OBJECTIVE: To investigate the effect on ovine footrot of repeated daily footbathing in a solution of zinc sulphate with surfactant. DESIGN: Merino sheep were allocated to control and treatment groups of 119 sheep each at week 0. The sheep had a history of S1, U1, T and/or U6 types of Dichelobacter nodosus in interdigital and underrunning footrot lesions. Feet were not pared prior to treatment. PROCEDURE: Treatment sheep were footbathed in a 15 to 18% (w/v) solution of zinc sulphate with surfactant for 10 min on five consecutive days during week 1. At week 2, and fortnightly to week 52, all feet were inspected, lesion scores were recorded and samples were taken for laboratory tests. At week 53, all feet with no lesions at week 52, but with underrunning lesions prior to week 1, were pared and samples were taken. RESULTS: After footbathing, there were no lesions in any treatment sheep at any inspection to week 52. The percentage of feet of control sheep with lesions increased from 9% (391 of 4,284) between weeks 20 and 36, to 14% (593 of 4,284) between weeks 36 and 52. Ninety-five of 96 control sheep with no lesions at week 20 were still asymptomatic at week 52. D nodosus was not isolated from samples taken from 99 and 87 pared feet of treatment and control sheep, respectively. CONCLUSION: Repeated daily footbathing combined with prolonged exposure to a dry environment eradicated footrot in sheep with both interdigital and underrunning lesions in feet that were not pared prior to treatment.  相似文献

OBJECTIVE: To investigate the occurrence of S1, U1 and T strains of Dichelobacter nodosus in new clinical lesions in sheep exposed to footrot associated with multi-strain infections. DESIGN: Seventy-seven donor sheep were grazed with 84 recipients for 33 weeks. The donor sheep were Merinos with a history of clinically virulent footrot associated with protease type S1, U1 and T strains of D nodosus that hybridised with gene sequences pJIR314B, pJIR318 and/or pB645-335. The recipient sheep were Merinos with no history of footrot. PROCEDURE: Each fortnight, all feet were examined, their lesion scores were recorded and samples of lesion material were taken for laboratory tests. RESULTS: Eighty-nine percent (299 of 336) of feet of recipient sheep developed new clinical lesions. S1, U1 and T strains of D nodosus were recovered from 58%, 22% and 18%, respectively, of these lesions at a ratio that remained constant during two apparent peaks in footrot transmission. Gene sequences homologous to pJIR314B and pB645-335 were detected in 56% (93 of 166) and 29% (48 of 166), respectively, of S1 strains of D nodosus at a ratio that was not constant during the experiment. CONCLUSIONS: S1 was the dominant protease type of D nodosus in new clinical lesions. The occurrence of S1 strains did not increase relative to U1 and T strains of D nodosus during the experiment. S1, U1 and T strains of D nodosus remained in equilibrium despite changes in environment, genetic types in the population of S1 strains, and host resistance to footrot.  相似文献

Trials were conducted on 2 commercial sheep flocks in the Gippsland region of Victoria to determine the efficacy of treating ovine virulent footrot by footbathing in aqueous zinc sulphate solution (20% w/v). The effects of foot paring, parenteral penicillin, vaccination and addition of sodium lauryl sulphate (SLS) to the footbaths were assessed. Trial 1 comprised 297 sheep with an initial prevalence of footrot of 33% and most lesions were severe and chronic. Treatment of sheep with unpared feet by zinc sulphate footbathing for 1h did not result in a significant reduction in footrot prevalence (n = 120, cure rate 33%) whereas a significant (P less than 0.01) response was obtained by footbathing for 1h with zinc sulphate/SLS (n = 120, cure rate 55%). Trial 2 comprised 1,042 sheep with a pretreatment footrot prevalence of 71% and predominantly severe lesions. In this flock all treated sheep were footbathed in zinc sulphate/SLS for 1h on 2 occasions, 5 days apart and the effects of additional surgical and parenteral treatments were assessed. Foot paring had a significant detrimental effect on cure rate (P less than 0.01). The administration of procaine penicillin at the time of the first footbathing with zinc sulphate/SLS made no significant improvement to the rate of cure. Footrot vaccine given 8 and 2 weeks prior to footbathing did not cure significantly more sheep than footbathing alone, but the results were significantly better than from foot paring plus footbathing, and from combined foot paring, footbathing and parenteral penincillin treatment (P less than 0.01). The cure rate was 84% for sheep that were only footbathed, 72% for those foot pared and footbathed, 72% for those foot pared, footbathed and given penicillin, and 88% for those vaccinated and footbathed.  相似文献

