The purpose of this study was to determine the optimal interval between 2 consecutive milk progesterone samples for the detection of cyclicity in dairy cows. Two hundred and thirty-six postpartum periods were monitored with thrice-weekly whole milk progesterone assay. Cyclicity was determined from elevation of the progesterone level. Animals which had started to cycle by 50 or 60 days post partum were included in the study. The last of the 2 samples was taken at 50 or 60 days post partum, respectively.The lowest percentage of false diagnoses (9.0%) in cows which had started to cycle by 50 days post partum was obtained when the samples were taken at 8 days’ interval. In cows which had started to cycle by 60 days post partum the lowest percentage of false diagnoses (1.1%) was obtained when the samples were taken at 10 days’ interval.  相似文献

A within cow comparison was made between milk progesterone levels in healthy and mastitic quarters. Material was collected from cows with mastitis induced by bacterial inoculation, or by inoculation with bacterial endotoxin. Furthermore material from cows with spontaneous subclinical mastitis was used. Milk progesterone levels were lowered due to mastitis. However, the decrease was not large enough to cause misinterpretation of where in the oestrous cycle (luteal phase or non-luteal phase) the samples were taken.  相似文献

为了完善已建立的孕酮免疫生物传感器的检测性能,本试验应用碳化二亚胺法将11α-羟基孕酮半琥珀酸和OVA制备成孕酮完全抗原,其浓度达0.6 mg/m L。在此基础上建立了以乳汁为基质的孕酮生物传感器的标准曲线(y=-4.9461x+8.7982,R2=0.993 2),进一步确立了检测乳汁孕酮的线性范围为0.31 ng/m L~50ng/m L,检测限为0.31 ng/m L,灵敏度为0.15 ng/m L,而且孕酮与皮质酮、皮质醇、雄烯二酮的免疫交叉反应较低,表明特异性较好。批内、批间变异系数分别为7.7%和7.5%,回收率为95.8%~115.7%,生物传感器稳定性在4℃保存4 d。该优化的奶牛乳汁孕酮免疫生物传感器的性能参数符合规定的标准,为检测妊娠、乏情和繁殖障碍疾病的奶牛乳汁孕酮含量提供了工具。  相似文献

The main objective of the study was to describe the relationship of high somatic cell count (SCC) with the incidence of abnormal postpartum resumption of ovarian cyclicity and reproductive performance in dairy cows. The factors influencing SCC were also investigated. Four hundred and forty-seven cows from six dairy herds in Japan were monitored for SCC and postpartum resumption of ovarian cyclicity. Cows with high SCC (200,000 to 500,000) had a higher incidence of prolonged luteal phase (P < 0.01) than cows with an SCC of 50,000 to 100,000. The high SCC cows (> 500,000) also showed a higher incidence of delayed first ovulation post partum than cows with an SCC ≤500,000 (P < 0.05) during the first month post partum. Cows with an SCC of 200,000 to 500,000 showed lower conception and pregnancy rates, and more days from calving to conception than cows with an SCC of less than 200,000 (P < 0.05). Cows in parity 5 or more had a higher incidence of high SCC than cows in the first and second parities (P < 0.05). It is concluded that cows with a high SCC have a higher incidence of abnormal postpartum resumption of ovarian cyclicity, leading to reduced reproductive performance.  相似文献

Background

Phosphorus (P) supplementation is costly and can result in excess P excretion. This study investigated the effects of reducing dietary P on milk production and P excretion in dairy cows over a full lactation.

Method

Forty-five multiparous Holstein dairy cows were divided into 15 blocks according to expected calving date and previous milk yield, and assigned randomly to one of the three dietary treatments: 0.37, 0.47, and 0.57% P (DM basis); these P levels represent the NRC recommendations, Chinese recommendations, and the amount of dietary P commonly fed by Chinese dairy farmers, respectively. Average daily feed intake was calculated from monthly data on feed offered and refused. Milk yields of individual cows were recorded weekly, and milk samples were taken for analysis of protein, fat, solids-not-fat, lactose, and somatic cell count. Blood samples were collected on days −6, −3, 0, 3, 6 relative to calving, and then monthly throughout lactation, and analyzed for P and Ca concentrations. Spot samples of feces and urine were collected for 3 consecutive d during weeks 12, 24, and 36, and P concentrations were analyzed. Reproduction and health data were recorded.

