Chlorophyll and glycoalkaloid synthesis in potato (Solanum tuberosum L.) tubers occur in direct response to light. The two processes are concurrent, but independent. Color photographic indices to subjectively grade fresh market potatoes for the extent of greening were developed under lighting conditions consistent with those of retail markets. Total glycoalkaloid (TGA) and chlorophyll accumulation for four cultivars were determined over the respective greening scales, thus calibrating the scales for TGA content. On average, TGA concentrations in complete longitudinal sections of tubers (flesh samples) were highest in Dark Red Norland followed by Russet Norkotah, Yukon Gold, and White Rose. TGA concentrations of flesh samples of White Rose and Yukon Gold tubers were somewhat variable and did not increase in direct proportion to greening level and chlorophyll content, particularly at higher levels of greening. TGA concentrations in Dark Red Norland and Russet Norkotah tubers were highly correlated (P < or = 0.001) with greening level and chlorophyll concentrations. When averaged over greening levels, skin samples contained 3.4- to 6.8-fold higher concentrations of TGAs than flesh samples, depending on the cultivar. The TGA concentration in periderm samples ranged from 37 to 160 mg/100 g of dry wt. Regardless of greening level, concentrations of TGAs in the flesh samples (including attached periderm) remained within limits presumed safe for human consumption. Discrimination of greened tubers on the basis of perceived glycoalkaloid toxicity is likely unfounded for the cultivars and greening levels studied.  相似文献

Vegetable flavor is an important factor in consumer choice but a trait that is difficult to assess quantitatively. The purpose of this study was to assess the levels of the major umami compounds in boiled potato tubers, in cultivars previously assessed for sensory quality. The free levels of the major umami amino acids, glutamate and aspartate, and the 5'-nucleotides, GMP and AMP, were measured in potato samples during the cooking process. Tubers were sampled at several time points during the growing season. The levels of both glutamate and 5'-nucleotides were significantly higher in mature tubers of two Solanum phureja cultivars compared with two Solanum tuberosum cultivars. The equivalent umami concentration was calculated for five cultivars, and there were strong positive correlations with flavor attributes and acceptability scores from a trained evaluation panel, suggesting that umami is an important component of potato flavor.  相似文献

Total and individual fatty acid contents were determined in raw tubers of four genotypes from each of the two species Solanum phureja and S. tuberosum. The four S. phureja genotypes contained statistically significantly higher concentrations of total fatty acids, with their average value being 37% greater than that for the mean of the four S. tuberosum cultivars. In both species, a total of 17 fatty acids were detected in quantifiable amounts, and in all genotypes the predominant fatty acid was linoleic followed by alpha-linolenic and palmitic acids. Unusually, 15-methyl hexadecanoate was present as a minor acid in both species. Although a number of statistically significant differences in the fatty acid percentage compositions were found between and within the two species, these were generally small. Averaged over all species and genotypes, tuber storage resulted in an initial small but statistically significant increase in total fatty acid content, but prolonged storage resulted in a fall to the initial values detected close to harvest. The same trend was evident for S. phureja alone (for mean values of all genotypes), but for S. tuberosum the total fatty acid content remained constant over the whole storage period. For both species, the contents (both as absolute levels and as percent compositions) of linoleic acid decreased and alpha-linolenic acid increased in tubers over the whole storage period, and possible mechanisms are discussed. Also, the absolute levels of these two acids were greater in S. phureja than in S. tuberosum, and this is discussed in relation to the development of flavor-related compounds during cooking. The polar lipids of one representative of S. tuberosum and of S. phureja were qualitatively similar. There were only minor differences in the polar lipid percentage compositions and in the corresponding fatty acid compositions of the individual polar lipids between the two species, although the absolute levels of the total, and of some individual, polar lipids were higher in S. phureja.  相似文献

