect of increasing and decreasing natural photoperiods on selected parameters of boar ejaculates. The study material consisted of 17 boars: six Polish Large White (PLW) breed, five Polish Landrace (PL) breed, and six Duroc×Pietrain (D×P) crossbreed, all aged between 8 and 12 months at the beginning of the research. Analyses were conducted on 612 ejaculates, which were collected in two experimental periods: an increasing photoperiod (IP) (January–June) and a decreasing photoperiod (DP) (July–December). A statistically proven impact of photoperiod on the volume of semen was observed in all the studied breeds (P=0.004). During the decreasing photoperiod the mean volume of semen was 261.16±75.20 ml and this was almost 17 ml higher than that for the increasing period. For boars involved in the experiment, day length also had a significant impact on the total number of motile spermatozoa (P=0.037). In the increasing photoperiod the mean number was 3.26×109 lower. A decreasing photoperiod has a positive affect on both boars and the parameters of the collected ejaculates, which were observed in the higher number of insemination doses per ejaculate. Between the different breeds, the reactions of boars to photoperiod differed and the most significant influence of photoperiod on semen parameters was noted among D×P breed boars. Least susceptible to changes in day length were PLW breed boars.  相似文献

The possibility of selecting boars for deep freezing of spermatozoa was evaluated by tests of frozen-thawed spermatozoa. 20 of 31 ejaculates were used for artificial insemination of 37 gilts. Boar seminal plasma and OLEP were used as thawing diluents. The thermoresistance test and the extracellular concentration of aspartate aminotransferase (ASAT) were used as indicators of fertility. Spermatozoa from 4 boars were utilized. 1 of the boars appeared to be of superior fertility, while the spermatozoa from another was infertile. The latter animal had been used for fresh artificial insemination trials, and had shown higher pregnancy rates than the average. Samples from this boar exhibited the lowest degree of motility after 3 hours storage at 37 degrees C, the greatest relative decrease of motility during the thermoresistance test, the greatest release of ASAT after thawing by OLEP, and greatest relative release of ASAT. These variations were statistically significant (p less than .05). The results show that there is a possibility of detecting boars with spermatozoa with potential low freezability.  相似文献

The prevalence of abaxial sperm tails, accessory tails and double tails, was determined from semen evaluation records of 600 western Canadian range bulls and 449 bulls in Canadian artificial insemination centers. Spermatozoa with abaxial tails were produced by 10.5% of bulls, however, only 0.48% produced sperm with greater than 50% abaxial tails, and 0.86% of the 1049 bulls produced sperm with accessory and double tails. Three experiments were done to determine the importance of abaxial sperm tails to fertility. In experiments 1 and 2, frozen semen with 88%, 50% and 0% abaxial tails was used to inseminate synchronized heifers, which were allowed to develop pregnancy, and superovulated heifers, which were slaughtered prior to embryo recovery seven days after breeding. In experiment 3, a bull which produced 100% abaxial sperm was used in a competitive mating situation with three control bulls in a herd of 118 cows. The combined results of these experiments indicate that spermatozoa with abaxial tail attachment fertilize ova at a normal rate and are not associated with any increase in embryonic death.  相似文献

The aim of the study was to determine the relation between the semen quality, frequency of sperm defects, sperm dimensions and shape, and the ejaculate volume of Large White and Landrace boars. A total of 648 ejaculates collected from 31 Large White and 30 Landrace boars were divided into three groups according to the criterion of the ejaculate volume. In this study Landrace boars produced ejaculates with higher volume, sperm concentration, and total numbers of spermatozoa than Large White boars. Landrace boars also showed a lower frequency of sperm with morphological abnormalities (P < 0.05). Landrace boars sperm had larger heads, which were by 0.15 μm longer, and by a larger perimeter and area (P < 0.05). Landrace boar spermatozoa also had a longer flagellum and were generally larger and by 2.07 μm longer than Large White boar sperm (P < 0.05). Significant differences were also found in the shape of sperm of the two breeds (P < 0.05). Landrace boars sperm had more elongated heads, and the ratio of head size to flagellum length was lower than in Large White boars sperm (P < 0.05). Sperm from ejaculates with low volume had a shorter flagellum and a greater head length/flagellum length ratio than sperm from medium- and high-volume ejaculates (P < 0.05).  相似文献

