The influence of plane of nutrition, growth hormone (GH) treatment and dietary polyunsaturated fat on serum concentrations of GH and insulin (INS) and binding capacities of GH, INS and prolactin (PRL) in liver, mammary parenchyma and adipose tissue was assessed in prepubertal ewe lambs. Ten lambs were assigned to each of four treatment groups. Treatments included: (A) lambs with ad libitum access to a high-energy ration; (G) lambs fed as group A and treated with bovine GH (.08 mg/kg/d); (R) lambs with feed intake restricted to limit ADG to about 120 g; and (S) lambs with ad libitum access to a ration including formaldehyde-protected sunflower seed. Diets, all approximately isonitrogenous and isocaloric, were fed from about 7 to 22 wk of age. Weekly blood samples were collected during wk 6 to 14 of the trial. Averaged across sampling dates, mean serum concentrations of GH were elevated in G lambs (P less than .05) and INS concentrations differed in the order G greater than A greater than R = S (P less than .05). Crude membranes for binding assays were prepared from liver, mammary parenchyma and adipose tissue. Mean concentrations of GH receptors in liver and PRL receptors in mammary parenchyma were elevated in group S lambs (P less than .01). Dietary polyunsaturated fat increased the number of GH receptors in liver and PRL receptors in mammary parenchyma. Increased availability of receptors may mediate the stimulation of mammary growth observed in lambs fed polyunsaturated fat.  相似文献

Effects of plane of nutrition, growth hormone treatment and dietary polyunsaturated fat on mammary development were assessed in prepubertal ewe lambs. Ten lambs (15.6 kg initial body weight) were assigned to each of four treatment groups. Treatments included: (A) lambs given ad libitum access to a high-energy ration; (G) fed as group A but injected s.c. daily with bovine growth hormone (.08 mg/kg BW); (R) feed intake restricted to achieve a growth rate of about 120 g/d; and (S) given ad libitum access to a ration including a formaldehyde-protected sunflower seed supplement. Diets were isonitrogenous and isocaloric and were fed from about 7 to 22 wk of age. At slaughter, the udder was removed and divided into halves. One udder half was trimmed and the parenchyma was dissected from surrounding stroma for compositional analyses. Final body weight and rate of gain averaged 41.2, 42.8, 27.3 and 46.0 kg, and 251, 267, 112 and 311 g/d for groups A, G, R and S, respectively. In the same treatment order, mammary parenchymal weight averaged 15.3, 20.3, 14.2 and 25.6 g. Parenchymal dry, fat-free tissue and DNA content were 1.7, 2.5, 1.3 g and 12.6, 24.1, 10.4 and 18.8 mg, respectively. Mammary parenchymal development was stimulated by polyunsaturated fat but was not affected by plane of nutrition. Supplemental dietary lipid may promote mammogenesis in ruminants.  相似文献

To examine effects of nutritional plane and Se supplementation on colostrum quality and mammary development, individually fed, pregnant Rambouillet ewe lambs were allotted randomly to 1 of 6 treatments in a 2 x 3 factorial arrangement. Main effects included dietary Se level, which began at breeding (d = 0) [adequate Se (9.5 mug/kg of BW) vs. high Se (81.8 mug/kg of BW)], and plane of nutrition, which began at d 50 of gestation [60% (RES), 100% (CON), and 140% (HIGH) of requirements]. Upon parturition, lambs were immediately separated from dams and weighed. Three hours after lambing, colostrum yield was determined, and samples were obtained for components and immunoglobulin G (IgG) analysis. Ewes were slaughtered within 24 h of parturition, and mammary tissues were collected for determination of alveolar secretory epithelial cell proliferation index and luminal area. Gestation length was reduced (P < 0.01) in HIGH ewes compared with RES and CON ewes. Although birth weights were reduced (P < 0.01) in RES and HIGH compared with CON ewes, there was little effect of diet on placental size. Mammary gland weight was reduced (P /= 0.15) on mammary gland weight, colostrum quantity, or IgG concentration in pregnant ewe lambs. Improper nutrition from mid to late pregnancy in ewe lambs altered colostrum quality and quantity and reduced offspring birth weight, which may have negative implications for lamb health and survival during the early postnatal period.  相似文献

