In controlled experiments in an insect-free stable, cattle became infected with Parafilaria bovicola when Musca lusoria, infected with the larvae of this worm, were allowed to feed on a fresh skin incision, and when infective larvae were placed on fresh skin incisions, injected subcutaneously or into the jugular vein, or instilled into the eyes. The sites of blood spots caused by ovipositing P. bovicola females and the sites of carcass lesions were seldom close to the site of infection, an indication that the worms had migrated. The prepatent period of P. bovicola in 4 cattle which developed blood spots ranged from 242--319 days. Neither of the infected cattle that were kept continuously in a shady stable showed blood spots, but 4 out of 7 infected cattle which spent some time in the sun bled. However, carcass lesions on shaded cattle were similar in appearance to those on cattle kept outdoors. Infective larvae were stimulated to escape from the mouth-parts of infected M. lusoria and Musca xanthomelas s.s. when these were fed citrated ox blood warmed to 38--40 degrees C. No escape took place when the flies were fed warmed saline or warmed 15% sucrose solution.  相似文献

The possible adverse effect of arsenical tick control dips on Parafilaria bovicola infections was investigated in 48 artificially infected cattle. A treatment group of 24 cattle was dipped in a plunge dip containing 1600 ppm arsenic trioxide. A control group of the same size was dipped in an organophosphate dip containing a mixture of chlorfenvinphos and dioxathion. Regular weekly to 3-weekly dipping had no effect initially on the prevalence of ovipositional blood spots of P. bovicola in either group. However, from 4 months after bleeding commenced there was a significant reduction in blood spots in the arsenic-dipped cattle and, on slaughter at 12-14 months after infection, the arsenic group had significantly fewer live worms and fewer carcass lesions. Arsenic residues in muscle samples of treated cattle were 11.6 times higher than in the controls. It is proposed that arsenic residues in the sub-cutaneous muscle layers increase with repeated dipping until a level toxic to P. bovicola is finally reached. Older cattle would therefore be refractory to infection and their carcasses at slaughter would not be affected.  相似文献

Ivermectin treatment of all cattle on a badly infected farm failed to interrupt the transmission of P. bovicola, even though ovipositional blood spots were drastically reduced in numbers for an entire summer season following treatment. Regular weekly to fortnightly dipping of all cattle in 50 ppm deltamethrin immediately reduced vector fly numbers to less than 1 fly per cow face. Sustained dipping for 9 months effectively reduced P. bovicola transmission from approximately 50% to less than 2%. However, cessation of fly control led to a return to predipping P. bovicola infection levels. Ovipositional blood spot counts and the ELISA technique for evaluating P. bovicola infection in a herd were compared and were both effective methods. Best results for the blood spot method, however, are obtained in spring at the peak of the bleeding season whereas the ELISA method does not have this limitation.  相似文献

The filaricidal effects of trichlorphon, arsenic trioxide, sodium antimony biscathecol disulphonate and nitroxynil against Parafilaria bovicola were the subject of this investigation. Levamisole hydrochooride was retested in a separate trial to assess the time required for healing after successful treatment. A comparison of carcass lesions and the percentage of lesion area in untreated controls with those in treated animals reconfirmed the efficacy of levamisole hydrochlorice, while nitroxynil gave more promising results. Both lesions and lesion area were reduced by 76% by the former and by 93% by the latter compound. The other drugs only slightly suppressed P. bovicola activity. After treatment with levamisole hydrochloride, active lesions were still present for 1-4 weeks and visible lesions on slaughter for 8 weeks. Since the visible lesions during the 8 weeks varied from acute to chronic and as they disappeared 9 weeks post treatment, it is suggested that provision should be made for a healing period of at least 9 weeks.  相似文献

