A restriction fragment length polymorphism (RFLP) based linkage map of a cross between two diploid Hordeum bulbosum (2n = 2x = 14) clones, PB1 and PB11, was constructed from 46 recombinant progeny clones. Since both parents are heterozygous, separate and combined parental maps were constructed. All of the RFLP markers screened had previously been mapped in barley (H. vulgare L.) so that comparative maps could be produced. The PB1 linkage map consists of 20 RFLP marker loci assigned to four linkage groups covering 94.3 cM. The PB11 linkage map consists of 27 RFLP marker loci assigned to six linkage groups covering 149.1 cM. Thirteen markers polymorphic in both parents were used as ‘anchors’ to create a combined linkage map consisting of 38 loci assigned to six linkage groups and covering a genetic distance of 198 cM. Marker order was highly conserved in a comparison with the linkage map of H. vulgare (Laurie etal., 1995). However, in contrast, the genetic distances for the same markers were very different being 649 cM and 198 cM respectively, a genetic distance ratio of 1: 3.3. Thus although the map was short, it can be presumed to cover half the genome of H. bulbosum. This study provides further confirmation of the close relationship between the two species and gives a basis for the development of marker mediated introgression through interspecific hybridisation between the two species. Sorghum (Sorghum bicolor L. Moench) is regarded a drought‐tolerant alternative to maize as a bioenergy and fodder crop, but its early‐stage chilling sensitivity is obstructing a successful implementation in temperate areas. While several studies have identified quantitative trait loci (QTL) underlying chilling tolerance‐related traits in sorghum lines, little is known about the inheritance of these traits in F1 hybrids. We have conducted a comprehensive approach to analyse heterosis, combining ability and the relation between line per se and hybrid performance for emergence and early shoot and root development comprising both field trials and controlled environment experiments including chilling tests. To our best knowledge, this is the first study analysing heterosis for sorghum root parameters under chilling. Our results show that most traits are heterotic and that the mid‐parent values are rather poor predictors of hybrid performance. Hybrid breeding programmes should focus on efficient GCA tests and the establishment of genetically diverse pools to maximise heterosis rather than on a too strict selection among lines based on their per se performance. The medium‐to‐high heritabilities estimated for seedling emergence and juvenile biomass suggest that a robust breeding progress for these complex traits is feasible.  相似文献

Many important apple (Malus × domestica Borkh.) fruit quality traits are regulated by multiple genes, and more information about quantitative trait loci (QTLs) for these traits is required for marker-assisted selection. In this study, we constructed genetic linkage maps of the Japanese apple cultivars ‘Orin’ and ‘Akane’ using F1 seedlings derived from a cross between these cultivars. The ‘Orin’ map consisted of 251 loci covering 17 linkage groups (LGs; total length 1095.3 cM), and the ‘Akane’ map consisted of 291 loci covering 18 LGs (total length 1098.2 cM). We performed QTL analysis for 16 important traits, and found that four QTLs related to harvest time explained about 70% of genetic variation, and these will be useful for marker-assisted selection. The QTL for early harvest time in LG15 was located very close to the QTL for preharvest fruit drop. The QTL for skin color depth was located around the position of MYB1 in LG9, which suggested that alleles harbored by ‘Akane’ are regulating red color depth with different degrees of effect. We also analyzed soluble solids and sugar component contents, and found that a QTL for soluble solids content in LG16 could be explained by the amount of sorbitol and fructose.  相似文献

This paper attempts to review the present status and offers a perspective on sorghum [Sorghum bicolor (L.) Moench] use and breeding in Europe. Topics such as botany, origin, utilization, agronomic performances and improvement of sorghum are discussed. Clearly, producers and users of sorghum should be more familiar with the crop so that they can take advantage of some of the unique characteristics that are associated with sorghum as it relates to Europe.  相似文献

