In the first-litter sows lower serum levels were found for all three Ig classes as compared to multiparous sows. The same was true for IgA in lacteal secretions and in piglet serum during the first days of life, while no differences were found for IgG levels. In contrast to these findings, IgM levels were found to be higher in lacteal secretions of first-litter sows and in piglet serum during the first days of life as compared to their counterparts. From three weeks after birth Igs found in piglet serum mainly originate from de novo synthesis. In this period piglets of first litter sows showed a higher IgA level up to the 6th week of life and higher IgG and IgM levels up to the end of the investigation period. Results are discussed in terms of maternal-neonatal immune regulation, focussing on the apparent suppressive role of maternally-derived IgG on total de novo Ig synthesis by suckling piglets.  相似文献

The concentrations of IgG, IgM and IgA in sera collected from 3855 sows (3208 pregnant and 647 lactating) at a single time point were determined. This experimental design allowed changes in serum immunoglobulin over the reproductive cycle to be studied without bias from seasonal influence. The concentrations of the three immunoglobulins changed independently during the reproductive cycle. Serum levels of IgM and IgG began a progressive postpartal decline during the 14th–17th week of gestation. At the onset of lactation serum IgG levels progressively increased while IgM levels continued to decline, the latter reaching their lowest level during the third week of lactation. In contrast to IgM and IgG, serum IgA levels increased 35% during weeks 14–17 of gestation and continued to increase throughout lactation, reaching their highest serum levels in the third week of lactation; the serum IgA concentration at this time was twice that observed during the first 13 weeks of gestation. Results of these studies allowed the reproductive cycle to be classified into four phases on the basis of serum immunoglobulin concentrations: (1) weeks 1–4 of gestation; (2) weeks 5–13 of gestation; (3) weeks 14–17 of gestation and (4) lactation.  相似文献

The aim of the study was the determination of IgA, IgM and IgG concentrations in porcine serum and colostrum, in order to evaluate their variations in the perinatal period, as well as to clarify whether there is a correlation between colostrum intake, initial level of immunoglobulins (Ig) in piglet serum and development of their own immunity. The mean IgA, IgM and IgG concentrations in sow serum 10 days before parturition were 1.58, 6.12 and 39.56 mg/ml, respectively. Seven days later only the IgG level was insignificantly lower (34.94 mg/ml, p = 0.55), while concentrations of IgA and IgM increased to 2.25 and 7.25 mg/ml, respectively (p = 0.23 and 0.62, respectively). The mean initial IgG concentration in colostrum at farrowing was 118.5 mg/ml and differed between sows. The average value of IgA in colostrum at birth was 23.8 mg/ml and decreased to 7.85 mg/ml at 6 hours (h) and to 4.59 mg/ml at 24 h after the onset of farrowing. IgM concentration at birth was 12.1 mg/ml and decreased to 4.23 mg/ml at 24 h postpartum. Positive relationships were found between concentrations of IgM and IgA in serum of piglets at 14 and 56 days of life (r = 0.41 and 0.80, respectively, p < or = 0.05) as well as for IgG concentration in the piglets serum at 7 days and 56 days of age (r = 0.48, p < or = 0.05). The above observations suggest that there is a correlation between the level of Ig in piglet serum in the first days of life and improvement of their own immunity.  相似文献

Colostrum from sows and gilts inoculated with virulent transmissible gastroenteritis virus was fractionated into the 3 major immunoglobulin classes, IgA, IgG, and IgM-IgA fractions, by chromatographic and gel-filtration procedures. Each fraction was assayed for purity with rabbit anti-porcine serum and rabbit monospecific anti-porcine IgG, anti-porcine IgA, and anti-porcine IgM. These analyses showed that the IgG and IgA fractions were pure. The IgM fraction contained some IgA in the polymeric form and was designated the IgM-IgA fraction. Each Ig was assayed for virus-neutralizing activity on swine testes cells by the plaque-reduction method before and after conjugation with fluorescein isothiocyanate. On the basis of activity per milligram of protein, the virus-neutralizing titers were 1:641, 1:44, and 1:6.8 for the IgA, IgG, and IgM-IgA fractions respectively; the fluorescent antibody titers were 1:31.3, 1:0.1, and 1:15.6, respectively, for the same Ig.  相似文献

