Microorganisms are the regulators of decomposition processes occurring in soil, they also constitute a labile fraction of potentially available N. Microbial mineralization and nutrient cycling could be affected through altered plant inputs at elevated CO2. An understanding of microbial biomass and microbial activity in response to belowground processes induced by elevated CO2 is thus crucial in order to predict the long-term response of ecosystems to climatic changes. Microbial biomass, microbial respiration, inorganic N, extractable P and six enzymatic activities related to C, N, P and S cycling (β-glucosidase, cellulase, chitinase, protease, acid phosphatase and arylsulphatase) were investigated in soils of a poplar plantation exposed to elevated CO2. Clones of Populus alba, Populus nigra and Populus x euramericana were grown in six 314 m2 plots treated either with atmospheric (control) or enriched (550 μmol mol−1 CO2) CO2 concentration with FACE technology (free-air CO2 enrichment). Chemical and biochemical parameters were monitored throughout a year in soil samples collected at five sampling dates starting from Autumn 2000 to Autumn 2001.

The aim of the present work was: (1) to determine if CO2 enrichment induces modifications to soil microbial pool size and metabolism, (2) to test how the seasonal fluctuations of soil biochemical properties and CO2 level interact, (3) to evaluate if microbial nutrient acquisition activity is changed under elevated CO2.

CO2 enrichment significantly affected soil nutrient content and three enzyme activities: acid phosphatase, chitinase and arylsulphatase, indicators of nutrient acquisition activity. Microbial biomass increased by a 16% under elevated CO2. All soil biochemical properties were significantly affected by the temporal variability and the interaction between time and CO2 level significantly influenced β-glucosidase activity and microbial respiration. Data on arylsulphatase and chitinase activity suggest a possible shift of microbial population in favour of fungi induced by the FACE treatment.

Future high levels of atmospheric carbon dioxide (CO2) may increase biomass production of terrestrial plants and hence plant requirements for soil mineral nutrients to sustain a greater biomass production. Phosphorus (P), an element essential for plant growth, is found in soils both in inorganic and in organic forms. In this work, three genotypes of Populus were grown under ambient and elevated atmospheric CO2 concentrations (FACE) for 5 years. An N fertilisation treatment was added in years 4 and 5 after planting. Using a fractionation scheme, total P was sequentially extracted using H2O, NaOH, HCl and HNO3, and P determined as both molybdate (Mo) reactive and total P. Molybdate-reactive P is defined as mainly inorganic but also some labile organic P which is determined by Vanado-molybdophosphoric acid colorimetric methods. Organic P was also measured to assess all plant available and weatherable P pools. We tested the hypotheses that higher P demand due to increased growth is met by a depletion of easily weatherable soil P pools, and that increased biomass inputs increases the amount of organic P in the soil. The concentration of organic P increased under FACE, but was associated with a decrease in total soil organic matter. The greatest increase in the soil P due to elevated CO2 was found in the HCl-extractable P fraction in the non-fertilised treatment. In the NaOH-extractable fraction the Mo-reactive P increased under FACE, but total P did not differ between ambient and FACE. The increase in both the NaOH- and HCl-extractable fractions was smaller after N addition. The results showed that elevated atmospheric CO2 has a positive effect on soil P availability rather than leading to depletion. We suggest that the increase in the NaOH- and HCl-extractable fractions is biologically driven by organic matter mineralization, weathering and mycorrhizal hyphal turnover.

Rapid response of soil protozoa to elevated CO2
Short-term changes in bacterial and protozoan populations from the soil of plants grown under elevated atmospheric CO2 were quantified. We grew Brassica nigra at either ambient or twice-ambient CO2 levels within open-top chambers in the field for 4 weeks. Plant biomass, above- and belowground, was unaffected by elevated CO2. Direct count bacterial density was unchanged under elevated CO2. Flagellate density tended to increase, whereas amoebal density significantly declined under elevated CO2. This change in protozoan community structure suggests trophic transfer of the elevated CO2 fertilization effect through the soil food chain.