QTL analysis and mapping of pre-harvest sprouting resistance in Sorghum   总被引:2,自引:0,他引:2  
One of the most important agronomic problems in the production of sorghum [Sorghum bicolor (L.) Moench] in humid climates is pre-harvest sprouting (PHS). A molecular linkage map was developed using 112molecular markers in an F2 mapping population derived from a cross between IS 9530 (high resistance to PHS) and Redland B2 (susceptible to PHS). Two year phenotypic data was obtained. By means of interval mapping analysis, two significant QTL were detected in two different linkage groups with LOD scores of 8.77and 4.39. Each of these two QTL individually explained approximately 53% of the phenotypic variance, but together, in a two-QTL model, they explained 83% of the phenotypic variance with a LOD score of 12.37.These results were corroborated by a one-way ANOVA in which the four flanking markers of the most likely QTL positions displayed highly significant values in theF-test, and significant variation in trait expression was associated with marker genotypic classes. The four markers with highest effect in the one-way ANOVA were also detected in the second year replication of the F2 population, and significant genotype × environment interactions was observed. The putative relationship between PHS resistance in sorghum and the maize Vp1 gene is also discussed. For intrachromosomal mapping of the dominant GA-sensitive dwarfing gene Rht12 and the vernalization response gene Vrn1 on chromosome 5 A, an F2 population was established using a wide (synthetic) wheat cross. In addition to restriction fragment length polymorphism (RFLP) probes four microsatellite markers were incorporated. Rht12 was mapped distally to four RFLP loci (Xmwg616, Xpsr164, Xwg114, Xpsr1201) and three microsatellite markers (Xgwm179, Xgwm410, Xgwm291), known to be located on the segment of chromosome SAL which was ancestrally translocated and is homoeologous to Triticeae 4 L. The map position of Rht12 suggests that it is homoeologous to the dominant GA-sensitive dwarfing gene Ddw1, present on chromosome 5RL. The vernalization response gene Vrn1 showed linkage to Xwg644, as might be expected from comparative maps.  相似文献

Four intergeneric hybrids were produced by crossing sorghum [ Sorghum bicolor (L.) Moench, 2n =20] as a female parent with sugarcane ( Saccharum officinarum L., 2n = 112). The hybrids were morphologically similar to the sugarcane parent, but lacked vegetative vigour. Sorghum characters present in the hybrids included soft texture of the leaves, tight clasping of the leaf sheaths, presence of aerial roots and triangular ligule. All the hybrids were highly susceptible to natural incidence of mites. Somatic chromosome number of the hybrids ranged from 2n = 62–66, indicating n+n transmission. Target region amplification polymorphism (TRAP) is a relatively new PCR-based technique that detects large numbers of loci in a single reaction without extensive pre-PCR processing of samples. The aim of this study was to integrate TRAP markers in an EST-derived SSR linkage map of a RIL mapping population from the cross of the durum wheat cultivars Ciccio and Svevo, for a more general purpose of establishing a high-throughput system for genetic map saturation. Primer combinations producing PCR products with at least 4–5 polymorphic bands were selected and analyzed across the mapping population. The PCR reactions produced a total of 2,881 fragments with an average of 52 peaks per reaction. A total of 142 new TRAP markers were mapped and found to be randomly distributed in the genome. The total length of the map was 2,043.0 cM, with an average chromosome length of 145.9 cM. Homoeologous group one had the highest number of TRAP markers (38 loci) and the longest map length (407.9 cM) for a total of 87 markers, while the homoeologous group five had the lowest TRAP marker number (5 loci) and the shortest map length (232.5 cM). The distribution of markers among the seven homoeologous groups was random. The results indicate that TRAP is highly efficient in genetic mapping, generating a large number of markers scattered across the genome. This closes many existing gaps in marker coverage and may join otherwise separate linkage groups.  相似文献

