Nine trees from a single, natural population of black cottonwood (Populus trichocarpa Torr. et Gray) in Alaska were screened for randomly amplified polymorphic DNA (RAPD) markers with ten different 10-base random oligonucleotide primers in order to evaluate the use of RAPD analysis for distinguishing black cottonwood clones. Nine primers amplified the genomic DNA targets; two primers were able to differentiate all clones. Eight clones were distinguished among the nine tree samples assayed. Two trees showed identical banding patterns with all primers used; therefore it is suggested that these trees are from the same clone. The RAPD fingerprinting method is simple and powerful-one primer can distinguish different clones, while the use of multiple primers reduces fingerprint similarity and resolves discrepancies. 相似文献

利用RAPD分析药用石斛的遗传多样性及亲缘关系 总被引:3,自引:0,他引:3
通过对9种药用石斛进行遗传多样性和亲缘关系的分析,为更好地开发利用石斛资源奠定基础。采用RAPD技术,应用NTSYS软件对9种石斛的RAPD-PCR结果进行分析。利用从103条随机引物中筛选出的70条随机引物对供试材料DNA进行随机扩增,共得到520条带,其中多态性带有491条,多态性百分率为94.42%。采用UPGMA类平均法对扩增出的谱带进行遗传聚类分析,得出反映各种间亲缘关系的树状图。在遗传距离(D)=0.44处,可将供试材料分为2类,RAPD对基因组的分析结果与传统分类学的结果差异不大。试验结果表明,RAPD技术可有效地应用于药用石斛的遗传多样性及亲缘关系的研究。 相似文献

从210个随机引物中筛选出的16个引物,共检测到118条带,其中多态性带95个,总多态位点比率为80.37%,充分说明了马尾松无性系间遗传背景的复杂性和多样性。采用Nei指数计算的建园无性系马尾松有效等位基因数平均值为1.3447,总基因多样性为0.2445,遗传多样度(h)平均值为0.2445,Shannon′s信息指数(I)平均值为0.3558,与一些松柏纲植物的RAPD数据资料相比,马尾松的遗传多样性处于较高水平,显示出该无性系马尾松种子园内各马尾松的遗传基础较宽,具备营建出遗传品质好、遗传多样性高、稳定性好的马尾松人工造林的能力,能满足林木育种的需要。 相似文献

We studied the genetic polymorphism among 29 clones of shisham (Dalbergia sissoo Roxb) belonging to different geographic regions using random amplified polymorphic DNA (RAPD) markers. Out of 30 primers used, only 20 primers generated polymorphism in amplified product. In total 232 bands were amplified with 20 primers, of which 192 (82%) were polymorphic with an average of 9.6 bands/primer. The resolving power (Rp) ranged from 2.14 (Primer 5) to 11.93 (Primer 4). Primer 4 and Primer 3 possessed high Rp value. Polymorphism information content (PIC) ranged from 0.15 (Primer 5) to 0.37 (Primer 4). Primer 4 amplified total 18 bands in 29 genotypes with PIC value of 0.37 hence; this set of primer was most informative. The similarity coefficient analysis revealed two clusters. The first cluster comprised of only 10 clones and the second major cluster comprised of 19 clones. The genetic similarity among 29 clones ranged from 25.86% (clone 10 and 235) to 100% (clone 19 and 59), suggesting a wide genetic base in shisham clones. 相似文献

The salt tolerance of various poplar and willow species and clones in the laboratory and field. The resistance of 10 black and 10 grey poplar clones and of 36 willow clones was examined by dipping shoots in a higly concentrated NaCl solution. All the black and most of the grey poplars showed high resistance. Only eleven willow-clones showed sufficient resistance. 运用随机扩增多态DNA(RAPD)技术,对黄心夜合[Michelia martinii(Levl.)Levl.]自然分布区的6个自然种群遗传多样性进行了研究。从100个随机引物中筛选出能产生清晰、稳定的多态性标记引物18个,共检测出110个位点,其中多态性位点为96个,占87.27%;在物种水平上,有效等位基因数目(Ne),Nei’s基因多样性系数(He)和Shannon表型多样性指数(I)分别为1.735 2,0.416 3和0.597 1;总的遗传变异量(Ht)为0.339 8,其中居群内遗传变异量(Hs)为0.258 7,各居群间的遗传分化系数(Gst)达到0.323 1.应用UPGMA法对遗传距离进行聚类分析并构建树系图,结果表明:黄心夜合自然种群具有较高的遗传多样性,其种群间的遗传差异与其地理分布有关。 相似文献

