Objective-To determine glycohistochemical characteristics of enzootic nasal tumors (ENTs) of sheep, compare results for ENT with those of histologically normal nasal mucosa of sheep, and identify the histologic origin of ENT. Sample-ENT and nasal mucosa samples obtained from cadavers of 5 adult Lacaune sheep with ENT and 5 Lacaune sheep unaffected by ENT, respectively. Procedures-Samples of ENT and nasal mucosa were collected from cadavers of sheep and sectioned. Conventional and lectin histochemical analyses were used to identify glycoconjugates in tissue sections on the basis of their principal chemical groups and principal terminal or internal oligosaccharidic glucidic residues, respectively. Results-ENTs had papillary and tubular portions. Cells in the papillary portion of ENTs had secretion and surface glycoconjugates, which included sulfated glycosaminoglycans and neutral and sialilated glycoproteins. Cells in the tubular portion of ENTs had surface glycoconjugates, which included neutral and sialilated glycoproteins. Both portions of ENTs had C(4)-acetylated sialoderivatives that were not detected in sections of histologically normal nasal mucosa. Conclusions and Clinical Relevance-The papillary portion of ENTs in sheep may originate from respiratory glands and goblet cells. The tubular portion of ENTs in sheep may originate from olfactory glands. Presence of C(4)-acetylated sialoderivatives in cells of ENTs could confer resistance against pathogens to those cells.  相似文献

Olfactory ensheathing cells (OECs) have been reported to promote axonal regeneration when transplanted to rodent spinal cord injury models. OECs are available from the olfactory bulb (OB) and olfactory mucosa (OM). Although harvesting OECs from the OM is less traumatic, OECs originating from the OM are less proliferative than those from the OB (OB-OECs). One possible reason for this difference is coexisting fibroblasts. Here, we examined the effect of coculturing either fibroblasts from the OB (OB-Fibs) or fibroblasts from the OM (OM-Fibs) on the proliferation of OB-OECs. Proliferation of OB-OECs was significantly higher in 5:5 coculture with OB-Fibs and in 7:3 and 5:5 cocultures with OM-Fibs than without fibroblasts. These results indicated that coculture with both OB-Fibs and OM-Fibs promoted the proliferation of OB-OECs.  相似文献

Three dimensional vascular architecture in the omasal laminae of sheep was studied by scanning electron microscopy of corrosion casts. The omasal laminae were equipped with well-developed subepithelial capillary network with various capillary loops and inter-papillar undulations. The capillary loops were more highly developed in the omasal papillae than in the inter-papillar space and more in the oral region than in the aboral. The capillary loops were inconspicuous in the inter-papillar space of the middle and aboral regions and in the omasal papillae of the aboral region. On the other hand, the inter-papillar undulation of the capillary network was more conspicuous in the aboral region than in the other regions of the omasal mucosa. These findings may indicate a regional diversity in the function of the omasal laminae.  相似文献

Transfer of six amino acids through rumen epithelium of sheep has been studied on mucous and serous sides. Amino acids were classified into two groups: 1. lysine-arginine-glutamic acid, 2. glycine-alanine-leucine. The transfer of amino acids in triads was measured after 60 min. incubation. To determine amino acids transfer through rumen epithelium, three concentrations of amino acids were used: 1500: 150 and 15 mumol per 50 ml. In the first group of amino acids, the highest transfer of arginine was found in comparison with lysine and glutamic acid in all concentrations. The lowest transfer was found in glutamic acid. It follows from this that initial concentration of amino acids in the first group had no effect on amino acids transfer as far as the transfer of amino acids was as follows in all concentrations: arginine greater than lysine greater than glutamic acid. On the other hand, the composition of amino acids and initial concentration of amino acids in the second group exhibited the marked dependence on the transport of amino acids. The transfer of amino acids at the concentration of 1500 mumol per 50 ml was as follows: the passage of amino acids was the highest while this was lowest in alanine and leucine, at the concentration 150 mumol per 50 ml the highest transfer was found in alanine, it was lower in glycine and lowest in leucine and at the concentration 15 mumol per 50 ml the transfer of glycine was highest, lower in leucine and lowest in alanine.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

