Mammalian intestinal organoids are multicellular structures that closely resemble the structure of the intestinal epithelium and can be generated in vitro from intestinal stem cells under appropriate culture conditions. This technology has transformed pharmaceutical research and drug development in human medicine. For the insect gut, no biotechnological platform equivalent to organoid cultures has been described yet. Comparison of the regulation of intestinal homeostasis and growth between insects and mammals has revealed significant similarities but also important differences. In contrast to mammals, the differentiation potential of available insect cell lines is limited and can not be exploited for in vitro permeability assays to measure the uptake of insecticides. The successful development of in vitro models could be a result of the emergence of molecular mechanisms of self‐organization and signaling in the intestine that are unique to mammals. It is nevertheless considered that the technology gap is a consequence of vast differences in knowledge, particularly with respect to culture conditions that maintain the differentation potential of insect midgut cells. From the viewpoint of pest control, advanced in vitro models of the insect midgut would be very desirable because of its key barrier function for orally ingested insecticides with hemolymphatic target and its role in insecticide resistance. © 2020 Society of Chemical Industry  相似文献

Precipitation, a natural feature of weather systems in the Earth, is vitally important for the environment of any region. Under global climate change condition, the characteristics of precipitation have changed as a consequence of enhanced global hydrological cycle. The source region of the Yellow River(SRYR), locating within the Qinghai-Tibet Plateau, is sensitive to the global climate change due to its complex orography and fragile ecosystem. To understand the precipitation characteristics and its impacts on the environment in the region, we studied the characteristics of rainy days and precipitation amount of different precipitation classes, such as light(0–5 and 5–10 mm), moderate(10–15, 15–20 and 20–25 mm) and heavy(≥25 mm) rains by analyzing the precipitation data of typical meteorological stations in the SRYR during the period 1961–2014, as well as the trends of persistent rainfall events and drought events. Results showed that annual average precipitation in this area had a non-significant(P>0.05) increasing trend, and 82.5% of the precipitation occurred from May to September. Rainy days of the 0–5 mm precipitation class significantly decreased, whereas the rainy days of 5–10, 10–15, and 20–25 mm precipitation classes increased and that of ≥25 mm precipitation class decreased insignificantly. The persistent rainfall events of 1-or 2-day and more than 2-day showed an increasing trend, with the 1-or 2-day events being more frequent. Meanwhile, the number of short drought periods(≤10 days) increased while long drought periods(>10 days) decreased. Since the 0–5 mm precipitation class had a huge impact on the grasslands productivity; the 5–10, 10–15, and 20–25 mm precipitation classes had positive effects on vegetation which rely on the deep soil water through moving nutrients and water into the root zone of these vegetation or through the plant-microbe interactions; the ≥25 mm precipitation class contributed to the floods; and more persistent rainfall events and fewer long drought events inferred positive effects on agriculture. Thus, these results indicate grassland degradation, less risk of floods, and the upgrading impact of climate change on agriculture. This study may provide scientific knowledge for policymakers to sustain the eco-environmental resources in the SYSR.  相似文献

Pesticide resistance is a major factor affecting world food and fibre production, but that has been contained so far by the availability of diverse modes of action. Overcoming resistance by switching to a new mode of action is a concept easily grasped by growers but threatened by losses through resistance and new registration requirements. Opportunities for innovation and development of a diversity of novel modes of action exist through harnessing recent advances, fundamental to all eukaryotes and largely funded for medical rather than agricultural objectives, in understanding cell biology and development. The cystoskeleton, cell wall synthesis, signal transduction and RNAi are discussed as examples where new targets are now exposed. However, new modes of action will be delivered not only by sprayer or seed treatment but also through transgenic crops, although these still need to be transferred from experiment to practice. Improvements in modelling protein structures and target-site changes, supplemented by rapid diagnostics to detect resistance early, will improve resistance risk management and integrate chemical, biopesticide, transgenic and conventional breeding around the concept of diversity in modes of action. However, before agronomy can translate this into practical antiresistance strategies, there is a need to direct more resources to the biochemistry and cell biology of pests, diseases and weeds to translate these new discoveries into key tools needed to manage resistance in the future.  相似文献