A study was conducted to determine if the purification of Parafilaria bovicola antigens can increase the specificity of serodiagnosis of parafilariasis in enzyme-linked immunosorbent assay. Antigens released from adult worms of P. bovicola were separated by chromatofocusing on a polybuffer exchanger of the pH range 7.3-4.0 Polypeptide analysis by sodium dodecyl sulphate polyacrylamide gel electrophoresis showed the presence of four major polypeptides with MWs of 41, 36, 24 and 20 kDa. Additional biochemical characterization identified the 24- and 20-kDa polypeptides as hydrophobic glycoproteins. The chromatofocusing purification procedures were also applied for separation of a whole-worm extract. Again, the 41- and 36-kDa antigens were identified in separate peak fractions. Using ELISA, it was shown that the 41- and 35-kDa antigens were recognized by bovine antibodies specific for P. bovicola, but not by other sera collected from cattle infected by Onchocerca gutturosa, Onchocerca lienalis, Ostertagia ostertagi and Dictyocaulus viviparus. The serological evaluation strongly suggests that the 41- and 36-kDa antigens are P. bovicola specific.  相似文献

Parafilariosis was first described in South Africa in 1964, thereafter being discovered at numerous localities in the country. When it became obvious that Parafilaria bovicola, for which no treatment was known, caused considerable economic losses, trials involving a series of compounds were conducted to identify candidate remedies. This paper describes an anthelmintic test for evaluating the efficacy of compounds for registration for field use. Recovery of Parafilaria worms is impractical for anthelmintic testing, and consequently the lesion sizes of treated and control groups of cattle are compared statistically, using appropriate statistical tests. The seasonal incidence of mature worm infection in cattle in South Africa is such that trials should commence after June and be completed before the end of January, allowing a lapse of 70 days between treatment and slaughter for resolution of the lesions. The presently available parafilaricidal compounds while of value for treating slaughter stock, when used alone will probably not be effective for control of infection in the field.  相似文献

More than 23 000 cattle of both sexes and different ages were examined for blood spots caused by egg-laying females of P. bovicola. Although these studies extended over four years and involved 5 farms in different parts of the Transvaal Bushveld, the overall results were the same. Ovipositional bleeding was strongly seasonal with blood spots first appearing in winter (June), reaching a peak in spring (September-November) and thereafter declining rapidly as summer progressed. In a single year at Zoutpan Research Station up to 92,1% of the 1st year heifers had already bled by November and this proportion increased only slightly to 95,1% by the end of the bleeding season (May). The number of blood spots per animal showed a similar seasonal abundance except for a second peak of abundance in June for 1st year heifers and oxen. The prevalence of blood spots in cattle of different ages and sex varied markedly. At Mara Research Station half as many oxen bled in their 2nd year as in their 1st year, while at Zoutpan 19,2% fewer heifers bled in their 2nd year than in their 1st. Bulls bled the most, then 1st year oxen, 1st and 2nd year heifers and 2nd year oxen, with breeding cows bleeding the least. A high female hormone level appears to be associated with the development of immunity. The shortest period from birth to 1st blood spot (the apparent prepatent period) was 191 days, while 81,8% of oxen bled for the 1st time within 279 days after birth.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Parafilaria bovicola was recorded in Swedish cattle for the first time in 1978. The parasite causes carcass lesions with a maximum incidence from February to July but already in the previous November the carcass lesions appear regularly in the form of yellow oedema. In February and onward the oedema may be extensive with a greenish-yellow colour. The lesions are most prominent in bulls and more or less negligible in cows. However, the cows are considered to be important reservoirs for the parasite from one season to the next. Bleedings are seen from March until August and the infective larvae are transmitted to the cattle from early June by the intermediate host Musca autumnalis. Parafilaria lesions look remarkably like those caused by contusions during handling and transport previous to slaughter. Eosinophile infiltrations can be regarded as a constant diagnostic feature of parafilariosis.  相似文献

A total of 20 375 flies collected off cattle on 12 farms over 36 months were identified and examined for 3rd stage P. bovicola. The 3 vector species accounted for 64.1% of the flies collected and were the only fly species found to be infected. Musca lusoria was clearly the dominant vector fly, although large numbers of Musca sp. A appeared regularly between February and April each year. This phenomenon, coupled with high numbers of M. lusoria throughout most of the year, led to an increase in the numbers of vector flies from their lowest level in June to a peak in February-April. Of the 13 070 vector flies examined for 3rd stage larvae only 64 (0.52%) were positive; of these 41 were M. lusoria and 17 Musca sp. A. No positive male flies were found. Incubation of wild-caught flies for up to 13 days at 27 degrees C noticeably increased the larval recovery rate. Flies were found to be infected mainly from August-March. Infected M. lusoria were recorded from July-March and infected Musca sp. A from January-May. Only 6 infected M. xanthomelas were collected and this was during the period August-December, when most ovipositional blood spots occur on cattle. It is concluded that P. bovicola transmission in the Bushveld is not correlated with peak periods of bleeding but rather with high numbers of vector flies, the various species augmenting each other so that transmission may take place almost throughout the year.  相似文献