An extramedullary plasmacytoma was found in a 10-year-old sheep. The tumor involved the mediastinum, where a 25 x 15 x 10-cm encapsulated mass was found. The lungs had multiple metastases ranging from 0.5 to 2 cm in diameter, and the portal vein contained a 10-cm-long mass. The cytologic and histopathologic analyses were consistent with a moderately differentiated plasmacytoma. The immunophenotype of the tumor cells was lambda light chain IgG+, CD79a-, and CD3-. Occasional granulomas were observed at the periphery of the mediastinal and pulmonary tumors. Microbiologic culture yielded growth of Corynebacterium from these granulomas. This is the first report of plasmacytoma in sheep. The tumor most likely arose from mediastinal lymph nodes and metastasized to the lungs and portal vein.  相似文献

BackgroundThe olfactory mucosa (OM) is crucial for odorant perception in the main olfactory system. The terminal carbohydrates of glycoconjugates influence chemoreception in the olfactory epithelium (OE).ObjectivesThe histological characteristics and glycoconjugate composition of the OM of Korean native cattle (Hanwoo, Bos taurus coreae) were examined to characterize their morphology and possible functions during postnatal development.MethodsThe OM of neonate and adult Korean native cattle was evaluated using histological, immunohistochemical, and lectin histochemical methods.ResultsHistologically, the OM in both neonates and adults consists of the olfactory epithelium and the lamina propria. Additionally, using periodic acid Schiff and Alcian blue (pH 2.5), the mucus specificity of the Bowman’s gland duct and acini in the lamina propria was determined. Immunohistochemistry demonstrated that mature and immature olfactory sensory neurons of OEs express the olfactory marker protein and growth associated protein-43, respectively. Lectin histochemistry indicated that numerous glycoconjugates, including as N-acetylglucosamine, mannose, galactose, N-acetylgalactosamine, complex type N-glycan, and fucose groups, were expressed at varied levels in the different cell types in the OMs of neonates and adults at varying levels. According to our observations, the cattle possessed a well-developed olfactory system, and the expression patterns of glycoconjugates in neonatal and adult OMs varied considerably.ConclusionsThis is the first study to describe the morphological assessment of the OM of Korean native cattle with a focus on lectin histochemistry. The findings suggest that glycoconjugates may play a role in olfactory chemoreception, and that their labeling properties may be closely related to OM development and maturity.  相似文献

In recent publications, it was shown that disease-associated prion protein (PrP(d)) accumulates in the lymphoid tissue of the rectal mucosa of a high proportion of scrapie-infected sheep at clinical and preclinical stages, regardless of several host factors; PrP(d) can also be detected in biopsy specimens of rectal mucosa, with an increased probability proportional to age or incubation period and with an efficiency almost identical to that of tonsil biopsies. Rectal biopsies have the advantages of providing higher numbers of lymphoid follicles and of being simpler to perform, which makes them suitable for scrapie screening in the field. In biopsy samples, PrP(d) could be demonstrated by immunohistochemical (IHC) and Western immunoblotting methods, and the purpose of the present study was to optimize and evaluate a "rapid test" for the diagnosis of scrapie in rectal biopsy samples. The HerdChek CWD (chronic wasting disease) antigen EIA (enzyme immunoassay) test was chosen and, once optimized, provided specificity and sensitivity figures of 99.2% and 93.5%, respectively, compared with IHC results in the same samples obtained at a postmortem. The sensitivity of the assay increased from 82.1%, when a single rectal mucosa sample was tested to 99.4% for those sheep in which 3 or more samples were analyzed. Similarly, sensitivity values of the HerdChek CWD antigen EIA test on biopsy samples increased from 95% to 100% for sheep subjected to 1 or 2 sequential biopsies 4 months apart, respectively. Thus, a preclinical diagnosis of scrapie in live sheep can be achieved by a combination of a simple sampling procedure, which can be repeated several times with no detrimental effect for the animals, and a rapid and efficient laboratory method.  相似文献

A total of 214 sheep with lesions of the oral mucosa were recorded at the Veterinary Laboratories Agency regional laboratories between March 15, 2002 and February 28, 2003. Using denominator data from the scrapie abattoir survey a prevalence of 0.95 per cent (95 per cent CI 0.82 to 1.10 per cent) was recorded, and using the fallen stock survey a prevalence of 1.15 per cent (95 per cent CI 0.64 to 1.91 per cent) was recorded. The lesions varied widely and included dental pad lesions in 20 per cent of cases. In total, 248 sites were affected, the commonest being the lower gum below the incisors, which was affected in 100 sheep. Most of the lesions were 1 cm or less in diameter. Ninety per cent of the 251 lesions examined histologically were erosions, ulcers, healed ulcers, focal epithelial necrosis or haemorrhages, and the changes observed indicated that trauma was the most likely primary cause.  相似文献

