外源ABA对NaCl胁迫下黄瓜幼苗不同离子含量的影响   总被引:2,自引:0,他引:2  
研究了外源ABA对NaCl胁迫下黄瓜幼苗不同器官中Na+、Cl-、K+和Ca2+含量的影响。结果表明,NaCl胁迫下,黄瓜幼苗体内Na+、Cl-含量升高;叶片中K+、Ca2+含量下降,Na+/K+、Na+/Ca2+增大,SK、Na、SCa、Na降低。外源ABA减少了叶片、根中Na+、Cl-的积累,抑制了叶片K+、Ca2+含量的降低,提高了K+、Ca2+的吸收和运输选择性。外源ABA可通过调控盐胁迫下植株体内的离子平衡,提高黄瓜幼苗的耐盐性。  相似文献

以酸枣水培幼苗为试验材料,研究了不同浓度NaCl胁迫对其活性氧、细胞膜透性、根系活力、不同器官Na+、K+、Ca2+和Mg2+离子含量的影响。结果表明:NaCl处理6 d,50、100 mmol ? L-1处理酸枣叶片无萎蔫症状,150、200 mmol ? L-1处理叶片萎蔫。在150、200 mmol ? L-1 NaCl处理下叶片H2O2积累增加,50、100、150、200 mmol ? L-1 NaCl处理下均有积累。随着NaCl处理浓度的升高,根、茎、叶中Na+含量均显著增加,叶片中K+、Ca2+和Mg2+含量显著下降,根和茎的K+和Mg2+含量变化不显著,Ca2+在200 mmol ? L-1NaCl处理下显著下降。根、茎、叶中K+/Na+、Ca2+/Na+、Mg2+/Na+均显著下降,但是根的K+/Na+值始终大于1。总之,NaCl胁迫下,维持根和茎中相对稳定的K+、Mg2+水平、保持根部K+/Na+比值 > 1,是酸枣幼苗对盐胁迫的一种适应。  相似文献

用体积浓度225mmol/L的NaCl对耐盐性不同的2个黄瓜品种进行处理,研究黄瓜盛果期NaCl胁迫下不同品种叶片和根中K+和Na+含量的变化。结果表明,盐胁迫下,黄瓜不同耐盐品种植株根和叶片中Na+含量明显增加,K+含量明显下降;耐盐品种叶片中Na+含量显著低于盐敏感品种,耐盐品种K+含量降低幅度显著小于盐敏感品种;植株的Na+、K+比值提高,耐盐品种Na+、K+比值显著低于盐敏感品种;耐盐品种SNa+,K+显著低于盐敏感品种。  相似文献

采用营养液栽培方法,研究NaCl胁迫下Ca2+和TFP对"津春2号"黄瓜幼苗叶片中多胺含量和抗氧化酶活性的影响。结果表明:Ca2+预处理能显著提高NaCl胁迫下黄瓜幼苗叶片中亚精胺(Spd)和精胺(Spm)含量,降低腐胺(Put)含量,同时叶片中SOD、POD和CAT活性也相应提高,使得株高相对生长率和干重明显增加;TFP预处理能显著增加Put含量,降低黄瓜幼苗叶片中Spd、Spm含量和(Spd+Spm)/Put比值,加重了NaCl胁迫对黄瓜幼苗生长的抑制。表明TFP阻碍Ca2+.CaM信使功能,从而降低盐胁迫下黄瓜幼苗的抗逆性。  相似文献

采用营养液栽培,进行NaCl盐胁迫处理,以耐盐性较强的葫芦为砧木嫁接西瓜,研究嫁接对盐胁迫下嫁接苗根部和地上部、根组织各部位Na+和K+含量的影响.结果表明,嫁接苗地下部比地上部截留了较多的Na+,而地上部的K+含量明显高于地下部;嫁接苗对Na+的根部截流和K+的地上部吸收能力强于自根苗.说明西瓜嫁接苗可阻隔根系内皮层对Na+向中柱导管中的运输以及限制Na+向中柱导管中的装载,进而将Na+较多地截流在根部,阻止其向地上部分运输;同时将K+较多地运输到地上部分,使地上部保持较高的[K+]/[Na+]比值,从而保持西瓜嫁接苗较强的耐盐性.  相似文献

