选取2002~2007年从上海市分离鉴定的11株鸡新城疫病毒(Newcastle disease virus,NDV)强毒株,克隆其融合蛋白(Fusion,F)基因片段,测序后登录GenBank,序列号分别为NDV 022433(EU258661)、022435(EU258662)、0210149(EU258653)、0210154(EU258654)、0210227(EU258656)、0210252(EU258658)、03024(EU258637)、04011(EU258639)、05013(EU258640)、07011(EU258642)和07023(EU258643)。序列分析结果显示:上述分离株的F基因扩增片段为1 654 bp,编码551个氨基酸,其核苷酸序列同源性为94.6%~100%,推导的F蛋白氨基酸序列同源性为93.0%~100%;各分离株F蛋白裂解位点序列均为112R-R-Q-K-R-F117,为基因Ⅶ型NDV强毒株。系统发育分析表明:这11株NDV分离株之间的遗传距离较近,而与传统疫苗株B1、La Sota、V4和F48E9的遗传距离较远。此外,这11株NDV强毒株均具有大多数鹅源NDV毒株特有的973和1 249 nt RsaⅠ位点。  相似文献

为研究油橄榄叶提取物对灰葡萄孢(Botrytis cinerea)的抑制作用及机理,本试验采用溶剂浸提法制备油橄榄叶提取物,再与灰葡萄孢作用,通过测定菌落直径、菌丝生物量探究提取物对灰葡萄孢菌体生长的抑制作用,通过测定相对电导率、核酸和蛋白质泄露量考察油橄榄叶提取物对菌体细胞膜完整性的影响;并基于平板培养体系,采用碘化丙啶(PI)染色法研究提取物对灰葡萄孢细胞膜通透性、活性氧(ROS)代谢的影响。结果表明,灰葡萄孢对油橄榄叶90%乙醇提取物具有高敏感性,经油橄榄叶提取物处理后其菌落直径和菌丝生物量分别为对照的20%、8.91%,膜电导率、核酸以及蛋白质泄漏量显著增加;孢内超氧阴离子(O2·-)、过氧化氢(H2O2)含量显著增加,还原型辅酶Ⅱ氧化酶(NOX)、过氧化氢酶(CAT)、过氧化物酶(POD)活性上升,超氧化物歧化酶(SOD)、谷胱甘肽还原酶(GR)、抗坏血酸过氧化物酶(APX)活性下降。综上,油橄榄叶90%乙醇提取物处理后的灰葡萄孢细胞膜受到损伤,导致细胞内容物外泄,并诱导ROS在胞内积累,...  相似文献

番茄黄化曲叶病毒(tomato yellow leaf curl virus,TYLCV)是一种由烟粉虱(Bemisia tabaei)和嫁接传播的双生病毒,在热带、亚热带地区给番茄生产造成严重威胁。根据番茄黄化曲叶病毒的保守序列设计一对引物,运用PCR技术从上海地区的感病番茄中扩增出一条575bp的特异带,而健康植株无此带。测序表明该序列与番茄黄化曲叶病毒具有极高的同源性(97%~99%)。将健康接穗嫁接到感染番茄黄化曲叶病毒的番茄砧木上,间隔15d和30d,分别提取接穗的DNA,并用PCR法检测病毒,发现嫁接15d后在部分接穗中检测到TYLCV病毒,嫁接30d后在所有的接穗中均检测到病毒,因此,嫁接法可以作为番茄黄化曲叶病毒病的接种鉴定方法。  相似文献

水稻几丁质酶的原核表达、复性及体外抑菌活性的研究   总被引:3,自引:0,他引:3  
利用RT-PCR反应,从纹枯菌诱导的水稻叶片cDNA中克隆了1个水稻几丁质酶基因RC7的全长编码序列(ORF)。通过亚克隆方法,RC7与原核表达载体pGEX-4T-1上的GST融合,构建成GST-RC7重组蛋白的原核表达载体pGST-RC7,然后,将其转化到大肠杆菌细胞中。经异丙基-β-D-硫代半乳糖苷(IPTG)诱导,获得了以包涵体形式高效表达的GST-RC7。经过洗涤、溶解(变性)、复性及纯化等处理,包涵体中GST-RC7得到溶解、复性和纯化。纯化的复性GST-RC7蛋白具有几丁质酶活性。对6种重要作物病原真菌进行体外抑菌活性分析的结果表明, RC7可显著抑制水稻纹枯病菌、小麦纹枯菌、小麦赤霉菌、棉花黄萎菌、烟草赤星菌的菌丝生长,说明RC7可作为上述植物病害抗性基因育种的重要基因。  相似文献