In a newly established closed specific pathogen-free (SPF) swine herd, gilt/sow suckling and weaned pig rotavirus specific antibody titers were followed for three lactations by enzyme-linked immunosorbent assay (ELISA) to gain insight into the dynamics of herd antibody titers to group A rotavirus. Among gilts/sows, serum antirotavirus IgG titers increased during each lactation with a subsequent drop in titer between farrowings. Serum antirotavirus IgM titers declined during each lactation and with subsequent parity. Serum antirotavirus IgA titers remained constant during lactations and among parities. In colostrum and milk, antirotavirus IgA antibody was abundant. Differences in titer were not noticed between gilts and second litter sows but third litter sows had significantly higher titers than the first two groups. Antirotavirus IgG was high in colostrum but nearly nonexistent in milk. This titer did not vary significantly within or among parities. There was a linear regression in the titers of baby pig serum antirotavirus IgG from the post colostral sample through to seven weeks old, after which titer began to increase. No difference in baby pig serum antirotavirus IgG was noted among the three litters. Serum antirotavirus IgA and IgM were undetectable in baby pig sera after 2-3 weeks of age. Coproantibody to rotavirus was sporadically present in pig feces for 2-3 weeks after birth with highest titers in the IgA fraction. We conclude that although it is probable that age resistance of pigs to rotavirus diarrhea occurs, humoral immunity as measured by ELISA rotavirus antibody titers may not be intimately involved in virus clearance since in our studies baby pigs passively received large amounts of antibody but still excreted pathogenic virus. The finding of increasing levels of serum antirotavirus IgG in gilt/sow serum suggest that exposure to antigen of dams occur without significant increases in antirotavirus IgG titers in either colostrum, milk, or baby pig serum.  相似文献

以MM-3基因工程活菌苗口服免疫妊娠73d、83d、103d母猪各4头,口服及肌注免疫妊娠92d母猪各5头,用间接BA-ELISA法检测血清及乳清中IgG、IgA和IgM型抗K_(88ac)及LTB抗体的动态变化.结果表明,无论口服还是肌注免疫母猪,抗K_(88ac)及抗LTB抗体均极显著增高.肌注以IgG增加为主,口服以IgA增加为主.肌注免疫IgG型抗K_(88ac)抗体滴度极显著地高于口服免疫(P<0.01);口服免疫IgA型抗LTB抗体滴度显著高于肌注免疫(P<0.05).口服及肌注免疫乳清中抗K_(88ac)及抗LTB抗体滴度很高,达1:3200~204800,但在1周内即降低,约减少一半(约3~5个滴度),2~3周内各型抗体下降缓慢,维持在对照组周内水平,4周后达最低水平.妊娠不同时期口服免疫,其血清中抗K_(88ac)抗体的动态变化相似,但乳汁中抗体则差异非常明显,妊娠83d和92d免疫组IgG、IgA和IgM均显著增加,且高于103d组(P<0.01)和73d组(P<0.05).口服免疫的最佳时间为分娩前26~30d,肌注免疫最佳免疫时间为分娩前21d.  相似文献

试验选用相近胎次(4-6胎)和分娩日龄(相差3d左右)的长×大二元母猪20头,随机分成4个处理组,各处理组哺乳仔猪均从5日龄开始补料,哺乳仔猪日粮中分别添加0、50、100和200 mg/kg的酵母-β1,3/1,6-葡聚糖,研究酵母β-1,3/1,6-葡聚糖对哺乳仔猪生长性能和免疫机能的影响。结果表明:哺乳仔猪日粮中添加酵母β-1,3/1,6-葡聚糖,对生产性能无显著影响,有降低仔猪腹泻率和死亡率的趋势;可显著提高外周血淋巴细胞转化率,血清IgA、IgG和IgM浓度(P<0.05),有提高血清溶菌酶活性的趋势。综合全期生长性能和免疫调节效果来看,哺乳仔猪日粮中β-1,3/1,6-葡聚糖适宜的添加剂量为50mg/kg。  相似文献

Background: Greyhound dogs have significant physiologic, hematologic, and biochemical differences when compared with other breeds, including significantly lower serum globulin concentration owing to decreases in the α‐ and β‐globulin fractions. The specific proteins that account for differences in globulin concentrations are not known, but IgA and IgM, both β‐globulins, are potential candidates. Objectives: The aims of this study were to measure serum IgG, IgA, and IgM in clinically healthy retired racing Greyhounds and compare the results with those of age‐ and sex‐matched non‐Greyhound dogs. Methods: Study animals included 25 Greyhound and 20 non‐Greyhound dogs. Total protein, albumin, and total globulin concentrations were determined. IgG, IgA, and IgM concentrations were measured using a commercially available radial immunodiffusion kit. The Student t‐test assuming equal variances was used to compare concentrations of immunoglobulins between groups. Results: Serum concentrations of IgA and IgM in Greyhounds (IgA=49±20 mg/dL; IgM=132±47 mg/dL) were significantly lower than concentrations in non‐Greyound dogs (IgA=70±39 mg/dL; Ig M=212±78 mg/dL). Concentrations of IgG did not differ between groups. Conclusions: Mean serum IgA and IgM concentrations in Greyhounds were lower than those in non‐Greyhound dogs. This may contribute to low serum concentrations of β‐globulins in Greyhounds. Specific reference intervals are recommended for Greyhounds to avoid possible misdiagnosis of IgA or IgM deficiency.  相似文献