Based on daily precipitation data from 163 meteorological stations, this study investigated precipitation changes in the mid-latitudes of the Chinese mainland(MCM) during 1960–2014 using the climatic trend coefficient, least-squared regression analysis, and a non-parametric Mann-Kendall test.According to the effects of the East Asian summer monsoon on the MCM and the climatic trend coefficient of annual precipitation during 1960–2014, we divided the MCM into the western MCM and eastern MCM. The western MCM was further divided into the western MCM1 and western MCM2 in terms of the effects of the East Asian summer monsoon. The main results were as follows:(1) During the last four decades of the 20~(th) century, the area-averaged annual precipitation presented a significant increasing trend in the western MCM, but there was a slight decreasing trend in the eastern MCM, where a seesaw pattern was apparent. However, in the 21~(st) century, the area-averaged annual precipitation displayed a significant increasing trend in both the western and eastern MCM.(2) The trend in area-averaged seasonal precipitation during 1960–2014 in the western MCM was consistent with that in the eastern MCM in winter and spring. However, the trend in area-averaged summer precipitation during1960–2014 displayed a seesaw pattern between the western and eastern MCM.(3) On an annual basis,both the trend in rainstorms and heavy rain displayed a seesaw pattern between the western and eastern MCM.(4) The precipitation intensity in rainstorms, heavy rain, and moderate rain made a greater contribution to changes in the total precipitation than precipitation frequency. The results of this study will improve our understanding of the trends and differences in precipitation changes in different areas of the MCM. This is not only useful for the management and mitigation of flood disasters, but is also beneficial to the protection of water resources across the MCM.  相似文献

ABSTRACT The root-knot nematode Meloidogyne incognita is a major pathogen of a range of important crops. Currently, control is typically achieved by the use of nematicides. However, recent work suggests that manipulating the ability of roots to slough off border cells, which then act as a decoy to the nematode, can significantly decrease damage to the roots. We investigated the attractiveness of border cells to M. incognita and the response of the nematode to border cells in close proximity. We found very limited attraction, in that nematodes did not preferentially alter direction to move toward the border cells, but a large and significant increase in nematode speed was observed once they were in the immediate vicinity of border cells. We discuss the results in the context of physical and biological mechanisms in relation to the control of pathogenic nematodes.  相似文献

Detecting near-surface soil freeze-thaw cycles in high-altitude cold regions is important for understanding the Earth's surface system, but such studies are rare. In this study, we detected the spatial-temporal variations in near-surface soil freeze-thaw cycles in the source region of the Yellow River(SRYR) during the period 2002–2011 based on data from the Advanced Microwave Scanning Radiometer for the Earth Observing System(AMSR-E). Moreover, the trends of onset dates and durations of the soil freeze-thaw cycles under different stages were also analyzed. Results showed that the thresholds of daytime and nighttime brightness temperatures of the freeze-thaw algorithm for the SRYR were 257.59 and 261.28 K, respectively. At the spatial scale, the daily frozen surface(DFS) area and the daily surface freeze-thaw cycle surface(DFTS) area decreased by 0.08% and 0.25%, respectively, and the daily thawed surface(DTS) area increased by 0.36%. At the temporal scale, the dates of the onset of thawing and complete thawing advanced by 3.10(±1.4) and 2.46(±1.4) days, respectively; and the dates of the onset of freezing and complete freezing were delayed by 0.9(±1.4) and 1.6(±1.1) days, respectively. The duration of thawing increased by 0.72(±0.21) day/a and the duration of freezing decreased by 0.52(±0.26) day/a. In conclusion, increases in the annual minimum temperature and winter air temperature are the main factors for the advanced thawing and delayed freezing and for the increase in the duration of thawing and the decrease in the duration of freezing in the SRYR.  相似文献

Worldwide, there is increasing focus on implementation of Quality Assurance systems (QA-systems) in plant health diagnostic laboratories. Several laboratories are in the development or implementation phase and some laboratories have gained accreditation through approval by national accreditation boards. To initiate the process of developing and implementing QA-systems, management and staff need a strong motivation factor. First, because it is a time-consuming and demanding process to go through. Second, because plant health testing does not fit very well into the QA-systems that traditionally were developed for chemical or physical testing laboratories. External pressure is often the only way to generate this motivation factor amongst staff and management to initiate the development of QA-systems. The principal motivation factor in our laboratory was a national requirement that official testing laboratories should implement QA-systems. At the Danish Plant Directorate (PD) we have gained experiences with accreditation of plant health diagnostic methods during the past 5 years. The focus of this paper is a presentation of the consequences and the practical approach to comply with the requirements of ISO 17025 in our plant health diagnostic laboratory. This includes the themes: staff competence and responsibilities, documentation and traceability, and continuous assessment and improvement of the QA-system.  相似文献

