Anthelmintic efficacy of ivermectin (greater than or equal to 80% 22,23-dihydroavermectin B1a and less than or equal to 20% 22,23-dihydroavermectin B1b) was evaluated in a controlled experiment against immature nematodes. Twenty calves raised to about 8 weeks of age under nematode-free conditions were allocated into 4 groups. Infective larvae of Haemonchus contortus, Ostertagia ostertagi, Trichostrongylus axei, T colubriformis, Cooperia oncophora, C punctata, Oesophagostomum radiatum, and Dictyocaulus viviparus were given orally to each calf on a staggered schedule 6 to 14 days before treatments. The drug was given subcutaneously when the nematodes were in the early 4th stage of development, and the calves were killed 23 or 24 days later. Group 1 nonmedicated control calves had a geometric mean of 25,102 nematodes; groups 2, 3, and 4 calves given ivermectin at dose rates of 50, 100, or 200 micrograms/kg had reductions of 88.2%, 98.0%, and 99.8%, respectively.  相似文献

The anthelmintic activity of ivermectin when administered orally and subcutaneously at a dosage of 0.2 mg/kg of body weight was evaluated in 4 trials involving 20 dromedary camels (Camelus dromedarius). Fecal egg counts were made on the day of treatment and at 2, 4, and 10 weeks after treatment. Trichostrongylid egg counts were reduced by a maximum of 100% when ivermectin was administered orally and by greater than 88% when given subcutaneously. Egg counts of Trichuris spp were reduced by greater than 85% with oral administration, but increased following subcutaneous treatment. No adverse local or systemic reactions were observed following treatment with ivermectin by either route.  相似文献

Four groups of 16 crossbred beef calves were used in evaluating different anthelmintic treatment schedules: group 1 was given ivermectin (IVM) at weaning only (October 31) and grazed on initially safe pasture; group 2 was given IVM at weaning, on January 28, and on April 22, and grazed on contaminated pasture; and group 3 was given IVM at weaning and on April 22, and grazed on contaminated pasture; and group-4 was group treated with fenbendazole (FBZ) at weaning only, with provision for individual salvage treatment, and grazed on contaminated pasture. The investigation was from Oct 31, 1984, to Oct 9, 1985. Initially high fecal egg counts at weaning were more effectively reduced by IVM than by FBZ, and the effect of safe pasture was evident in minimal worm burdens in tracer calves grazed with group-1 cattle during November and least amount of weight loss in group-1 yearlings during winter. Fecal egg counts, pasture larval counts, and plasma pepsinogen concentrations remained low in group 2 after the January treatment. Fecal egg counts of the other groups increased substantially during late winter and spring, but pasture larval counts increased only on group-1 and group-4 pastures. During spring, highest worm burdens were found in group-1 and group-4 tracer calves (grazed in April) and in group-3 and group-4 yearlings (slaughtered in early April). Six cattle of group 4 were salvage treated with FBZ in February and April. A series of randomized, controlled, masked, field (veterinary patient) studies were conducted in the USA and Europe to evaluate the efficacy of selamectin, a novel macrocyclic lactone of the avermectin subclass, in the treatment of naturally acquired gastrointestinal nematode infections in cats. After confirmation of ascarid and/or hookworm infection, 298 cats of various ages and breeds were randomly assigned to treatment with selamectin (n=202) or an existing commercially approved positive-control product (n=96). Unit doses of selamectin (providing a minimum dosage of 6mgkg(-1)) were administered topically to the skin in a single spot at monthly intervals. Quantitative fecal examinations were performed on days 0 (before treatment), 30, and 60. In the selamectin-treated cats, fecal ascarid egg counts were reduced by 99.6 to 100% on day 30, and by 99.9 to 100% on day 60. Fecal hookworm egg counts were reduced by 98.3% on day 30, and by 100% on day 60 in the selamectin-treated cats. The positive-control products achieved reductions in egg counts of 96.5 to 100% (ascarids) and 98.9 to 99.9% (hookworms). These studies have shown that monthly topical administration of selamectin is safe and highly effective in the treatment of naturally acquired ascarid and hookworm infections in cats.  相似文献