Results

Dietary P did not affect dry matter intake or milk yield (P > 0.10). Milk fat content was slightly higher in cows fed 0.37% P than in cows fed 0.47% P (P = 0.05). Serum concentrations of P and Ca did not reflect dietary P content (P > 0.10). Fecal and urinary P both declined linearly (P < 0.05) as dietary P decreased from 0.57 to 0.37%. Fecal P content was 25% less when dietary P was 0.37% compared to 0.57%. Health events and reproductive performance were not associated with dietary P content (P > 0.05).

Conclusions

Lowering dietary P from 0.57 to 0.37% did not negatively affect milk production, but did significantly reduce P excretion into environment.  相似文献

A delayed rise in post-ovulatory progesterone is associated with poor embryo development in the cow, although the underlying cause of this aberrant luteal function is poorly understood. The objective of this study was to develop a novel model, in which a delayed progesterone rise could be induced by manipulating the dynamics of the follicular phase. Luteolysis was induced in 20 dairy cows in the presence of either a larger follicle > 10 mm (LF, n = 11) or a smaller follicle < 10 mm (SF, n = 9) and transrectal ultrasonography was performed to determine follicle and CL growth and timing of ovulation. Plasma progesterone and oestradiol were analysed 3x daily. Cows were slaughtered on either day 4 (n = 4 per group) or day 7 (SF, n = 5; LF, n = 7) after ovulation. The pre-ovulatory follicle was larger in the LF group than the SF group at luteolysis (13.5 +/- 0.4 mm versus 6.7 +/- 0.7 mm, P < 0.001) and ovulation (16.7 +/- 0.3 mm versus 13.6 +/- 0.6 mm, P < 0.001). The LF group experienced a shorter follicular phase and ovulated 36 h earlier than the SF group (P < 0.001). At luteolysis, plasma oestradiol concentrations were greater in the LF group (P < 0.001), although peak concentrations were not different (P > 0.05). Moreover, higher progesterone concentrations were observed in the LF group during the early luteal phase (P < 0.05). Luteal weights were positively correlated with plasma progesterone concentrations on day 5 (P < 0.05) but not day 8. In conclusion, a model has been developed which has shown that the dynamics of follicle development during the pre-ovulatory period is an important determinant of subsequent CL development and function.  相似文献

The aim of this study was to validate a direct time-resolved fluoroimmunoassay (TR-FIA) for quantifying progesterone concentrations in milk during the bovine oestrous cycle. Holstein-Friesian and suckled and non-suckled Japanese Black cows were used to demonstrate the relationship between milk and plasma progesterone concentrations and to monitor progesterone profiles in milk and plasma during the oestrous cycle. The minimum detection level of the assay was 1.53ng/mL. Progesterone concentrations in milk and plasma changed in a similar manner throughout the oestrous cycle in dairy and beef cows, and milk and plasma progesterone profiles were significantly correlated (P<0.001). The study confirmed that a direct TR-FIA can be used to monitor the oestrous cycle in cattle and to quantify progesterone concentrations in whole milk.  相似文献

Milk samples were collected from 91 dairy cows on days 18, 22, and 24 postbreeding. The progesterone values of these samples were assayed, using enzyme-immuno-assay for pregnancy diagnosis. These animals were classified by nonpregnant (low progesterone values, 5 ng/ml or less) and pregnant (high progesterone values, more than 5 ng/ml). The accuracy of diagnosis on days 18, 22, and 24 postbreeding were 56%, 78%, and 79% for pregnant cows and 90%, 93%, and 93% for nonpregnant animals. The sampling date had a significant effect on diagnostic accuracy (P less than 0.05).  相似文献

Effect of lameness on milk yield in dairy cows   总被引:1,自引:0,他引:1  
OBJECTIVE: To examine the relationship between lameness and milk yield in dairy cows. DESIGN: Cohort study. ANIMALS: 531 dairy cows. PROCEDURE: Cows affected with lameness were classified into 1 of 3 groups on the basis of type of diseases or lesions observed, including interdigital phlegmon (foot rot), papillomatous digital dermatitis (foot warts), or claw lesions. Cows not affected with lameness were classified as healthy. From Dairy Herd Improvement Association records, 305-day mature equivalent milk yield data were collected at the end of lactation or when the cow left the herd. Milk yield was compared between cows affected with lameness and healthy cows. RESULTS: 167 (31%) cows were affected with lameness during lactation. Lame cows had claw lesions (60%), papillomatous digital dermatitis (31%), or interdigital phlegmon (9%). Milk yield in lame cows with interdigital phlegmon (mean, 17,122 lb) was significantly less, compared with healthy cows (19,007 lb). CONCLUSIONS AND CLINICAL RELEVANCE: In this herd, interdigital phlegmon was associated with a 10% decrease in milk production. Lame cows with claw lesions or papillomatous digital dermatitis produced less milk than healthy cows, but the difference was not significant.  相似文献