Responses to prepartum injection of sows with Se and vitamin E (E) were evaluated by determining immunoglobulin (IgA, IgM, IgG) levels in the colostrum and serum of the sows and the serum of their offspring. Fifty-four sows (40 multiparous, 14 primiparous) receiving diets adequate in E and Se according to current NRC (1988) standards were randomly allotted to four treatment groups in which a single i.m. injection of saline (controls), 5 mg of Se, 1,000 IU of E, or both Se and E were given on d 100 of gestation. Sows were bled prior to and 7 d after injection, at farrowing and on d 14 and 28 of lactation. Colostral samples were collected at the initiation of farrowing. Pigs were bled 20 h postpartum and at 14 and 28 d of age. Major immunoglobulin changes in the serum of the sows due to treatment were not seen prior to parturition. Injections of Se and(or) E resulted in higher colostral IgM levels (8.4, 10.7, 9.8 and 9.6 mg/ml, respectively), but only the response from Se was significant (P less than .05). Concentrations of colostral IgA or IgG were not affected by treatment (P greater than .30). Compared with controls, all three treatments increased (P less than .10) IgM concentrations in serum from pigs at birth (28.3, 33.3, 36.0 and 33.5 mg/ml, respectively), whereas IgA and IgG concentrations were not affected (P greater than .30).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Immunoglobulin (Ig) concentrations in serum and in nasal secretions were correlated with pneumonia and diarrhea during the first 12 weeks of life in 56 calves. The peak onset of pneumonia occurred between 2 and 4 weeks of age when the calves' serum IgG1, IgG2, and IgA concentrations were lowest. As IgG2 concentrations increased, fewer calves developed pneumonia. Peak onset of pneumonia was also correlated with the lowest IgG and IgA concentrations in the calves' nasal secretions. Most calves developed pneumonia when serum concentrations of IgG1 were less than 1.5 g/dl, IgG2 less than 0.3 g/dl, IgA less than 0.1 g/dl, and IgM less than 0.2 g/dl and when the combined IgG and IgA values in nasal secretions were less than 0.2 mg of Ig/mg of protein. In study A, diarrhea preceded pneumonia in 63% of 56 calves. In study B, 38% of 23 calves had diarrhea and/or hemorrhagic feces before pneumonia. Seemingly, there was a relationship between diarrhea and pneumonia. Furthermore, pneumonia occurred at or just after the time when IgG1, IgG2, and IgA concentrations in serum and the combined IgG and IgA concentrations in nasal secretions were lowest. Pneumonia is a common disease of calves between 1 and 5 months of age, a period coinciding with the usual low point in serum immunoglobulin (Ig) concentrations due to catabolism of passively acquired antibodies. Studies were made of the immunoglobulin (Ig) in serums from umbilical cord of newborn pigs and maternal placenta. The neutralization test for porcine parvovirus and Japanese encephalitis virus was carried out with the serum of the sow and that of the umbilical cord of the newborn pig. Comparative studies of the serums from the dam and the umbilical cord were also done with gel filtration. Of 20 umbilical cord serum samples, IgG was seen in 5 samples (25%), IgA in 1 sample (5%), and IgM in 9 samples (45%). The amount of any 1 of the 3 classes of Ig in the serums was between 13.5 and 28.0 mg/dl. Among the samples examined, 1 had both IgG and IgA and 1 had IgG and IgM, but none had both IgA and IgM and none had 3 classes of immunoglobulins (i.e., IgG, IgA, and IgM). Only 7 samples (35%) did not have any class of Ig. The IgG disappeared from the blood of hysterectomy-produced colostrum-deprived pigs at 3 days of age, and IgM disappeared when pigs were 7 days of age. Neutralization antibodies of porcine parvovirus and Japanese encephalitis virus in maternal serum were not transferred to the fetus through the placenta. Results of immunohistologic surveys indicated that the sow's Ig were not transferred to the fetus through the placenta. Therefore, it is believed that the Ig in the porcine fetus might be synthesized in certain cells in the placental tissue, and the degree of production of the Ig in the placental tissues may differ in each case. The component, which seems to be Ig, was observed as the obscure band of the beta- to gamma-globulin area in serum of the umbilical cord. Comparison was made, with gel filtration, of maternal serum and serum from the umbilical cord of the newborn pig originating from the same sow. Seemingly, the IgG in the umbilical cord serum is mainly in the lower molecular weight fraction, whereas IgG in the sow's serum was distributed in the high to low molecular weight fractions.  相似文献