To investigate the influence of maternal Se supply and plane of nutrition on lamb morbidity, mortality, and passive transfer of IgG, pregnant ewe lambs were used in 2 experiments with 2 × 3 factorial treatment arrangements. Supplementation of Se began at breeding and was either adequate Se (ASe, 9.5 μg/kg of BW) or high Se (HSe, 81.8 μg/kg of BW) in Exp. 1 or ASe (11.5 μg/kg of BW) or HSe (77.0 μg/kg of BW) in Exp. 2. On d 50 or 40 of gestation for Exp. 1 or 2, respectively, ewes were assigned randomly to 1 of 3 nutritional planes: 60% (RES), 100% (control, CON), or 140% (HI) of NRC requirements. This resulted in the following treatments: ASe-RES, ASe-CON, ASe-HI, HSe-RES, HSe-CON, and HSe-HI. Upon parturition, lambs were separated from their dams and serum samples obtained. Lambs were fed artificial colostrum for the first 20 h and then placed on milk replacer and grain pellets until completion of the study (Exp. 1, 57 d; Exp. 2, 21 d). Twenty-four hours after parturition, lamb serum samples were collected for IgG analysis. All lambs were reared similarly and morbidity and mortality assessed. Main effects were considered significant when P ≤ 0.05. In Exp. 1, there was a Se × plane of nutrition interaction (P ≤ 0.01) for lamb morbidity from birth to weaning and for 24-h IgG concentration. Lambs from ASe-RES and HSe-HI ewes were treated more frequently (P < 0.01) for respiratory and gastrointestinal disease, and lambs from HSe-HI ewes had the smallest (P < 0.01) 24-h serum IgG concentration. In Exp. 1, lambs from HI ewes also had the greatest (P < 0.01) mortality rates from birth to weaning compared with lambs from CON and RES ewes. In Exp. 2, there was an effect (P < 0.01) of maternal plane of nutrition with lambs from RES ewes having increased 24-h IgG compared with lambs from CON and HI ewes. There was no effect of maternal Se supplementation on lamb 24-h IgG in Exp. 2; however, there was a Se × plane of nutrition interaction (P < 0.01) for morbidity. From birth to 21 d of age, lambs from ASe-CON ewes had fewer (P < 0.01) treatment days compared with lambs from any of the other treatment groups. There also tended (P = 0.08) to be an effect of maternal Se supplementation on lamb mortality with increased mortality observed in lambs from HSe ewes. Results from the studies show a restricted maternal plane of nutrition can increase lamb serum IgG concentration. Selenium results were not consistent between the 2 experiments and may be due to differences in maternal Se.  相似文献

The objective of this study was to investigate the expression and localization of HSP60 in the heart, liver, and kidney of acutely heat-stressed broilers at various stressing times. The plasma creatine kinase (CK) and glutamic pyruvic transaminase (GPT) concentrations statistic increased following heat stress. After 2 h of heat stress, the tissues showed histopathological changes. Hsp60 expressed mainly in the cytoplasm of parenchyma cells heat stress. The intensity of the cytoplasmic staining varied and exhibited an organ-specific distribution pattern. Hsp60 levels in the hearts of heat-stressed chickens gradually increased at 1 h (p < 0.05) and peaked (p < 0.05) at 5 h; Hsp60 levels in the liver gradually decreased at 3 h (p < 0.05); Hsp60 levels in the kidney had no fluctuation. It is suggested that Hsp60 expression is tissue-specific and this may be linked to tissue damage in response to heat stress. The Hsp60 level is distinct in diverse tissues, indicating that Hsp60 may exert its protective effect by a tissue- and time-specific mechanism.  相似文献