The reliability of clinical ovarian findings was assessed as an indicator of luteal function in primiparous dairy cows. The postpartum period of 103 cows following their first parturition was studied by thrice weekly rectal palpation of ovaries and whole milk progesterone assay from 1 week after parturition to the first insemination. The relationship between milk progesterone levels and 1101 ovarian findings was compared during the follicular phases, short luteal phases and during the early, mid and late thirds of normal luteal phases. The compatibility between elevated progesterone and palpable corpus luteum was 71%, and between low progesterone and lack of corpus luteum 77%. In 10% of all rectal examinations the finding was unspecified; i.e. the clinician could not differentiate between luteal and follicular activity. During the acyclic period prior to the initiation of luteal function, the proportion of false corpus luteum findings was 11%. The corpora lutea of the short oestrous cycles were more difficult to palpate than those of normal cycles. During early dioestrus the corpus luteum was significantly more difficult to palpate than during the rest of dioestrus. The percentage of unspecified findings was highest during early dioestrus.The paper discusses the reliability of rectal examination as a method of diagnosing cyclicity and of evaluating the responsiveness of a cow to prostaglandin treatment.  相似文献

Two field trials were carried out by five experienced veterinary surgeons to evaluate the milk progesterone test as an aid to the differential diagnosis of cystic ovaries in dairy cows. Of 200 cystic cows, 71 per cent were diagnosed clinically as follicular and 29 per cent as luteal cases; 80 per cent of the cases occurred in the six winter months November to April. The progesterone assay indicated that the diagnosis was correct in 84 per cent of the follicular cases but only 54 per cent of the luteal cases. Seventy-four per cent of the confirmed cystic cases responded to treatment within two weeks according to milk progesterone tests and there were no significant treatment differences. Overall, 80 per cent of the cows became pregnant on average 38 days after treatment having received an average of 1.70 inseminations each. Milk yield data in the first trial indicated that the cystic cows were not significantly higher yielders than their herd-mates.  相似文献

Crossbreeding effects on milk production traits of Czech dual‐purpose and dairy cattle breeds were estimated. Nearly 370 000 cows with known gene proportions from Czech Pied, Ayrshire or Holstein cattle were selected from the national milk recording data base. Single‐trait animal models were calculated for milk, fat and protein yield, fat and protein content. The model of Dickerson including additive, additive maternal, heterotic and recombination effects was used for the part of the animal model describing the crossbreeding effects in all calculations. For milk yield, the additive genetic effect (defined as deviation from Czech Pied cattle) was 850–900 kg for Holstein and 240–480 kg for Ayrshire. The maternal effects were low and negative. Low significant positive heterotic effects were observed being up to approximately 100 kg for Czech Pied × Holstein. The recombination effects were negative and statistically significant for Czech Pied × Holstein. The results for fat and protein yield were similar to the results for milk yield. For fat and protein content, nearly no statistically significant crossbreeding effects were found.  相似文献

Three hundred and sixty inseminated Holstein cows were monitored in a controlled trial for low milk progesterone concentration 21-23 days post-breeding. Information about low progesterone cows was given to herdsmen for the experimental group only. Of the cows in the experimental group with milk progesterone of less than or equal to 4.5 ng/ml, 56.0% were inseminated within 2 days of monitoring compared to 28.8% of the control animals. Of the cows reaching the 42-day rectal pregnancy diagnosis, 13.7% and 43.6% were negative for the experimental and the control groups respectively. Inter-service intervals were 34.5 +/- 14.5 days and 44.3 +/- 17.7 days for the experimental and control groups respectively. Pregnancy rates were the same for 264 cows served on routine progesterone monitoring and for 318 cows bred on behavioural oestrus when matched to farm and day of breeding. A gain of US $32.5 (an equivalent of 85 l milk) per lactation for a cow in the monitored herd was established using the data in a model of a 250-cow herd with a total pregnancy goal of 82.4%.  相似文献