The composition of sow colostrum and milk was quantitated in 25 sows at 14 time points throughout lactation. All animals belonged to the same experimental herd of German Landrace, farrowed within 4 d, and were of various lactation numbers and various litter sizes. In the first 6 h of lactation colostrum total solids (TS) and protein contents were higher, while fat and lactose contents were lower than in mature milk. Decreased total protein and whey protein contents and concomitantly increased fat and lactose content, with nearly unchanged TS levels, indicate transition from colostrum to mature milk. The high protein content of colostrum was largely due to immunoglobulin (Ig). During the first 6 h, IgG accounts for nearly all the protein in colostrum but plays a decreasing role in sow milk as lactation proceeds. After 2 wk, IgA levels begin to increase and at the end of lactation, IgA constitutes 40% of the total whey protein. No influences of lactation number and litter size on milk composition could be ascertained in this study.  相似文献

Rotavirus antibodies were demonstrated in lacteal secretions and sera of 20 parturient sows and in sera of their newborn by an enzyme-linked immunosorbent assay blocking technique, using bovine rotavirus cell culture antigen and monospecific antibody to bovine rotavirus. Antibodies to rotavirus occur in the 3 immunoglobulin (Ig) classes IgM, IgA, and IgG in lacteal secretions. High and long-persisting antibody activity was mainly associated with the IgA class. The IgM and IgG decreased to undetectable concentrations in most sows during the 14-day investigation period. Serum antibodies of newborn pigs nursing their dams also decreased rapidly during this time. The heterologous enzyme-linked immunosorbent assay blocking technique was a reliable and rapid procedure for the demonstration of rotavirus antibodies.  相似文献

The present results suggest that mannanoligosaccharides (MOS) included in a sow nutrition may affect its immune system and humoral antibody production in colostrum and milk, and thus increase the piglet immunity at the postnatal period. The studies involved sows of the Polish Landrace breed mated with boars (Hampshire x Duroc). In each experiment, the sows were assigned to two groups: control and experimental (MOS). Each group consisted of 16 sows managed in pens (2 animals in each) during pregnancy, whereas at farrowing and lactation period they were placed in individual pens. The basal diet during pregnancy (PR-S) and lactation (LC-S) period contained wheat (40% in experiment I--groups 1 and 2) or triticale (40% in experiment II--groups 3 and 4), as well as barley, soybean meal, soybean oil and mineral-vitamin premix. Throughout both experiments, the sows from the experimental group had a dietary supplement of mannanoligosaccharides (MOS) preparation for 4 of weeks prepartum and 4 weeks of post partum period. A level of the MOS supplementation (8 g of MOS per sow daily) based on the recommendations of the manufacturer. Blood samples were collected from the sows on days 84 (the start of trial) and 110 of pregnancy, after farrowing, and on day 21 of lactation period, while from the piglets at birth and on day 21 of age. Colostrum was collected between 1-3, 12, 24 and 48 h after farrowing. The blood samples taken from sows and piglets as well as the samples of sow colostrum and milk were evaluated for the presence of IgG, IgA and IgM antibodies. The present study has provided considerable evidence that MOS supplementation of sows feedstuff before and after farrowing (4 weeks before and 4 weeks after) exerts a positive effect on IgG content in the colostrum and plasma of sows and following this on serum IgG level in the suckling piglets. Higher level of colostral (passive) immunity influences positively body weight gain and survival rate of the piglets at weaning.  相似文献