Anthracnose, one of the destructive foliar diseases of sorghum growing in warm humid regions, is incited by the fungus Colletotrichum graminicola.The inheritance of anthracnose resistance was studied using the parental cultivars of Sorghum bicolor (L.) Moench, HC 136 (susceptible to anthracnose) and G 73 (anthracnose resistant). The F1 and F2 plants were inoculated with the local isolates of C. graminicola cultures. The F2 plants showed a segregation ratio of 3 (susceptible): 1(resistant) indicating that the locus for resistance to anthracnose in sorghum accession G 73 segregates as a recessive trait in a cross to susceptible cultivar HC 136. RAPD (random amplified polymorphic DNA) marker OPJ 011437 was identified as marker closely linked to anthracnose resistance gene in sorghum by bulked segregant analysis of HC 136 × G73 derived recombinant inbred lines (RILs) of sorghum. A total of 84 random decamer primers were used to screen polymorphism among the parental genotypes. Among these, only 24 primers were polymorphic. On bulked segregant analysis, primer OPJ 01 amplified a 1437 bp fragment only in resistant parent G 73 and resistant bulk. The marker OPJ 011437 was cloned and sequenced. The sequence of RAPD marker OPJ 011437 was used to generate specific markers called sequence characterized amplified regions (SCARs). A pair of SCAR markers SCJ 01-1 and SCJ 01-2 was developed using Mac Vector program. SCAR amplification of resistant and susceptible parents along with their respective bulks and RILs confirmed that SCAR marker SCJ 01 is at the same loci as that of RAPD marker OPJ 011437 and hence, is linked to anthracnose resistance gene. Resistant parent G 73 and resistant bulk amplified single specific band on PCR amplification using SCAR primer pairs. The RAPD marker OPJ 011437 was mapped at a distance of 3.26 cM apart from the locus governing anthracnose resistance on the sorghum genetic map by the segregation analysis of the RILs. Using BLAST program, it was found that the marker showed 100 per cent alignment with the contig{_}3966 located on the longer arm of chromosome 8 of sorghum genome. Therefore, these identified RAPD and SCAR markers can be used in the resistance-breeding program of sorghum anthracnose by marker-assisted selection.An erratum to this article can be found at  相似文献

For mapping the Sec2 and Sec5 loci of rye which determine expression of 75K γ-secalins, a partial genetic map of chromosome 2R spanning 64 cM was constructed. The map was developed using an F2 population of 103 plants from a cross between two inbred lines. Both loci were mapped distally on the short arm of chromosome 2R and clearly tagged in relation to 12 restriction fragment length polymorphism (RFLP) markers. The Sec2 locus was localized between the Xiag57 and Xpsr109a loci in an 11 cM interval. The Sec5 locus co-segregated to Xiag57 and was tightly linked to the Sec2 locus at a map distance of 0.5cM.  相似文献

Seed traits are important considerations for improving yield and product quality of chickpea (Cicer arietinum L.). The purpose of this study was to construct an intraspecific genetic linkage map and determine map positions of genes that confer double podding and seed traits using a population of 76 F10 derived recombinant inbred lines (RILs) from the cross of ‘ICCV-2’ (large seeds and single pods) × ‘JG-62’ (small seeds and double podded). We used 55 sequence-tagged microsatellite sites (STMS), 20 random amplified polymorphic DNAs (RAPDs), 3inter-simple sequence repeats (ISSR) and 2 phenotypic markers to develop a genetic map that comprised 14 linkage groups covering297.5 cM. The gene for double podding (s) was mapped to linkage group 6 and linked to Tr44 and Tr35 at a distance of7.8 cM and 11.5 cM, respectively. The major gene for pigmentation, C, was mapped to linkage group 8 and was loosely linked to Tr33 at a distance of 13.5 cM. Four QTLs for 100 seed weight (located on LG4 and LG9), seed number plant-1 (LG4), days to 50% flower (LG3) were identified. This intraspecific map of cultivated chickpea is the first that includes genes for important morphological traits. Synteny relationships among STMS markers appeared to be conserved on six linkage groups when our map was compared to the interspecific map presented by Winter et al. (2000). 脂肪及脂肪酸组分的改良是大豆油脂品质育种的主要方面。本研究旨在构建遗传图谱,定位大豆脂肪及脂肪酸组分的QTL,为大豆油脂品质育种提供参考。以Essex×ZDD2315的114个BC1F1单株为作图群体,构建了250个SSR标记和1个形态标记,具有25个连锁群的遗传图谱,覆盖大豆基因组2 963.5 cM,平均每个连锁群上10.0个标记,标记平均间距11.8 cM。用BC1F3家系3个重复的表型平均值代表相对应的BC1F1单株表型值,采用Win QTL Cartographer 2.5复合区间作图法(CIM)检测到18个控制脂肪及脂肪酸组分含量的QTL,位于9个不同的连锁群上,表型贡献率为9.6%~34.5%;多区间作图法(MIM)检测到与CIM区间相同的7个QTL(fat-1, pal-1, st-1, ole-1, lin-1, lin-4和lio-2),区间相近的2个QTL(ole-4和lin-5),位于6个不同的连锁群上,表型贡献率为8.2%~39.3%。CIM法检测到的其他9个QTL有待进一步验证。大豆脂肪及脂肪酸组分含量的主效QTL数量不多,效应大的不多,可能还受许多未能检测出来的微效基因控制,育种中既要注意主效QTL的利用,又要考虑微效多基因的积聚。  相似文献