Ovine footrot is a contagious disease of sheep that occurs in temperature climates. It is caused by the strict anaerobe, Dichelobacter nodosus. Benign and virulent organisms are differentiated according to serotype and protease production. This study was conducted to identify the presence of virulent serotypes of D. nodosus in sheep flocks in Alberta and British Columbia. Dichelobacter nodosus was detected in lame sheep from 11 of 15 (73%) flocks in Alberta and in 4 of 5 (80%) British Columbia flocks. It was recovered from 57 of 107 (53%) lame sheep. In Alberta, 4 distinct serotypes were isolated from the 11 positive flocks while in British Columbia a total of 6 different serotypes were isolated. One British Columbia isolate could not be classified into existing serotypes. Of the 19 field strains tested, all but 3 were defined as virulent based upon the rapid rise in protease activity in vitro which was maintained between 3 and 5 d. The knowledge of the serotype and virulence of the D. nodosus isolated from affected animals can assist in the control and prevention of ovine footrot.  相似文献

Bacteroides nodosus was recovered from naturally occurring cases of virulent ovine footrot (VFR) and benign footrot (BFR) using an artificial culture medium which incorporated trypticase, arginine, serine and 5% agar. A degrading proteinase (DP) test was developed which measured the proteinase activity of broth cultures of B. nodosus for a period of time after organism death to assess the stability of the enzyme. The spectrophotometric measurement of the release of dye from a hide powder--azure conjugate by the action of proteinase provided an objective analysis of enzyme activity. The DP test differentiated VFR and BFR isolates and promises to be a useful laboratory method for the diagnosis of benign and virulent footrot in sheep.  相似文献

SUMMARY: Virulent ovine footrot was transmitted accidentally to a group of 23 adult Merino sheep (flock B) after holding for 1 hour in sheep yards, which earlier the same day had contained another flock (flock A) with < 1% prevalence of sheep with footrot lesions. Sheep in flock B were rendered susceptible to virulent footrot by grazing 600 mm high unimproved pasture dominated by paspalum ( Paspalum dilatatum ) and kangaroo grass ( Themeda australis ) during warm, humid and wet weather. In addition to moisture, interdigital abrasions caused by the pasture might have predisposed the interdigital skin to infection with Dichelobacter nodosus .  相似文献

A field trial was carried out to compare 3 topical treatments for ovine footrot. They were preliminary hoof paring with weekly footbathing either in 10% formalin, or in 10% zinc sulphate + 0.2% anionic surfactant ('Teepol'), or twice-weekly footbathing in the same zinc sulphate-surfactant mixture without hoof paring. Control sheep were not treated. Within 21 days, the number and severity of active lesions of footscald and footrot among treated sheep were substantially reduced by comparison with the controls; no significant differences were observed between treatments. Treatment effects were also comparable against mild footrot infection of the sole or heel region, hut in the more advanced lesions the healing response to paring and zinc footbathing was superior to that from paring and formalin footbathing (P<0.01). Twice-weekly footbathing in zinc-surfactant solution was effective in preventing new footrot infection. No chronic toxic effects attributable to zinc sulphate at a concentration of 10% were demonstrable in the hoof integument either clinically or histologically.  相似文献

Objective To test the hypothesis that ovine footrot associated with a thermostable protease strain of Dichelobacter nodosus undergoes self cure or is sustained as an annually recurring disease, depending on the environment.
Design and procedure Forty Merino sheep from a single blood line were infected with a protease thermostable strain of D nodosus a t each of five sites in Western Australia. Footrot lesions and microscopic evidence of D nodosus were recorded every fortnight for 2.5 years, supplemented by laboratory culture. Rainfall, soil and air temperature, pasture quantity and composition and soil types were also recorded. Flocks that apparently self cured were relocated to a more favourable site for footrot in the final spring season.
Results The maximum prevalence of feet with clinical footrot lesions was 80.6, 1.3, 14.4, 3.8 and 88.1% at the five sites. Severe footrot occurred for three consecutive spring seasons at one site that had clay loam soil and at least 3500 kg/ha total pasture dry matter annually. However, the infection was asymptomatic for up to 10 weeks between outbreaks. D nodosus was isolated from flocks for 2.5 years at only two sites, although there was microscopic evidence of the organism at other sites in the final year. A thermolabile variant (strain U6) of D nodosus was isolated from the two sites where footrot persisted.
Conclusion Depending on time and location, ovine footrot induced by a protease thermostable strain of D nodosus either self cured or persisted as annual outbreaks interspersed with periods of asymptomatic infection.  相似文献