Vinblastine sulphate (VBS) is an anticancer drug that acts by disrupting microtubule dynamics of highly mitotic tissue cells. The consequences of VBS on the olfactory mucosa (OM), a tissue with high mitotic numbers, are not clearly understood. We used qualitative and quantitative methods to determine the structural changes that may be produced on the rabbit OM by VBS. Following a single dose (0.31 mg/kg) of this drug, the structure of the mucosa was greatly altered on the first 3–5 days. The alteration was characterized by disarrangement of the normal layering of nuclei of the epithelia, degeneration of axonal bundles, occurrence of blood vessels within the bundles, localized death of cells of Bowman's glands and glandular degeneration. Surprisingly on or after day 7 and progressively to day 15 post‐exposure, the OM was observed to regenerate and acquire normal morphology, and the vessels disappeared from the bundles. Relative to control values, bundle diameters, olfactory cell densities and cilia numbers decreased to as low as 53.1, 75.2 and 71.4%, respectively, on day 5. Volume density for the bundles, which was 28.6% in controls, decreased to a lowest value of 16.8% on day 5. In contrast, the volume density for the blood vessels was significantly lower in controls (19.9%) than in treated animals at day 2 (25.8%), day 3 (34.3%) and day 5 (31.5%). These findings suggest that the changes induced on the rabbit OM by VBS are transient and that regenerative recovery leads to the restoration of the normal structure of the mucosa.  相似文献

Sengis are eutherian insectivores belonging to superorder Afrotheria, a recently defined clade of mammals that diverged from other placentals over 100 million years ago. In this study, a histological and ultrastructural analysis was carried out on the olfactory mucosa (OM) of the rufous sengi (Elephantulus rufescens) and the data were compared with those reported earlier in the dog (Canis familiaris) and the sheep (Ovis aries), whose dietary lifestyles are carnivorous and herbivorous, respectively. Qualitatively, the microstructure of the sengi's OM was basically similar to that of the other eutherian mammals except for differences in the pattern of cilia projection from the dendritic knobs of the olfactory receptor neurons (ORNs) and distribution of Bowman's glands within the lamina propria of the OM. On morphometry, significant differences (p < .05) were recorded with respect to olfactory epithelial (OE) thickness between the sengi (65.4 ± 2.6 μm) and the other species. ORN packing density and cilia number/ORN knob varied markedly only between the sengi (73.8 ± 5.4 mm?2 × 103 and 15 ± 4, respectively) and the sheep. No remarkable differences were noted in regard to ORN bundle diameters between sengis (62.7 ± 12.5 μm) and the other species. The observed differences in OM structural refinement may be attributed to olfactory function demand levels related to feeding lifestyles and ecology. Myrmecophagous insectivory, social monogamy, absentee maternal care and exposed sheltering habits are behavioural features that may warrant substantial OM modification in sengis.  相似文献

A 2-year old Thoroughbred filly was examined for a 1-month history of persistent nasal discharge. Contrast radiography revealed a circumscribed mass within the right maxillary sinus which extended to the frontal sinus and ethmoid labyrinth. A discrete attachment of the mass to the ethmoid labyrinth was identified at surgery. Surgical removal of the mass eliminated the nasal discharge. On gross examination, the external structure of the mass was similar to a turbinate with a thin bony wall covered by a smooth mucosal membrane. The internal structure of the lesion had a lining membrane with multiple 1-3 cm in diameter fluid filled cystic structures. The histological appearance of the multiloculated structure was similar to the ethmoid labyrinth. This, combined with the single site of attachment to the ethmoid labyrinth, suggested that the cyst was ethmoidal in origin.  相似文献

Aerosol transmission in equine Getah virus (GV) infection was examined by intranasal inoculation with 10(3.0) to 10(7.0) TCID50 of the MI-110 strain in 7 experimental horses. The establishment of intranasal infection of GV was confirmed in all these horses by detecting serum neutralizing antibody against the MI-110 strain. Horses inoculated with more than 10(4.0) TCID50 of the virus manifested mild pyrexia, eruptions, serous nasal discharge, lymphopenia or monocytosis. Viremia ranging from 10(1.0) to 10(3.5) TCID50/0.2 ml occurred in horses inoculated with 10(5.0) TCID50, or more. Virus recovery from the nasal cavity was observed only in horses inoculated with 10(7.0) TCID50, and the viral titers recorded were 10(3.0) TICD50/ml or less. From these results, it is assumed that GV disseminated from the nasal cavity of naturally infected horses, except for intranasal infection with a lot of the virus, is probably very low in titer. So it seems to be rare that GV in natural cycles is spread from horse to horse by aerosol transmission.  相似文献