Ecosystem exposure to elevated atmospheric CO2 concentration can often leads to increased ecosystem carbon (C) fluxes, as well as greater net primary production. Changes in the soil C pool with elevated [CO2] are more difficult to measure and therefore remain poorly understood. In this study, we carried out a series of laboratory soil incubations, in order to determine whether 8 years of ecosystem exposure to elevated [CO2] altered decomposition dynamics of two age classes of soil C in a temperate coniferous forest. Our objectives were to determine whether there were differences in the decomposition kinetics of soil C up to 8 years old (Cpost-tr) and soil C older than 8 years (Cpre-tr), in the absence of concurrent plant activity. We collected soil from the Duke Forest Free Air CO2 Enrichment site in North Carolina and incubated whole and crushed (all macroaggregates dispersed) soil from two depth increments (0–5 cm and 5–15 cm) for 102–127 days. We found that mineral soil from the treatment plots had higher respiration rates in the absence of concurrent plant activity than mineral soil from plots under ambient CO2 conditions. These differences in respiration rate were only significant in 0–5 cm soil and could be largely explained by higher initial respiration rates of soil collected from the CO2-treated plots. Disruption of soil macroaggregates did not result in a difference in efflux rate in soil from this forest under ambient or elevated CO2 conditions at either depth. The specific respiration rate of Cpost-tr was higher than that of Cpre-tr in the top 5 cm of soil, while the opposite was true for 5–15 cm of soil. Even though Cpost-tr was assimilated by the ecosystem more recently than Cpre-tr, their decay constants were similar at both depths. These results suggest that, in the absence of plant activity, the mineralization of soil C of different ages in this forest may be under similar biological and/or biochemical control. Therefore, if the higher initial rates of decomposition of Cpost-tr seen in these experiments are sustained in the field, greater labile pool size of recently added C, and potentially faster cycling of this pool, may in part explain higher soil respiration rates and limited soil C accumulation under elevated [CO2] in this forest.

Agricultural soil CO2 emissions and their controlling factors have recently received increased attention because of the high potential of carbon sequestration and their importance in soil fertility. Several parameters of soil structure, chemistry, and microbiology were monitored along with soil CO2 emissions in research conducted in soils derived from a glacial till. The investigation was carried out during the 2012 growing season in Northern Germany. Higher potentials of soil CO2 emissions were found in grassland (20.40 µg g?1 dry weight h?1) compared to arable land (5.59 µg g?1 dry weight h?1) within the incubating temperature from 5°C to 40°C and incubating moisture from 30% to 70% water holding capacity (WHC) of soils taken during the growing season. For agricultural soils regardless of pasture and arable management, we suggested nine key factors that influence changes in soil CO2 emissions including soil temperature, metabolic quotient, bulk density, WHC, percentage of silt, bacterial biomass, pH, soil organic carbon, and hot water soluble carbon (glucose equivalent) based on principal component analysis and hierarchical cluster analysis. Slightly different key factors were proposed concerning individual land use types, however, the most important factors for soil CO2 emissions of agricultural soils in Northern Germany were proved to be metabolic quotient and soil temperature. Our results are valuable in providing key influencing factors for soil CO2 emission changes in grassland and arable land with respect to soil respiration, physical status, nutrition supply, and microbe-related parameters.

ABSTRACT

Elevated concentrations of carbon dioxide (e[CO2]) affect plant growth and physiological characteristics, including metal accumulation, and the activity of anti-oxidant enzymes. These effects were investigated in cadmium (Cd) tolerant wheat (Triticum aestivum L.) and sorghum (Sorghum bicolor (L.) Moench.) cultivars. Plants were grown at the ambient and elevated CO2 levels, with four concentrations of Cd (0, 10, 20 and 40 mg kg?1) added to the soil. After 60 days, subsamples were tested for chlorophylls and carotenoids, protein, enzyme activities and morphological characteristics.

