Fusarium head blight (FHB) resistance of 50 cultivars from the National List of winter wheat cultivars approved for sale (or were undergoing trails for approval in 2003) in the UK was compared with 21 reference cultivars from continental Europe which had previously been characterized for resistance. Only three UK National List cultivars (Soissons, Spark and Vector) had stable resistance over trial sites that was significantly greater than that of the FHB susceptible cultivar Wizard. In addition, under moderate disease pressure, 21 of the National List cultivars had levels of the trichothecene mycotoxin deoxynivalenol (DON) above the proposed European Union limit of 1·25 ppm in grain. Surveys show that levels of FHB and DON in the UK crop are currently very low, however, should disease pressure increase for any reason, then an improvement in the overall levels of FHB resistance of UK winter wheat germplasm will be required. In order to infer the origin of resistance and to identify potentially novel resistance, allele sizes of microsatellite (simple sequence repeat, SSR) markers linked to quantitative trait loci (QTL) for FHB resistance were compared between the test cultivars and known, characterized resistance sources. The major FHB resistance QTL from the Chinese cv. Sumai-3 (3BS, 5A and 6B), the Romanian cv. Fundulea F201R (1B and 5A) and the French cv. Renan (5AL) were screened with 17 SSRs. No National List cultivar had haplotypes similar to any of these QTL. However, the highly resistant German reference cultivar Petrus had an identical haplotype to cv. Fundulea F201R on 1B indicating that this cultivar has an allelic FHB resistance QTL at that location.  相似文献

The effect of small temperature differentials (16 vs. 20°C) on the pathogenicity of deoxynivalenol producing single isolates of Fusarium culmorum and F. graminearum and on the fusarium head blight (FHB) response of eight wheat cultivars was examined. Fusarium culmorum inoculation caused greater visual disease symptoms at 20°C than at 16°C, both overall and on an individual cultivar basis (overall AUDPC = 13·5 and 9·6, respectively) ( P  < 0·05). In contrast, F. graminearum inoculation caused greater overall visual disease symptoms at 16°C than at 20°C, both overall and at the individual cultivar level (overall AUDPC = 12·8 and 10·9, respectively) ( P  < 0·05). Results showed both F. culmorum and F. graminearum inoculations caused a greater loss in yield at 20°C (54·3 and 46·9% relative 1000-grain weight, respectively) compared with 16°C (73·3 and 66·9% relative 1000-grain weight, respectively) ( P  < 0·05). Fusarium culmorum -inoculated heads contained similar amounts of fungal DNA at both 16 and 20°C (1·9 and 1·7 ng mg−1 of plant material, respectively) (not significant), while for F. graminearum inoculation, plants contained higher amounts of fungal DNA at 20°C (2·0 and 1·0 ng mg−1 of plant material, respectively) ( P  < 0·05). Overall, there was a significant negative correlation between AUDPC and percentage relative 1000-grain weight at both 16 and 20°C ( r  =−0·693 and −0·794, respectively, P  < 0·01).  相似文献

ABSTRACT Over 4 years, the environmental conditions and the causal agents of Fusarium head blight (FHB) disease of wheat were determined in field sites in four European countries: Hungary, Ireland, Italy, and the United Kingdom. Polymerase chain reaction-based methods were used to detect each species causing FHB and quantify its DNA (as a measurement of fungal abundance) in the samples. Canonical correspondence analysis (CCA) was used to determine the relationship of the incidence and abundance of each species with weather variables. CCA indicated that little variability in the species prevalence data was explained by the weather variables. In contrast, a greater proportion of variability in abundance data was accounted for by the weather variables. Most samples contained two or more species and statistical analysis suggested that these species tended to coexist at field sites. CCA also indicated that there were differences in the relationships of the prevalence and abundance of the six FHB species with environmental variables. Fusarium poae was associated with relatively drier and warmer conditions, whereas F. graminearum was associated with warmer/humid conditions. F. avenaceum and F. culmorum were both associated with niches of cooler/wet/humid conditions. Two Microdochium species were associated with regions of relatively cool/moderate temperatures and frequent rainfalls of short duration. The results also suggested that environmental conditions differentially affect the infection and colonization processes, and the comparative abundance of the six species.  相似文献