Diverse procedures have been reported for the separation and analysis by HPLC of the two major glycoalkaloids present in potatoes, alpha-chaconine and alpha-solanine. To further improve the usefulness of the HPLC method, studies were carried out on the influence of several salient parameters on the analysis of the two potato glycoalkaloids. Effects on retention (elution, separation) times of the (a) composition and pH of the mobile phase (acetonitrile and phosphate buffer), (b) concentration of the phosphate buffer, (c) capacity values of column packing of four commercial HPLC amino columns, (d) column temperature were studied. Except for pH, all of the variables significantly influenced the retention times. The results make it possible to select analysis conditions that produce well-separated as well as symmetrical peaks of the two glycoalkaloids. This improved HPLC method (limit of detection of approximately 150 ng) was evaluated with extracts from the cortex of one whole potato variety (May Queen) grown in Japan and the freeze-dried peel and flesh from the following eight cultivars grown in the United States: Atlantic, Dark Red Norland, Ranger Russet, Red Lasoda, Russet Burbank, Russet Norkota, Shepody, and Snowden. In addition, the same samples were analyzed by GC-MS for the presence of two water-soluble nortropane alkaloids, calystegine A(3) and calystegine B(2), reported to be potent glycosidase inhibitors. The following ranges for the eight varieties of total glycoalkaloid and calystegine levels were observed: dry flesh, 5-592 and 6-316 mg/kg; dry peel, 84-2226 and 218-2581 mg/kg; dry whole potatoes, 40-883 and 34-326 mg/kg; wet flesh, 1-148 and 1-68 mg/kg; wet peel, 12-429 and 35-467 mg/kg; wet whole potatoes, 7-187 and 5-68 mg/kg. The possible significance of the results to plant and food sciences is discussed.  相似文献

Potatoes (Solanum tuberosum L.) supplemented with increasing amounts of sodium selenate were analyzed for glycoalkaloid (GA) content. GAs were extracted with 5% acetic acid from freeze-dried tubers of two potato cultivars, Satu and Sini, harvested 10 weeks after planting as immature. The GAs alpha-solanine and alpha-chaconine were quantified by reverse-phase high-performance liquid chromatography (RP-HPLC) with diode array detection. Two independent experiments were performed. In the first experiment, the total GA concentration +/- standard error of the tubers ranged between 105 +/- 9 and 124 +/- 10 mg kg(-1) fresh weight in Satu and between 194 +/- 26 and 228 +/- 10 mg kg(-1) fresh weight in Sini. The ratio of alpha-solanine to alpha-chaconine was 0.2 in Satu and 0.5-0.6 in Sini. In the second experiment, the total GA concentration +/- standard error was 75 +/- 4 to 96 +/- 11 mg kg(-1) fresh weight, and the ratio of alpha-solanine to alpha-chaconine was 0.3-0.4 in Satu. A high sodium selenate supplementation (0.9 mg of Se kg(-1) quartz sand) slightly decreased the GA content in Satu, but this decrease was not statistically significant. Furthermore, at this addition level the Se concentration increased to a very high level of 20 microg g(-1) dry weight, which cannot be recommended for human consumption. In both experiments, the Se concentration in tubers increased with increasing sodium selenate application levels. Our results show that acceptable application levels of selenate did not have an effect on the GA concentration in immature potato tubers.  相似文献

Steroidal glycoalkaloids occur in potatoes and are reported to impart resistance to phytopathogens including bacteria, fungi, and insects. Because glycoalkaloids can be passed to progenies during breeding programs designed to develop improved potatoes, it is of importance to determine the quality of desired characteristics and the composition of glycoalkaloids of new somatic hybrids. The objective of this study was to determine the appearance, size, and shape (morphological characters) as well as the glycoalkaloid content of potato tubers of somatic hybrids between tetraploid Solanum tuberosum cv. Dejima (2n = 4x = 48 chromosomes) and the dihaploid clone ATDH-1 (2n = 2x = 24 chromosomes) induced by anther culture from Solanum acuale-T (acl-T, 2n = 4x = 48 chromosomes). Tuber size and shape in somatic hybrids were in accord with those of cv. Dejima, whereas the tuber skin color resembled that of ATDH-1. Thin-layer chromatography, high-performance liquid chromatography, and gas-liquid chromatography/mass spectrometry studies showed that the two steroidal glycoalkaloids (alpha-chaconine and alpha-solanine) were present in the tubers of S. tuberosum, whereas acl-T and ATDH-1 tubers were found to contain alpha-tomatine and demissine. The concentrations of total glycoalkaloids in both acl-T and ATDH-1 was >100 mg/100 g of fresh weight tuber cortex, much higher than in S. tuberosum. All somatic hybrids, except one clone, contained four glycoalkaloids (alpha-chaconine, alpha-solanine, alpha-tomatine, and demissine) derived from the fusion parents. The lack of alpha-tomatine in the remaining clone may be due to somaclonal variation. The results show that character expression is influenced by ploidy level and that total glycoalkaloid levels in most somatic hybrids were intermediate between those of the fusion parents. The possible significance of these findings for plant breeding and food safety is discussed.  相似文献