以甜瓜品种"金辉1号"(耐盐型)和"一品天下208"(盐敏感型)为试材,采用深液流水培栽培,研究了不同频率(每天、每2d、每3d喷1次)喷施50mmol/Lγ?氨基丁酸(GABA)对80mmol/L Ca(NO_3)_2胁迫下甜瓜幼苗生长、可溶性总糖和脯氨酸含量的影响。结果表明:Ca(NO_3)_2胁迫显著降低了甜瓜幼苗株高、茎粗、地上部干鲜重、根系干鲜重、叶片和根系的可溶性总糖含量,显著提高了脯氨酸(Pro)含量,严重抑制了甜瓜幼苗的生长;Ca(NO_3)_2胁迫下,外源喷施GABA可明显增加甜瓜幼苗株高、茎粗、地上部干鲜重、根系干鲜重和可溶性总糖含量,降低叶片和根系脯氨酸(Pro)含量,对Ca(NO_3)_2胁迫引起的甜瓜幼苗生长的抑制作用有所缓解,且喷施频率为每2d1次时,这种缓解抑制作用的效果最佳;同时,试验研究发现,外源喷施GABA对盐敏感品种"一品天下208"的盐胁迫缓解效果更好。由此表明,每2d叶面喷施1次50mmol/L GABA最有利于提高甜瓜幼苗对Ca(NO_3)_2胁迫逆境的适应能力,且对盐敏感品种"一品天下208"缓解效果更好。  相似文献

以2年生盆栽富士苹果树(砧木为平邑甜茶)为试材,研究了NaCl胁迫对其叶片膜透性、脯氨酸及矿质营养含量的影响,结果表明:0.3%盐胁迫处理的电导率在中前期高于对照,1%盐处理中期高于对照;盐胁迫后半期0.3%NaCl处理的脯氨酸含量高于对照。NaCl胁迫下,苹果体内各器官Na+含量大小的顺序是:根>新梢>主干韧皮部>叶>主干木质部,在0.1%NaCl胁迫下,新梢和叶片中的Na+没有明显变化,根、主干韧皮部和主干木质部含量明显提高,在0.3%NaCl胁迫下各器官中Na+含量明显提高,地上部叶片、主干韧皮部和新梢中Na+含量提高幅度最大。NaCl胁迫对各器官的N、P、K+含量没有明显影响,但胁迫下N/Na+、P/Na+和K+/Na+比值不同程度的降低,从而使树体内的元素平衡受到破坏,导致树体受害。N、K+含量在叶片中含量最高,P在新梢中含量最高,N、P、K+在主干木质部中含量均最低。  相似文献

 采用水杨酸(SA)叶面喷施法,研究了NaCl胁迫下外源SA对菊花根系Na+、K+、Ca2+ 、Mg2+含量以及根系质膜ATPase、液泡膜ATPase和PPase活性的影响。结果表明,盐胁迫增加了菊花根系中Na+的含量而减少了K+、Ca2+、Mg2+的含量,而外源SA处理减少了Na+的含量并增加了K+、Ca2+、Mg2+的含量。NaCl胁迫下菊花根系质膜ATPase、液泡膜ATPase和PPase活性均明显下降,外源SA处理减小了盐胁迫引起的质膜ATPase、液泡膜ATPase和PPase活性的下降幅度,而且在胁迫10 d时,外源SA处理的根系质膜ATPase、液泡膜ATPase和PPase活性分别比NaCl处理提高了17.67%、16.47%和18.65%。说明盐胁迫下外源SA可以通过诱导质子泵活性来提高菊花根系对K+、Ca2+ 、Mg2+的吸收和减少对Na+的吸收,从而缓解盐胁迫对菊花植株的伤害程度。  相似文献

以黄瓜品种‘津优4号’为材料,采用营养液栽培,研究了外源腐胺(Putrescine,Put)对NaCl 胁迫下幼苗叶片光系统Ⅱ(PSⅡ)光化学特性和器官离子分布的影响。结果表明,外源Put提高了NaCl胁迫下黄瓜幼苗叶片最大光化学效率(Fv/Fm)、PSⅡ实际光化学效率(ΦPSⅡ)和保护性热耗散(ΦNPQ),显著增强光耐受能力和电子传递能力,降低了光损伤程度,抑制了植株体内Na+ 和Cl-向地上部的运输,显著降低了叶中Na+ 和Cl-的含量。说明外源Put能够减少NaCl胁迫下盐离子进入植株叶中,降低离子毒害,减轻不可逆光抑制,进而缓解盐胁迫对黄瓜幼苗光合机构的伤害。  相似文献

以盐地碱蓬幼苗为试材,研究了相同Na+浓度的NaCl和Na2 SO42种盐胁迫对盐地碱蓬幼苗含水量、叶绿素、有机酸、脯氨酸和可溶性糖含量的影响.结果表明:NaCl胁迫能促进盐地碱蓬幼苗含水量的增加,而Na2SO4胁迫则抑制其增加;NaCl和Na2 SO4胁迫均导致其叶绿素a、叶绿素b和叶绿素总量的减少以及有机酸含量的下降;而脯氨酸含量则随盐浓度的增大而增加,可溶性糖含量随盐浓度的增大呈先升高后降低的变化趋势.总体表现出Na2 SO4胁迫对盐地碱蓬的伤害作用弱于NaCl.  相似文献