Nutrition has been shown to influence several reproductive functions, including hormone production, oocyte competence and fertilization, and early embryonic development. To determine the effects of maternal diet on in vitro fertilization (IVF) and early embryonic development, ewes (n = 18; 47.0 +/- 1.5 kg of initial BW) were divided into control and underfed (60% of control) nutritional planes for 8 wk before oocyte collection. Pelleted diets containing 2.4 Mcal of ME/kg and 13% CP (DM basis) were fed once daily. During the first 4-wk acclimation phase, control and underfed ewes were fed 1,000 and 600 g/d, respectively. From wk 4 to 8, control (adequate) ewes were fed to maintain BW and offered 720 g/d, whereas underfed ewes received 432 g/d (60% restricted). Synchronization of estrus was performed using progestagen sponges for 14 d. Follicular development was induced by twice daily injections of FSH on d 13 (5 units/injection) and 14 (4 units/injection) of the estrous cycle. Oocytes were collected from all visible follicles on d 15 of the estrous cycle. After IVF, the proportion of developing embryos was evaluated throughout an 8-d culture period. Under-nutrition decreased (P < 0.006) the rate of cleavage, number of blastocysts per ewe, and rate of blastocyst formation (from 79 to 64%; from 3.3 to 0.8; and from 31 to 8%, respectively). However, the number of visible follicles, total number of oocytes, number of healthy oocytes, percentage of healthy oocytes, number of cleaved oocytes, and morula formation per ewe were similar for control and underfed ewes. These data indicate that undernutrition of donor ewes, resulting in lower BW and BCS, has a negative effect on oocyte quality, which results in lower rates of cleavage and blastocyst formation.  相似文献

为了研究3种营养水平(A1-0.7×NRC、A2-0.8×NRC、A3-0.9×NRC)全混合日粮对舍饲羔羊生产性能、养分表观消化率和屠宰性状的影响,本试验选择48只南非肉用美利奴×甘肃高山细毛羊杂交一代羔羊,随机分为3个处理组,进行为期60 d的育肥试验。结果显示,在全混合日粮饲喂技术下,饲喂A2、A3营养水平日粮组的羔羊全期平均日增重极显著高于A1营养水平组(P<0.01),A2和A3营养水平组差异不显著(P>0.05),公羔全期平均日增重极显著高于母羔(P<0.01);A因子对钙、总磷、中性洗涤纤维、酸性洗涤纤维消化率均有极显著影响(P<0.01),性别仅对钙、总磷消化率有极显著影响(P<0.01),但营养水平与性别没有显著交互作用(P>0.05);营养水平对育肥羔羊的屠宰性能没有显著影响(P>0.05),但杂种羔羊的屠宰率、熟肉率、眼肌面积等高于甘肃高山细毛羊。以上结果表明采用0.8×NRC营养水平全混合日粮育肥南非肉用美利奴×甘肃高山细毛羊杂交一代羔羊较为适宜,在同一营养水平下,公羔生长性能优于母羔。  相似文献

Splanchnic metabolite flux was measured in young lambs given access to a high-concentrate diet either ad libitum (ADLIB) or at a maintenance level (MAINT) for 21 d. Net fluxes of urea N (UN), ammonia N (NH3 N), alpha-amino N (AAN), amino acids, glucose (G), and lactate (L) across the liver and portal-drained viscera (PDV) were measured in 11 crossbred ram lambs (35 kg) surgically fitted with indwelling catheters in the portal, hepatic, and mesenteric veins and mesenteric artery. During the 21-d period, daily N and ME intakes were 24.6 and 10.7 g N/d and 3.02 and 1.28 Mcal/d, respectively, for ADLIB and MAINT lambs. Intakes, thus, were 42% lower for MAINT than for ADLIB lambs. Net portal fluxes of UN, NH3 N, AAN, and L in MAINT lambs were 46%, 84%, 50%, and 74%, respectively, of that in ADLIB lambs. Expressed as a percentage of N intake, the proportion of AAN absorbed by the PDV was higher in MAINT lambs (P less than .05) than in ADLIB lambs. There was no net portal glucose absorption in either group of lambs; however, net hepatic glucose production in MAINT lambs was 48% of that in ADLIB lambs. There was net utilization of glutamine by the PDV; net glutamine flux in MAINT lambs was 49% of that in ADLIB lambs. The liver utilized AAN and NH3 N and produced UN. Splanchnic tissues modulate metabolite flux following changes in feed intake in young ruminants.  相似文献