Abstract

Earlier harvest of potatoes (Solanum tuberosum L.) can be achieved by different treatments of the seed tubers. This can be important, for example, in areas with a short growing season and in organic farming. In a pot experiment, different pre-sprouting methods were evaluated for four cultivars. The experiment consisted of (a) untreated seed tubers stored at 4 °C until planting, (b) traditional pre-sprouted tubers (P) and (c) pre-sprouted tubers with stimulation of adventitious root formation (PR). Pre-sprouted seed tubers have earlier and faster tuber formation, but often lower yield than non pre-sprouted tubers if there is no restriction of the growing season. The PR method developed here, where tubers were sprayed with water to stimulate adventitious root formation and give faster development and tuber initiation than conventional pre-sprouting, can be of value when the growing season is restricted or if early harvest is important. Initial emergence of the pre-sprouted potatoes was monitored in two pot experiments. Emergence was faster for the pre-sprouted treatments compared with the control, but there were no significant differences between the two pre-sprouting methods. At harvest, the pre-sprouted treatments gave higher yield than the control for all growing periods, methods and cultivars, although the differences were not significant for every combination. The cultivars Matilda, Cicero, Ovatio and Superb seemed to respond more positively in terms of yield to PR treatments than cv. Ditta. The new method for stimulating adventitious root formation developed here will substantially help growers to achieve earlier harvests.  相似文献

A PVYO virus-resistant potato (Solanum tuberosum L. cv. Golden Valley) was identified, and further, from its tubers, a small (5.57 kDa) antiviral peptide potide-G was isolated. Application of potide-G on virus susceptible potato (cv. Winter valley) expressed robust resistance to PVYO infection and showed no virus infected morphology. We found that PVYO infection spreads up completely within 3 days post inoculation (dpi) in susceptible cultivar. PVYO was more accumulated toward the basal leaves, when infection occurred longer. Combined results of morphology of PVYO infection, ELISA, RT-PCR, and real-time PCR showed the resistance to the PVYO infection depends on the expression of Ry gene. Indeed, the real-time PCR result showed that the Ry gene up-regulated to 3 times higher in PVYO infected cv. Golden valley. Golden crude protein was found to be active against PVYO infection in the in vivo test. In addition, application of potide-G in a virus susceptible potato potently reduced the viral infection actively with 50 times lower concentration than that of the Golden protein. Further identification of a host-specific resistant gene in a plant and the peptide derived from it offers new opportunities for the development of novel bio-pesticides against plant virus.  相似文献