The aim of this study was to assess, by a clinical trial, the efficacy of an ivermectin‐based pour‐on treatment against gastrointestinal parasitic nematodes in naturally infected horses using 2 groups of mature indigenous Pura Raza Galega grazing mares. Faecal and blood samples were collected individually over a 21 week period. Faeces were analysed by the coprological flotation, sedimentation and migration techniques. Changes in circulating blood cells were monitored over the study period. The administration of the ivermectin suppressed the eggelimination of ascarids and pinworms throughout the study and no strongyle‐eggs were observed in the treatment group between the 3rd and 10th weeks. The numbers of red cells increased significantly after the anthelmintic therapy, and a statistical reduction in circulating leucocytes was recorded. No side effects were observed. The pour‐on ivermectin formulation was highly successful against gastrointestinal nematodes and appears to be a useful therapeutic routine for large groups of horses.  相似文献

Twelve calves (mean weight, 175.5 kg) were used to confirm efficacy of ivermectin delivered from a prototype sustained-release bolus against naturally acquired gastrointestinal nematodes including early fourth-stage (inhibited) larvae of Ostertagia ostertagi. The calves were allocated by restricted randomization on weight to 1 of 2 groups: controls, to which a placebo bolus was given orally, and treated calves, to which a sustained-release bolus designed to deliver 8 mg of ivermectin/day at a steady rate was given orally. After treatment, the 2 groups were housed in separate pens with concrete flooring. Twenty-eight days after treatment, all calves were euthanatized and necropsied. The ivermectin-treated calves had no larval or adult Ostertagia spp and significantly (P less than 0.01) fewer adult Trichostrongylus axei and adult Cooperia (C oncophora, C punctata and C surnabada) than control calves. Efficacy of ivermectin was greater than 99% for Cooperia spp, and 100% for other parasites. Drug-related adverse reactions were not observed.  相似文献

Levamisole, morantel, fenbendazole, or ivermectin was administered at 2-week intervals from May 1 through Sept 14, 1985, to breeding ewes (20 ewes/drug) infected with various gastrointestinal nematodes. All ewes had fewer gastrointestinal nematode eggs per gram of feces (epg) after 2 treatments, compared with pretreatment epg counts. Ewes administered ivermectin continued to have a low mean epg (0 to 3) throughout the study. The mean epg counts of ewes treated with levamisole increased from 3 to 483 during the study. This increase was similar to that of ewes treated with morantel (7 to 485 epg). The mean epg count of fenbendazole-treated ewes increased from 4 to 192 during the study. By the end of the study, the mean epg counts when expressed as a percentage of the pretreatment epg counts were 4% (ivermectin), 249% (fenbendazole), 627% (levamisole), and 630% (morantel). With the exception of the ivermectin-treated ewes, the epg count increased almost linearly in the ewes after the 2nd anthelmintic treatment. These data indicate that the gastrointestinal nematodes (including Haemonchus contortus) may have developed more resistance to levamisole and morantel than to fenbendazole. On the basis of the epg counts, resistance to ivermectin did not develop during the 4.5-month treatment period. The percentage of ewes shedding eggs after 2, 4, and 6 anthelmintic treatments was lowest for ewes treated with ivermectin (20%) and was similar (40%) for ewes treated with 1 of the other 3 anthelmintics. The efficacy of a morantel sustained release bolus (MSRB) for control of gastrointestinal nematodes in yearling steers was evaluated over a 6-month grazing period commencing on 26 March 1982. Three groups of 15 steers were allotted to the following treatments: Group 1 -- one MSRB at start of trial; Group 2 -- one therapeutic dose of thiabendazole at start of trial; Group 3 -- untreated control. The treatment groups were grazed separately. Parasite egg counts (EPG), herbage larval counts, pepsinogen levels and weight gains were monitored. Every other month, sets of 2 parasite-free tracer calves were placed in the pasture grazed by each treatment group and allowed to graze for 3 weeks before being subsequently necropsied for worm counts. At the end of the trial, 6 animals from each group were also necropsied for worm counts. The MSRB treatment resulted in significantly lower egg counts, fewer infective larvae on pasture, lower pepsinogen levels and lower worm burdens in tracer calves than was the case for the untreated group, but generally the levels were not significantly different from those associated with the thiabendazole treatment. The mean weight gain for the MSRB treated steers showed a significant advantage (70.9 lb) over the untreated animals, but was not significantly different from those which received thiabendazole. Dairy replacement heifers given two strategic treatments with ivermectin (0.2 mg/kg) three and eight weeks after turnout to spring pastures in northern USA had mean adjusted weight gains (62.3 kg) that were 62 per cent (P less than 0.02) greater than those of control heifers (38.5 kg) at the time of winter housing. Pasture infectivity was consistently higher in control pastures and there was a sixfold difference between treatment (56 L3/kg) and control (358 L3/kg) pastures by the time of winter housing on October 18. This study confirmed earlier studies in northern USA demonstrating the value of two strategic treatments in achieving significant increases in weight gains and hastening the time of first breeding by one to three months at a saving of $40 to $140 per heifer. It was concluded that this dosing strategy is a satisfactory method of parasite control for dairy replacement heifers in northern USA without undue selection for drug resistance.  相似文献