Introduction of alien genes into wheat has been proposed as a strategy to breed cultivars with improved resistance to Fusarium seedling blight (FSB) and Fusarium head blight (FHB). In this study, we co-transformed different anti-fungal peptides (AFPs) into an elite wheat cultivar Yangmai11. We identified the genetically stable transgenic wheat lines carrying single or multiple genes by PCR, qRT-PCR and Southern blot analyses. Transgenic wheat lines 451 and 513 expressing two AFPs displayed a consistent, significantly improved overall resistance to FSB and FHB, whereas only FHB resistance was observed from other lines. Furthermore, crude proteins extracted from the lines 451 and 513 showed a clear inhibitory activity against F. graminearum in vitro. Taken together, it was essential to properly combine and express AFPs in transgenic wheat in order to obtain an improved overall resistance to Fusarium pathogens.  相似文献

Fusarium culmorum causes head blight, produces toxins and reduces yield and quality of cereals. To prevent damage caused by fusarium head blight (FHB), azole fungicides are mainly applied. The occurrence of insensitivity to azoles is a major problem in agriculture. The present study shows that a tebuconazole insensitive strain of F. culmorum can be readily produced in the laboratory, but that the resulting strain of the fungus is of lower fitness in vitro. Insensitivity was confirmed microscopically and by cell viability and metabolic activity. The tebuconazole insensitive strain shows cross insensitivity to nine important azoles. In addition, plants inoculated with the insensitive F. culmorum strain showed no reduction of FHB symptoms and deoxynivalenol (DON) content after tebuconazole treatment, compared to an inoculation with the sensitive strain. Use of wheat cultivars carrying a high resistance level (i.e. cv. Toras) was the most effective method for reducing symptoms and decreasing DON content, independent from the level of fungicide insensitivity of the F. culmorum strain. In conclusion, resistant cultivars and a fungicide mixture which combines different mechanisms of action in fungal metabolism should be applied to avoid fungicide insensitivity of Fusarium spp. in future.  相似文献

Seven spring wheat, 13 barley, 14 oats and 20 winter wheat genotypes were inoculated at flowering in 1993 and 1994 with mixed conidial suspensions of 8 isolates of Fusarium culmorum. Four weeks after inoculation, head blight was recorded in the field. After harvest, seed infection was assessed by a Freezing Blotter Test in the laboratory. Seed samples were also analyzed for deoxynivalenol (DON) content. Differences were found in head blight rating, the levels of infected seeds and the DON content between wheat, barley, and oats and between cultivars. Highly significant correlations were found between the percentage of heavily infected seed and the DON content. The weighted mean value of infected seeds and DON content were also significantly correlated. No significant correlation was found between head blight rating in the field and DON content. The level of infected seeds, as determined by the Freezing Blotter Test, was a better indication of the DON content in the seeds than the head blight rating. This visual assessment of levels of infected seeds gives a reliable estimate of resistance to Fusarium.  相似文献

Fusarium head blight (FHB), caused by Fusarium graminearum sensu stricto (s.s.), causes tremendous annual yield losses in wheat worldwide. Variation of aggressiveness of isolates from individual field populations in terms of FHB infection and deoxynivalenol (DON) concentration in the host are important population parameters reflecting parasitic ability. Our main objective was to estimate the variation of both traits within three populations of F. graminearum s.s., each consisting of 30 single-spore isolates collected from small wheat fields in Germany, and to compare it with 11 isolates of a collection (F. graminearum collection) from four countries. The same isolates were characterized using 19 single-sequence repeat markers. All isolates were spray inoculated on a moderately resistant spring wheat cultivar at two field locations over 2 years (i.e., in four environments). The genotypic proportion of phenotypic variance (σ(2)(G)) within populations was significant (P < 0.01) for both traits, and the σ(2)(G) × environment interaction was even more important for mean FHB severity. Ranges in mean FHB severity and DON concentration in the host were only slightly smaller for the field populations than for the F. graminearum collection. Both traits were significantly (P < 0.05) correlated within and across populations. A further partitioning of σ(2)(G) revealed 72% of σ(2)(G) within and 28% of σ(2)(G) across populations for both traits. Molecular variance of the three populations was similarly distributed (73.6% within versus 26.4% between populations). In view of this high within-field variation for traits of parasitic ability and selection, neutral molecular markers, multiple resistance genes of different origin should be employed in wheat breeding programs to obtain a long-term stable FHB resistance.  相似文献