Carotenoids in white- and red-fleshed loquat fruits   总被引:2,自引:0,他引:2  
Fruits of 23 loquat ( Eriobotrya japonica Lindl.) cultivars, of which 11 were white-fleshed and 12 red-fleshed, were analyzed for color, carotenoid content, and vitamin A values. Color differences between two loquat groups were observed in the peel as well as in the flesh. beta-Carotene and lutein were the major carotenoids in the peel, which accounted for about 60% of the total colored carotenoids in both red- and white-fleshed cultivars. beta-Cryptoxanthin and, in some red-fleshed cultivars, beta-carotene were the most abundant carotenoids in the flesh, and in total, they accounted for over half of the colored carotenoids. Neoxanthin, violaxanthin, luteoxanthin, 9- cis-violaxanthin, phytoene, phytofluene, and zeta-carotene were also identified, while zeaxanthin, alpha-carotene, and lycopene were undetectable. Xanthophylls were highly esterified. On average, 1.3- and 10.8-fold higher levels of colored carotenoids were observed in the peel and flesh tissue of red-fleshed cultivars, respectively. The percentage of beta-carotene among colored carotenoids was higher in both the peel and the flesh of red-fleshed cultivars. Correlations between the levels of total colored carotenoids and the color indices were analyzed. The a* and the ratio of a*/ b* were positively correlated with the total content of colored carotenoids, while L*, b*, and H degrees correlated negatively. Vitamin A values, as retinol equivalents (RE), of loquat flesh were 0.49 and 8.77 microg/g DW (8.46 and 136.41 microg/100 g FW) on average for white- and red-fleshed cultivars, respectively. The RE values for the red-fleshed fruits were higher than fruits such as mango, red watermelon, papaya, and orange as reported in the literature, suggesting that loquat is an excellent source of provitamin A.  相似文献

Anthocyanins gain more and more importance as natural colourants. Besides many other plant species, coloured potatoes could serve as a potential source. Therefore, 27 potato cultivars and four breeding clones of different origin were analysed for their anthocyanin contents in fresh weight (fw). On average the highest amounts of anthocyanins were found in the skin (0.65 g kg−1 fw) of potatoes. The corresponding values of samples taken from whole tubers (0.31 kg−1 fw) and flesh (0.22 kg−1 fw) were significantly lower (p<0.03). The analysis revealed considerable differences in the amounts of anthocyanins between the 31 cultivars/breeding clones. Among them ȁ8Peru Purpleȁ9 revealed the highest anthocyanin content in the skin with 2.96 g kg−1 fw. A similar high value was reached by ȁ8Violettfleischigeȁ9 and clone 1.81.202–92 N. Also for the other two types of samples, flesh and whole tubers, these three genotypes exhibited the highest level of anthocyanins. Interestingly, different rates of nitrogen fertilization at 100 and 200 kg ha−1 had no significant effect on the pigment content of potatoes. The same was observed with respect to the year or the location of plant growth. There were also no significant changes in the anthocyanin contents of tubers during storage for 135 days. In dry matter, starch and protein contents the coloured potato cultivars/breeding clones were comparable with traditional cultivars. Further tests have shown that the glycoalkaloids were mainly localized in the skin of coloured potatoes.  相似文献

酸性转化酶是植物体内降解蔗糖为还原糖(葡萄糖、果糖)的关键酶,而还原糖在马铃薯(Solanum tuberosum L.)低温贮藏中的快速积累是影响油炸加工品质的重要因素。为了实现对马铃薯块茎低温糖化的品质改良,本研究利用所克隆的酸性转化酶基因构建了RNA干涉载体,并转化马铃薯品种N2。经PCR、Northern鉴定,酸性转化酶基因的干涉片段已经成功导入马铃薯植株中。对干涉转基因株系的试管苗和试管薯的酸性转化酶活性进行测定,结果显示,植株的酸性转化酶的活性平均下降69.8%(Ni-1除外),最大降幅为78%(Ni-4),而试管薯的酶活性最大降幅为68%。与反义RNA的表现较好的转基因株系进行比较,RNA干涉对酶活的调节作用与之相当。研究结果表明,RNA干涉对马铃薯内源酸性转化酶活性的调节作用显著,这种转录表达的调控技术对马铃薯低温糖化改良具有良好的应用前景。  相似文献

The total ascorbate content of potato tubers from 33 Solanum tuberosum genotypes grown at three geographically diverse sites in Europe in each of two years was determined immediately postharvest and after approximately 4 months of storage at 4 degrees C. Statistically significant differences in total ascorbate concentration were observed between genotypes both at harvest and after storage. In all genotypes, the levels of ascorbate decreased during storage. These results are discussed in terms of their implications for diet and health as well as in terms of breeding for improved vitamin C content in potatoes.  相似文献