Factors influencing boar sperm cryosurvival   总被引:1,自引:0,他引:1
Optimal sperm cryopreservation is a prerequisite for the sustainable commercial application of frozen-thawed boar semen for AI. Three experiments were performed to identify factors influencing variability of postthaw sperm survival among 464 boar ejaculates. Sperm-rich ejaculate fractions were cryopre-served using a standard freezing-thawing procedure for 0.5-mL plastic straws and computer-controlled freezing equipment. Postthaw sperm motility (assessed with a computer-assisted semen analysis system) and viability (simultaneously probed by flow cytometry analysis after triple-fluorescent stain), evaluated 30 and 150 min postthaw, were used to estimate the success of cryopreservation. In the first experiment, 168 unselected ejaculates (1 ejaculate/boar), from boars of 6 breeds with a wide age range (8 to 48 mo), were cryopreserved over a 12-mo period to evaluate the predictive value of boar (breed and age), semen collection, transport variables (season of ejaculate collection, interval between collections, and ejaculate temperature exposure), initial semen traits, and sperm quality before freezing on sperm survival after freezing-thawing. In Exp. 2, 4 ejaculates from each of 29 boars, preselected according to their initial semen traits and sperm quality before freezing, were collected and frozen over a 6-mo period to evaluate the influence of interboar and intraboar ejaculate variability in the survival of sperm after cryopreservation. In Exp. 3, 12 ejaculates preselected as for Exp. 2, from each of 15 boars with known good sperm cryosurvival, were collected and frozen over a 12-mo period to estimate the sustainability of sperm cryosurvival between ejaculates over time. Boar and semen collection and transport variables were not predictive of sperm cryosurvival among ejaculates. Initial semen traits and sperm quality variables observed before freezing explained 23.2 and 10.9%, respectively, of the variation in postthaw sperm motility and viability. However, more that 70% of total variance observed in postthaw sperm quality variables among ejaculates was explained by boar. This indicates that boar is the most important (P < 0.001) factor explaining the variability among ejaculates in sperm cryosurvival, with most (14 of the 15 boars in Exp. 3) showing consistent (P > 0.05) sperm cryosurvival over time.  相似文献

A porcine in vitro fertilization (IVF) system and seminal quality parameters of frozen–thawed boar semen were used to assess the effectiveness of two different thawing rates of frozen boar semen, and to address the question of whether differences between fertility of ejaculates could be predicted in a limited field trial. In the first experiment, two thawing procedures were analysed (37°C, 30 s; 50°C, 12 s) and no differences in sperm quality were found. However, when the procedure was 50°C, 12 s the IVF results showed a higher number of sperm per penetrated oocyte and a near 10 points higher rate of pronuclear formation. In the second experiment, the fertility results obtained in the limited field trial show to be efficient enough for application in a commercial use, especially for three of the employed boars (fertility ≥80%). In this limited study, the conventional seminal parameters are not accurate enough to discriminate good and bad boars in relation to fertility. On the contrary, parameters of in vitro penetrability are more precise to predict subsequent fertilities. As conclusion, the IVF fertilization system seems to be a good tool to evaluate the quality of frozen–thawed boar semen previous to its commercial way, to verify the bank semen storage quality and a good way to assay new sperm freezing procedures, as it is the more precise evaluating method in estimating the potential fertilizing ability.  相似文献

Several laboratory assays have been designed to assess the fertility potential of a semen sample before insemination, but none have been consistent and accurate predictors of fertility. To determine whether zona-binding ability may be a useful fertility predictor, we validated and used an in vitro competitive assay to measure the ability of porcine sperm to bind to the zona pellucida. The zona-binding ability of sperm from 11 boars that exhibited a broad range in average litter size and farrowing rate was determined. Sperm from each boar were compared directly with sperm from eight other boars in a systematic, pairwise fashion. Sperm from two semen samples were labeled with fluorophores at concentrations that did not affect motility or zona-binding ability. An equal number of labeled sperm from each boar was coincubated with homologous oocytes. Least squares means from analysis of variance were used to rank boars based on zona-binding ability. The competitive assay was effective in establishing a ranking of the boars (R2 = 0.62). Furthermore, there was a correlation between zona-binding ability and fertility when estimated by average litter size (r = 0.64, P < 0.05) but not when estimated by farrowing rate (r = -0.28). The explanation for this difference was that litter size and farrowing rate were poorly correlated (r = 0.14). In conclusion, a competitive zona-binding assay distinguished boars that sired either small or large litters. Competitive zona-binding ability may be useful for identifying boars with reduced fertility that produce smaller litters following insemination.  相似文献