Summary Nine cowpea (Vigna unguiculata (L.) Walp.) breeding lines were grown in 13 experiments under sole crop and intercrop management, with and without insecticide application, in Nigeria. Cowpea was intercropped with cassava (Manihot esculenta Cranz), maize (Zea mays L.) and maize-cassava in the forest zone and with sorghum (Sorghum bicolor Moench.) and millet (Pennisetum glaucum (L.) R.Br.) in the savanna zone. Line by cropping system interactions were significant in 5 of 13 experiments and line by insecticide treatment interactions were significant in 2 of 13 experiments. Cowpea yield was severely depressed in intercropped plots and in plots to which no insecticide was applied. Among-environment variation in cowpea yield was greater when no insecticide was applied. Within each management treatment, the yield performance and stability of the cowpea lines was examined. Stability was assessed both by examining among-environment coefficients of variation and by regression analysis. Several of the breeding lines evaluated appeared promising under different levels of management and in a diversity of environments.  相似文献

For the long‐term sustainability of the dairy industry in the Northeastern USA, manure nutrient application rates should not exceed crop nutrient removal once above‐optimum soil fertility levels are reached. Dairy producers have shown a growing interest in brown midrib (BMR) forage sorghum (Sorghum bicolor (L.) Moench.) × sudangrass (Sorghum sudanense Piper) hybrids (S × S) as a more environmentally sound alternative to maize (Zea mays L.) but data on S × S nutrient removal rates are scant. Our objectives were to determine N, P, K, Ca and Mg removal with harvest as impacted by N application rate, using six N rate studies in New York. One of the six sites had a recent manure history. Although site‐to‐site differences existed, N application tended to decrease P and K and increase N, Ca and Mg concentrations in BMR S × S forage. Nutrient removal and yield were highly correlated for all sites except one location that showed a K deficiency. The crop removed large amounts of P and K in the manured site, suggesting that BMR S × S is an excellent scavenger of these nutrients. If manure is applied mid‐season, forage K levels are likely too high for feeding to non‐lactating cows.  相似文献

To develop a high density linkage map in faba bean, a total of 1,363 FBES (Faba bean expressed sequence tag [EST]-derived simple sequence repeat [SSR]) markers were designed based on 5,090 non-redundant ESTs developed in this study. A total of 109 plants of a ‘Nubaria 2’ × ‘Misr 3’ F2 mapping population were used for map construction. Because the parents were not pure homozygous lines, the 109 F2 plants were divided into three subpopulations according to the original F1 plants. Linkage groups (LGs) generated in each subpopulation were integrated by commonly mapped markers. The integrated ‘Nubaria 2’ × ‘Misr 3’ map consisted of six LGs, representing a total length of 684.7 cM, with 552 loci. Of the mapped loci, 47% were generated from multi-loci diagnostic (MLD) markers. Alignment of homologous sequence pairs along each linkage group revealed obvious syntenic relationships between LGs in faba bean and the genomes of two model legumes, Lotus japonicus and Medicago truncatula. In a polymorphic analysis with ten Egyptian faba bean varieties, 78.9% (384/487) of the FBES markers showed polymorphisms. Along with the EST-SSR markers, the dense map developed in this study is expected to accelerate marker assisted breeding in faba bean.  相似文献