The immunological memory (anamnestic) responses in sheep recovered from virulent footrot (VFR) can be aroused by subcutaneous injection of outer membrane protein (OMP) antigens of Dichelobacter nodosus. The magnitude of this response is directly correlated to the highest antibody response attained during infection and memory lasts at least a year after recovery from VFR. However, some older animals show non-specific responses to OMP antigens. In this study an evaluation of D. nodosus pilus antigen for the anamnestic diagnosis of footrot in sheep was undertaken. The results indicated that the primary and anamnestic responses to pilus were similar in character to OMP antigen but were highly specific. The sensitivity of the procedure for detection of sheep with a history of VFR was approximately 80%. A low proportion of sheep with mild lesions due to virulent strains of D. nodosus reacted to anamnestic challenge. Anamnestic challenge with 10 microg pilus was used in a VFR surveillance program in migratory sheep flocks in Nepal. Conventional diagnostic methods could not be applied during the disease transmission periods in these flocks because of their migration to alpine pastures far away from human habitation. The results supported clinical and bacteriological findings suggesting that virulent strains of D. nodosus have apparently been eliminated from these flocks in Nepal.  相似文献

Soluble outer membrane protein of Bacteroides nodosus extracted with potassium thiocyanate (KSCN) was employed as antigen in an enzyme linked immunosorbent assay (ELISA) to detect serum antibody in sheep naturally infected with a heterologous serogroup. Serum antibody responses in 55 sheep were monitored for 2 years and maximum levels were directly related to the severity of clinical foot lesions. Serum antibody levels rose 2 weeks after foot lesions developed and declined within several months of resolution of lesions. After the first footrot transmission period, antibody levels persisted significantly (P less than 0.001) longer in sheep that did not become affected in the next transmission period compared with sheep in which footrot recurred. Antibody response did not appear to result in resolution of foot lesions. ELISA using KSCN antigen gave similar results to whole cell ELISA where cells prepared from an homologous serogroup were used as antigen. Both these assays were more sensitive than ELISA in which heterologous whole cell antigen was used. Proteins extracted from the outer membrane of B. nodosus, which are known to be immunogenic in natural infection and common to different serogroups of B. nodosus, appear to be useful antigens for serological investigations of ovine footrot.  相似文献

Programmes based on the identification and treatment of cases and the culling of animals refractory to treatment had failed to eradicate virulent footrot from two districts in the western region of Nepal. From 1993 to 1996 vaccination against two endemic virulent strains of Dichelobacter nodosus was tested for its potential to contribute to the eradication of footrot from the region. Only sheep and goats which had been free of signs of footrot at three inspections at monthly intervals before their annual migration to alpine pastures were eligible for inclusion. From November 1992, the treatment of cases identified during inspections included the injection of specific vaccine. Successfully treated cases migrated with their flocks but were excluded from the vaccine trial. Non-responding cases were culled. Forty combined flocks of sheep and goats (approximately 9500 animals) were used initially to compare three vaccination regimens. Eleven flocks (sheep and goats) were treated with two doses of specific vaccine (group A), nine (sheep and goats) were treated with commercial vaccine followed by specific vaccine (group B) and 10 (sheep and goats) were treated with two doses of commercial vaccine (group C) in March to April 1993 before the annual migration; 10 flocks (sheep and goats) remained unvaccinated (group D). Only sheep and goats free of signs of footrot were allowed to migrate. Nevertheless, virulent footrot recurred in many flocks three months later. However, its prevalence was significantly lower in group A than in the other three groups combined. Groups A, B and C then received the specific vaccine before their migrations in 1994 to 1996; group D remained unvaccinated. The annual programme of inspection and identification and treatment of cases continued for seven years, but the vaccinations ceased after four years. There was no recurrence of virulent footrot after November 1993. After the first season the virulent strains of D nodosus used in the specific vaccine could no longer be isolated, although antigenically distinct, benign strains of the organism persisted in cases of benign footrot.  相似文献

