本试验探讨了促性腺激素(FSH、LH)对猪卵母细胞体外成熟的影响,最终找到了其合理的使用剂量。在成熟液中添加LH浓度分别为1、5、10IU/mL时,与没有添加FSH与LH的对照组(15.9%)相比,成熟率有显著提高,成熟率分别为49.1%、30.0%、36.3%。LH浓度为1IU,mL的实验组成熟率最高,显著高于对照组(P〈0.05);然后以成熟液中只添加1IU/mLLH为对照组,试验组分别添加1、5、10IU/mLPSH。试验结果表明:添加FSH浓度为5和10IU/ml时其成熟率分别为40.1%、27.6%,低于对照组(49.0%)。添加FSH浓度为1IU/mL时其成熟率(49.1%)高于对照组(49.0%),但差异不显著(P〉0.05)。两组试验同时得出,随着LH与FSH添加浓度的增加,成熟率呈下降趋势。通过本试验结果得出,在体外成熟培养猪卵母细胞时,各添加1IU/mL LH与FSH时,得到最佳的成熟效果。 相似文献

Background: Determination of adrenocorticotropic hormone (ACTH) concentration is a commonly used test in the evaluation of endocrine causes of equine laminitis, but the concentration in healthy horses can be high at certain times of year, which alters the specificity of the ACTH test. Objective: To determine if circulating concentrations of ACTH, cortisol, glucose, insulin, and thyroxine vary month to month in healthy horses and in horses with equine metabolic syndrome (EMS). Animals: Nine healthy adult horses were studied on their farm/stable over the course of 1 year. After the diagnosis of EMS, 10 laminitic horses residing at the same farm/stable were also studied. Methods: Prospective study of healthy and laminitic horses. Plasma/serum samples were analyzed for concentrations of hormones and glucose. Results: ACTH was the only analyte to show a discrete seasonal pattern, with concentrations in healthy and EMS horses frequently outside of the reference range (9–35 pg/mL) in August through October. Insulin was elevated (>40 μIU/mL) in EMS horses during most months and median serum glucose was generally higher in EMS horses (100 mg/dL, range, 76–163 mg/ dL) than in controls (94 mg/dL, range, 56–110 mg/dL), but no seasonal patterns for insulin or glucose were found. Conclusions and Clinical Importance: An increased ACTH concentration in horses in late summer or autumn should be interpreted with caution. In contrast, insulin concentration is maintained within the reference range throughout the year in healthy horses, thus an increased insulin concentration at any time of year should raise suspicions of EMS, ECD, or both. 相似文献

The concentrations of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) in the serum of the Bactrian camel (Camelus bactrianus) throughout pregnancy were analysed by radioimmunoassay. The samples were withdrawn at intervals of half a month. The serum LH concentrations varied significantly between 2.2±0.9 and 20.3±18.8 ng/ml during the pregnancy, with the highest value on the day of artificial insemination, which indicated that a preovulatory LH peak appeared. Thereafter, the LH concentration tended to fall, reaching a fairly constant low level by the eighth month of pregnancy. The serum FSH concentrations varied between 7.0±0.2 and 28.9±0.4 ng/ml during gestation, with the peak value being reached at 4.5 months, followed by a marked drop to 7.0±0.2 ng/ml at 7.5 months. 相似文献

为了研究促黄体素(luteinizing hormone,LH)在牛卵母细胞成熟培养体系中的时间依从性,试验通过向牛卵母细胞体外成熟培养的不同时间段添加LH来观察其核成熟的情况。首先用无LH的基础成熟液I和添加LH的成熟液Ⅱ分别培养,在成熟的不同时间点(3、6、9h)用醋酸地衣红染色观察各自的生发泡破裂率。然后在体外成熟培养的不同时间段(0~3、0~6、0~9、0~24h)添加LH,24h后统计各自的第一极体率。结果表明,添加LH组在6h的生发泡破裂率显著低于对照1组(P0.05),24h添加LH组的第一极体率显著高于对照2组(P0.05)。因此,添加LH能延迟牛卵母细胞的核成熟,成熟培养中24h全程添加LH较不添加组能显著提高核成熟率。 相似文献