Endocannabinoids are fatty acid amides (FAE; oleoylethanolamide and anandamide) which have orexigenic, anorexigenic or anti‐inflammatory properties. We examined mRNA expression via qPCR of endocannabinoid receptors (CNR1 and CNR2), enzymes that synthesize FAE (HRASLS5 and N‐acyl phosphatidylethanolamine phospholipase D), enzymes that degrade FAE [fatty acid amide hydrolase (FAAH), N‐acylethanolamine acid amidase (NAAA) and monoglyceride lipase (MGLL)], and the hormone precursor proopiomelanocortin (POMC) in liver at ?14, 7, 14 and 30 days around parturition from cows fed with a control (CON; NEL = 1.34 Mcal/kg) or moderate‐energy (OVER; NEL = 1.62 Mcal/kg) diet during the dry period. Expression of CNR2 and POMC was greater at 7 days in cows fed with OVER because of a decrease in expression between ?14 and 7 days in cows fed with CON. Cows fed with CON had an increase in expression of FAAH, HRASLS5, NAA, MGLL and POMC between 7 and 14 days; for FAAH and HRASLS5, such response led to greater expression at 14 days vs. cows fed with OVER. Cows fed with OVER vs. CON had a approximately twofold increase in expression of MGLL between ?14 and 7 days followed by a gradual decrease through 30 days at which point expression was still greater in OVER vs. CON. FAAH, MGLL and HRASLS5 were the most abundant genes measured. Expression of the hepatic endocannabinoid system and POMC was altered by plane of dietary energy prepartum particularly during the first 2‐week postpartum. Such responses may play a role in the physiological adaptations to the onset of lactation, including energy balance and feed intake.  相似文献

To examine the effect of a beta-adrenergic agonist (BAA) on muscle growth, proteinase activities, and postmortem proteolysis, 16 wether lambs were randomly assigned to receive 0 or 4 ppm of L644,969 in a completely mixed high-concentrate diet for 6 wk. Weight of the biceps femoris was 18.6% heavier in treated lambs. At 0 h after slaughter, treated lambs had higher cathepsin B (35.6%), cathepsins B + L (19.1%), calpastatin (62.8%), and m-calpain (24.6%) than control lambs, but both groups had similar mu-calpain activities. In both longissimus and biceps femoris muscles, treated lambs had higher protein and RNA and lower DNA concentrations. However, total DNA was not affected, indicating that the increase in muscle mass was probably due to muscle hypertrophy rather than to hyperplasia. The pattern of postmortem proteolysis was significantly altered by BAA feeding. In treated lambs, postmortem storage had no effect on the myofibril fragmentation index and degradation of desmin and troponin-T. These results indicate that the ability of the muscle to undergo postmortem proteolysis has been dramatically reduced with BAA feeding. Similar proteolytic systems are thought to be involved in antemortem and postmortem degradation of myofibrillar proteins, so BAA-mediated protein accretion is probably due, at least in part, to reduced protein degradation. To examine whether protein synthesis was altered with BAA feeding, the level of skeletal muscle alpha-actin mRNA was quantified. Longissimus muscle alpha-actin mRNA abundance was 30% greater in BAA-fed lambs. Collectively, these results indicate that dietary administration of BAA increases muscle mass through hypertrophy and that the increase in muscle protein accretion is due to reduced degradation and possibly to increased synthesis of muscle proteins.  相似文献

Over a period of years it has become standard practice in certain areas of New Zealand to take precautions against tetanus following lamb marking. These areas are fairly well defined and the main regions are those of Hawke's Bay and the southern half of the South Island. In these areas, losses following lamb marking, many being due to tetanus, can be high if no preventive measures are taken. It has frequently been observed that conditions favouring the development of Clostridium tetani accompany certain operations at marking, and that this risk has increased because of the use of rubber rings and searing irons at marking. This was to be expected since anaerobic conditions in deep-seated wounds will favour the growth of Cl. tetani.  相似文献