Drought-prone grasslands provide a critical resource for the millions of people who are dependent on livestock for food security.However,this ecosystem is potentially vulnerable to climate change(e.g.,precipitation)and human activity(e.g.,grazing).Despite this,the influences of precipitation and grazing on ecological functions of drought-prone grasslands in the Tianshan Mountains remain relatively unexplored.Therefore,we conducted a systematic field investigation and a clipping experiment(simulating different intensities of grazing)in a drought-prone grassland on the northern slopes of the Tianshan Mountains in China to examine the influences of precipitation and grazing on aboveground biomass(AGB),soil volumetric water content(SVWC),and precipitation use efficiency(PUE)during the period of 2014–2017.We obtained the meteorological and SVWC data using an HL20 Bowen ratio system and a PR2 soil profile hydrometer,respectively.We found that AGB was clearly affected by both the amount and seasonal pattern of precipitation,and that PUE may be relatively low in years with either low or excessive precipitation.The PUE values were generally higher in the rapid growing season(April–July)than in the entire growing season(April–October).Overall,moderate grazing can promote plant growth under water stress conditions.The SVWC value was higher in the clipped plots than in the unclipped plots in the rapid growing season(April–July),but it was lower in the clipped plots than in the unclipped plots in the slow growing season(August–October).Our findings can enhance the understanding of the ecological effects of precipitation and grazing in drought-prone grasslands and provide data that will support the effective local grassland management.  相似文献

One goal of integrated pest management (IPM) as it is currently practiced is an overall reduction in fungicide use in the management of plant disease. Repeated and long‐term success of the early broad‐spectrum fungicides led to optimism about the capabilities of fungicides, but to an underestimation of the risk of fungicide resistance within agriculture. In 1913, Paul Ehrlich recognized that it was best to ‘hit hard and hit early’ to prevent microbes from evolving resistance to treatment. This tenet conflicts with the fungicide reduction strategies that have been widely promoted over the past 40 years as integral to IPM. The authors hypothesize that the approaches used to implement IPM have contributed to fungicide resistance problems and may still be driving that process in apple scab management and in IPM requests for proposals. This paper also proposes that IPM as it is currently practiced for plant diseases of perennial systems has been based on the wrong model, and that conceptual shifts in thinking are needed to address the problem of fungicide resistance. © 2013 Society of Chemical Industry  相似文献

The progress of colonization of ash stems from ascospore inocula of Hymenoscyphus fraxineus was examined by light and electron microscopy. The main aim of the study was to characterize the cytology of the biotroph to necrotroph transition during lesion formation. Following direct penetration into epidermal cells, the fungus produced intracellular hyphae that invaded up to five cells before plant cells died. A lack of close attachment between the hyphal cell wall and plant cell membrane was revealed by plasmolysis of epidermal cells. Plant cells died at the centre of the infection but hyphae at the edge were typically found in living plant cells even around large lesions. During biotrophic invasion, the cytoplasm of penetrated plant cells showed very little response despite the plant cell membrane being in direct contact with the fungal cell wall. Before plant cell death, dark staining of the cytoplasm and proliferation of small vesicles was noted, but organelles retained normal ultrastructure. Dead plant cells contained dark brown, osmiophilic droplets. Penetration between epidermal or collenchyma cells was usually targeted to shared pits and involved constriction of hyphae. The transition to necrotrophy was not associated with a clear change in hyphal morphology. Biotrophic intracellular hyphae contained dense cytoplasm but hyphae in dead plant cells were more vacuolated. Remarkably little plant cell wall degradation was observed despite the fungus penetrating up to 18 cells deep into stem tissue. Features of the development of the ash dieback fungus are compared with other hemibiotrophic pathogens.  相似文献

The northeastern coast of the Caspian Sea is one of the youngest territories for plant colonization in Kazakhstan,and the flora of the most recent portion of this coastal area,the New Caspian marine plain,was chosen to study phytogeographical characteristics of plant species,especially as related to alien origin and invasive potential.Because of the recent formation of this flora,I expected that a large proportion of the species may be alien and invasive.After compiling a comprehensive species list from previous research,I identified the taxonomic,life form,ecological,and geographical structures of the flora.The area belongs to the northern type of deserts with continental climate and low annual precipitation.Taxonomic and life form structures show that the flora is specific to Central Asian deserts.Ecological structure demonstrates a high percentage of halophytes,which resulted from their adaptation to soil salinity and shallow ground waters that occur throughout the study area.Geoelements of the flora indicate allochtonous properties of the flora(i.e.species derived from areas outside the study area) with a low proportion of native(Caspian) plants.Alien species are estimated to comprise 12%-19% of the flora.Although some species have invasive features,severe arid environmental conditions and high salinity of soils and ground waters decrease invasive capacity of exotic plants.However,a threat of expansion of potentially invasive plants exists in the New Caspian marine plain due to the fast growth of anthropogenic habitats associated with recent oil developments.  相似文献