The efficacy of an oral formulation of ivermectin plus praziquantel in the reduction of nematode and cestode egg counts in horses was assessed in 273 horses under field conditions at 15 sites in North America (n = 6) and Europe (n = 9). Horses were confirmed by fecal examination to have natural infections of strongyles (100%) and tapeworms (76%). Replicates of four horses were formed at each site, and in each replicate three animals received ivermectin (0.2 mg/kg body weight) plus praziquantel (1 mg/kg body weight) oral paste and one animal remained untreated or received vehicle paste. Fecal samples were collected for fecal nematode and cestode egg counting before and 7, 8, 9, 14, 15, and 16 days after treatment. Horses treated with ivermectin plus praziquantel oral paste had significantly (P <.01) lower posttreatment strongylid and cestode egg counts (reductions of 98% or more) than controls. Combined site analyses revealed that 95% or 96% of the horses positive for cestode eggs before treatment that were treated with ivermectin plus praziquantel were negative for cestode eggs at each posttreatment fecal examination. No adverse reactions attributable to ivermectin plus praziquantel oral paste treatments were observed. The results of the studies demonstrated that ivermectin plus praziquantel paste was highly effective in reducing egg shedding by gastrointestinal nematodes and cestodes, and no adverse reactions were observed in horses treated under field conditions.  相似文献

Alternatives to chemical dewormers are needed to counter anthelmintic resistance and improve worm control in organic management systems. The objective was to examine the effectiveness of grazing sericea lespedeza (SL) compared with grass pastures for control of gastrointestinal nematodes (GIN) in lambs. In Experiment 1, Katahdin lambs grazed bermudagrass (BG; n=14), tall fescue (TF; n=7), or SL (n=19) pastures during early summer months. In Experiment 2, lambs grazed TF (n=15) or SL (n=13) pastures during late summer. Stocking rate of pastures was based on forage availability; additional lambs grazed pastures in Experiment 2, but were not sampled. Lambs were dewormed with 0.5 g COWP if FAMACHA(?) score was >3. In Experiment 1, FEC were reduced within 35 days in SL compared with BG lambs (forage by time, P=0.03). The PCV was more resilient to changes over time in SL compared with other groups of lambs (forage by time, P=0.001). In Experiment 2, FEC were lower (P=0.02) and PCV tended to be higher (P=0.09) in lambs grazing SL compared with TF forage. Incidence of deworming was similar among forage groups in both experiments. Grazing SL reduced FEC in lambs in early and late summer, despite reluctance by lambs to graze. Grazing forage and selective deworming using COWP was effective in lambs.  相似文献