Thirty-two mature crossbred (Fine Wool Synthetic, FWS) sheep developed for fine wool production in India were treated for superovulation and oestrus synchronization in spring season. The ewes were randomly allocated to four treatment groups in a factorially designed experiment. For induction of superovulation, pregnant mare serum gonadotrophin (PMSG) was administered alone (group 1), in combination with gonadotrophin releasing hormone (GnRH) (group 2) or with follicle stimulating hormone (FSH) (group 3) and with both (group 4). Oestrus was synchronized in all the ewes by two injections of prostaglandin F2 (PGF, 10 mg each) administered at an interval of 10 days. Superovulation treatment started 48 h prior to the second PGF injection. The proportion of ewes in oestrus did not differ significantly (p>0.05) in the four groups. The use of GnRH set the ewes into oestrus earlier than the ewes in the other groups. Treatment with PMSG (800 IU) in conjunction with 4 g of Buserilin (GnRH) increased the ovulation rate (9.1±2.6 corpora lutea (CL)) above that observed when PMSG was used alone (3.0±0.7 CL). The use of FSH (0.5 U ovagen) in conjunction with PMSG was characterized by a decrease in the proportion of ewes with 2 CL (4/8 vs 7/8; p<0.05) and in the number of ovulations, i.e. CL observed (2.4±0.6 vs 9.1±2.6), and a nonsignificant increase in the incidence of large follicles (LF) (4.6±1.28 vs 3.25±0.6; p>0.05). The interaction between treatments of FSH and GnRH was not significant (p>0.05).It is concluded that use of GnRH, in conjunction with either PMSG alone or PMSG plus FSH treatment, advanced the onset of oestrus and increased the ovulation rate in FWS sheep. 相似文献

In general, follicle-stimulating hormone (FSH) and luteinizing hormone (LH) play important roles in the regulation of cumulus cell expansion and oocyte maturation. We investigated the effects of supplementation of FSH or LH in in vitro maturation (IVM) medium on the incidence of cumulus cell expansion and nuclear maturation in canine oocytes. Cumulus-oocyte complexes (COCs) were cultured in TCM-199 supplemented with 10% foetal bovine serum (FBS), 1 mg/ml cysteine, 0.2 mm pyruvic acid and different concentrations of FSH or LH (control, 0.5, 5 or 50 microg/ml) at 38.5 degrees C, 5% CO(2) in air for 72 h. The cumulus cell expansion was measured by microscopic visualization, and nuclear maturation of denuded oocytes was determined by staining with 10 microg/ml Hoechst33342 for 30 min. The cumulus cell expansion in the 5 microg/ml FSH group (397.2 +/- 64.3 microm) was significantly higher than those in the control, 0.5, and 50 microg/ml FSH groups (168.3 +/- 19.1, 286.0 +/- 69.7 and 300.0 +/- 84.3 microm, respectively; p < 0.05). However, there was no difference in cumulus cell expansion among the control, 0.5, 5 and 50 microg/ml LH groups (165.6 +/- 20.2, 160 +/- 26.5, 172 +/- 20.5 and 168 +/- 23.1 microm, respectively; p > 0.05). After 72 h of IVM, the proportion of nuclear development to the MI-MII stage in the 0.5 microg/ml FSH group (15.1%) was higher than those in the control, 0.5 and 50 microg/ml FSH groups (0.9%, 6.5% and 8.0%, respectively; p < 0.05). However, there was no significant difference in nuclear maturation to the MI-MII stage among control, 0.5, 5 and 50 microg/ml LH groups (4.6%, 2.3%, 5.4% and 8.6%, respectively; p > 0.05). This study indicated that a FSH supplement in IVM medium can increase cumulus cell expansion and nuclear maturation, while the nuclear maturation rate remained low. Further studies are required to improve the nuclear development to the MI-MII stages in canine oocytes. 相似文献

本试验用原子吸收光谱法(AAS)和ELISA法分别测定了48头奶牛血清中Zn、Fe含量和促黄体激素(LH)、促卵泡激素(FSH)的浓度。结果表明,不同年龄奶牛血清Zn和Fe含量以7岁和5岁为最高;LH以4岁为最高(P0.01),FSH至4岁是最低(P0.01),然后升高;Zn与FSH、Fe与LH和FSH均呈负相关(P0.05);这些都和回归方程中截距A的正负号是完全一致的,同一种激素与不同元素回归方程的截距和回归系数均明显不同。可以得出不同年龄奶牛血清激素和微量元素含量变化较大,微量元素和激素间存在明显的相关性。 相似文献