A total of 124 Duroc and 99 Landrace primiparous and multiparous sows were assigned, within breed and contemporary group, to control (N) or 10% added fat (F) diets on d 105 of gestation based on parity and genetic line (control or selected for improved sow productivity), to determine the effects of genetic line and fat addition to the lactation diet on sow and litter performance. Weekly feed intake was not affected (P greater than .10) by genetic line for Duroc and Landrace sows but feed intake was reduced (P = .08) during wk 1 to 4 for Duroc sows and during wk 1 and 4 for Landrace sows (P less than .05) when they were fed diet F compared with diet N. Select (S)-line Duroc and Landrace sows lost more weight during lactation (P less than .01) than did control (C)-line sows. Select-line Landrace sows lost more backfat during lactation (P less than .05) than did C-line sows. Landrace sows lost less weight during lactation (P less than .05) when fed diet F than when fed diet N. The total number of pigs born, born alive, and alive at 21 d and at weaning were higher (P less than .01) for S-line Duroc sows, and litter size at 21 d and at weaning was higher (P less than .01) for S-line Landrace sows than for C-line litters within each breed. 本试验旨在研究母猪饲粮添加β-羟基-β-甲基丁酸(HMB)对母猪繁殖性能及仔猪生长性能和免疫功能的影响。选取20头3~6胎次、妊娠74 d的"长×大"母猪,随机分成2组,每组10个重复,每个重复1头母猪,对照组饲喂基础饲粮,试验组在对照组基础饲粮中添加2 000 mg/kg HMB。试验从母猪妊娠第74天开始,至仔猪21日龄断奶结束。结果表明:与对照组相比,试验组仔猪初生个体重显著提高14.1%(P<0.05),仔猪7、21日龄窝重分别显著提高16.6%、11.7%(P<0.05),仔猪初生至7日龄、初生至21日龄窝增重分别显著提高28.3%和12.5%(P<0.05);与对照组相比,试验组母猪在妊娠第98天和分娩当天血清免疫球蛋白G(IgG)含量分别提高35.3%(P<0.01)、13.0%(P>0.05),免疫球蛋白M(IgM)含量分别提高34.7%(P<0.05)、12.6%(P>0.05);与对照组相比,试验组母猪初乳中IgG含量显著提高19.0%(P<0.05),分娩后第14天常乳中IgM含量显著提高21.4%(P<0.05);与对照组相比,试验组仔猪7日龄血浆IgG、IgM含量分别显著提高11.4%和40.1%(P<0.05)。上述结果提示,母猪妊娠后期及哺乳期饲粮中连续添加2 000 mg/kg HMB,可显著提高仔猪初生重和哺乳期增重,改善母猪及仔猪免疫功能。  相似文献

Samples of urine and serum from 45 newborn rottweiler puppies from six litters, and milk from their mothers, were taken 24, 48 and 72 hours and seven and 14 days after birth. Urine total protein and creatinine concentrations were determined and the ratios calculated. The immunoglobulin (Ig) concentrations of IgG, IgM and IgA in urine, serum and milk were determined with a commercially available elisa kit. The concentration of total protein in urine decreased from 1.64 to 0.29 mg/ml, and it and the ratio of total protein to creatinine in the urine of the neonatal puppies exceeded the normal values for adult dogs, but all the puppies developed normally. The average concentration of IgG in urine decreased from 0.0035 to 0.0003 mg/ml, that of IgA from 0.0035 to 0.0002 mg/ml and that of IgM from 0.0006 mg/ml to undetectable levels after two weeks. After two weeks, 47 per cent of the puppies had measurable levels of IgA and 70.6 per cent had measurable levels of IgG, but none of them had measurable levels of IgM.  相似文献

The development of the antibody concentration against lipopolysaccharide (LPS) of S. Typhimurium und S. Choleraesuis in rearing pigs during the fattening period and in breeding sows of the corresponding age was recorded. The studies revealed the following results. Antibodies of isotypes IgG1 and IgG2 revealed a more pronounced specificity against the according Salmonella serovar than IgM antibodies. The calculated "antibody percent value" based on the total amount of Salmonella antibodies is mainly determined by the IgM antibodies in sera and meat juice, respectively. In fattening pigs a significant increase of antibodies against IgM and total Ig was observed between week 3 and 10 after beginning of the rearing period. In breeding pigs this increase was detectable already earlier. In only 3 out of 10 groups an increase of IgG1 and IgG2 was also seen. The detected significant increase of total Ig and IgM in the other groups might be the result of a less intensive exposure to salmonellas or it might be due to an increase of unspecific antibodies induced by other antigens. Serological investigations represent a valuable tool to record the intensity and development in time of the Salmonella exposure in pigs farms. Examination of total Ig is an appropriate method to detect pig herds with a high level of Salmonella exposure, for detailed epidemiological studies in pig farms the examination of antibody isotypes will give more comprehensive information.  相似文献