AIM:To observe the effect of acupuncture on CD4+ CD25+ Foxp3(forkhead box P3)+ regulatory T-cells(Treg cells) in rats with embryo implantation failure. METHODS:One hundred and forty-four pregnant rats were randomly divided into control group(N), mifepristone treatment group(M), mifepristone+acupuncture treatment group(A) and mifepristone+progestin treatment group(W). The rats in groups M, A and W were treated with mifepristone-sesame oil solution on day 1, while the rats in group N were injected with the same amount of sesame oil. The Housanli(ST36) and Sanyinjiao(SP6) points were selected for acupuncture. From day 1 to the time of death, the rats in group A were fasten up and then the acupuncture was performed. Accordingly, the rats in group N and group M were only fixed, and the rats in group W were given progestin. Implanted embryos in each group were counted. The proportions of CD4+ CD25+ Foxp3+ Treg cells in peripheral blood and CD4+ Foxp3+ Treg cells in the endometrium were detected by flow cytometry. The mRNA and protein levels of Foxp3 were determined by real-time PCR and Western blotting, respectively. RESULTS:Compared with group N, the number of implanted embryos, the percentages of CD4+ CD25+ Foxp3+ Treg cells in peripheral blood and CD4+ Foxp3+ Treg cells in the endometrium, and the expression of Foxp3 protein and mRNA in the endometrium were significantly decreased in group M(P<005). Compared with group M, the above indexes in group A and group W were significantly increased. CONCLUSION: The effect of acupuncture in rats with embryo implantation failure may be closely correlated with the modulation of CD4+ CD25+ Foxp3+ Treg cells.  相似文献

以重瓣百合'Thalita'为试材,采用4个不同浓度(10、20、40、80 mg·L-1)的GA4+7+氯吡脲(1∶1),对其进行叶面喷施处理,以清水为对照,测定其株高、茎粗、叶面积等指标,探讨GA4+7+氯吡脲对重瓣百合'Thalita'生长的影响,以期为其推广提供参考.结果 表明:20 mg·L-1 GA4+7+氯吡脲对'Thalita'花苞直径和叶面积促进作用最大,分别比对照高14.18%和55.13%,均达显著差异;茎粗随着GA4+7+氯吡脲的浓度升高表现出一定抑制作用;20 mg·L-1GA4+7+氯吡脲在花苞开放率和花期上综合效果最好;80 mg·L-1 GA4+7+氯吡脲对'Thalita'株高促进效果最好,比对照高2.67%;所有处理组种球鲜质量和根长均比对照低,其中20 mg·L-1 GA4+7+氯吡脲效果最为明显,分别比对照低36.77%和48.18%,均达显著差异.综合比较来看,20 mg·L-1 GA4+7+氯吡脲对重瓣百合'Thalita'生长影响最大,效果更佳.  相似文献

 以自根和嫁接黄瓜幼苗为试材, 研究叶片Mn、Cu、Zn和Fe含量及低温胁迫下SOD活性的差异, 结果表明: 嫁接黄瓜叶片Mn、Cu和Zn的含量均显著高于自根黄瓜, 而Fe的含量则显著低于自根黄瓜; 低温胁迫第3天, 嫁接黄瓜叶片SOD、Cu /Zn-SOD和Mn-SOD活性均显著高于自根黄瓜, Fe-SOD则显著低于自根黄瓜。为探索嫁接黄瓜叶片高含量Mn、Cu、Zn与SOD活性之间的关系, 向低温胁迫第3天的自根黄瓜叶片SOD 提取液中加入Mn2+ 、Cu2+和Zn2+ , 使之与嫁接黄瓜叶片SOD 提取液对应组合的Mn2+ 、Cu2+和Zn2+浓度相等, 研究此时的SOD及其同工酶活性的变化, 结果发现: Mn2+ 、Cu2+和Zn2+加入自根黄瓜叶片提取液后均能显著增强SOD活性, 其增强作用的大小依次为Mn2+ +Zn2+, Mn2+ + Cu2+,Mn2+, Cu2+, Cu2+ + Zn2+ +Mn2+, Cu2+ + Zn2+和Zn2+ 。3种离子的加入都不影响Fe-SOD的活性。这一结果说明黄瓜叶片Mn、Cu和Zn的含量是影响SOD活性大小的重要因素, 嫁接黄瓜吸收Mn、Cu和Zn的能力较强是其SOD活性显著高于自根黄瓜的重要原因之一。  相似文献