An enzyme-linked immunosorbent assay (ELISA) for the detection of antibodies in bovine sera against Parafilaria bovicola nematodes was developed and its sensitivity was compared with the immunodiffusion (ID) method. An exoantigen of P. bovicola which was shown to contain four major polypeptides was used in these procedures. The serological reactivity of the antigen polypeptides was defined by using the enzyme-linked immunoelectrotransfer blot technique (EITB) and whole-worm extract proteins. It identified only four serologically reactive polypeptides with sera from one experimentally infected calf and a verified field case. These two positive sera reacted mainly with four major antigens which coincided in molecular weights of the polypeptides of the exoantigenic preparation, namely, 43, 39, 28 and 25 KDa. Calves experimentally infected with P. bovicola showed a positive reaction with ELISA at 4 months after inoculation, and after this period a rapid increase in serum antibody response occurred. In these cases the ID reaction was observed for the first time at 7 months after inoculation. The specificity of an ELISA method using crude exoantigen preparation of P. bovicola was tested for the diagnosis of bovine parafilariasis. No cross-reactivity was detected when the P. bovicola exoantigen preparation was tested against sera from calves experimentally infected with Onchocerca lienalis, as well as against the sera from cattle naturally infected with Dictyocaulus viviparus or from cattle chronically infected with Ostertagia ostertagi. In addition, testing of 740 field sera from cattle in areas non-endemic and endemic for P. bovicola indicated a specificity of the antigen preparation used. Forty sera from laboratory-confirmed field cases of P. bovicola infection were tested by ELISA and immunodiffusion. All of these sera were ELISA positive, whereas only 70% of these were positive in the ID test. Seven (2.1%) of 328 sera from 21 herds from non-endemic P. bovicola areas were ELISA positive, as opposed to none in the ID test. Of the 94 sera from six herds in areas endemic for P. bovicola infection, 51 (54%) were ELISA positive whereas only 24 (26%) were positive in the ID test. When 56 slaughtered cattle, with varying degrees of meat condemnations due to parafilariasis, were tested for P. bovicola specific antibody, 91% of the serum samples were positive by ELISA. These results suggest that the exoantigen of P. bovicola can be used in a sensitive and reliable serological detection of parafilariasis by ELISA.  相似文献

Thirty-two cattle were included in a study to confirm the efficacy of ivermectin administered at 200 μg kg−1 against Parafilaria bovicola and to determine the time required for resolution of the lesions caused by the parasite. Four treated and four control animals were slaughtered over 2 days starting 15, 30, 50 or 70 days after treatment. The number, distribution and surface area of carcass lesions, and the weight of tissue trimmed to render the carcasses aesthetically acceptable, were recorded and parasites were recovered from subcutaneous tissues.

Significantly (P < 0.01) fewer worms were recovered from ivermectin-treated cattle slaughtered 50 or 70 days after treatment than from controls. Reductions in the mean number and surface area of lesions, and in the weight of tissue trimmed, were statistically significant (P < 0.05) for the ivermectin-treated group slaughtered 70 days after treatment and approached significance (P < 0.1) for the ivermectin group slaughtered 50 days after treatment.  相似文献

The aim of this study was to identify the prevalence of pathological lesions in pigs from small‐scale farms and to determine associations between pathological lesions and hematological parameters, and carcass and meat quality in slaughtered pigs. The study was conducted on 625 pigs (~115 kg) originating from 20 small‐scale farms. Any signs of pneumonia, pleurisy, pericarditis, and liver milk spots were recorded as present or absent. Complete blood count was investigated. The following carcass quality parameters were measured: live, hot and cold carcass weights, cooling loss, dressing percentage, backfat thickness, and meatiness. Meat pH and temperature were measured 45 min postmortem. Of the 625 examined pigs, 41.8% had pneumonia, 23.5% pleurisy, 2.7% pericarditis, and 29.9% liver milk spots. The presence of pathological lesions in slaughtered pigs adversely affected hematological parameters, reduced live, hot and cold carcass weights, and meatiness and had deleterious effects on meat quality (higher pH45min and higher prevalence of dark, firm and dry meat). In conclusion, this study showed a high prevalence of pathological lesions in slaughtered pigs, indicating serious health problems in smallholder pig production systems. The presence of single and, especially, multiple pathological lesions in slaughtered pigs negatively affected hematological parameters, and carcass and meat quality.  相似文献