小麦赤霉病(Fusarium Head Blight,FHB)是一种世界性病害,也是我国小麦生产中主要病害之一,由镰刀菌属(Fusarium spp.)真菌引起的。其中最主要的是禾谷镰刀菌(Fusarium graminearum)。小麦赤霉病不仅导致减产,而且病原菌产生毒素降低了小麦的品质,危害人、畜的健康。防治小麦赤霉病主要在小麦扬花期进行化学防治,但是药剂防治使病原菌产生了严重的抗药性。本文综述了小麦赤霉病抗药性的国内外研究进展,主要介绍了赤霉病抗药性发生现状、抗药性机制和抗药性治理,对小麦赤霉病的高效防治、减少或延缓抗药性的发生以及对抗药性的治理具有重要的理论和现实意义。  相似文献

为探索生物炭对小麦赤霉病的防治效果和对产量的影响,设置小麦播种前、返青拔节期单施或两个时期均施生物炭处理,在小麦分蘖期人工接种病麦粒,在抽穗扬花期采用孢子捕捉器对穗层空气中的赤霉病菌孢子数量进行动态监测,并在乳熟期调查各处理的病穗率和病情指数,成熟期测定各处理的理论产量、实际产量和籽粒中脱氧镰刀菌烯醇(deoxynivalenol,DON)毒素的含量。结果表明,施用生物炭后穗层赤霉病菌孢子的数量显著降低,赤霉病病情指数降低,且以基施和返青拔节期追施13 500 kg/hm~2生物炭的处理最优;施用生物炭后小麦千粒重和株高均显著增加(P0.05),其中基施和追施13 500 kg/hm~2生物炭的处理较对照处理增产29.5%(P0.05)。  相似文献

A dynamic simulation model for the risk of Fusarium head blight on wheat was elaborated based on systems analysis. The model calculates a daily infection risk based on sporulation, spore dispersal and infection of host tissue of the four main species causing the disease (Gibberella zeae, Fusarium culmorum, Gibberella avenacea, Monographella nivalis). Spore yield and dispersal are calculated as functions of temperature, rainfall and relative humidity, while the main factors affecting the infection rate are temperature, wetness and the host growth stage. The model also calculates a risk for mycotoxin production by G. zeae and F. culmorum in the infected head tissue. First validations against field data, collected in some wheat‐growing areas in northern Italy and not used in model elaboration, produced satisfactory results.  相似文献

为了解湖北省小麦品种(系)的抗病性水平,对2001年以来湖北省审定的46个小麦品种和在湖北省进行区域试验的447份小麦品系进行赤霉病、条锈病、白粉病和纹枯病等4种主要病害的抗病性评价。结果表明:1)参试品系中有1份材料对赤霉病表现高抗,未发现对纹枯病表现高抗或免疫的材料;对赤霉病和纹枯病的平均抗性评价分值分别为14.30和3.49,介于中感和高感之间;对条锈病和白粉病的平均抗性评价分值分别为13.43和8.34,介于中抗和中感之间。2)审定品种的综合抗性水平不高,对条锈病的抗性评价分值为12.47,介于中抗和中感之间,对其他3种病害的抗性介于中感和高感之间;审定品种的单产呈增加趋势,株高在不断降低,生育期相应延长,综合抗病性也在增强。3)审定品种的综合抗病性与对条锈病抗性的相关系数最高,为0.738;对纹枯病的抗性与株高呈显著正相关,与单产呈显著负相关;对赤霉病的抗性与株高无显著相关。鉴于此,湖北省需加强抗病育种力度,同时应充分做好小麦病害的防控。  相似文献