AIM: To check the physical interaction between GST- Na+-K+-ATPase domain and recombinant human augmenter of liver regeneration (rhALR) by GST pull down assay. METHODS: With PCR and genetic recombinant techniques, the coding region of β subunit of Na+-K+-ATPase was cloned into expressing plasmid pGEX-4T and identified by endonuclease digestion and sequencing methods. Under the inducing of 0.1 mmol/L IPTG, the fusion protein GST- Na+-K+-ATPase domain was highly expressed by E.coli DH-5α. After hypersound quassating, the GST- Na+-K+-ATPase domain was purified by glutathione agarose beads and the physical interaction with rhALR was checked by GST pull down assay. RESULTS: Analysis by SDS-PAGE showed the rhALRs of monomer and dimmer in GST- Na+-K+-ATPase domain lane. The Western blotting of the GST-pull down assay showed the same results as well. CONCLUSION: The Na+-K+-ATPase domain is associated with rhALR specifically in vitro.  相似文献

以单季茭白品种蒋墅茭和双季茭白品种葑红早为试材,采用土壤为栽培介质,对Cd2+,Pb2+的单一及其复合胁迫,进行有机肥和无机肥处理,测定了茭白根、短缩茎、叶及产品器官中Cd2+.Pb2+的积累量.研究结果表明,Cd22+,Pb2+的积累量在茭白不同器官间表现为根>短缩茎>叶>肉质茎;两品种中复合胁迫时的Cd2+.Pb2+积累量均明显高于单一胁迫;不同肥料间以有机肥处理下茭白各器官中Cd2+.Pb2+的积累量同比均显著低于无机肥处理.品种问存在明显差异,蒋墅茭对Pb2+的积累能力强于葑红早,对Cd2+的积累能力则低于葑红早.茭白根系对Cd2+,Pb2+的富集系数表现为无机肥处理和复合胁迫处理时分别显著大于有机肥处理和单一胁迫.  相似文献

在固体和液体静置纯培养条件下,研究了3种重金属Cu、Zn和Pb在不同浓度处理下对橙黄疣柄牛肝菌菌丝生长的影响,并利用EC50值评价真菌对Cu,Zn和Pb的耐性。结果表明:通过半致死浓度评价橙黄疣柄牛肝菌耐受性发现,橙黄疣柄牛肝菌对Zn的耐受性强于Cu和Pb;菌根真菌培养后,基质pH降低与真菌生物量有关,说明菌根真菌在重金属胁迫下能通过调节自身pH环境缓解压力。  相似文献

以单性结实的龟井和自花结实的鄂柑1号柑橘品种为材料,对果实的整个发育期的子房(幼果)、果皮和果肉的总钙、可溶性Ca2+含量和Ca2+-ATPase活性进行了测定。结果表明龟井花前至花期子房总钙含量就已很高,花后趋下降,而鄂柑1号花前至花期子房总钙含量相对较低,花后有一显著上升;龟井可溶性Ca2+含量在花期及花后有一明显下降,而对应鄂柑1号在花后却有一显著上升;龟井花前至花期Ca2+-ATPase活性较高,花后下降,而对应鄂柑1号花前较低,花期即始上升;果实增大期内两品种果皮的总钙均趋上升,对应果肉总钙却趋下降;而果皮和果肉的可溶性Ca2+含量和Ca2+-ATPase活性在增大期内均有一明显增长过程或居相对较高水平。  相似文献

AIM:To investigate the proliferation of CD4+CD25+ T cells from PBMCs of the gastric cancer patients and the inhibitory effect on CD4+CD25- T cells in vitro. METHODS:Magnetic activated cell sorting (MACS) method was used to separate CD4+CD25+T and CD4+CD25-T cells from peripheral blood monocytic lymphocytes in the gastric cancer patients, and then the purity and activity of CD4+CD25+T cells were analyzed with flow cytometer. After stimulated with anti-CD3 Ab, anti-CD28 Ab and rh IL-2, CD4+CD25- and CD4+CD25+ T cells were cocultured. The inhibitory effect of CD4+CD25+T on CD4+CD25-T cells was assayed by [3H] thymidine proliferation experiment. RESULTS:(1)After sorting, CD4+CD25+ T cells purity in healthy control and gastric cancer patients were 83.80%±1.84% and 84.13%±2.77%, respectively. No significant difference between the two groups (P>0.05) was observed. (2)The activity of CD4+CD25+ and CD4+CD25- T cells in healthy control and the gastric cancer patients after sorting were 98.52%±0.72% and 97.80%±0.95%. There was no significantly difference between the two groups (P>0.05). (3) CD4+CD25+ T cells obviously inhibited the CD4+CD25-T cell proliferation in vitro. The inhibition achieved to maximum in coculture of CD4+CD25+ T cells together with CD4+CD25- T cells (ratio of 1∶〖KG-*2〗1). CONCLUSION:The MACS system can effectively isolate CD4+CD25+ and CD4+CD25- T cells. After sorting, CD4+CD25+T cells obviously inhibit the proliferation of CD4+CD25- T cells in vitro and the inhibitory effect display an effect-target ratio relationship.  相似文献