Five raccoon dogs (Nyctereutes procyonoides) and two domestic dogs (Canis familiaris) were subcutaneously infected with 100 infective larvae (L3) of Dirofilaria immitis. Two and five worms, respectively, were collected from two of three raccoon dogs. Villous endarteritis was found in the raccoon dog with five worms and two dogs at 116 days after infection. The number of recovered worms in the raccoon dogs was significantly smaller than that of the domestic dogs having 22 and 29 worms, while histopathological features and the severity of the lesions in the raccoon dogs were similar to those in the domestic dogs. The vascular lesions in two chronically-infected raccoon dogs turned into much severe at 565 and 590 days after inoculation.  相似文献

Thirteen mixed-breed beef bulls, 1 to 4 years old, were used to determine the effect of live and dead filarial nematodes, Setaria labiatopapillosa, placed in the vaginal cavity of the testes. When dead worms were used, granulomatous lesions developed on the tunica vaginalis parietalis in 7 of 8 testes. The lesions were similar to those seen in some clinical cases of periorchitis. Similar lesions developed in 5 of 6 testes after live worms were implanted in the vaginal cavity of the testes and tetramisole (8 mg/kg) was injected subcutaneously 6 days after implantation. When live worms were implanted and tetramisole was not given, lesions developed in 3 of 6 testes. It was concluded that the granulomatous reaction was a local response to dead or killed S labiatopapillosa.  相似文献

The aim of this study was to compare a split marketing (SM) strategy, in which the heaviest pigs in a group are removed and slaughtered earlier than the others, with an all-out (AO) marketing strategy, in which all pigs are removed from the pen simultaneously and slaughtered on the same day, in terms of welfare, performance, and carcass traits of noncastrated (i.e., intact) male and female pigs. The experimental treatments were arranged in a 2 × 2 factorial array with 1) marketing strategy (SM vs. AO) and 2) sex (males vs. females), which yielded 4 treatment groups of 14 pigs (73.1 ± 4.8 kg): male SM, male AO, female SM, and female AO (7 replicates/group). Pigs in AO groups were all slaughtered after 6 wk on trial, whereas in SM groups the 3 heaviest pigs were removed and slaughtered 2 wk before the remainder of the group, which were slaughtered at the same time as the AO pigs. Pigs were fed a liquid diet from a long trough 3 times daily. Behavioral observations were conducted before and after SM, the day of SM, and 1 and 2 wk later. Behavior was recorded both during and between feed events, and skin lesions were scored on all, except the 3 pigs removed from SM groups before and 2 wk after SM. Growth performance, feed efficiency, and carcass traits were recorded. The number of aggressive interactions during feed events decreased after the 3 pigs were removed from SM groups. This reduction in aggressive interactions was observed on the day of SM in male groups (before SM: 24.3 vs. the day of SM: 14.7, SED = 3.31, P < 0.05 for interaction) and in subsequent observations in female groups (before SM: 21.4 vs. days after SM: 13.4, SED = 3.31, P < 0.05 for interaction). However, SM had no effect on behaviors recorded between feed events or on the number and severity of skin lesions (P > 0.10). There were no differences between the 11 remaining pigs in SM groups and the 14 pigs in AO groups in terms of growth performance, feed efficiency, and carcass traits of female or intact male pigs (P > 0.10). However, reduced within-pen CV in carcass weight was detected in pigs from SM groups compared with pigs from AO groups (8.6 vs. 10.9, SEM = 0.72, P < 0.05). Therefore, in restrictively fed pigs, a SM strategy improved the welfare of both female and intact male pigs by reducing aggressive interactions during feeding but had no effect on performance or carcass traits.  相似文献