Peripubertal ewe lambs (44.3 +/- 1.1 kg of initial BW) were used in a 2 x 3 factorial design to test the effects of plane of nutrition (diet) and stage of gestation on maternal visceral tissue mass, intestinal cellularity, crypt cell proliferation, and jejunal mucosal vascularity. Singleton pregnancies to a single sire were established by embryo transfer, and thereafter ewes were offered a control (Control) or high (High) amount of a complete diet (2.84 Mcal/kg and 15.9% CP; DM basis) to promote slow or rapid maternal growth rates. After d 90 of gestation, feed intake of the Control group was adjusted weekly to maintain BCS and meet the increasing nutrient demands of the gravid uterus. Ewes were slaughtered at 50 d (n = 6 Control; n = 5 High), 90 d (n = 8 Control; n = 6 High), or 130 d (n = 8 Control; n = 6 High) of gestation. Ewes were eviscerated and masses of individual organs were recorded. The jejunum was sampled and processed for subsequent analyses. Final ewe BW for Control-fed ewes was similar at d 50 and 90 and increased (P = 0.10) from d 90 to 130 (46.0, 48.9, and 58.2 +/- 1.6 kg, respectively), whereas final BW increased (P 相似文献

Effects on udder development and colostrum production of changing the level of ewe nutrition were investigated. Udder weights at term were derived from their linear dimensions and yields of colostrum were obtained by hand milking on three occasions between one and 18 hours after birth. The linear dimensions of the udder increased progressively in all animals between 100 days of gestation and term (about 145 days), but udder development was retarded significantly within three days of the onset of underfeeding at 105 days of gestation. Underfeeding reduced the total yield of colostrum during the first 18 hours after birth by decreasing the prenatal accumulation of colostrum and its subsequent rates of secretion. Improving the ewe's nutrition from one hour after birth tended to increase the secretion rates of colostrum between 10 and 18 hours. Refeeding previously underfed ewes to high levels from about five days before birth had no effect on udder growth but did increase the prenatal accumulation of colostrum and its subsequent rates of secretion to levels which, when corrected for differences in udder weight, were similar to those of ewes which were fed well throughout. Calculations revealed that by ignoring udder growth the Agricultural Research Council recommendations for the energy requirements of pregnant ewes underestimate the cost of tissue synthesis during the last four weeks of pregnancy by about 100 per cent.  相似文献

The effects of hemi-castration upon compensatory hypertrophy, serum gonadotropin and testosterone concentrations, testicular gonadotropin receptors and daily sperm production (DSP) were studied in 10 crossbred ram lambs. At 4 mo of age lambs were either hemi-castrated (HC; n = 5) or left intact (INT; n = 5). Blood samples were collected every 2 h for the first 24 h post-surgery, every 6 h for the next 24 h and then three times weekly for the following 14 wk. Serial blood samples (15-min intervals for 8 h) were collected during the 4th, 8th and 12th week following hemi-castration. Individual mean testicular and epididymal weights increased (P less than .05) 48 and 33% in HC compared with INT rams, respectively. Serum follicle stimulating hormone (FSH) increased (P less than .05) within 8 h after HC, reached peak concentrations within 1 wk and remained elevated for 4 wk before returning to concentrations of INT rams. Neither mean serum luteinizing hormone (LH) nor pulse patterns of LH or FSH were different (P greater than .05) between these two groups at any period examined. Serum testosterone (T) concentrations were lower (P less than .05) during the first 48 h post-surgery in HC rams, but by 1 wk concentrations were similar (P greater than .05) to those in INT rams. Remaining testes from HC and INT rams were removed at 7 mo of age, 3 mo after initial gonadal manipulation. Sixty-four white-faced rams and wethers were dressed with the aid of a commercial pelt puller. The effects of age, castration, and season on difficulty of pelt removal and pelt damage were evaluated. Lambs were divided into two age groups (5 and 12 mo) within gender (ram and whether) and season (spring and fall). A greater force (P less than .05) was required to remove pelts from rams than from wethers in both 5- and 12-mo-old groups. Older lambs slaughtered in the fall required more force (P less than .05) to remove their pelts than did those slaughtered in the spring, but differences by season did not exist for 5-mo-old lambs. The difference between rams and wethers in percentage of live weight that was closely shorn pelt weight was not significant (P greater than .05). The area of grain crack in the flank expressed as a percentage of total area of the skin was lower (P less than .05) for skins from 5-mo-old lambs and ram lambs than it was for skins from 12-mo-old lambs and wether lambs, respectively. Factors involved in difficulty of pelt removal in ram lambs included crosscut shoulder weight, fat firmness, and carcass weight. Difficulty of pelt removal in wether lambs was best predicted by including crosscut shoulder weight and bodywall thickness in multiple regression equations.  相似文献