Results showed that e[CO2] increased plant height, leaf area, and the dry weight of shoots and roots (P < 0.01). In addition, it decreased the Cd concentration in the shoots and roots of wheat, and increased the same concentrations for sorghum. With increasing Cd, the activities of the anti-oxidants, SOD and GSH-px increased in wheat. The differences in enzyme activity parallel the changes in Cd concentration in the plants of both species.

Considering that even contaminated soils are a potential resource for agricultural production, it is essential to develop a set of cropping systems to allow a safe and sustainable agriculture on contaminated lands while avoiding any transfer of toxic trace elements to the food chain. In this review, three main strategies, i.e., phytoexclusion, phytostabilization, and phytoextraction, are proposed to establish cropping systems for production of edible and non-edible plants, and for extraction of elements for industrial use. For safe production of food crops, the selection of low-accumulating plants/cultivars and the application of soil amendments are of vital importance. Phytostabilization using non-food energy and fiber plants can provide additional renewable energy sources and economic benefit with minimum cost of agricultural measures. Phytoextracting trace elements (e.g., As, Cd, Ni, and Zn) using hyperaccumulator species is more suitable for slightly and moderately polluted sites, and phytomining of Ni from serpentine soils has shown a great potential to extract Ni-containing bio-ores of economic interests. We conclude that appropriate combinations of soil types, plant species/cultivars, and agronomic practices can restrict trace metal transfer to the food chain and/or extract energy and metals of industrial use and allow safe agricultural activities.

The effects of elevated atmospheric CO2 on root dynamics were studied in a semi-natural grassland in central Sweden during five consecutive summer seasons. Open-top chambers were used for ambient and elevated (+350 μmol mol?1) concentrations of CO2, and chamberless rings were used for control. Root dynamics were observed in situ with minirhizotrons during the five summers and root biomass production was measured with root in growth cores during the last two years, from which total root biomass was estimated for each of the five years. The elevated CO2 treatment showed both a greater increase in root numbers during the early summer and a greater decline in root numbers during autumn and winter than the ambient CO2 treatment. Mean root production under elevated CO2 was 50% greater than ambient CO2 during the five years, and the difference increased from +25% in the first year to +80% in the last two years. Conversely, during the same period, the elevated to ambient CO2 difference in shoot biomass decreased from +50% to +5%. This resulted in a dramatic change in root to shoot ratios in elevated CO2 compared with the ambient treatment, which increased from ?15% in 1996 to +70% in 2000. Similar differences were seen between elevated CO2 and the chamberless grown control plants, where root to shoot ratios increased steadily from ?47% in 1996 to +27% in 2000. Less dynamically, the root to shoot ratios of ambient CO2 grown plants compared with the chamberless control plants were consistently ?29%±6% during the experimental period. In conclusion, during the 5 years this grassland was studied, there was a clear shift in plant biomass partitioning from above to below ground for plants exposed to elevated CO2.