Progesterone plays an important role in maintenance of pregnancy. It is hypothesized that insufficient progesterone early in pregnancy may result in embryonic loss, and that supplemental progesterone would decrease pregnancy loss in dairy cows. In Experiment 1, 84 cows and 16 heifers from a single dairy operation were selected randomly. Within each age category, controlled internal drug release (CIDR) devices were inserted into the vagina of every other female on Day 4 post-insemination and removed on Day 18 post-insemination. Transrectal ultrasonography was performed to determine pregnancy at 4 time periods [days 30 to 37 (week 5), days 44 to 51 (week 7), days 58 to 65 (week 9), and days 86 to 93 (week 13)]. Progesterone supplementation had no effect on pregnancy rate. In Experiment 2, there were no differences in progesterone concentrations between cows that did and did not receive a CIDR. Further, cows receiving CIDR devices did not have an increase in circulating progesterone concentrations 30 min or 1 h after CIDR insertion. It appears that progesterone supplementation does not increase circulating levels of progesterone in the early pregnant lactating dairy cow. Alterative methods to influence progesterone concentrations and/or early embryonic loss need to be investigated.  相似文献

More than 80% of the milk stored in the udder, i.e. the alveolar fraction, only available after milk ejection is induced by the release of oxytocin and myoepithelial contraction. Milk ejection is induced by tactile teat stimulation, either manually or by the milking machine. The time from the start of a tactile stimulation until the occurrence of milk ejection spans from 40 s up to more than 2 min and increases with decreasing degree of udder filling. Therefore, cows need a longer pre-stimulation in late stages of lactation or if the milking is performed shortly after the previous milking whereas in full udders, pre-stimulation is less important. Milk ejection can be disturbed under several conditions, for example, during milking in unfamiliar surroundings or for several weeks after parturition in many primiparous cows. We found that a disturbed milk ejection is due to a reduced release or complete absence of oxytocin from the pituitary. During milkings with disturbed milk ejection in unfamiliar surroundings, the concentrations of cortisol and beta-endorphin were elevated. While exogenous glucocorticoids could not induce the disturbance, morphine caused inhibition of oxytocin release during milking. However, spontaneously occurring disturbance of milk ejection could not be abolished by the opioid-antagonist naloxone. On the other hand, severity of disturbed milk ejection and coping capacity towards novel milking environment was inversely related to the cortisol release in response to ACTH, i.e. adrenal cortex activity. Therefore, the susceptibility of individual cows to the inhibition of oxytocin release and milk ejection could be predicted by an ACTH challenge test.  相似文献

The progesterone level of milk was measured daily in 20 Icelandic dairy cows for 50-90 days post-partum and for 30-50 days post-conception by radioimmunoassay. In 40% of the cows the 1st post-partum ovarian cycle was shorter and the progesterone level lower than in normal cycles. The average duration of the 1st post-partum luteal phase was significantly shorter and the progesterone values lower than in the 2nd and 3rd phases. The progesterone level rose to a plateau at the onset of luteal function within 5 +/- 2 (mean +/- s.d.) days, whereas the decrease in progesterone at the end of the luteal phase was more rapid i.e. 3 +/- 1 days. The progesterone value for a normal interluteal phase was 1.5 +/- 1.3 nmol/l. However in 9 of 48 interluteal phases the progesterone values did not decline below 3 nmol/l. The average progesterone concentration was significantly increased during the first 30 days post-conception. In 4 of 20 cows a significant drop in progesterone concentration was found 15-19 days post-conception. The results indicate that the luteal activity of the 1st post-partum ovarian cycle is inferior to that of the following cycles. There is a significant correlation in the intensity of the luteal activity between cycles. Ovarian cyclicity continues in early pregnancy in, at least, 20% of cows. The sampling frequency is of importance in studies concerning the luteal activity.  相似文献

根据宁夏地区某奶牛场的DHI检测获得的体细胞数据(SCS),利用SAS软件实现单因素方差分析,探讨不同乳中SCS对产奶量和乳中主要成分的影响,并对它们之间的相关性进行分析。结果表明:乳中SCS与日产奶量呈显著负相关(P0.01);与乳糖和胎次呈正相关(P0.01);乳中SCS水平与乳脂率、乳蛋白率和尿素氮没有明显的相关性。本试验通过对SCS的研究为乳品质的测定提供一定依据和参考。  相似文献