Two groups of adult Merino sheep, initially grazed on pasture, were dosed daily with zinc sulphate (1 mg Zn/kg LW) or zinc oxide (15 mg Zn/kg LW) for six and seven weeks, respectively. On the 18th day, both groups were transferred to indoor pens together with unmedicated control sheep, and five days later the feet of all animals were infected artificially with a virulent strain of Bacteroides nodosus. By the fourth week after challenge, 85% or more of the feet challenged had developed advanced footrot, and no significant differences in the incidence and severity of lesions between dosed and un-dosed groups were recorded. Plasma zinc concentrations, monitored throughout the experiment, remained at similar levels in sheep receiving the lower dose rate and in the controls. At the higher dose rate, plasma zinc levels increased till Day 23, but thereafter declined to values comparable to those of undosed animals. At post mortem, no evidence of zinc toxicosis was found, and only the kidneys of sheep receiving the higher dosage showed a significant accumulation of zinc compared with the controls.  相似文献

Footrot is a mixed bacterial infection of the hooves of sheep. The gram-negative anaerobic bacterium Dichelobacter nodosus is the principal causative agent, with different strains causing diseases of different severity, ranging from benign to virulent. In Australia, in the state of New South Wales (NSW), only virulent footrot is subject to regulatory action, including quarantine. However, it is often difficult to distinguish benign footrot from virulent footrot in the initial stages of infection, or under adverse climatic conditions. The gelatin gel test, which measures the thermostability of secreted bacterial proteases, is the laboratory test most widely used in Australia to aid in the differential diagnosis of footrot. The proteases of virulent strains are, in general, more thermostable than the proteases of benign strains. However, there are some false positives in the gelatin gel test, which may lead to unnecessary quarantine procedures. We used Southern blot analysis on 595 isolates of D. nodosus from 124 farms on which sheep had benign or virulent footrot to test for the presence of the intA gene. We found that for D. nodosus strains which are stable in the gelatin gel test, there is a high correlation between the presence of the intA gene and the ability of the strain to cause virulent footrot. We also developed a PCR-based assay for the rapid detection of intA, which can be used to test DNA extracted from colonies grown on plates, or DNA extracted from cotton swabs of culture plates.  相似文献

Characterisation of virulent and benign strains of Bacteroides nodosus   总被引:2,自引:0,他引:2  
The extracellular proteases of 395 isolates of B. nodosus from ovine, bovine and caprine foot lesions were classified as either thermostable or thermolabile. Stable protease was associated with one and unstable protease with four distinctive isoenzyme patterns, each pattern differentiated by the relative mobility of paired isoenzymes. Pathogenicity tests on 64 isolates showed a correlation between the production of stable protease and the production of virulent ovine footrot lesions. The mean values for total protease activity, twitching motility and colony diameter were significantly higher for virulent compared to benign isolates, but the range of values overlapped. SDS-PAGE whole-cell electrophoretic profiles of virulent isolates were similar to the profiles of some benign isolates.  相似文献

Groups of 10-12 Romney and Merino wethers were challenged simultaneously with homologous experimental footrot infection after having received the second of 2 doses of Bacteroides nodosus (strain 198) vaccine 0, 4, 8, 12, 16 and 20 weeks previously. Inoculations were carried out 28 days apart and unvaccinated sheep of both breeds were challenged as controls. Most Romneys that had been vaccinated up to 16 weeks prior to challenge were resistant to footrot whereas 8 of 10 controls were susceptible. This resistance was lost by about 20 weeks after vaccination. By contrast, protection against challenge in vaccinated Merinos lasted only about 4-5 weeks, although residual benefits of vaccination were apparent after longer intervals from the reduced number and severity of foot lesions among vaccinates compared with controls. Agglutinin titres, which did not differ markedly after similar intervals between Romneys and Merinos, reached maximum levels between 4 and 8 weeks after the second vaccine dose and subsequently declined. Although peak titres were generally recorded at the time of maximum protection in Merinos, the relationship between agglutinin levels and resistance in the Romneys was ill-defined.  相似文献