Background, Aims, and Scope  The genetic structure and the functionality of soil microbes are both important when studying the role of soil in the C cycle in elevated CO2 scenarios. The aim of this work was to investigate the genetic composition of the fungal community by means of PCR-DGGE and the functional diversity of soil micro-organisms in general with MicroResp-based community level physiological profiling (CLPP) in a poplar plantation (POPFACE) grown under elevated [CO2] with and without nitrogen fertilization. Materials and Methods  The POPFACE experimental plantation and FACE facility are located in central Italy, Tuscania (VT). Clones of Populus alba, Populus nigra and Populus x euramericana were grown, from 1999 to 2004, in six 314 m2 plots treated either with atmospheric (control) or enriched (550 μmol mol−1) CO2 with FACE (Free Air CO2 Enrichment) technology in each growing season. Each plot is divided into six triangular sectors, with two sectors per poplar genotype: three species × two nitrogen levels. After removal of the litter layer one soil core per genotype (10 cm wide, 20 cm depth) was taken inside each of the three sectors in each plot, for a total of 36 soil cores (3 replicates × 2 [CO2] × 2 fertilization × 3 species) in October 2004 and in July 2005. DNA was extracted with a bead beating procedure. 18S rDNA gene fragments were amplified with PCR using fungal primers (FR1 GC and FF390). Analysis of CLPP was performed using the MicroResp method. Carbon substrates were selected depending on their ecological relevance to soil and their solubility in water. In particular rhizospheric C sources (carboxylic acids and carbohydrates) were chosen considering the importance of root inputs for microbial metabolism. Results  The fertilization treatment differentiated the fungal community composition regardless of elevated [CO2] or the poplar species; moreover the number of fungal species was lower in fertilized soil. The effect of elevated [CO2] on the fungal community composition was evident only as interaction with the fertilization treatment as, in N-sufficient soils, the elevated [CO2] selected a different microbial community. For CLPP, the differ ent poplar species were the main factors of variation. The FACE treatment, on average, resulted in lower C utilization rates in un-fertilized soils and higher in fertilized soils. Discussion  Fungal biomass and fungal composition depend on different factors: from previous studies we know that the greater quantity and the higher C/N ratio of organic inputs under elevated [CO2] influenced positively the fungal biomass both in fertilized and in un-fertilized soil, whereas nitrogen availability resulted to be the main determinant of fungal community composition in this work. Whole active microbial community was directly influenced by the soil nutrient availability and the poplar species. Under elevated CO2 the competition for N with plants strongly affected the microbial communities, which were not able to benefit from added rhizospheric substrates. Under Nsufficient conditions, the increase of microbial activity due to [CO2] enrichment was related to a more active microbial community, favoured by the current availability of C and N. Conclusions  Different factors influenced the microbial community at different levels: poplar species and root exudates affected the functional properties of the microbial community, while the fungal specific composition (as seen with DGGE) remained unaffected. On the other hand, factors such as N and C availability had a strong impact on the community functionality and composition. Fungal community structure reflected the availability of N in soils and the effect of elevated [CO2] on community structure and function was evident only in N-sufficient soils. The simultaneous availability of C and N was therefore the main driving force for microbial structure and function in this plantation. Recommendations and Perspectives  Using the soil instead of soil extracts for CLPP determination provides a direct measurement of substrate catabolism by microbial communities and reflects activity rather than growth because more immediate responses to substrates are measured. Further applications of this approach could include selective inhibition of different microbial functional groups to investigate specific CLPPs. To combine the structural analysis and the catabolic responses of specific microbial communities (i.e. fungi or bacteria) could provide new outlooks on the role of microbes on SOM decomposition. The effect of an elevated concentration of atmospheric CO2 and the application rate of nitrogen fertilizers on the microbial biomass and maximum specific growth rate of microorganisms in the soil and rhizosphere was studied in a long-term field experiment involving the growing of sugar beets and winter wheat. It was shown that the treatment of field plots with carbon dioxide at a concentration higher than that in the atmosphere (550 ppm) for three-four years resulted in the formation of a microbial community with a higher maximum specific growth rate and a larger share of R-strategy microorganisms as compared to the soil under the control plants. No reliable differences in the total microbial biomass in the soil under the winter wheat were revealed between the treatments with the ambient and elevated CO2 concentrations; in the soil under the beet plants, a reliable increase in the total microbial biomass at the elevated CO2 concentration was noted only in the close vicinity of the plant roots.