本试验旨在研究日粮赖氨酸水平对绿壳蛋种鸡生产性能、血清激素及卵巢FSH受体(FSHR)、LH受体(LHR)mRNA表达的影响。试验选用30周龄的新杨父母代绿壳蛋鸡720只,随机分成6组,每组5个重复,每个重复24只鸡,分别饲喂赖氨酸水平为0.60%、0.65%、0.70%、0.75%、0.80%和0.85%的日粮。结果表明:0.65%赖氨酸水平组产蛋率显著高于0.85%水平组(P0.05),0.70%赖氨酸水平组料蛋比显著低于0.60%水平组(P0.05);血清FSH和LH含量均以0.70%赖氨酸水平组最高(P0.05);与0.75%和0.85%赖氨酸水平组相比,0.65%水平组卵巢FSHR mRNA的表达量显著提高(P0.05);血清FSH含量与卵巢FSHR、LHR mRNA表达量呈现较强相关性(r~20.85)。二次曲线分析表明,以产蛋率和料蛋比为主要评价指标,绿壳蛋种鸡赖氨酸需要量分别为0.70%和0.73%。综上所述,日粮赖氨酸水平为0.70%~0.73%时能够提高新杨绿壳蛋种鸡(31~43周龄)生产性能、血清促性腺激素含量及卵巢FSHR基因的表达。 相似文献

试验旨在研究暖季放牧加补饲对褐牛公犊生长过程中不同时期血清激素水平变化的影响。选取10月龄新疆褐牛公犊70头,分为对照组(放牧)和试验组(放牧加补饲),分别于0、30、60、90和120 d时,测定公犊体重,并采集血样进行生长激素(growth hormone,GH)、睾酮(testosterone,T)、胰岛素(insulin,INS)、生长抑制素(somatostatin,SS)、甲状腺素(T4)、三碘甲腺原氨酸(T3)及糖皮质激素(corticosteroid,CORT)含量的检测。结果表明,在试验全期,两组公犊牛血清中GH含量差异均不显著(P>0.05);在90 d时,试验组公犊牛血清中T含量显著高于对照组(P<0.05),30、60和120 d时,两组差异均不显著(P>0.05);在60和90 d时,试验组公犊牛血清T4含量极显著高于对照组(P<0.01);在30、60和120 d时,两组公犊牛T3含量差异均不显著(P>0.05),90 d时,试验组公犊牛血清T3含量显著高于对照组(P<0.05)。提示,暖季天然草场放牧新疆褐牛公犊为其补饲合理的蛋白质和能量等营养物质能极显著提高公犊牛的日增重(P<0.01)。 相似文献

This experiment was conducted to investigate the effect of naloxone (NAL), an opioid receptor antagonist, on pituitary LH secretion in anovulatory Holstein cows during the early postpartum period when cows were expected to be in negative net energy balance. Twenty-three cows (11 primiparous) received either saline (n = 12) or 1 mg/kg BW NAL i.v. (n = 11) on Day 14 or 15 postpartum. Jugular blood samples were collected at 15-min intervals for 2 hr before and 2.5 hr after NAL or saline. All cows received 3 ug gonadotropin releasing hormone (GnRH) at 2.5 hr post-NAL or -saline and blood collection was continued for 1 hr. Mean serum progesterone concentration was 0.33 ± 0.2 ng/ml. Mean net energy balance for all cows was -5.5 ± 0.6 Mcal/day. Naloxone caused a transient increase (P < 0.05) in serum LH concentrations in both primi- and multiparous cows within 45 min after administration. In contrast, serum LH concentrations remained unchanged in saline-treated cows. GnRH increased (P < 0.05) LH and there was no effect of treatment. These results suggest that modulation of LH secretion, at least in part, may be mediated via endogenous opioids in dairy cows before first postpartum ovulation. 相似文献