The incidence of mange in dairy buffalo in India has increased significantly in recent years. The authors record an outbreak of mange affecting a dairy herd stocking about 30,000 buffalo and 1000 cows. The mange mites were either Sarcoptes scabiei or Psoroptes ovis, or a mixed infestation of both. The morbidity rate was 5-30% varying from group to group, with 100% in a severely affected group. Signs noticed were progressive dermatitis, alopecia, keratinization, skin thickened and wrinkled, intense itching and marked loss of condition often ending in death. Great losses of young animals from mange and gastrointestinal nematodes are very common in dairy herds in India. In view of their economic importance, the activity of ivermectin against naturally occurring mange and parasitic infections of adult buffalo and buffalo calves was determined. Ivermectin was administered by subcutaneous injection (IVOMEC 1% w/v - MSD AGVET) at a dose of 200 mcg kg-1 body weight. The efficacy was ascertained from the disappearance of mites from skin scrapings and the reduction in numbers of worm eggs in the faeces. The results of the treatment were dramatic: the mites disappeared within 2 weeks of the drug being administered in the majority of animals, with marked improvement in skin lesions. Four buffalo which had their entire body surface affected with mange needed a second dose on Day 28 for complete recovery. The effect on the nematodes was equally spectacular, with infections of Neoascaris vitulorum, Trichostrongylidae, Oesophagostomum spp. and Bunostomum being eliminated within 1 week of treatment.  相似文献

Prevalence of anthelmintic resistance in gastrointestinal nematodes (GIN) of goats has necessitated studies for alternative means of control. The objective was to determine the effectiveness of dietary copper sulfate for control of GIN in meat goats. Naturally infected buck kids received 0 (LC), 78 (MC) or 158 (HC) mg copper sulfate (CS)/kid daily mixed with concentrate supplement for 63 days. After 42 days the HC group was re-randomized into either the LC or MC treatment. In another study, naturally infected yearling does were offered trace mineral mix with and without CS and intake was estimated to be 140-200mg CS/day. Goats grazed bermudagrass pastures and were rotated among two or three pastures to minimize pasture effects. Fecal egg count (FEC) and packed cell volume (PCV) were determined every 7 days in the first experiment and 14 days in the second experiment, and goats were weighed every 28 days. On Days 49 and 56 FEC were lower in the HC-treated kids (copper by day, P<0.02), but FEC were similar on all other days in the first experiment and were similar between the two groups of does in the second experiment. Blood packed cell volume was similar among treatment groups throughout both studies. Body weight was greater in LC compared with MC or HC-supplemented kids on Days 42 and 63 (copper by day, P<0.04). Body weights of does were similar on Days 0 and 56, but were reduced in those consuming trace mineral with CS on Day 28 (copper by day, P<0.03). On the basis of worm counts in naturally infected Angora-X goats, albendazole at either 3.8 mg/kg repeated after 24 hours or as a single dose at 7.6 mg/kg, was >99% effective in removing adult Ostertagia and Trichostrongylus spp. Small numbers of adult Haemonchus contortus, Cooperia sp. and Chabertia ovina were present in control but not treated animals. The single dose of 7.6 mg/kg removed >99% of adult Oesophagostomum venulosum whereas the repeated dose of 3.8 mg/kg removed only 96%.  相似文献

Thirteen Standardbred horses, two to five years of age, were treated with ivermectin paste per os at 200 μg/kg of body weight and 13 were untreated. Two weeks after treatment, previously untreated horses were given the paste. Fecal samples were collected from all horses at the time of treatment and periodically thereafter up to 14 weeks and were examined for nematode eggs using the Cornell-McMaster dilution and the Cornell-Wisconsin double centrifugation procedures.

All horses consumed the paste readily and had no signs of toxicosis. Strongyle eggs were found in the feces of all horses before treatment but not at two to three weeks after treatment. At five to six weeks after treatment only two horses had eggs in the feces. At eight, ten, 12 and 14 weeks after treatment 27, 69, 88 and 100% of the horses examined, respectively, had a few strongyle eggs but these were no greater than 18% of that of the pretreatment samples. Ivermectin oral paste, therefore, appeared to be highly effective against both adult and immature strongyles.

Selected cardiorenal functions for conscious and chaired rhesus macaques were compared directly and indirectly with corrections for body weights and surface areas to well-established base-line values for human subjects. Certain functional differences between species and conditions of measurement do not allow all experimental results from the rhesus macaque to be extrapolated to man. However, this comparative study provides evidence that can be useful in attempting to evaluate cardiorenal data from macaques in relationship to human studies.  相似文献