用RAPD技术研究了紫胶虫7种主要生产种12个群体之间的亲缘关系,研究中每个群体使用了24个标本,共有288个个体用于研究.在试用的45种随机引物中,筛选出9种引物用于随机引物扩增,共得到121个清晰稳定的位点,其中118个为多态性位点,多态百分率为97.52%,每条引物检测到的位点数为11~16个,长度为150~3 000 bp.根据扩增结果,用UPGMA法对Nei's遗传距离进行聚类分析,构建分子系统树.7种紫胶虫聚类结果显示:紫胶虫各群体间的遗传距离为0.044 3~0.791 7,其中,种间的遗传平均距离为0.443 0,种内的遗传平均距离为0.170 7.根据聚类分析,讨论了紫胶虫主要生产种的亲缘关系. 相似文献

从80个随机扩增多态性DNA(RAPD)随机引物中筛选出22个谱带清晰且重复性好的引物用于PCR扩增,探讨夹竹桃科(Apocynaceae)10属12种植物的遗传关系。获得162个位点用于计算Nei’s遗传相似性系数,并应用NTSYS程序UPGMA法构建了系统发育树。黄蝉(Allamanda schottiiPohl.)与软枝黄蝉(A.catharticaL.)、红鸡蛋花(Plumeria rubraL.)与鸡蛋花(P.rubraL.‘Acutifolia’)之间的遗传相似性最高;在遗传相似性系数0.73处将12种植物划分为10个属,并在0.58处分为3支,但并不完全支持3个独立的亚科。RAPD分析结果与形态学的分析结果基本一致,可为夹竹桃科系统分类学提供分子生物学的证据。 相似文献

在海滨湿地分别4个盐度梯度,12种杨树、柳树新无性系田间扦插试验结果表明:在苗期,不同杨树、柳树无性系的成活率、保存率、生长量等均与土壤含盐量呈负相关;在出苗期的耐盐能力,1-74杨、1-69杨可达0.241%,35杨为0.188%,J30-16、J35-13、J4-34、J31-17柳为0.089%,其余5个柳树无性系均在0.060%以下;在苗木生长期,其耐盐极限分别为0.343%,0.333%和0.201%。 相似文献

能源林作为生产并提供生物质能源的一种重要方式, 以其可再生、生物量大、环境友好及适应地域广而倍受国际社会关注。文中依据其主要用途将能源林分为燃油能源林、生物发电能源林与薪炭能源林等3类。燃油能源林树种包括续随子、霍霍巴、油楠等, 生物发电能源林树种包括柳树、桉树、杨树等, 薪炭能源林树种相对较多。生物发电能源林与薪炭能源林可统称为燃料能源林。目前, 国外十分重视燃料能源林树种选育及高产培育技术研究和实践。不同树种、无性系、种源的燃料能源林生物产量差异很大, 瑞典选育出蒿柳和毛枝柳等, 南魁北克选育出了柳树无性系SX64与SX61。造林密度与收获周期紧密相关, 巴西的澳洲金合欢树3年收获时密度以10000株/hm2生物量最大; 欧洲2年收获与3年收获的柳树能源林适宜密度为15000株/hm2。另外, 文中对能源林类型、燃料能源林树种选育、立地条件、造林整地、造林密度、收获周期、抚育管理等技术进行了介绍, 希望对能源林的栽培提供依据。 相似文献

Malabar glory lily (Gloriosa superba L.) is a medicinally potent plant species used for the production of alkaloid colchicine. With ever increasing demand, there is a pressing need to conserve it through biotechnological approaches. A large number of complete plantlets were obtained by direct regeneration from the non-dormant tuber explants on Murashige and Skoog (MS) medium supplemented with 2.0 mg/l 6-benzylaminopurine (BAP) + 0.5 mg/l α-naphthalene acetic acid (NAA). Large number of plants can be produced in vitro under aseptic conditions, but there is always a danger of producing somaclonal variants by tissue culture technology. Thus, the genetic stability of micropropagated clones was evaluated using random amplified polymorphic DNA (RAPD) and inter simple sequence repeat (ISSR) analysis. During the study a total of 80 (50 RAPD and 30 ISSR) primers were screened, out of which 10 RAPD and 7 ISSR primers produced a total of 98 (49 RAPD and 49 ISSR) clear, distinct and reproducible amplicons. The amplification products of the regenerated plants showed similar banding patterns to that of the mother plant thus demonstrating the homogeneity of the micropropagated plants. This is the first report that evaluates the use of genetic markers to establish genetic fidelity of micropropagated G. superba using RAPD and ISSR, which can be successfully applied for the mass multiplication, germplasm conservation and further genetic transformation assays for colchicine production to meet the ever increasing demand of this medicinally potent plant for industrial and pharmaceutical uses. 相似文献