The influence of 2 weeks and 3 months of dark storage upon light-induced chlorogenic acid accumulation within tuber tissue of four potato cultivars and upon 5-, 4-, and 3-caffeoylquinic acid concentrations within cv. King Edward was determined. Storage period significantly affected (P < 0.05) the magnitude of the light-induced chlorogenic acid response with accumulation rates 3-4 times higher in tubers exposed to light after 2 weeks compared with those placed under light after 3 months. Comparison of chlorogenic acid concentrations in controls after 2 and 3 months of dark storage indicated that tuber chlorogenic concentrations decline during prolonged cold store at 5 degrees C. Rates of accumulation in response to light were cultivar-dependent with cv. Fianna the most light-sensitive and cv. Maris Piper relatively light-insensitive. In virtually all cases exposure to sodium and fluorescent light promoted higher rates of accumulation than did exposure to high-pressure mercury light sources. Chlorogenic acid values steadily increased over 15 days of illumination with, in the majority of cases, no indication of cessation. Light exposure increased 5-, 4-, and 3-caffeoylquinic acid accumulation rates in cv. King Edward. Irrespective of storage period and light source, ratios of 5-:4-:3-caffeoylquinic acid were ca. 85:15:0 at day 0 and 52:42:6 by day 15.  相似文献

Peroxidases (POD; EC 1.11.1.7) can cross-link cell wall polymers and may have an impact on the final textural quality of potato tubers. Because heat treatments are important during processing, the thermal properties of isoPODs from soluble and ionically and covalently bound fractions were studied from both potato tubers and sprouts. For both tissues, the ionically bound fraction was the most thermostable; approximately 20% of POD activity remained after a heat treatment of 10 min at 90 degrees C (for sprouts). The temperature profile of the ionically bound sprout fraction appeared to be nonlinear and suggested the presence of a very thermostable POD, which still showed activity after a heat treatment at 100 degrees C. Visualization by using isoelectric focusing confirmed the occurrence of a thermostable isoPOD with an IEP of 9.5, which displayed regeneration of activity after heat inactivation. This cationic POD was further purified by chromatography techniques, and by SDS-PAGE its molecular mass was estimated at 38 kDa.  相似文献

The phenolic compounds of 25 peach, nectarine, and plum cultivars were studied and quantified by HPLC-DAD-ESIMS. Hydroxycinnamates, procyanidins, flavonols, and anthocyanins were detected and quantified. White and yellow flesh nectarines and peaches, and yellow and red plums, were analyzed at two different maturity stages with consideration of both peel and flesh tissues. HPLC-MS analyses allowed the identification of procyanidin dimers of the B- and A-types, as well as the presence of procyanidin trimers in plums. As a general rule, the peel tissues contained higher amounts of phenolics, and anthocyanins and flavonols were almost exclusively located in this tissue. No clear differences in the phenolic content of nectarines and peaches were detected or between white flesh and yellow flesh cultivars. There was no clear trend in phenolic content with ripening of the different cultivars. Some cultivars, however, had a very high phenolic content. For example, the white flesh nectarine cultivar Brite Pearl (350-460 mg/kg hydroxycinnamates and 430-550 mg/kg procyanidins in flesh) and the yellow flesh cv. Red Jim (180-190 mg/kg hydroxycinnamates and 210-330 mg/kg procyanidins in flesh), contained 10 times more phenolics than cultivars such as Fire Pearl (38-50 mg/kg hydroxycinnamates and 23-30 mg/kg procyanidins in flesh). Among white flesh peaches, cultivars Snow King (300-320 mg/kg hydroxycinnamates and 660-695 mg/kg procyanidins in flesh) and Snow Giant (125-130 mg/kg hydroxycinnamates and 520-540 mg/kg procyanidins in flesh) showed the highest content. The plum cultivars Black Beaut and Angeleno were especially rich in phenolics.  相似文献