Carbon dioxide emissions from soils beneath canopies of two Mediterranean plants, Artemisia absinthium L. and Festuca pratensis Huds. cv. Demeter, were monitored over a 7-day period that included an artificial precipitation event of 4 cm. The experiments were conducted using 0.2 m3 soil microcosms inside greenhouses with CO2 concentrations of either 360 or 500 μmol mol−1. Carbon dioxide flux from the soil surface, as calculated using a diffusive transport model agreed well with CO2 flux measurements made using a dynamic flow system. Soil CO2 emissions did not differ significantly between the 360 and 500 μmol mol−1 CO2 treatments when soils were dry (volumetric soil moisture content ≤9%). A simulated precipitation event caused an immediate exhalation of CO2 from soil, after which CO2 emissions declined slightly and remained constant for approximately 36 h. CO2 emissions from soil microcosms with F. pratensis plants growing in 500 μmol mol−1 CO2 then rose to levels that were significantly greater than CO2 emissions from soils in the microcosms exposed to 360 μmol mol−1 CO2. For A. absinthium growing in 500 μmol mol−1 CO2, the rise in soil CO2 emissions following the wetting event was not significantly greater than emissions from soils with A. absinthium growing under 360 μmol mol−1 CO2. A. absinthium above ground biomass increased by 46.1 ± 17.9% (mean ± S.E., n = 4, P ≤ 0.05). Above ground biomass did not significantly increase for F. pratensis (14.4 ± 6.5%, P ≥ 0.10). Root biomass, on the other hand, increased for both species; by 50.6 ± 17.9% (P ≤ 0.05) for A. absinthium and by 55.9 ± 12.7% (P ≤ 0.05) for F. pratensis. Our results demonstrate two events following precipitation onto dry soils, an immediate release of CO2 followed by a gradual increase from enhanced biological activity The gradual increase was greater for the herbaceous ruderal perennial F. pratensis under elevated CO2.

Abstract

An anaerobic incubation experiment was conducted to investigate methane (CH4) production potential in soil samples collected from a paddy field after exposure to free-air CO2 enrichment (FACE). The FACE experiment with two CO2 levels, ambient and ambient + 200 p.p.m.v CO2 during the rice growing season, was conducted at Shizukuishi, Iwate Prefecture, Japan. The soil was a wet Andosol. Soil samples were taken from the surface (0–1 cm) and the sub-surface (1–10 cm) soil layers 2 months after rice harvest. Sub-samples of the fresh soils were put into glass bottles and submerged under N2 gas headspace during the incubation. The results showed that, prior to incubation, the contents of total C and dissolved organic C (DOC) were significantly greater in FACE soil than ambient soil. During the incubation, CH4 production potential was approximately 2–4-fold higher in FACE soil than ambient soil and approximately 500–1,000-fold greater in surface soil than sub-surface soil. In general, the FACE soil contained more DOC than ambient soil, particularly in the surface soil layer. These findings suggest that FACE treatment exerted long-term positive effects on CH4 production and increased organic C content in this paddy soil, particularly in the surface soil layer.

We compared, from 2004 through 2006, rates of soil–atmosphere CH4 exchange at permanently established sampling sites in a temperate forest exposed to ambient (control plots; ∼380 μL L−1) or elevated (ambient + 200 μL L−1) CO2 since August 1996. A total of 880 observations showed net atmospheric CH4 consumption (flux from the atmosphere to the soil) from all static chambers most of the time at rates varying from 0.02 mg m−2 day−1 to 4.5 mg m−2 day−1. However, we infrequently found net CH4 production (flux from the soil to the atmosphere) at lower rates, 0.01 mg m−2 day−1 to 0.08 mg m−2 day−1. For the entire study, the mean (±SEM) rate of net CH4 consumption in control plots was higher than the mean for CO2-enriched plots, 0.55 (0.03) versus 0.51 (0.03) mg m−2 day−1. Annual rates of 184, 196, and 197 mg m−2 for net CH4 consumption at control plots during the three calendar years of this study were 19, 10, and 8% higher than comparable values for CO2 enriched plots. Differences between treatments were significant in 2004 and 2005 and nearly significant in 2006. Volumetric soil water content was consistently higher at CO2-enriched sites and a mixed-effects model identified a significant soil moisture x CO2 interaction on net atmospheric CH4 consumption. Increased soil moisture at CO2-enriched sites likely increases diffusional resistance of surface soils and the frequency of anaerobic microsites supporting methanogenesis, resulting in reduced rates of net atmospheric CH4 consumption. Our study extends our observations of reduced net atmospheric CH4 consumption at CO2-enriched plots to nearly five continuous years, suggesting that this is likely a sustained negative feedback to increasing atmospheric CO2 at this site.