Chickens were inoculated intranasally with encapsulated or nonencapsulated strains of Haemophilus paragallinarum. The nasal mucosa of the chickens were examined, using scanning and transmission electron microscopy. In chickens given the encapsulated variant, marked loss of cilia and microvilli, infiltration of leukocytes, and deposition of a mucopurulent substance were seen on the surface of the nasal mucosa; the number of microvillous cells were markedly increased, and infiltration of mast cells into the lamina propria of the mucous membrane was observed. Numerous encapsulated organisms were found near cilia and on microvillous cells, and the capsule surrounding the organisms appeared to mediate attachment of the organisms to the cilia. In contrast, the nasal mucosa of the chickens given the nonencapsulated variants changed little morphologically. A few nonencapsulated organisms were found near cilia. Colonization of the nasal mucosa by encapsulated organisms probably was essential to induce the morphologic changes seen in the nasal mucosa.  相似文献

AIMS: Both adult and larval Ostertagia circumcincta infections raise abomasal pH and serum gastrin and pepsinogen concentrations, either because of physical effects of the worms or from their chemical secretions. The study was designed to examine whether adult worms require contact with the gastric mucosa to effect changes in gastric secretion. METHODS: Abomasal pH and serum gastrin and pepsinogen concentrations were measured in four groups of sheep: Group A (n = 4): abomasal contents containing about 18,000 adult O. circumcincta were obtained from donor sheep, concentrated and infused into 5 microm mesh porous bags attached to surgically implanted abomasal cannulae. A second worm transplantation was carried out 1 week later to assess worm survival after 16 hours. Group B (n = 4): about 9000 adult worms, recovered after migration out of abomasal contents set in agar, were placed in small 5 microm mesh bags which were inserted through indwelling abomasal cannulae and left for 3 days. Group C (n = 2): about 3000 adult worms from the population recovered from agar were infused through abomasal cannulae which allowed. free movement in the abomasum. Group D (n = 3) was left uninfected. RESULTS: Worms transplanted directly into the abomasum (Group C) caused rapid and marked effects on abomasal secretion. Adult O. circumcincta died within 16 hours of transfer into the abomasum when they were restrained within porous bags. Nevertheless, in sheep receiving 18,000 worms, abomasal pH increased soon after new feed was presented on Days 1, 2 and 4 after worm transfer; serum gastrin was elevated in three sheep from 113 hours and serum pepsinogen increased in one animal. Sheep receiving 9000 worms showed similar trends but the results were equivocal. CONCLUSIONS: Adult O. circumcincta prevented from physical contact with the gastric mucosa by restraint in porous bags are able to raise abomasal pH. This study implicates parasite excretory-secretory products in mediating changes in gastric secretion caused by adult abomasal worms.  相似文献

Nineteen gnotobiotic piglets, four to six days old, from three different litters, were inoculated intranasally on three consecutive days with a pure culture of Bordetella bronchiseptica. Necropsy 10 to 30 days after inoculation revealed atrophic rhinitis lesions in all and pneumonia in 13 (73 per cent)infected piglets. One died of septicaemia two days after inoculation. Agglutinating antibodies were detected in the sera of all piglets necropsied from 17 to 30 days after inoculation. Five control piglets showed no lesions or serological changes.  相似文献

Olfactory glial cells have been the focus of much recent research interest because of their possible future use as cellular transplants in repair of spinal cord injury. Although olfactory glial cells can be collected from the olfactory bulb for in vitro culture, alternative sites would be preferable for safer surgical access. This study was designed to investigate the distribution of olfactory sensory neurons and olfactory glial cells within the canine peripheral olfactory system. Using immunohistochemistry and electron microscopy on perfused tissue we demonstrate that olfactory sensory neurons are found in both the caudal nasal and the frontal sinus epithelia. Olfactory ensheathing glia were found in the mucosa at both these sites implying that surgical access for harvesting cells for transplantation would be straightforward.  相似文献