考虑到轴流泵叶轮出口导叶进口区域水流复杂,该文在常规设计导叶基础上通过改变导叶叶片前掠和后掠的角度,期望导叶能够尽可能的回收叶轮出口的速度环量,提高轴流泵段的效率。该文采用计算流体动力学软件研究后置导叶在不同的扫掠角度下对轴流泵段水力性能的影响。以常规设计导叶为基础,一共研究计算了6种不同导叶扫掠方案,每种导叶扫掠方案又计算了8个不同流量工况点。根据数值模拟结果,分析了不同导叶扫掠角度对轴流泵段能量特性的影响,对导叶和出水弯管的水力损失进行了定量计算。最后对前掠16°导叶进行了泵段能量性能试验,并结合数值模拟对该文主要结论进行验证分析。研究结果表明:导叶扫掠角度对轴流泵段性能影响主要体现在小流量工况,且导叶叶片前掠效果比后掠好;导叶叶片前掠16°时,整流效果最好,导叶损失和出水弯管损失最小,效率最高;试验数据与数值模拟结果各点误差在3%以内,验证了数值模拟结果的可靠性、准确性。研究结果不仅有助于导叶水力性能的优化设计,同时对提高泵段的效率提供了参考。  相似文献

X-ray absorption was used as an index to detect the quality of mango based on its relationship with density, moisture content, soluble solids, titratable acidity, and pH. A regression equation was obtained between the computed tomography (CT) number and the physicochemical properties based on the principle of X-ray absorption capacities of the fruit. The CT numbers of intact mangoes were positively correlated with density, moisture content, and titratable acidity. The soluble solids and pH were inversely related with CT number. The results suggest that CT number measurements on intact fruit can be used as a nondestructive indicator of the quality of mango.  相似文献

探讨叶面喷施和土施外源硒对芒果叶片和果实硒含量、果实矿质元素含量及营养品质的影响,以期为富硒芒果的安全生产提供理论依据和技术支持。以“桂热 82号”芒果为试材,叶面喷施以亚硒酸钠为硒源,设置0.0、25.0、50.0、100.0、150.0和 200.0 mg/L 6个浓度水平,于生理落果结束后,每 15 d喷施一次,连喷 2次。土施设置亚硒酸钠处理和硒酸钠处理,施肥量为 10 g/株,于花期沟施。结果表明,芒果叶片和果实中的硒含量随着叶面喷硒浓度的增加而递增。叶面喷施 50 mg/L亚硒酸钠可显著提高果实 Vc含量,200 mg/L亚硒酸钠可显著提高果实K、可滴定酸含量,100~ 200 mg/L亚硒酸钠可显著降低果实Zn、Ca含量。土施相同量的亚硒酸钠和硒酸钠均可显著提高芒果叶片和果实硒含量。土施硒酸钠叶片和果实硒含量显著高于亚硒酸钠。土施 10 g/株亚硒酸钠显著降低果实 Ca、Mg含量,土施 10 g/株硒酸钠显著提高果实 K、Vc含量,降低果实 Ca含量。叶面喷硒和土壤施硒均可显著提高芒果叶片和果实硒含量,其中叶面喷硒提高幅度远大于土壤施硒提高幅度。土施硒酸钠比亚硒酸钠更容易被芒果吸收。富硒芒果生产上推荐叶面喷施 25.0~ 50.0 mg/L亚硒酸钠,或者土施 10 g/株硒酸钠。  相似文献