Pups of Nigerian mongrel dogs randomly bought from two village markets for experimental purposes at the University of Nigeria, Nsukka, were observed to be ill within 4 days of purchase. By 13 days after purchase 52.5 per cent of all the animals had died. All sick animals were anorectous and suffered from diarrhoea. The faeces were mostly dark yellow to light brown and a few contained spots of blood. Few animals responded to treatment with fluids, antibiotics and vitamins and death occurred within 24 hours of the onset of clinical signs. In most cases, post mortem and histopathological lesions were similar to those characteristic of canine parvovirus. The sera of the surviving animals tested for haemagglutination-inhibition antibodies to canine parvovirus, using porcine red blood cells, were positive.  相似文献

The lateral transmission of Histomonas meleagridis in turkeys was studied in floor pens without the presence of Heterakis gallinarum. Battery-reared poults (120) were transferred at 2 wk of age to concrete-floored floor pens with fresh pine shavings litter (40/group). One group received no exposure. In other groups, either 10% or 25% of the birds were inoculated per cloaca with cultured H. meleagridis (200,000/bird) and placed in the pens as seeder birds. Inoculated birds died at 10-18 days postinfection (PI) showing typical liver and cecal lesions of histomoniasis. Birds in the high-exposure group died of histomoniasis beginning 16 days PI and continuing to 100% mortality by day 23 PI. Birds in the low-exposure (LE) group died beginning on day 19 PI and continuing through day 31 PI. All but one LE bird alive on day 31 PI had severe liver and cecal lesions of histomoniasis at necropsy. There was no evidence of histomoniasis in unexposed birds. No cecal worms (H. gallinarum) were found at necropsy of dead birds or in unexposed birds at the end of the experiment. Even though H. gallinarum is the only known reservoir for H. meleagridis, these results suggest that lateral transmission of histomoniasis through a flock can occur readily through normal contact between uninfected birds and infected birds and their droppings in the total absence of cecal worms.  相似文献

A killed Mycoplasma hyopneumoniae vaccine was evaluated in a single swine herd in which the farrowing barn and weaner rooms were on one Mycoplasma-free farm, while the growing and finishing barn was on a separate farm on which Mycoplasma was present. The study was carried out in a cohort of pigs born in a 12-week period. Pigs born in 6 of the 12 wk were vaccinated and the rest were left as controls. The vaccine was administered twice at approximately 3 and 6 wk of age. Carcass characteristics, lung lesions, and growth rates were recorded on 893, 390, and 220 pigs, respectively. The vaccine reduced the prevalence of pneumonic lesions in slaughter hogs from 69% to 36% (P < 0.001). It also appeared to reduce the prevalence of pleuritis from 20% to 13%, but the difference was only statistically significant at P = 0.07. The vaccine had no effects on carcass characteristics except that carcasses of vaccinated pigs were, on average, 1 kg heavier than those of nonvaccinated pigs, and a smaller percentage of vaccinated pigs were shipped "light" (carcass weight < 70 kg). Two methods were used to estimate the effect of the vaccine on growth rates (as measured by days to 80 kg carcass weight) resulting in estimates of 11 and 2 d reduction attributable to vaccination, respectively. The latter estimate was probably an underestimate for reasons discussed in the paper.  相似文献

为研究日粮磷水平对22~42日龄肉仔鸡胴体性能和肉品质的影响,选用500只1日龄爱拔益加肉鸡公雏,于1~21日龄统一饲喂相同的玉米—豆粕型配合日粮(含非植酸磷实测值为0.39%),于22日龄从中选取360只鸡,按体重相近原则随机分成10个处理组,每个处理组6个重复笼,每个重复笼6只鸡,分别饲喂不添加无机磷的玉米—豆粕型日粮(对照组,含非植酸磷0.09%)及在基础日粮中分别添加0.05%、0.10%、0.15%、0.20%、0.25%、0.30%、0.35%、0.40%和0.45%无机磷(饲料级CaHPO4),饲养21 d。结果表明,日粮添加适宜水平的无机磷可显著提高42日龄肉仔鸡屠宰率和全净膛率(P<0.1),但日粮磷水平对42日龄肉仔鸡肉品质各指标及其他胴体性能指标均无显著影响(P>0.1)。  相似文献