Contents Determination of the osmotic resistance of boar spermatozoa and the relations to sperm freezing and long storage The osmotic behaviour of fresh boar spermatozoa (482 ejaculates from 34 A.I. boars) was examined in a standard medium but with different osmotic pressures ranging from 150 mosm/kg till 500 mosm/kg. The percentage of spermatozoa with normal acrosomal ridge (NAR) was determined after incubation of 0.2 ml fresh semen in 3 ml of the osmotic test solutions at 39° C for 15 and 120 min. In the non-isotonic media % NAR decreased and could be quantified depending to incubation time and to the different osmolalities; NAR decreased strongest at 150 or 500 mosm/kg and after 120 min incubation time. Differences in % NAR were large between ejaculates and between boars but low within boars and statistically significant correlations had been found to % NAR and motility in froren/thawed semen samples or stored samples. Still higher correlations were found by calculation of an osmotic resistance test-value (ORT-value) which is defined as follows: ORT = 1/2 % NAR in isotonic medium/(after 15 min) +% NAR in 150 or 500 mosm/kg/(after 15 or 120 min) ORT-values can be calculated for hypotonic (150 mosm/kg) as well as for hypertonic (500 mosm/kg) test solutions and for 15 and 120 minutes incubation time. The hypotonic ORT-value for 120 min. is the most accurate; the values varied from 34 till 83 between ejaculates and from 44 till 76 between boars with a low variability within boars. Grouping of boars by their ORT-values in “good”, “moderate” and “poor” corresponded to a similar grouping of these boars by their semen quality (% NAR and motility) in preserved semen samples. Also a grouping of single ejaculates by their ORT-values corresponded to a similar qualitative classification of these ejaculates. ORT seems to become a good and practicable assessment test for the selection of boars suitable dor deep freezing or long storage of their spermatozoa. On the other hand preservation techniques too can be examined by ORT. But more investigations with this test must be done especially its relation to the fertilization capacity of semen samples with different ORT-values.  相似文献

This work aimed to establish whether the temperature humidity index (THI) under different intertropical zones affects the retention of cytoplasmic droplets (CDs), sperm function and DNA integrity in boars. With this purpose, two separate studies were devised. In the first one, 49 boars from six farms were collected every 45 days (230 ejaculates). THI were measured daily, and sperm parameters were evaluated. Boars were classified into three groups based on the incidence of ejaculates having more than 25% spermatozoa with CDs: persistent (at least three consecutive ejaculates), moderate (less than three ejaculates) and absent (no ejaculate having ≥25% spermatozoa with CDs). Farms were classified based on THI through cluster analysis into two groups. In the second study, 32 liquid-stored semen samples were classified based on three cluster analysis: low and high incidence of proximal (PCDs), distal (DCDs) CDs and a combination of PCD and DCDs. high THI farms presented significantly (p < .05) higher proportions of boars with moderate and persistent incidence of CD than those with low THI. In study 2, the presence of PCDs was negatively correlated with sperm DNA integrity (r = −0.691; p < .01). However, differences between groups were more apparent when ejaculates were classified based on both PCDs and DCDs than when PCDs or DCDs were considered separately. In conclusion, classification of boars according to the severity and persistence of CDs in boars allows understanding more clearly the dynamics of CD retention and the effects of ambient temperature and relative humidity. Additionally, the joint analysis of both PCDs and DCDs is necessary in routine sperm quality analyses.  相似文献

The aim of this study was to address the question of whether differences in farrowing rate and litter size after the use of different ejaculates could be predicted using the standard semen parameters under commercial conditions. In this study, a total of 1818 sows were used to evaluate the fertility predictive value of different sperm parameters. Logistic regression analysis (univariate and multivariate) was used to correlate the dichotomous farrowing rate data to the sperm parameters. Linear regression was also used to determine the relationship between litter size and semen parameters (Pearson correlation and multiple regression). Receiver-operating curves (ROC) were used to determine the overall performance characteristics of each semen variable in the logistic regression model. Semen analysis, under commercial conditions, allows to identify ejaculates with very low fertility potential but the pre-selection of the samples, the high number of sperm per doses and the high quality of the semen used in artificial insemination (AI) programmes reduces the variability. Therefore, it is unlikely to detect fertility differences associated with seminal parameters.  相似文献

The aim of the present study was to determine whether the levels of reactive oxygen species (ROS) substances production and the levels of lipid peroxidation of the sperm membrane were related to the quality that the ejaculates exhibited after cryopreservation in boars. Ejaculates from 42 healthy boars were used in this study and they were cryopreserved with the lactose‐egg yolk extender (LEY). Several sperm quality parameters were assessed by flow cytometry in samples incubated for 30 and 150 min at 37°C after thawing: the percentage of sperm with intact plasma membrane (SIPM), intracellular reactive oxygen substances production through mean of DCF fluorescence intensity of total sperm (mean‐DCF) and the percentage of viable and non‐viable sperm containing oxidized BODIPY (VSOB and NVSOB). In addition, the percentages of total motile (TMS) and progressively motile sperm (PMS) were assessed at the same incubation times with a computer‐assisted sperm analysis system. The classification of the ejaculates into good or bad freezers was performed through hierarchical cluster analysis from SIPM and TMS at 150 min post‐thawing. The ejaculates of those males classified as good freezers exhibited higher (p < 0.05) SPIM, TMS and PMS than the bad freezers, although both groups presented similar (p > 0.05) VSOB, NVSOB and mean‐DCF. Therefore, these results show that lipid peroxidation and the amount of reactive oxygen substances in the sperm after cryopreservation are similar between boars classified as good or bad freezers.  相似文献