Researching into changes in thermal growing season has been one of the most important scientific issues in studies of the impact of global climate change on terrestrial ecosystems. However, few studies investigated the differences under various definitions of thermal growing season and compared the trends of thermal growing season in different parts of China. Based on the daily mean air temperatures collected from 877 meteorological stations over northern China from 1961 to 2015, we investigated the variations of the thermal growing season parameters including the onset, ending and duration of the growing season using the methods of differential analysis, trend analysis, comparative analysis, and Kriging interpolation technique. Results indicate that the differences of the maximum values of those indices for the thermal growing season were significant, while they were insignificant for the mean values. For indices with the same length of the spells exceeding 5°C, frost criterion had a significant effect on the differences of the maximum values. The differences of the mean values between frost and non-frost indices were also slight, even smaller than those from the different lengths of the spells. Temporally, the starting date of the thermal growing season advanced by 10.0–11.0 days, while the ending dates delayed by 5.0–6.0 days during the period 1961–2015. Consequently, the duration of the thermal growing season was prolonged 15.0–16.0 days. Spatially, the advanced onset of the thermal growing season occurred in the southwestern, eastern, and northeastern parts of northern China, whereas the delayed ending of the thermal growing season appeared in the western part, and the length of the thermal growing season was prolonged significantly in the vast majority of northern China. The trend values of the thermal growing season were affected by altitude. The magnitude of the earlier onset of the thermal growing season decreased, and that of the later ending increased rapidly as the altitude increased, causing the magnitude of the prolonged growing season increased correspondingly. Comparing the applicability of selected indices and considering the impacts of frost on the definitions are important and necessary for determining the timing and length of the thermal growing season in northern China.  相似文献

Microclimates play important roles in controlling water budgets and water vapor transportation, as well as vegetation growth characteristics. In order to understand the differences in meteorological parameters under different vegetation cover (VC) and terrain conditions, wind velocity, air temperature, relative humidity (RH), photosynthetically active radiation (PAR), and solar radiation were simultaneously monitored on shifting, semi-fixed and fixed sandy lands, and an oasis. The air temperature and RH differences among the four landscape types were obvious in the period from May to October. It was found that the higher elevations of semi-fixed sandy land can influence PAR and solar radiation during sand-blowing weather. The differences of air temperature and RH among the four sites during the dust-storm and rainy days were not obvious, but their differences during sand-blowing weather were greater than during rainy weather and less than during floating-dust and sunny weather. The differences of PAR and solar radiation among the four landscape types were most obvious during the dust-storm event. During most of the weather types studied, significant positive correlations were found between wind velocity and temperature, PAR, and solar radiation, as well as between temperature and PAR, and solar radiation. Meanwhile, significant negative correlations were found between RH and wind velocity, temperature, PAR, and solar radiation. VC and topography were found to be the main factors influencing the changes in meteorological parameters between desert–oasis ecotone and oasis.  相似文献

ABSTRACT

The aim of this study was to evaluate whether foraging decisions of Acromyrmex lundi in the field are affected by the extract of Aristolochia argentina. A free choice test was performed using Rosa x hybrida leaves treated with the extract and control with acetone. Two leaves (treated and control) were presented on either side of a foraging trail. The percentage of removed material was recorded for 90 minutes of observation on two consecutive days. Dry weight and repellency index were calculated. Ant activity, the percentage of carried leaves and weight data were analyzed using a “t”-test for paired data, and a General Linear Mixed Model test was used to evaluate the different variables and their interactions. A. argentina extract (1%) did not affect the foraging activity of A. lundi, whereas the 5% dose caused significant differences in foraging activity as well as between the factors and the interactions. A repellency index of above 95% was obtained at the 5% dose. The extract of A. argentina could be considered for future management of this insect.  相似文献

To elucidate the early epidemic stages of septoria tritici blotch, especially the relationship between the onset of epidemics, the local availability of primary inoculum, and the presence of wheat debris, the early disease dynamics and airborne concentration in Zymoseptoria tritici ascospores were concomitantly assessed at a small spatiotemporal scale and over two years, using spore traps coupled with a qPCR assay. One plot, with the crop debris left, provided a local source of primary inoculum, while the other plot, without debris, lacked any. According to the assay's limits of detection, daily spore trap samples were classified as not detectable or not quantifiable, detectable, and quantifiable. The proportions of samples assigned to the different classes and numbers of spores in samples classified as quantifiable were significantly different between years, time periods (from November to March) and spore trap location (field with or without debris). The effect of year on the airborne ascospore concentration was high: 22 daily peaks with more than 230 ascospores m?3 day?1 were identified in the autumn of 2012/13, but none in the autumn of 2011/12. The local presence of wheat debris had no obvious effect on the amount of airborne ascospores or on the earliness of the two epidemics, especially in the year with high inoculum pressure (2012/13). These results suggest that the amount of primary airborne inoculum available in a wheat crop is not a limiting factor for the onset of an epidemic.  相似文献