蓖麻叶片粗提物对黄瓜瓜绢螟的作用   总被引:1,自引:0,他引:1  
为了测定蓖麻对瓜绢螟的作用,也为黄瓜害虫无公害控制提供新的手段,用不同浓度的蓖麻叶片粗提物处理黄瓜叶片,测定瓜绢螟取食量、存活率及体内解毒酶活性的变化。结果显示,蓖麻叶片粗提物对瓜绢螟的取食和存活有明显的抑制作用,处理浓度越大,取食量和存活率越小。如用稀释200倍的蓖麻叶片粗提液处理黄瓜叶片,瓜绢螟的取食面积(1.89 cm2)仅为对照(3.51 cm2)的53.7%,存活率为对照的2.3%;600倍浸提液处理的瓜绢螟取食面积(3.43 cm2)为对照的97.7%,存活率为对照的51.4%。瓜绢螟幼虫虫龄对粗提物的作用有明显影响,粗提物对低龄幼虫的作用明显强于高龄幼虫,200倍液处理24 h后,1龄幼虫的存活率(22.22%)仅为5龄幼虫(66.67%)的33.33%。不同剂量蓖麻叶片粗提物处理后,瓜绢螟体内的谷胱甘肽转移酶、羧酸脂酶、多功能氧化酶等解毒酶活性迅速上升,随着处理剂量的增加,解毒酶的活性上升幅度增大。取食用稀释200倍的蓖麻粗提液处理的黄瓜叶片24 h后,1龄幼虫和5龄幼虫体内谷胱甘肽转移酶活性分别较对照高23.79%和78.02%,羧酸脂酶活性分别较对照高72.42%和23.84%,多功能氧化酶活性分别较对照高100.58%和122.39%。结果表明,蓖麻叶片粗提物对瓜绢螟有明显的拒食和毒杀作用。  相似文献

Mango (Mangifera indica L.) is one of the most economically grown fruits in the tropical and subtropical areas around the world. In the present study, start codon targeted (SCoT) markers were employed to investigate the genetic diversity of 73 mango accessions obtained from Guangxi province, China. A total of 275 bands were amplified by thirty-four SCoT primers, of which 203 (73.82%) were polymorphic. Genetic similarity between accessions was in the range of 66.2–94.2% with an average of 78.8%. The observed highest genetic similarity value (94.2%) was found between ‘Ren Mian Mango’ and ‘Hong Hua Mango’, the observed lowest genetic similarity value (66.2%) was found between‘Xia Mao Xiang Mango’ and ‘India No. 15’. These coefficients were utilized to construct a dendrogram using the unweighted pair group of arithmetic means. All the accessions were grouped into four (A, B, C, D) clusters and correspond well with their geographical origin and their known history. These results have an important implication for mango’s rue germplasm characterization, improvement, management and conservation.  相似文献

In parallel with the rising popularity of exotic fruits in Europe, allergy against mango is of increasing importance. Because mangoes are also consumed as processed products such as chutneys or beverages, the influences of different process conditions on their allergenicity were investigated. Mango purees and nectars were manufactured at small pilot-plant scale, and the allergenic potencies of the resulting intermediate and final products were determined by means of sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), immunoblotting and inhibitive enzyme allergosorbent tests (EAST-inhibition), using a pool serum of 9 individuals with manifest mango allergy. The mango allergens were shown to be very stable during technological processing. Irrespective of enzymatic matrix decomposition, mechanical tissue disintegration and heating during peeling, mash treatment, and pasteurization, significant loss of allergenicity could not be observed in the extracts of mango purees and nectars derived thereof. These results were confirmed by analogous investigation of commercial mango drinks and nectars. Hence, conventional mango processing into pulp-containing products typical for this species obviously does not allow complete elimination of the allergenic potency.  相似文献

This paper reports a very simple and fast method to collect eluates with high amounts of hydroxytyrosol, biotransforming Olea europaea L. leaf extract by a thermophilic beta-glycosidase immobilized on chitosan. Some phenolic compounds in the leaf tissue and in the eluates obtained by biotransformation are identified. To propose the eluates as natural substances from a vegetal source, their antioxidant properties have been compared with those of the leaf extract from which they are originated. The eluates possess a higher concentration of simple phenols, characterized by a stronger antioxidant capacity, than those available in extra virgin olive oils and in many tablets of olive leaf extracts, commercially found as dietetic products and food integrators.  相似文献

Information about the nutritional status of a plant is a basic prerequisite for its adequate nutrition and is crucial to achieve high-yield productivity. Assessing the annual amount of nutrient that a tree needs to absorb in order to successfully complete a vegetative and reproductive growth is a fundamental step for developing rational fertilization in orchards. We are unaware of any report describing macro- and micronutrient dynamics in fruit at different growth stages of mango. Seven cultivars of mango (Dashehari, Langra, Mahmood Bahar, Menka, Sabri, Sundar Langra and Zardalu) were selected for the study. Different macronutrients such as nitrogen, phosphorus, potassium, calcium and magnesium (N,P, K, Ca, and Mg) and micronutrients such as iron, manganese, zinc and copper (Fe, Mn, Zn, and Cu) were analyzed at four growth stages, namelymarble stage, prestone formation stage, stone hardening stage and harvest stage. There was a significant variation in nutrient content of fruits throughout the developmental stages irrespective of cultivars. There was no definite trend of nutrient variation among cultivars but in general, most of the nutrients like N, P, K, Mg, Mn, Zn, and Cu were the highest at the marble stage, and Ca and Fe at the stone hardening stage.  相似文献