Intercropping represents an alternative to maize (Zea mays L.) monoculture to provide substrate for agricultural biogas production. Maize was intercropped with either sunflower (Helianthus annuus L.) or forage sorghum [Sorghum bicolor (L.) Moench] to determine the effect of seasonal water supply on yield and quality of the above‐ground biomass as a fermentation substrate. The two intercrop partners were grown in alternating double rows at plant available soil water levels of 60–80 %, 40–50 % and 15–30 % under a foil tunnel during the years 2006 and 2007 at Braunschweig, Germany. Although the intercrop dry matter yields in each year increased with increasing soil moisture, the partner crops responded quite differently. While maize produced significantly greater biomass under high rather than low water supply in each year, forage sorghum exhibited a significant yield response only in 2006, and sunflower in none of the 2 years. Despite greatly different soil moisture contents, the contribution of sorghum to the intercrop dry matter yield was similar, averaging 43 % in 2006 and 40 % in 2007. Under conditions of moderate and no drought stress, sunflower had a dry matter yield proportion of roughly one‐third in both years. In the severe drought treatment, however, sunflower contributed 37 % in 2006 and 54 % in 2007 to the total intercrop dry matter yield. The comparatively good performance of sunflower under conditions of low water supply is attributable to a fast early growth, which allows this crop to exploit the residual winter soil moisture. While the calculated methane‐producing potential of the maize/sorghum intercrop was not affected by the level of water supply, the maize/sunflower intercrop in 2006 had a higher theoretically attainable specific methane yield under low and medium than under high water supply. Nevertheless, the effect of water regime on substrate composition within the intercrops was small in comparison with the large differences between the intercrops.  相似文献

Little information exists concerning how crops will respond to the predicted increased night temperatures. The objective of this work was to determine if respiration and growth of sorghum [Sorghum bicolor (L.) Moench], a C, plant, and sunflower (Helianthus annuus L.), a C3 plant, are affected when the night temperature is increased by 5°C compared to the long-term (19 year) average night temperature in June in Kansas. Sorghum and sunflower were grown in two walk-in growth chambers with either the ambient night temperature (21C) or a high night temperature (26C). Day temperature was the same for all plants (27C). Both sunflower and sorghum had higher respiration rates under the elevated night temperature than under the ambient temperature. The average respiration rate of sunflower grown under elevated night temperature increased by 19% (0.89 vs. 0.75 μmol m?2 s?1) and that of sorghum by 44 % (0.52 vs. 0.36μmol m?2s?1). After 74 days, sunflower plants grown under the ambient night temperature were 30.2 cm taller than sunflower plants grown under the elevated night temperature; sorghum plants under the ambient temperature were 24.8 cm taller. Sunflower plants grown under the elevated night temperature formed flowers one week earlier than those grown under the ambient temperature. Sorghum formed no flowers by 74 days. Results suggest that, if climate change does increase night temperature, respiration will be increased more in C4, than C3 plants.  相似文献

A total of 415 sorghum (Sorghum bicolor (L.) Moench) accessions representing different regions of Ethiopia, Eritrea and a group of introduced lines were evaluated for 15 quantitative characters to determine the extent and geographical pattern of morphological variation. The extent of variation was highly pronounced for agronomically important characters for sorghum. These characters included plant height, days for 50% flowering, peduncle exsertion, panicle length and width, number and length of primary branches per panicle and thousand seed weight. Significant regional variation was also observed for most of the characters. The results implied that environmental factors such as altitude, rainfall, temperature and growing period are important in regional variation. Mean for plant height and for days for 50% flowering showed clinal variation along the gradients of rainfall pattern and growing period in Ethiopia. Moreover, there were significant positive correlation coefficients between most of the characters. This included the correlation between agronomic characters of primary interest in sorghum breeding such as plant height and days for 50% flowering and also between various characters and the altitude of the collection sites. The implications of the results in plant breeding, germplasm collection and conservation as well as the probable sources of the wide range of variation are discussed.