为明确不同杀菌剂防治小麦赤霉病和减少小麦籽粒中DON(脱氧雪腐镰刀菌烯醇)毒素积累的效果,于大田人工接菌条件下进行了不同杀菌剂对小麦赤霉病的防治试验,并于收获期测定了不同药剂处理的小麦籽粒中DON毒素含量。结果表明:两种处理方式下,50%多菌灵WP,30%多·酮SC,25%戊唑醇WP,25%氰烯菌酯SC和70%甲基硫菌灵WP处理中,除先喷药后接菌条件下25%戊唑醇WP在病粒率和籽粒中DON积累上没有显著防治效果,接菌后1d喷药条件下50%多菌灵WP、25%戊唑醇WP在病粒率上没有显著防治效果;30%多·酮SC在病粒率和籽粒中DON积累上没有显著防治效果外,各处理对小麦的病穗率、病情指数、病粒率和籽粒中DON积累都有显著的防治效果(P0.05)。各处理的病穗率防效在42.4%~83.5%,病情指数防效在44.9%~88.2%,病粒率防效在34.7%~69.4%,籽粒中DON防效在48.0%~86.9%。上述几种杀菌剂具有明显的保护作用,其在先喷药后接菌条件下的防治效果均明显优于接菌后1d喷药的防治效果。两种处理方式下,30%己唑醇悬浮剂、25%嘧菌酯悬浮剂和12.5%烯唑醇可湿性粉剂处理在病穗率、病情指数、病粒率和籽粒中DON积累与清水对照处理没有显著性差异(P0.05)。  相似文献

Summary An infection of bread wheat by fusarium head blight contaminates the crop with mycotoxins, particularly deoxynivalenol (DON) and nivalenol (NIV). The toxicity and natural occurrence of these mycotoxins in wheat are reviewed. Based on 8 years data of fusarium head blight epidemics of wheat in the Netherlands, DON contamination of the grain was estimated. Fusarium head blight ratings averaged an infection of 1.7% of all spikelets; estimates for DON contamination averaged 0.9 mg kg–1. Taking a guideline level for DON in uncleaned bread wheat of 2 mg kg–1, in 1979 and 1982 a wheat crop was produced with estimated DON concentrations above the limit of tolerance. Human and animal exposure to mycotoxins in the Netherlands appears to be small but chronic. The information presented in this paper illustrates the need for an annual evaluation of the crop for fusarium head blight incidence and mycotoxin content, and the necessity of fusarium head blight resistant wheat cultivars.Samenvatting Aaraantasting van tarwe doorFusarium culmorum enFusarium graminearum leidt tot vorming van mycotoxinen in het graan, waarvan deoxynivalenol (DON) en nivalenol (NIV) de belangrijkste toxinen zijn. In dit artikel wordt een overzicht gegeven van de toxicologische aspecten, en het voorkomen van deze toxinen in tarwe. Informatie over DON en NIV in tarwe in West-Europa is schaars. Gebaseerd op gegevens vanFusarium epidemieën in de jaren 1979–1986 wordt een schatting gegeven van de concentratie DON in Nederlandse tarwe. Rekening houdend met de herkomst en verwerking van tarwe, blijken zowel in dierlijk als menselijk voedsel lage concentraties DON chronisch voor te komen. Op basis van een maximaal toelaatbare dagelijkse dosis DON van 3 g kg–1 lichaamsgewicht is de schatting van de dagelijkse opname van DON in het jaar volgend op de oogst van 1982 net op de grens. Zowel een jaarlijkse inventarisatie vanFusarium aantasting en DON besmetting van het graan, als de ontwikkeling vanFusarium-resistente rassen zijn noodzakelijk.  相似文献

Fusarium head blight (FHB) of wheat is a destructive fungal disease worldwide and has become more severe over the last two decades. Development of FHB resistant wheat varieties is the most effective way to manage FHB. The middle and lower reaches of the Yangtze River are the traditional FHB epidemic areas in China. The landraces and germplasms with resistance to FHB originating from this region were used to identity FHB resistance quantitative trait loci (QTLs). Yangmai 158 and Ningmai 9 are the most popular varieties for commercial wheat production in this region and both have moderate FHB resistance. A high-density genetic map was constructed using 282 recombinant inbred lines (RILs) from a cross between Ningmai 9 and Yangmai 158. Ten QTLs related to Type II FHB resistance were identified, and QFhb-3B.1 and QFhb-5A were stably detected across all environments. Based on position alignment, QFhb-3B.1 from Ningmai 9 is likely to be Fhb1 and QFhb-5A from Yangmai 158 is a novel QTL not previously described. A competitive allele-specific PCR (KASP) marker closely linked to QFhb-5A was developed and could be used for marker-assisted selection. Distribution of QFhb-5A was tested with numerous accessions from a widespread core collection. The results suggest that QFhb-5A has undergone both natural and artificial selection. Some RILs with both Fhb1 and QFhb-5A presented better FHB resistance than the parents and could be used in FHB resistance breeding.  相似文献