Abstract

This investigation was conducted during two successive seasons (2014/2015) and (2015/2016) using 15?years old productive mango (Mangifera indica) trees cv. Zebda. The trees were grown at AlMalak Valley Farm, El-Sharkeya Governorate- Egypt (30–51° North; 32–53° East). Trees were planted 8?×?8 meter within and between rows in sandy soil under drip irrigation system using the Nile water. The objective of this study is to alleviate alternate bearing in cv. Zebda using mineral nutrients (nitrogen in the on year and boron in the off year). Treatments included three concentrations of nitrogen (1000, 1250, 1500?g/tree/year) and three concentrations of boron (0.0, 250, 500?mg L?1). Nitrogen was applied to the soil as ammonium sulfate and boron was applied as foliar spray of boric acid. The extra amount of nitrogen fertilizer (250 and 500?g N/tree) was applied at three installments in (May, June and July). Treatment was arranged in a factorial Completely Randomized Block Design with three replicates for each treatment. Results show that the on-year nitrogen fertilization significantly increased mango tree vegetative growth (number of shoot/branch, shoot length, shoot thickness, number of leaves/meter and leaf area) and yield. The average yield in the on year is 85.5?kg/tree at 1250?g N/tree but 67.4?Kilogram/tree at 1000?g N/tree (the control treatment). While in the off year boron foliar application resulted in a significant increase in flowering, initial fruit set, final fruit set and fruit yield. The average yield in the off year is 47?kg/tree at 250?mg L?1 boron but 9?kg/tree at 0.0?mg L?1 boron rate (the control treatment).The interaction treatment of 250?mg L?1 boron + 1500?g nitrogen/tree is the best treatment as it resulted in the highest values for all the tested parameters. The average yield of this treatment is 53.5?kg/tree. This treatment helps alleviate alternate bearing phenomenon by 41% and obtain the highest economic yield in the off-year, i.e. increased yield by 5.9 fold.  相似文献

A rapid method for quantitative determination of beta-carotene, including cis-isomers, in dried mango has been developed. Applicability of available methods to dried products was limited because of formation of artifacts caused by extraction and preparation. The analytical procedure was based on the extraction of carotenoids from dried mango mesocarp using a mixture of methanol and acetone/hexane, allowing the separation of disturbing fibers. No saponification was required. Furthermore, carotenoid determination by HPLC on a C30 stationary phase was achieved. This method was applied to determine beta-carotene and its stereoisomers in fresh, dried, and solar-dried mango slices of four cultivars. Drying resulted in a complete and partial degradation of xanthophylls and all-trans-beta-carotene, respectively. Isomerization was shown to depend on the drying process. Whereas conventionally dried mangoes were characterized by elevated amounts of 13-cis-beta-carotene, solar-dried mango slices contained additional amounts of the 9-cis-isomer. Calculation of vitamin A values was based on the real amount of the beta-carotene stereoisomers and ranged from 113 to 420 and from 425 to 1010 RE/100 g for fresh and dried mango slices, respectively.  相似文献

An aqueous decoction of mango (Mangifera indica L.) stem bark (MSB) has been developed in Cuba on an industrial scale to be used as a nutritional supplement, cosmetic, and phytomedicine, with antioxidant, anti-inflammatory, analgesic, and immunomodulatory properties. The concentration of major and trace elements was determined for 16 varieties of MSB belonging to two cultivars and grown in Cuba in the same soil (red ferralytic). Plants were classified into two groups, according to the tree age (12 and 26 year olds) and were analyzed for As, Ca, Cd, Cu, Fe, Hg, K, Mg, Pb, Se, and Zn content by means of ICP-AES technique. Experimental data were processed by ANOVA and principal component analysis in terms of elements, variety, and plant age, to choose the most adequate varieties for industrial purposes.  相似文献