Effects of Se fertilization on potato processing quality, possible changes in Se concentration and form in tubers during storage, and retransfer of Se from seed tubers were examined. Potato plants were grown at five selenate (SeO4(2-)) concentrations. Tubers were harvested 16 weeks after planting and were stored at 3-4 degrees C prior to analysis. The results showed that the Se concentration did not decrease during storage for 1-12 months. In tubers, 49-65% of total Se was allocated in protein fraction, which is less than found in plant leaves in a previous study. The next-generation tubers produced by the Se-enriched seed tubers had increased Se concentrations, which evidenced the relocation of Se from the seed tubers. At low levels, Se improved the processing quality of potato tubers by diminishing and retarding their raw darkening. The value of Se-enriched potato tubers as a Se source in the human diet was discussed.  相似文献

There are concerns of potential food chain transfer of metals in crops grown on lead–arsenate-contaminated soils. The objective of this study was to investigate lead and arsenic uptake by four potato (Solanum tuberosum L.) cultivars grown on lead–arsenate-contaminated soils with lead and arsenic concentrations ranging from 350 to 961 and 43 to 290 mg kg?1, respectively. Yield was not reduced due to treatment. Potato tubers were washed thoroughly before peeling. Lead concentration in both peeled tubers and peel was below instrument detection limit. Arsenic concentration in peeled tubers grown on the lead–arsenate soils ranged from 0.24 to 1.44 mg kg?1. Arsenic concentration was 60% higher in the peel than in the peeled tuber. The relatively high arsenic levels in the peel demonstrated that arsenic was taken up into the potato peel tissue. It is recommended that if potatoes are grown on these soils they should be peeled before consumption.

The antioxidant activities of eight apple cultivars were studied by using the ferric reducing/antioxidant power (FRAP), the beta-carotene-linoleic acid model system (beta-CLAMS), and the photochemiluminescent (PCL) assays. The antioxidant activity of apples is highly correlated to the total phenolic content (TPC) measured by the Folin-Ciocalteu method and the total polyphenolic index (TPI) obtained by HPLC. Extracts of the peel and flesh were analyzed and assayed separately. The FRAP activities of both peel and flesh extracts correlate well with the TPC (r = 0.95 and 0.99, respectively) and the TPI (r = 0.82 and 0.99, respectively). Similar results were found in the beta-CLAMS activities, showing correlation coefficients of r = 0.90 and 0.91 with the TPC for the peel and flesh extracts and of r = 0.90 and 0.84 with the TPI for the peel and flesh extracts, respectively. The antioxidant activity measured by the PCL assay was not correlated with TPC or TPI due to the lack of integratable lag phase in this method with the flavan-3-ols/procyanidins. Among the five major polyphenolic groups, flavan-3-ols/procyanidins had the highest positive correlation with the FRAP and beta-CLAMS activities: r = 0.84 and 0.88 for the peel extracts, respectively; and r = 0.98 and 0.87 for the flesh extracts, respectively. At individual compound level, epicatechin and procyanidin B2 were the major contributors to the antioxidant activity of apple. Hydroxycinnamic acids may have a significant role in the flesh.  相似文献

The antioxidant potential of eight clingstone peach cultivars was investigated by determining phenolic compounds and inhibition of low-density lipoprotein (LDL) oxidation. Cultivars low in polyphenol oxidase (PPO) were also selected to minimize enzymatic browning. Inhibition of LDL oxidation varied from 17.0 to 37.1% in peach flesh extract, from 15.2 to 49.8% in whole peach extract, and from 18.2 to 48.1% in peel extract. Total phenols were 432.8-768.1 mg/kg in flesh extract, 483.3-803.0 mg/kg in whole extract, and 910.9-1922.9 mg/kg in peel extract. The correlation coefficient between relative LDL antioxidant activity and concentration of total phenols was 0.76. Peel PPO activity was higher than flesh activity in most cultivars. The lowest PPO and specific activities were found in the Walgant cultivar, followed by Kakamas and 18-8-23. These three cultivars combine the desirable characteristics of strong antioxidant activity, low PPO activity, and lower susceptibility to browning reactions.  相似文献