The consequences of the use of swirl assessment at a bovine artificial breeding centre were studied. Data relating to the amount of semen discarded, fertility of accepted semen, the relationship between discard rates and age of bull and ejaculate number have been presented. The overall differences in fertility between semen with swirl ratings, 5, 6 and 7 were very small. About 50% of all ejaculates collected in the period considered were discarded at the first inspection. The discard rate tended to be greater in young and in older bulls. The discard rate was significantly greater (P less than or greater to 0.001) for second ejaculates. It was considered that a proportion of rejected semen may have been of good fertility. Semen wastage after freezing was low.  相似文献

This study was conducted to investigate the predictive capacity of fertility and litter size of sperm head morphometric measurements when the ejaculates fulfilled the minimum requirements commonly used in artificial insemination (AI). Semen samples from 11 rabbits (77 ejaculates) were evaluated for sperm motility, abnormal spermatozoa and sperm head morphometry using computer automated sperm analysis system. Morphometric dimensions for length, width, area and perimeter were analysed. Only ejaculates with more than 70% of motility rate and <15% of abnormal sperm were used for AI. A total of 1031 individual AI were performed in commercial rabbitries. Our results showed significant differences among animals for all sperm head measurements. The mean values for fertility and litter size obtained were 68.4 ± 0.01% and 9.3 ± 0.1% respectively. To assess the predictive value of morphometric dimensions in fertility, a logistic regression analysis was applied. Moreover, multiple linear regression analyses were used to examine the relationship between litter size and sperm head morphometric parameters. Logistic regression analysis rendered a significant model between fertility and area and perimeter, explaining the 0.65% variation. Multiple linear regression analysis rendered a significant model between litter size and width, area and perimeter that explained the 1.3% variation. By conclusion, the sperm head morphometric parameters assay showed low potential to predict fertility and litter size when the ejaculates fulfilled the minimum requirements commonly used in AI (motility and abnormal spermatozoa) in rabbit.  相似文献

This study was performed to determine the relationship between selenium (Se) content and Se-dependent glutathione peroxidase (GSH-Px) activity in blood and semen, and semen quality of boars at different age used in an artificial insemination (AI) station. Routine macroscopic and microscopic analyses of semen quality were accompanied by measurement of Se content and GSH-Px activity in blood and semen. The Se concentration in blood plasma, seminal fluid and spermatozoa was measured by fluorometric method, the GSH-Px activity by a method based on NADPH-coupled reaction. A total of 155 ejaculates and 58 blood samples were investigated. The results of this study showed that there was no direct relationship between the Se content and GSH-Px activity in blood plasma and semen, and semen quality of sexual matured boars. The mechanisms controlling Se content and GSH-Px activity in blood and semen seem to be independent. The age of boars as a differentiating factor for Se content and GSH-Px activity in blood and semen is possible. It is concluded that a determination of Se status and/or GSH-Px activity in organism before Se supplementation is indicated.

Contents Functional epididymal disorders in boars. In a total of 1100 routine sperm evaluations of different boars (mostly of the Belgian Landrace), ejaculates of 112 boars were affected by tail abnormalities of a varying degree (10–25 % abnormal tails in 33 boars, more than 25 % in the remainders, of which 10 boars with about 70 % abnormal tails). The most common defect is the hairpin curved sperm defect followed by the single bent tail, the partially coiled midpiece (pseudo-DAG defect) and coiled tails. Electronmicroscopical pictures (scanning and transmission) of the defect were similar with those, found in literature. Some factors, which possibly could induce the defect are discussed: zinc content in the sperm cell is significantly increased in boars which were affected in a high percentage compared to normal ones. Exhaustion tests in affected boars seemed not to improve sperm morphology while analogue tests in controls never provoked sperm tail abnormalities. Also, on the other hand low sexual activity (1 ejaculate a week) for several months did not decrease the percentage of abnormal tails in the ejaculate. In search for the active motility of the sperm cells inseminated into oestrous gilts, it was clear after slaughter, that morphologically normal sperm cells reached the tip of the uterine horn earlier than sperm cells with tail abnormalities. It was proved by postmortem examination of the genital tract of the affected boars, that the tail defects originated from the epididymides and not from the testes. Perhaps, the defect can be explained by the hypothesis that the main piece is reflected in the direction of the nucleus around the distal protoplasmadroplet, followed by fusion of the cell membranes.  相似文献