为探明不同杀菌剂对小麦赤霉病和小麦籽粒DON毒素(包括DON、3-ADON和15-ADON)的控制效果, 采用菌丝生长速率法测定了12种药剂对禾谷镰刀菌野生型菌株PH-1的室内活性, 同时采用液相色谱-串联质谱法(LC-MS)测定了这些药剂对DON毒素的抑制效果, 并开展了小麦赤霉病及籽粒DON毒素的田间防治试验。结果表明, 12种原药对菌丝生长抑制活性强弱依次为氟唑菌酰羟胺>咪鲜胺>戊唑醇>丙硫菌唑>叶菌唑>氰烯菌酯>氟环唑>多菌灵>甲基硫菌灵>吡唑醚菌酯>嘧菌酯>井冈霉素A。氟环唑EC50和EC90离体胁迫均刺激DON毒素产生, 其他杀菌剂EC50和EC90胁迫均抑制DON毒素产生。田间试验结果表明, 200 g/L氟唑菌酰羟胺SC、30%丙硫菌唑OD和20%叶菌唑WP病指防效和DON防效为87.68%~94.77%; 430 g/L戊唑醇SC、25%氰烯菌酯SC、45%咪鲜胺EW、25%氟环唑SC、50%多菌灵WP和70%甲基硫菌灵WP病指防效和DON防效为57.63%%~85.49%; 250 g/L吡唑醚菌酯EC和250 g/L嘧菌酯SC病指防效分别为72.18%和51.98%, DON防效分别为43.06%和-7.96%; 24%井冈霉素A AS病指防效和DON防效分别为42.37%和62.87%。药剂离体和田间控毒效果不完全一致, 赤霉病有效防控是DON防控的前提, 病害防效与DON防效不完全一致, 本研究为小麦赤霉病及籽粒DON毒素防控提供了科学依据。  相似文献

本研究明确了黄淮冬麦区主栽小麦品种对赤霉病的抗侵染、抗扩展、抗毒素积累和抗籽粒侵染能力以及几种抗性之间的相互关系,并检测了各品种是否带有FHB1抗性基因,旨在为该区小麦赤霉病抗性鉴定评价、抗性品种培育和利用提供科学依据。小麦赤霉病综合抗性鉴定结果表明:22个黄淮冬麦区主栽品种中,有20个为感病品种,只有‘郑麦9023’和‘西农979’为中感品种,所有品种均不含FHB1基因;长江中下游麦区的9个品种中,‘扬麦17’和‘宁麦9号’等6个品种表现中抗,‘扬麦23’表现中感,‘苏麦3号’和‘扬麦21’表现抗,‘扬麦14’‘扬麦17’和‘扬麦23’不含有FHB1基因,其他品种均含FHB1基因。小麦品种的抗扩展能力与抗侵染能力无显著相关性(r=0.27,P0.05);两种接种条件下小麦品种的病粒率与抗脱氧雪腐镰刀菌烯醇(DON)毒素积累能力呈极显著正相关(r=0.86,P0.01;r=0.88,P0.01);单小花滴注法接种条件下,小麦品种的平均病级与病粒率和籽粒中DON含量都呈极显著正相关(r=0.71,P0.01;r=0.81,P0.01);喷雾接种条件下,小麦品种的病小穗率与平均病级、病粒率、籽粒中DON含量和ZEN含量都呈极显著正相关(r=0.78,P0.01;r=0.73,P0.01;r=0.78,P0.01;r=0.63,P0.01)。在毒素积累抗性上,DON含量和ZEN含量呈极显著正相关(r=0.70,P0.01)。在目前黄淮冬麦区没有中抗品种的情况下,可以增加育种和鉴定目标为抗籽粒侵染和抗毒素积累的品种,在小麦品种推广过程中加以运用,可以达到较好的效果。  相似文献