SUMMARY Benign footrot was studied in 1 1/2-years-old Merinos on 2 farms in central Victoria from September 1987 to August 1990, Inclusive. Treatment groups of 100 sheep grazed together with the remaining untreated sheep. Inspections were carried out every 3 weeks during the spring transmission period until the number of lesions greater than score 2 dropped below 3%. At each inspection, each sheep was weighed and lesion scores for each foot and digit were recorded, the treated group of sheep was treated by standing in 20% (w/v) zinc sulphate-sodium lauryl sulphate for 1 hour, and bacteriological samples were randomly collected from 5 sheep with and 5 without lesions. Dichelobacter nodosus organisms were obtained from sheep in both groups.
Laboratory tests indicated benign organisms in flock A and low virulence, intermediate organisms in flock B. During the first 2 years, the number and severity of lesions were greater in flock A than in flock B. However, in the third year, with an early 'autumn break', there was a rapid and severe outbreak of footrot in flock B; 98% of the flock had lesions at the first inspection in July 1989. Flock A had a less dramatic increase in lesions of footrot. Both treated and untreated groups in flock B recovered rapidly between the third and fourth inspections. A later increase in lesions for both flocks coincided with damage caused by barley grass seeds. During this period there was a significant difference (P / 0.001) in body weight between the treated and untreated sheep on farm B. An extra 0.2 kg of skirted fleece was obtained from treated sheep when shorn in 1990, compared with the untreated group (P = 0.04). Treated sheep had fewer tender fleeces, 11% compared with 25%. Wool from the untreated group was 0.5 micron finer but the value was 1.79 a head (7%) less than that from the treated group.  相似文献

Contagious ovine digital dermatitis (CODD) is a recently recorded, apparently new infection of the ovine hoof, which differs clinically from footrot caused by Dichelobacter nodosus and which fails to respond well to accepted treatment practices for footrot. Despite the welfare implications of such an infection, very little research has been performed on CODD to date and the aetiology remains confused. Suggestions have been made that there is a potential role for treponemes in the pathogenesis of CODD but that D. nodosus is apparently not involved. Six farms were therefore targeted in this study to provide a more in-depth investigation into the bacterial flora of CODD lesions. Dark ground microscopy, culture and PCR techniques were used, concentrating on the presence of D. nodosus and spirochaetes, particularly those of the genus Treponema. The results demonstrated that isolates of D. nodosus were indeed present in a high percentage (74%) of CODD lesions compared with 31% of apparently healthy feet. The isolates were shown to be of similar virulence type to those reported previously in cases of footrot, and the range of serogroups was also found to be similar to footrot, with serogroup H being prevalent. Treponemes were present in 70% of CODD lesions and 38% of apparently healthy feet, supporting a possible association between CODD and treponemes. However, any further progress on the aetiology of CODD and the potential for novel, effective treatment will depend on an improved ability to culture these organisms routinely in the laboratory thereby enabling their complete characterisation.  相似文献

Footrot and contagious ovine digital dermatitis (CODD) are common causes of foot disease of sheep in the UK. The study reported here is a split flock randomised treatment trial undertaken on a group of 748 fattening lambs on a UK sheep farm affected by CODD and footrot. The sheep were randomly assigned to one of two treatment protocols. In protocol A, all sheep were given two doses of footrot vaccine (Footvax, MSD), plus targeted antibiotic therapy (long-acting amoxicillin, Betamox LA, Norbrook Pharmaceuticals) to sheep with foot lesions likely to be associated with a bacterial infection. In protocol B, the sheep only received targeted antibiotic therapy. Sheep were re-examined and foot lesions recorded five and nine weeks later. New infection rates in the footrot vaccinated group were lower compared with the vaccinated group for both CODD (18.2 per cent compared with 26.4 per cent, P=0.014) and footrot (12.55 per cent compared with 27.5 per cent, P<0.001). Recovery rates were unaffected for CODD (80.46 per cent compared with 70.97 per cent, P=0.14) but higher for footrot (92.09 per cent compared with 81.54 per cent, P=0.005) in sheep which received the vaccine. On this farm, a footrot vaccine efficacy of 62 per cent was identified against footrot and 32 per cent against CODD infection. An association between a sheep having footrot at visit 1 and subsequently acquiring CODD was identified (odds ratio [OR] 3.83, 95 per cent CI 2.61 to 5.62, P<0.001). These results suggest a role for infection with Dichelobacter nodosus in the aetiopathogenesis of CODD on this farm.  相似文献