Twenty-four-hour Profiles of Serum Prolactin and Luteinizing Hormone in Anoestrous Crossbred Bitches
The dynamics of prolactin (PRL) and luteinizing hormone (LH) secretion in the anoestrous bitch is poorly known. Therefore, the objective of this study was to characterize the 24 h profiles of serum PRL and LH in crossbred anoestrous bitches and to assess whether a relationship exists between the secretory patterns of these two hormones. Serum PRL and LH concentrations were measured in 10 healthy anoestrous crossbred bitches at 145 min intervals for 24 h. During the experiment the animals received continuous artificial illumination and remained undisturbed except at the time of blood sampling. Serum PRL was measured by a homologous enzyme‐linked immunosorbent assay, whereas LH and progesterone (P4) were determined by radioimmunoassay. The anoestrous state of the bitches was assessed by vaginal cytology, vaginoscopy and physical examination. Two groups of animals were identified according to their PRL levels: a high PRL group (n=3, mean ± SEM 12.3 ± 2.7 ng/ml) and a low PRL group (n=7, mean ± SEM: 2.5 ± 0.9 ng/ml). In the low PRL group, the PRL profiles were flat and did not show any significant circadian pattern. Nevertheless, occasional single‐point peaks were detected in some of the bitches. In the high PRL group the individual PRL profiles were variable. To detect the presence of a circadian change of PRL concentrations, two different sets of time windows (TW) of sampling were studied. The first set was: day [TW1A, samples 1–5 (0900–1840 h)] and night [TW1B, samples 6–10 (2105–0645 h)]. The second set was chosen after visual inspection of the average PRL profiles for both (high and low) groups: [TW2A, samples 3–7 (1350–2330 h) and TW2B, samples 1–2 and 8–10 (0155–1125 h)]. PRL concentrations were not significantly different between day and night. In the high PRL group, but not in the low PRL group, average serum PRL was significantly (p < 0.01) higher in TW2A than TW2B. In both groups serum LH levels were more homogeneous than PRL levels. Neither TW showed circadian changes in LH patterns of secretion (TW1A versus TW1B, p < 0.69; TW2A versus TW2B, p < 0.88). On the other hand, bitches in the high PRL group showed significantly (p < 0.01) lower serum LH levels than those in the low PRL group of animals. Serum PRL concentrations presented a significant inverse correlation with LH concentrations (r=?0.21, p < 0.03) and a significant positive correlation with P4 concentrations across the study (0.92, p < 0.01). It is concluded that in anoestrous crossbred bitches serum PRL is highly variable and inversely related to LH. No circadian rhythm of PRL secretion appears to exist in most anoestrous bitches. 相似文献

Kisspeptin (KP),a neuroendocrine regulator of reproduction,is hypothesized to be an integrator of metabolism and hormones critical to the regulation of reproduction.Lactation is associated with enhanced growth hormone (GH) responsiveness and reduced fertility.Our study was designed to determine the effects of lactation on KP-stimulated GH and luteinizing hormone (LH) secretion.Five nonlactating and five lactating dairy cows were used in the study.Experiments were conducted with lactating cows at weeks 1,5 and 11 after parturition.The experimental treatments (saline and KP [ 100 and 400 pmol/kg body weight] ) were given intravenously and blood was collected and plasma was stored until later assay to determine concentrations of GH,LH,progesterone and non-esterified fatty acids.We found that neither dose of KP stimulated an increase in GH secretion.The low dose of KP increased (P <0.05)LH concentrations only in lactating cows.The higher dose of KP elicited an increase in circulating LH concentrations in both lactating and non-lactating cows.The lower dose of KP increased ( P < 0.05 ) the area under the curve for LH only in cows during week 5 of lactation,and the area under the curve of LH following the highest dose of KP was greater ( P < 0.05 ) in cows during week 5 of lactation than that for the other groups of cows.In summary,lactation status and stage of lactation did not change the sensitivity of the GH system to KP.However,an effect of stage of lactation on KP-stimulated LH secretion was detected in the dairy cows.Study of the KP system during lactation in dairy cows may provide critical insights into the mechanisms for lactation-associated changes in the reproductive axis. 相似文献

试验旨在研究日粮中添加不同诱食剂对安格斯犊牛生长性能、养分表观消化率及部分血清激素的影响。采用单因子随机区组设计,选用平均体重165.1 kg±35.5 kg、约6月龄的健康安格斯肉犊牛24头,随机分为A、B、C、D 4组,每组6个重复。A组为对照组,饲喂全混合日粮(TMR);B、C、D组分别饲喂TMR+0.24%绿色素、TMR+1.2 g/d XTRACT® 7065(香味剂)、TMR+0.4%甜蜜素。预饲期10 d,正饲期30 d。结果显示,与对照组相比,B、C、D组平均干物质采食量(ADMI)均显著提高(P < 0.05)。各诱食剂均能提高犊牛营养物质表观消化率,但各诱食剂的影响程度不同,其中B组酸性洗涤纤维(ADF)表观消化率显著高于其他诱食剂组(P < 0.05),B组中性洗涤纤维(NDF)表观消化率显著高于对照组(P < 0.05)。在试验第31天采食后1 h,各组间神经肽Y(NPY)和5-羟色胺(5-HT)含量差异均不显著(P > 0.05),B、D组NPY含量高于对照组,C组则低于对照组,B、D组5-HT含量均低于对照组,而C组则高于对照组。综合考虑,日粮中添加绿色剂、香味剂及甜味剂均可在一定程度上提高犊牛生长性能及营养物质消化率;绿色剂和甜味剂可以提高犊牛血清中NPY含量,而香味剂则使犊牛血清中5-HT含量升高。 相似文献

本试验采用了 3个生产阶段的蛋用鹌鹑。 35日龄开产期蛋用鹌鹑日粮中添加 3mg/kg大豆黄酮 ,鹌鹑的产蛋率和血液中T3 水平均发生显著性提高 (P <0 0 5 )。 7月龄蛋用鹌鹑日粮中添加 3mg/kg大豆黄酮 ,可显著提高鹌鹑的产蛋率 (P <0 0 5 )和血液中T3 水平 (P <0 0 5 ) ;而日粮中添加 6mg/kg大豆黄酮则使鹌鹑的产蛋率显著下降 (P <0 0 5 )。 12月龄蛋用鹌鹑日粮中添加 6mg/kg大豆黄酮 ,鹌鹑的产蛋率提高 10 3% (P <0 0 1) ;血液中T3 、T4和IGF 1水平分别提高 34 8% (P <0 0 1)、2 7 6 % (P <0 0 5 )和 30 8% (P <0 0 5 )。本次研究结果表明 ,日粮中添加一定剂量大豆黄酮能显著增加开产期、高峰期和后期蛋用鹌鹑的产蛋率 ,并影响其内分泌机能 相似文献

The growth hormone (GH) and growth hormone releasing hormone (GHRH) contents in serum of 3 weights (90,110 and 130 kg) of Chuanzang Black swine hybrid and DLY pigs were detected by enzyme-linked immune sorbent assay (ELISA),their relationship with cross sectional area of muscle fiber (CSA) and intramuscular fat (IMF) content were also analyzed.The results showed that the GH contents of Chuanzang Black swine hybrid and DLY pigs of 3 weights were 14.25,14.61,14.11 and 17.86,16.98,16.77 μg/L,respectively,while the GHRH contents of which were 22.88,23.98,24.33 and 27.72,27.47,28.39 μg/L,respectively.There was no significant difference in different weight of the same breeds (P >0.05) except the significant difference between 90 and 130 kg of DLY pigs (P<0.05).The GH and GHRH contents of Chuanzang Black swine hybrid were both significant lower than DLY pigs of the same bodyweight (P< 0.05) except GHRH content of 110 kg weight (P >0.05).Relevance analysis showed that GH and GHRH contents had a significant positive correlation with CSA (P< 0.05),and an extremely significant negative correlation with IMF content (P< 0.01),which prompted that GH and GHRH might regulate the growth of muscle fiber and IMF deposition through blood. 相似文献

采用ELISA方法检测了3个不同体重梯度(90、110和130 kg)的川藏黑猪配套系和DLY猪血清中生长激素(GH)和生长激素释放激素(GHRH)的含量变化,并分析了其与肌纤维面积(CSA)和肌内脂肪(IMF)含量的关系。结果显示,90、110和130 kg体重梯度川藏黑猪配套系和DLY猪血清中GH含量分别为14.25、14.61、14.11及17.86、16.98、16.77 μg/L,GHRH含量分别为22.88、23.98、24.33及27.72、27.47、28.39 μg /L,除DLY猪90和130 kg血清中GH含量差异显著(P<0.05)外,品种内不同体重梯度间差异均不显著(P >0.05);不同猪种间,除110 kg梯度GHRH水平差异不显著(P >0.05)外,其余川藏黑猪配套系均显著低于DLY猪(P< 0.05)。相关性分析结果显示,GH、GHRH水平与CSA呈显著正相关(P< 0.05),与IMF含量呈极显著负相关(P< 0.01)。结果表明GH和GHRH可能通过血液对猪肌纤维的生长和肌内脂肪的沉积进行调控。 相似文献
